Experimental studies have highlighted the potential influence of contaminants on marine mammal immune function and anthropogenic contaminants are commonly believed to influence the development of diseases observed in the wild. However, estimates of the impact of contaminants on wild populations are constrained by uncertainty over natural variation in disease patterns under different environmental conditions. We used photographic techniques to compare levels of epidermal disease in ten coastal populations of bottlenose dolphins (Tursiops truncatus) exposed to a wide range of natural and anthropogenic conditions. Epidermal lesions were common in all populations (affecting > 60% of individuals), but both the prevalence and severity of 15 lesion categories varied between populations. No relationships were found between epidermal disease and contaminant levels across the four populations for which toxicological data were available. In contrast, there were highly significant linear relationships with oceanographic variables. In particular, populations from areas of low water temperature and low salinity exhibited higher lesion prevalence and severity. Such conditions may impact on epidermal integrity or produce more general physiological stress, potentially making animals more vulnerable to natural infections or anthropogenic factors. These results show that variations in natural environmental factors must be accounted for when investigating the importance of anthropogenic impacts on disease in wild marine mammals.  相似文献

Contaminant related health risks to marine mammals are typically inferred from the levels of contaminants measured in blubber. Such measurements alone are insufficient to indicate the likelihood of biological effects from contaminant exposure, especially for contaminants that do not bioaccumulate. Cytochrome P450 1A1 (CYP1A1) in mammals is induced by, and involved in, the metabolism of planar halogenated aromatic hydrocarbons and polycyclic aromatic hydrocarbons, chemicals of concern in aquatic systems. CYP1A induction is a molecular response to exposure to these inducers in many vertebrates. Using immunohistochemistry, we semiquantitatively measured CYP1A1 expression in integument (epidermis and blubber) collected by biopsy or at necropsy from 17 species of cetaceans. CYP1A1 expression was detected in all species and, in some cases, varied both within and between species. CYP1A1 expression in mysticetes was comparable to that in odontocetes. Assessing how the differences in contaminant burdens, life history parameters, and physiological condition between individuals, populations, or species affect CYP1A1 expression in cetacean integument is essential to the interpretation of this induction as a biomarker of exposure to and effects of contaminants. Detection of CYP1A1 expression in integument samples offers a relatively simple, non-lethal technique to study biological changes associated with contaminant exposure in cetaceans.  相似文献

Marine mammals have been subject to heavy anthropogenic pressure by direct killing and chemical pollution all over the world. Most studies of contamination and biomarker responses in marine mammals have been conducted using animals killed by hunting out of a total of 12 cetacean species studied, 45 of the specimens were obtained by sacrificing the animal; out of a total of eight pinniped species studied, 40 of the specimens were obtained by killing. The development of a series of non destructive techniques to evaluate biomarker responses and residue levels is recommended for the hazard assessment and conservation of endangered species of marine mammals. Here we review the current status of the non destructive biomarker approach in marine mammals, describing the biological materials available for non destructive tests in stranded brain, liver, blood, skin, subcutaneous blubber, muscle and fur and free ranging animals blood, skin biopsy, fur and faeces and the respective biomarker techniques mixed function oxidase activity and DNA damage in skin biopsy samples; porphyrins in faeces and fur; esterases, porphyrins, clinical biochemical parameter, vitamin A and micronuclei in blood samples. Residue analysis can be carried out in the various biological materials. Virus-associated mass mortalities among several marine mammal populations inhabiting industrialized coastal areas have generated an interest in wildlife immunotoxicology. Despite the isolation of previously uncharacterized viruses from victims, a contribution of immunotoxic contaminants to the severity of the outbreaks could not be ruled out. Fish-eating marine mammals, including seals, occupy high trophic levels in the aquatic food chain, and accumulate high levels of contaminants including polychlorinated biphenyls (PCBs), polychlorinated dibenzo-p-dioxins (PCDDs), and polychorinated dibenzofurans (PCDFs). Such chemicals have been found to be immunotoxic at low doses in studies of laboratory animals. While associations have been established between environmental contaminants and various adverse biological effects in certain free-ranging seal populations, evidence for immunotoxicity has, until recently, been lacking. To this end, we carried out an immunotoxicological study, in which captive harbor seals were fed herring from either relatively uncontaminated sites of the Atlantic Ocean, or from the highly contaminated Baltic Sea. In this review, we summarize the contaminant-related immunosuppression observed in the captive group of seals fed herring from the Baltic Sea, and discuss these results in the context of what is currently known about outbreaks of virus infection, comparative immunology, and environmental contaminants. We also describe two parallel studies, in which laboratory rats exposed as adults or perinatally to the contaminants in the Baltic Sea herring, exhibited immunotoxicity. On the basis of these and other studies, we conclude that complex mixtures of environmental contaminants may represent a real immunotoxic risk to free-ranging marine mammals in many areas of Europe and North America.  相似文献

Fatty acid signature analysis (FASA) is a powerful ecological tool that uses essential fatty acids (FA) from the tissues of animals to indicate aspects of diet. However, the presence of vertical stratification in FA distribution throughout blubber complicates the application of FASA to marine mammals. Blubber biopsy samples were collected from adult female southern elephant seals (Mirounga leonina) from Macquarie Island (n=11), and blubber cores were divided into inner and outer sections to determine the degree to which the blubber layer was stratified in FA composition, we found 19 FA from both blubber layers in greater than trace amounts (>0.5%). The inner and outer blubber layers could be separated using principal components analysis based on the relative proportion of FA in each layer. Dietary polyunsaturated fatty acids (PUFA) were also observed in significantly higher proportions in the inner blubber layer. Due to the degree of FA stratification in southern elephant seals, we concur with other marine mammal studies that sampling only the outer blubber layer will result in a loss of recently accumulated information regarding diet structure (as indicated by 'surplus' PUFA from the diet). This finding suggests that differential mobilization/deposition of certain FA may result in a modified signature from prey to predator. Thus, sampling animals to recover the inner blubber layer is important for studies attempting to describe aspects of marine mammal diet. This can be achieved in animals such as pinnipeds where the whole blubber layer can be readily sampled.  相似文献

Coastal dolphins are regarded as indicators of changes in coastal marine ecosystem health that could impact humans utilizing the marine environment for food or recreation. Necropsy and histology examinations were performed on 35 Indian Ocean bottlenose dolphins (Tursiops aduncus) and five Indo-Pacific humpback dolphins (Sousa plumbea) incidentally caught in shark nets off the KwaZulu-Natal coast, South Africa, between 2010 and 2012. Parasitic lesions included pneumonia (85%), abdominal and thoracic serositis (75%), gastroenteritis (70%), hepatitis (62%), and endometritis (42%). Parasitic species identified were Halocercus sp. (lung), Crassicauda sp. (skeletal muscle) and Xenobalanus globicipitis (skin). Additional findings included bronchiolar epithelial mineralisation (83%), splenic filamentous tags (45%), non-suppurative meningoencephalitis (39%), and myocardial fibrosis (26%). No immunohistochemically positive reaction was present in lesions suggestive of dolphin morbillivirus, Toxoplasma gondii and Brucella spp. The first confirmed cases of lobomycosis and sarcocystosis in South African dolphins were documented. Most lesions were mild, and all animals were considered to be in good nutritional condition, based on blubber thickness and muscle mass. Apparent temporal changes in parasitic disease prevalence may indicate a change in the host/parasite interface. This study provided valuable baseline information on conditions affecting coastal dolphin populations in South Africa and, to our knowledge, constitutes the first reported systematic health assessment in incidentally caught dolphins in the Southern Hemisphere. Further research on temporal disease trends as well as disease pathophysiology and anthropogenic factors affecting these populations is needed.  相似文献

Marine pollution has been studied under the following groupings of effects; harm to living resources, hazards to human health, reduction of amenities and interference with other users of the sea. This paper is concerned mainly with the first two categories and their interrelation. Apart from certain seabirds affected by oil, the major stocks of marine animals show no evidence of reduction by pollution. Pollution effects are generally insignificant in relation to other factors governing reproductive success, survival, growth and population size. Even in the North Sea, which has received a greatly increased pollution load during the last three decades, both total production of fish and catch per unit of effort (a measure of abundance) of cod, haddock and plaice increased during the 20 years 1950--69. Very recent decreases have been due to over-exploitation but, except in certain estuaries and immediate coastal waters, direct damage by pollution to marine populations and ecosystems is not evident. Pollution effects can, however, be detected by chemical analysis. The paper examines human health risks arising in the marine environment, particularly from contaminated seafood, especially in relation to sewage pollution, metals such as mercury, cadmium and lead, synthetic organic substances and oil.  相似文献

Trends in wrack composition and biomass, and its relationship with the anthropogenic impact were studied along a coastal area in Nuevo Gulf (south Patagonia) in front of Puerto Madryn city. Beach-cast macroalgae composition was sampled from 1992 to 1999 in the Puerto Madryn beaches and in several other nearby beaches in 1993, 1996 and 1998. Historical information was based on local knowledge and observations reported by marine biologists who worked in the area. The botanical composition of the beach-cast macroalgae in Puerto Madryn indicates a succession in the dominance from Codium spp. to Ulva spp. during the 1990s and from Ulva to Undaria pinnatifida since 1998, accompanied by a significant decrease in biomass of Gracilaria gracilis and Macrocystis pyrifera. The increase of the opportunist species such as Ulva may be supported by the continuous delivery of waste waters into the Nuevo Gulf while the dominance of U. pinnatifida may be associated with port activities. During the sampling period the highest wrack biomass values were recorded in spring and summer. The beach-cast seaweed biomass harvested by the municipality of Puerto Madryn during beach cleaning operation, ranged between 2500 and 12000 t year–1 ( 200 and 960 t dry weight). Wrack harvesting produces an environmental impact by removing sand from the beach and affecting coastal communities. Composting of wrack is proposed as one of the environmental alternatives to land disposal.  相似文献

Once the moult patterns have been taken into account, feather methylmercury levels can be used to accurately measure the mercury burdens of seabirds. We used body feathers from live seabirds and from museum collections to examine geographical and temporal patterns of mercury contamination in the North Sea. This approach identifies an increase in mercury concentrations in seabirds of the German North Sea coast during the last 100 years, especially high levels during the 1940s, and reduced contamination in the last few years. Comparisons among populations suggest that some increases in mercury levels are predominantly due to local pollution inputs, as on the German coast, while in other areas deposition from jet stream circulation of global contamination may be the major contributor. Mercury levels are far higher in seabirds from the German North Sea coast than in populations from the north and west North Sea or from most areas of the North Atlantic. We advocate the use of museum collections of birds for studies of long-term changes in levels of mercury contamination.  相似文献

A series of high profile outbreaks of newly described diseases in humans, domestic animals and wildlife has attracted widespread interest in the topic of Emerging Infectious Diseases (EIDs). Marine mammals are no exception: since 1987 several mass mortalities have been observed following infection with viruses previously undescribed in the populations or species in question. As with terrestrial examples, some of these outbreaks have followed either migrations associated with large-scale ecological changes or the introduction of virus from domestic animals. However, marine mammals warrant special concern in the context of emerging infectious diseases: they typically occupy high trophic levels and can therefore be highly contaminated with immunotoxic chemicals. Persistent Organic Pollutants (POPs), including polychlorinated -biphenyls (PCBs), dibenzo-p-dioxins (PCDDs), -dibenzofurans (PCDFs) and related compounds, are demonstrated immunotoxicants in laboratory animals, as well as marine mammals. Immunotoxic contaminants may represent a factor that facilitates disease emergence, and may lead to the creation of susceptible “reservoirs” for new pathogens in contaminated marine mammal populations. The factors underlying the emergence and exchange of pathogens among marine mammals, domestic animals, and humans demand multidisciplinary study and invite regulatory and conservation scrutiny. The complexity of this issue may be best addressed through an integrated human and ecological risk assessment framework.  相似文献

Until recently the commercial collection of beach-cast seaweeds in New Zealand was prohibited but the legislation has recently been amended to allow permitting of this activity. This review collates existing information on the role of beach-cast seaweed in coastal ecosystems to describe the nature and extent of the effects that commercial removals of beach cast seaweed may have on the marine environment. It outlines the amount of beach-cast seaweed available for harvest in New Zealand and the fate of seaweed when not collected; reviewing current information on the importance of beach-cast seaweeds and its inhabitants on: feeding and nesting shorebirds, and nutritional contribution of seaweed inhabitants to nearshore coastal ecosystems when seaweed is washed back into the sea It also identifies key research gaps related to any environmental impacts that the removal of beach cast seaweed might have.  相似文献

Environmental pollution often accompanies the expansion and urbanization of human populations where sewage and wastewaters commonly have an impact on the marine environments. Here, we explored the potential for faecal bacterial pathogens, of anthropic origin, to spread to marine wildlife in coastal areas. The common zoonotic bacterium Campylobacter was isolated from grey seals (Halichoerus grypus), an important sentinel species for environmental pollution, and compared to isolates from wild birds, agricultural sources and clinical samples to characterize possible transmission routes. Campylobacter jejuni was present in half of all grey seal pups sampled (24/50 dead and 46/90 live pups) in the breeding colony on the Isle of May (Scotland), where it was frequently associated with histological evidence of disease. Returning yearling animals (19/19) were negative for C. jejuni suggesting clearance of infection while away from the localized colony infection source. The genomes of 90 isolates from seals were sequenced and characterized using a whole‐genome multilocus sequence typing (MLST) approach and compared to 192 published genomes from multiple sources using population genetic approaches and a probabilistic genetic attribution model to infer the source of infection from MLST data. The strong genotype‐host association has enabled the application of source attribution models in epidemiological studies of human campylobacteriosis, and here assignment analyses consistently grouped seal isolates with those from human clinical samples. These findings are consistent with either a common infection source or direct transmission of human campylobacter to grey seals, raising concerns about the spread of human pathogens to wildlife marine sentinel species in coastal areas.  相似文献

Although eutrophication is a widespread problem in marine waters, its effects are often difficult to separate from normal fluctuations of pelagic coelenterate populations and from other anthropogenic changes due to industrial pollution, construction, introductions, global warming and overfishing. The least complex situations are in small coastal water bodies such as the Caribbean lagoons and Scandinavian fjords. Typically, the diversity of pelagic coelenterates decreases, but the biomass of a small number of species (such as the hydromedusae Aglantha digitale and Rathkea octopunctata and the scyphomedusae Aurelia aurita and Cassiopea spp.) may increase. Adaptations that may allow these species to survive under eutrophic conditions are discussed.  相似文献

Organochlorine (OC) pesticides and polychlorinated biphenyls (PCBs) have been detected in a variety of marine mammal species at levels associated with adverse health effects. Little is known about OC levels and impacts on health in pinnipeds with different life histories. We determined the health and levels of 18 OC pesticides and 16 PCB congeners in blubber samples from 20 Steller sea lions and 39 Pacific harbor seals stranded from Oregon and Southern Washington. The most commonly detected OC at the highest concentration was p,p′- dichlorodiphenyldichloroethylene (DDE). PCBs were detected in all samples as well. Hypothesis testing indicated that diseased Steller sea lions (males and females combined) had higher contaminant concentrations than healthy Steller sea lions, and diseased Pacific harbor seals had higher concentrations of total OCs than healthy animals. Differences were also noted between diseased and healthy animals when looking at individual sexes of each species. Diseased Steller sea lions had higher mean contaminant levels than diseased harbor seals and healthy Steller sea lions had higher mean contaminant concentrations than healthy Pacific harbor seals. These results show that species differences exist in both contaminant loads and sensitivity to contaminants, which may be due to differences in life histories and physiology.  相似文献

This is of importance in assessing water quality regarding pollution with genotoxic compounds, food safety analysing edible aquatic animals and in terms of ecotoxicology. Covalent modification of DNA is considered a crucial event in chemical carcinogenesis and thus may be considered a biomarker of an early genotoxic effect. Measuring DNA adducts is unique in that these lesions may be considered a biomarker of both exposure and effect. A number of studies have described the analysis of the DNA isolated from the liver of both freshwater and marine fish. Considerable levels of DNA adducts have been observed in some animals from contaminated lakes or rivers. Low levels were observed in DNA from the liver of marine fish. The background levels of DNA adducts have to be determined in animals from appropriate uncontaminated control sites that are matched for species, gender, age and season of the year. Despite the openness of the oceanic environment, limited dispersal and tight social structure often induce genetic structuring in marine organisms, even in large animals such as cetaceans. In the bottlenose dolphin, mitochondrial and nuclear DNA analyses have revealed the existence of genetic differentiation between pelagic (or offshore) and coastal (or nearshore) ecotypes in the western North Atlantic, as well as between coastal populations. Because previous studies concentrated on continental margins, we analysed the population structure of bottlenose dolphins in two of the most isolated archipelagos of the North Atlantic: the Azores and Madeira. We analysed 112 samples collected on live animals in the two archipelagos, and nine samples collected on stranded animals in Madeira and mainland Portugal. Genetic analyses consisted in molecular sexing, sequencing of part of the mitochondrial hyper-variable region, and screening of ten microsatellite loci. We predicted that: (1) there is at least one pelagic and one or more coastal populations in each archipelago; (2) populations are differentiated between and possibly within archipelagos. Contrary to these predictions, results indicated a lack of population structure in the study area. In addition, comparison with published sequences revealed that the samples from the Azores and Madeira were not significantly differentiated from samples of the pelagic population of the western North Atlantic. Thus, bottlenose dolphins occurring in the pelagic waters of the North Atlantic belong to a large oceanic population, which should be regarded as a single conservation unit. Unlike what is known for coastal populations, oceanic bottlenose dolphins are able to maintain high levels of gene flow.  相似文献

Sublethal effects of pollution may be significant to survival of a stock of marine fish or even a species. Such effects sometimes lead to reproductive failure and have been identified so far only in freshwater systems. Atlantic salmon have disappeared from many streams in Europe and eastern North America, partly as a result of pollution in their freshwater spawning areas and in their estuarine nursing grounds. Reductions in populations of marine fishes due to pollution solely have not yet been demonstrated. However, Baltic Sea seals, where reproductive failure is apparently associated with high concentrations of DDT and polychlorinated biphenyl in the blubber, may have suffered a decline owing to the presence of these organochlorines. Sublethal effects of pollutants have been studied in the laboratory, essentially under four categories: (1) physiology (growth, swimming performance, respiration, circulation); (2) biochemistry/cell structure (blood chemistry, enzyme activity, endocrinology, histochemistry); (3) behaviour/neurophysiology; and (4) reproduction. Not all pollutants elicit meaningful responses in all categories, and a response is not always linear with pollutant concentration. For application to survival of populations the response has to be ultimately related to a healthy progression through a full life cycle, including successful reproduction. In recent time, physiological studies have moved into polluted marine environments with mobile laboratories having continuous sampling capability, to observe effects of pollutants in situ on marine organisms. The Controlled Ecosystem Pollution Experiment (Cepex) in Saanich Inlet, British Columbia, endeavours to investigate the effects of low concentrations of pollutants on marine organisms in large plastic silos having a slow replacement of water.  相似文献
