海藻多糖抑制白细胞呼吸爆发作用研究   总被引:15,自引:0,他引:15  
采用ESR、自旋捕集及自旋氧探针技术,研究了海藻硫酸多糖(SPS)对豆蔻酰佛波醇乙酯(PMA)刺激的多形核白细胞(PMN)呼吸爆发的影响.结果表明,SPS能显著抑制PMN呼吸爆发,10 g/L和5 g/L SPS几乎完全清除PMN呼吸爆发产生的自由基,1 g/L SPS可清除53.2%;10 g/L SPS对PMN的耗氧量也有较明显的抑制作用.  相似文献

稀土La3+跨PC12细胞膜行为研究   总被引:1,自引:0,他引:1  
使用AR-CM-M1C阳离子测定系统,发展Fura-2荧光测定技术,将其应用于测定细胞内游离稀土离子La3+,并以此研究了La3+跨PC12细胞(大鼠嗜铬细胞瘤细胞)膜的行为.结果表明:在模拟细胞内离子组分,pH=7.05的溶液中,测得La3+-Fura-2的表观解离常数为3.27×10-11 mol·L-1.对于PC12细胞,静息条件下La3+不能跨越细胞膜进入胞内.与钙离子通道相关的KCl和去甲肾上腺素均不能刺激稀土La3+过膜.用哇巴因(ouabain)使胞内Na+超载后,La3+可过膜进入细胞内,且过膜量与胞外La3+浓度和胞内Na+超载程度有一定的浓度依赖关系,提示La3+可以经由Na+/La3+交换机制过膜而进入细胞内.  相似文献

真核细胞内空泡细胞器,如高尔基体、内质网、溶酶体等,膜上存在的质子泵ATPase 与线粒体类型的质子泵 ATPase 类似.近几年对该类型 H+-ATPase 的结构、作用机制进行了深入的研究,证明这是一类新型质子泵,在进化的过程中与线粒体类型的 H+-ATPase 有密切的亲缘关系.  相似文献

脉冲电场引起的红血球内钠离子浓度变化的研究   总被引:1,自引:0,他引:1  
利用位移试剂和23Na-NMR的方法研究脉冲电场对正常人红血球内Na+浓度的影响,实验结果给出在高强度电场作用下,细胞内Na+浓度增加,并且随脉冲强度的增加而增加,比指数关系还快.在低强度电场作用下,细胞内Na+浓度减少.乌苯苷能抑制细胞内Na+浓度的减少,抑制程度随乌苯苷浓度的增加而增强,从而证实了低强度的脉冲电场对Na+,K+-ATPase的激活作用,直接测定脉冲电场对红血球血影膜的Na+,K+-ATPase活性的影响,进一步证实了这一结果.并对在电场作用下细胞膜的通透性和电场对酶的激活作用及电场等外界物理信号是否能跨过细胞膜等进行了讨论.  相似文献

Na+/H+交换泵(Na+/H+ exchanger, NHE)是存在于所有脊椎动物细胞中的重要跨膜蛋白,该蛋白质涉及细胞的多种功能,包括细胞内pH值调节、细胞体积的控制以及离子转运等.目前已克隆了五个亚型NHE的cDNA,它们构成了脊椎动物细胞离子转运泵的一个基因家族. 这五个亚型的表达水平及活性可受多种因素的调节.在肿瘤、高血压及糖尿病等疾病中,已发现NHE-1亚型的表达水平和活性显著增高.因此,研究NHE-1的转录及活性调节机制,将可能为这些疾病的诊治提供新的手段.  相似文献

以二硫苏糖醇(DTT)或L-抗坏血酸(Vit C)为终止剂改进的经典邻苯三酚法测定超氧化物歧化酶(SOD)活性.反应体系:50mmol/L邻苯三酚,pH8.20,总体积9ml,25℃,加一滴DTT(100mmol/L)或Vit C(5%),约50μl,终止自氧化反应.终止后,反应体系的420nm光吸收在lh内保持恒定.终止剂法的邻苯三酚自氧化率及酶活测定灵敏度与经典法相近,一个酶活单位相当于纯SOD 100μg/L.  相似文献

研究了5个氨基酸的磷光特性、光谱、寿命和最小检测量.以色氨酸(Trp)的磷光最强,在λexem=290/438 nm,最小检测量为1 ng,酪氨酸(Tyr)其次(284/390 nm)为Trp的1/10,苯丙氨酸(Phe)在276/386 nm,脯氨酸(Pro)在308/454 nm,组氨酸(His)在320/466 nm,其磷光只有Trp的1%.Phe与Trp磷光寿命最长,在7 s左右;His最短,只有0.49 s;Tyr 2.8 s,Pro 1.34 s.另外研究氨基酸在甲醇,乙醇,丙醇,丁醇中及pH对氨基酸磷光性质的影响.还计算了Stokes位移能量及激化态pK*a值.  相似文献

用ESR自旋捕集技术研究了人多形核白细胞(PMN)受促癌剂佛波醇(PMA)刺激产生O2-和NO自由基的相互作用.发现加L-精氨酸使在PMA刺激PMN体系中捕捉到的O2-明显减少,加NG-甲基精氨酸(NGMA)使在PMA刺激PMN体系中捕捉的O2-明显增加.用黄嘌呤/黄嘌呤氧化酶和光照核黄素体系证明,加L-精氨酸使PMA刺激PMN产生NO-·自由基与O2-结合生成ONOO-是加入L-精氨酸使PMA刺激PMN体系捕捉的O2-减少的主要原因,并且推算了加入不同浓度L-精氨酸PMN产生NO-·自由基的量.用羟基自由基清除剂和ONOO-氧化DMPO和DMSO及其对pH的依赖关系,证明了ONOO-分解并没有直接生成羟基自由基.用依赖鲁咪诺的化学发光法研究了人多形核白细胞受促癌剂PMA刺激产生NO-·自由基动力学过程.另外,用化学合成的NO-·自由基和过氧亚硝基在模型体系研究了它们的ESR和化学发光特征.说明PMA刺激PMN生成的NO-·和O2-自由基反应形成的ONOO-是引起化学发光的主要形式.  相似文献

通过气溶胶发生系统模拟PM2.5颗粒的发生,运用15N示踪技术研究了欧美杨107(Populus euramericana Neva.)对PM2.5中水溶性无机成分NH+4和NO-3的吸收与分配规律。结果表明,欧美杨能够有效吸收PM2.5中的NH+4和NO-3。轻度和重度污染下,欧美杨叶片对NH+4和NO-3的吸收速率均于处理后第1天达到峰值,之后,轻度污染下对NH+4和NO-3的吸收速率迅速降低以后趋于稳定,而重度污染下对NH+4和NO-3的吸收速率缓慢下降至趋于稳定。轻度污染下的欧美杨叶片的15N含量在处理后第1天达到峰值,15N(NH+4)的含量为0.11 mg/g,干重,15N(NO-3)的为0.14 mg/g,干重,之后15N含量迅速下降至趋于稳定。重度污染下的叶片15N含量在处理第1天迅速增长,之后缓慢增长至处理后第7天达到最高值,15N(NH+4)的含量为0.11 mg/g,干重,15N(NO-3)的为0.13 mg/g,干重。处理7 d后,欧美杨不同组织器官吸收或通过再分配获取的15N含量存在差异。轻度污染下,细根对NH+4和NO-3的吸收量最高,树皮、叶柄、叶片次之,髓最低。而重度污染下,叶片对NH+4和NO-3的吸收量最高,细根、叶柄、树皮次之,髓最低。欧美杨各组织器官中NH+4和NO-3的含量均表现为重度污染大于轻度污染,且两种污染程度下的欧美杨各组织器官对NO-3的吸收均大于对NH+4的吸收。重度污染下,欧美杨茎木质部对15N(NH+4和NO-3)的吸收征调能力(Ndff,Nitrogen derived from fertilizer)最大,其次为髓,叶片最小;欧美杨各组织器官中的15N分配率表现为叶片细根叶柄树皮粗根茎木质部髓。研究结果对进一步揭示植物吸收PM2.5的机制及有效利用植物降低颗粒物污染、净化环境提供了重要的科学理论依据。  相似文献

G protein coupled receptor (GPCR) signalling is mediated by transactivation independent and transactivation dependent pathways. GPCRs transactivate protein tyrosine kinase receptors (PTKRs) and protein serine/threonine kinase receptors (PS/TKR). Since the initial observations of transactivation dependent signalling, there has been an effort to understand the mechanisms behind this phenomena. GPCR signalling has evolved to include biased signalling. Biased signalling, whereby selected ligands can activate the same GPCR that can generate multiple signals, but drive only a unique response. To date, there has been no focus on the ability of biased agonists to activate the PTKR and PS/TKR transactivation pathways differentially. As such, this represents a novel direction for future research. This review will discuss the main mechanisms of GPCR mediated receptor transactivation and the pathways involved in intracellular responses.  相似文献

在植物细胞内,除了顺向的信号转导通路,即核基因控制着质体基因的转录和翻译之外,还存在着逆向的信号转导通路,即质体的代谢状况作为一种信号去调控核基因的表达。过去对这条逆向的信号转导通路,亦称质体因子,研究得非常少。近几年来,随着对基因组解偶联突变体的深入研究,人们对这条通路的认识大大加深了。现着重介绍质体中的四吡咯代谢中间产物参与信号的产生,以及质体向细胞质搬运这些中间产物启动了对编码质体蛋白的核基因的表达调控。  相似文献

Guanosine 3', 5'-cyclic monophosphate (cGMP) signalling has received increasing attention over the last decade, since the discovery of the gaseous signalling molecule, nitric oxide, which activates cGMP synthesis. Furthermore, research into cGMP signalling has also been stimulated by the development of Viagra and pharmacologically active related compounds, which act to prevent cGMP breakdown. While much is known about the biochemical aspects of components of the cGMP signalling pathway, the precise in vivo roles of such components have only recently come to light through work in model organisms. This review outlines recent work utilising the genetic model organism Drosophila melanogaster in studies of organotypic cGMP signalling. While organisms such as Drosophila may not be the obvious choice for such studies, use of this model has proved that unique and detailed insights for cGMP signalling can be achieved.  相似文献

Bioactive peptides are a group of diverse intercellular signalling molecules. Almost half a century of research on this topic has resulted in an enormous amount of data. In this essay, a general perspective to interpret all these data will be given. In classical endocrinology, neuropeptides were thought of as simple signalling molecules that each elicit one response. However, the fact that the total bioactive peptide signal is far from simple puts this view under pressure. Cells and tissues express many different bioactive peptides and they are also able to respond to many different bioactive peptides, indicating that multiple receptors and signal transduction pathways are present in a single cell. Therefore, the authors suggest that the bioactive peptide signalling system should be regarded in the context of network and systems biology. Bioactive peptides can best be viewed as an extension of the protein interaction network that allows regulating and fine‐tuning the metabolism of the different cells and tissues in the body. The cell thus responds to the ‘peptidome’ instead of to a single peptide. The intracellular part of this signalling network consists of the various signalling transduction cascades. Recently, new systems biology approaches have emerged for the modelling of cell signalling. The network and systems biology approach is also able to shed new light on the evolution of intercellular signalling.  相似文献

Reciprocal altruism has been the backbone of research on the evolution of altruistic behaviour towards non-kin, but recent research has begun to apply costly signalling theory to this problem. In addition to signalling resources or abilities, public generosity could function as a costly signal of cooperative intent, benefiting altruists in terms of (i) better access to cooperative relationships and (ii) greater cooperation within those relationships. When future interaction partners can choose with whom they wish to interact, this could lead to competition to be more generous than others. Little empirical work has tested for the possible existence of this 'competitive altruism'. Using a cooperative monetary game with and without opportunities for partner choice and signalling cooperative intent, we show here that people actively compete to be more generous than others when they can benefit from being chosen for cooperative partnerships, and the most generous people are correspondingly chosen more often as cooperative partners. We also found evidence for increased scepticism of altruistic signals when the potential reputational benefits for dishonest signalling were high. Thus, this work supports the hypothesis that public generosity can be a signal of cooperative intent, which people sometimes 'fake' when conditions permit it.  相似文献

Bone is a complex, evolving tissue, architecturally defined by the activities of osteoclasts and osteoblasts that continually resorb and replace the mineralised matrix. Numerous regulatory mechanisms exist to control bone remodelling and the maintenance of bone mass. The consequences of inappropriate or uncoupled bone resorption and formation are significant and invariably lead to different disease states, the most prevalent being osteoporosis. In recent years, much attention has focused on unravelling the systemic and local signalling interactions that influence the differentiation and function of bone cells with a view to developing our understanding of bone biology and identifying potential new targets for therapeutic intervention. Several lines of evidence indicate that neurotransmitters and neuromodulators have influential roles to play in the regulation of bone remodelling and much of this research has involved analysis of the excitatory amino acid glutamate. This review will summarise current understanding of glutamate signalling in bone cells, addressing specifically the function of N-methyl-D-aspartate (NMDA)-type glutamate receptor signalling mechanisms, and will address the functional significance and future prospects for this area of research.  相似文献

Protein phosphorylation is a common signalling mechanism in both prokaryotic and eukaryotic organisms. Whilst the focus of protein phosphorylation research has primarily been on protein serine/threonine or tyrosine phosphorylation, there are other phosphoamino acids that are also biologically important. Two of the phosphoamino acids that are functionally involved in the biochemistry of protein phosphorylation and signalling pathways are phosphoaspartate and phosphoglutamate, and this review focuses on their chemistry and biochemistry. In particular, we cover the biological aspects of phosphoaspartate and phosphoglutamate in signalling pathways and as phosphoenzyme intermediates. In addition, we examine the synthesis of both of these phosphoamino acids and the chemistry of the acyl phosphate group. Although phosphoaspartate is a major component of prokaryotic two-component signalling pathways, this review casts its net wider to include reports of phosphoaspartate in eukaryotic cells. Reports of phosphoglutamate, although limited, appear to be more common as free phosphoglutamate than those found in phosphoprotein form.  相似文献

The innate immune system of insects relies on both humoral and cellular immune responses that are mediated via activation of several signalling pathways. Haemocytes are the primary mediators of cell-mediated immunity in insects, including phagocytosis, nodulation, encapsulation and melanization. The last years, research has focused on the mechanisms of microbial recognition and activation of haemocyte intracellular signalling molecules in response to invaders. The powerful tool, RNA interference gene silencing, helped several regulators involved in immune responses, to be identified. In this review, we summarize recent advances in understanding the role(s) of receptors and intracellular signalling molecules involved in immune responses.  相似文献
