Genetic diversity analysis by RAPD in Cathaya argyrophylla Chun et Kuang   总被引:5,自引:0,他引:5  
Genetic diversity level of Cathaya argyrophylla was confirmed by random amplified polymorphic DNA (RAPD) markers. Seventy five samples (individuals), collected from Hunan and Sichuan provinces of China were used in the study. 21 10-mer oligonucleotide primers detected 106 sites, and 34 (32% ) of them were polymor-phic. The level of genetic variation in C. argyrophylla was lower than those of other conifers, and was considered to be associated with the complexity of habitats. The percentages of polymorphic sites (PPS) in the Hunan and Sichuan pop-ulations were 18% and 25% respectively. 7.99% of genetic variation existed between the two populations; this value was higher than the mean value (6.8%) among populations in conifers displayed by allozyme. Some subpopulations of C. argyrophylla were greatly differentiated because of site mutation and genetic drift. The highest value of genetic dif-ference between subpopulations amounted to 16. 23% . 对蒙古羊、湖羊、滩羊、小尾寒羊、乌珠穆沁羊、藏绵羊、阿勒泰羊7个地方绵羊品种和无角陶赛特羊、德国美利奴羊、萨福克羊3个引入品种基因组DNA进行了RAPD分析。结果表明:(1)RAPD可作为一种有效的标记用于绵羊品种之间遗传亲缘关系的分析。(2)在所使用的43种随机引物中,有35种引物扩增出多态谱带,多态频率为66.24%,说明RAPD技术用于研究绵羊核DNA的遗传变异具有较高的检出率和灵敏度。(3)总群体平均遗传多样性指数(HSP)为0.9139,说明绵羊群体具有较为丰富的遗传多样性。(4)我国地方绵羊品种间的分子聚类关系与其所处的地理位置、考古学结果,以及细胞遗传学研究结果基本一致,引入品种间的分子聚类关系也与其育成史基本一致。 Abstract:The genetic polymorphism and relationship of 7 indigenous sheep breeds of China and 3 imported sheep breeds were studied using random amplified polymorphic DNA (RAPD).The results indicated that the RAPD was an effective marker for the analysis of genetic relationship among sheep breeds.Among 43 arbitrary primers,35 were polymorphic.The percentage of polymorphic markers was 66.24%,which indicated that the RAPD had higher efficiency of polymorphism detection and sensitivity in studying the genetic variation among sheep breeds.The average index of genetic polymorphism for whole population (Hsp) was 0.9139,which showed that the genetic polymorphism was abundant between sheep populations.The genetic relationship between different indigenous sheep breeds in China was in accord with their localities,the results from archeology and cytogenetics and the genetic relationship between imported sheep breeds was in accord with their breeding history.  相似文献

用RAPD技术对蓖麻蚕基因组DNA进行多态性研究,分析了5个蓖麻蚕品种间的遗传差异。结果表明,所采用的40个随机引物中,有27个引物扩增谱带清晰且重复性较好,扩增总片段数达243个,单个引物的扩增片段数在4~17之间,平均为9条,片段大小在0.33~3.0kb之间。不同蓖麻蚕品种间的遗传距离(D)在0.0683~0.1603之间,根据D值,由UPGMA聚类分析软件绘制了它们的聚类分子树。 Abstract:Random amplified Polymorphic DNA (RAPD) was used to analyze the genetic diversity among eri sickworm. The genetic variance of five erisickworm was studied. The result showed that: 27 of 40 arbitrary primers could amplify clearly with repeatable bands.243 fragments were obtained.Each primer gave 4~17 bands and the average was 9.The length of the band was 0.33~3.0kb. Random amplified Polymorphic DNA(RAPD) analysis is a new technology of molecular marking which has proved very powerful in detecting genetic diversity at the level of population.The genomic DNAs used in our experiment were extracted from fresh leaves taken from 59 individuals sampled from three natural populations in Yan an,Shanxi Province.Through more than 2,000 PCRs,deep-going RAPD analysis was carried out on DNA samples from 49 inviduals.The percentage of polymorphic RAPD loci found in these three populations were respectively 27.2%,18.6% and 5.4%;the average genetic distances within population,0.055,0.026 and 0.008;the average genetic distances between populations (I-II),(I-III) and (II-III),0.105,0.096 and 0.060.The genetic diversity of A.brachypus within and between populations was found,for the first time,to be rather poor,thus revealing innate factors as the cause contributing to its endangered status.In addition,our work also provides basic materials for elucidating the underlying cause of its endangerment and for its protection biology.  相似文献

Randomly amplified polymorphic DNA (RAPD) analysis was used to investigate the genetic variation among populations, between populations, and within populations, relationships between genetic distance and geographic distance, and the molecular variation and population size. The effects of geographic and genetic distances, as well as of genetic differentiation and population size, on genetic variations of Leymus chinensis (Trin.) Tzvel. are discussed. The present study showed that there was significant RAPD variation between the Baicheng region population and the Daqing region population, with a molecular variance of 6.35% (P 〈 0.04), and for differentiation among area populations of the Daqing region, with a molecular variance of 8.78% (P 〈 0.002). A 21.06% RAPD variation among all 16 populations among two regions was found (P 〈 0.001), as well as 72.59% variation within populations (P 〈 0.001). The genetic diversity of Stipa grandis P.Smirn and its relationship with the climatic variables were studied using the RAPD technique for 90 genes from five natural populations sampled in the Xilingol steppe, China. Sixteen oligonucleotides screened from 100 random primers were used to amplify 310 trackable RAPD loci, which were all polymorphic. By analyzing the RAPD data using POPGENE software, different geographic S. grandis populations were studied, which indicated a high level of genetic diversity, and the maximum variation was observed within the populations with a 28% variation observed among the populations. Using Pearson correlation analysis, significant (P < 0.05) or highly significant (P < 0.01) relationships were found between gene diversity indexes and temperature factors (≥10°C cumulative temperature in a year, annual mean temperature and mean temperature in January). Mantel's tests showed that there was no significant correlation between Nei's unbiased genetic distance and the geographic distance of S. grandis populations (r = 0.184, P = 0.261). However, there were significant or highly significant correlations between Nei's genetic distance and the several climatic divergences in pairwise S. grandis populations. All results indicated that natural selection resulting from variations in water and temperature was responsible for the adaptive eco-geographical differentiation indicated by the RAPD markers of different S. grandis populations, and that immigration and gene drift did not play an important role in affecting the differentiation of S. grandis populations.  相似文献

本研究应用RAPD技术对湄洲岛和平潭岛上4个烟豆小种群进行的生态遗传学研究,结果显示,总的多态位点比率为1.0,4个种群中最小为0.41,最大为0.82,表现出高度的多态性。利用相似系数及遗传距离进行的聚类分析结果表明,在种群间地理距离较大时,遗传分化与地理距离有一定的相关性,在小范围内则无明显相关性。 Abstract:The ecological genetic research on Glycine tabacina populations was based on RAPD technique,which revealed 100% polymorphisms,with minimum value of 0.41 in population MM,and maximum value of 0.82 in population PT.Cluster analysis showed that the populations' genetic variation was correlation to the environment gradient when the geographic distance among populations was big.In small geographic range,however,no correlation exists between genetic structure and ecological factors because of random genetic drift.  相似文献

为探讨分子遗传标记在鸵鸟(Struthiocamelus)育种中的应用,利用RAPD技术分析了我国养殖的三个主要鸵鸟品种内和品种间的遗传变异.蓝颈鸵鸟、澳洲灰和非洲黑三个品种在品种内和品种间显示出低水平的变异.品种间的遗传距离较好地说明了三个品种的育种改良过程,同时预示出用澳洲灰与其他品种鸵鸟进行杂交育种的可行性.选择的20条在鸡中能稳定扩增的RAPD引物其75%能在鸵鸟基因组中扩增出产物,显示出两种禽类DNA具有同源性,为今后利用鸡的微卫星引物对鸵鸟进行基因组扫描(genomescan)分析奠定基础。 Abstract:To evaluate the molecular genetic markers on ostrich (Struthio camelus)breeding,we have analyzed the genetic variability of three main ostrich breeds by using RAPD.The studies show that the Blue,Australia Grey and African Black have low genetic variability.The genetic distance between breeds clearly illustrates the breeding history,at the same time,it also showes the possibility that cross breeding can be applied to Australia Grey and the breeds.75 percent selected RAPD primers working well in chicken could stably amplify products in ostriches,which implied DNA identity between chicken and ostrich.The work provided a foundation for genomic scanning.  相似文献

首次利用RMAPD标记对43只洋县人工饲养朱鹮群体的遗传多态性进行了分析。结果表明,RMAPD技术的稳定性与多态性比RAPD高。12对引物共扩增出2147条带纹,93条具有多态性。朱鹮的条带共享率为0.718,遗传多样性指数为3.664,反映群体内个体间相似程度较高,遗传多态性较低,需要加强对朱鹮的遗传多样性保护。  相似文献

利用蛋白质电泳、RAPD和微卫星DNA技术,对我国濒危保护动物———人工饲养朱进行遗传多态性研究。结果表明,40只朱在前白蛋白(Pre)、白蛋白(Alb)、后白蛋白(Pa)、运铁蛋白(Tf)共4个蛋白质位点上没有发现多态性。用40个随机引物对40只朱进行RAPD分析,其中35个引物扩增出完全相同的RAPD带纹,仅筛选到5个多态性引物,朱群体的平均带纹相似率为0.86。利用其它物种的22对微卫星引物对30只朱的DNA进行检测,共筛选出4对多态性微卫星引物,扩增出13个等位基因。朱在这4对微卫星位点的平均杂合度为0.3760,平均多态信息含量为0.3382,平均等位基因数为2.075。表明朱人工饲养群体的遗传多态性比发现之初得到了一定程度的恢复,但仍然比较贫乏  相似文献

构建高密度遗传连锁图谱是冰草抗性、品质、产量等重要性状QTL精细定位及标记辅助育种研究的基础。该试验以四倍体杂交冰草F2群体的202个分离单株及其亲本为材料,利用SRAP分子标记技术和Join Map 4.0作图软件对冰草的遗传连锁图谱进行了构建。结果表明:(1)共筛选出22对多态性好、标记位点清晰稳定的SRAP适宜引物,对冰草杂种F2分离单株的基因组DNA进行PCR扩增,共获得510个SRAP多态性标记位点,其比率占88.2%。(2)偏分离分析表明,偏分离标记比率仅为14.12%,符合遗传作图的要求。(3)成功构建了冰草的SRAP分子标记遗传连锁图谱,该图谱有14个连锁群、510个标记,连锁群间长度范围86.4~179.0cM,覆盖基因组总长度1 912.9cM,标记间平均间距3.75cM,为高密度遗传图谱。  相似文献

运用随机扩增多态DNA(RAPD)技术,对天目木兰(Magnolia amoena)居群的遗传多样性进行了研究.从40个10-mer随机引物中筛选出14个能得到清晰、稳定扩增带的引物进行扩增,14个引物共检测了94个位点,其中多态性位点为23,占24.4%,计算了12个居群之间的遗传相似度和遗传距离,并运用UPGMA法进行了聚类分析,结果显示相同严地个体间(居群内)的遗传距离较小,遗传多样性水平很低;不同产地个体间(居群间)遗传距离较大,遗传多样性水平较前者高,即天目木兰个体间遗传多样性水平与它的地理分布有关,天目木兰总体较低的遗传多样性是导致它濒危的原因之一。  相似文献

朱曾广泛分布于中国、日本、朝鲜半岛和俄罗斯的远东地区。其它国家的朱先后灭绝,目前只栖息于我国,为我国所特有,是我国四大国宝之一,当今世界极濒危级鸟类。1981年朱在陕西洋县重新被发现时其种群数量只有7只,经20余年的保护,目前我国朱的总数已达到700余只,人工繁殖也取得巨大成功。笔者想通过本文唤起人们对生活在西部边远地区的我国珍稀鸟类———朱的关注与保护。  相似文献

Construction of a genetic linkage map is necessary to apply marker-assisted selection tools in a crop improvement program. Except for the recent studies from two laboratories, most of the previous studies have shown little or no DNA polymorphism in cultivated groundnut (Arachis hypogaea L.). In the present study, 70 selected genotypes, representing variability for several morphological, physiological, and other characters, were studied for polymorphism employing random amplified polymorphic DNA (RAPD) assay with 48 oligonucleotide primers. Of the 48 oligonucleotide primers only 7 (14.6%) yielded polymorphic amplification products. The total number of bands from the 7 primers was 408, of which 27 were polymorphic. Detection of polymorphism in cultivated groundnut opens up the possibility of development of its molecular map by judicious selection of genotypes that show DNA polymorphism. This approach will be useful for developing marker-assisted selection tools for genetic enhancement of groundnut for desirable traits.  相似文献

采用简单序列重复区间扩增(ISSR)分子标记技术对广西地不容3个野生居群和1个引种居群共92个个体进行了遗传多样性研究。10个引物共扩增出61条带,其中60条具多态性,多态性位点百分率为98.36%。4个居群多态性百分率在73.77%~86.89%。Nei’s基因多样性指数(H)为0.3379,Shannon信息多样性指数(Ⅰ)为0.5055。3个野生居群Nei’s遗传分化系数(Gst)表明:83.87%遗传变异分布在居群内,16.13%的遗传变异分布在居群间。引种居群与3个野生居群间的遗传一致度达0.8846。引种居群有效地保护了广西地不容的遗传多样性。  相似文献

利用随机扩增多态DNA 技术检测了3 类不同蜘蛛的系统发生关系.用12 个随机引物对各实验蜘蛛的基因组DNA 进行扩增,选择其中扩增谱带清晰的8 个引物进行分析并计算不同类蜘蛛间的遗传距离.结果表明:所有8 个引物获得的RAPD谱带均表现为不同程度的多态性;地穴型蜘蛛与结网型蜘蛛间的遗传距离及结网型与游猎型之间的遗传距离,均比地穴型与游猎型之间的遗传距离近,体现了蜘蛛由地穴→结网→游猎的系统演化进程.这一结论与根据古生物学、胚胎学、形态学及生态学等得出的结论一致,从而进一步从DNA 分子水平上为蜘蛛系统演化提供了新的证据.  相似文献

In this paper we present a method for the generation of randomly amplified polymorphic DNA (RAPD) markers for sweet potato. These were applied to produce genetic fingerprints of six clonal cultivars and to estimate genetic distances between these cultivars. The level of polymorphism within the species was extremely high. From the 36-decamer random primers used, 170 fragments were amplified, of which 132 (77.6%) were polymorphic. Ten primers resulted in no detected amplification. Of the remaining 26 primers for which amplification was achieved, only one did not reveal polymorphism. Six primers used alone enabled the discrimination of all six genotypes. Pattern analysis, which employed both a classification and ordination method, enabled the grouping of cultivars and the identification of primers which gave greatest discrimination among the cultivars.  相似文献

Genomic-level analyses of DNA from non-invasive sources would facilitate powerful conservation and evolutionary studies in natural populations of endangered and otherwise elusive species. However, the typical low quantity and poor quality of DNA that is extracted from non-invasive samples have generally precluded such work. Here we apply a modified DNA capture protocol that, when used in combination with massively-parallel sequencing technology, facilitates efficient and highly-accurate resequencing of megabases of specified nuclear genomic regions from fecal DNA samples. We validated our approach by comparing genetic variants identified from corresponding fecal and blood DNA samples of six western chimpanzees (Pan troglodytes verus) across more than 1.5 megabases of chromosome 21, chromosome X, and the complete mitochondrial genome. Our results suggest that it is now feasible to conduct genomic studies in natural populations for which constraints on invasive sampling have otherwise long been a barrier. The data we collected also provided an opportunity to examine western chimpanzee genetic diversity at unprecedented scale. Despite high mitochondrial genome diversity (π = 0.585%), western chimpanzees have a low ratio (0.42) of X chromosomal (π = 0.034%) to autosomal (chromosome 21 π = 0.081%) sequence diversity, a pattern that may reflect an unusual demographic history of this subspecies.  相似文献
