The effects on the Schwann cell electrical potential of external ionic concentrations and of K-strophanthoside were investigated. Increasing (K)o depolarized the cell. The potential is related to the logarithm of (K)o in a quasi-linear fashion. The linear portion of the curve has a slope of 45 mv/ten-fold change in (K)o. Diminutions of (Na)o and (Cl)o produced only small variations in the potential. Calcium and magnesium can be replaced by 44 mM calcium without altering the potential. Increase of (Ca)o to 88 mM produced about 10 mv hyperpolarization. The cell was hyperpolarized by 11 mv and 4 mv within 1 min after applying K-strophanthoside at concentrations of 10-3 and 10-5 M, respectively. No variations of cellular potassium, sodium, or chloride were observed 3 min after applying the glycoside. The hyperpolarization caused by 10-3 M K-strophanthoside was not observed when (K)o was diminished to 1 or 0.1 mM or was increased to 30 mM. At a (K)o of 30 mM, 10-2 M strophanthoside was required to produce the hyperpolarizing effect. In high calcium, the cell was further hyperpolarized by the glycoside. The initial hyperpolarization caused by the glycoside was followed by a gradual depolarization and a decrease of the cellular potassium concentration. The results indicate that the Schwann cell potential of about -40 mv is due to ionic diffusion, mainly of potassium, and to a cardiac glycoside-sensitive ion transport process.  相似文献

Measurements have been made of K influx in squid giant axons under internal solute control by dialysis. With [ATP]i = 1 µM, [Na]i = 0, K influx was 6 ± 0.6 pmole/cm2 sec; an increase to [ATP]i = 4 mM gave an influx of 8 ± 0.5 pmole/cm2 sec, while [ATP]i 4, [Na]i 80 gave a K influx of 19 ± 0.7 pmole/cm2 sec (all measurements at ∼16°C). Strophanthidin (10 µM) in seawater quantitatively abolished the ATP-dependent increase in K influx. The concentration dependence of ATP-dependent K influx on [ATP]i, [Na]i, and [K]o was measured; an [ATP]i of 30 µM gave a K influx about half that at physiological concentrations (2–3 mM). About 7 mM [Na]i yielded half the K influx found at 80 mM [Na]i. The ATP-dependent K influx responded linearly to [K]o from 1–20 mM and was independent of whether Na, Li, or choline was the principal cation of seawater. Substances tested as possible energy sources for the K pump were acetyl phosphate, phosphoarginine, PEP, and d-ATP. None was effective except d-ATP and this substance gave 70% of the maximal flux only when phosphoarginine or PEP was also present.  相似文献

The cells of cat right ventricular papillary muscles were depleted of K and caused to accumulate Na and water by preincubation at 2–3°C. The time courses of changes in cellular ion content and volume and of the resting membrane potential (Vm) were then followed after abrupt rewarming to 27–28°C. At physiological external K concentration ([K]o = 5.32 mM) recovery of cellular ion and water contents was complete within 30 minutes, the maximal observable rates of K uptake and Na extrusion (Δmmol cell ion/(kg dry weight) (min.)) being 3.4 and 3.6, respectively. The recovery rate was markedly slowed at [K]o = 1.0 mM. Rewarming caused Vm measured in cells at the muscle surface to recover within from <1 to 9 minutes, but only slight restoration of cellular ion contents (measured in whole muscles) had occurred after 10 minutes. Studies of recovery in NaCl-free sucrose Ringer''s solution made it possible to separate the ouabain-insensitive outward diffusion of Na as a salt from a simultaneous ouabain-sensitive Na extrusion which is associated with a net cellular K uptake. A hypothesis consistent with these observations is that rewarming may activate a ouabain-sensitive "electrogenic" mechanism, most probably the net active transport of Na out of the cell, from which net K uptake may then follow passively.  相似文献

Studies on HeLa cells in spinner culture at pH 7.0 and 37° have shown that [Na]i decreased and [K]i increased with increasing [Ca]o. In Na-free (choline) medium [K]i remained high whether or not Ca was present in the medium. [Na]i and [K]i approached a new steady state within 1 min after transfer to Ca-free medium and returned to the initial values within 15 min upon readdition of Ca. 40% of the cell Ca exchanged within 1 min followed by a slow exchange of the remaining Ca over several hours. [Ca]i increased with decreasing [Na]o but was independent of [K]o. Equimolar Mg did not substitute for Ca in maintaining low [Na]i and high [K]i. Under steady-state conditions about 50% of the cell Na exchanged in accordance with a single rate constant. The initial Na influx was 270, 100, and 2.5 µM/liter of cell water/sec for 0, 0.10, and 1.0 mM [Ca]o, respectively. When Na transport was inhibited with strophanthidin and [Na]i and [K]i allowed to reach a steady state, Na influx was more rapid for cells incubated in Ca-free medium than for cells incubated in medium containing 1.0 mM Ca. These results suggest that Ca competes with Na at the cell membrane and thus controls the passive diffusion of Na into the cell.  相似文献

Using area under the contracture curve to quantitate contractures, the diffusion coefficient of calcium ions within the frog toe muscle during washout in a calcium-free solution and subsequent recovery after reintroduction of calcium to the bathing solution was calculated to be about 2 x 10-6 cm2/sec. The diffusion coefficient measured during washout was found to be independent of temperature or initial calcium ion concentration. During recovery it was found to decrease if the temperature was lowered. This was likely due to the repolarization occurring after the depolarizing effect of the calcium-free solution. The relation between contracture area and [Ca]o was found to be useful over a wider range than that between maximum tension and [Ca]o. The normalized contracture areas were larger at lower calcium concentrations if the contractures were produced with cold potassium solutions or if NO3 replaced Cl in the bathing solutions. Decreasing the potassium concentration of the contracture solution to 50 mM from 115 mM did not change the relation between [Ca]o and the normalized area. If the K concentration of the bathing solution was increased, the areas were decreased at lower concentrations of Ca.  相似文献

Potassium and sodium transport across single distal tubules of Amphiuma   总被引:6,自引:0,他引:6  
The transport properties of potassium (K) and sodium (Na) were studied in single distal tubules of Amphiuma using free-flow micropuncture techniques and stationary microperfusion methods. The transepithelial movement of labeled potassium was measured utilizing a three-compartment system in series in which the time course of tracer disappearance from the lumen was followed. Under control conditions, in blood- and doubly-perfused kidneys, extensive active net reabsorption of sodium and potassium obtains along single distal tubules. Tubular potassium reabsorption is abolished by ouabain at a concentration of 5 x 10-6 M. Significant net secretion of K can be induced by exposing Amphiuma to a high K environment (100 mM KCl) or by adding acetazoleamide (1 x 10-4 M) to the perfusion fluid. Transepithelial movement of potassium involves mixing with only a small fraction of total distal tubular cell potassium. This transport pool of potassium increases significantly with the transition from tubular net reabsorption to net secretion. Indirect evidence is presented which indicates that increased active K uptake across the peritubular cell boundary may be of prime importance during states of net K secretion.  相似文献

The influence of several ions on the membrane potential of the somatic muscle of Ascaris has been investigated by changing their concentration in the surrounding solution. When [K]o is increased at the expense of [Na]o leaving [Cl]o constant, the membrane potential is first seen to increase. [K]o higher than 45 mM reduces the membrane potential with a slope of 23 mv for a tenfold change in [K]o. However, when [K]o is increased keeping [Na]o and [Cl]o low and constant, the line relating the membrane potential with log [K]o has a slope of almost 50 mv. If [Cl]o is reduced in the absence of external Na, after the [K]o is increased to 45 mM, the membrane potential decreases with a slope of 59 mv per tenfold change in [Cl]o in close agreement with the Nernst equation. If Cl- is replaced by SO4 2-, a depolarization is produced, while chloride replacement by NO3 -, Br-, and I- results in a hyperpolarization of the membrane. Removal of the external Na+ ions increases the average membrane potential by 17 mv.  相似文献

Correlation of the localization of La+++ with its effects on Ca++ exchange in cultured rat heart cells is examined with the use of a recently developed technique. 75% of cellular Ca++ is exchangeable and is completely accounted for by two kinetically defined phases. The rapidly exchangeable phase has a t ½ = 1.15 min and accounts for 1 1 mmoles Ca++/kg wet cells or 43% of the exchangeable Ca++ (cells perfused with [Ca++]o = 1 mM) Phase 2 has a t ½ = 19.2 min and accounts for 1.5 mmoles Ca++/kg wet cells or 57% of the exchangeable Ca++. 0.5 mM [La+++]o displaces 0 52 mmoles Ca++/kg wet cells—all from phase 1—and almost completely abolishes subsequent Ca++ influx and efflux The presence of La+++ in the washout converts the washout pattern to a single phase system with a t ½ = 124 min. The effects upon Ca++ exchange are coincident with abolition of contractile tension but regenerative depolarization of the tissue is maintained Electron microscope localization of the La+++ places it exclusively in the external lamina or basement membrane of the cells. Sodium uptake by rainbow trout gills has been investigated with a small-volume system enabling rapid, successive flux measurements in different solutions. Sodium influx obeys a Michaelis-Menten type relation, with a Km of 0.46 mM, and uptake proceeds unimpaired in the absence of penetrating counter-ions. This suggests a coupled cation exchange. Ammonia output is about the same as the Na+ influx when external [Na+] is 1 mM, but at higher or lower Na+ influxes, the correlation does not hold. A progressive downward shift in the pH of the irrigating medium as Na+ influx increases indicates that the exchanging cation is hydrogen. In support of this, acetazolamide, which inhibits Na+ uptake, also prevents the downward pH shift. The potential across the gill is about 10 mv, body fluids positive, in NaCl solutions up to 10 mM, and is little affected by changes in Na+ concentration below that. Finally, evidence for locating the rate-limiting step at the outer membrane of the epithelium is presented.  相似文献

Measurements were made of the sodium outflux rate constant, o k Na, and sodium influx rate constant, i k Na, at varying concentrations of extracellular (Nao) and intracellular (Nac) sodium. o k Na increases with increasing [Nao] in the presence of extracellular potassium (Ko) and in solutions containing ouabain. In K-free solutions which do not contain ouabain, o k Na falls as [Nao] rises from 0 to 6 mM; above 6 mM, o k Na increases with increasing [Nao]. Part of the Na outflux which occurs in solutions free of Na and K disappears when the cells are starved or when the measurements are made in solutions containing ouabain. As [Nao] increases from 0 to 6 mM, i k Na decreases, suggesting that sites involved in the sodium influx are becoming saturated. As [Nac] increases, o k Na at first increases and then decreases; this relation between o k Na and [Nac] is found when the measurements are made in high Na, high K solutions; high Na, K-free solutions; and in (Na + K)-free solutions. The relation may be the consequence of the requirement that more than one Na ion must react with the transport mechanism at the inner surface of the membrane before transport occurs. Further evidence has been obtained that the ouabain-inhibited Na outflux and Na influx in K-free solutions represent an exchange of Nac for Nao via the Na-K pump mechanism.  相似文献

1. The aerobic transport of d-glucose and d-galactose in rabbit kidney tissue at 25° was studied. 2. In slices forming glucose from added substrates an accumulation of glucose against its concentration gradient was found. The apparent ratio of intracellular ([S]i) and extracellular ([S]o) glucose concentrations was increased by 0·4mm-phlorrhizin and 0·3mm-ouabain. 3. Slices and isolated renal tubules actively accumulated glucose from the saline; the apparent [S]i/[S]o fell below 1·0 only at [S]o higher than 0·5mm. 4. The rate of glucose oxidation by slices was characterized by the following parameters: Km 1·16mm; Vmax. 4·5μmoles/g. wet wt./hr. 5. The active accumulation of glucose from the saline was decreased by 0·1mm-2,4-dinitrophenol, 0·4mm-phlorrhizin and by the absence of external Na+. 6. The kinetic parameters of galactose entry into the cells were: Km 1·5mm; Vmax 10μmoles/g. wet wt./hr. 7. The efflux kinetics from slices indicated two intracellular compartments for d-galactose. The galactose efflux was greatly diminished at 0°, was inhibited by 0·4mm-phlorrhizin, but was insensitive to ouabain. 8. The following mechanism of glucose and galactose transport in renal tubular cells is suggested: (a) at the tubular membrane, these sugars are actively transported into the cells by a metabolically- and Na+-dependent phlorrhizin-sensitive mechanism; (b) at the basal cell membrane, these sugars are transported in accordance with their concentration gradient by a phlorrhizin-sensitive Na+-independent facilitated diffusion. The steady-state intracellular sugar concentration is determined by the kinetic parameters of active entry, passive outflow and intracellular utilization.  相似文献

Cation composition of frog smooth muscle cells was investigated. Fresh stomach muscle rings resembled skeletal muscle, but marked Na gain and K loss followed immersion. Mean Na (49.8–79.7 mM/kg tissue) and K (61.8–80.1 mM/kg tissue) varied between batches, but were stable for long periods in vitro. Exchange of 6–30 mM Na/kg tissue with 22Na was extremely slow and distinct. Extracellular water was estimated from sucrose-14C uptake. Calculated exchangeable intracellular Na was 9 mM/kg cell water, and varied little. Thus steady-state transmembrane cation gradients appeared to be steep. K-free solution had only slight effects. Ouabain (10-4 M) caused marked Na gain and reciprocal K loss; at 30°C, Na and K varied linearly with time over a wide range of contents, indicating constant net fluxes. Net fluxes decreased with temperature decrease. 22Na exchange in ouabain-treated tissue at 20–30°C was rapid and difficult to analyze. The best minimum estimates of unidirectional Na fluxes at 30°C were 10–12 times the constant net flux; constant pump efflux may explain these findings. The rapidity of Na exchange may not reflect very high permeability, but it does require a high rate of transport work.  相似文献

Arterially cannulated rabbit interventricular septal tissue was exposed to 5–40 µM La in 2.5 mM Ca perfusate. Immediately following perfusion with La two concurrent events were consistently observed: (a) a rapid decline of active tension to a lesser steady-state value, and (b) an abrupt, release of short duration of tissue-bound Ca. The magnitude of both events was directly related to the [La]o. If the duration of exposure to La was brief, contractility returned toward normal upon return to the La-free perfusate. Elevation of [Ca]o during exposure to La counteracted its effect and induced a concurrent displacement of tissue-bound La. Cellular action potentials recorded during brief perfusion with La demonstrated that essentially normal regenerative depolarization was maintained. Analysis of the quantities of 45Ca released following exposure to 10 µM La indicated that this La-susceptible Ca was being displaced from a homogeneous pool—the one previously shown by Langer to represent contractile dependent Ca. These data led to the following conclusions: During perfusion with 2.5 mM Ca contractile dependent Ca was derived primarily from "superficially" located sites. La effected the release of contractile dependent Ca by modifying the normal permselectivity of this "superficial" membrane for activator Ca. These and other data infer that contractile dependent Ca is derived primarily from superficially located sites.  相似文献

Slices from the cortex corticis of the guinea pig kidney were immersed in a chilled solution without K and then reimmersed in warmer solutions. The Na and K concentrations and the membrane potential Vm were then studied as a function of the Na and K concentrations of the reimmersion fluid. It was found that Na is extruded from the cells against a large electrochemical potential gradient. Q10 for net Na outflux was ∼2.5. At bath K concentrations larger than 8 mM the behavior of K was largely passive. At the outset of reimmersion (Vm > EK) K influx seemed secondary to Na extrusion. Na extrusion would promote K entrance, being limited and requiring the presence of K in the bathing fluid. At bath K concentrations below 8 mM, K influx was up an electrochemical potential gradient. Thus a parallel active K uptake is apparent. Q10 for net K influx was ∼2.0. Dinitrophenol inhibited net Na outflux and net K influx, Q10 became <1.1 for both fluxes. The ratio between these fluxes varied. Thus at the outset of reimmersion the net Na outflux to net K influx ratio was >1. After 8 minutes it was <1.  相似文献

Frog toe muscles were bathed in isotonic, sodium-free Tris chloride, methanesulfonate, or sulfate solutions containing sucrose or mannitol and varying in ionic strength from 0.006 to 0.291. By decreasing the ionic strength the curve relating the peak tension of the K contractures to the log [K] was reversibly shifted to lower [K]. Increasing the [Ca] from 1 to 4 mM almost abolished this effect. The resting uptake of 45Ca was increased more than two times by decreasing the ionic strength from 0.125 to 0.039. It was not increased significantly by raising [Ca] from 1 to 4 mM at low or normal ionic strength. The additional uptake of 45Ca during contractures provoked by 120 mM K was not significantly different at the two levels of ionic strength. The rate of emergence of 45Ca from muscles loaded with 45Ca at reduced ionic strength, was decreased. The effects of low ionic strength are discussed in terms of changes in the potential difference across a membrane with fixed negative charges on the outer surface.  相似文献

L-Alanine and 3-O-methyl-D-glucose accumulation by mucosal strips from rabbit ileum has been investigated with particular emphasis on the interaction between Na and these transport processes. L-Alanine is rapidly accumulated by mucosal tissue and intracellular concentrations of approximately 50 mM are reached within 30 min when extracellular L-alanine concentration is 5 mM. Evidence is presented that intracellular alanine exists in an unbound, osmotically active form and that accumulation is an active transport process. In the absence of extracellular Na, the final ratio of intracellular to extracellular L-alanine does not differ significantly from unity and the rate of net uptake is markedly inhibited. Amino acid accumulation is also inhibited by 5 x 10-5 M ouabain. 3-O-methyl-D-glucose accumulation by this preparation is similarly affected by ouabain and by incubation in a Na-free medium. The effects of amino acid accumulation, of ouabain, and of incubation in a Na-free medium on cell water content and intracellular Na and K concentrations have also been investigated. These results are discussed with reference to the two hypotheses which have been suggested to explain the interaction between Na and intestinal nonelectrolyte transport.  相似文献

Leaves of Vitis californica Benth. (California wild grape) exposed to a photon flux density (PFD) equivalent to full sun exhibited temperature-dependent reductions in the rates or efficiencies of component photosynthetic processes. During high-PFD exposure, net CO2 uptake, photon yield of oxygen evolution, and photosystem II chlorophyll fluorescence at 77 Kelvin (Fm, Fv, and Fv/Fm) were more severely inhibited at high and low temperatures than at intermediate temperatures. Sun leaves tolerated high PFD more than growth chamber-grown leaves but exhibited qualitatively similar temperature-dependent responses to high-PFD exposures. Photosystem II fluorescence and net CO2 uptake exhibited different sensitivities to PFD and temperature. Fluorescence and gas exchange kinetics during exposure to high PFD suggested an interaction of multiple, temperature-dependent processes, involving both regulation of energy distribution and damage to photosynthetic components. Comparison of Fv/Fm to photon yield of oxygen evolution yielded a single, curvilinear relationship, regardless of growth condition or treatment temperature, whereas the relationship between Fm (or Fv) and photon yield varied with growth conditions. This indicated that Fv/Fm was the most reliable fluorescence indicator of PSII photochemical efficiency for leaves of different growth conditions and treatments.  相似文献

Unidirectional Na fluxes in isolated fibers from the frog''s semitendinosus muscle were measured in the presence of strophanthidin and increased external potassium ion concentrations. Strophanthidin at a concentration of 10-5 M inhibited about 80 per cent of the resting Na efflux without having any detectable effect on the resting Na influx. From this it is concluded that the major portion of the resting Na efflux is caused by active transport processes. External potassium concentrations from 2.5 to 7.5 mM had little effect on resting Na efflux. Above 7.5 mM and up to 15 mM external K, the Na efflux was markedly stimulated; with 15 mM K the Na influx was 250 to 300 per cent greater than normal. On the other hand, Na influx was unchanged with 15 mM K. The stimulated Na efflux with the higher concentrations was not appreciably reduced when choline or Li was substituted for external Na, but was completely inhibited by 10-5 M strophanthidin. From these findings it is concluded that the active transport of Na is stimulated by the higher concentrations of K. It is postulated that this effect on the Na "pump" is produced as a result of the depolarization of the muscle membranes and is related to the increased metabolism and heat production found under conditions of high external K.  相似文献

Some factors influencing sodium extrusion by internally dialyzed squid axons   总被引:15,自引:12,他引:3  
Squid giant axons were internally dialyzed by a technique previously described. In an axon exposed to cyanide seawater for 1 hr and dialyzed with an ATP-free medium, the Na efflux had a mean value of 1.3 pmole/cm2sec when [Na]i was 88 mM, in quantitative agreement with flux ratio calculations for a purely passive Na movement. When ATP at a concentration of 5–10 mM was supplied to the axoplasm by dialysis, Na efflux rose almost 30-fold, while if phosphoarginine, 10 mM, was supplied instead of ATP, the Na efflux rose only about 15-fold. The substitution of Li for Na in the seawater outside did not affect the Na efflux from an axon supplied with ATP, while a change to K-free Na seawater reduced the Na efflux to about one-half. When special means were used to free an axon of virtually all ADP, the response of the Na efflux to dialysis with phosphoarginine (PA) at 10 mM was very small (an increment of ca. 3 pmole/cm2sec) and it can be concluded that more than 96% of the Na efflux from an axon is fueled by ATP rather than PA. Measurements of [ATP] in the fluid flowing out of the dialysis tube when the [ATP] supplied was 5 mM made it possible to have a continuous measurement of ATP consumption by the axon. This averaged 43 pmole/cm2sec. The ATP content of axons was also measured and averaged 4.4 mM. Estimates were made of the activities of the following enzymes in axoplasm: ATPase, adenylate kinase, and arginine phosphokinase. Values are scaled to 13°C.  相似文献

The exchange of cell K with K42, J K, has been measured in cat right ventricular papillary muscle under conditions of a steady state with respect to intracellular K concentration. Within the limits of the measurement, all of cell K exchanged at a single rate. Cells from small cats are smaller and have larger surface/volume ratios than cells from large cats. The larger surface/volume ratio results in larger flux values. J K increases in an approximately linear manner as the external K concentration is increased twentyfold, from 2.5 to 50 mM, at constant intracellular K concentration. The permeability for K ions, P K, calculated from the influx and membrane potential, remains very nearly constant over this range of external K concentrations. J K is not affected by replacement of O2 by N2, or by stimulated contractions at 60 per minute, but K influx decreases markedly in 10-5 M and 10-8 M ouabain.  相似文献
