De-differentiation of transformed root cultures of Datura stramonium has previously been shown to cause a loss of tropane alkaloid synthetic capacity. This indicates a marked shift in physiological status, notably in the flux of primary metabolites into tropane alkaloids. Nitrogen metabolism in transformed root cultures of D. stramonium (an alkaloid-producing system) and de-differentiated suspension cultures derived therefrom (a non-producing system) has been compared using Nuclear Magnetic Resonance (NMR) spectroscopy. (15)N-Labelled precursors [((15)NH(4))(2)SO(4) and K(15)NO(3)] were fed and their incorporation into nitrogenous metabolites studied using Heteronuclear Multiple Bond Coherence (HMBC) NMR spectroscopy. In both cultures, the same amino acids were resolved in the HMBC spectra. However, marked differences were found in the intensity of labelling of a range of nitrogenous compounds. In differentiated root cultures, cross-peaks corresponding to secondary metabolites, such as tropine, were observed, whereas these were absent in the de-differentiated cultures. By contrast, N- acetylputrescine and gamma-aminobutyric acid (GABA) accumulated in the de-differentiated cultures to a much larger extent than in the root cultures. It can therefore be suggested that the loss of alkaloid biosynthesis was compensated by the diversion of putrescine metabolism away from the tropane pathway and toward the synthesis of GABA via N-acetylputrescine.  相似文献

Hairy root cultures were obtained from diploid and induced tetraploid plants of Datura stramonium and analyzed by gas chromatography/mass spectrometry. Twenty alkaloids (19 for diploid and 9 for tetraploid hairy root cultures) were identified. A new tropane ester 3-tigloyloxy-6-propionyloxy-7-hydroxytropane was identified on the basis of mass spectral data. Hyoscyamine was the main alkaloid in both diploid and tetraploid cultures. In contrast to diploid hairy roots, the percentage contributions of the alkaloids, with exceptions for hyoscyamine and apoatropine, were higher in the total alkaloid mixture of tetraploid hairy roots.  相似文献

Thirty-six alkaloids were identified in the organs of Datura ceratocaula by GC/MS. Thirty-three of them have not been previously reported for the species. Furthermore, a new tropane ester was tentatively identified as 3-(3'-formyloxytropoyloxy)tropane on basis of its mass spectral fragmentation. Hyoscyamine was the main alkaloid in the plant organs.  相似文献

Isotopic labelling experiments have been carried out in Datura stramonium root cultures with the following isotopically labelled precursors; [2H3]- [2-13C, 2H3]-, [1-13C, 18O2]-acetates, 2H2O, [2H3-methyl]-methionine, [2-13C]-phenyllactate, [3-2H]-tropine and [2'-13C, 3-2H]-littorine. The study explored the incorporation of isotope into the tropane ring system of littorine 1 and hyoscyamine 2 and revealed that deuterium from acetate is incorporated only into C-6 and C-7, and not into C-2 and C-4 as previously reported. Oxygen-18 was not retained at a detectable level into the C(3)-O bond from [1-13C, 18O2]-acetate. The intramolecular nature of the rearrangement of littorine 1 to hyoscyamine 2 is revealed again by a labelling study using [2'-13C, 3-2H]-littorine, [2-13C]-phenyllactate and [3-2H]-tropine.  相似文献

Alkaloid profile of Datura stramonium plant parts collected at different stages of development were investigated by GC–MS. Sixty-four tropane alkaloids were detected and 48 of them were determined. Two new tropane alkaloids, 3-phenylacetoxy-6,7-epoxynortropane and 7-hydroxyapoatropine were tentatively identified. The alkaloids scopoline, 3-(hydroxyacetoxy)tropane, 3-hydroxy-6-(2-methylbutyryloxy)tropane, 3β-tigloyloxy-6-hydroxytropane, 3,7-dihydroxy-6-tigloyloxytropane, 3-tigloyloxy-6-propionyloxytropane, 3-phenylacetoxy-6,7-epoxytropane, 3-phenylacetoxy-6-hydroxytropane, aponorscopolamine, 3β,6β-ditigloyloxytropane and 7-hydroxyhyoscyamine are reported for the first time for this species and 3-acetoxy-6-isobutyryloxytropane for the family Solanaceae. The number and the type of alkaloids in the different plant organs depend on the stages of development. The variability by types of alkaloids (monosubstituted, 3,6- and 3,7-disubstituted, 3,6,7-trisubstituted and 3-substituted-6,7-epoxytropanes) decreases from the roots of the plants in vegetative growth and immature fruits to the seeds of senile plants (mainly monosubstituted and 3-substituted-6,7-epoxytropanes).  相似文献

Fifty three alkaloids were identified in the organs (roots, stems, leaves, flowers, and seeds) of Datura innoxia by GC/MS. Seventeen of them are reported for the first time for this species and one nor-derivative, 3-phenylacetoxynortropane (28), for the genus Datura. Furthermore, four new tropane esters were tentatively identified as 3-acetoxy-6,7-epoxytropane (acetylscopine) (10), 3-acetoxy-6-propionyloxy-7-hydroxytropane (15), 6,7-dehydro-3-phenylacetoxytropane (25), and 3-(2'-phenylpropionyloxy)-6,7-epoxynortropane (dihydroaponorscopolamine) (37) on the basis of their mass spectral data. Hyoscyamine (44) and scopolamine (48) figure as main alkaloids in the roots and aerial parts, respectively.  相似文献

The effects of elicitation, carbon, and nitrogen sources, and precursors on cell growth and tropane alkaloid production in Datura stramonium cell cultures were studied. D. Stramonium cell cultures responded very well to elicitors in the late exponential phase. Addition of cell wall fragments of Phytophotora megasperma (Pmg) enhanced the final tropane alkaloid yield by fivefold compared with control culture. Supply of carbon culture. Supply of carbon and nitrogen sources, at a ratio (C/N) of up to 70, to cell cultures in the early stationary phase, suppressed tropane alkaloid production; whereas C/N rations beyond about 100 increased the final product yield by more than 100% compared to that of the control experiment. Total alkaloid production in the cell culture supplemented with phenylalanine and ornithine was five times higher that that in the control culture. Higher rations of tropine to tropic acid also stimulated alkaloid production. At a ratio of 20, the productivity was seven times higher that that in the control culture. Adding precursors at high concentrations (e.g., 3 to 10 mM) to the cell culture reduced the final cell yield by less than 40%, while elicitation did not affect the cell yield. On the other hand, cell yield in the cultures supplemented with carbon and nitrogen sources was influenced by the C/N ratio. The highest cell yield was obtained at C/N = 70. Datura plants were grown on a clayed support and subjected tosalinity stress (153.8 mol m–3 NaCl) at the 6-leaf stage.Salt treatment increased total alkaloid content in young leaves.The results indicated that at the organ level tropane alkaloidaccumulation was related to plant growth. The kinetics of tropane alkaloids accumulation in different organs such as roots, leaves, stems, flowers and seeds of Datura innoxia was investigated by GC-MS. Twenty-six tropane alkaloids were detected. The ester derivatives of tropine (3alpha-tigloyloxytropine and 3-tigloyloxy-6-hydroxytropine) are the major compounds. Undifferentiated callus were established from the stem explants of Datura innoxia using Murashige and Skoog (MS) medium supplied with 6-benzylaminopurine (BA, 1 mg l(-10) and indole-3-acetic acid (IAA, 0.5 mg l(-1)) in combination for 6 weeks. Callus differentiation was initiated by subculture onto solid MS medium, free from hormones, for more than 10 months. Initially, shoots were formed after four weeks from subculture. Further subculturing in basal MS medium without growth regulators initiated the rooting of a shooty callus after 6 weeks. Investigation of the alkaloid content of the unorganized and organized callus revealed that callus (either green or brown) yielded only trace amounts of alkaloids. On the other hand, re-differentiated shoots contained mainly scopolamine while re-differentiated roots biosynthesized hyoscyamine as the main alkaloid.  相似文献

Tropine (tropan-3 alpha-ol) is an intermediate in the formation of hyoscyamine. An acyltransferase activity that can acetylate tropine using acetylcoenzyme A as cosubstrate has been found in transformed root cultures of Datura stramonium. A further acyltransferase activity that acetylates pseudotropine (tropan-3 beta-ol) with acetyl-coenzyme A is also present. These two activities can be partially resolved by anion-exchange chromatography, some fractions containing only the pseudotropine-utilizing activity. The basic properties of these two enzymes are reported and their roles in forming the observed alkaloid spectrum of D. stramonium roots discussed.  相似文献

Summary Three sexual generations of the somatic hybrids from Datura innoxia+Datura stramonium (Datura straubii) and from Datura innoxia+Datura discolor (Datura hybrida) were investigated (Schieder, 1978). Self fertility of the somatic hybrids proved to be comparable to that of the parental plant species. The good fertility is attributed to the low number of quadrivalents formed in meiosis I. The growth rate of the somatic hybrids is superior to the parental plant species and also to the autotetraploid somatic hybrids of Datura innoxia. Datura innoxia is the species showing the best growth rate of all species used for the hybridization experiments. Summary A stabilized two-year old suspension of a Datura innoxia cell line, producing small amounts of tropane alkaloids (scopolamine and hyoscyamine) was used in this study. Calcium alginate immobilization has been shown to be able to increase secondary metabolite (i. e. alkaloid) production. 用水蒸气蒸馏法从曼陀罗中提取挥发油,并用气相色谱/质谱联用技术(GC-MS)对挥发油的化学成分进行分离鉴定,用气相色谱峰面积归一化法确定各组分的相对含量.结果从曼陀罗挥发油中鉴定出58种化合物,占总挥发油量的92.37%.其中主要成分为5,6-二氢-6-戊基-2H-吡喃-2-酮(44.29%)、二苯胺(12.50%)、四十四烷(10.41%)、二十烷(4.19%)、(E)-3-己烯-1-醇(2.38%)、3,7,11,15-四甲基-2-十六碳烯-1-醇(2.28%)等.  相似文献

This review illustrates the distribution of tropane alkaloids within the families Solanaceae, Erythroxylaceae, Proteaceae, Euphorbiaceae, Rhizophoraceae, Convolvulaceae and Cruciferae. Whereas tropane alkaloids are characteristic of the genera Datura, Brugmansia (tree datura) and Duboisia of the Solanaceae, the distribution is more widespread with novel tropane derivatives in families not traditionally associated with these bases. The chemical nature of more recently discovered water-soluble calystegines and the di- and trimeric forms from the Convolvulaceae (e.g. schizanthines from Schizanthus spp.), truxillines from Bolivian coca leaves and moonines of Erythroxylum moonii are highlighted. Where possible and appropriate, links between the phytochemistry and taxonomy are discussed.  相似文献

The conversion of littorine to hyoscyamine has been investigated by feeding deuterium labelled (RS)-[2-(2)H]-, [3, 3-(2)H(2)]-, [2, 3, 3-(2)H(3)]- phenyllactic acids to transformed root cultures of Datura stramonium. Isolation and GC-MS analyses of the isotope incorporation into the resultant hyoscyamine does not support the involvement of a vicinal interchange process operating during the isomerisation of littorine to hyoscyamine. Additionally a metabolism study with [1'-13C, 3', 3'-(2)H(2)]-hyoscyamine has established that the alkaloid is metabolically stable at C-3' with no evidence for a reversible in vivo oxidation process to the corresponding aldehyde. The data do not support an S-adenosy-L-methionine (SAM 5)/co-enzyme-B(12) mediated process for the isomerisation of littorine to hyoscyamine.  相似文献

Transformed root cultures of Datura stramonium and of related species contain both aliphatic and aromatic tropane esters. It has been shown that these esters are produced by the action of several acyl transferases that transfer the acidic moiety to tropan-3-ol (tropine) or tropan-3-ol (pseudotropine) from various acyl-coenzyme A thioesters. The presence of these enzymes has been examined in a range of tropane-alkaloid-producing and non-tropane-alkaloid-producing species. Activities that esterify tropine appear to be confined to species that accumulate tropane alkaloids, whereas a number of species that do not accumulate tropane alkaloids possess some ability to esterify pseudotropine.The present state of knowledge of these enzymes is reviewed.
