Intraspecific variation in cellular DNA content was measured in five Coregonus autumnalis spawning populations from the Mackenzie River drainage, Canada, using flow cytometry. The rivers assayed were the Peel, Arctic Red, Mountain, Carcajou, and Liard rivers. DNA content was determined from whole blood preparations of fish from all rivers except the Carcajou, for which kidney tissue was used. DNA content measurements of kidney and blood preparations of the same fish from the Mountain River revealed statistically indistinguishable results. Mosaicism was found in blood preparations from the Peel, Arctic Red, Mountain, and Liard rivers, but was not observed in kidney tissue preparations from the Mountain or Carcajou rivers. The Liard River sample had significantly elevated mean DNA content relative to the other four samples; all other samples were statistically indistinguishable. Significant differences in mean DNA content among spawning stocks of a single species reinforces the need for adequate sample sizes of both individuals and populations when reporting "C" values for a particular species.  相似文献

The saline springs of Gypsum Hill in the Canadian high Arctic are a rare example of cold springs originating from deep groundwater and rising to the surface through thick permafrost. The heterotrophic bacteria and autotrophic sulfur-oxidizing bacteria (up to 40% of the total microbial community) isolated from the spring waters and sediments were classified into four phyla (Actinobacteria, Bacteroidetes, Firmicutes, and Proteobacteria) based on 16S rRNA gene analysis; heterotrophic isolates were primarily psychrotolerant, salt-tolerant, facultative anaerobes. Some of the isolates contained genes for thiosulfate oxidation (soxB) and anoxygenic photosynthesis (pufM), possibly enabling the strains to better compete in these sulfur-rich environments subject to long periods of illumination in the Arctic summer. Although leucine uptake by the spring water microbial community was low, CO(2) uptake was relatively high under dark incubation, reinforcing the idea that primary production by chemoautotrophs is an important process in the springs. The small amounts of hydrocarbons in gases exsolving from the springs (0.38 to 0.51% CH(4)) were compositionally and isotopically consistent with microbial methanogenesis and possible methanotrophy. Anaerobic heterotrophic sulfur oxidation and aerobic autotrophic sulfur oxidation activities were demonstrated in sediment slurries. Overall, our results describe an active microbial community capable of sustainability in an extreme environment that experiences prolonged periods of continuous light or darkness, low temperatures, and moderate salinity, where life seems to rely on chemolithoautotrophy.  相似文献

Summary

At high latitudes Saxifraga oppositifolia exists with distinct ecotypes adapted to differences in growing season length. In areas with late snow-lie and cold, wet soils increased metabolic rates and rapid shoot production compensate for ultra-short growing seasons but do not conserve carbohydrate or water for adverse periods. An opposing strategy is evident in ecotypes living in sites with an earlier resumption of growth where soils are warmer and drier and the growing season longer. Here metabolic rates are lower and result in a greater ability to conserve both carbohydrate and water. The existence of opposing strategies for survival in warm and cold habitats, suggests that even in the minimal thermal conditions of the high Arctic a high degree of biodiversity gives the species as a whole a wide ecological amplitude. This degree of diversity not only increases the sites in which the species can survive but confers an ability to adapt to climate change by changing ecotype frequencies to accommodate climatic fluctuations. This facility may have contributed to the survival of populations of this and other species in the high Arctic during the Weichselian glaciation.  相似文献

A lively debate has taken place in anthropology in recent years on field‐work and its representation in ethnographic accounts. At the same time, historians and cultural critics have dissected the ideology and rhetoric of early explorations. Here I examine the writings of the anthropologist and explorer Vilhjalmur Stefans‐son (1879–1962) in the light of current debates about textual representation, drawing upon recent discussions of ethnography, gender, and the power relations of early twentieth‐century explorations. Stefansson went on lengthy expeditions into the Alaskan and Canadian Arctic and wrote extensively on his encounters with indigenous groups, I argue that while Stefansson was a perceptive ethnographer and explorer, he was silent in both his publications and his diaries about important aspects pertaining to his fieldwork, in particular the ethnographic contributions of his Inuit companions and his intimate relations with a native woman named Pannigabluk. This silence, I suggest, contradicts the narrative trope Stefansson generally adopted, summed up in his concept of the friendly Arctic’  相似文献

  • 1.1. Freshwater-resident Arctic charr acclimated for 2 months at 8°C, 15% were divided into four experimental groups in July and exposed to 1 and 8°C in 15 and 34% salinity.
  • 2.2. Only slight changes in gill Na-K-ATPase activity, blood plasma osmolality and blood plasma concentrations of Cl and Mg2+ were found for the fish exposed to 1 or 8°C in brackish water.
  • 3.3. When exposed to sea-water at 8°C, an increase in osmolality and in concentrations of Cl and Mg2+ took place during the first 2–3 days, after which it levelled off.
  • 4.4. If exposed to sea-water at 1°C, however, marked increases were found for all parameters measured and all the fish were dead within 5 days of exposure.
  • 5.5. These results show that freshwater-resident Arctic charr—if acclimated to brackish water—can survive in sea-water during summer if the environmental temperature is not too low.

Arctic relict populations, which persist in disjunct locations far south of a species’ normal range, are at the frontline of climate change and may be especially susceptible to the negative impacts of climate warming. Further, these relict populations may face increasing contact with, or become outcompeted by, invasive species if the invasive taxa are spreading along with the warming climate. Relict populations are simultaneously of particular conservation importance due to their unique genetic make-up and potential for adaptations to warmer temperatures compared to populations at the core of the species range. In this study, we used genotyping-by-sequencing to study the population genetics of Euphrasia hudsoniana, a polyploid arctic disjunct of conservation concern, at the southern edge of its range along the northwestern shore of Lake Superior. In addition, we examined evidence for hybridization with its invasive congener, E. stricta. Overall, we found clear differentiation between the native and invasive species indicated by nearly all analyses. There was limited evidence for gene flow from the invasive into the native species, but patterns were consistent with more extensive gene flow in the opposite direction. Differentiation among native populations was low, yet two of the five populations fell into a separate, distinct group based on STRUCTURE analyses. Continued genetic monitoring of these populations will help elucidate whether hybridization with invasives is a burgeoning threat for this arctic relict.  相似文献

The Arctic is an excellent model system for the study of polyploidy. It is one the Earth's most polyploid‐rich areas, in particular of high‐level and recently evolved polyploids. Here we re‐address previous hypotheses on arctic polyploidy based on a new analysis of the circumarctic flora, and review recent molecular, cytological and reproductive studies. The frequency and level of polyploidy strongly increase northwards within the Arctic. We found no clear‐cut association between polyploidy and the degree of glaciation for the arctic flora as a whole, which contains many widespread species. However, for ‘arctic specialist’ taxa with restricted distributions, the frequency of diploids is much higher in the Beringian area, which remained largely unglaciated during the last ice age, than in the heavily glaciated Atlantic area. This result supports the hypothesis that polyploids are more successful than diploids in colonizing after deglaciation. There is abundant molecular evidence for recurrent formation of arctic polyploids at different scales in time and space. Examples are given of low‐level polyploids formed after the last glaciation and of repeated and successively more high‐level polyploidizations throughout the Quaternary. Recurrent polyploid origins, followed by interbreeding within and across ploidal levels, provide a major explanation for the taxonomic complexity of the arctic flora. In the well‐studied, recently deglaciated archipelago of Svalbard, most species are mainly self‐fertilizing or clonal. All Svalbard polyploids examined so far are genetic allopolyploids with fixed heterozygosity at isozyme loci. The level of heterozygosity in 65 taxa increases dramatically from diploids to high‐level polyploids. In the circumarctic area, there is evidence for numerous recently evolved sibling species within diploid taxonomic species. Rapid evolution of crossing barriers at the diploid level promotes further diversification after expansion from different refugia, and may provide new raw materials for allopolyploid formation. We conclude that the evolutionary success of polyploids in the Arctic may be based on their fixed‐heterozygous genomes, which buffer against inbreeding and genetic drift through periods of dramatic climate change. © 2004 The Linnean Society of London, Biological Journal of the Linnean Society, 2004, 82 , 521–536.  相似文献

The heavy and light subunits of myosin from white and red muscles of Atlantic salmon parr, smolt and adult individuals were analyzed by SDS-PAGE and two-dimensional electrophoresis. Tropomyosin was identified by comigration with rat tropomyosins in two-dimensional gels in the presence and absence of urea. These myofibrillar proteins were compared to those of Arctic charr.
  • 1.1. The myosin heavy chain from Atlantic salmon red muscles was associated with two types of light chain, 1S and 2S, that comigrated with the light chains 1S and 2S of Arctic charr.
  • 2.2. As in the Arctic charr, four myosin light chain spots were detected in white muscles: two fast myosin light chains type 1, one of which comigrated with its analogous in the Arctic charr; one fast myosin light chain type 2, differing slightly in isoelectric point from that of Arctic charr; and one fast myosin light chain type 3 with higher electrophoretic mobility than that of Arctic charr.
  • 3.3. Three tropomyosin spots were detected. White muscles contained only one type of β-tropomyosin and red muscles two types of α-tropomyosin. These three tropomyosin spots comigrated with those of Arctic charr.
  • 4.4. Two myosin heavy chain bands were observed in red muscles of salmon parrs but only one in the rest of the red muscles analyzed.
  • 5.5. Only one myosin heavy chain band was detected in white muscles by SDS-glycerol-polyacrylamide gel electrophoresis. Alfa-chymotryptic peptide mapping of these white myosin heavy chain bands revealed differences attributed to the presence of a new type of myosin heavy chain first detected several months after smoltification.

It was shown that in the years when the numbers of the Arctic foxes are high, even though the lemming numbers are high as well, Brent geese nest considerably closer to owls’ nests than in the years with low Arctic fox numbers. At values of the Arctic fox densities greater than one breeding pair per 20 km2, the factor of lemming numbers ceases to affect the distance between owl and geese nests. This distance becomes dependent on the Arctic fox density (numbers). When the Arctic fox density is greater than the pronounced threshold, the owl-Brent internest distance is inversely and linearly related to the Arctic fox density.  相似文献

The contamination of polar regions due to the global distribution of anthropogenic pollutants is of great concern because it leads to the bioaccumulation of toxic substances, methylmercury among them, in Arctic food chains. Here we present the first evidence that microbes in the high Arctic possess and express diverse merA genes, which specify the reduction of ionic mercury [Hg(II)] to the volatile elemental form [Hg(0)]. The sampled microbial biomass, collected from microbial mats in a coastal lagoon and from the surface of marine macroalgae, was comprised of bacteria that were most closely related to psychrophiles that had previously been described in polar environments. We used a kinetic redox model, taking into consideration photoredox reactions as well as mer-mediated reduction, to assess if the potential for Hg(II) reduction by Arctic microbes can affect the toxicity and environmental mobility of mercury in the high Arctic. Results suggested that mer-mediated Hg(II) reduction could account for most of the Hg(0) that is produced in high Arctic waters. At the surface, with only 5% metabolically active cells, up to 68% of the mercury pool was resolved by the model as biogenic Hg(0). At a greater depth, because of incident light attenuation, the significance of photoredox transformations declined and merA-mediated activity could account for up to 90% of Hg(0) production. These findings highlight the importance of microbial redox transformations in the biogeochemical cycling, and thus the toxicity and mobility, of mercury in polar regions.  相似文献

Early observers of plant evolution in the Arctic have noted a floristic similarity with temperate alpine regions and a predominance of high ploidy levels. The aim of our study was to survey these and other traits in multiple closely related but independently evolved lineages of Artemisia. Our phylogenetic study was based on 133 taxa using 3'-ETS and ITS, and on data on morphology, karyology, distribution, and ecological preferences. We compared Arctic lineages with sister groups and tested whether patterns were significantly different. We found: (1) Artemisia has independently adapted to Arctic habitats 13-18 times; (2) There were no ecological preferences of putative progenitors that might determine the colonization success in the Arctic, although most sister groups were centered in steppe habitats; (3) Plant height was distinctly reduced in Arctic lineages; (4) Arctic lineages contained no more polyploids than their respective sister groups or taxa from other habitats; (5) Enlarged flower heads have evolved repeatedly, probably for better pollinator attraction. This strategy could be a substitute for polyploidy, which is typical in other Arctic taxa. Stronger pollinator attraction should result in better outcrossing and higher heterozygosity in the offspring, which is among the main effects of polyploidy.  相似文献

Microbes in high arctic snow and implications for the cold biosphere   总被引:1,自引:0,他引:1  
We applied molecular, microscopic, and culture techniques to characterize the microbial communities in snow and air at remote sites in the Canadian High Arctic (Ward Hunt Island, Ellesmere Island, and Cornwallis Island, latitudes 74 to 83(o)N). Members of the Bacteria and Eukarya were prevalent in the snow, and their small subunit (SSU) rRNA gene signatures indicated strong local aerial transport within the region over the preceding 8 months of winter snowpack accumulation. Many of the operational taxonomic units (OTUs) were similar to previously reported SSU rRNA gene sequences from the Arctic Ocean, suggesting the importance of local aerial transport processes for marine microbiota. More than 47% of the cyanobacterial OTUs in the snow have been previously found in microbial mats in the region, indicating that this group was also substantially derived from local sources. Viable cyanobacteria isolated from the snow indicated free exchange between the snow and adjacent mat communities. Other sequences were most similar to those found outside the Canadian Arctic but were from snow, lake and sea ice, glaciers and permafrost, alpine regions, Antarctica, and other regions of the Arctic, supporting the concept of global distribution of microbial ecotypes throughout the cold biosphere.  相似文献

A hormonal-challenge protocol was used to compare the stress response of males of Arctic ground squirrels and red squirrels during the breeding season (May). These squirrels live in the same boreal forest of the Yukon, but have very different life histories and utilize the forest in markedly different ways. Red squirrels had levels of total cortisol, maximum corticosteroid-binding capacity, and free cortisol that were 5, 7, and 2 times, respectively, those of Arctic ground squirrels. Red squirrels were resistant to suppression by an artificial glucocorticoid, dexamethasone (DEX); Arctic ground squirrels were not. Cortisol levels in red squirrels responded slowly but continuously to the ACTH injection; Arctic ground squirrels responded rapidly and then stabilized. Testosterone levels in red squirrels were extremely sensitive to the challenge, being suppressed by both DEX and ACTH; levels in Arctic ground squirrels were resistant to the challenge, being modestly suppressed by DEX and stimulated by ACTH. Energy mobilization, as measured by glucose and free fatty acid responses, was not affected. Red squirrels had four times the levels of white blood cells and higher proportions of lymphocytes and lower proportions of eosinophils than Arctic ground squirrels, indicating that the latter were in worse condition immunologically. Our evidence suggests that the functions associated with the hypothalamic-pituitary-adrenal axis are compromised in breeding male Arctic ground squirrels, but not in red squirrels. We propose that in male red squirrels this axis has evolved in the context of a stable social system based on long-lived animals with individual territories which are needed to deal with unpredictable winter food supplies. In contrast, Arctic ground squirrels escape the rigors of winter by hibernation and this hormonal axis has evolved in short-lived males in the context of intense intra-sexual competition in a social system based on female kin groups and regular male dispersal to avoid inbreeding. Sedimentary records from three Canadian High Arctic ponds on Ellesmere Island, spanning the last several thousand years, show major shifts in pond communities within the last ~200 years. These paleolimnological data indicate that aquatic insect (Diptera: Chironomidae) populations rapidly expanded and greatly increased in community diversity beginning in the 19th century. These invertebrate changes coincided with striking shifts in algal (diatom) populations, indicating strong food‐web effects because of climate warming and reduced ice‐cover in ponds. Predicted future warming in the Arctic may produce ecological changes that exceed the large shifts that have already occurred since the 19th century.  相似文献

Root-associated bacteria capable of reducing acetylene to ethylene (biological nitrogen fixation) were isolated from various native plants grown in the Canadian High Arctic. All the strains belonged to the genus Pseudomonas but varied in several physiological characteristics. The rates of acetylene reduction at 14 or 20 degrees C were higher than at 25 or 9 degrees C. Six strains reduced acetylene at 4 degrees C. All the strains exhibited chemotaxis to l-asparagine in semisolid agar at 4 to 25 degrees C. Eleven strains colonized roots of canola (Brassica campestris cv. Tobin) in field soil at population densities of log 4.3 to log 5.1 CFU/g of fresh root at 14 degrees C and log 4.0 to log 5.2 CFU/g of fresh root at 25 degrees C. Some of these nitrogen-fixing pseudomonad strains demonstrated a competitive advantage for root colonization over other rhizosphere bacteria at low temperatures. The combined capabilities of nitrogen fixation and root colonization by diazotrophic pseudomonads may be useful for the development of a biofertilizer inoculant for temperate and cold regions.  相似文献

A continuous flow-through system incubated in situ was used to model oil biodegradation in Arctic coastal waters. High numbers of oil-degrading microorganisms were found in the Arctic coastal waters examined in this study. The microbial community underlying oil slicks increased and showed a population shift to a greater percentage of hydrocarbon-utilizing microorganisms. Microbial populations and oil biodegradation were increased by the addition of nitrogen and phosphorus. Both abiotic and biodegradative losses were lower than expected, perhaps due to the unusually harsh, ice-dominated Arctic summer, during which these tests were conducted. Chromatographic and spectrometric analyses showed that residual oils contained similar percentages of individual components and classes of hydrocarbons, regardless of the amount of degradation, indicating that most components of the oil were being degraded at similar rates.  相似文献
