The amino acid sequence of cytochrome c from niger-seed has been determined by sequence analysis of chymotryptic and tryptic peptides using the dansyl-phenylisothiocyanate method and by qualitative analysis of peptide composition by the dansyl method. Although the spectral ratios indicated the protein was not completely pure, no indication of impurity was found during the sequence analysis and no peptides in addition to those given here were obtained. In certain cases the alignment of peptides was by homology with other cytochromes c. Four residues in the proposed sequence, alanine-1, cysteine-25, histidine-26 and lysine-61 were identified only from peptide compositions. The amino-terminus of the protein is acetylated. The sequence contains two residues of ϵ-N-trimethyllysine.  相似文献

The amino acid sequence of the ferredoxin from Sambucus nigra consists of a single polypeptide chain of 97 amino acid residues, 5 of which are cysteine. The positions of the 4 cysteine residues which bind the iron atoms of the active centre are identical to those of other ferredoxins. Due to difficulties of obtaining pure protein, residues 87–90 have only been identified from the amino acid analysis of peptide C 10 and by homology with other higher plant ferredoxins.  相似文献

Continuous far red light, acting through phytochrome, stimulates the rate of incorporation of density label into amino acids in the cotyledons of Sinapis alba. It is shown that such stimulation leads to increased incorporation of label into proteins. This has important consequences for experiments in which rates of enzyme synthesis in light treated and dark grown plants are compared by labelling methods. The results of some such experiments are re-evaluated.  相似文献

Proposed amino acid sequence of cytochrome c from Enteromorpha intestinalis is presented. The cytochrome is a basic protein, homologous with higher plant and animal cytochromes c of mitochondrial origin. Peptides from chymotryptic and tryptic digests were analysed by the dansyl-phenylisothiocyanate method and aligned by comparison with other homologous cytochromes c. The proposed sequence consists of a single polypeptide chain of 111 residues which is acetylated at its N-terminus. The sequence contains one residue of the unusual amino acid ?-N-trimethyllysine in position 80.  相似文献

The amino acid sequence of plastocyanin from lettuce (Lactuca sativa L.) was determined by using a Beckman 890C automatic sequencer and by dansyl-phenylisothiocyanate analysis of peptides obtained by enzymic digestion of purified CNBr fragments. The protein consists of a single polypeptide chain of 99 residues, and shows close homology with other higher plant plastocyanins. We previously reported two free D-amino acids, D-2-aminopimelic acid (D-APA) and trans-3,4-dehydro-D-2-aminopimelic acid (D-Δ-APA), from Asplenium unilaterale. In the present work we isolated 4-hydroxy-2-aminopimelic acid (OH-APA) from the same plant and determined it to be the α-L-form. We also investigated the configurations of these amino acids isolated from A. prolongatum and A. wilfordii which are morphologically distinct from A. unilaterale. In A. prolongatum, APA was the D- and OH-APA was the L-isomer. In contrast, APA from A. wilfordii was partially racemized and the degree of racemization was significantly different in plant material collected in July and November, L:D = 3:2 and 3:7, respectively. In A. wilfordii OH-APA was almost pure L- and Δ-APA was mostly the D-isomer.  相似文献

A new non-protein amino acid, erythro-γ-hydroxyhomo-L-arginine has been isolated from seed of Lonchocarpus costaricensis by exploiting its property of interacting with borate ions. For structural comparisons, threo-γ-hydroxyhomo-L-arginine was isolated from seed of Lathyrus tingitanus and erythro-γ-hydroxyarginine from Vicia unijuga by novel procedures. The reasons for the interaction of borate with the erythro- but not the threo-forms of these amino acids are discussed.  相似文献

The amino acid sequence of Epiteromorpha cytochrome c has been added to an affinity tree relating the cytochrome c sequences of animals, plants, fungi, protozoans and one bacterium, cytochrome c2 from Rhodospirillum. The Enteromorpha sequence lies on the line of descent of the higher plant sequences; it is not closely related to the cytochrome c of the photosynthetic protozoan, Euglena. 白芷是我国常用中药材的一种,关于白芷镇痛的有效成分,主要认为是异欧前胡素、欧前胡素等香豆素类具有解痉、镇痛等作用的挥发油类化学成分,但白芷镇痛的成分及其机理尚不十分清楚,是否有其他物质也具有一定的药理作用也不得而知,比如说某些药效性氨基酸。该研究采用自动氨基酸分析仪测定了湖南道地茶陵白芷的蛋白质类氨基酸和游离氨基酸的含量,并分析其氨基酸组成。结果表明:总氨基酸检出除Asn以外的其他16种蛋白质氨基酸,含量最高的Arg占总氨基酸的31.21%,接近1/3;必需氨基酸总量达27.01%,其中含量最高的是Leu,占总必需氨基酸的24.14%;药效氨基酸比例较高,在酸水解产物总氨基酸和游离氨基酸中分别达到73.89%和85.78%,其中Arg的含量最高,达1.383g·100g-1,占比为42.24%,游离氨基酸中γ-氨基丁酸、鸟氨酸含量也较高。此外,还含有少量的高半胱氨酸和鹅肌肽等游离非蛋白质氨基酸和短肽;必需氨基酸的组成接近WHO/FAO的建议摄入值,但Met+Cys的RC值最小,为第一限制性氨基酸。这些药效性氨基酸和游离氨基酸可能是茶陵白芷具有良好药效的一个因素,而且茶陵白芷的氨基酸组成和配比较合理、符合人体需要,在强化含硫氨基酸的基础上具有开发成为新型药食同源保健性食品的潜力。研究结果可为进一步研究白芷的药理作用提供依据。  相似文献

The yeast Saccharomyces cerevisiae was grown in the presence of 1.0 mM l-methionine and the half-life of degradation of glutathione determined for the strains Σ1278b (444 min) and the amino-acid-uptake deficient mutant 2512c (368 min). There is no significant difference in these values, yet the rate of uptake of l-methionine is 5–7 times lower in the mutant. In neither strain is the turnover of glutathione sufficient to account for amino acid uptake. We conclude that there is no correlation between the γ-glutamyl cycle and amino acid uptake by this east.  相似文献

Effects of various kinds of dietary protein on growth of the silkworm, Bombyx mori, were determined using semi-synthetic diets. Also, the ingestion, digestion and utilization of dry matter and of nitrogen were measured. Nutritive effects of dietary proteins and supplementation of limiting amino acids on haemolymph protein and amino acids pattern were also investigated. Larval growth was largely dependent on the dietary proteins. When the larvae were reared on a diet containing weakly nutritive proteins such as gluten and zein, haemolymph protein was decreased and uric acid excretion was markedly accelerated. The free amino acid composition of the haemolymph manifested characteristic patterns according to the kinds of dietary protein.The supplementation of gluten and zein with their limiting amino acids resulted in a rise of haemolymph protein and a drop in uric acid excretion. The amino acid patterns in the haemolymph were greatly changed according to supplementation.  相似文献

Thirty two thermophilic amino acid aminotransferases (AATs) were expressed in Escherichia coli as soluble and active proteins. Based on their primary structures, the 32 AATs were divided into four phylogenetic groups (classes I, II, IV, and V). The substrate specificities of these AATs were examined, and 12 AATs were found capable of synthesizing ring-substituted phenylglycine derivatives such as hydroxyl-, methoxy-, and fluorophenylglycines. Eleven out of the 12 AATs were enzymes belonging to two phylogenetic groups namely, one subgroup of the class I family and the class IV family. AATs in these two groups may thus be useful for the synthesis of a variety of ring-substituted phenylglycine derivatives.  相似文献
