The tumor promoter 12-O-tetradecanoyl phorbol-13-acetate is a potent inhibitor of mitochondrial respiration in both normal and methylcholanthrene-transformed C3H 10T12 mouse fibroblasts. This inhibition is seen at concentrations of tumor promoter in the range of 10?8M, occurs within a few minutes after exposure of the intact cells, and is not seen with a biologically inactive analog. The effect appears to be exerted through inhibition of the function of an oligomycin-sensitive ATPase. It is possible, therefore, that alterations in mitochondrial function are associated with the process of tumor promotion.  相似文献

The tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA), which reversibly inhibits the adipose conversion of BALBc 3T3 preadipose cells, increases lactate production by these cells. The stimulation of lactate production requires 4–7 days for optimal effect. Once TPA is removed from the cultures, the rate of lactate production falls to control levels. The concentration dependence for the TPA-mediated stimulation of lactate production is similar to that for its inhibitory effect on adipose conversion. Exogenous lactate in the absence of TPA also inhibits adipose conversion. These results suggest that the ability of TPA to interfere with the normal pattern of glucose metabolism may be important in the inhibitory effect of TPA on triglyceride accumulation in these cells.  相似文献

Application of the tumor-promoting phorbol diester 12-O-tetradecanoylphorbol-13-acetate to mouse epidermis causes a large increase in the activity of ornithine decarboxylase and in polyamine accumulation. Concurrent application of fluocinolone acetonide, an anti-inflammatory steroid that dramatically inhibits tumor promotion, resulted in a dose-dependent decrease in the 12-O-tetradecanoylphorbol-13-acetate-stimulated ornithine decarboxylase activity and the subsequent rise in spermidine levels. Spermine and putrescine levels were not greatly affected by fluocinoline acetonide treatment except that maximal putrescine values occurred later in time. Doses of the glucocorticoid as low as 0.1 μg inhibited the 12-O-tetradecanoylphorbol-13-acetate-induced ornithine decarboxylase activity by as much as 50% and the rise in spermidine accumulation by 30% after coincident treatment of female Sencar mice.  相似文献

The tumor promoter phorbol 12-myristate 13-acetate rapidly induces alterations in both Ca++ content and transport in cultured differentiated chick myoblasts. At 4 ng/ml (6nM), the promoter caused a 25 ± 12% decrease in total intracellular Ca++ within 5 h after its addition. Measurement of 45Ca++ transport at this time revealed a 15 ± 6% decrease in the rate constants for both efflux and influx. Values of t12 for the cytosolic Ca++ pool in control and treated cells were 9.1 and 10.7 min, respectively, for efflux and 8.6 and 10.4 min, respectively, for influx. Ca++ influx was decreased maximally within 90 sec after promoter addition. No effect was observed on 86Rb+ uptake or intracellular concentration at equilibrium. The Ca++ response is among the most rapid yet reported and may play a primary role in altering cellular metabolism.  相似文献

Previous results (Castagna et al. (1979) FEBS Lett. 100, 62–66; Fisher et al. (1979) Biochem. Biophys. Res. Commun. 86, 1063–1068) indicated us that the active tumor promoter TPA (12-O-tetradecanoylphorbol 13-acetate) decreased fluorescence polarisation of diphenylhexatriene in lymphoblastoid and rat embryo cells. In the present study, experiments aimed at examining the molecular interactions of tumor promoters with cell membrane components are performed with fully hydrated multibilayers of 1,2-diacyl-sn-glycero-3-phosphocholine (DPPC) into which increasing amounts of TPA are inserted. The thermotropic behaviour of both the phospholipid bilayers and the interbilayer water was investigated using the differential scanning calorimetry (DSC) and the approach of Ter-Minassian-Saraga et al. ((1982) J. Colloïd Interface Sci. 81, 369–383). The major effects of the tumor promoter are confined to concentrations up to 20% mol fractions of TPA. In this range of concentrations the incorporation of TPA into liposomes decreases the phase-transition temperature but dit not affect ΔHDPPC. Furthermore TPA increases the hydration of the multibilayers. Above 20% mol fractions of TPA, a different thermal behaviour of the system which might suggest morphological rearrangements was observed. The lipid state in TPA-treated liposomes was monitored by fluorescence polarisation using diphenylhexatriene as a lipophilic fluorescent probe and the phase-transition temperature was calculated. The phase transition temperatures determined by both methods were in good agreement. The lowering of this temperature and the decay of fluorescence anisotropy of diphenylhexatriene were parallel. Those effects are consistent with the ‘fluidising’ effect of TPA on DPPC.  相似文献

Several aspects of polyamine biosynthesis were compared in low-passage hamster embryo fibroblasts and transformed hamster fibroblasts. Earlier studies had demonstrated a larger and longer-lasting induction of ornithine decarboxylase activity in transformed cells than in hamster embryo fibroblasts. The increases in intracellular polyamine concentrations after serum stimulation were much greater in chemically transformed HE68BP cells than in normal hamster fibroblasts. Treatment of confluent cultures with the tumor promoter, 12-O-tetradecanoylphorbol-13-acetate, greatly potentiated ornithine decarboxylase induction by fresh medium in HE68BP cells, but not in hamster fibroblasts. A similar synergistic effect was observed when transformed cells, but not normal cells, were treated with the combination of insulin and promoter. HE68BP cells were capable of growth in medium containing serum concentrations as low as 0.5%, whereas only concentrations of 5% or more supported the growth of hamster embryo fibroblasts. Low serum concentrations induced ornithine decarboxylase in HE68BP cells but not in normal cells, and a given serum concentration always produced a greater induction of ornithine decarboxylase in transformed than in normal cells.Another enzyme involved in polyamine synthesis, S-adenosyl-L-methionine decarboxylase was induced in normal and transformed cells by serum-containing medium or tetradecanoylphorbol acetate, but in contrast to ornithine decarboxylase, no synergistic effect was seen in transformed cells exposed to the combination of fresh medium and the tumor promoter. A macromolecular inhibitor of ornithine decarboxylase was readily detected in hamster fibroblast cultures treated with high concentrations of putrescine, but little or none of this inhibitor was found in HE68BP cultures. In both cell types, however, serum induction of ornithine decarboxylase was inhibited under conditions of excess putrescine.The results demonstrate several differences between normal and transformed hamster cells in the regulation of polyamine synthesis.  相似文献

Latent collagenase activity was detected in the media of a well-characterized line of human breast carcinoma cells maintained for over two years in culture. The media also contained sufficient plasminogen activator to convert extrinsically added plasminogen to plasmin which in turn activated the collagenase. During culture of the breast carcinoma in serum-free medium, collagenase activity was maximum on day 12 whereas plasminogen activator activity changed little with time. Using type I collagen as a substrate, the activated breast tumor collagenase produced 34 ? 14 fragments consistent with a mammalian collagenase. These findings suggest a pathologic role of plasminogen activator in the activation of latent collagenase during tumor invasion.A number of investigators have postulated that proteases may play a role in tumor invasion (1–5). Collagenase is one such protease which is active at neutral pH and specifically cleaves triple helical collagen into two (34 ? 14 fragments (6). Secretion of collagenase by tumor cells migrating from the primary mass provides an attractive hypothesis for the mechanism of tumor invasion of surrounding host connective tissue—since the local environment would likely be at neutral pH. Consequently, a number of investigators have reported significant levels of collagenase activity in a wide variety of tumors (7–14). Abramson (13) has correlated aggressive in vivo growth in carcinomas of the head and neck with collagenase activity, and Kuettner et al. (14) have postulated that inhibitors of collagenase may prevent tumors from invading cartilage.Collagenase is produced in both latent and active forms (6). The latent form can be activated with brief protease treatment (15). Since one of the proteases capable of activating collagenase is plasmin (15), the possibility arose that tumor cells could activate collagenase through plasminogen activator. Plasminogen activator secreted by tumor cells (4, 5) could convert plasminogen zymogen to plasmin which would in turn activate latent tumor collagenase. Testing this hypothesis in vitro was the subject of the present study.Previous studies on collagenase from human carcinoma (7, 13, 14) have suffered from the drawback that contaminating inflammatory cells and fibroblasts may have been the source of the collagenase. Therefore, we have studied collagenase production from cultured human breast carcinoma cells which have been well characterized to be mammary epithelial in origin, malignant in karyotype, and able to grow in nude mice. Production of collagenase from these cells is therefore unequivocally of human carcinoma origin. The time course of latent collagenase and plasminogen activator secretion by these cultured tumor cells was studied following withdrawal of serum. To test whether plasminogen activator was secreted in sufficient amounts to indirectly activate latent collagenase, collagenase activity of the culture media was studied after the extrinsic addition of plasminogen. Finally, to verify that the tumor-secreted collagenase cleaved type I collagen at a single locus, enzyme degradation products were studied by gel electrophoresis.  相似文献

A set of virulent mutants of bacteriophage lambda have been selected from λv2 v3. The sites of mutation form two microclusters, both close to v3. Some of the mutants, selected for their ability to grow on a λ-lysogen, can also grow on a λdv carrier strain. They are called “supervirulent” and a mutation conferring super-virulence is called vs. The sites of mutation to vs lie between the presumed promoter mutants (x3, x7) and x13, implying that the operator and promoter interpenetrate each other. The relative affinities of λ repressor for binding, in vitro, to λv+, λv3, λvs326, and λv3 vs327 DNA were 1, 14, 120, and 14000, respectively. We suggest that two separate mutations in the right operator are needed to confer virulence because promoter sites lie within the operator.  相似文献

The invitro DNA dependent synthesis of ribosomal protein L12 and the β subunit of RNA polymerase has been investigated using DNA from a plasmid which contains the genetic information for ribosomal protein L12 and the β subunit of RNA polymerase. This DNA, however, lacks the promoter region and the genetic information for the first 26 amino acids of ribosomal protein L10. It was found that L12 and the β subunit of RNA polymerase are efficiently synthesized invitro from this DNA. These results suggest that L12 and the β subunit of RNA polymerase can be synthesized from a promoter situated within the L10 gene.  相似文献

Isolated porcine Graafian follicles which were explanted in vitro and maintained in organ culture were used as a test-system for the meiosis-inducing action of PMSG and hCG. The addition of either PMSG or hCG alone (10 or 20 IU/ml, respectively) to the culture medium was not effective, whereas the simultaneous administration of these hormones (1515IU/ml) induced resumption of meiosis in 90.3% (3741). The same hormone concentrations were used in a second series of experiments in which the inhibition and induction of meiosis of isolated oocytes were tested by transferring them into host follicles. In host follicles containing up to 12 foreign eggs, which were cultured in control media, meiosis was prevented in 86.0% of all oocytes (104121). By adding PMSG (15 IU/ml) simultaneously with hCG (15 IU/ml) to the medium, meiosis was induced in 95.0% of all oocytes (133140).The assumption is made that the signal initiating resumption of meiosis of the isolated and transferred oocytes is mediated by the follicular fluid, since intimate contact with the membrana granulosa of the host follicle was prevented by using a roller technique.  相似文献

Purified chloroplasts from leaves of Spinacia oleracea L. (spinach) incorporated glycerol 3-phosphate into diacylglycerol, monoacylglycerol, phosphatidylglycerol, phosphatidic acid, and lysophosphatidic acid. The omission of ATP or CTP, CoA or illumination decreased the incorporation markedly. The fraction of incorporated glycerol 3-phosphate found in phosphatidylglycerol was greatly reduced by the omission of bicarbonate, acetate, and ATP, or in darkness, low-osmolarity medium, or high magnesium ion concentration (10 mM). Incorporation of glycerol 3-phosphate into lipid and specifically into phosphatidylglycerol was optimal at a Mg2+CTP ratio of 1, whereas the optimal ratio for Mg2+ATP was closer to 2. The Mg2+CTP gave lower total incorporation but a higher fraction of incorporation in phosphatidylglycerol. Triton X-100 inhibited incorporation of glycerol 3-phosphate into lipid, especially into phosphatidylglycerol.  相似文献

The major form of bovine erythrocyte carbonic anhydrase has been prepared by a new method in which the conventional chloroform-ethanol treatment is replaced by chromatography on DEAE-Sephadex. Single crystals, suitable for high resolution X-ray diffraction studies, have been obtained from enzyme prepared by this method. The space group is P6122. The unit cell contains 12 enzyme molecules and has the dimensions a = b = 68 A?, c = 244 A?.  相似文献

ADP and Pi-loaded membrane vesicles from l-malate-grown Bacillus alcalophilus synthesized ATP upon energization with ascorbateN,N,N′,N′-tetramethyl-p-phenylenediamine. ATP synthesis occurred over a range of external pH from 6.0 to 11.0, under conditions in which the total protonmotive force Δ\?gmH+ was as low as ?30 mV. The phosphate potentials (ΔGp) were calculated to be 11 and 12 kcal/mol at pH 10.5 and 9.0, respectively, whereas the Δ\?gmH+ values in vesicles at these two pH values were quite different (?40 ± 20 mV at pH 10.5 and ?125 ± 20 mV at pH 9.0). ATP synthesis was inhibited by KCN, gramicidin, and by N,N′-dicyclohexylcarbodiimide. Inward translocation of protons, concomitant with ATP synthesis, was demonstrated using direct pH monitoring and fluorescence methods. No dependence upon the presence of Na+ or K+ was found. Thus, ATP synthesis in B. alcalophilus appears to involve a proton-translocating ATPase which functions at low Δ\?gmH+.  相似文献

The sugar composition of the growth medium influenced the NAD+NADH ratio, pyruvate and lactate production, and ATP levels in both normal and transformed fibroblast cell lines growing in tissue culture. Removal of glucose led to a rapid three- to fourfold rise in the NAD+NADH ratio, followed by a slower decline in the content of ATP. However, there was no change in the adenylate energy charge [(ATP + 12ADP)/(ATP + ADP + AMP)] over a 2-h period. The NAD+NADH ratio was restored to the original level within 10 s of glucose readdition. The NAD+NADHratios in cell lines growing on galactose were as high as for those incubated without sugars; growth on mannose or fructose produced intermediate ratios. There was an inverse relationship between the NAD+NADH ratio and pyruvate-lactate production for glucose, fructose and galactose. Thus, all cell lines had a high production of pyruvate and lactate but a low NAD+NADH ratio when grown on glucose. In contrast, when galactose served as the sugar source, acid production was low, while the ratio was high. All cell lines had comparable hexokinase activity, and glucose was the best substrate, mannose intermediate and fructose poorest. Hexokinase activity did not correlate with the relative degree of utilization of the sugars. These results suggest that the sugar composition of the growth medium affects the metabolic pattern of a cell line, including the NAD+NADH ratio, the ATP content and the production of pyruvate and lactate.  相似文献

Two acidic proteins from the 50 S subunit of Bacillus stearothermophilus ribosomes, namely B-L13 (homologous to Escherichia coli protein L7L12) and B-L8, form a complex. Radioactive B-L13, added to ribosomes before dissociation, does not appear in the complex after electrophoresis, so the (B-L13 · B-L8) complex must exist in the ribosome before dissociation. Digestion of B. stearothermophilus ribosomes with polyacrylamide-bound trypsin causes the appearance of new B-L8 and B-L13 spots on two-dimensional polyacrylamide gel electrophoresis, in a pattern which suggests that single molecules of B-L13 are being sequentially cleaved from a four-to-one complex of B-L13 and B-L8.  相似文献

Monoclonal antibodies have been used to study the distribution of several proteins in cleavage and blastoderm stages of Drosophila melanogaster. These antigens are known to be associated with hnRNA-containing particles in tissue culture cells. Protein blotting shows that they are present in the embryo 1 hr after egg deposition. A redistribution from the cytoplasm into the somatic nuclei can be observed during developmental stage 1213, one stage prior to the formation of the cellular blastoderm. Yolk nuclei become stained by these antibodies at about the same time. The shift into pole cell nuclei, however, occurs 112 hr later, during the migration of these cells into the posterior midgut rudiment.  相似文献

In this study, the cells producing T-cell growth factor (TCGF) in the rat MLR were characterised with respect to the antigens defined by W313, W325, and OX8 monoclonal antibodies. Unfractionated lymphocytes and cells depleted of OX8 positive cells were found to be fully capable of producing TCGF, whereas lymphocytes depleted of W313andW325 positive cells had lost this ability. Parallel experiments demonstrated that cells selected by the fluorescence-activated cell sorter for the expression of W313andW325 defined antigens were potent producers of TCGF. Further studies suggested a functional role for the antigen defined by W325 antibody because the addition of this antibody to a MLR abrogates TCGF production. These findings suggest that the important immunomodulatory functions of W325 positive lymphocytes could be exercised via the synthesis of essential lymphokines.  相似文献

The replication defective transducing phage λplac5O29P3 carries a portion of the E.colilac operon in the b2 region of the lambda phage. This lac operon segment contains the lac promoter, the lac operator, and the β-galactosidase z gene, but does not contain the lac repressor i gene. The z gene can be expressed from both the inserted lac promoter and the phage promoter. When E.coli strain 594 (z?, i+) or JC6256 (Δlac) is infected by λplac5O29P3 in the absence of additional cyclic AMP, β-galactosidase synthesis is shown to be expressed from the phage promoter. When 594 (λ+) or JC6256 (λ+) is infected by λplac5O29P3 in the presence of additional cyclic AMP and IPTG, β-galactosidase synthesis is shown to be expressed from the inserted lac promoter.The ability to separate the phage promoter from the inserted lac promoter for β-galactosidase expression will simplify the interpretation whenever λplac5 is used.  相似文献

The relative significance of alterations in precursor supply and enzyme activities for the rate of triacylglycerol synthesis was studied in isolated hepatocytes and perfused livers. Precursor availability was varied in vitro by changing the fatty acid concentration in the incubation medium or adding ethanol to the perfusion medium in order to increase the cellular glycerol 3-phosphate concentration. The rate of glycerolipid synthesis in hepatocytes, measured in terms of the label incorporated into the various lipid classes from tritiated glycerol, was strongly dependent on the fatty acid concentration up to 2 mm of oleate (fatty acid/albumin molar ratio 71). Ethanol in vitro increased the incorporation of labeled oleate into phosphatidic acid and diacylglycerol in the isolated perfused liver, but its effect on the incorporation into triacylglycerol was small. Ethanol in vitro increased the label incorporation into both diacylglycerol and triacylglycerol in the livers from cortisol-treated rats. Although cortisol treatment increased the soluble phosphatidate phosphohydrolase activity 4.4-fold in the hepatocytes, it had no effect on the rate of triacylglycerol synthesis, whereas fasting increased this rate about 3-fold, although only a moderate concomitant increase in soluble phosphatidate phosphohydrolase activity was observed. Neither cortisol treatment nor fasting affected the microsomal glycerol-3-phoshate acyltransferase activity. The results demonstrate that substrate availability can override enzyme modulations in the regulation of triacylglycerol synthesis and that phosphatidate phosphohydrolase is not the main regulator of triacylglycerol synthesis.  相似文献
