Three-day-old germinating embryos of Zea mays were incubatedwith [8-14C]adenine for 8 and 24 h. Use of high performanceliquid and thin layer chromatography provided evidence for theincorporation of adenine by isolated germinating embryos intotrans-zeatin. Kinetin and zeatin(100 mmol m–3)reversald the ABA-mediated(100mmol m-2)closure of stomata of young maize leaves but did notaffect stomatal apertures of these leaves when applied alone.As leaves aged, kinetin or zeatin alone promoted increased stomatalapertures, while abscisic acid (ABA) applied alone had a reducedeffect on stomata. Root to Shoot Communication in Maize Plants of the Effects of Soil Drying   总被引:44,自引:2,他引:44  
Seedlings of Zea mays L. (John Innes hybrid) were grown withroots divided between two containers such that part of the rootsystem could reduce the water potential of the soil in its immediatevicinity while the rest of the root system was well suppliedwith water. When compared to plants rooted in two pots of moistsoil, drying of part of the root system resulted in partialclosure of stomata, even though leaf water potential, turgorand abscisic acid (ABA) content remained unaffected. When leafpieces were removed from the two groups of plants and incubatedunder conditions favourable for stomatal opening, stomata ofthe ‘half-watered’ plants still showed restrictedapertures. Incubation in kinetin (10 mmol m–3) or zeatin(100 mmol m–3) reversed the closure of stomata stimulatedby soil drying. These results suggest that a continuous supplyof cytokinin from roots may be necessary to sustain maximalstomatal opening and an interruption of this supply due to soildrying may act as an indicator of inhibited root activity, resultingin restricted stomatal opening and thereby restricted wateruse. The endogenous cytokinins present in dryZea mays seed were determined using both radioimmunoassay and gas chromatography—mass spectrometry. Similar values for bases and ribosides were obtained by the two methods. The cytokinins present in embryo and endosperm were estimated separately using radioimmunoassay; similar levels of cytokinins were found in these two tissues. The major cytokinins detected on a whole-seed basis were dihydrozeatin riboside, O-glucosyldihydrozeatin riboside, zeatin 9-glucoside, zeatin, and the nucleotides of zeatin, dihydrozeatin, and isopentenyladenine. Cytokinin levels in the mature dry seed were considerably lower than cytokinin levels published in the literature for immature seed. Unexpected activity in the radioimmunoassays was detected in the wash from the DEAE cellulose column chromatography step. The compound(s) responsible for this activity did not have the solvent partitioning characteristics of a cytokinin base or riboside. They eluted as a single fraction following high-performance liquid chromatography on a Zorbax C8 column; this fraction showed no activity in theAmaranthus bioassay for cytokinins, but inhibited the activity of authentic zeatin riboside present at an optimal concentration.  相似文献

Intact Zea mays L. kernels attached to cob tissue develop tomaturity when grown in vitro. This experiment was designed todetermine if it is possible to prolong kernel growth by refreshingthe culture medium. Blocks of maize kernels were grown in vitroon media containing several concentrations of sucrose. Kernels,at all concentrations of sucrose, developed to maturity at 30–35d post-pollination, indicating that it is not possible to extendthe kernel growth phase by supplying a carbohydrate source.Kernels grown on media containing 80 g 1–1 or higher sucroseconcentration had a significantly greater percentage of kernelsthat developed to maturity, and had greater weight and starchcontent per seed. Embryos obtained from developing kernels of maize (Zea maysL.) were incubated in a bathing medium, to measure the effectof the osmotic environment on the balance between uptake andrelease of assimilates by the embryo. This report describes the isolation and identification of endogenouscytokinins from Citrus ovaries. Cytokinin active fractions wereobtained by extraction with 80% (v/v) ethanol, followed by purificationwith hexane, n-butanol and a polyvinylpyrrolidone and SephadexLH-20 column chromatography. Five fractions with cytokinin activity were found in the organicphase, using the tobacco callus assay. The main active compoundsin these fractions were separated by HPLC, bioassayed and identifiedby GC—MS as ribosyl zeatin, zeatin and isopentenyl adenosine.Hydrolysis of the first fraction with B-glucosidase gave cytokininactive compounds that in paper chromatography had RF's similarto those of zeatin and ribosyl zeatin. Treating the aqueousphase with alkaline phosphatase produced a cytokinin activecompound that in paper chromatography had the same RF as isopentenyladenosine indicating that their ribotide was probably the majorphosphorylated cytokinin present in Citrus ovaries. Cytokinin contents in cotyledon, hypocotyl and root of etiolatedsquash (Cucurbita maxima Duch.) seedlings were determined byinstrumental analysis using 14C-benzyladenine (14C-BA) as aninternal standard. Crude extracts were purified using insolublepolyvinylpyrrolidone, cellulose-phosphate column and SEP-PAKC18 cartridge, then applied to a Sephadex LH-20 column to separatezeatin riboside (ZR), isopentenyl adenosine, isopentenyl adenine,14C-BA and a mixture of zeatin (Z) and dihydrozeatin (DHZ).The recovery rate for the cytokinin fractions after LH-20 wascorrected by 14C-BA. Each cytokinin fraction was further purifiedby HPLC which also separated Z and DHZ in the LH-20 fraction.Before permethylation, 14C-BA was added to each of the cytokininfractions to correct the methylation rates. Each methylatedcytokinin fraction was again purified by HPLC, then subjectedto gas chromatography with a capillary column and flame thermionicdetector. The detection limit of cytokinins by this system was0.1 ng. cis-ZK was the most abundant cytokinin in all tissues of theetiolated squash seedlings. Active cytokinins such as trans-ZRand trans-Z were mostly found in cotyledons with lesser amountsin the roots. DHZ was most abundant in the cotyledon. To provide insight into the regulatory roles of auxin and cytokininin endosperm development, defective kernel (dek) mutants ofmaize (Zea mays L.) were examined. The mutants dek6, dek18 anddek 26 had substantially lower indole-3-acetic acid (IAA) levelsthan wild-type counterparts while dek8, dek12 dek24 had higherIAA levels than wild-type counterparts. The mutant dek6 hada somewhat lower zeatin level than its wild-type counterpart.The mutation-induced changes in IAA levels and effects of exogenously-appliedauxin were not consistently related to changes in numbers ofnuclei per endorsperm; however, there was a correspondence betweenauxin level in endosperm and nuclear diameter, which is proportionalto the extent of post-mitotic DNA synthesis (endoreduplication).We conclude that auxin may be involved in stimulating endoreduplication.Copyright1993, 1999 Academic Press Zea mays L., maize, defective kernel mutant, dek, auxin, cytokinin, hormone, ELISA, cell number, cell division, endopolyploidy, endoreduplication, DNA amplification  相似文献

The nature of the cytokinins present in mature, expanded leavesof alder (Alnus glutinosa (L.) Gaertn.) was investigated usinga chromatographic system capable of partially resolving zeatinfrom (?)-dihydrozeatin. A zeatin-like cytokinin, present asboth a ‘free’ form and as a polar conjugate, postulatedto the the O-ß-D-glucoside of zeatin, accounted forthe bulk of the cytokinin activity (detected by the soybeancallus assay). Studies on the fate of [8-14C]zeatin supplied to detached alderleaves indicated this cytokinin to undergo extensive metabolism.An appreciable proportion of the radioactivity was incorporatedinto 80% methanol-insoluble and soluble acidic/neutral fractions,while adenosine- and adenine-like peaks were prominent metabolitesin the basic n-butanol-soluble fraction. Small amounts of glucosides,with the properties of both zeatin-O-ß-D-glucosideand dihydrozeatin-O-ß-D-glucoside were formed, thelatter becoming the most prominent with time. The ability of alder leaves to form glucoside directly from(?)-dihydrozeatin was assessed using the soybean callus assay.(?)-Dihydrozeatin was subject to a relatively rapid, continuousand substantial conversion to its putative O-ß-D-glucosideand cytokinin activity was consequently conserved while, incontrast, leaves supplied with zeatin exhibited a progressiveloss of cytokinin activity and, in agreement with the radioactivityexperiments, produced only a small amount of activity attributableto the putative O-glucosides. The significance of the observed cytokinin metabolism is discussedin relation to the endogenous cytokinin status of the leaf.  相似文献

As part of a search for biochemical indicators of seed withgood survival characteristics during storage, polyamine (PA)levels were measured in embryos for Zea mays seeds. PA levelswere determined in individual embryos of Mean Generation Analysis(MGA) of crosses between lines which show good (C2) and poor(C6) storage characteristics. A correlation was found betweenhigher PA contents (putrescine and spermidine) and superiorsurvival characteristics within the populations that were analysedby MGA. Titres of putrescine (PUT) and spermidine (SPD) showedthat three-quarters of the F2 resembled the C2 parent and aquarter resembled the C6 parent. Furthermore, the backcrossC6F1 showed that half of the embryos had PUT levels above theparental C6 line. A maternal inheritance effect was not observed.Neither spermine nor cadaverine content was correlated withthe survival inheritance pattern. Given the survival kineticsobtained, it is suggested that both the PA titres and the seedsurvival characteristics are determined by nuclear dominantfactors. Young leaf segments of Zea mays L. seedlings were cultured onMurashige and Skoog's basal nutrient medium supplemented with2 mg l–1 2, 4-D and sub-cultured on medium containing8 mg l–1 2,4-D. Two types of callus tissues appeared—embryogenicand non-embryogenic. The embryogenic callus tissue producednumerous somatic embryos which on transfer to media containinglow amounts of 2,4-D or ABA produced plantlets. Uptake of the dinitroaniline herbicide oryzalin (3,5-dinitro-N4,N4-dipropylsulfamlamide) and its effect on root growth werestudied using 5 mm corn (Zea mays L.) and pea (Pisum sativum)root apices. Pea root growth was much less susceptible to oryzalinthan corn root growth. Uptake studies showed that pea root apicesalso accumulated much less [14C]oryzalin and had a lower bindingaffinity for this herbicide. [14C]oryzalin was not metabolizedin root apices from either species. Thus, the differential susceptibilityto oryzalin in the case of corn versus pea can be explained,at least in part, by differences in oryzalin uptake and accumulationby roots. The time course of xylem differentiation was determined in explantsof lettuce pith parenchyma (Lactuca sativa L. cv. Romana) culturedon Murashige and Skoog (1962) medium using different concentrationsof auxin (IAA) and one cytokinin (zeatin or kinetin). Increasinglevels of auxin from I mg 1–1 to 15 mg 1–1 in thepresence of a constant level of a cytokinin (zeatin or kinetin)yielded up to 10 mg 1–1 IAA, an increase in the numberof tracheary element formations. Cytokinin concentrations aboveand below o.1 mg 1–1 interacting with an optimal xylogenicamount of auxin inhibited xylogenesis. The IAA (10 mg 1–1)-zeatin(0.1 mg 1–1) treatment produced the greatest number oftracheids, while kinetin compared to zeatin did not producesuch an effect. The different effectiveness of zeatin and kinetinin inducing tracheary element formations was not due to a differentcapacity of the two cytokinins to stimulate cell division butit seems likely that zeatin, because of interaction with IAA,is more active than kinetin in the determination of the dividingcells in a specific type of cytodifferentiation. The IAA (10mg 1–1)-zeatin (0.1 mg 1–1) treatment produced about6.9 per cent tracheids with respect to cell division while IAA(10 mg 1–1)-kinetin (0.1 mg 1–1) produced 4.2 percent. These results are discussed with reference to the problemsof hormonal control of xylem differentiation.  相似文献

Endogenous cytokinin and gibberellin-like activity were measuredby bioassay in developing fruit of the orchid Epidendrum ibaguense.Cytokinins decline during the first 30 d after pollination,then begin to accumulate, with very high levels (1–13µ g zeatin eq. g–1 dry wt. ) in the mature fruitand seed. The major structural change in developing fruit duringthe first 30 d is the ongoing cell division in the fruit wall.By day 30 most ovules have been fertilized and embryo developmentbegins. The increase in cytokinin activity thus coincides withthe onset of embryo development. Gibberellin levels declinein the fruit throughout development, although high activity(0.9 µ g GA3 eq. g–1 dry wt. ) is observed in themature seed. The mature embryo shows no obvious structural differentiationinto embryonic axis and cotyledon and no endosperm develops.  相似文献

In order to assess the potential role of endogenous cytokininsin the germination of Zea mays L. caryopses, cytokinin activitywas determined in mature kernels and in kernels 1, 2, and 3d after imbibition. Cytokinin activity was also recorded inthe endosperm and embryo tissue of mature kernels and of kernels3 d after imbibition to establish the localization and changesin the levels of endogenous cytokinins during imbibition andearly germination. Using chromatographic, chemical, and enzymic techniques, compoundswith chromatographic properties resembling those of zeatin,zeatin riboside, zeatin glucoside, zeatin riboside glucoside,and their respective dihydro derivatives were detected in extractsfrom mature maize kernels. From analyses of the endosperm andembryo tissue at the two stages, it appears that the cytokininglucosides present in the endosperm are transported to the embryonicaxis for utilization in growth and development of the seedling.This concept is supported by the fact that the levels of ß-glucosidaseactivity detected were highest in the embryos, particularlyin the radicles, 3 d after imbibition.  相似文献

Epidermal strips and leaf fragments of Commelina and leaf fragmentsof maize were incubated on solutions containing naturally-occurringor synthetic cytokinins and/or ABA. The effects of these treatmentson stomatal behaviour were assessed. Cytokinins alone did notpromote stomatal opening in either species but concentrationsof both zeatin and kinetin from 10–3 to 10–1 molm–3 caused some reversal of ABA-stimulated closure ofmaize stomata. The reversal of the ABA effect increased withincreasing cytokinin concentration. Cytokinins had no effecton ABA-stimulated closure of Commelina stomata. When appliedalone, at high concentration (10–1 mol m–3), toCommelina epidermis or leaf pieces both zeatin and kinetin restrictedstomatal opening. Previous results indicate that cytokinin action requires formationof Ca-calmodulin. We evaluated that hypothesis for cytokinin(zeatin) induced growth and chlorophyll formation in excisedcucumber (Cucumis sativus L.) cotyledons using several compoundsexpected to increase or decrease cytosolic levels of Ca2+ andthereby perturb formation of Ca-calmodulin. We also studiedeffects of eight drugs known to inhibit action of Ca-calmodulinin animals. Most of the results indicate no special requirementof Ca-calmodulin for cytokinin action in these organs.
