Alginate concentrations between 2 and 4% had little effect on the degradation rate of phenol by alginate-immobilized Pseudomonas putida. Ten-degree shifts from 25°C resulted in approximately 30% slower degradation. Maximal degradation rates were favored at pH 5.5–6.0. The response of degradation rate to increased air flow in the bubble column used was almost linear and an optimal higher than 16 vol vol−1 was indicated, although free cells appeared in the reaction medium above 12 vol vol−1. When the initial phenol concentration was raised, degradation rate was not significantly affected until levels higher than 1200 mg ml−1 where performance was markedly reduced. Increasing the ratio of total bead volume to medium volume gave progressively smaller increases in degradation rate. At a medium volume to total bead volume ratio of 5:1, the maximum degradation rate was 250 mg L−1 h−1. Received 24 November 1998/ Accepted in revised form 27 January 1999  相似文献

AIMS: To study the effect of co-contaminants (phenol) on the biodegradation of pyridine by freely suspended and calcium alginate immobilized bacteria. METHODS AND RESULTS: Varying concentrations of phenol were added to free and calcium alginate immobilized Pseudomonas putida MK1 (KCTC 12283) to examine the effect of this pollutant on pyridine degradation. When the concentration of phenol reached 0.38 g l(-1), pyridine degradation by freely suspended bacteria was inhibited. The increased inhibition with the higher phenol levels was apparent in increased lag times. Pyridine degradation was essentially completely inhibited at 0.5 g l(-1) phenol. However, immobilized cells showed tolerance against 0.5 g l(-1) phenol and pyridine degradation by immobilized cell could be achieved. CONCLUSIONS: This works shows that calcium alginate immobilization of microbial cells can effectively increase the tolerance of P. putida MK1 to phenol and results in increased degradation of pyridine. SIGNIFICANCE AND IMPACT OF THE STUDY: Treatment of wastewater stream can be negatively affected by the presence of co-pollutants. This work demonstrates the potential of calcium alginate immobilization of microbes to protect cells against compound toxicity resulting in an increase in pollutant degradation.  相似文献

A mathematical model is proposed to analyze the mass transfer limitations in phenol biodegradation using Pseudomonas putida immobilized in calcium alginate. The model takes into account internal and external mass transfer limitations, substrate inhibition kinetics and the dependence of the effective diffusivity of phenol in alginate gel on cell concentration. The model is validated with the experimental data from batch fermentation. The effect of various operating conditions such as initial phenol concentration, initial cell loading, alginate gel loading on the biodegradation of phenol is experimentally demonstrated. Phenol degradation time is found to decrease initially and reach stationary value with increase in cell loading as well as gel loading. The model predicts these trends reasonably well and shows the presence of external mass transfer limitations. A new concept of effectiveness factor is introduced to analyze the overall performance of batch fermentation.  相似文献

Granular activated carbon (GAC) was incorporated into hollow fiber membrane bioreactors for the biodegradation of 1,000 mg phenol l−1 through immobilization of Pseudomonas putida. The phenol was removed within 25 h in the hybrid bioreactor, comparing with 31 h for a GAC-free bioreactor. AIMS: The aim of this study is to evaluate the utility of aerobically grown microbial granules for the biological treatment of phenol-containing wastewater. METHODS AND RESULTS: A column-type sequential aerobic sludge blanket reactor was inoculated with activated sludge and fed with phenol as the sole carbon source, at a rate of 1.5 g phenol l-1 d-1. Aerobically grown microbial granules first appeared on day 9 of reactor operation and quickly grew to displace the seed flocs as the dominant form of biomass in the reactor. These granules were compact and regular in appearance, and consisted of bacterial rods and cocci and fungi embedded in an extracellular polymeric matrix. The granules had a mean size of 0.52 mm, a sludge volume index of 40 ml g-1 and a specific oxygen utilization rate of 110 mg oxygen g VSS-1 h-1 (VSS stands for volatile suspended solids). Specific phenol degradation rates increased with phenol concentration from 0 to 500 mg phenol l-1, peaked at 1.4 g phenol g VSS-1 d-1, and declined with further increases in phenol concentration as substrate inhibition effects became important. CONCLUSIONS: Aerobically grown microbial granules were successfully cultivated in a reactor maintained at a loading rate of 1.5 g phenol l-1 d-1. The granules exhibited a high tolerance towards phenol. Significant rates of phenol degradation were attained at phenol concentrations as high as 2 g l-1. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study to demonstrate the ability of aerobically grown microbial granules to degrade phenol. These granules appear to represent an excellent immobilization strategy for microorganisms to biologically remove phenol and other toxic chemicals in high-strength industrial wastewaters.  相似文献

Summary The semicontinuous and continuous degradation of phenol by Pseudomonas putida P8 which was immobilized on activated carbon was investigated. The amount of bacteria immobilized on the activated carbon surface dependend on the cell concentration in the suspension and on the type of activated carbon. In a continuous process running for four weeks the biomass, which accumulated in the activated carbon fixed bed, was removed periodically. The average phenol degradation rate in this process was 360 mg/1 h. The degradation activity of the bacteria for phenol, measured by the activity of the catechol-2,3-dioxygenase, was stimulated by the activated carbon. During the fermentation processes the carbon particles were covered with a biofilm. The bacteria grew, especially in the caverns and the entrances of the macropores, whereby the phenol adsorption by the activated carbon was decreased.  相似文献

Density functional theory (DFT) calculations performed at the PBE/DZP level using the DFT-D2 method were utilized to investigate the adsorption of phenol on pristine activated carbon (AC) and on activated carbon functionalized with OH, CHO, or COOH groups. Over the pristine AC, the phenol molecule undergoes weak physical adsorption due to van der Waals interactions between the aromatic part of the phenol and the basal planes of the AC. Among the three functional groups used to functionalize the AC, the carboxylic group was found to interact most strongly with the hydroxyl group of phenol. These results suggest that functionalized AC-COOH has great potential for use in environmental applications as an adsorbent of phenol molecules in aqueous phases.  相似文献

The purpose of this paper is to clarify the effect of the two different GAC types (steam activated or chemically activated) on DOC biodegradation in biological activated carbon (BAC) columns. For this purpose, raw water taken from a surface reservoir was fed to continuous-flow lab-scale biofiltration columns which were run for more than 18,000 bed volumes. The effect of pre-ozonation on DOC removal was also evaluated. Experimental results showed that biological activity inside the BAC columns extended the service life and the choice of filter material was crucial in BAC systems. The DOC biodegradation was higher in thermally activated carbon columns compared to the chemically activated one. The ability of GAC to better adsorb and retain organic compounds increased the chance of biodegradation. Contrary to expectations, pre-ozonation did not significantly enhance DOC biodegradation. Despite the high increase in biodegradable dissolved organic carbon (BDOC) upon ozonation, overall DOC biodegradation efficiencies did not differ from raw water. Overall, the DOC biodegradation in columns was higher than in most of the studies. This observation was primarily attributed to the low specific ultraviolet absorption (SUVA) values in raw water indicating a high biodegradability.  相似文献

This article reports studies on a continuous pulsed plate bioreactor (PPBR) with the cells of Pseudomonas desmolyticum (NCIM2112) immobilized on granular activated carbon (GAC) used as a biofilm reactor for biodegradation of phenol. Almost complete removal of 200 ppm phenol could be achieved in this bioreactor. Biofilm structure and characteristics are influenced by hydrodynamic and shear conditions in bioreactors. In this article, the effect of shear stress induced by frequency of pulsation on biofilm characteristics during the startup period in the PPBR is reported. The startup time decreased with the increase in frequency of pulsation. The formation of biofilm in PPBR was found to have three phases: accumulation, compaction, and plateau. The effect of frequency on production of exoploymeric substances (EPS) such as, protein, carbohydrate, and humic substance is reported. An increase in shear stress induced by the frequency of pulsation increased the production of exopolymeric substances in the biofilm during startup of the bioreactor. Increase in shear stress caused a decrease in biofilm thickness and an increase in dry density of the biofilm. Increase in shear stress resulted in a smoother and thinner biofilm surface with more compact and dense structure.  相似文献

Summary A defined mixed culture of the yeast Cryptococcus elinovii H1 and the bacterium Pseudomonas putida P8 was immobilized by adsorption on activated carbon and sintered glass, respectively. Depending on its adsorption capacity for phenol the activated carbon system could completely degrade 17 g/l in batch culture, whereas the sintered glass system was able to degrade phenol up to 4 g/l. During semicontinuous degradation of phenol (1 g/l) both systems reached constant degradation times with the fourth batch that lasted 8 h when using the activated carbon system and 10 h in the sintered glass system. In the course of continuous degradation of phenol the activated carbon system reached a maximum degradation rate of 9.2 g l–1 day–1 compared to 6.4 g l–1 day–1degraded by the sintered glass system. 2-Hydroxymuconic acid semialdehyde could be identified and quantitatively determined as a metabolite of phenol degradation by P. putida P8. Summary Addition of powdered activated carbon to a continuous-flow activated-sludge culture in a gas-lift bioreactor growing on o-cresol in a mineral-salts medium resulted in a reduction of the effluent o-cresol concentration during steady-state conditions as well as during shock-load conditions. During the latter conditions the activated carbon acted as a buffer. Bioregeneration of loaded activated carbon did occur, resulting in enhanced biodegradation of o-cresol. Adsorption of o-cresol by activated carbon was not sufficient to explain the total difference in effluent o-cresol concentration between the system with activated carbon and the system without activated carbon. Summary Phenol degradation by a defined mixed culture of Pseudomonas putida P8 and Cryptococcus elinovii H 1, which were immobilized by adsorption on activated carbon, was studied.The immobilized mixed culture was able to degrade phenol up to 17 g/l and degraded it faster than the pure cultures, depending on a complementary metabolism of the two microorganisms.Storage experiments revealed an excellent longterm storage capability of the biocatalyst: activated carbon with adsorbed cells of Pseudomonas putida P8 and Cryptococcus elinovii H1 could be stored up to 12 months without decrease on degradation capacity.Scanning electron micrographs showed that Pseudomonas putida P8 had grown through the pore system of the activated carbon into the inside of the carbon particles.  相似文献

农业土壤和黑碳(BC)两种不同的吸附剂吸附苯酚平衡后分离,每组一部分不做处理,另一部分通过加入无酚灭菌溶液脱附平衡后分离,制备得到在不同吸附位点上吸附有苯酚的两类不同类型的4种吸附苯酚的吸附剂,研究了在不同Pseudomonasputida ATCC 11172菌密度条件下吸附在这4种吸附剂上的苯酚的脱附行为.结果表明,土壤及BC对苯酚的吸附均呈现明显的非线性,可用Freundlich模型描述.吸附态的苯酚能否被微生物利用取决于微生物及吸附剂的性质,BC具有发达的微孔结构,微孔小于假单胞菌细胞尺寸,导致假单胞菌无法直接利用吸附在BC上的苯酚;土壤基本无微孔结构,微生物较易与吸附的苯酚发生表面接触,直接利用吸附态苯酚.BC和土壤上的吸附态苯酚的脱附行为能用三元位点模型很好地描述,模型计算结果表明BC上的苯酚脱附主要受慢速脱附和极慢速脱附控制,微生物降解速率受脱附控制,降解可加速BC上的慢速脱附和极慢速脱附;土壤上的苯酚脱附主要受快速脱附控制,微生物降解不受脱附速率限制,对土壤上的脱附行为基本无影响.  相似文献

Respirometry is a precious tool for determining the activity of microbial populations. The measurement of oxygen uptake rate is commonly used but cannot be applied in anoxic or anaerobic conditions or for insoluble substrate. Carbon dioxide production can be measured accurately by gas balance techniques, especially with an on-line infrared analyzer. Unfortunately, in dynamic systems, and hence in the case of short-term batch experiments, chemical and physical transfer limitations for carbon dioxide can be sufficient to make the observed carbon dioxide evolution rate (OCER) deduced from direct gas analysis very different from the biological carbon dioxide evolution rate (CER).To take these transfer phenomena into account and calculate the real CER, a mathematical model based on mass balance equations is proposed. In this work, the chemical equilibrium involving carbon dioxide and the measured pH evolution of the liquid medium are considered. The mass transfer from the liquid to the gas phase is described, and the response time of the analysis system is evaluated.Global mass transfer coefficients (K(L)a) for carbon dioxide and oxygen are determined and compared to one another, improving the choice of hydrodynamic hypotheses. The equations presented are found to give good predictions of the disturbance of gaseous responses during pH changes.Finally, the mathematical model developed associated with a laboratory-scale reactor, is used successfully to determine the CER in nonstationary conditions, during batch experiments performed with microorganisms coming from an activated sludge system. Phenol bioconversion by Pseudomonas stutzeri OX1 using either free or immobilized cells was investigated with the aim of searching for optimal operating conditions of a continuous bioconversion process. The study was developed by analyzing: (a) free-cell growth and products of phenol bioconversion by batch cultures of P. stutzeri; (b) growth of P. stutzeri cells immobilized on carrier particles; (c) bioconversion of phenol-bearing liquid streams and the establishment and growth of an active bacterial biofilm during continuous operation of an internal-loop airlift bioreactor. We have confirmed that free Pseudomonas cultures are able to transform phenol through the classical meta pathway for the degradation of aromatic molecules. Data indicate that bacterial growth is substrate-inhibited, with a limiting phenol concentration of about 600 mg/L. Immobilization tests revealed that a stable bacterial biofilm can be formed on various types of solid carriers (silica sand, tuff, and activated carbon), but not on alumina. Entrapment in alginate beads also proved to be effective for P. stutzeri immobilization. Continuous bioconversion of phenol-bearing liquid streams was successfully obtained in a biofilm reactor operated in the internal-circulation airlift mode. Phenol conversion exceeded 95%. Biofilm formation and growth during continuous operation of the airlift bioreactor were quantitatively and qualitatively assessed.  相似文献

The effect of the presence of an alternate toxiccompound (phenol) on the p-nitrophenol(PNP)-degrading activity of freely suspended andcalcium alginate immobilized Nocardioides sp.NSP41 was investigated. In the single substrateexperiments, when the concentration of phenol and PNPwas increased to 1400 mg l-1 and 400 mg l-1,respectively, the initial cell concentrations in thefreely suspended cell culture should be higher than1.5 g dry cell weight l-1 for completedegradation. In the simultaneous degradationexperiment, when the initial concentration of phenolwas increased from 100 to 400 mg l-1, thespecific PNP degradation rate at the concentration of200 mg l-1 was decreased from 0.028 to 0.021h-1. A freely suspended cell culture with a highinitial cell concentration resulted in a highvolumetric degradation rate, suggesting the potentialuse of immobilized cells for simultaneous degradation.In the immobilized cell cultures, althoughsimultaneous degradation of PNP and phenol wasmaintained, the specific PNP and phenol degradationrate decreased. However, a high volumetric PNP andphenol degradation rate could be achieved byimmobilization because of the high cell concentration.Furthermore, when the immobilized cells were reused inthe simultaneous degradation of PNP and phenol, theydid not lose their PNP- and phenol-degrading activityfor 12 times in semi-continuous cultures. Takentogether, the use of immobilized Nocardioidessp. NSP41 for the simultaneous degradation of PNP andphenol at high concentrations is quite feasiblebecause of the high volumetric PNP and phenoldegradation rate and the reusability of immobilizedcells.  相似文献

A bacterial strain isolated from activated sludge and identified as Bacillus amyloliquefaciens could biodegrade phenol, but 2,4,6-trichlorophenol (TCP) inhibited phenol biodegradation and biomass growth. UV photolysis converted TCP into dichlorocatechol, monochlorophenol, and dichlorophenol, and this relieved inhibition by TCP. Phenol-removal and biomass-growth rates were significantly accelerated after UV photolysis: the monod maximum specific growth rate (μ max) increased by 9 % after TCP photolysis, and the half-maximum-rate concentration (K S) decreased by 36 %. Thus, the major benefit of UV photolysis in this case was to transform TCP into a set of much-less-inhibitory products.  相似文献

Tert-butyl alcohol (TBA) is a metabolite of methyl tert-butyl ether and is itself possibly a fuel oxygenate. The goals of this study were to enrich and characterize TBA-degrading micro-organism(s) from a granular activated carbon (GAC) unit currently treating TBA. The results reported herein describe the first aerobic, TBA-degrading cultures derived from GAC. Strains KR1 and YZ1 were enriched from a GAC sample in a bicarbonate-buffered freshwater medium. TBA was degraded to 10% of the initial concentration (2–5 mM) within 5 days after initial inoculation and was continuously degraded within 1 day of each re-amendment. Resting cell suspensions mineralized 70 and 60% of the TBA within 24 h for KR1 and YZ1, respectively. Performance optimization with resting cells was conducted to investigate kinetics and the extent of TBA degradation as influenced by oxygen, pH and temperature. The most favorable temperature was 37°C; however, TBA was degraded from 4 to 60°C, indicating that the culture will sufficiently treat groundwater without heating. This is also the first report of psychrotolerant or thermotolerant TBA biodegradation. The pH range for TBA degradation ran from 5.0 to 9.0. Phylogenetic data using a partial 16S rRNA gene sequence (570 bases) suggest that the primary members of KR1 and YZ1 include uncharacterized organisms within the genera Hydrogenophaga, Caulobacter, and Pannonibacter.  相似文献

In polluted soil or ground water, inorganic nutrients such as nitrogen may be limiting, so that Monod kinetics for carbon limitation may not describe microbial growth and contaminant biodegradation rates. To test this hypothesis we measured14CO2 evolved by a pure culture ofAcinetobacter johnsonii degrading 120 µg14C-phenol per ml in saturated sand with molar carbon:nitrogen (CN) ratios ranging from 1.5 to 560. We fit kinetics models to the data using non-linear least squares regression. Phenol disappearance and population growth were also measured at CN1.5 and CN560.After a 5- to 10-hour lag period, most of the14CO2 evolution curves at all CN ratios displayed a sigmoidal shape, suggesting that the microbial populations grew. As CN ratio increased, the initial rate of14CO2 evolution decreased. Cell growth and phenol consumption occurred at both CN1.5 and CN560, and showed the same trends as the14CO2 data. A kinetics model assuming population growth limited by a single substrate best fit the14CO2 evolution data for CN1.5. At intermediate to high CN ratios, the data were best fit by a model originally formulated to describe no-growth metabolism of one substrate coupled with microbial growth on a second substrate. We suggest that this dual-substrate model describes linear growth on phenol while nitrogen is available and first-order metabolism of phenol without growth after nitrogen is depleted.  相似文献

The objectives of this study are to obtain the time courses of the amount of chlorophenol adsorbed onto granular activated carbon (GAC) in the simultaneous adsorption and biodegradation processes involving 4-chlorophenol (4-CP) and 2,4-dichlorophenol (2,4-DCP), respectively, and to quantify the bioregeneration efficiency of GAC loaded with 4-CP and 2,4-DCP by direct measurement of the amount of chlorophenol adsorbed onto GAC. Under abiotic and biotic conditions, the time courses of the amount of chlorophenol adsorbed onto GAC at various GAC dosages for the initial 4-CP and 2,4-DCP concentrations below and above the biomass acclimated concentrations of 300 and 150 mg/L, respectively, were determined. The results show that the highest bioregeneration efficiency was achieved provided that the initial adsorbate concentration was lower than the acclimated concentration. When the initial adsorbate concentration was higher than the acclimated concentration, the highest bioregeneration efficiency was achieved if excess adsorbent was used.  相似文献
