【目的】小菜蛾是危害十字花科蔬菜的世界性重要害虫。通过分析抗生素对小菜蛾的毒性效应,了解肠道细菌对小菜蛾适合度的影响,有助于更好地阐明小菜蛾肠道细菌的功能。【方法】利用抗生素处理含有高丰度肠道细菌的萝卜苗饲养品系(FZss)小菜蛾幼虫,同时利用抗生素处理饲料饲养的无肠道细菌(SLss)小菜蛾幼虫,分析抗生素及肠道细菌对小菜蛾适合度的影响。【结果】抗生素处理FZss品系小菜蛾导致了小菜蛾发育历期延长,虫重、蛹重、化蛹率、产卵量和成虫寿命降低。利用抗生素处理无肠道菌的SLss品系小菜蛾幼虫,小菜蛾化蛹率和单雌产卵量均显著降低,而对发育历期、虫重和蛹重则无影响。【结论】综合两个研究的结果发现抗生素处理后宿主适合度的降低一方面是由于抗生素的毒性效应导致,另一方面是由于小菜蛾肠道细菌的缺失引起。抗生素的毒性效应主要表现为对化蛹率和单雌产卵量的影响,而肠道细菌则对小菜蛾的发育历期、虫重、蛹重以及成虫寿命具有重要的促进作用。  相似文献

用Bt制剂和Bt毒素Cry1Ac分别对源自深圳田间的小菜蛾Plutella xylostella在室内进行抗性种群选育,获得相对抗性倍数分别为24.36、38.16倍的抗性种群DBM.1Ac-R30和DBM.Bt-R46。对这2个抗性种群及其敏感种群(DBM.Bt-S)的生长发育、存活及繁殖特征进行了详细地观察与比较,并以甘蓝为饲喂材料构建了2个抗性实验种群的生命表。结果表明,DBM.1Ac-R30和DBM.Bt-R46种群较DBM.Bt-S种群的产卵量和孵化率下降,幼虫发育历期延长,雌雄比显著降低,雌成虫数量、寿命减少。DBM.1Ac-R30和DBM.Bt-R46种群相对于DBM.Bt-S种群的相对适合度分别为0.75和0.65,抗性种群在繁殖能力上存在明显的生存劣势。  相似文献

本文报道不含有活菌的苏云金杆菌可湿性粉剂在田间应用的药效试验。结果表明:经过辐射处理杀灭活孢子后的苏云金杆菌对稻纵卷叶螟和小菜蛾有良好的防治效果,与未经过处理的Bt可湿性粉剂的效果相比不存在明显的差异。由于该种制剂不含有活菌,对生态环境无不良影响,在田间应用将更加安全。  相似文献

【目的】本研究旨在探讨葡萄糖氧化酶(glucose oxidase, GOX)在黏虫Mythimna separata发育、消化和免疫防御中的作用。【方法】采用转录组测序技术克隆了黏虫GOX基因的cDNA序列。qRT-PCR检测该GOX基因在黏虫不同发育阶段(1-6龄幼虫、蛹和1日龄成虫)和4龄第1天幼虫不同组织(前肠、中肠、后肠、下唇腺、马氏管、脂肪体和表皮)、以及取食用不同浓度(0.01%, 0.1%, 1%和10%)葡萄糖溶液浸泡10 s的玉米叶片及饥饿和再取食条件下的4龄第1天幼虫中的特异性表达模式。通过RNAi和生物测定研究该GOX基因在黏虫对苏云金芽孢杆菌Bacillus thuringiensis抗性中的功能。【结果】克隆获得了一条新的长2 187 bp的黏虫GOX基因MsGOX cDNA序列(GenBank登录号: KY348779),其开放阅读框长1 821 bp,编码606个氨基酸,预测分子量为66.4 kD。发育表达谱结果表明MsGOX在黏虫不同发育阶段表达量不同,在4龄幼虫期表达量最高;组织表达谱结果表明MsGOX在黏虫4龄第1天幼虫各组织中均有表达,在下唇腺中表达量最高。饲喂幼虫不同浓度葡萄糖对MsGOX转录的诱导效果不同,在葡萄糖浓度为10%时,该基因的表达水平达到最高。随着饥饿时间的延长,4龄第1天幼虫中MsGOX的表达水平逐渐升高,并在饥饿24 h时达到高峰,饥饿后再用玉米叶片饲喂幼虫,MsGOX的表达水平逐渐回升。注射dsMsGOX 48 h后,MsGOX的表达水平较对照(注射dsEGFP)降低了88.3%; 与同一时间点的对照相比,注射dsMsGOX后48和72 h幼虫 的体重、体长和消化系数均显著降低,苏云金芽孢杆菌侵染48和72 h提高了幼虫的校正死亡率。【结论】MsGOX可能参与了黏虫中肠的消化和抗菌,研究结果为进一步研究黏虫MsGOX的功能并提出新的黏虫防治策略提供了依据。  相似文献

摘要:【目的】通过比较Cry1Ac蛋白抗性及敏感棉铃虫中肠细菌群落的结构组成,研究中肠微生物是否与棉铃虫Bt抗性产生有关。【方法】首先提取了棉铃虫中肠微生物基因组DNA,通过PCR扩增获得了16S rDNA全长片段及V3区。采用基于16S rDNA 的免培养技术—16S rDNA文库建立和变性梯度凝胶电泳(DGGE)研究了国内特有的Bt抗性和敏感品系棉铃虫中肠细菌群落组成,并对其进行分析和比较。【结果】16S rDNA文库测序结果表明,抗性品系与敏感品系棉铃虫中肠细菌群落特别是优势菌群非常相似,但在部分劣势菌群上存在差异。抗性品系中主要优势菌有:不可培养微生物(Uncultured bacterium)占56.4%,鹑鸡肠球菌(Enterococcus gallinarum)占17.0%,铅黄肠球菌(Enterococcus casseliflavus)占17.0%;敏感品系中主要优势菌为不可培养微生物(Uncultured bacterium)60.2%,鹑鸡肠球菌(Enterococcus gallinarum)占19.3%,铅黄肠球菌(Enterococcus casseliflavus)占14.7%。随后进行的PCR验证表明,部分有差异的劣势菌在两种品系虫体都存在。DGGE图谱分析表明,这两个品系棉铃虫中肠菌群相似性达到92.3%。【结论】敏感品系与抗性品系棉铃虫肠道菌群组成极其相似,推测抗性的产生与肠道微生物无直接关系。  相似文献

【目的】构建带有苏云金芽孢杆菌cry3a基因非芽孢依赖启动子和绿色荧光蛋白基因gfp(Green Fluorescent Protein)的原核表达载体,并转化从桑粒肩天牛幼虫肠道分离的两株常驻细菌短短芽孢杆菌CQUBb和苏云金芽孢杆菌CQUBt,以检测cry3a启动子在昆虫肠道常驻菌中的启动子活性,获得GFP标记菌株,为常驻菌在昆虫幼虫肠道中的定殖情况和杀虫工程菌的构建奠定基础。【方法】采用重叠延伸PCR将cry3a基因启动子和gfp基因进行融合,并与pHT304载体连接构建重组质粒pHT3AG,获得的重组质粒以电脉冲转化肠道常驻菌短短芽孢杆菌CQUBb和苏云金芽孢杆菌CQUBt,于可见光和荧光显微镜下观察荧光并通过SDS-PAGE分析重组菌株的蛋白表达情况,然后对重组菌株进行生长动力学分析和稳定性测试。【结果】重组菌在营养期大量组成型表达GFP,经电泳分离在凝胶上出现约29kDa的特异蛋白条带;重组菌生长曲线与出发菌没有显著差异,说明外源质粒未对宿主菌的生长带来明显不利影响;抗性条件下传代30次后两菌株外源质粒稳定性仍可达95%、67%;两个菌株比较,CQUBb比CQUBt质粒转化率高、重组菌GFP表达时间长、表达量大,并且重组菌株稳定性好。【结论】成功地将cry3a基因核心启动子和gfp基因转入桑粒肩天牛幼虫肠道常驻菌,实现了该启动子在Bt之外的菌株中发挥作用,构建了两个GFP标记菌株;重组基因工程菌株表达量大,稳定性好,可以用作昆虫肠道内微生态研究和芽孢杆菌表达系统以及杀虫菌株的构建。  相似文献

Potential trap crops for the diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), were evaluated through a series of ovipositional preference and larval survival experiments in outdoor screenhouses in 2002 and 2003. Hosts examined as trap crops were glossy and waxy collards, Brassica oleracea L. variety acephala; Indian mustard, Brassica juncea (L.) Czern; and yellow rocket, Barbarea vulgaris (R. Br.) variety arcuata. More eggs were laid on the potential trap crops, with the exception of waxy collards, than on cabbage. When P. xylostella was offered multiple hosts at the same time, numbers of eggs laid on glossy collards, Indian mustard, and yellow rocket were 3, 18, and 12 times greater than on cabbage, respectively. Similarly, when P. xylostella was offered a single trap crop host and cabbage, numbers of eggs laid on glossy collards, Indian mustard, and yellow rocket were 300, 19, and 110 times greater than on cabbage, respectively. Our studies suggest differences in oviposition between the potential trap crops and cabbage were likely due to host volatiles, leaf morphology and color, or a combination of these factors, rather than to total leaf areas, leaf shape, or plant architecture. Two-choice tests with a Y-tube olfactometer indicated that plant volatiles were major factors in P. xylostella host preference. The percentage larval survival from egg to pupation was 22.2% on cabbage, 18.9% on waxy collards, and 24.4% on Indian mustard, whereas survival was significantly lower on glossy collards (6.7%) and yellow rocket (0%). Based on our tests, it seems that yellow rocket may be the best candidate for use as a trap crop for P. xylostella because it is highly attractive for oviposition, but larvae do not survive on it.  相似文献

【目的】昆虫肠道微生物对于其食物消化、生长发育以及环境适应性等方面都具有重要作用,本研究旨在探究小菜蛾 Plutella xylostella (L.)幼虫肠道可培养细菌的菌群结构及抗生素敏感性。【方法】对小菜蛾3龄幼虫粪便进行了细菌分离培养和16S rDNA分子鉴定,并采用纸片扩散法对之进行了药敏试验。【结果】小菜蛾3龄幼虫肠道有5属6种细菌,即蒙氏肠球菌 Enterococcus mundtii、欧文氏菌属 Erwinia 2种、成团泛菌 Pantoea agglomerans 、类芽孢杆菌Paenibacillus sp.和栖稻假单胞菌 Pseudomonas oryzihabitans,均与小菜蛾中肠细菌已知种不同,其中蒙氏肠球菌数量最多;蒙氏肠球菌、桃色欧文氏菌和成团泛菌对青霉素、氨苄青霉素、麦迪霉素、克拉霉素和洁霉素均不敏感,而对磷霉素、万古霉素和强力霉素等表现出相似的敏感性。【结论】小菜蛾幼虫肠道细菌菌群组成具有多样性,且存在天然耐药性现象。本研究为进一步开展小菜蛾幼虫肠道细菌微生物区系及功能细菌的研究提供了菌株材料和研究基础。  相似文献

羧酸酯酶介导的小菜蛾对氟虫腈的抗性   总被引:3,自引:0,他引:3  
【目的】羧酸酯酶(carboxylesterases, CarEs)是昆虫重要的解毒代谢酶之一,可以介导靶标昆虫对多种杀虫剂的代谢抗性。本研究检测了羧酸酯酶对小菜蛾Plutella xylostella 抗药性的介导功能,旨在阐明羧酸酯酶在小菜蛾代谢解毒中的生理生化和分子机理。【方法】采用点滴法测定氟虫腈对小菜蛾敏感种群和抗氟虫腈种群的毒力,以及羧酸酯酶抑制剂磷酸三苯酯(triphenyl phosphate, TPP)对氟虫腈的增效作用;以LC30和LC50浓度的氟虫腈处理抗性小菜蛾,测定药剂处理后CarEs酶活性的变化;利用qRT-PCR技术分析Pxae22和Pxae31两个基因在小菜蛾不同发育阶段、组织和种群的表达模式;利用dsRNA干扰Pxae22和Pxae31后观察基因的表达变化和小菜蛾3龄幼虫对药剂敏感性的变化。【结果】TPP可以削弱小菜蛾3龄幼虫对氟虫腈的抗性,增效倍数约为6倍;使用较低剂量(LC30和LC50)氟虫腈处理小菜蛾3龄幼虫后,处理组CarEs比活力明显高于对照,提示氟虫腈对小菜蛾CarEs活性具有诱导作用。对羧酸酯酶基因Pxae22和Pxae31在小菜蛾不同发育阶段、4龄幼虫不同组织和不同种群3龄幼虫中的表达模式分析发现,这两个基因在小菜蛾4龄幼虫中的表达量最高;在4龄幼虫中以中肠组织中的表达量较高,头、表皮、脂肪体中的表达量很低; 抗性种群中的表达量显著高于敏感种群。通过干扰 Pxae22和 Pxae31后的qRT-PCR验证,两个基因的表达量均显著降低,进一步的氟虫腈毒力测定发现,干扰P xae22和 Pxae31后的小菜蛾3龄幼虫对氟虫腈的敏感性分别增加了1.63倍和1.73倍。【结论】羧酸酯酶在小菜蛾对氟虫腈解毒代谢中具有重要作用;Pxae22和Pxae31是小菜蛾的两个抗性相关基因,其表达水平的变化直接影响小菜蛾对氟虫腈的敏感性。  相似文献

Abstract Field trials were conducted in China in 2008 and 2009 to evaluate the efficacy of mating disruption (MD) on diamondback moth, Plutella xylostella, in cabbage, Brassica oleracea var. capitata. Effectiveness was positively correlated with the MD dispenser density in the field. A density of 167 MD dispensers per ha produced an average population decrease of about 50% compared to the conventional‐practice field. Significant fewer males were captured in pheromone‐treated and conventional‐practice fields than in the blank control field, but the difference was not significant between the pheromone‐treated and conventional‐practice fields. In addition, fewer eggs and larvae were observed in pheromone‐treated fields. Our results suggest mating disruption coupled with minimal insecticidal supplements is a promising solution for resistance management and control of diamondback moth infestation.  相似文献

A total of 1700 Japanese strains of Bacillus thuringiensis, belonging to at least 47 H serogroups, were examined for insecticidal activity against larvae of the diamondback moth, Plutella xylostella. The high-level toxicity was associated with 612 isolates (36.0%). Of these, 608 isolates (99.3%) fell into 13 H serogroups belonging to the low-numbered H serotypes, H1-H10. Conversely, most isolates belonging to the high-numbered serotypes (>H10) had little or no larvicidal activity; only one isolate of the serovar japonensis H23 was active. P xylostella larvae were susceptible to 89.8% of the serovar morrisoni H8a:8b strains and 85.7% of galleriae H5a:5b strains. High values of 60-80% were also obtained in six serovars (thuringiensis H1, alesti H3a:3c, kurstaki H3a:3b:3c, kenyae H4a:4c, aizawai H7, and tolworhi H9), while relatively low values of <60% in two other common serovars, sotto H4a:4b and darmstadiensis H10a:10b. Five selected isolates, belonging to H serovars other than kurstaki and aizawai, were 10-60 times less toxic than the reference strain HD-1 (serovar kurstaki). Parasporal inclusion proteins of these strains were immunologically unrelated to those of the strain HD-1 and the aizawai type strain.  相似文献

RNA干扰(RNA interference, RNAi)是一种调控基因表达的方法, 其通过体外合成一段与内源靶基因同源的双链RNA(dsRNA)或siRNA, 导入生物体内, 使内源靶基因中同源mRNA降解, 从而达到阻抑基因表达的目的。类钙粘蛋白(cadherin-like protein)是位于昆虫中肠刷状缘膜囊(brush border membrane vesicles, BBMV)上与钙粘蛋白(cadherin)结构相似的物质, 是多种昆虫体内Bt杀虫蛋白的受体。本研究利用基因特异引物通过RT-PCR扩增了小菜蛾类钙粘蛋白基因的2个片段(CAD1和CAD2), 合成相对应的双链RNA(double-stranded RNA, dsRNA); 并将dsRNA通过显微注射导入小菜蛾3龄幼虫体内, 测定了不同靶位点、不同剂量、不同检测时间对目的基因mRNA表达量的影响。结果表明: 将70 nL CAD1对应的dsRNA注射到幼虫体内48 h后, 基因表达量显著下降, 72 h后恢复。免疫印迹检测结果表明, 类钙粘蛋白在注射dsRNA 48 h后幼虫BBMV中的含量明显下降。本实验成功实现了小菜蛾类钙粘蛋白基因的沉默, 该体系的成功建立为利用RNAi技术分析小菜蛾及其他鳞翅目昆虫基因的功能提供了参考。  相似文献

取食不同食物对小菜蛾幼虫肠道细菌多样性的影响   总被引:2,自引:0,他引:2  
【目的】植食性昆虫肠道细菌的组成与其食物密切相关。本研究旨在探究小菜蛾Plutella xylostella幼虫肠道细菌多样性与其取食食物之间的关系以及它们之间相互适应的过程。【方法】本研究选取小菜蛾人工饲料品系(S)及其转寄主到结球甘蓝Brassica oleracea var. capitata、结球白菜Brassica rapa subsp. pekinensis和花椰菜Brassica olerocea var. botrytis饲养后第1代(分别为G1C, G1CC和G1WC)和第3代(分别为G3C, G3CC和G3WC)的4龄幼虫,提取小菜蛾肠道细菌基因组DNA,利用Illumina MiSeq二代高通量测序技术,分析其肠道细菌多样性和丰度。【结果】α多样性指数分析发现,取食不同食物的小菜蛾4龄幼虫肠道细菌多样性高低顺序为G1WC>G1CC>S>G1C。在菌群组成上,以人工饲料为食的S样品肠道细菌主要由厚壁菌门(Firmicutes)组成,转寄主植物后的G1C, G1CC和G1WC肠道中厚壁菌门(Firmicutes)相对丰度显著下降,G1C和G1CC小菜蛾肠道中变形菌门(Proteobacteria)相对丰度显著上升成为优势菌群,G1WC肠道中拟杆菌门(Bacteroidetes)成为优势菌群。在寄主植物上连续饲养3代后,与第1代相比,小菜蛾肠道细菌α多样性指数没有显著性改变,但在结球甘蓝和结球白菜上小菜蛾肠道菌群结构却发生了变化,相比G1C,G3C肠道中芽孢杆菌目(Bacillales)的相对丰度显著下降;相比G1CC, G3CC肠道中放线菌门(Proteobacteria)、芽单胞菌门(Gemmatimonadetes)和硝化螺旋菌门(Nitrospirae)的相对丰度均显著上升。【结论】取食人工饲料和不同寄主植物的小菜蛾幼虫肠道细菌多样性和群落构成存在显著差异,寄主植物对小菜蛾肠道微生物的结构组成具有重要的影响,且小菜蛾肠道微生物对寄主植物可能存在一个长期适应的过程。本研究为进一步探讨影响小菜蛾肠道细菌变化的因素,以及后续研究肠道细菌与寄主植物之间的互作奠定了良好的基础。  相似文献

AlthoughPlutella xylostella (L.) is a worldwide pest of cruciferous crops, relatively little is known about its oviposition behaviour. This study was undertaken to provide necessary information about mechanisms involved inP. xylostella host selection. Four oviposition behaviours were described. Moths were given artificial substrates treated with water, sinigrin, orBrassica napus (cv. Westar) squashes, combined withB. napus volatiles and/or grooves in the substrate. No eggs were deposited in the absence of olfactory and gustatory stimuli. Moths given gustatory but not olfactory stimuli deposited similar numbers of eggs but spent significantly more time performing olfactory-related behaviours. Conversely, moths given olfactory but not gustatory stimuli did not oviposit.SSubstrate grooves did not influence egg numbers but appeared to influence egg location. The order in which oviposition behaviours occurs and the relative importance of stimuli type are discussed.  相似文献

[目的]肠道细菌桃色欧文氏菌Erwinia persicina是小菜蛾Plutella xylostella幼虫肠道的优势细菌,本研究旨在阐明桃色欧文氏菌的代谢表型特征.[方法]采用BIOLOG细胞表型芯片技术系统研究了桃色欧文氏菌的细胞表型;采用PM l-10代谢板,对桃色欧文氏菌的950种代谢表型进行了测定.[结果]桃色欧文氏菌能代谢39.47%的碳源、89.74%的氮源、100%的硫源和100%的磷源;高效代谢的碳源为有机酸类和碳水化合物类,高效代谢的氮源为氨基酸类.该肠道细菌表现出261种不同的氮源代谢通路和95种生物合成通路.桃色欧文氏菌具有广泛的适应性,能在分别具有高达9%氯化钠、4%氯化钾、5%硫酸钠、20%乙二醇、6%甲酸钠、2%尿素、6%乳酸钠、200mmol/L磷酸钠(pH 7.0)、20 mmol/L苯甲酸钠(pH 5.2)、100 mmol/L硫酸铵(pH 8.0)、100 mmol/L硝酸钠和100 mmol/L亚硝酸钠的渗透溶液中正常代谢;其适应pH值范围为4.5 ~10,最适7.0.在多种氨基酸的作用下,桃色欧文氏菌均表现出脱羧酶和脱氨酶活性.[结论]桃色欧文氏菌的代谢特征增加了我们对该肠道细菌,特别是其与宿主昆虫的互作及其在肠道环境中的适应性的认识,同时表明通过降低桃色欧文氏菌密度来防治小菜蛾的可能性.  相似文献
