The localization and identification of the target cells for estradiol (E2) and dihydrotestosterone (DHT) were studied in the pituitary gland of male and female baboons (Papio cynocephalus) using autoradiography and autoradiography combined with immunocytochemistry. The largest number of cells which sequestered 3H-E2 were located in the pars distalis of both the males and females. Immunocytochemistry revealed that these target cells were predominantly gonadotrophs and mammotrophs, albeit a few somatotrophs and thyrotrophs were also labeled. A distinct sexual dimorphism was not observed with respect to the uptake and retention of the tritiated estrogen but was clearly evident in those animals given 3H-DHT. Significantly more cells in all three lobes of the male baboon, the partes distalis, intermedia and nervosa, concentrated the radiolabeled androgen as compared to that found in the females. Regardless of sex, however, the gonadotrophs were the major cell type in the pars distalis which was labeled. A small population of mammotrophs, somatotrophs and thyrotrophs also retained 3H-DHT. In both males and females, the pars nervosa was the portion of the hypophysis most sensitive to 3H-DHT. These findings indicate that all three lobes of the hypophysis of the male and female baboon contain target cells for E2 and DHT and that the propensity for DHT uptake is greater in males than in females.  相似文献

Glycoconjugates were localized by light microscopy with lectin-peroxidase conjugates and by electron microscopy with the periodic acid-thiocarbohydrazide-silver proteinate (PA-TCH-SP) sequence in immunocytochemically or morphologically identified cell types in rat pituitary. Lectin histochemistry demonstrated sialic acid and glycoconjugates with N-glycosidically linked oligosaccharides in gonadotrophs, thyrotrophs, and corticotrophs. Galactose penultimate to sialic acid was observed mostly in gonadotrophs. The terminal galactose-N-acetylgalactosamine disaccharide was detected in a few gonadotrophs and in a moderate number of mammotrophs. Fucose was localized in only corticotrophs with two fucose-binding lectins and in thyrotrophs with another. Several different monosaccharides were seen in glycoconjugates in melanotrophs and in Herring bodies. Melanotrophs displayed heterogeneous staining with fucose-binding lectins. A small number of nonsecretory cells were also visualized in the pars distalis by virtue of their glycogen content. PA-TCH-SP staining revealed complex carbohydrates in secretory granules and some Golgi cisternae in all types of hormone-producing cells in the pars distalis except for the somatotrophs. Melanotrophs of pars intermedia exhibited stained secretory granules and irregular dense bodies containing a stained meshwork. Corticotrophs of the pars distalis lacked the latter bodies, although they form the same glycoprotein precursor hormone as melanotrophs. Lectin conjugates and the PA-TCH-SP sequence stained some groups of secretion granules in Herring bodies, possibly representing vasopressin-containing granules as well as other cell types in the pars nervosa.  相似文献

Donor tissue containing Rathke's pouch (RP) with its associated mesenchyme and neural lobe was isolated from 15-day fetal rats and stereotaxically transplanted either to hypothalamic hypophysiotropic sites or to cerebral cortex of adult females for 30 days. Hosts either were intact or had been hypophysectomized 2-4 weeks prior to transplantation of Rathke's pouch. Grafts in the hypothalamus of either intact or hypophysectomized hosts were pleomorphic and large, often as wide as 1-2 mm, and occasionally larger. Grafts in the cortex of either intact or hypophysectomized hosts were nodular and occasionally projected upward in association with the meninges (cortex/meninges grafts). Certain features were characteristic of the grafts in all experimental groups, i.e., development of histotypic pars distalis with cell cords and fenestrated capillaries. In all experimental groups gonadotrophs and somatotrophs, when present, were localized at the graft margin adjacent to the connective-tissue interface; mammotrophs, when present, were distributed throughout the graft. Features specific to each experimental group also were apparent. Grafts in the hypothalamus of both intact and hypophysectomized hosts typically were encapsulated by a labyrinthine meshwork of cell processes, whereas cortex/meninges grafts directly abutted dense connective tissue or neural tissue. In hypothalamic grafts in intact hosts, moderately differentiated mammotrophs, gonadotrophs, and somatotrophs could be identified by their cytological features and immunopositivity for prolactin, luteinizing hormone, and growth hormone, respectively. In hypothalamic grafts in hypophysectomized hosts, mammotrophs were absent, and gonadotrophs and somatotrophs were poorly granulated and not abundant. Grafts in the cortex of intact hosts contained numerous, well-differentiated mammotrophs, gonadotrophs, and somatotrophs. Many of the mammotrophs in these grafts were hypertrophied, and profiles of exocytosis were common. In grafts in the cortex of hypophysectomized hosts, mammotrophs were either absent or very few, whereas gonadotrophs and somatotrophs were numerous. Gonadotrophs in these grafts were dramatically hypertrophied, although exocytosis was rare. The results indicate that development of histotypic pars distalis may occur in hypophysiotropic and non-hypophysiotropic brain sites and that the hormonal state of the host as well as implantation site modulate cytodifferentiation of specific pars distalis cell types.  相似文献

Summary The pars distalis of parturient harp seals (Pagophilus groenlandicus) is divisible into three distinct zones based on the major cellular components of the different regions. A clear zonation is lacking in neonate seals, the medial basophilic wedge and the anterior gonadotrophic were small and difficult to identify. Five cell types were identified based on cytological characteristics: somatotrophs, mammotrophs, thyrotrophs, gonadotrophs and stellate cells; corticotrophs could not be identified, with any certainty, in any of the preparations, nor could the gonadotrophs be classified into LH, FSH, and ICSH cells.The pars intermedia enlarged during the early post-partum period and was large in the parturient females.Abbreviations Used LH luteinizing hormone - FSH follicle stimulating hormone - ICSH interstitial cell stimulating hormone - PAS periodic acid-Schiff We wish to thank Mrs. Lucy Lin for her technical assistance. Summary The postnatal differentiation of somatotrophs and mammotrophs in the pars distalis of normal and dwarf (dw/dw) mice was analyzed by means of immunocytochemistry at the electron-microscopic level. Thin sections of Araldite-embedded pituitaries from postnatal mice ranging in age from 2 days to 3–4 months were exposed to antigrowth hormone serum (anti-GH) or to anti-prolactin serum (anti-PRL) and were processed immunocytochemically by means of the colloidal-gold technique. In the normal adult pars distalis, somatotrophs reacted strongly with antiGH, and mammotrophs showed a positive anti-PRL reaction. In the dwarf adult, the granulated cells were unreactive with either anti-GH or anti-PRL. However, at 6 days postnatally, some cells in the dwarf pars distalis showed a positive reaction with anti-GH, though the reaction was not as strong as in the 6-day normal pars distalis. The 6-day dwarf was only faintly reactive with anti-PRL, and by 14 days the dwarf was unreactive with anti-GH as well as anti-PRL, in contrast to the strong positive reactivity to these two antisera in the normal 14-day pars distalis.Supported by USPHS grant HD12308  相似文献

Chromogranins A, B and C, three distinct groups of proteins found in bovine chromaffin granules, were also found to be present in the pituitary using immunoblotting techniques. Their distribution was therefore studied in the normal ram pituitary using an immunoperoxidase technique applied to semithin serial sections and compared with that of some of the hormones of the anterior pituitary. Chromogranin-immunoreactivity was found in gonadotrophs (all three), thyrotrophs (A with some positive for C) and corticotrophs (a fraction with A and fewer with B and C). The mammotrophs and somatotrophs were negative. Chromogranin C was the only one of the three to be located in the pars nervosa, whilst chromogranin B was rarely found in the pars intermedia. The results suggest that chromogranins A, B and C are not always stored together, some hormone-containing cells do not contain immunohistologically detectable levels of the chromogranins.  相似文献

The cellular pathology and postnatal differentiation of somatotrophs and mammotrophs in the pars distalis of little (lit/lit) and normal (+/+; lit/+) mice were studied by means of electron microscopy. The results indicate that in little the pituitary at birth contains recognizable somatotrophs and mammotrophs; however, between 14 and 24 days of age a contrast between little and normal becomes conspicuous with respect to the somatotrophs. Little somatotrophs are less heavily populated with granules, have granules of smaller size, and show less developed organelles than do normal somatotrophs at comparable stages. Beyond 24 days the little somatotrophs become more difficult to locate, and those that do occur show a minimal increase in complexity from that present at 14 days. In contrast, the mammotrophs in little are similar in appearance to normal mammotrophs and show increasing complexity as development proceeds, often sending forth cellular processes between neighboring cells, as do normal mammotrophs.  相似文献

Summary The pars distalis of the pituitary gland of the C57BL mouse was studied by means of electron microscopy during postnatal stages of males ranging in age from newborn through 24 days, with particular emphasis on the somatotrophs and mammotrophs. During this period, growth curves were plotted in order to correlate postnatal growth patterns with the state of differentiation of the somatotrophs in the pars distalis. In the newborn, the somatotrophs show well developed organelles, including rough endoplasmic reticulum and Golgi complexes. These cells are not as densely packed with granules as the adult somatotrophs; however, from days 5 through 24, they show a progressive accumulation of granules. Although mammotrophs are scarce in the newborn, they are readily distinguishable in the pars distalis at 5 days. Male mice in small litters show a progressive increase in body weight between birth and 11–12 days, at which time the rate slackens until 18–19 days when the rate again increases. Growth curves for mice from large litters are similar to those from smaller litters, except that the transitory decrease in rate is more prominent.Supported by NIH grant no. The pituitary of Heteropneustes fossilis is divisible into rostral (RPD) and proximal (PPD) pars distalis, and pars intermedia (PI). Neurohypophysis extensively ramifies in the PI. The RPD is formed of acidophilic prolactin cells and lead hematoxylin (H. Pb) positive ACTH cells. The latter is arranged around the neurohypophysial extensions. The PPD is comprised of thyrotrophs (TSH) and gonadotrophs which are stainable with PAS, AF, and aniline blue. Acidophilic somatotrophs (STH) and chromophobes are scattered throughout the PPD. In the pars intermedia PAS positive and H. Pb positive cells are present. The latter is believed to secrete MSH whereas the function of the former is yet to be confirmed. In the partly and completely thyroidectomised H. fossilis the TSH cells exhibited marked hypertrophy. They are located in the middle of the PPD flanked by the gonadotrophs on either side. Hypertrophy of the ACTH cells were also evident. But the other cell types did not show any marked changes as compared to the controls.  相似文献

The RPD (rostral pars distalis) is formed of two cell types. The erythrosinophilic acidophils were hypertrophied in response to ammonium sulphate treatment and dark background adaptation. The response to former may be due to osmoregulatory involvement. The PbH positive corticotrophs were markedly hypertrophied and degranulated in response to metopirone and ammonium sulphate treatments. They were moderately activated in the dark and white background adapted fish. The PPD (proximal pars distalis) is composed of two types of cyanophils and acidophils. The thyrotrophs are identified by their hypertrophic response to radiothyroidectomy and thiourea treatment. They are distributed in the ventral part of the PPD, posterior neurohypophysis and PI (pars intermedia). The gonadotrophs which largely occupy the dorsal aspect of the PPD increase numerically as the gonads mature and become the major cell type before spawning. The acidophilic somatotrophs are distributed all over the PPD. They are the predominant cell type in the PPD of fish with immature gonads. They gave strong reaction to Baker's acid haematein indicating the presence of phospholipid. Both PbH and PAS cells of PI were stimulated in fish kept in dark background whereas they did not exhibit any obvious change under white background adaptation.  相似文献

The neuroendocrine hypothalamic-pituitary axis undergoes morphological and biochemical changes throughout gestation. In viscacha, pregnancy lasts approximately 154 days, and three stages can be described: early, mid- and late pregnancy. The aim of this work was to study the pituitary LH-gonadotrophs, FSH-gonadotrophs, somatotrophs, corticotrophs and thyrotrophs of non-pregnant and pregnant adult viscachas by immunohistochemistry and morphometric analysis. Immunopositive percentage area (%IA), cell percentage in the pars distalis (%PDC), number of cells per reference area (n°cell/RA), and major cellular (MCD) and nuclear (ND) diameters were analyzed. The different cell populations showed a well-defined morphology, immunolabeling patterns and regionalization in the pars distalis (PD). In the early pregnancy of animals the morphometric analysis demonstrated that %IA, %PDC and n°cell/RA increased in the FSH-gonadotrophs and decreased in the somatotrophs in relation to non-pregnant animals. In mid-pregnancy, there was an increase in %IA, %PDC, and n°cell/RA of LH-gonadotrophs, FSH-gonadotrophs, somatotrophs, and thyrotrophs. The MCD of LH-gonadotrophs and FSH-gonadotrophs increased. In late pregnancy, the %IA, %PDC and n°cell/RA of LH-gonadotrophs, FSH-gonadotrophs, somatotrophs and corticotrophs decreased whereas the values of the thyrotrophs remained constant. The MCD of LH-gonadotrophs, FSH-gonadotrophs and corticotrophs decreased. No significant changes were observed in the ND of the studied cell types. In conclusion, this work provides evidence for histological and morphometric changes in the different cell types of the pituitary PD in viscachas during pregnancy, probably according to the requirements of this physiological stage.  相似文献

We examined by immunocytochemistry the localization of cathepsin B in endocrine cells of rat anterior pituitary lobe, using a monospecific antibody to cathepsin B. By light microscopy, granular immunodeposits for cathepsin B were detected in most endocrine cells of anterior pituitary lobe. Cells immunoreactive for luteinizing hormone (LH) were diffusely immunostained by anti-cathepsin B. By electron microscopy, immunogold particles for cathepsin B were localized in lysosomes of thyrotrophs, somatotrophs, and mammotrophs. In mammotrophs, immunogold particles for cathepsin B were also detected in crinophagic bodies. Double immunostaining co-localized immunogold particles for LH and cathepsin B in secretory granules of gonadotrophs. Immunocytochemistry was also applied to demonstrate localization of renin and prorenin in LH-producing gonadotrophs; immunogold particles for renin were co-localized with those for LH, cathepsin B, or prorenin in their secretory granules. Immunogold particles for prorenin were also co-localized with those for LH or cathepsin B in secretory granules, but prorenin-positive granules appeared less frequently than renin-positive granules. These results suggest that cathepsin B not only plays a role in the protein degradation in lysosomes of anterior pituitary endocrine cells but also participates in the activation of renin in gonadotrophs, as has been demonstrated in secretory granules of juxtaglomerular cells.  相似文献

Summary Using the electron-microscopic immunogold method, vasotocin, isotocin, somatostatin (SRIF), gonadotrophin-releasing hormone (LHRH) and corticotrophin-releasing factor (CRF)-like immunoreactivities were localized in separate neurosecretory fibres in the pituitary of a teleost fish Poecilia latipinna. Antigenicities were preserved in sections of conventionally fixed tissue, except in the case of LHRH and CRF-like substances which were sensitive to osmium postfixation. Under the same fixation conditions, ultrastructural differences were observed between the 5 fibre types, and morphometric analysis of their granule sizes revealed significant differences in mean diameter except between vasotocin and isotocin fibres.Terminal-like regions of each type were identified on blood vessels, glial cells or other fibres in the neurohypophysis, on the basement lamina of the adenohypophysis, or directly on adenohypophysial endocrine cells. The fibres containing the two neurohypophysial hormones, originating from separate preoptic perikarya, were intermingled with, and may form endings near all the adenohypophysial cell types except those secreting prolactin. Although both types had similar mean granule diameters, the granules in the vasotocin fibres (mean 135 nm) were markedly less electron dense than those in the isotocin fibres (mean 140 nm). SRIF-immunoreactive fibres (mean 101 nm) appeared to form synapse-like endings on the somatotrophs, and a few thyrotrophs in the proximal pars distalis, and near the pars intermedia cells. An LHRH-positive type (mean 103 nm) contacted only the gonadotrophs of the proximal pars distalis. The rarer CRF-like fibres (mean 116 nm) appeared to project mainly towards the pars intermedia, but a few appeared to terminate rostrally near the adrenocorticotrophic cells.The significance of these observations is discussed in relation to the direct neurosecretory control of adenohypophysial function in teleosts.  相似文献

Summary The chronological appearance of endocrine cells in the pituitary of sea-bream (Sparus auratus) larvae was studied using antisera against salmon prolactin, trout growth hormone, salmon gonadotropin and N-terminal human adrenocorticotropin. The larval pituitary (1–12 days after hatching) was oval in shape and was composed of a dense mass of cells with few neurohypophysial fibres. By 60 days after hatching it began to resemble the adult and was divisible into a distinct rostral pars distalis containing prolactin and adrenocorticotropin cells; a proximal pars distalis containing somatotrophs and gonadotrophs and a pars intermedia. Cells immunoreactive with antisera against growth hormone were observed immediately after hatching (2 days post-fertilization). Weakly staining prolactin cells were observed 2 days later in the region corresponding to the rostral pars distalis. Cells immunoreactive with anti-gonadotropin and anti-adrenocorticotropin sera were observed in the pituitary 6 and 8 days after hatching, respectively. All the cell-types studied were immunoreactive from the time they were first identified until the final samples 90 days after hatching.  相似文献

The morphological characteristics and percentage of the cellular associations between gonadotrophs (LH- and FSH-secreting cells) and other cellular types were studied in pituitary pars distalis of adult male viscachas (Lagostomus maximus maximus) by double immunohistochemistry using specific antibodies to LH, FSH, PRL, GH, ACTH, TSH and S-100 protein (by folliculostellate cells; FSC), during long and short photoperiods. Bihormonal gonadotrophs were observed in ventro-medial and dorsal regions, interspersed between monohormonal gonadotrophs, and their number increased in short photoperiod. LH- and FSH-gonadotrophs were found around lactotrophs, enclosed by somatotrophs in the dorsal region, and associated with irregular corticotrophs. Gonadotrophs and thyrotrophs were associated along blood vessels and follicular structures. The cytoplasmic prolongations of FSC were in contact with both gonadotrophs. The percentage of LH–FSH, LH–ACTH, LH–FSC, FSH–LH, FSH–PRL, FSH–GH, FSH–ACTH, FSH–TSH and FSH–FSC associations decreased, whereas LH–PRL increased in short as compared to long photoperiod. The most abundant associations were LH–GH and LH–TSH during long photoperiod, but LH–GH and LH–PRL during short photoperiod. FSH–GH and FSH–PRL were the most numerous associations, and LH–FSC and FSH–FSC were the less abundant ones in both photoperiods. These results provide the morphological evidence for specific cellular associations between gonadotrophs and other cellular types of viscacha pituitary.  相似文献

The hypophysial portal vessels and anterior pituitary gland of adult male Wistar rats were exposed surgically. A hypophysial portal vessel was cannulated and infused for one minute with saline or thyrotrophin (TRH). Anterior pituitary glands were collected at 1,5,15,30 or 60 minutes after cessation of infusion, for light and electron microscopic examination. Before and immediately after cannulation of a portal vessel, a 1-ml sample of blood was collected at 1,5,15,30, or 60 minutes, from the femoral vein for radioimmunoassay (RIA) of growth hormone. Thyrotrophs from anterior pituitary glands of rats infused with TRH displayed emiocytic activity at all time-periods studied. Rough endoplasmic reticular (RER) cisternae were dilated at 15 minutes following infusion and remained dilated at 30 and 60 minutes. TRH was observed to stimulate emiocytic activity in most pituitary cell-types. Extensive dilations of RER cisternae were also observed in mammotrophs and gonadotrophs, but were not observed in somatotrophs or adrenocorticotrophs. The demonstration that thyrotrophs, mammotrophs, somatotrophs, and gonadotrophs respond to TRH suggests that some common features may be shared by these cells. Preliminary analysis of the RIA data show that TRH was potent in elevating radioimmunoassayable growth hormone levels. Significant increases (p less than 0.02) in plasma GH levels were present at the earlier time periods studied (1,5, and 15 minutes) following the infusion of TRH, but no at 30 or 60 minutes. These findings provide additional support for the non-specific action of TRH upon hte various adenohypophysial cell types, and demonstrate that TRH stimulates these cells by a direct action on the adenohypophysis.  相似文献

We examined orexin-like immunoreactivity in the pituitary of the red-bellied piranha (Pygocentrus nattereri). Orexin-B-immunoreactive (IR) cells corresponded to luteinizing hormone (LH)-containing cells in the pars distalis, and orexin-B-IR fibers corresponded to melanin-concentrating hormone (MCH)-containing fibers in the pars nervosa. In the pars distalis, orexin-B-IR puncta that were also immunoreactive for MCH were observed around the orexin-B-IR cells. In the ventral hypothalamus, orexin-B-IR and MCH-IR neurons were found in the nucleus lateralis tuberis. Immunoelectron-microscopic analysis revealed that the orexin-B-like substance co-localized with LH in secretory granules and with MCH in MCH-containing neurons. Some of the MCH secreted in the pituitary might participate in the modulation of LH secretion from the gonadotrophs, together with orexin-B, leading to food intake by the stimulation of growth hormone secretion from the somatotrophs.  相似文献

The uptake of L-dopa into the cells of the adenohypophysis of the rat was studied during the postnatal development and at adult age using the formaldehyde-induced fluorescence method (FIF). The cells taking up L-dopa were classified by Alcian blue-PAS-Orange G staining. The correlation between the cells taking up L-dopa and those containing tryptophyl-peptide was estimated during the postnatal period and in adult rats. The cells containing tryptophyl-peptide were demonstrated using fluorescence induced by treatment with combined formaldehyde and acetyl chloride vapour. The following observations were made: 1) Great majority of the cells taking up L-dopa did not contain tryptophyl-peptide. Thus the accumulation of L-dopa into the cells of pars distalis is not due to accumulation of L-dopa into the cells by the same transport mechanism as the amino acids for tryptophyl-peptide. 2) Of the cells taking up L-dopa in the adult rats 96% were chromophobes, 2.0% acidophilic cells (somatotrophs and cells producing prolactin), 0.9% R-mucoid cells (corticotrophs), and 1.2% S1- and S2-mucoid cells (gonadotrophs and thyrotrophs). At 10 and 25 days' age the relative numbers of the cells taking up L-dopa were about the same. 3) Pretreatment with nialamide caused only a slight increase in the number of the cells taking up L-dopa. The decrease in the number of the cells uptaking L-dopa of the pars distalis, which takes place after 5 weeks' age is thus not caused by the increased MAO-activity. 4) Strongly chromophilic cells did not take up L-dopa. At the light of our results it seems evident that L-dopa is taken up by the chromophobic cells when these differentiate into chromophilic cells. The accumulation of L-dopa may be a sign of an active transport of amino acids into the cells. The accumulation of L-dopa into the chromophobic stellate and follicular cells may reflect their metabolic activity. These cells probably have an important role in the production of the hormones of the pars distalis.  相似文献

Summary The freeze-etching technique was used in studies of cell surface phenomena during the release of secretory products from the adenohypophysis and from neurosecretory terminals of rats in which exocytosis had been stimulated by the administration of hypothalamic extracts (somatotrophs, thyrotrophs, gonadotrophs and mammotrophs) or severe hemorrhage (neurohypophysis). The observations suggest that secretory granules are extruded through an opening at the tip of a protrusion of the cellular surface. The protrusions seem to result from the abutting of secretory granules on the inner surface of the plasma membrane. Summary The immunocytochemical distribution of gonadotropin (GTH) in the goldfish pituitary gland was studied applying the peroxidase-antiperoxidase (PAP) method and the protein A-gold technique at lightand electron-microscopic levels, respectively, with an antiserum raised against silver carp GTH. In the light-microscopic immunocytochemistry, PAS-positive cells in the proximal pars distalis showed strong reaction with the antiserum. Gold particles were concentrated both on globules (large) and on granules (small) of the gonadotrophs (PAS-positive cells) in the electron-microscopic immunocytochemistry. Other cells in the pituitary gland, including thyrotrophs, displayed no immunoreactivity with the antiserum at the dilutions tested. These results indicate, not only immunocytochemical distribution of GTH both in globules and in granules in the gonadotrophs, but also the high purity of the antigen (silver carp GTH) and specificity of the antiserum.  相似文献
