To test the validity of previous results the inducibility of amylase as well as other biochemical parameters was measured using 45 homozygous strains of Drosophila melanogaster from Akayu, Japan. Only the inducibility (but not protein contents or specific activity of the enzyme) was highly correlated with productivity measured using a starch food regime (rp = 0.41, P < 0.005, rg = 0.73 ± 0.21). Inducibility was also negatively correlated with developmental time using starch food; namely, the one with high inducibility developed the fastest. Population cage experiments using 1600 genomes from the same natural population showed that the inducibility responded positively to natural selection (1.6-fold increase in inducibility in cages using starch food relative to those using normal food), but little frequency change of allozymes was observed. All of these results were consistent and indicated that polymorphisms of inducing factors or regulatory genes were major determinants of fitness differences in a particular environment and may be the genetic materials responsible for the adaptive evolution of organisms, at least in amylase loci.  相似文献

Fifty lethal-free, sterility-free isogenic lines of Drosophila melanogaster that were randomly sampled from a natural population were tested for net fitness and other components of fitness by competition with D. hydei. Larval viability and developmental time were also measured using the balanced marker method. Distribution patterns of these fitness components were similar, but correlation between the fitness components varied depending on the combinations used. The highest correlations were obtained between net fitness and productivity (rp = 0.6987, rg = 0.9269). The correlation between net fitness and total larval viability was much lower (rp = 0.1473 and rg = 0.2171). These results indicate that measuring net fitness, not just a component of fitness, is necessary for the good understanding of the genetic structures of natural populations.  相似文献

Heritability of bone mineral density (BMD) varies across skeletal sites, reflecting different relative contributions of genetic and environmental influences. To quantify the degree to which common genetic variants tag and environmental factors influence BMD, at different sites, we estimated the genetic (rg) and residual (re) correlations between BMD measured at the upper limbs (UL-BMD), lower limbs (LL-BMD) and skull (SK-BMD), using total-body DXA scans of ∼4,890 participants recruited by the Avon Longitudinal Study of Parents and their Children (ALSPAC). Point estimates of rg indicated that appendicular sites have a greater proportion of shared genetic architecture (LL-/UL-BMD rg = 0.78) between them, than with the skull (UL-/SK-BMD rg = 0.58 and LL-/SK-BMD rg = 0.43). Likewise, the residual correlation between BMD at appendicular sites (re = 0.55) was higher than the residual correlation between SK-BMD and BMD at appendicular sites (re = 0.20–0.24). To explore the basis for the observed differences in rg and re, genome-wide association meta-analyses were performed (n∼9,395), combining data from ALSPAC and the Generation R Study identifying 15 independent signals from 13 loci associated at genome-wide significant level across different skeletal regions. Results suggested that previously identified BMD-associated variants may exert site-specific effects (i.e. differ in the strength of their association and magnitude of effect across different skeletal sites). In particular, variants at CPED1 exerted a larger influence on SK-BMD and UL-BMD when compared to LL-BMD (P = 2.01×10−37), whilst variants at WNT16 influenced UL-BMD to a greater degree when compared to SK- and LL-BMD (P = 2.31×10−14). In addition, we report a novel association between RIN3 (previously associated with Paget''s disease) and LL-BMD (rs754388: β = 0.13, SE = 0.02, P = 1.4×10−10). Our results suggest that BMD at different skeletal sites is under a mixture of shared and specific genetic and environmental influences. Allowing for these differences by performing genome-wide association at different skeletal sites may help uncover new genetic influences on BMD.  相似文献

Selective breeding of animals for increased disease resistance is an effective strategy to reduce mortality in aquaculture. However, implementation of selective breeding programs is limited by an incomplete understanding of host resistance traits. We previously reported results of a rainbow trout selection program that demonstrated increased survival following challenge with Flavobacterium psychrophilum, the causative agent of bacterial cold water disease (BCWD). Mechanistic study of disease resistance identified a positive phenotypic correlation between post-challenge survival and spleen somatic-index (SI). Herein, we investigated the hypothesis of a genetic correlation between the two traits influenced by colocalizing QTL. We evaluated the inheritance and calculated the genetic correlation in five year-classes of odd- and even-year breeding lines. A total of 322 pedigreed families (n = 25,369 fish) were measured for disease resistance, and 251 families (n = 5,645 fish) were evaluated for SI. Spleen index was moderately heritable in both even-year (h2 = 0.56±0.18) and odd-year (h2 = 0.60±0.15) lines. A significant genetic correlation between SI and BCWD resistance was observed in the even-year line (rg = 0.45±0.20, P = 0.03) but not in the odd-year line (rg = 0.16±0.12, P = 0.19). Complex segregation analyses of the even-year line provided evidence of genes with major effect on SI, and a genome scan of a single family, 2008132, detected three significant QTL on chromosomes Omy19, 16 and 5, in addition to ten suggestive QTL. A separate chromosome scan for disease resistance in family 2008132 identified a significant BCWD QTL on Omy19 that was associated with time to death and percent survival. In family 2008132, Omy19 microsatellite alleles that associated with higher disease resistance also associated with increased spleen size raising the hypothesis that closely linked QTL contribute to the correlation between these traits. To our knowledge, this is the first estimation of spleen size heritability and evidence for genetic linkage with specific disease resistance in a teleost fish.  相似文献

The number of ovarioles of the Drosophila melanogaster ovary is a trait thought to be associated with female fecundity, and therefore is expected to be under strong natural selection. This hypothesis may be tested by examining patterns of genetic and environmental variation for ovariole number in natural populations, and by determining the association between ovariole number and fitness in isogenic lines derived from a natural population. We measured ovariole number, and competitive fitness and its components, for 48 homozygous chromosome 3 substitution lines in a standard inbred background; and body size in a sample of 15 chromosome 3 substitution lines. We found significant segregating genetic variation for ovariole number, with a broad-sense heritability (H2) of 0.403 and correspondingly high coefficients of genetic variation (CVC = 20.8) and residual variation (CVR = 25.3). Estimates of quantitative genetic parameters for body size (H2 = 0.191, CVG = 2.15, and CVR = 3.87) are similar to those previously reported for this trait. Although the isogenic chromosome 3 substitution lines varied significantly for components of fitness, there was no significant linear or quadratic association of ovariole number and body size with fitness. There was, however, highly significant sex × genotype interaction for fitness among these lines. This special case of genotype × environment interaction for fitness may contribute to the maintenance of genetic variation for fitness in natural populations.  相似文献

In the Atacama Desert from northern Chile (19–24°S), Prosopis (Leguminosae) individuals are restricted to oases that are unevenly distributed and isolated from each other by large stretches of barren landscape constituting an interesting study model as the degree of connectivity between natural populations depends on their dispersal capacity and the barriers imposed by the landscape. Our goal was to assess the genetic diversity and the degree of differentiation among groups of Prosopis individuals of different species from Section Algarobia and putative hybrids (hereafter populations) co‐occurring in these isolated oases from the Atacama Desert and determine whether genetic patterns are associated with dispersal barriers. Thirteen populations were sampled from oases located on three hydrographic basins (Pampa del Tamarugal, Rio Loa, and Salar de Atacama; northern, central, and southern basins, respectively). Individuals genotyped by eight SSRs show high levels of genetic diversity (H O = 0.61, A r = 3.5) and low but significant genetic differentiation among populations (F ST = 0.128, F ST‐ENA = 0.129, D JOST = 0.238). The AMOVA indicates that most of the variation occurs within individuals (79%) and from the variance among individuals (21%); almost, the same variation can be found between basins and between populations within basins. Differentiation and structure results were not associated with the basins, retrieving up to four genetic clusters and certain admixture in the central populations. Pairwise differentiation comparisons among populations showed inconsistencies considering their distribution throughout the basins. Genetic and geographic distances were significantly correlated at global and within the basins considered (p < .02), but low correlation indices were obtained (r < .37). These results are discussed in relation to the fragmented landscape, considering both natural and non‐natural (humans) dispersal agents that may be moving Prosopis in the Atacama Desert.  相似文献

About 400 second chromosomes were extracted from the Aomori population, a northernmost population of D. melanogaster on Honshu in Japan, and the following experimental results were obtained. (1) The frequency of lethal chromosomes was 0.23. (2) The effective size of the population was estimated to be about 3000, from the allelism rate of lethal chromosomes and their frequency. (3) The detrimental and lethal loads for viability were 0.243 and 0.242, respectively, and the D/L ratio became 1.00. (4) The average degree of dominance for mildly deleterious genes was estimated to be 0.178 ± 0.056. (5) Additive (σ2A) and dominance (σ2D) variances of viability were estimated to be 0.00276 ± 0.00090 and 0.00011 ± 0.00014, respectively. (6) There was no significant difference in environmental variances between homozygotes and heterozygotes. Using these estimates, we discuss the maintenance mechanisms of genetic variability of viability in the population. The mutation-selection balance explained these experimental results.  相似文献

In a large experiment, using nearly 200 population cages, we have measured the fitness of Drosophila melanogaster homozygous (1) for the second chromosome, (2) for the third chromosome, and (3) for both chromosomes. Twentyfour second chromosomes and 24 third chromosomes sampled from a natural population were tested. The mean fitness of the homozygous flies is 0.081 ± 0.014 for the second chromosome, 0.080 ± 0.017 for the third chromosome, and 0.079 ± 0.024 for both chromosomes simultaneously. Assuming that fitnesses are multiplicative (the additive fitness model makes no sense in the present case because of the large selection coefficients involved), the expected mean fitness of the homozygotes for both chromosomes is 0.0066; their observed fitness is more than ten times greater. Thus, it appears that synergistic interactions between loci are considerable; and that, consequently, the fitness function substantially departs from linearity. Two models are tentatively suggested for the fitness function: a "threshold" model and a "synergistic" model.—The experiments reported here confirm previous results showing that the concealed genetic load present in natural populations of Drosophila is sufficient to account for the selective maintenance of numerous polymorphisms (of the order of 1000).  相似文献

Genetic parameters for meat quality traits and their relationships with body weight and breast development were estimated for a total of 420 male turkeys using REML. The birds were slaughtered in a commercial plant and the traits measured included pH at 20 min (pH20) and 24 h post-mortem (pHu) and colour of the breast and thigh meat. The heritabilities of the rate and the extent of the pH fall in the breast muscle were estimated at h2 = 0.21 ± 0.04 and h2 = 0.16 ± 0.04, respectively. Heritabilities ranging from 0.10 to 0.32 were obtained for the colour indicators in the breast muscle. A marked negative genetic correlation (rg = -0.80 ± 0.10) was found between pH20 and lightness (L*) of breast meat, both traits corresponding to PSE indicators. The pH20 in the thigh muscle had a moderate heritability (h2 = 0.20 ± 0.07) and was partially genetically related to pH20 in the breast muscle (rg = 0.45 ± 0.17). Body weight and breast yield were positively correlated with both initial and ultimate pH and negatively with the lightness of breast meat.  相似文献

Uncovering how natural selection and genetic drift shape the evolutionary dynamics of virus populations within their hosts can pave the way to a better understanding of virus emergence. Mathematical models already play a leading role in these studies and are intended to predict future emergences. Here, using high-throughput sequencing, we analyzed the within-host population dynamics of four Potato virus Y (PVY) variants differing at most by two substitutions involved in pathogenicity properties. Model selection procedures were used to compare experimental results to six hypotheses regarding competitiveness and intensity of genetic drift experienced by viruses during host plant colonization. Results indicated that the frequencies of variants were well described using Lotka-Volterra models where the competition coefficients βij exerted by variant j on variant i are equal to their fitness ratio, rj/ri. Statistical inference allowed the estimation of the effect of each mutation on fitness, revealing slight (s = −0.45%) and high (s = −13.2%) fitness costs and a negative epistasis between them. Results also indicated that only 1 to 4 infectious units initiated the population of one apical leaf. The between-host variances of the variant frequencies were described using Dirichlet-multinomial distributions whose scale parameters, closely related to the fixation index F ST, were shown to vary with time. The genetic differentiation of virus populations among plants increased from 0 to 10 days post-inoculation and then decreased until 35 days. Overall, this study showed that mathematical models can accurately describe both selection and genetic drift processes shaping the evolutionary dynamics of viruses within their hosts.  相似文献

Individual selection based on female performance only was conducted in four lines of mice: L+ for increased litter size, W+ for increased 6-week body weight, L-W+ for a selection index aimed at decreasing litter size and increasing 6-week body weight and L+W- for a selection index aimed at increasing litter size and decreasing 6-week body weight. A fifth line (K) served as an unselected control. All litters were standardized to eight mice at one day of age. Expected heritability was based on twice the regression of offspring on dam (h2d), which contains additive genetic variance due to direct (σ2Ao) and maternal (σ2Am) effects and their covariance (σAoAm). Responses and correlated responses were measured either deviated (method 1) or not deviated (method 2) from the control line. Realized heritabilities (h2R) for litter size were 0.19 ± 0.04 (1) and 0.16 ± 0.03 (2), which were similar to h 2d of 0.17 ± 0.04. The h2 R for 6-week body weight of 0.55 ± 0.07 (1) and 0.44 ± 0.07 (2) agreed with h2d of 0.42 ± 0.02. Realized genetic correlations (r*GR) between litter size and 6-week body weight calculated from the double-selection experiment were 0.52 ± 0.10 (1) and 0.52 ± 0.13 (2), which were not significantly different from the base population estimate of r* Gd = 0.63 ± 0.14. Divergence (L-W + minus L+W-) in the antagonistic index selection lines was 0.21 ± 0.01 index units (I = 0.305 PW - 0.436 PL, where P W and PL are the phenotypic values for 6-week body weight and litter size, respectively.). The h2 R of index units of 0.14 ± 0.02 calculated from divergence agreed with h2d of 0.14 ± 0.04. Divergences in litter size (-0.19 ± 0.07) and 6-week body weight (0.46 ± 0.10) were in the expected direction. Antagonistic index selection yielded about one-half the expected divergence in litter size, while divergence in 6-week body weight was only slightly less than expected. Realized genetic correlations indicated that litter size, 6-week body weight and index units each showed positive pleiotropy with 3-week body weight, postweaning gain and weight at vaginal introitus and negative pleiotropy with age at vaginal introitus. Sex ratio and several components of fitness (days from joining to parturition, percent fertile matings and percent perinatal survival) did not change significantly in the selected lines.  相似文献

The evolution of population dynamics in a stochastic environment is analysed under a general form of density-dependence with genetic variation in r and K, the intrinsic rate of increase and carrying capacity in the average environment, and in σe2, the environmental variance of population growth rate. The continuous-time model assumes a large population size and a stationary distribution of environments with no autocorrelation. For a given population density, N, and genotype frequency, p, the expected selection gradient is always towards an increased population growth rate, and the expected fitness of a genotype is its Malthusian fitness in the average environment minus the covariance of its growth rate with that of the population. Long-term evolution maximizes the expected value of the density-dependence function, averaged over the stationary distribution of N. In the θ-logistic model, where density dependence of population growth is a function of Nθ, long-term evolution maximizes E[Nθ]=[1−σe2/(2r)]Kθ. While σe2 is always selected to decrease, r and K are always selected to increase, implying a genetic trade-off among them. By contrast, given the other parameters, θ has an intermediate optimum between 1.781 and 2 corresponding to the limits of high or low stochasticity.  相似文献

Background

Naturally occurring coinfections of pathogens have been reported in salmonids, but their consequences on disease resistance are unclear. We hypothesized that 1) coinfection of Caligus rogercresseyi reduces the resistance of Atlantic salmon to Piscirickettsia salmonis; and 2) coinfection resistance is a heritable trait that does not correlate with resistance to a single infection.

Methodology

In total, 1,634 pedigreed Atlantic salmon were exposed to a single infection (SI) of P. salmonis (primary pathogen) or coinfection with C. rogercresseyi (secondary pathogen). Low and high level of coinfection were evaluated (LC = 44 copepodites per fish; HC = 88 copepodites per fish). Survival and quantitative genetic analyses were performed to determine the resistance to the single infection and coinfections.

Main Findings

C. rogercresseyi significantly increased the mortality in fish infected with P. salmonis (SI mortality = 251/545; LC mortality = 544/544 and HC mortality = 545/545). Heritability estimates for resistance to P. salmonis were similar and of medium magnitude in all treatments (h 2 SI = 0.23±0.07; h 2 LC = 0.17±0.08; h 2 HC = 0.24±0.07). A large and significant genetic correlation with regard to resistance was observed between coinfection treatments (rg LC-HC = 0.99±0.01) but not between the single and coinfection treatments (rg SI-LC = −0.14±0.33; rg SI-HC = 0.32±0.34).

Conclusions/Significance

C. rogercresseyi, as a secondary pathogen, reduces the resistance of Atlantic salmon to the pathogen P. salmonis. Resistance to coinfection of Piscirickettsia salmonis and Caligus rogercresseyi in Atlantic salmon is a heritable trait. The absence of a genetic correlation between resistance to a single infection and resistance to coinfection indicates that different genes control these processes. Coinfection of different pathogens and resistance to coinfection needs to be considered in future research on salmon farming, selective breeding and conservation.  相似文献

Tilia cordata Mill. is a valuable tree species enriching the ecological values of the coniferous‐dominated boreal forests in Europe. Following the historical decline, spreading of Tilia sp. is challenged by the elevated inbreeding and habitat fragmentation. We studied the geographical distribution of genetic diversity of Tilia cordata populations in Lithuania. We used 14 genomic microsatellite markers to genotype 543 individuals from 23 wild‐growing populations. We found that Tilia cordata retained high levels of genetic diversity (population F IS = 0–0.15, H o = 0.53–0.69, H e = 0.56–0.75). AMOVA, Bayesian clustering, and Monmonier''s barrier detection indicate weak but significant differentiation among the populations (F ST = 0.037***) into geographically interpretable clusters of (a) western Lithuania with high genetic heterogeneity but low genetic diversity, bottleneck effects, (b) relatively higher genetic diversity of Tilia cordata on rich and most soils of midland lowland, and (c) the most differentiated populations on poor soils of the coolest northeastern highland possessing the highest rare allele frequency but elevated inbreeding and bottleneck effects. Weak genetic differentiation among the Tilia cordata populations in Lithuania implies common ancestry, absence of strong adaptive gradients, and effective genetic exchange possible mediated via the riparian networks. A hypothesis on riparian networks as gene flow mediators in Tilia cordata was raised based on results of this study.  相似文献

Genic variation in male haploids and male diploids was compared assuming constant fitnesses (derived from computer-generated random numbers) and infinite population size. Several models were studied, differing by the fitness correlation between the sexes (rs) and genotypes (rg), and by the intensity of selection as measured by the coefficient of variation (CV) of the fitness distribution. Genic variation was quantified using the proportion of polymorphic loci, P, the gene diversity at polymorphic loci, Hp, and the gene diversity over all loci, Ha. The two genetic systems were compared via variation ratios: variation in male haploidy/variation in male diploidy.—P and Ha were markedly lower for male-haploids than for male diploids, the variation ratios declining with increasing rs, rg and CV, but the two genetic systems were similar for Hp. Except for male diploids with rs = 1, the two sexes had different equilibrium gene frequencies but the sample sizes required to detect such differences reliably were larger than usually possible in surveys of natural populations.—Data from natural populations fit the above trends qualitatively, but the variation ratios are much lower than those from our analyses, except that for Hp, which is higher when Drosophila is excluded. Also, the frequency distribution of most common alleles from electrophoretic data has a deficiency of intermediate frequencies compared to that from the computer-generated sets of fitnesses, possibly reflecting either the influence of stochastic processes shifting frequencies away from equilibrium or the involvement of alleles under selection-mutation balance.——While electrophoretic data suggest that Drosophila has unusually high levels of genic variation, unusually low levels of genic variation in male haploids compared with male diploids are not strongly indicated. However, if further data confirm male haploids as having low levels of genic variation, likely explanations are that the bulk of electrophoretically detected variation involves fixed-fitness balancing selection, selection-mutation balance involving slightly deleterious recessive alleles, new favorable male haploid alleles moving more rapidly to fixation than under male diploidy and thus carrying linked loci to fixation faster, or some combination of these possible factors.  相似文献

Hyadaphis foeniculi (Passerini) (Hemiptera: Aphididae) is a cosmopolitan species and the main pest of fennel in northeastern Brazil. Understanding the relationship between temperature variations and the population growth rates of H. foeniculi is essential to predict the population dynamics of this aphid in the fennel crop. The aim of this study was to measure the effect of constant temperature on the adult prereproductive period and the life table fertility parameters (infinitesimal increase ratio (rm), gross reproduction rate (GRR), net reproduction rate (R0), finite increase ratio (λ), generation time (GT), the time required for the population to double in the number of individuals (DT), and the reproduction value (RVx)) of the fennel pest H. foeniculi. The values of lx (survival of nymphs at age x) increased as the temperature rose from 15 to 28°C and fell at 30°C, whereas mx (number of nymphs produced by each nymph of age x) increased from 15 to 25°C and fell at 28 and 30°C. The net reproduction rates (R0) of populations of H. foeniculi increased with temperature and ranged from 1.9 at 15°C to 12.23 at 28°C for each generation. The highest population increase occurred with the apterous aphids at 28°C. The rate of population increase per unit time (rm) (day) ranged from 0.0033 (15°C) to 0.1995 (28°C). The highest values of rm were recorded at temperatures of 28°C and 30°C. The rm values were a good fit to the models tested, with R2 > 0.91 and R2 adj > 0.88. The models tested (Davidson, Sharpe and DeMichele modified by Schoolfield et al., Logan et al., Lamb, and Briere et al.) were very good fits for the rm values observed, with R2 > 0.91 and R2 adj > 0.88. The only exception was the Davidson model. Of the parameters studied, the reproductive capacity was higher in the apterous aphids, with the unique exception of daily fecundity at 28°C, which was higher in the alate aphids of H. foeniculi. Parameters relating to the age-specific fertility table for H. foeniculi were heavily influenced by temperature, with the highest biotic potential and population growth capacity found at 34°C. Therefore, the results obtained in this study could be of practical significance for predicting outbreaks of fennel aphids and improving the management of this aphid in fennel crops.  相似文献

The dynamics of a gene in a haploid population can be explained approximately by considering the average reproductive value of the gene. The dynamics of the average reproductive value are similar to those of a gene in a population with nonoverlapping generations with the following modifications: The effective population size, Ne, replaces N; the average mutation rates µ* and ν* replace µ and ν; the average overall selection r*+(T-1)s** replaces s; and time is measured in terms of generations, T. Phylogeographic research concerning Central China has been rarely conducted. Population genetic and phylogeography of Ziziphus jujuba var. spinosa (also called sour jujube) were investigated to improve our understanding of plant phylogeographic patterns in Central China. Single‐copy nuclear gene markers and complete chloroplast genome data were applied to 328 individuals collected from 21 natural populations of sour jujube in China. Nucleotide variation of sour jujube was relatively high (π = 0.00720, θ w = 0.00925), which resulted from the mating system and complex population dynamics. Analysis of molecular variation analysis revealed that most of the total variation was attributed to variation within populations, and a high level of genetic differentiation among populations was detected (F st = 0.197). Relatively low long‐distance dispersal capability and vitality of pollen contributed to high genetic differentiation among populations. Differences in the environmental conditions and long distance among populations further restricted gene flow. Structure clustering analysis uncovered intraspecific divergence between central and marginal populations. Migrate analysis found a high level of gene flow between these two intraspecific groups. Bayesian skyline plot detected population expansion of these two intraspecific groups. Network and phylogeny analysis of chloroplast haplotypes also found intraspecific divergence, and the divergence time was estimated to occur at about 55.86 Ma. Haplotype native to the Loess Plateau was more ancient, and multiple glacial refugia of sour jujube were found to locate at the Loess Plateau, areas adjacent to the Qinling Mountains and Tianmu Mountains. Species distribution model analysis found a typical contraction‐expansion model corresponding to the Quaternary climatic oscillations. In the future, the distribution of sour jujube may shift to high‐latitude areas. Backgrounds and Aims

The stem growth habit, determinate or indeterminate, of soybean, Glycine max, varieties affects various plant morphological and developmental traits. The objective of this study is to identify the effect of stem growth habit in soybean on the stomatal conductance of single leaves in relation to their leaf morphology in order to better understand the ecological and agronomic significance of this plant trait.

Methods

The stomatal conductance of leaves on the main stem was measured periodically under favourable field conditions to evaluate gmax, defined as the maximum stomatal conductance at full leaf expansion, for four varieties of soybean and their respective determinate or indeterminate near isogenic lines (NILs). Leaf morphological traits including stomatal density, guard cell length and vein density were also measured.

Key Results

The value of gmax ranged from 0·383 to 0·754 mol H2O m−2 s−1 across all the genotypes for both years. For the four pairs of varieties, the indeterminate lines exhibited significantly greater gmax, stomatal density, numbers of epidermal cells per unit area and total vein length per unit area than their respective determinate NILs in both years. The guard cell length, leaf mass per area and single leaf size all tended to be greater in the determinate types. The variation of gmax across genotypes and years was well explained by the product of stomatal density and guard cell length (r = 0·86, P < 0·01).

Conclusions

The indeterminate stem growth habit resulted in a greater maximum stomatal conductance for soybean than the determinate habit, and this was attributed to the differences in leaf structure. This raises the further hypothesis that the difference in stem growth habit results in different water use characteristics of soybean plants in the field. A thermoanaerobe (Thermoanaerobacter sp.) grown in TYE-starch (0.5%) medium at 60°C produced both extra- and intracellular pullulanase (1.90 U/ml) and amylase (1.19 U/ml) activities. Both activities were produced at high levels on a variety of carbon sources. The temperature and pH optima for both pullulanase and amylase activities were 75°C and pH 5.0, respectively. Both the enzyme activities were stable up to 70°C (without substrate) and at pH 4.5 to 5.0. The half-lives of both enzyme activities were 5 h at 70°C and 45 min at 75°C. The enzyme activities did not show any metal ion activity, and both activities were inhibited by β- and γ-cyclodextrins but not by α-cyclodextrin. A single amylolytic pullulanase responsible for both activities was purified to homogeneity by DEAE-Sepharose CL-6B column chromatography, gel filtration using high-pressure liquid chromatography, and pullulan-Sepharose affinity chromatography. It was a 450,000-molecular-weight glycoprotein composed of two equivalent subunits. The pullulanase cleaved pullulan in α1,6 linkages and produced multiple saccharides from cleavage of α-1,4 linkages in starch. The Kms for pullulan and soluble starch were 0.43 and 0.37 mg/ml, respectively.  相似文献
