The stability constants and the thermodynamic parameters of the formation of the binary complexes of trivalent Am3+, Cm3+ and Eu3+ with CDTA and of their ternary complexes with CDTA + IDA were determined by solvent extraction measurements in aqueous solutions of I = 6.60 m (NaClO4) at temperatures of 0-60 °C. The endothermic enthalpy and the positive entropy values reflect the significant effects of cation dehydration and of the rigidity of the ligand structure in the formation of these complexes. TRLFS and NMR (1H and 13C) data provided information on the structure of the ternary complexes in solution. The size and rigidity of CDTA affect the binding mode of IDA in the complexation of M(CDTA)(IDA)(H2O)3− and M(CDTA)(IDA)3− in which IDA has a bidentate coordination mode in the former and a tridentate coordination mode in the latter. 相似文献

The binary complexation of Am3+, Cm3+and Eu3+ with citrate has been studied at I = 6.60 m (NaClO4), pcH 3.60 and in the temperatures range of 0-60 °C employing a solvent extraction technique with di-(2-ethylhexyl)phosphoric acid/heptane. Two complexes, MCit and , were formed at all temperatures. For the three metal ions, the log β101 was between 5.9 and 6.2 and log β102 between 10.2 and 10.6 at 25 °C. The thermodynamic parameters for the Am-Cit system have been calculated from the temperature dependence of the β101 and β102 values. Positive enthalpy and entropy values for the formation of both complexes are interpreted as due to the contributions from the dehydration of the metal ions exceeding the exothermic cation-anion pairing. The formation of the ternary complex M(EDTA)(Cit)4− (M = Cm and Eu) was measured to have large stability constants (log β111 between 20.9 and 24.4) at 25 and 60 °C. Time resolved laser luminescence spectroscopy and lifetime measurement data validated the nature of the complexes of Eu(III) formed in the presence of Cit and EDTA + Cit in 6.60 m (NaClO4) solution. 相似文献

The iridium 1,1,1-tris(diphenylphosphinomethyl)ethane (triphos) complexes [{κ2(C1,C4)-CRCRCRCR}{CH3C(CH2PPh2)3}Ir(NCMe)]BF4 (2-NCMe, R = CO2Me) and [{κ2(C1,C4)-CRCRCRCR}{CH3C(CH2PPh2)3}Ir(CO)]BF4 (2-CO, R = CO2Me) serve as models for proposed iridium-vinylidene intermediates of relevance to the [2 + 2 + 1] cyclotrimerization of alkynes. The stability constants of the binary and the ternary complexes of Pu4+ have been measured for certain carboxylate and aminocarboxylate ligands in aqueous solution of I = 5.0 M (1 M perchloric acid + 4 M ionic strength perchlorate media) and temperatures of 0-45 °C by the solvent extraction technique. The stability constants of the binary and the ternary complexes increased with increased temperature. The complexation enthalpy and entropy of the binary Pu-Ox and the ternary Pu-EDTA-Ox and DGA complexes indicated the stability of these complexes is due to the highly favorable entropy contribution while complexation enthalpies either oppose complexation or are weakly favorable. 相似文献

Reaction of [CuIIL⊂(H2O)] (H2L = N,N′-ethylenebis(3-ethoxysalicylaldimine)) with nickel(II) perchlorate in 1:1 ratio in acetone produces the trinuclear compound [(CuIIL)2NiII(H2O)2](ClO4)2 (1). On the other hand, on changing the solvent from acetone to methanol, reaction of the same reactants in same ratio produces the pentametallic compound [(CuIIL)2NiII(H2O)2](ClO4)2·2[CuIIL⊂(H2O)]·2MeOH (2A), which loses solvated methanol molecules immediately after its isolation to form [(CuIIL)2NiII(H2O)2](ClO4)2·2[CuIIL⊂(H2O)] (2B). Clearly, formation of 1 versus 2A and 2B is solvent dependent. Crystal structures of 1 and 2A have been determined. Interestingly, compound 2A is a [3 × 1 + 1 × 2] cocrystal. The cryomagnetic profiles of 1 and 2B indicate that the two pairs of copper(II)···nickel(II) ions in the trinuclear cores in both the complexes are coupled by almost identical moderate antiferromagnetic interaction (J = −22.8 cm−1 for 1 and −26.0 cm−1 for 2B). 相似文献

Aneuploid cells are frequently observed in human tumors, suggesting that aneuploidy may play an important role in the development of cancer. In this review, I discuss the processes that may give rise to aneuploid cells in normal tissue and in tumors. Aneuploid cells may arise directly from diploid cells through errors in chromosome segregation, as a consequence of incorrect microtubule-kinetochore attachments, or through failure of the spindle checkpoint. A second route to formation of aneuploid cells is through a tetraploid intermediate, where division of tetraploid cells can yield very high rates of chromosome missegregation as a consequence of multipolar spindle formation. Diploid cells may become tetraploid through a variety of mechanisms, including endoreduplication, cell fusion, and cytokinesis failure. Although aneuploid cells may arise from either diploid or tetraploid cells, the fate of the resulting aneuploid cells may be distinct. It is therefore important to understand the different pathways that can give rise to aneuploid cells, and how the varied origins of these cells affect their subsequent ability to survive or proliferate. 相似文献

Establishing the origin of electrophoretic mobility of sarcoplasmic reticulum (SR) vesicles is the primary goal of this work. It was found that the electrophoretic mobility originates from ionizable amino acids of cytoplasmic domains of the Ca2+-ATPase, the calcium pump of SR. The mobility was measured at pH 4.0, 4.7, 5.0, 6.0, 7.5, and 9.0 in the region of ionic strength from 0.05 to 0.2 M. Mobility measurements were supplemented by studies of SR vesicles by photoelectron microscopy. The median diameter of SR vesicles was 260 nm. Ca2+-ATPases were not resolved. The mobility data were standardized by interpolation to a reference ionic strength of 0.1 M. The mobility of the SR vesicles is determined by the charge of the Ca2+-ATPase. It is due to the ionizable amino acids selected from the amino acid sequence of SERCA1a Ca2+-ATPase. The pH dependence of charge residing in various domains of Ca2+-ATPase was computed using pKa values in free water. The charge correlated with measured mobility. It was shown that a linear relationship exists between the mobility of the SR vesicles, μ, and the total computed charge, Q, on three cytoplasmic domains of Ca2+-ATPase: A, P, and N. It is given by μ = α + β Q where the fitted values β = (0.043 ± 0.002) × 10−8 m2 V−1 s−1 e−1 and α = (0.16 ± 0.02) × 10−8 m2 V−1 s−1. Since β and α values do not change from pH 4 to pH 9, one concludes that the hydrodynamic friction of the cytoplasmic domains of SR is independent of their charge. 相似文献

New ternary transition metal complexes of formulations [Co(bpa)(p-HB)2](bpa = 2,2′-bipyridylamine, p-HB = p-hydroxybenzenecarboxylic acid) (1), [Ni(bpa)(p-HB)(H2O)2]+(NO3)− · H2O (2), , [Cu(bpa)(p-HB)Cl] (4) and [Zn(bpa)(p-HB)2]2 · 0.5H2O (5) are prepared, their structural features are characterized by crystal structural studies, and their DNA binding propensity has been evaluated by fluorescence method. The molecular structure of complex 1 shows the six coordinate octahedral geometry with one bpa and two p-HB ligands, complex 2 is the cationic complex and has the six coordinate octahedral structure with one bpa, one p-HB and two aqua ligands, complex 3 is also the cationic complex of octahedral coordination with two bpa and one p-HB ligands, complex 4 is five coordinate distorted square pyramidal with one bpa, one p-HB and chloride ligands and complex 5 has the distorted octahedral coordination with two p-HB and one bpa ligands. In all of the complexes, both bpa and p-HB act as the bidentate N and O-donor ligands, respectively. The intermolecular H-bond networks, together with π-π interaction in their solid state are also described. The complexes show the competitive inhibition of ethidium binding to DNA, and the DNA binding propensity can be reflected as the relative order: 3 > 2 > 1 > 5 > 4, in which the cationic charged Ni(II) complexes 2 and 3 show the most effective inhibition ability. 相似文献

This work is an experimental and theoretical study of electrostatic and hydrodynamic properties of the surface of sarcoplasmic reticulum (SR) membrane using particle electrophoresis. The essential structural components of SR membrane include a lipid matrix and a dense layer of Ca2 +-ATPases embedded in the matrix. The Ca2 +-ATPase layer both drives and impedes vesicle mobility. To analyze the experimental mobility data, obtained at pH 4.0, 4.7, 5.0, 6.0, 7.5, and 9.0 in 0.1 M monovalent (1:1) electrolyte, an analytical solution for the vesicle mobility and electroosmotic flow velocity distribution was obtained by solving the Poisson–Boltzmann and the Navier–Stokes–Brinkman equations. The electrophoretic mobility model includes two sets of charges that represent: (a) charged lipids of the lipid matrix of the vesicle core, and (b) charged amino acid residues of APN domains of Ca2 +-ATPases. APN domains are assumed to form a charged plane displaced from the surface of lipid matrix. The charged plane is embedded in a frictional layer that represents the surface layer of calcium pumps. Electrophoretic mobility is driven by the charged APN domain and by lipid matrix while the surface layer provides hydrodynamic friction. The charge of APN domain is determined by ionized amino acid residues obtained from the amino acid composition of SERCA1a Ca2 +-ATPase. Agreement between the measured and the predicted mobility is evaluated by the weighted sum of mobility deviation squared. This model reproduces the experimental dependence of mobility on pH and predicts that APN domains are located in the upper half of the SR vesicle surface layer. 相似文献

Cobalt(III) and rhodium(III) complexes of the series of [MIIICl3 − n(P)3 + n]n+ (M = Co or Rh; n = 0, 1, 2 or 3) have been prepared with the use of 1,1,1-tris(dimethylphosphinomethyl)ethane (tdmme) and mono- or didentate phosphines. The single-crystal X-ray analyses of both series of complexes revealed that the M-P and M-Cl bond lengths were dependent primarily on the strong trans influence of the phosphines, and secondarily on the steric congestion around the metal center resulting from the coordination of several phosphine groups. In fact, the M-P(tdmme) bonds became longer in the order of [MCl3(tdmme)] < [MCl2(tdmme)(PMe3)]+ < [MCl(tdmme)(dmpe)]2+ (dmpe = 1,2-bis(dimethylphosphino)ethane) < [M(tdmme)2]3+ for both CoIII and RhIII series of complexes, while the M-Cl bond lengths were shortened in this order (except for [M(tdmme)2]3+). Such a steric congestion around the metal center can also account for the structural and spectroscopic characteristics of the series of complexes, [MCl(tdmme)(dmpm, dmpe or dmpp)]2+ (dmpm = bis(dimethylphosphino)methane, dmpp = 1,3-bis(dimethylphosphino)propane). The X-ray analysis for [CoCl(tdmme)(dmpm or dmpe)](BF4)2 showed that all Co-P bonds in the dmpm complex were shorter by 0.03-0.04 Å than those in the dmpe complex. Furthermore, the first d-d transition energy of the CoIII complexes and the 1JRh-P(tdmme) coupling constants observed for the RhIII complexes indicated an unusual order in the coordination bond strengths of the didentate diphosphines, i.e., dmpm > dmpe > dmpp. 相似文献

A synthetic strategy for the covalent anchoring of nickel β-diketonate complexes on Si(1 0 0) has been examined. Engineered Si(1 0 0) surfaces were prepared by the Si-grafting of 10-undecylenic acid methyl ester followed by hydrolysis of the ester to free the carboxylic functions suited for the anchoring of the Ni complex. Bis(pentane-2,4-dionate)Ni(II) was bonded to the functionalized surface from the gas phase by the exchange of the acetylacetonate ligand with the grafted acid. The surface density of the anchored Ni complex was controlled by tuning the surface concentration of carboxylic groups adopting a mixed monolayer of undecylenic acid and 1-decene used as a spectator spacer. The nickel decorated silicon surfaces were characterized by attenuate total reflectance infrared absorption spectroscopy (ATR-IRAS) and angle resolved X-ray photoelectron spectroscopy (AR-XPS). 相似文献

Three new binary Cu(II) complexes of norfloxacin have been synthesized and characterized. We also report the synthesis, characterization and X-ray crystallographic structures of a new binary compound, [Cu(HNor)(2)]Cl(2).2H(2)O (2) and two new ternary complexes norfloxacin-copper(II)-phen, [Cu(Nor)(phen)(H(2)O)](NO(3)).3H(2)O (4), and [Cu(HNor)(phen)(NO(3))](NO(3)).3H(2)O (5). The structure of 2 consists of two crystallographically independent cationic monomeric units of [Cu(HNor)(2)](2+), chloride anions, and uncoordinated water molecules. The Cu(II) ion is placed at a center of symmetry and is coordinated to two norfloxacin ligands which are related through the inversion center. The structures of 4 and 5 consist of cationic units ([Cu(Nor)(phen)(H(2)O)](+) for 4 and [Cu(HNor)(phen)(NO(3))](+) for 5), nitrate counteranions, and lattice water molecules that provide crystalline stability through a network of hydrogen-bond interactions. The complexes exhibit a five coordinated motif in a square pyramidal environment around the metal center. The ability of compounds 4 and 5 to cleave DNA has also been studied. Mechanistic studies with different inhibiting reagents reveal that hydroxyl radicals, singlet oxygen, and superoxide radicals are all involved in the DNA scission process mediated by these compounds. 相似文献

The reaction of [RuCl2(PPh3)3] and [OsBr2(PPh3)3] precursors with a series of heterocyclic bidentate (N, X) ligands, X = S, Se, gave complexes [M(R-pyS)2(PPh3)2], (R = H, 3-CF3, 5-CF3, 3-Me3Si); [M(R-pymS)2(PPh3)2], (R = 4-CF3, 4,6-MeCF3) and [M(R-pySe)2(PPh3)2], (R = H, 3-CF3, 5-CF3), where M is Ru or Os, pyS and pymS the anions of pyridine-2-thione and pyrimidine-2-thione, respectively, and pySe is the anion produced by the reductive cleavage of the Se-Se bond in the dipyridyl-2,2′-diselenide. All of the compounds obtained were characterized by microanalysis, IR, FAB, NMR spectroscopy and by cyclic voltammetry. Compounds [Ru(3-CF3-pyS)2(PPh3)2] · 2(CH2Cl2) (2), [Ru(3-Me3Si-pyS)2(PPh3)2] (4), [Ru(4-CF3-pymS)2(PPh3)2] (5), [Ru(3-CF3-pySe)2(PPh3)2] · 2(CH2Cl2) (8), [Os(3-CF3-pyS)2(PPh3)2] · (CHCl3) (11), [Os(3-Me3Si-pyS)2(PPh3)2] (13), [Os(3-CF3-pySe)2(PPh3)2] · 2(CH2Cl2) (17), [Os(5-CF3-pySe)2(PPh3)2] · 2(H2O) (18) and [OsCl2(4,6-MeCF3-pymS)(PPh3)2] (19) were also characterized by X-ray diffraction. Phosphoryl-transfer reactions have long been of interest due to their importance in maintaining numerous cellular functions. A phosphoryl-transfer reaction results in two possible stereochemical outcomes: either retention or inversion of configuration at the transferred phosphorus atom. When the product is phosphate, isotopically-labeled [16O, 17O, 18O]-phosphate derivatives can be used to distinguish these outcomes; one oxygen must be replaced by sulfur or esterified to achieve isotopic chirality. Conventionally, stereochemical analysis of isotopically chiral phosphate has been based on 31P NMR spectroscopy and involves complex chemical or enzymatic transformations. An attractive alternative would be direct determination of the enantiomeric excess using chiroptical spectroscopy. (S)-Methyl-[16O, 17O, 18O]-phosphate (MePi∗), 7 and enantiomeric [16O, 17O, 18O]-thiophosphate (TPi∗), 10, were previously reported to exhibit weak electronic circular dichroism (ECD), although with 10 the result was considered to be uncertain. We have now re-examined the possibility that excesses of 7 and 10 enantiomers can be detected by ECD spectrometry, using both experimental and theoretical approaches. 7 and both the (R) and (S) enantiomers of 10 (10a, 10b) were synthesized by the ‘Oxford route’ and characterized by 1H, 31P and 17O NMR, and by MS analysis. Weak ECD could be found for 7, with suboptimal S/N. No significant ECD could be detected for the 10 enantiomers.Time-dependent DFT (TDDFT) calculations of the electronic excitation energies and rotational strengths of the same three enantiomers were carried out using the functional B3LYP and the basis set 6-311G∗∗. The isotopically-perturbed geometries were predicted using the anharmonic vibrational frequency calculational code in GAUSSIAN 03. In the case of 10, calculations were also carried out for the hexahydrated complex to investigate the influence of the aqueous solvent. The predicted excitation wavelengths are greater than the observed wavelengths, a not unusual result of TDDFT calculations. The predicted anisotropy ratios are 2.9 × 10−5 for 7, −5.3 × 10−6 for 10a/b, and 1.7 × 10−6 for 10a/b⋅(H2O)6. For 7 the predicted anisotropy ratio approximates that observed in this work, 4.5 × 10−5 at 208 nm. For 10a/b, the upper limits of the experimental anisotropy ratios (<5 × 10−6 at 225 nm, pH 9; <5 × 10−6 at 236 nm, pH 12) are comparable to the predicted magnitude of the value for 10a/b. The lower predicted value for 10a/b · (H2O)6 suggests that the aqueous environment affects the ECD significantly. Altogether, the TDDFT calculations together with a stereochemical analysis based on NMR and the MS data support the conclusion that the experimental ECD results for MePi∗ and TPi∗ may be reliable in order of magnitude. 相似文献

Aim
To investigate the association between interleukin-6 (IL-6) − 174G > C and − 572C > G polymorphisms and risk for ischemic stroke (IS) in young patients.Methods
We genotyped IL-6 − 174G > C and − 572C > G in a case–control study of 430 young IS patients and 461 control subjects. An unconditional multiple logistical regression model was used to calculate the effects of IL-6 − 174G > C and − 572C > G polymorphisms on IS risk.Results
Higher body mass index, diabetes, hypertension, obesity, and smoking were associated with risk of ischemic stroke. Multivariate regression analyses showed that subjects carrying the − 174CC genotype (OR = 1.69, 95% CI = 1.16–2.57) and C allele (OR = 1.37, 95% CI = 1.09–1.67) had a small but significant increased risk of IS. Similarly, those carrying the − 572GG genotype (OR = 2.12, 95% CI = 1.18–3.82) and G allele (OR = 1.43, 95% CI = 1.14–1.83) had a moderate increased risk of IS. We found the − 174G > C and − 572C > G polymorphisms interact with hypertension and obesity.Conclusion
Our results suggest that polymorphisms in IL-6 − 174G > C and − 572C > G are associated with IS risk in young patients, and that these polymorphisms interact with hypertension, obesity and etiologic subtypes. Calcium-dependent distance changes have been determined by resonance energy transfer in binary and ternary troponin complexes in order to collect evidence for the structural rearrangements which are part of the hypothetical trigger mechanism of skeletal muscle contraction. Donor and acceptor fluorophores were either intrinsic tryptophans in subunits with a favourable sequence from different species, quasi-intrinsic Tb3+ ions bound to troponin C or extrinsic labels attached to specific cysteine or methionine residues. All chemically modified subunits proved fully active in conferring calcium sensitivity onto myosin ATPase. Nine distances were determined between five sites which allowed construction of a three-dimensional lattice representing the spatial distribution of four sites in the ternary complex of troponin C, I and T. Distances in binary complexes were nearly unaltered upon addition of the third subunit. Regulatory calcium binding caused distance changes of the order of 0.7-1.1 nm. In view of the large displacements of the hypothetical mechanism, they turned out to be smaller than anticipated. The fluorophoric sites selected may be localized in a zone of the troponin complex which happens to be relatively little affected by the mechanism. Alternatively, amplification of the moderate changes seen here would require the complete set of thin filament proteins. 相似文献

Neutral, hexacoordinated “3 + 2” mixed ligand oxorhenium (1) and oxotechnetium (2) complexes of the general formula MO[SNO][NN], where M = Re or 99Tc, SNO is 2-mercaptoethyl-N-glycine and NN is 2,2′-bipyridine (bpy), were synthesized by simultaneous action of the tridentate SNO and the bidentate NN ligand on ReOCl3(PPh3)2 or 99TcO-gluconate precursors in a 1:1:1 molar ratio. Both complexes were characterized by elemental analysis, IR and NMR spectroscopy. X-ray structure determination of rhenium complex 1 revealed a distorted octahedral coordination geometry where the SNO donor atoms of the tridentate ligand and one bpy nitrogen atom occupy the equatorial positions of the octahedron, whereas the second bpy nitrogen atom and the oxo-group fill the apical positions. 相似文献
