为了提高细菌视紫红质(BR)的可组装怀,使之使用于生物传感器等生物器件,利用生物素对紫膜进行修饰,使之可以被亲和素识别,从而可以定向的固定于固体支撑物表面。实验结果表明:生物素可以修饰紫膜表面的赖氨酸,修饰的程序依修饰的时间不同而有所不同,但即使被修饰24小时的紫膜,其表面的赖氨酸仍然没有被完全修饰。同时,生物素的修饰不会影响紫膜的结构和功能,但是不同的结合位点对M412的衰减会产生不同的影响。这  相似文献

本文用吸收光谱和可见圆二色谱研究了不同浓度的山莨菪碱对紫膜中菌紫质结构的影响,并设计了用不同浓度的去垢剂Triton X-100作为脂环境的扰动剂,研究山莨菪碱对菌紫质的影响与膜脂关系的实验.结果表明山莨菪碱不仅影响菌紫质分子本身的构象变化而且扰动了菌紫质分子之间的激子偶联作用.通过吸收差光谱技术表明山莨菪碱对菌紫质结构的影响与膜脂密切相关并指出紫膜中菌紫质的三体结构对膜功能的贡献是不容忽视的.  相似文献

在很宽波长和PH范围内用紫外及可见区光吸收、圆二色及共振拉曼测定了酰化诱导的紫膜溶液的光谱变化。结构表明:酰化引起的表面电位的改变诱导了蛋白局部构象的改变。这种改变在很大程度上离域到整个蛋白分子中而影响了视黄醛结合位点的性质及生色团的结构,从而导致可观察的光谱变化。与未酰化紫膜比较,紫膜对PH的稳定性减小了。  相似文献

为进一步验证麦胚凝集素作用下膜血型糖蛋白A空间构象的变化,本文用傅里叶变换红外技术,定量测定了麦胚凝集素作用下,溶液血型糖蛋白A和脂蛋白体膜血型糖蛋白A的二级结构的改变,发现麦胚凝集素结合于血型糖蛋白A导致血型糖蛋白Aα—螺旋减少,β—结构增加,麦胚凝集素的抑制剂N—乙酰葡萄糖胺对麦胚凝集素诱导的血型糖蛋白A二级结构的改变有抑制作用。本文同时用扫描隧道显微术直接观察了麦胚凝集素结合前后膜血型糖蛋白A分子形态的变化。  相似文献

Brain slices were prepared from 17-day old rats, and incubated with [3H]glycine or [3H]-leucine to label proteins. Myelin was isolated from the slices, and the proteins were separated by discontinuous gel electrophoresis in buffers containing sodium dodecyl sulfate. Radioactive basic and Wolfgram proteins appeared in myelin at similar initial rates, and their entry was nearly linear between 15 and 120 min with no detectable lag. Radioactive proteolipid protein appeared in myelin at one-fourth the rate of the basic and Wolfgram proteins between 0 and 30 min, then entered at a rate comparable to the other proteins between 45 and 120 min. When cycloheximide (0.2 mM) or puromycin (1.0 mM) was added, appearance of newly labeled basic and Wolfgram proteins in myelin stopped while proteolipid protein continued to appear in myelin at a normal rate for at least 30 min. Chase experiments with unlabeled glycine had similar effects. These results indicate the existence of a previously synthesized precursor pool of proteolipid protein with a 30-min interval between synthesis of proteolipid protein and its appearance in myelin. Incorporation of [3H]fucose into glycoprotein of the myelin sheath was studied, as was inhibition of incorporation of radioactivity by the use of either cycloheximide, or dilution with unlabeled fucose. The results indicated fucosylation of a sizable pool of presynthesized protein and a delay of 30 min between fucosylation of these polypeptides and their subsequent appearance in myelin as glycoproteins.  相似文献

Direct evidence has been presented to confirm the existence of a spiral in the myelin sheaths of the central nervous system. An account of some of the variations in structure of central myelin sheaths has been given and it has been shown that the radial component of myelin sheaths has the form of a series of rod-like thickenings of the intraperiod line. These thickenings extend along the intraperiod line in a direction parallel to the length of the axon. The relative position of the internal mesaxon and external tongue of cytoplasm has been determined in a number of transverse sections of sheaths from the optic nerves of adult mice, adult rats, and young rats. In about 75 per cent of the mature sheaths examined, these two structures were found within the same quadrant of the sheath, so that the cytoplasm of the external tongue process tends to lie directly outside that associated with the internal mesaxon. The frequency with which the internal mesaxon and external tongue lie within the same quadrant of the sheath increases both with the age of the animal and with the number of lamellae present within a sheath. The possible significance of these findings is discussed.  相似文献

A study of the effects of hypertonic solutions on the structure of peripheral nerve myelin reveals an expansion rather than a contraction of the layer spacing. This suggests the absence of "free" water between the myelin layers. Hypotonic solutions bring about a change in radial repeat period from 171 A to 250 to 270 A. 紫膜蛋白菌紫质(bacteriorhodopsin,BR)处于不同的膜脂环境中,通过吸收光谱、荧光光谱和闪光动力学光谱的测定,比较了3种不同膜脂环境对菌紫质分子结构和功能的影响。实验结果表明:天然膜脂是BR最稳定的膜脂环境,可以形成以三聚体为单位的二维六角形品格结构。二豆蔻酰脂酰胆碱(dimyristoyl phosphatidylcholine,DMPC)也是BR分子的稳定脂环境,但以单体形式存在于膜脂环境中,功能受到一定影响。经TritonX-100增溶处理的紫膜膜脂环境中BR为单体,不稳定,容易发生结构和功能的变化。  相似文献

本文报道了pH7.5时血红蛋白和红细胞膜的结合效应.在10—45℃温度范围内观察到血红蛋白对膜脂质流动性的限制作用.看来这种限制作用不是脂质过氧化所致,而是血红蛋白和红细胞膜直接作用的结果.对流动性大的膜,血红蛋白的效应也随之增大.高铁血红蛋白及红细胞膜去胆固醇皆能修饰血红蛋白和膜的相互作用.  相似文献

本文以荧光探针为手段,以人红细胞膜为材料,测量了膜偏振度的改变,荧光探针能量转移,荧光峰的蓝移和甲素激发峰的分裂。结果表明在有竹红菌甲素存在时,红细胞膜偏振度增加,探针荧光强度减小,荧光峰蓝移。甲素浓度增加时,上述现象更加明显,即它们之间有正的相关关系。同时,甲素激发光谱的a带发生分裂。据此,我们认为甲素对红细胞膜内脂双层产生明显微扰,甲素与红细胞膜间存在着相互作用。在甲素浓度较大时,它主要是渗入到红细胞膜脂双层的深层部位(膜脂肪酸链的12—16位)。  相似文献

In the study of the drying kinetics of nerve fibres, at least five "phases" of water evaporation can be distinguished. A consideration of the accompanying changes in low-angle x-ray diffraction patterns permits a tentative identification of the "phases" and a quantitative interpretation of the data in terms of the water distribution in nerve fibres. These results suggest that the myelin sheath of frog sciatic nerve contains 40 to 50 per cent water, and it is suggested further that the greater part of this water is "organised" in relation to the hydrophilic groups of the lipide and protein components.  相似文献

Abstract— Microsomal and myelin membrane fractions were prepared from the brains of warm-adapted (room temperature) and hibernating Syrian hamsters ( Mesocricetus auratus ). Lipid extracts of these preparations were assayed for phospholipid and galactosphingolipid composition, and for cholesterol levels. In both myelin and microsomes, plasmenlethanolamine levels decreased while total ethanol-amineglycerophospholipid levels remained constant with hibernation. Cerebroside levels changed slightly, increasing in microsomes while decreasing in myelin. No changes in cholesterol levels were detectable. Fatty acid analyses of microsomal ethanolamineglycerophospholipids and phosphatidylserine showed predominantly increases in 18:1 and 20:4 (n-6), and decreases in 18:0 and 22:6 (n-3), in both lipid classes with hibernation. Myelin ethanolamineglycerophospholipids exhibited a decrease in 20:1 and an increase in 20:4 (n-6). Aldehyde analyses of plasmenylethanolamines revealed a decrease in 16:0 and an increase in 18:1 in microsomes, and an increase in 18:O in myelin. The hydroxylated fatty acids of myelin cerebrosides showed no discernible changes in composition with hibernation. It is proposed that these lipid changes aid in the maintenance of the structure and function of brain membranes at the reduced temperatures encountered during hibernation.  相似文献

用小角X射线衍射(SAXD)和差示扫描量热(DSC)方法研究胆固醇对二棕榈酸磷脂酰胆碱多层膜结构的影响。在不同胆因醇浓度及不同温度下的衍射实验和热吸收曲线实验显示纯磷脂的尖锐的凝胶-液晶相变随胆固醇含量增加而逐渐消失。当胆固醇含量约为40(mol)%或更多时,衍射曲线和热吸收曲线均显示存在分相现象。  相似文献

The effects on myelin of autolysis in situ after death and after purification were studied in normal brains and spinal cords and in those made edematous as a result of chronic triethyl tin (TET) feeding. Myelin prepared from normal and edematous brains and spinal cords autolyzed for 12 h at 4°C contained only slightly less basic protein than that prepared from freshly killed animals. The amounts of a light lipid-protein fraction (dissociated myelin) usually obtained during purification of myelin from edematous CNS were about the same in tissue from freshly killed rats and those autolyzed for 12 h at 4°C. Autolysis for 12 h at room temperature resulted in formation of large amounts of dissociated myelin and loss of basic protein, but more dissociation and basic protein loss occurred in CNS from edematous brains and spinal cords than from the normal. Purified myelin prepared from freshly-killed normal and TET-fed rats was incubated at 37°C in media of several ionic strengths. In Krebs-Ringer bicarbonate (physiological extracellular fluid) extensive dissociation of myelin occurred with much less in 0.04 M-Tris buffer, pH 7.2, and only small amounts were formed in 0.01 M-Tris. In all cases myelin from edematous CNS formed more dissociated fraction than did the normal myelin. Basic protein loss was also proportional to the ionic strength of the media, but there was no difference in loss between normal and TET-myelin. Two different factors, proteolysis and physical extraction of basic protein by salt solutions, may be contributing to myelin dissociation and loss of basic protein.  相似文献
