Triacylglycerol, which was one of the minor lipid components in immature seeds of tobacco, accumulated dramatically between 7 and 27 days after flowering and, in mature seeds at 37 days, the fatty acid methyl esters of the triacylglycerols comprised 96.3% of those of the total lipids. Diacylglycerols and sterol ester also increased significantly during seed development. Phosphatidylcholine and phosphatidylethanolamine, which were major components in immature seeds, decreased constantly with increasing maturation as well as the quantities of phosphatidylinositol and phosphatidylglycerol. Monogalactosyldiacylglycerols, digalactosyldiacylglycerols and sulfoquinovosyldiacylglycerols also decreased and disappeared in mature seeds. In the triacylglycerols the percentages of palmitate, stearate and linolenate fell with increasing seed age, while that of linoleate increased up to 75.3% in mature seeds. A similar trend was observed in the fatty acid composition in the diacylglycerols and sterol ester. Generally, in the phospholipids the proportions of linoleate and linolenate decreased with concomitant increases of stearate and oleate.  相似文献

Maturation of mustard (Sinapis alba) seed proceeds with a sharp decrease in the amounts of palmitic and linoleic acids in the total lipids up to 6 weeks after flowering (WAF). Concomitantly, the concentration of oleic acid increases, reaching a plateau at 4 WAF, which is followed by chain elongation of oleic acid to gadoleic and erucic acids. Compositional changes in constituent fatty acids of individual lipid classes indicate that the very long-chain monounsaturated fatty acids (C20 and C22), as opposed to common long-chain fatty acids (C16 and C18), are metabolized to triacylglycerols mainly by esterification to preformed diacylglycerols and monoacylglycerols, rather than via esterification to glycerol-3-phosphate or lysophosphatidic acids.  相似文献

High levels of ‘alkaline’ invertase activity occur in dormant mung bean seeds. During germination this activity decreases rapidly and is replaced by high ‘acid’ invertase activity. Cycloheximide prevented the formation of the latter activity and also inhibited germination. It is suggested that de novo synthesis of ‘acid’ invertase occurs during germination. Both enzymes bind to concanavalin A and, hence, are presumed to be glycoproteins. Affinity-purified enzyme samples show similar ratios of ‘acid’ and ‘alkaline’ invertase activities to the crude preparations indicating that specific enzyme inhibitors or activators are probably not involved in controlling the activities in vivo.  相似文献

Fatty acid composition and lipid synthesis in developing safflower seeds   总被引:1,自引:0,他引:1  
Linoleic acid predominated in every lipid class during the whole period of seed development of safflower, while linolenic acid decreased with increasing maturation and it was not detected in mature seeds. Just before the initiation of triacylglycerol accumulation, the fatty acid composition of triacylglycerols changed more rapidly than those of phospholipids and glycolipids. Saturated fatty acids tended to accumulate at the 1- and 3-positions of the glycerol molecule and the more highly unsaturated acids at the 2-position. The fatty acid compositions at the 1- and 3-positions were similar in all cases investigated, but in none of the triacylglycerols was the distribution completely symmetrical. The positional distribution of linolenic acid in triacylglycerols prepared from the immature seeds 2 days after flowering and from the leaves was unusual; in spite of its highest degree of unsaturation, it was preferentially esterified at the 1- and 3-positions. When triacylglycerol was most rapidly accumulated (14–18 days after flowering), the incorporation of acetate-[U- 14C] into total lipids was also maximum and dienoic fatty acids were the principal acids labelled. Diacylglycerols and compound lipids reached the highest rate of synthesis 15 days after flowering, and then a maximum incorporation into triacylglycerol occurred 18 days after flowering. Incubation temperature affected the synthesis of individual lipid classes. Triacylglycerol was more rapidly synthesized at 32° than at 10°, while diacylglycerols and compound lipids were accumulated under the low-temperature condition. A rise of incubation temperature caused a depression in dienoic acid synthesis.  相似文献

Electroblotting method employing a semidry blotting apparatus for the subsequent protein microsequence analysis (Hirano, 1987) was improved. This method is convenient and allows rapid and efficient transfer of the proteins from a polyacrylamide gel (1 mm thick) onto the Polybrene-coated glass-fiber sheet or polyvinylidene difluoride membrane filter in only 20 min. The electroblotted proteins could be sequenced directly with the gas-phase protein sequencer at a 20-pmole level. This method was applied to the sequence analysis of winged bean seed proteins. A portion of the crude extracts from only one-twentieth of a seed of the winged bean was separated by two-dimensional polyacrylamide gel electrophoresis and electroblotted, and the N-terminal amino acid sequences of the blotted proteins were analyzed. The sequences of about 60% of the blotted major proteins, including nine Kunitz trypsin inhibitor-like proteins with heterogeneity in the N-terminal sequences, a protein that has a homologous sequence to the leghaemoglobin, nitrogen-fixing root nodule-specific protein, and a soybean basic 7S globulin-like protein could be easily identified.  相似文献

The neutral fraction of nonstarch lipids in developing brown rice (Oryza sativa L., cv IR42) was accumulated up to 16 days after flowering (DAF), but phospholipids and glycolipids increased only up to 8 DAF. Fatty acids accumulated in nonstarch lipids until 12 DAF. However, the proportion of linolenic acid in the lipid fraction decreased and that of oleic acid increased during this period. Accumulation of fat-by-hydrolysis in the brown rice occurred until 20 DAF and followed closely that of starch. The proportion of linolenic acid decreased and that of linoleic acid increased until 16 DAF. The fatty acid composition of fat-by-hydrolysis and starch lipids were identical and fat-by-hydrolysis accounted for 48% by weight of starch lipids. Nonstarch lipids were mainly composed of triglycerides and were located in the bran and embryo of mature brown rice. Starch lipids were mainly composed of lysophosphatidyl choline, free fatty acids and lysophosphatidyl ethanolamine, and were located in the endosperm.  相似文献

Etiolated Arachis hypogaea seedlings grown in the presence of sodium chloride and sodium sulphate were analysed for their percent fatty acid composition. Seeds of natural populations of Capsella bursa-pastoris, collected from temperate regions, weighed less and had a higher lipid content than those from colder regions. The long-chain (16:0, 18:0, 18:1, 18:2 and 18:3) and very long-chain (20:0, 20:1, 20:2 and 20:3) fatty acid compositions were, however, quite similar in the lipids of all the seed samples which indicates a rigid genetic, rather than environmental, control of fatty acid biosynthesis. Characteristics of the seeds of the diploid species C. rubella and C. grandiflora were similar to those of the tetraploid C. bursa-pastoris, with the exception of the distinctly lower lipid content in C. grandiflora seeds.  相似文献

Phosphorylase has been fractionated during development and germination of seeds of smooth and wrinkled-seeded peas. The total phosphorylase levels have been compared. In addition, a number of other pea tissues and other legumes have been examined. Some kinetic properties of the two enzymes present have been measured. Both enzymes have been further purified by affinity chromatography on Sepharose 4B-starch columns and by sequential gel filtration in the absence and presence of amylopectin. The MW and sub-unit structures of the two enzymes have been examined and their possible roles discussed.  相似文献

通过气相色谱 GC 分析了华山松籽油的主要脂肪酸组成.结果表明:华山松籽含油率为56.5%,其中主要含有亚油酸63%,油酸26%,棕榈酸5%,硬脂酸2%,花生酸1%,芥酸1%,不饱和脂肪酸总量达90%.抗氧化研究采用Schaal烘箱法63±1℃,即在松籽油中加入几种不同的抗氧化剂进行加速氧化.结果表明:抗氧化剂二丁基对甲苯酚、没食子酸丙酯、特丁基对苯二酚对松籽油有明显的抗氧化作用,其中特丁基对苯二酚效果较好,柠檬酸对其具有协同增效作用,添加50mg/kg柠檬酸,抗氧化效果显著.VE无显著作用.  相似文献

The objective of this study was to identify single nucleotide polymorphisms (SNPs) in the thioesterase (TE) domain of the bovine fatty acid synthase (FASN) gene and to evaluate the extent to which they were associated with beef fatty acid composition. The four exons in FASN that encode for the TE domain were sequenced, and three SNPs, AF285607:g.17924A>G, g.18663T>C and g.18727C>T, were identified. Purebred Angus bulls (n = 331) were classified into three genotype groups, g.17924AA (n = 121), g.17924AG (n = 168) and g.17924GG (n = 42). The g.17924A>G genotype was significantly associated with fatty acid composition of longissimus dorsi muscle of Angus bulls. Cattle with the g.17924GG genotype had lower myristic acid (C14:0; P < 0.0001), palmitic acid (C16:0, P < 0.05) and total saturated fatty acid contents (P < 0.01), greater health index (P < 0.001), oleic acid content (C18:1; P < 0.001) and total monounsaturated fatty acid concentration (P < 0.01) in the total lipids and triacylglycerols fraction than did those with the g.17924AA genotype. Because of the linkage disequilibrium between SNPs g.17924A>G and g.18663T>C, similar significant associations of fatty acid contents with the g.18663T>C genotypes were observed. In conclusion, the SNPs g.17924A>G and g.18663T>C may be used as DNA markers to select breeding stock that have a healthier fatty acid composition.  相似文献

Seeds of Norway maple ( Acer platanoides L.) differing in water content (10, 20 and 30%) were stored for 6 weeks at 20 to 30°C. During this period changes in phospholipids and fatty acids as well as in seed viability and germination capacity were studied. A considerable decrease in the phospholipid content was observed, which depended on the water content in the seeds and was related to the decrease of the seed germination capacity. The level of linoleic (18:2) and linolenic (18:3) acids in the phospholipid fraction decreased considerably in the course of the accelerated seed ageing. The results obtained suggest that phospholipid degradation and peroxidation of unsaturated fatty acids, followed by membrane destruction, play a considerable role in maple seed ageing.  相似文献

Winged seeds, or samaras, are believed to promote the long‐distance dispersal and invasive potential of wind‐dispersed trees, but the full dispersive potential of these seeds has not been well characterised. Previous research on the ecology of winged seeds has largely focussed on the initial abscission and primary dispersal of the samara, despite it being known that the primary wind dispersal of samaras is often over short distances, with only rare escapes to longer distance dispersal. Secondary dispersal, or the movement of the seeds from the initial dispersal area to the site of germination, has been largely ignored despite offering a likely important mechanism for the dispersal of samaras to microhabitats suitable for establishment. Herein, we synthesise what is known on the predation and secondary dispersal of winged seeds by multiple dispersive vectors, highlighting gaps in knowledge and offering suggestions for future research. Both hydrochory and zoochory offer the chance for samaroid seeds to disperse over longer distances than anemochory alone, but the effects of the wing structure on these dispersal mechanisms have not been well characterised. Furthermore, although some studies have investigated secondary dispersal in samaroid species, such studies are scarce and only rarely track seeds from source to seedling. Future research must be directed to studying the secondary dispersal of samaras by various vectors, in order to elucidate fully the invasive and colonisation potential of samaroid trees.  相似文献

Winged bean (Psophocarpus tetragonolobus) is of rapidly increasing interest as a high-protein multipurpose crop; in the future the winged bean could become as important as the soybean in world agriculture, with the added bonus of yielding substantial quantities of edible, high-protein root tubers. During the survey of root-knot nematodes (Meloidogyne species) on vegetable crops in Uttar Pradesh (India), it was observed that winged bean was restricted to home gardens and backyard cultivation where it was found severe galling of roots and reduces tuber production and may affect pod and seed yield. Sometimes leaf anthracnose disease was also observed but it does not cause severe loss to the crop.  相似文献

Polar lipids were extracted from the leaves of Passiflora species which varied in their resistance to chilling injury. The fatty acid compositions of the 8 major polar lipid classes from P. caerulea (chill-resistant) were generally similar to those of the corresponding lipids from P. flavicarpa (chill-sensitive). Using ESR spectroscopy, the motion of spin-labelled molecules was measured in phospholipids isolated from a range of Passiflora species. The temperature dependence of the motion of the spin labels showed a change at 1° for lipids of the most chill-resistant species and at 9° for the lipids of the most chill-sensitive species. Lipids from other species showed changes at intermediate temperatures, and the greater the chilling sensitivity of the species, the higher was the temperature of the change. It is concluded that pronounced differences in chilling sensitivity of the Passiflora species are correlated with physical differences in their membrane lipids; however, the degree of unsaturation of the lipids is not a reliable guide to chilling sensitivity.  相似文献

A protein inhibitor of phosphoinositol kinase has been detected in the later stages of ripening of mung bean seeds. This has been isolated and purified from the ungerminated seeds. It migrated as a single protein band when subjected to polyacrylamide gel electrophoresis. The MW of the inhibitor is approx. 86 000. The phosphoinositol kinase inhibition has been found to be dependent on the protein concentration of the purified inhibitor. It seems that 1 molecule of the inhibitor is necessary to inhibit 1 molecule of enzyme. The nature of the inhibition has been found to be non-competitive, the Ki of which is around 1·47 × 10?6 M. The enzyme inhibitor complex dissociates on gel electrophoresis without any loss of enzyme activity.  相似文献

Seed release mechanisms, seed production, seed weights, and fatty acid composition are investigated for Loasaceae subfam. Loasoideae. A total of ca. 60 species are studied. Nearly all species have erect, xerochasious capsules which restrict the release of the (balistochorous and/or anemochorous) seeds to dry, windy conditions. Very few taxa have tardily dehiscent capsules that are dispersed as a unit (anemochoria, epizoochoria, Blumenbachia sect. Blumenbachia, Klaprothia mentzelioides) or one-seeded cypselas (anemochoria, Kissenia). Dehiscent, but non-xerochasious capsules are only found in Blumenbachia, and Blumenbachia sylvestis (B. sect. Angulatae) has seeds with good floating ability and may be occasionally hydrochorous. The seeds of all other Loasoideae taxa tested rapidly sink when placed in water. Seed weights range from less than 0.02 g per 1000 seeds (Huidobria, Presliophytum) to over 23 g per 1000 seeds (Loasa ser. Macrospermae), thus spanning 3 orders of magnitude. Seed weight seems to correlate to some degree with substrate and seeds are heavier in species from “deep substrates” such as gravel and leaf litter. Seed numbers per fruit range from 1 (Kissenia) to over 3000 (Presliophytum) and annual seed production of individual mature plants ranges from less than 100 (Kissenia) to over 4 ×106 (Presliophytum). Fatty acid composition of Loasoideae seeds is mostly of common fatty acids, but exact composition varies considerably between groups. Most taxa have high levels of poly-unsaturated fatty acids (over 40% in Caiophora and Nasa), very low levels (<2%) are only found in Presliophytum. Nasa is the only genus of Loasaceae which has γ-linolenic acid and stearidonic acid in its seeds (at levels of 3.5–10% and 2–8.5% respectively). There is no obvious connection between the degree of unsaturation of the seed oils and habitat.  相似文献
