A protein inhibitor of phosphoinositol kinase has been detected in the later stages of ripening of mung bean seeds. This has been isolated and purified from the ungerminated seeds. It migrated as a single protein band when subjected to polyacrylamide gel electrophoresis. The MW of the inhibitor is approx. 86 000. The phosphoinositol kinase inhibition has been found to be dependent on the protein concentration of the purified inhibitor. It seems that 1 molecule of the inhibitor is necessary to inhibit 1 molecule of enzyme. The nature of the inhibition has been found to be non-competitive, the Ki of which is around 1·47 × 10?6 M. The enzyme inhibitor complex dissociates on gel electrophoresis without any loss of enzyme activity.  相似文献

High levels of ‘alkaline’ invertase activity occur in dormant mung bean seeds. During germination this activity decreases rapidly and is replaced by high ‘acid’ invertase activity. Cycloheximide prevented the formation of the latter activity and also inhibited germination. It is suggested that de novo synthesis of ‘acid’ invertase occurs during germination. Both enzymes bind to concanavalin A and, hence, are presumed to be glycoproteins. Affinity-purified enzyme samples show similar ratios of ‘acid’ and ‘alkaline’ invertase activities to the crude preparations indicating that specific enzyme inhibitors or activators are probably not involved in controlling the activities in vivo.  相似文献

用0.5ppm表油菜素内酯处理绿豆幼苗,显著促进上胚轴伸长生长,若切除真叶则可抑制表油菜素内酯诱导的效应。三碘苯甲酸(TIBA)也可抑制表油菜素内酯促进的伸长生长。外源IAA能部分恢复TIBA的抑制效应。经处理的上胚轴内源IAA含量明显高于对照。暗示表油菜素内酯可能通过对内源IAA的调节来促进绿豆上胚轴的伸长生长。 表油菜素内酯处理的绿豆上胚轴组织中,与生长素降解密切相关的IAA氧化酶以及过氧化物酶活性均明显低于对照。  相似文献

An aminopeptidase (EC 3.4.11.-) was purified to homogeneity, as judged by SDS-PAGE. from mung bean ( Vigna radiata ) cotyledons. The molecular mass of this peptidase was estimated as 75 kDa by gel filtration. When an oligopeptide consisting of 5 amino acid residues was used as substrate, amino acids were released in the order of the N-terminal sequence of the oligopeptide chain. This enzyme apparently requires free sulfhydryl for its activity, as judged by the effects of various proteinase inhibitors. Among aminoacyl- p -nitroanilides examined for the availability as substrates of the enzyme, p -nitroanilides with hydrophobic amino acids were preferred substrates. According to western immunoblot profiles, the enzyme level in cotyledons was high at the early stage of imbibition and declined rapidly after germination.  相似文献

Phosphoinositol kinase isolated and purified from germinating mung bean seeds has been further characterized. The rate of phosphorylation varies with different inositol phosphates and this is consistent with the Km and Vmax for each of the substrates. The phosphate transfer from ATP has been found to be mediated by a phosphoprotein intermediate. In a particular step of the reaction the immediate product of the reaction has been found to be most inhibitory, other products being less or non-inhibitory. The inhibition has been found to be competitive in nature. The Kis have been found to range between 0.6 and 1 × 10?4 M. ADP also inhibited non-competitively with respect to IP5. Ki for this has been found to be 2.3 × 10?4 M. The purified enzyme migrated as a single protein band on polyacrylamide gel electrophoresis. In the presence of sodium dodecyl sulphate it is dissociated into 3 subunits in the ratio 1 : 1 : 1. The MW of the three subunits are approx. 86 000, 56 000 and 35 000. The MW of the enzyme has been found to be approx. 177 000.  相似文献

The incorporation of radioactivity into sterols by transmethylation of methionine-[14C-methyl] was studied in mung bean hypocotyl sections. Young hypocotyl sections (1 cm) synthesized 4 times more radioactive sterols than older sections (5 cm). The transmethylation reactions may be rate limiting in older tissues. Wounding has only a quantitative effect on sterol biosynthesis, as seen by incorporation experiments with MVA-[2-14C]. Naphthalene acetic acid (NAA) stimulates sterol biosynthesis in both wounded surfaces and intact tissues of mung bean hypocotyl sections.  相似文献

Phosphoinositol kinase (adenosine triphosphate-inositolmonophosphate phospho—tranferase) has been isolated from cotyledons; about 300-fold purification has been achieved, with a recovery of 11%. The enzyme has a pH optimum at 7·4. It can mediate phosphorylation of lower inositol phosphates to their corresponding higher homologues, ATP being the phosphate donor. ATP can be replaced partially by UTP and PEP. The enzyme requires divalent cations for the reaction. Mn2+ has been found to be twice as effective as Mg2+, Ca2+ being inhibitory. Phosphoinositol kinase has been found to be different from inositol kinase.  相似文献

以离体油菜子叶为材料,研究了营养胁迫诱导的子叶衰老过程中吲哚乙酸氧化酶(IAA氧化酶)和细胞分裂素氧化酶活性的变化。在光照条件下,离体子叶在不含任何无机元素的0.8%的琼脂中培养9d后,出现明显的衰老迹象(叶绿素含量下降,丙二醛含量上升),15d时完全死亡。在营养胁迫诱导的衰老过程中,IAA氧化酶和细胞分裂素氧化酶的活性表现出相似的变化趋势,在诱导处理1d时,两种酶的活性均比处理前有明显下降,之后又随着衰老进程逐渐上升。IAA氧化酶活性在诱导处理11d时达到高峰,超出处理前30%以上;比对照高出1倍以上;而细胞分裂素氧化酶活性在诱导处理13d时达到高峰,比对照高出3倍以上,也超过了处理前的水平。衰老过程中IAA氢化酶和细朐分裂素氧化酶活性的上升可能是导致内源激素含量下降的重要原因。  相似文献

The activity of cysteine endopeptidase (EP) in the cotyledons of mung bean seeds increased with time after germination. When cotyledons were excised from the embryonic axis in the course of seedling growth, the activity of EP in the excised cotyledon markedly dropped during the following incubation of 1 d. However, the level of EP protein in excised cotyledons, as examined by immunoblotting, was similar to that in axis-attached cotyledons at the corresponding stage. Thus, it seems that the low activity of EP in excised cotyledons is not due to a decrease in the content of EP protein, but due to a loss of the activity of existing EP. Treatment of attached cotyledons with polyamines (PAs; putrescine and spermidine [Spd]) resulted in a decrease in EP activity, while the same PA-treatment brought about little alteration in the level of EP protein. This indicates that PAs somehow produce an inhibitory effect on the activity of EP. Axis-removal resulted in an accumulation of Spd in the cotyledon. The possibility is suggested that PA, especially Spd, is involved in the inhibition of EP activity in excised mung bean cotyledons.  相似文献

The presence of nine different glycosidases was demonstrated in the crude extract of mature mung bean seeds. N-Acetyl β-D-glucosaminidase, α-D-galactosidase and β-D-glucosidase were each resolved into two respective active forms by gel filtration. The other glycosidases showed single forms only. The apparent MWs of the glycosidases were determined. The glycosidases were absorbed to Con A-Sepharose column, with the exception of a small percentage of α-galactosidase and α-mannosidase which were eluted unretarded. The bound enzymes displayed varying affinities for the immobilized lectin, indicating differences in glycosylation. With the exception of β-galactosidase and invertase, all the glycosidase activities were detected in the protein bodies isolated from the seeds.  相似文献

Three new plant constituents were isolated from the primary leaves of Vigna radiata (= Phaseolus aureus) and their structures elucidated and characterized with the aid of negative-ion fast atom bombardment mass spectrometry (FAB MS), 1H NMR and UV spectroscopy, thin-layer, gas-liquid and high performance liquid chromatography. The new conjugates are (E)-p-coumaroyl-, (E)-caffeoyl- and (E)-feruloyltartronic acids. Their structures were unequivocally confirmed by comparison with synthetic material. The metabolism of the new hydroxycinnamic acid conjugates in young plants of Vigna radiata is described.  相似文献

研究了绿豆总黄酮的提取工艺。通过对固液比、乙醇体积分数、提取温度与提取时间的单因素实验确定水平点,设计4因素3水平实验,选用L9(3^4)正交表,优选绿豆总黄酮的最佳提取工艺为固液比1:50,φ(乙醇)为40%,提取温度70℃,提取时间120min。在此基础上得到绿豆皮中总黄酮的提取量为27.57mg/g,且5次平行的相对标准偏差为0.75%,总黄酮的平均回收率达97.5%。  相似文献

Several characteristics of mitochondrial respiration and energy status have been studied during growth and senescence of mung bean ( Phaseolus radiatus L.) cotyledons. The results showed that mitochondrial oxygen consumption, respiratory control, ADP:O ratios, and energy charge changed in the cotyledons during germination and growth of the seedlings. The respiration rate of intact cotyledons approximately reflected the trend of the oxidative activities of the isolated mitochondria. An increase was observed in both whole cotyledon respiration and mitochondrial oxygen uptake at the onset of senescence of mung bean cotyledons (day 3 after germination), which thereafter declined gradually. The capacity and activity of the alternative pathway increased markedly in mitochondria isolated from senescent cotyledons. After the onset of senescence, the mung bean cotyledon mitochondria exhibited a decrease both in the respiratory control ratios and ADP:O ratios, and the cotyledons exhibited a gradual decline in energy charge. All these results showed an irreversible deterioration of energy conservation in mung bean cotyledons. The role(s) of the alternative pathway in senescent mung bean cotyledons is discussed.  相似文献

The apparent absence of diamine oxidase in extracts of cotyledon of germinating groundnut seeds is due to the presence of a natural inhibitor. The inhibitor was associated with the lipid in the upper layer obtained on centrifugation of the cotyledon extract. It was non-dialysable, thermolabile and was inactivated by TCA and Triton X-100. The inhibitor was detected in the cotyledon extracts from early stages of seed development and was present up to 20 days after germination.  相似文献

The importance of the absolute and relative concentrations of monovalent and divalent cations and centrifugal speed (pressure) in the dissociation of mung bean 80S ribosomes has been examined. In the absence of Mg2+ ions, ribosome monomers yield 47S and 34S particles. Fixation with glutaraldehyde, however, indicates that this dissociation pattern is largely dependent upon high pressures developed during centrifugation and that in the absence of such artifacts the immediate product of Mg-free conditions is a 74S particle. Since 74S particles rapidly revert to the 80S form when Mg is replaced, this would appear to be a conformational change. Ribosomes were also dissociated in the presence of Mg2+ ions if the K+ ion concentration was raised. Three major particles were produced, 38S and 49S from the small ribosomal sub-unit and 60S from the large sub-unit. A proportion of the 80S monomer population is more resistant to dissociation. Experiments with puromycin indicate that the more resistant fraction probably represents ribosomes completed with nascent polypeptide resulting from polysome breakdown.  相似文献

Ribulose 1,5-bisphosphate carboxylase/oxygenase has been reported to occur in multiple forms in mung bean (Phaseolus aureus) using Sephadex G-200 chromatography. We have isolated this enzyme by identical methodology. The profile from Sephadex G-200 chromatography shows only one peak in contrast to the previous report and we find no evidence to corroborate the conclusions. Where Vc, Vo and Kc, Ko represent Vmax and Michaelis constants, respectively, the constant VcKo/VoKc for the single form is 70 at 40 μM CO2 and 1200 μM O2.  相似文献

Effects of Pb^2 on the growth of mung bean(Phaseolus radiatus L.)seedlings were conducted with an experiment.The results showed that the dry weight of roots increased by 13.01%,the dry weight of shoots decreased by 12.80%,the free proline and MDA content in leaves increased specially by 2.07% and 5.82%,the activity of POD decreased by 37.32% compared to control under 0.05mmol/L Pb^2 treatment.Under 0.5mmol/L Pb^2 treatment,the dry weights of roots and shoots decreased specially be 14.45% and 29.36%,the free proline and MDA content in leaves increased specially by 43.55% and 28.50,the activity of POD decreased by 59.37% compared to control.The results indicated that Pb^2 interfered the growth of mung bean seedlings by destroying cell membrane intergrity and decreasing the activity of POD in leaves.  相似文献

Heat shock increases chilling tolerance of mung bean hypocotyl tissue   总被引:5,自引:0,他引:5  
The effects of heat shock on the chilling tolerance of mung bean [Vigna radiata (L.) Wilczek] seedling tissue were studied by using two measurements of chilling injury: increased 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase activity and solute leakage. ACC oxidase activity (measured as ACC-induced ethylene production) of freshly excised mung bean hypocotyl segments was highly dependent on the temperature at which the seedlings were grown. However, this highly temperature-dependent level of ACC oxidase activity was probably a wound response since it was almost entirely eliminated by incubating the excised segments at 20°C for 3 h. In contrast, heating of excised segments to 40°C for up to 4 h resulted in a time-dependent increase in ACC oxidase activity which was sensitive to cycloheximide, indicating rapid protein synthesis during the heat treatment. ACC oxidase activity fell sharply during subsequent chilling at 2. 5°C. After 3 days of chilling, all treated segments, regardless of their initial ACC oxidase activity, showed a decline to the same low activity level and ACC oxidase activity continued to fall slowly for up to 9 days at 2. 5°C. Hypocotyl segments excised from seedlings held at 15°C showed no change in solute leakage, but leakage increased rapidly when seedlings were either chilled at 2. 5°C or heated to 32°C (just below the heat shock temperature). Chill-induced leakage from non-heat-shocked segments increased steadily with chilling duration and was unaffected by cycloheximide concentration up to day 6. Within the elevated rate of leakage on day 9, however, leakage was lower from segments exposed to 10 and 50 μM cycloheximide. Solute leakage was markedly reduced for up to 9 days when segments were heat shocked at 40°C for 3 or 4 h with or without 10 M cycloheximide, but the presence of 50 μM cycloheximide caused an initial doubling of solute leakage and a 3-fold increase after 3 days of chilling. Cycloheximide prevented formation of heat shock protection against chilling from the start at 50 μM and after 9 days at 10 μM. These results indicate that the protection afforded by heat shock against chilling damage is quantitative and probably involves protein synthesis.  相似文献
