The structure of afzelin (kaempferol 3-O-α-l-rhamnopyranoside) and paeonoside (kaempferol 3,7-bis-O-β-d-glucopyranoside) has been confirmed by total synthesis. Synthetic kaempferol 3-O-β-rutinoside had a mp of 190–192° suggesting that those natural kaempferol 3-O-rhamnoglucosides which melt in the same range are also 3-O-β-rutinosides.  相似文献

A new flavanone glycoside, naringenin-7-O-β-d-glucuronopyranoside, and a new flavonol glycoside, 6-hydroxykaempferol-7-O-β-d-glucuronopyranoside were isolated together with 12 known compounds, 5 flavone glycoside; hispidulin-7-O-β-d-glucuronopyranoside, apigenin-7-O-β-d-methylglucuronopyranoside, hispidulin-7-O-β-d-methylglucuronopyranoside, hispidulin-7-O-β-d-glucopyranoside, apigenin-7-O-β-d-glucopyranoside, a flavonol; kaempferol, two flavone; apigenin, and luteolin, a flavanone glycoside; eriodictyol-7-O-β-d-glucuronopyranoside, and three phenol glycoside; arbutin, salidroside, and 3,5-dihydroxyphenethyl alcohol-3-O-β-d-glucopyranoside from Centaurea urvillei subsp. urvillei. The structure elucidation of the new compounds was achieved by a combination of one- (1H and 13C) and two-dimensional NMR techniques (G-COSY, G-HMQC, and G-HMBC) and LC-ESI-MS. The isolated compounds were tested for their antiproteasomal activity. The results indicated that kaempferol, a well known and widely distributed flavonoid in the plant kingdom, was the most active antiproteasomal agent, followed by apigenin, eriodictyol-7-O-β-d-glucuronopyranoside, 3,5-dihydroxyphenethyl alcohol-3-O-β-d-glucopyranoside, and salidroside, respectively.  相似文献

Three covalent anthocyanin–flavonol complexes (pigments 1–3) were extracted from the violet-blue flower of Allium ‘Blue Perfume’ with 5% acetic acid-MeOH solution, in which pigment 1 was the dominant pigment. These three pigments are based on delphinidin 3-glucoside as their deacylanthocyanin and were acylated with malonyl kaempferol 3-sophoroside-7-glucosiduronic acid or malonyl-kaempferol 3-p-coumaroyl-tetraglycoside-7-glucosiduronic acid in addition to acylation with acetic acid.By spectroscopic and chemical methods, the structures of these three pigments 1–3 were determined to be: pigment 1, (6I-O-(delphinidin 3-O-(3I-O-(acetyl)-β-glucopyranosideI)))(2VI-O-(kaempferol 3-O-(2II-O-(3III-O-(β-glucopyranosylV)-β-glucopyranosylIII)-4II-O-(trans-p-coumaroyl)-6II-O-(β-glucopyranosylIV)-β-glucopyranosideII)-7-O-(β-glucosiduronic acidVI))) malonate; pigment 2, (6I-O-(delphinidin 3-O-(3I-O-(acetyl)-β-glucopyranosideI)))(2VI-O-(kaempferol 3-O-(2II-O-β-glucopyranosylIII)-β-glucopyranosideII)-7-O-(β-glucosiduronic acidVI))); and pigment 3, (6I-O-(delphinidin 3-O-(3I-O-(acetyl)-β-glucopyranosideI)))(2VI-O-(kaempferol 3-O-(2II-O-(3III-O-(β-glucopyranosylV)-β-glucopyranosylIII)-4II-O-(cis-p-coumaroyl)-6II-O-(β-glucopyranosylIV)-β-glucopyranosideII)-7-O-(β-glucosiduronic acidVI))) malonate.The structure of pigment 2 was analogous to that of a covalent anthocyanin–flavonol complex isolated from Allium schoenoprasum where delphinidin was observed in place of cyanidin. The three covalent anthocyanin–flavonol complexes (pigment 1–3) had a stable violet-blue color with three characteristic absorption maxima at 540, 547 and 618 nm in pH 5–6 buffer solution. From circular dichroism measurement of pigment 1 in the pH 6.0 buffer solution, cotton effects were observed at 533 (+), 604 (−) and 638 (−) nm. Based on these results, these covalent anthocyanin–flavonol complexes were presumed to maintain a stable intramolecular association between delphinidin and kaempferol units closely related to that observed between anthocyanin and hydroxycinnamic acid residues in polyacylated anthocyanins. Additionally, an acylated kaempferol glycoside (pigment 4) was isolated from the same flower extract, and its structure was determined to be kaempferol 3-O-sophoroside-7-O-(3-O-(malonyl)-β-glucopyranosiduronic acid).  相似文献

Three flavonoids were isolated from the aerial part of the wood horsetail (Equisetum silvaticum L.); two of them were found for the first time. The compounds were identified as kaempferol, kaempferol 3-O-β-D-galactopyranosyl-7-O-α-L-rhamnopyranoside and kaempferol 3-O-rutinosyl-7-O-L-rhamnopyranoside on the basis of the chemical transformations and IR, UV, 1H-NMR and mass spectra.  相似文献

Two novel delphinidin 3-(tri or di)-glycoside-7-glycosides were isolated from the violet–blue flowers of Vinca minor L. and V. major L. (Family: Apocynaceae), and determined to be delphinidin 3-O-[2-O-(β-xylopyranosyl)-6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-(2G-xylosylrobinobioside)-7-rhamnoside] as major floral anthocyanin of V. minor and delphinidin 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-robinobioside-7-rhamnoside] as major floral anthocyanin of V. major by chemical and spectroscopic methods. In addition, chlorogenic acid and kaempferol 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= kaempferol 3-robinobioside-7-rhamnoside (robinin)] were identified in these flowers. In this paper, the relation between the structure of floral anthocyanins and classification of Vinca species was discussed.  相似文献

Two new 5-methyl ether flavone glucosides (7,4′,5′-trihydroxy-5,3′-dimethoxyflavone 7-O-β-D-glucopyranoside and 7,4′-dihydroxy-5-methoxyflavone 7-O-β-D-glucopyranoside) were isolated from the leaves of Thai mangrove Bruguiera gymnorrhiza together with 7,3′,4′,5′-tetrahydroxy-5-methoxyflavone, 7,4′,5′-trihydroxy-5,3′-dimethoxyflavone, luteolin 5-methyl ether 7-O-β-D-glucopyranoside, 7,4′-dihydroxy-5,3′-dimethoxyflavone 7-O-β-D-glucopyranoside, quercetin 3-O-β-D-glucopyranoside, rutin, kaempferol 3-O-rutinoside, myricetin 3-O-rutinoside and an aryl-tetralin lignan rhamnoside. The structure of a lignan rhamnoside was found to be related to racemiside, an isolated compound from Cotoneaster racemiflora, and also discussed. Structure determinations were based on analyses of physical and spectroscopic data including 1D- and 2D-NMR.  相似文献

Catabolism of flavonol glucosides was investigated in plant cell suspension cultures using kaempferol 3-O-β-d-glucoside and kaempferol 7-O-β-d-glucoside labelled with 14C either in the glucose or in the flavonol moiety. Catabolic rates of glucosides were compared with those of free glucose and kaempferol. All substrates were degraded efficiently by cell cultures of mungbean, soybean, garbanzo bean and parsley. Based on 14CO2-formation, glucose from position 3 of kaempferol is 3–5 times more rapidly metabolized than that from position 7. The flavonol nucleus from both isomers is, however, oxidized to the same extent with a considerable portion of the flavonol being incorporated into insoluble polymeric cell material.  相似文献

The European dewberry (Rubus caesius) is a perennial shrub that is widely distributed in Europe but can also be found in North America. In folk medicine, the European dewberry is used to treat hyperglycaemia, diarrhoea and inflammation. LC-MS analysis of the European dewberry confirmed the presence of 35 compounds, mostly flavonoids, phenolic acids and derivatives of ellagic acid. Phytochemical analysis of R. caesius leaves led to the isolation of nine phenolics, namely: quercetin 3-O-β-D-rutinoside (1), kaempferol 3-O-β-D-glucuronide (2), quercetin 3-O-β-D-glucuronide (3), methyl brevifolincarboxylate (4), kaempferol 3-O-β-D-(6″-O-(E)-p-coumaroyl)-glucoside (5), kaempferol (6), quercetin (7), pedunculagin (8), and ellagic acid (9). Compounds 18 were isolated from this species for the first time. The chemophenetic significance was discussed.  相似文献

The dried fruits and seeds of Styphnolobium japonicum (L.) Schott (syn. Sophora japonica L.) are used in traditional Chinese medicine and known as Fructus Sophorae or Huai Jiao. The major flavonoids in these fruits and seeds were studied by LC-MS and other spectroscopic techniques to aid the chemical authentication of Fructus Sophorae. Among the flavonoids were two previously unreported kaempferol glycosides: kaempferol 3-O-β-glucopyranosyl(1 → 2)-β-galactopyranoside-7-O-α-rhamnopyranoside and kaempferol 3-O-β-xylopyranosyl(1 → 3)-α-rhamnopyranosyl(1 → 6)[β-glucopyranosyl(1 → 2)]-β-glucopyranoside, the structures of which were determined by NMR. Two further tetraglycosides were identified for the first time in S. japonicum as kaempferol 3-O-β-glucopyranosyl(1 → 2)[α-rhamnopyranosyl(1 → 6)]-β-glucopyranoside-7-O-α-rhamnopyranoside and kaempferol 3-O-β-glucopyranosyl(1 → 2)[α-rhamnopyranosyl(1 → 6)]-β-galactopyranoside-7-O-α-rhamnopyranoside; the latter was the main flavonoid in mature seeds. The chromatographic profiles of 27 recorded flavonoids were relatively consistent among fruits of similar ages collected from five trees of S. japonicum, and those of maturing unripe and ripe fruits were similar to a market sample of Fructus Sophorae, and thus provide useful markers for authentication of this herbal ingredient. The flower buds (Huai Mi) and flowers (Huai Hua) of S. japonicum (collectively Flos Sophorae) contained rutin as the main flavonoid and lacked the flavone glycosides that were present in flower buds and flowers of Sophora flavescens Ait., reported to be occasional substitutes for Flos Sophorae. The single major flavonoid in fruits of S. flavescens was determined as 3′-hydroxydaidzein.  相似文献

The ethyl ester of kaempferol 3-O-β-D-glucuronide, the methyl and ethyl esters of quercetin 3-O-β-D-glucuronide have been isolated from an aqueous acetone extract of the flowers of Tamarix nilotica. In addition kaempferol 3-O-sulphate-7,4′-dimethyl ether and the free aglycones were isolated. Chemical investigation of an acidic methanol extract of the whole plants of Datura metel resulted in the isolation of two new guainane sesquiterpenes, 1β,5α,7β-guaiane-4β,10α,11-triol (1) and 1α,5α,7α-11-guaiene-2α,3β,4α,10α,13-pentaol (2), along with eight known compounds: pterodontriol B (3), disciferitriol (4), scopolamine (5), kaempferol 3-O-β-d-glucosyl(1  2)-β-d-galactoside 7-O-β-d-glucoside (6), kaempferol 3-O-β-glucopyranosyl(1  2)-β-glucopyranoside-7-O-α-rhamnopyranoside (7), pinoresinol 4′′-O-β-d-glucopyranoside (8), (7R,8S,7′S,8′R)-4,9,4′,7′-tetrahydroxy-3,3′-dimethoxy-7,9′-epoxy-lignan-4-O-β-d-glucopyranoside (9), and (7S,8R,7′S,8′S)-4,9,4′,7′-tetrahydroxy-3,3′-dimethoxy-7,9′-epoxylignan-4-O-β-d-glucopyranoside (10). Their structures were elucidated by extensive spectroscopic methods, including 1D and 2D NMR and MS spectra. Compounds 2-4 and 6-10 were shown to have modest anti-inflammatory effects through inhibition of NO production in LPS-stimulated BV cells.  相似文献

Structural elucidation of kaempferol 3-O-β-d-xylopyranosyl(1  3)-(4-O-E-p-coumaroyl-α-l-rhamnopyranosyl(1  2))[β-d-glucopyranosyl(1  6)]-β-d-galactopyranoside-7-O-α-l-rhamnopyranoside (1) and kaempferol 3-O-β-d-xylopyranosyl(1  3)-(4-O-Z-p-coumaroyl-α-l-rhamnopyranosyl(1  2))[β-d-glucopyranosyl(1  6)]-β-d-galactopyranoside-7-O-α-l-rhamnopyranoside (2) was achieved using UV, NMR, and mass spectrometry, indicating the presence of trans or cis isomers of p-coumaric acid moiety in these novel structures. The antioxidant activity of the two compounds was assessed in the peroxynitrite assay.  相似文献

Two phenolic acids (1 and 2) and seven flavonoids (39) were isolated from the aerial parts of Alyssum alyssoides (Brassicaceae). All these compounds (19) were isolated from this particular species for the first time. Their structures were identified, on the basis of MS and NMR spectra as: p-hydroxy-benzoic acid (1), 3-methoxy-4-hydroxybenzoic acid (vanillic acid) (2), kaempferol 3-O-β-D-glucopyranoside (astragalin) (3), kaempferol 3-O-(6″-α-L-rhamnopyranosyl)-β-D-glucopyranoside (nicotiflorin) (4), quercetin 3-O-β-D-glucopyranoside (isoquercetin) (5), quercetin 3-O-β-D-galactopyranoside (hyperoside) (6), isorhamnetin 3-O-β-D-glucopyranoside (7), isorhamnetin 3-O-β-D-galactopyranoside (8) and isorhamnetin 3-O-(6″-α-L-rhamnopyranosyl)-β-D-glucopyranoside (narcissin) (9).
