The effect was studied of arginine-vasopressin (AVP) intraperitoneal administration in a doze of 0.001 mg/kg on elaboration of conditioned reaction (CR) of active avoidance in rats. Estimation of reaction dynamics by dynamic delta index allowed to increase the informational value of the experimental data with taking into account the direction of reaction shift and individual characteristic of the animal. AVP improved the elaboration of active avoidance CR, lowered variability of individual learning ability and did not influence the change of the number of intersignal reactions and short-latency escapes in the process of learning. It is suggested that the main site of AVP effect is the animal's reaction to conditioned sound stimulus.  相似文献

Previously the developmental switch to caspase-3 activation in the rat hippocampus has been shown during the third week of life. The goal of this study was to explore effects of caspase-3 inhibition during this period on learning in a two-way avoidance paradigm. On postnatal day 18, the pups were intracerebroventricularly administered with caspase-3 inhibitor Z-DEVD-FMK. Control groups were injected with either the control peptide Z-FA-FMK or saline. Caspase-3 inhibition, naturally activated in this critical period, was found to disturb the maturation of instrumental behavior. In particular, the young adult rats of Z-DEVD-FMK group displayed less effective elaboration of escape and active avoidance reactions in two-way avoidance paradigm, accompanied with a decrease in inter-trial crossings. However, associative components of the learning did not change after caspase-3 inhibition. Conditioned emotional behavior was, in general, similar in all groups, and the number of responses related to conditioned stimulus exploration did not differ in Z-DEVD-FMK and Z-FA-FMK groups. In spite of the deficit in active avoidance conditioning in Z-DEVD-FMK group, a significant increase in incomplete or preparatory reactions to conditioned stimulus was demonstrated suggesting that the association between predictive conditioned stimulus and possibility of crossing can be elaborated. The change of exploratory behavior is unlikely to be specific for caspase-3 inhibition, being similar in Z-DEVD-FMK and Z-FA-FMK groups.  相似文献

The experiments on rats have shown changes in the learning ability of animals. The learning of locomotor reactions of escape and avoidance conditioned reflex was accelerated in animals after pinealectomy. Training of rats in U-shaped and 16-door mazes was significantly affected after pinealectomy. The index of reflex retention (1 and 7 days later) was not changed in the operated animals. Epithalamin (0.5 mg/kg for 5 days) selectively facilitated learning of visual and space discrimination.  相似文献

Dopaminergic system and its D1 as well as D2 receptors are involved in the modulation of emotional behavior. This experiment investigated the role of dopaminergic activity in the inescapable stress-induced learned helplessness, a widely used depression animal model, by using the pharmacological manipulation through the apomorphine (APO), an agonist for D1 and D2 receptors, and sulpiride (SUL), a selective D2 antagonist. Male Sprague Dawley rats were used and tested in a shuttle box. In the day-1 session, the rats received a 10-trial (1 min/trial) inescapable stressor: a 3 sec conditioned stimulus (CS; 75 db sound and 250 lux red light) followed by a 10 sec unconditioned stimulus (UCS; electrical foot shock, 0.5 mA). In the day-2 session, a 15-trial active avoidance test, 3 sec CS followed by UCS, was performed 30 min after the administration of APO (0, 0.05, 0.5, 1, and 5 mg/kg, i.p.). The number of failures was counted and the UCS was stopped when the rats did not escape after 15 sec UCS. The results showed that APO at the dosage of 0.5 mg/kg had a tendency to enhance the avoidance behavior. In contrast, the treatment of higher dose of APO, 1 and 5 mg/kg, reduced the number of escape but increased the number of failure. Pretreatment of SUL (5 mg/kg, i.p.), 10 min before 1 mg/kg of APO, significantly enhanced the failure behavior. The present data suggest that the activity of D2 receptor may be associated with the adaptive or protective role in the prevention of escape deficits after exposure to inescapable stress. However, the excessive stimulation of D1 receptor may participate in the failure of coping behavior leading to learned helplessness and therefore in the pathophysiological mechanisms underling the development of depression.  相似文献

The extirpation of the foreleg area in the somatosensory cortex (SI) in dogs was performed in order to study the role of this area in motor control of the corresponding limb. Stable precise avoidance and escape reactions were established before the cortical lesion. Avoidance reactions disappeared during the first week after the surgery but then they were gradually restored without retraining. At the third weak there was a practically complete recovery of the precision of movements, the ability to fix the leg position and of the duration of successive phases of the reactions. The conclusion was drawn that the initial disturbances of avoidance reactions were most likely the result of diaschisis (shock). The initial disturbances of visually guided instrumental movements and escape reactions were much weaker than those of avoidance reactions (lifting of the foreleg in response to conditioned signal).  相似文献

Microinjection of dopamine and serotonin into the dorso-medial portions of the amygdalic complex of rats increased the latent period of conditioned defense and motor food reactions, spontaneous motor activity, the number of jerks and the value of the conditioned food reflex. Noradrenaline failed to influence the recorded indices of the conditioned food-procuring reflex, but facilitated the conditioned reaction of avoidance, significantly shortening its latent time. The results obtained pointed to the specificity of the neurochemical mechanisms realizing at the level of the amygdalic complex conditioned reflexes of various biological modality in rats.  相似文献

Lengthening the time interval between the conditioned stimulus and the unconditioned stimulus increases the number of active avoidance conditioned responses in subjects that have been trained to a stable level of performance in many previous conditioning sessions. In the present research, rats chosen from a population specially selected for low rates of avoidance conditioning have been used. In addition to this characteristic, subjects were chosen for the exhibition of an apparent absence of retention from one day to another. The dependency of the number of conditioned responses on the time interval between conditioned stimulus and unconditioned stimulus may lead to wrong evaluation of the subjects' conditioning level. In fact, the level of conditioning may be attributed to either learning or memory processes when in many cases it is determined only by the latency time of the conditioned response. The conditioned response has no possibility of manifesting itself when its latency time exceeds in length the time interval between conditioned stimulus and unconditioned stimulus.  相似文献

The dexamethasone suppression test (DST) was applied to male Wistar rats with different models of depression: group with the learned helplessness, group with informational neurosis provided by time-deficit conditioned avoidance training, as well as groups of rats of two strains selected for low (KLA)--and high (KHA) avoidance learning. The pre-dexamethasone basal and stress-induced corticosterone levels were similar in intact rats and those exposed to inescapable shock. The dexamethasone administration (5 mkg/kg) failed to decrease the serum corticosterone level in rats with learned helplessness. The informational neurosis increased significantly the basal corticosterone level and decreased the stress response. Serum corticosterone levels were similar in KLA and KHA rats. These results give evidence that two stress-induced rat models of depression with similar behavioural disturbances (reduction of escape/avoidance reactions) exhibit marked differences in the activity of hypothalamo-pituitary-adrenal (HPA) axis.  相似文献

A "preparatory" hypothesis according to which the bar-holding develops as a preparatory response placing an animal in an advantageous position for the reaction on the subsequent action of the electric current has been experimentally examined. Comparing the avoidance training under the condition allowing the bar-holding until the next conditioned and unconditioned stimuli, and under conditions excepting such long holding it has been established that in first case the number of bar-holding responses is more and the avoidance responses are less than in the second one. It contradicts the "preparatory" hypothesis. It has been shown that the dynamics of inter-trail and the bar-holding responses results from stimulus generalization and discrimination.  相似文献

In order to determine the optimun conditions suitable for a number of trials and the intensity of unconditioned stimulant (US) in the two-way shuttle-box avoidance test in Sprague-Dawley strain rats, which are used most frequently in reproduction studies, conditioned avoidance response was observed under various conditions for 30 and 60 trials and the low and high US levels. Investigation was also conducted in Wistar rats under a high US level with 30 and 60 trials. Latency time of the escape response in Sprague-Dawley rats was shortened with increasing trials. Body weight gains of both strains of rats in the high US level with the 60-trial group decreased during the observation period. These findings suggest that the high US level with the 60-trial group is not suitable for the two-way shuttle-box avoidance test. The rate and latency time of the avoidance response were lower in Wistar rats than in Sprague-Dawley rats, although those of the escape response were higher. Significant changes in the following were observed, mainly from first to third sessions: the avoidance rate of all groups in strains of rats, escape rate of 60-trial group in Sprague-Dawley rats, avoidance and escape latency time of the 60-trial groups in both strains of rats and escape latency time of the 30-trial group in Sprague-Dawley strain rats.  相似文献

Fear conditioning, escape and active avoidance reactions in two-way avoidance paradigm were compared in rats of different ages. Fear conditioning, but not escape and active avoidance reactions could be acquired on the 16-17th postnatal days, and the acquisition was more effective than in adults. Escape behavior matured beginning from the 18th postnatal day reaching the adult level within the 3d-4th postnatal weeks. Maturation of the mechanisms of Pavlovian (fear reaction) and instrumental (escape reaction) conditioning did not facilitate the acquisition of two-way avoidance until the 4th postnatal week, young animals displayed low acquisition in this period. The maturation of these memory processes is proposed to be related to developmental stages of different mechanisms of hippocampal plasticity.  相似文献

Levels of c-fos mRNA expression in mouse cerebral cortex and hippocampus at different stages of footshock escape and avoidance learning were studied by Northern hydridization. In the first series of experiments a mouse was presented with 30 electric footshock daily in a chamber where it could escape from the floor by jumping on the safe platform attached to the wall. A large increase in c-fos mRNA level in the cerebral cortex and hippocampus was observed during the first day of training. Mice that were trained for 9 consecutive days and acquired a footshock escape reaction showed no elevation of c-fos expression in the brain as compared to the quiet control group. In the second series of experiments the levels of c-fos expression were compared in individual mice trained to avoid the footshock by jumping on the platform in response to an auditory conditioned stimulus. Mice which acquired avoidance behavior more rapidly had lower c-fos mRNA levels than slow learners. There was no such to difference between the corresponding yoked control groups which consisted of animals matched the rapid and slow learners by the number of footshocks received. It is concluded that achievement of adaptive results in the course of learning leads to a suppression of further c-fos induction by motivational excitation.  相似文献

The paper deals with the character of changes in the EP form recorded from the visual cortex of a rat during elaboration of a system of positive and negative (with different degree of precision) conditioned defensive reflexes. It has been shown that in a state of a consolidated habit of pain stimulus avoidance in a shuttle box a negative component with peak latency up to 70 msec develops in EP to the differentiation stimulus of 0.5 c/s. This component is much less pronounced to the 2 c/s conditioned stimulus. When a more precise differentiation stimulus (1 c/s) was presented to the animal in this state the development of the studied component depended on the subsequent behavioural reaction: it developed if the animal reacted in the same way as to the differentiation stimulus. This fact suggests that the generation of this component is connected with the inhibitory processes developing at the time when the animal is choosing its behavioural reaction.  相似文献

Short-term (90 s) effects of the complex acoustic signal (CAS) with ultrasonic components on the development of defensive conditioned reflex of two-way active avoidance in a shuttle-box were studied in female Wistar rats. The learning ability of rats was measured on a scale designed in our laboratory. It was shown that CAS stimulation triggered an audiogenic seizure of different strength in 59% of animals. The CAS was extremely stressful for Wistar rats: it prevented the active avoidance learning in early terms after its application (the first training session in 4 or 6 days). This effect did not depend on the presence or intensity of audiogenic seizures during CAS. In the second training session in 9 days (the first session was in 4 days), learning was impaired as compared to control without CAS. However, during repeated training procedure 1.5 months after the CAS (the first session in 6 days), rats rapidly reached the criterion of learning (10 consecutive avoidance reactions). On the other hand, if the CAS was presented with different time lags (immediately, in 3 or in 45 days) after the first training session, the ability of animals to learn during the second session was not impaired both in early and late terms after exposure to the stressor. The results suggest that exposure to CAS prevents development of short-term memory but does not affect consolidation process and long-term memory.  相似文献

Decrease of serotonin concentration in the septo-hippocampal region of Wistar male rats was obtained by introduction of 5,7-dihydroxytryptamine in the raphe median nucleus. In seven days, after 20 pre-expositions of the conditioned stimulus (presentation of the experimental chamber) conditioned reaction of passive avoidance was elaborated. In sham-operated control the pre-exposition of the conditioned stimulus caused latent inhibition, determined by several parameters: low level of conditioned reaction reproduction, its prolonged preservation at the formed level and nonsubjection to amnesia. Introduction of haloperidol in a dose of 0.5 mg/kg one hour before learning restored the state of latent inhibition disturbed by switching off of the mesolimbic serotonin system. It is supposed that in the state of pre-exposition, the decrease of attention to nonreinforced stimulus takes place because of the intensification of inhibitory influence of the serotoninergic system and reciprocal decrease of dopaminergic system activity.  相似文献

After 10-15 food stimuli paired with electrical shock in semi-intact snail preparation, responses to strong tactile stimuli identified feeding behaviour neurones were studied. Inhibition evoked by tactile stimulation in these cells before learning procedure disappeared and in some cases noxious stimulus evoked synaptic activation corresponding to feeding reactions in the intact animal. Changes in second-order sensory neurones pre-synaptic to the command neurones of avoidance behaviour are suggested to be the mechanism of forward conditioned connection as well as the mechanism of backward conditioned connection.  相似文献

Dogs of 1.5 and 7 months and rats of 15, 20, 30, 45, 60, 80, and 100 days of age were given acquisition training in an escape task. The previous observations suggested that difficulties in defensive conditioning typically found in young animals were connected with their generalized perception of Igarning conditions. To facilitate the discrimination of the environmental factors a conditioned signal was not applied and the home cage was used as a "safe" compartment. The intertrial intervals were increased up to 2 hours for dogs and 24 hours for rats. Under such conditions an acquisition of escape reaction was most rapid in animals of early age. The period of effective learning (exaltation) in rats began from 20 and lasted to 60 days of age, the maximal efficiency being observed at 20 days. Pups of 1.5 months had learned more quickly than those of 7 months of age. Variations of learning efficiency in ontogeny of rats were positively correlated with changes of correct escape reaction search activity. Phenomenon of learning exaltation in ontogeny was supposed to be connected with the high level of activity of perception and association cerebral mechanisms being the result of immaturity of inhibitory structures.  相似文献

Sex-related peculiarities of dynamics of brain sex steroids in the process of learning and extinction of the conditioned reflex of passive avoidance have been studied in model experiment. Prior to learning of the conditioned reflex, female rats were found to be distinguished by manifestation of anxiety and fear as compared with male rats. At formation of the conditioned reflex, no significant sex-related differences were detected between males and females, whereas extinction of the conditioned reaction of passive avoidance in males occurred by 2–3 days faster than in females. At learning of conditioned reaction of passive avoidance, in sexually mature male rats there was revealed an increase of the testosterone content in various brain structures, especially in hippocampus and frontal cortex, while its level in blood plasma remained unchanged. Also shown was an elevation of estradiol concentration in female amygdale, whereas at extinction of the conditioned reaction of passive avoidance, a rise of estradiol values was noted in hippocampus and cingular cortex. At the same time, the testosterone level in blood plasma did not change, whereas after extinction of the conditioned reflex the estradiol concentration decreased statistically significantly. Different dynamics of changes of the sex steroid levels in brain and blood plasma can indicate a possibility of their formation in the nervous tissue. The performed correlation analysis confirms the concept of selective involvement of testosterone and estradiol of individual brain structures in realization of processes of learning and memory in sexually mature male and female rats.  相似文献

In rats aged 2-8 weeks cortical EP to CS (20 flash - tone combinations, 0,9/sec, reinforced since the 10th application by electric shocks to the hind leg) were studied within different kinds of behavioral responses during avoidance learning and extinguishing. In contrast to our results in freely moving rats no developmental trend was found in this kind of avoidance (lifting of the hind leg). Average EP within reinforced trials (with escape or no reactions) differed in isolated application of CS from those when both CS and US were acting together. In younger animals the EP to CS combined with US were characterized by an evident late negative wave which shifted later (5-6 weeks toward the early negative complex. The EP changes in the auditory cortex were more pronounced, whereas visual EP with CS-US combination were rather decreased. In the youngest animals (2 weeks) the auditory EP within trials with avoidance were characterized by a distinct short latency deflection of the first positive wave, whereas in EP to extinguished CS the second deflection of the first positive wave prevailed. Also in these phenomena, the typical changes were clearly revealed in the auditory cortex. At later developmental stages (starting the 3rd, more prominently the 4th and 5th week) the wave following primary positive - negative complex was shifted toward the negativity if the animal responded by an avoidance; on the contrary an ample positive, often a double-peak wave arose if the response was extinguished. The stimulus and reaction dependence in the cortical EP showed the role of not yet fully mature cerebral cortex in avoidance learning. Both, fast as well as with some delay running processes participated in the observed phenomena during the ontogenetical development.  相似文献

Excitability of the motor cortex (projection of the foreleg), judged by the threshold of flexion reaction to its stimulation during elaboration of conditioned transswitching (CT), was studied in dogs. CT was elaborated for instrumental reflexes of escape (1-st CT variant) and avoidance (2-nd CT variant). In the 1-st variant the dog escaped from the hydrochloric acid solution, if during its administration it produced an active flexion of the foreleg in situation A and an active extension of the same leg in situation B, with a permanent action of a switch (a fan). In the 2-nd CT variant, the dog avoided the acid, if in situation A, in response to a tone of 250 Hz, it actively lifted its foreleg, and if in situation B, it actively resisted a passive lift in response to the same stimulus. In escape CT as well as in avoidance CT, there was a statistically significant difference between the thresholds of flexion reaction. The threshold was lower in situation of instrumental flexion reflex formation in comparison to the situation of extension reflex formation.  相似文献
