The ultrastructure of whole X-Y pairs has been reconstructed by serial sectioning and model building. Seven X-Y pairs were completely reconstructed and the lengths of the cores of the sex chromosomes were measured. These X-Y pairs corresponded to zygonema, early, middle and late pachynema. Special regions of the X-Y pair were reconstructed from thinner sections. — It has been shown that two cores exist in the sex pair during the cited stages, and that their lengths and morphology are rather constant in specific stages. The long core averages 8.9 in length and the short core is 3.5 long. Both cores have a common end region in which a synaptonemal complex is formed from zygonema up to midpachynema. This synaptonemal complex shortens progressively up to mid-pachynema and at late pachynema becomes obliterated. Each core has a free end touching the nuclear membrane. During mid-pachynema an anomalous synaptonemal complex is developed on most of the length of the long core. This complex is asymmetric and disappears at late pachynema. The meaning of the cores and the complexes are discussed, and the existence of a homologous region in the X-Y pair of the mouse is interpreted to be proved. 相似文献

In the pygmy mouse, Mus dunni, the entire Y chromosome and the short arm of the X and distal region of its long arm are constitutively heterochromatic. Different banding studies on somatic chromosomes revealed the GC nature of the distally located heterochromatin of the long arms of both the X and Y chromosomes. The short arm of the X and the rest of the Y are AT-rich. During meiosis, the long arms of the X and Y paired extensively, sometimes more than half of the Y pairing with the X. This observation is in disagreement with that of Pathak and Hsu (1976) who reported end-to-end pairing between the long arm of the X and the short arm of the Y. The orientation observed by us is favourable to a successful meiotic recombination but whether this takes place remains to be demonstrated. 相似文献

Having compared the microanatomy of the toes of a terrestrial to two climbing species, adaptations were found in the flexor tendons and in the integument. In contrast to Crocidura russula, both Muscardinus avellanarius and Micromys minutus have a tendon-locking mechanism (TLM) that is engaged when the middle phalanx is bent. A ventral thickening of the flexor tendon is situated deep to a thickened portion of the ventral tendon sheath. When twigs or stalks are grasped, the TLM allows less muscular energy to be expended. In C. russula glands are restricted to the terminal pads, but in the climbing species they occur in the sole of the toes as well. In the reed-living M. minutus knob-shaped integumental thickenings, together with the digital pads, stabilize the grip. In contrast the arboreal M. avellanarius often climbs thick branches and shows adaptations for pressing the sole of the feet against the surface. Thereby the tendon attached to the plantar integument of the toes transfers the muscle force directly to the bark. Unlike the other digits on the forefeet of both climbing species, no TLM is present in the anterior digit. In M. minutus this short digit is twisted towards the palm and, with the carpal pads, provides an abutment against the grasping fingers. In M. avellanarius the anterior digit has very thin tendons and is that much reduced in length that it is completely integrated into the digital pad where it acts, at best, as a lateral support of the pad. © 1996 Wiley-Liss, Inc. 相似文献

Summary Studies of spermatogenesis in an XYY male, presenting at a subfertility clinic, confirm the tendency for the germ cells to lose the second Y chromosome but for some XYY cells to reach metaphase I (MI). Light microscope studies of MI revealed the presence of YY bivalents and EM studies of microspread, silver-stained pachytene stages showed 30% of the cells to have two Y chromosomes; 13 out of 16 of these showing a YY synaptonemal complex. Strikingly, the Y axes show only partial synapsis; in no case was synapsis of the long arm heterochromatic regions apparent. 相似文献

Data are presented on the breeding biology of Micromys minutus in Britain. It is shown that although litters are produced over a protracted period (May-December), 74% of the litters are born in August and September. Mean litter size is 5·40±0·16( n = 62), which is a significant reduction on the mean litter size of 6·75±0·40( n = 16) calculated from pre-1917 literature. Litter size was found to be constant throughout the breeding season, and there was no difference in mean litter size between two samples collected close to the northern and southern limits of the known British range. By comparing the mean litter size of samples 1–5, 6–10 and 11+ days old, no significant loss of individual mice within litters could be detected, although 12·3% of all litters died prior to weaning. This loss was predominantly of autumnal litters, cold and wet being important climatic factors that terminate the breeding season. Comparisons are made with data from other parts of the animal's range. 相似文献

Synaptic behaviour and the progression of morphological differentiation of the XY chromosome pair during pachytene was studied for the first time in three species of the South American subterranean rodents of the genus Ctenomys (tuco-tucos). In general, synapsis progression in the sex pair could be subdivided into four substages: (i) initial partial synapsis of the X and Y chromosome axes and beginning of the differentiation of the unsynapsed regions; (ii) complete or almost complete synapsis of the Y axis accompanied with morphological differentiation of the unsynapsed region of the X chromosome; (iii) a novel stage exclusive to Ctenomys perrensi consisting in a retraction of the free X axis, associated with the formation of a homogeneous and dense structure along the synaptic region, which leads to the achievement of full synapsis between sex chromosomes; or (iv) an increase in morphological complexity involving extreme splitting of the XY pair. The implications of the peculiar synaptic behaviour displayed by sex chromosomes in C. perrensi, a species complex highly polymorphic for Robertsonian translocations, are discussed in relation to both the triggering of the pachytene checkpoint and the avoidance of non-homologous associations between sex chromosomes and the asynaptic pericentromeric regions of trivalents in translocation heterozygotes. 相似文献

Summary In this paper observations are summarized and speculations discussed, and it is suggested that some loci on the distal short arm of the X chromosome (Xp) are not randomly inactivated in the female, because they are within the proximal part of the pairing segment between Xp and Yp. This peculiarity of gene expression may be a remnant of the evolutionary history of the sex chromosomes, the pairing segment of which may involve at least 27% of Xp and 95% of Yp. Crossing over seems to occur mostly in the terminal third of the X/Y pairing segment. However, crossing-over inhibition control may lapse, or may be on the X and Y (e.g. Xg, H-Y, STS, and perhaps others) might cross over with a variable frequency which is proportional to their distances from the telomeres of the short arms. It is postulated that the DNA of the pairing segment is composed in a way which may also permit unequal crossing over to occur between the X and the Y, thereby giving rise to exceptions to X-or Y-linked inheritance. The peculiarities of behaviour and the position of other loci on the sex chromosomes are also discussed briefly. 相似文献

Harvest Mouse (Micromys minutus) has a very wide range of distribution in Asia and Europe. However, the phylogenetic relationship of M. minutus is still uncertain. In this study, we determined the complete mitochondrial (mt) genome sequences of M. minutus, and used the complete mitochondrial genome sequences constructed the phylogenetic tree of Muroidea. The size of the genome is 16,232 bp in length and has a base composition of 33.6% A, 29.1% T, 24.8% C, and 12.5% G. The mitogenome structure was similar to that of typical vertebrate and other rodents' mitochondrial genomes, includes 13 protein-coding genes, 2 rRNA genes (12S rRNA and 16S rRNA), 22 tRNA genes, and 1 control region. We suggested a new initiation codon for ND5 (NADH dehydrogenase subunit), which has been never reported in the mitochondrial genome of vertebrate. The ML and BI phylogenetic trees, which based on the combination of the 12 protein-coding genes, supported strongly that the genus Micromys was represent an early offshoot within the Muridae with high support values (BI = 1.00, ML = 100). 相似文献

Organization of B chromosomes in the Korean field mouse Apodemus peninsulae was analyzed. We painted its metaphase chromosomes with whole and partial chromosome paints generated by microdissection and DOP-PCR. The results of the painting indicated that all B chromosomes contained a large amount of repeated DNA sequences. The repeats could be classified in terms of their homology and predominant location. Pericentromeric repeats of B chromosomes were present in many copies in pericentromeric C-blocks of all autosomes and in non-centromeric C-blocks of the sex chromosomes. B arm specific type 1 repeats comprised the main body of the arms of almost all B chromosomes and were present in the arms of A chromosomes as interspersed sequences. B arm-specific type 2 repeats were found at the ends of some B chromosomes that did not undergo compaction at the interphase- metaphase transition and remained uncondensed. On the basis of comparative analysis of localization of B chromosome repeats in the chromosomes of two related species, A. peninsulae and A. agrarius, we suggest a hypothesis of B chromosome origin and evolution in the genus Apodemus. 相似文献

Recombination between the X and Y chromosomes is limited to the pseudoautosomal region and is necessary for proper segregation of the sex chromosomes during spermatogenesis. Failure of the sex chromosomes to disjoin properly during meiosis can result in individuals with a 47,XXY constitution, and approximately one-half of these result from paternal nondisjunction at meiosis I. Analysis of individuals with paternally derived 47,XXY has shown that the majority are the result of meiosis in which the X and Y chromosomes have failed to recombine. Our studies of sperm have demonstrated that aneuploid 24,XY sperm have a decreased recombination frequency, compared with that of normal sperm. Some studies have indicated a relationship of increased paternal age with 47,XXY offspring and with the production of XY disomic sperm, whereas others have failed to find such relationships. To determine whether there is a relationship between paternal age and recombination in the pseudoautosomal region, single-sperm genotyping was performed to measure the frequency of recombination between a sex-specific locus, STS/STS pseudogene, and a pseudoautosomal locus, DXYS15, in younger men (age < or =30 years) compared with older men (age > or =50 years). A total of 2,329 sperm cells were typed by single-sperm PCR in 20 men who were heterozygous for the DXYS15 locus (1,014 sperm from 10 younger men and 1,315 sperm from 10 older men). The mean recombination frequency was 39.2% in the younger men and 37.8% in the older men. There was no heterogeneity in the frequency of recombination rates. There was no significant difference between the recombination frequencies among the younger men and those among the older men, when analyzed by the clustered binomial Z test (Z=.69, P=.49). This result suggests that paternal age has no effect on the recombination frequency in the pseudoautosomal region. 相似文献

Surface-spread, silver-stained primary spermatocytes from individuals of the Sitka deer mouse (Peromyscus sitkensis) were analyzed by electron microscopy. Pairing of the X and Y chromosomes is initiated at early pachynema and is complete by mid pachynema. The pattern of sex chromosome pairing is unique in that it is initiated at an interstitial position, with subsequent synapsis proceeding in a unidirectional fashion towards the telomeres of the homologous segments. One-third the length of the X and two-thirds the length of the Y are involved in the synaptonemal complex of the sex bivalent. Various morphological complexities develop in the heteropycnotic (unpaired) segments as pachynema progresses, but desynapsis is not initiated until diplonema. Analysis of C-banded diakinetic nuclei indicated that sex chromosome pairing involves the heterochromatic short arm of the X and the long arm of the heterochromatic Y. An interstitial chiasma between the X and Y was observed in the majority of the diakinetic nuclei. The observation of a substantial pairing region and chiasma formation between the sex chromosomes of these deer mice is interpreted as indicating homology between the short arm of the X and the long arm of the Y. 相似文献

Individuals in a colony of European harvest mice (Micromys minutus) were diagnosed with a variety of skin tumors including papillomas, trichoepitheliomas, and sebaceous carcinomas. Papillomavirus group-specific antigens and viruslike particles were detected in the papillomas. A 7.6-kilobase supercoiled circular DNA, which was cleaved once by EcoRI, was visualized in papilloma extracts by low-stringency Southern blot hybridization with a bovine papillomavirus type 2 probe. The molecule was cloned in pUC18, and a restriction map was generated. The molecule was shown to be colinear with the genome of human papillomavirus type 1a by partial sequence analysis. The DNA hybridized to human papillomavirus type 1a, rabbit oral papillomavirus, and the genome of Mastomys natalensis papillomavirus at Tm - 33 degrees C but not to the DNAs of 13 other papillomaviruses. Transformation of NIH 3T3 or C127I cells by tail papilloma extracts or transfected viral DNA was not observed. All 17 tumors examined contained large amounts of viral DNA in a supercoiled, unintegrated form as revealed by Southern blot hybridization. Furthermore, many extracts (25 of 35) from normal organs and skin of individuals with lesions elsewhere on their bodies contained viral DNA. This paper reviews the past known history of the Harvest mouse and presents the results of a survey of the distribution and status of the animal in Britain. Data are included which demonstrate the wide variety of habitats colonized by the Harvest mouse and the diversity of monocotyledonous plants that can be utilized for nest building. The survey has shown that there is no evidence of a decline in the distribution or abundance of this animal in Britain during the last century, and that the sparsity of earlier records was probably due to the lack of competent recorders. 相似文献

Conventional and microspread preparations of Microtus cabrerae spermatocytes were made to investigate the chromosomes of this species. Three different types of Y chromosomes, varying in size of the heterochromatic block, were observed; they were alike, however, in regard to synapsis, which was consistently absent. Our results suggest that the heterochromatic blocks are not involved in the lack of synapsis and that asynapsis is a cytological feature common to all species of the family Microtidae. In addition, the co-aligned configuration of the ends of the sex-chromosome axes of this species and the lack of silver-stainable threads or filaments connecting them suggest the existence of two mechanisms for association of the sex chromosomes during prophase I and metaphase I: attachment of the ends of both sex chromosome axes to the nuclear envelope and heterochromatin "stickiness." 相似文献

The harvest mouse (Micromys minutus) occurs throughout most of continental Europe. There are also two isolated and recently discovered populations on the Scandinavian peninsula, in Sweden and Norway. Here, we investigate the origin of these populations through analyses of mitochondrial DNA. We found that the two populations on the Scandinavian peninsula have different mtDNA haplotypes. A comparison of our haplotypes to published sequences from most of Europe showed that all Swedish and Norwegian haplotypes are most closely related to the haplotypes in harvest mice from Denmark. Hence, the two populations seem to represent independent colonisations but originate from the same geographical area. We discuss the age of the Swedish and Norwegian populations and suggest that both have been introduced recently. 相似文献

The replication pattern of the X and Y chromosomes at the beginning of the synthetic phase was studied in human lymphocyte cultures partially synchronized by the addition of 5-fluoro-2-deoxyuridine (FUdR). The data were evaluated statistically by an analysis of the distribution of silver grain counts over the X and Y chromosomes. —In cells from normal females, one of the X chromosomes began replication later than any other chromosomes of the complement. The short arm of the late replicating X chromosome started replication earlier than the long arm. The telomeric region of the short arm was a preferential site of DNA synthesis at the beginning of replication. —In partially synchronized lymphocyte cultures from a patient with the XXY syndrome, the Y chromosome started replication together with the late replicating X chromosome. The Y chromosome most frequently replicated synchronously with the short arm of the X. The centromeric region of the Y chromosome initiated synthesis before the telomeric region and appeared to replicate synchronously with the telomeric region of the short arm of the X. The mitotic and meiotic chromosomes of four male East-Asiatic mice, Apodemus peninsulae, having three to seven chromosomes in addition to the standard karyotype (2n = 48), were investigated. B-chromosomes were represented by medium-sized metacentric and dotlike chromosomes. Mosaicism of bone marrow cells due to a numerical variation of accessory chromosomes was established for the males examined. Capacity of B-chromosomes to form axial elements and synaptonemal complexes in meiotic prophase I was revealed by electron microscopy. The occurrence of univalents of different morphology, bivalents, and multivalents, corresponding to B-chromosomes, was demonstrated. An increase in the number of B-chromosomes was found in spermatocytes at zygotene-pachytene relative to the number in bone marrow cells, which may be evidence of B-chromosome accumulation in the germ cell line of the East-Asiatic mouse. 相似文献
