目的探讨EFS和DAP两种玻璃化冷冻方法对不同品系小鼠胚胎冷冻的效果。方法6个品系小鼠(KM、ICR、BALB/c、C57BL/6J、OB/OB、LAP/~TAOF59)的2-cell胚胎分别用EFS和DAP两种玻璃化冷冻方法进行冷冻和复苏,比较两种冷冻方法的胚胎复苏率和着床率。结果6个品系小鼠冷冻胚胎EFS方法的平均复苏率为69.97%(47.9%~83.6%),DAP方法的平均复苏率47.23%(26.3%-76.7%),EFS方法明显优于DAP方法。其中KM、ICR和BALB/c小鼠EFS方法的冷冻复苏率显著高于DAP方法(P〈0.01);冻融胚胎移植后EFS方法的平均着床率27.23%(1.75%一45.0%),DAP方法的平均着床率31.43%(7.0%一46.3%)。除KM、ICR小鼠外,其他4个品系小鼠的着床率DAP方法高于EFS方法。结论KM和ICR远交群小鼠胚胎适合用EFS方法冷冻保存;C57BL/6J、OB/OB、LAP/aTAOF59三个品系小鼠DAP方法优于EFS方法,但差异不大;BALB/c小鼠两种玻璃化冷冻方法的冻融胚胎着床率均较低,需进一步研究。  相似文献

Antibodies cytophilic for homologous and heterologous (guinea pig, rat) peritoneal cells of the mononuclear phagocyte type were demonstrated in the sera of mice immunized with sheep red cells. The formation of these antibodies can be induced by immunization with red cells with and without complete Freund's adjuvant. They are also present, in varying titres, in the sera of non-immunized mice. The peritoneal cavity of normal and immunized mice contains cells which adsorb sheep red cells. This phenomenon is significantly reduced by preincubating the peritoneal cells for two hoursin vitro at 37° C. The cytophilic activity of mouse sera can be abolished by absorption by spleen homogenate, but not by pulverized guinea pig kidneys. The haemolysin and haemagglutinin titres before and after absorption are practically the same. The cytophilic component of some sera was inhibited by 2-mercaptoethanol. Tests of the sera of mice revaccinated with sheep red cells showed, after separation on Sephadex G 200, that cytophilic activity was present mainly in the 7 S fraction, but also in the 19 S fraction. Cells adsorbing cytophilic antibodies stain supravitally with neutral red; in fixed and stained preparations they are typical macrophages and monocytoid mononuclears with varying degrees of affinity for basic dyes.  相似文献

The capacity of heat-killed meningococci and the polysaccharide-protein-lipopolysaccharide fraction ( PPLF ) isolated from the microbial cell wall for changing nonspecific immunological reactivity was studied. In this investigation CBA mice with high response and C57BL/6 mice with low response to sheep red blood cells (SRBC) were used. Heat-killed N. meningitidis, serogroup A, and PPLF , serogroups A and B, were found to enhance and suppress humoral response to the heterologous antigen. The effect of modulation depended on the dose of the antigen, the serogroup of meningococci, the scheme of the experiment and the strain of mice. The immune response of the vaccinated animals to the heterologous antigen was characterized by the following stages: the state of the adjuvant effect was replaced by the state of temporary immunodeficiency and then by enhanced response to SRBC.  相似文献

Summary The 3-methylcholanthrene (MCA)-induced BALB/c (H-2d) fibrosarcoma C-1 bears a strong tumor-specific transplantation antigen (TSTA) which, in previous studies, appeared to be distinct from H-2k alien antigens expressed by this tumor. To see whether a syngeneic anti-C-1 serum obtained by multiple immunizations with C-1 tumor cells contained anti-TSTA-specific antibodies, in vitro cytotoxicity tests were performed. The syngeneic anti-C-1 serum had a high cytotoxic activity on C-1 cells, which allowed an absorption analysis to be carried out. Absorption of the serum with C3Hf, AKR, or B10.BR normal lymphoid cells (all sharing H-2k antigens) reduced the cytotoxic activity on C-1 cells to 30%–50%. This residual activity could not be absorbed by FMR+ or G+ murine leukemias, by ecotropic endogenous virus obtained from SC-1 cells infected with the C-1 virus, by embryonic cells, or by normal BALB/c or C57BL/6 lymphoid cells. Conversely, the serum activity was abrogated by absorbing with the MCA-induced BALB/c fibrosarcomas C-1, ST2, C-3, GI-17, or CMS-1, and significantly lowered by the MCA-induced C3Hf fibrosarcoma C3H-7. A significant reduction of the anti-C-1 cytotoxicity was also obtained by absorbing with the two BALB/c fibrosarcomas teflon-9 and SCS (both lacking TSTA), by means of fresh newborn BALB/c or C3Hf muscle cells or of in vitro-cultured newborn BALB/c fibroblasts. These results suggest that, in spite of the strong transplantation immunity elicited by C-1 cells, antibodies to the individual TSTA of C-1 were undetectable in the syngeneic anti-C-1 serum obtained from animals highly resistant to the challenge of C-1 cells.  相似文献

The humoral immune responses of mice patently infected with Schistosoma mansoni and of mice vaccinated with radiation-attenuated cercariae were compared by radioimmunoassays and one- and two-dimensional polyacrylamide gel analyses of radioimmunoprecipitates. The binding observed with antibodies of mice vaccinated twice with radiation-attenuated cercariae over a period of 7 to 11 wk was less than 50% of the binding observed with antibodies of mice patently infected for 20 wk, but three to four times greater than that obtained with antibodies of mice infected for 6 wk, irrespective of whether the test antigen extracts were derived from schistosomula or adult worms. Sera of vaccinated mice precipitated a restricted number of predominantly high m.w. glycoproteins of both schistosomula and adult worms metabolically labeled with [35S] methionine. Each of the glycoproteins of 36 hr in vitro-cultured schistosomula that was precipitated by the sera of vaccinated mice was also precipitated by sera of infected mice. In contrast, sera of vaccinated mice uniquely precipitated a 38,000 m.w. glycoprotein of schistosomula cultured for 5 days and a 94,000 m.w. glycoprotein of adult male worms. Although radiation-attenuated larvae do not reach the adult stage, mice vaccinated with these still elicit a strong immune response against egg glycoproteins. In particular, an egg glycoprotein of 85,000 to 70,000 and isoelectric point of 4.8 showed an enhanced reactivity with sera of vaccinated mice in comparison with infected mice. These results show that the antibody response in mice vaccinated with radiation-attenuated larvae differs qualitatively and quantitatively from that of infected mice.  相似文献

Salmonella fIagellin has been repeatedly used as a carrier for heterologous peptide epitopes either as a parenterally delivered purified antigen or as a parenterally/orally-administered, flagellated, live, attenuated vaccine. Nonetheless, the ability to induce specific antibody responses against the flagellin moiety, fused or not with heterologous peptide, has not usually been reported in mice orally inoculated with a live, attenuated, flagellated Salmonella strain. In this work we evaluated the immunogenicity of flagellin in mice following oral inoculation with an aroA Salmonella enterica serovar Dublin SL5929 strain, which expressed plasmid-encoded recombinant hybrid flagellin fused to the CTP3 epitope (amino acids 50–64) of cholera toxin B-subunit. In contrast to parenterally immunized mice, no significant CTP3- or flagellin-specific antibody responses either in sera (IgG) or feces (IgA) were detected following repeated oral delivery of the recombinant Salmonella strain to C57BL/6 mice. Similarly, flagellin-specific antibody responses were also not detected in mice immunized with strain SL5930, which expressed a nonhybrid flagellin. The lack of flagellin-specific antibody responses was not associated with deficient Peyer patch colonization or spleen invasion. Moreover, stabilization of the flagellin-coding gene by integration into the host chromosome did not significantly improve flagellin-specific antibody responses following administration by the oral route. Taken together, these results suggest that flagellin does not represent an efficient peptide carrier for activation of antibody responses in mice orally immunized with live, attenuated Salmonella strains. The chromosomes of four different mouse strains were examined after administration of a single dose of cyclophosphamide, 100 mg/kg. Chromosomal breaks and rearrangements were produced in C3H mice three to four times as frequently and in AKR mice twice as frequently as in C57 B1 and XLII Orl mice. This indicates that cytogenetic investigations in mutation research using mice as in vivo test systems may be variable depending on the mouse strain employed. Because the effect of the alkylating agent was not pronounced in the two strains having a high incidence of cancer and leukemia (C3H and AKR), the possibility of a relationship between chromosomal instability and cancer proneness is discussed.  相似文献

Rabbit antisera were obtained against cytotoxic small peritoneal lymphocytes (IPEL) of CBA (H-2k) mice immune to alloantigens C57BL/6 (H-2b) and to the enriched 5-day MLC cytotoxic blast lymphocytes (MLC--CL). After appropriate absorption by cells and tissues of intact mice the cytotoxicity of the sera was lost relative to normal lymphoid cells. The absorbed anti-CPL serum inhibited, in the presence of complement, the cytotoxic effect of CPL but not that of MLC--CL on 51Cr-labeled allogeneic macrophages. This inhibition was restricted by idiotypic and strain specificity. Conversely, the absorbed anti-MLC--CL serum inhibited the cytotoxic effect of both CPL and MLC--CL of various mouse strains, irrespective of their immunologic specificity. It is supposed that the effect of the anti-CPL serum is mainly caused by antibodies againts idiotypic determinants of the killer T receptors, whereas the effect of the anti-MLC--CL serum is due to antibodies against differentiation antigens of the proliferating lymphocytes.  相似文献

The ganglioside patterns in the liver of different inbred and hybrid strains of mice were investigated. The inbred strains were Balb/cAnNCr1BR, C57BL/6NCr1BR, DBA/2NCr1BR. C3H/HeNCr1BR; the hybrid strain was the Swiss albino. The following major gangliosides were found to be present in mouse liver: GM3-NeuAc; GM3-NeuGl, GM2 [a mixture of one species carrying N-acetylneuraminic acid (NeuAc) and one carrying N-glycollylneuraminic acid (NeuGl)], GM1 and GD1a-(NeuAc,NeuGl). The qualitative and quantitative patterns of liver gangliosides were markedly different in the various inbred strains of mice; in Balb/cAnNCr1BR strain, ganglioside GM2 was preponderant (99.2% of total ganglioside content); in C57BL/6NCr1BR, the major ganglioside was GM2 (90.4%), followed by GM3-NeuAc (5.6%) and GM3-NeuGl (4.0%); in DBA/2NCr1BR, GM2 accounted for 77.1%, GD1a-(NeuAc,NeuGl) 18.9% and GM1 3.1% of gangliosides; in C3H/HeNCr1BR, GM2 constituted 50.6%, GM1 22.8% and GD1a-(NeuAc,NeuGl) 22.1%. In the hybrid Swiss albino mice, liver ganglioside composition markedly varied from one animal to another, GM3-NeuGl, GM2 and GD1a-(NeuAc,NeuGl) being the predominant gangliosides in the various cases.  相似文献
