对经紫外诱变,筛选出的突变菌株马疫链球菌SH-2,进行发酵条件的优化实验,通过单因素实验确定最佳摇瓶条件是种龄14-16h,初始pH7.6,发酵温度37℃,发酵时间44h;通过多因素正交实验摸索出营养培养基的最佳配比为:葡萄糖5%,酵母粉6.67%,牛肉膏3.33%,MgSO4·7H2O0.1%,MnSO4·4H2O0.02%,NaHCO30.3%,Na2HPO40.15%,NaCl0.3%,UTP0.05%,异甘草素0.08‰。  相似文献

目的:通过优化蜡状芽孢杆菌SWWL6产耐有机溶剂脂肪酶营养条件,产酶量有较大提高。方法与结果:通过单因素实验确定了产酶的最佳碳源、氮源及无机盐分别为可溶性淀粉、酵母膏、NH4NO3、MgSO4·7H2O和NaCl。部分因子实验结果表明初始培养基中酵母膏、NH4NO3的质量浓度对产酶的影响显著。通过最陡爬坡实验逼近最大响应区域。以中心组合设计和响应面分析法确定了最优培养基。结论:优化的培养基为酵母膏0.64%、NH4NO30.384%、可溶性淀粉1%、MgSO4·7H2O0.1%、NaCl0.25%、甲苯20%。优化后脂肪酶相对酶活为348.44%,比优化前提高了3.48倍。  相似文献

研究了不同碳源、氮源和无机盐对桑黄深层培养菌丝生物量和胞外多糖产量的影响,结果表明:在培养温度为26℃、摇床转速为160r/min、发酵时间为10d的条件下,以桑黄菌丝生物量为指标,最适碳源、氮源和无机盐分别是果糖或葡萄糖、酵母粉或酵母膏、KH2P04或MgSO4·7H2O;菌丝生物量分别达到1.51、1.31、1.69、1.52、1.52、1.36g/100mL;以胞外多糖为指标,最适碳源、氮源和无机盐分别是葡萄糖、酵母膏、ZnSO4·7H2O,胞外多糖产量分别达到0.49、0.45、0.22g/100mL。  相似文献

利用单因子实验和正交实验对假丝酵母(Candida sp.)突变菌株YQ5摇瓶发酵产生S-腺苷甲硫氨酸的培养基成分进行了优化。单因子实验结果表明, 发酵最适pH值为6.0, 最佳碳源为 8%蔗糖, 最佳氮源为1.5%胰蛋白胨, 酵母粉最适浓度为2%, MgSO4·7H2O、CaCl2、FeSO4·7H2O、CoCl2、CuSO4·5H2O、H3BO3等作为无机离子对胞内S-腺苷甲硫氨酸的积累均有促进作用。利 用正交实验获得了最终的发酵培养基配方。在最适条件下发酵48 h, S-腺苷甲硫氨酸含量可达1740.0 mg/L。  相似文献

溶剂稳定性蛋白酶产生菌Bacillus licheniformis YP1分离自油田土样。考察了碳源、氮源、金属离子等营养因素对YP1菌株发酵产溶剂稳定性蛋白酶的影响。YP1菌株发酵产胞外蛋白酶的最佳碳源为淀粉,果糖、甘露糖和乳糖显著抑制产酶;最佳氮源为酵母膏,干酪素、酵母粉和牛肉膏促进产酶,玉米浆和尿素显著抑制产酶。Mn^2+可以显著促进酶活,Mg^2+可以促进产酶,在初步优化的培养条件下,YP1菌株的胞外蛋白酶产量达980U。  相似文献

响应面法快速优化曲霉Aspergillus sp.F044产脂肪酶培养条件   总被引:1,自引:0,他引:1  
运用逐因子试验(Seriatim-Factorial Experiment)、Plackett-Burman、Response Surface Methodology(RSM)和单因子试验(Monofactorial Experiment)分析对产脂肪酶曲霉Aspergillus sp.F044产酶培养条件进行了快速优化。首先运用逐因子试验确定Aspergillus sp.F044产脂肪酶最适碳源和氮源,分别为麦芽糖和牛肉膏。在此基础上,通过Plackett-Burrman设计对影响其产酶相关因素进行评估并筛选出具有显著效应的橄榄油、牛肉膏、硫酸镁三个因素。然后通过实验拟合上述三因素与发酵液脂肪酶活力的一阶线性模型,沿着一阶模型给定的路径进行最速上升试验,在上升最高点处由中心组合试验和向应面分析确定其最优培养基组成;最后通过单因子试验确定最适发酵温度和摇床转速。试验优化的最优培养条件为:麦芽糖1.5%(w/v),硫酸铵7‰(w/v),磷酸氢二钾1‰(w/v),牛肉膏1.25%(w/v),硫酸镁2.11‰(w/v),橄榄油1.41%(v/v),自然pH,250r/min和30℃培养72h酶活达32.15U/mL。与初始11.18U相比,酶活提高2.88倍。  相似文献

本研究对枯草芽孢杆菌产中性蛋白酶的发酵条件进行优化。通过单因素试验探究碳源、氮源、无机盐离子及表面活性剂对枯草芽孢杆菌产中性蛋白酶的影响。结果表明,枯草芽孢杆菌产中性蛋白酶最佳碳源、氮源、无机盐离子及表面活性剂为蔗糖、牛肉膏、Ca Cl2及吐温-80。在单因素试验基础上,通过PlackettBurman试验设计和响应面分析法得到了产中性蛋白酶最佳发酵条件:蔗糖3%、牛肉膏2%、Ca Cl20.3%、吐温-80 0.55%、初始p H 7.5、发酵温度37℃、发酵时间50.4 h、接种量2.66%。在最佳发酵条件下,所产中性蛋白酶最大酶活为143.54 U,比优化之前提高1.3倍。  相似文献

枯草芽孢杆菌Bacillus subtilis B47菌株为番茄内生细菌, 也是玉米小斑病拮抗菌, 能产生对玉米小斑病菌有强烈抑制作用的抗菌物质。以B47菌株发酵液的无菌滤液对玉米小斑病菌的抗菌活性为检测指标, 测定B47菌株产抗菌物质培养所需的最佳碳、氮源和无机盐, 并通过正交试验法对该菌株产抗菌物质的培养基配方和摇瓶发酵条件进行优化。研究结果表明, B47菌株产抗菌物质最佳碳、氮源和无机盐分别为蔗糖、酵母浸膏和MgSO4·7H2O, 最优培养基是YSB (Yeast extract-sucrose-beef extract)培养基, 其配方为: 蔗糖2%, 酵母浸膏2%, 牛肉浸膏1.5%, MgSO4?7H2O 0.06%, FeSO4·7H2O 0.000 9%, 最优发酵条件组合为: 30 °C, pH 7.0, 170 r/min摇床培养6 d, 接种量为1%, 装液量为40 mL/200 mL。  相似文献

对莱氏野村菌(Nomuraea rileyi)菌株CQ031021产几丁质酶条件及酶学性质进行了研究。结果表明:该菌株最适产酶碳源为2.0%(W/V)葡萄糖,氮源为1.2%(W/V)复合氮源(蛋白胨、牛肉膏按1∶1的比例),接种量为孢悬液2mL(1×107个/mL),培养温度28℃,培养液初始pH6.0,培养时间6d;一定浓度的吐温-80对几丁质酶活性有促进作用,而SDS有抑制作用;粗酶液最适反应温度50℃,最适pH6.0,在40℃以下及pH5.5~6.5范围内酶活力较稳定。  相似文献

[目的]研究氮源、氮源和矿质元素对菌株MY07产吲哚乙酸量的影响。[方法]在单因素筛选和初始浓度确定试验的基础上,利用响应面分析法对细菌MY07产吲哚乙酸的液体发酵培养基进行优化,采用Box-Benhnken设计和响应面法对碳源、氮源和矿质元素水平进行分析。[结果]结果表明,菌株MY07产IAA的液体发酵培养基最佳组合为:0.94%甘露醇,2.35%KNO3,0.12‰Mg SO4·7H2O,0.13‰Ca Cl2·2H2O,在此条件下的验证试验表明,IAA产量可达到28.16μg/ml。[结论]通过响应面法优化菌株培养基后,有利于提高该菌产生IAA的能力。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
