? Premise of the study: Michelia maudiae is a threatened species in the Magnoliaceae. Microsatellite markers were developed and characterized in M. maudiae for further investigation of its conservation genetics. ? Methods and Results: Microsatellite markers were developed in M. maudiae using the Fast Isolation by AFLP of Sequences Containing Repeats protocol. Ten polymorphic microsatellites were assessed in two populations of M. maudiae. The number of alleles detected per locus varied from 1 to 8. Observed and expected heterozygosities ranged from 0.000 to 0.792 and from 0.000 to 0.826, respectively. Six primer pairs showed transferability in the two related species Michelia foveolata and Michelia chapensis. ? Conclusions: This set of microsatellite markers provides a useful tool for future studies of the conservation genetics of M. maudiae and other congeneric species.  相似文献

Genetic diversity and differentiation were analyzed in 11 populations of Magnolia stellata (Sieb. and Zucc.) Maxim. (Magnoliaceae) in the Tokai district, Japan. Variation at four nuclear microsatellite (nSSR) loci was examined, three chloroplast microsatellite (cpSSR) markers were developed and 13 haplotypes identified. The 11 populations were divided into three groups (A, B and C). Each population within the group was separated less than 40 km. Group B harbored the highest gene diversity (H) and allelic richness (Ar) for nSSR (H=0.74 and Ar=8.02). Group C had the highest diversity of chloroplast haplotypes (H=0.79 and Ar=6.8): 2.5 times more haplotypes than the other groups. Each population contributed differently to the total diversity, with respect to nSSR and cpSSR. AMOVA revealed that 58% of haplotypic and 15% of nSSR variation was partitioned among populations within groups. A Mantel test revealed significant correlations between population pairwise geographic ln(distance) and FST/(1−FST) for both nSSR (r=0.479; P=0.001) and cpSSR (r=0.230; P=0.040). Dendrograms of populations for nSSR, based on Nei’s genetic distance, were constructed using UPGMA and the neighbor-joining method. These results suggest that populations in group C have diverged from the other populations, while those in group B are similar to each other. For group B, fragmentation between populations should be avoided in order to maintain gene flow. For group C, the uniqueness of each population should be given the highest priority when planning genetic conservation measures for the species.  相似文献

Rhododendron simsii Planch. (Ericaceae) is a valuable horticultural and medicinal plant species. In this study, the genetic diversity of eight wild R. simsii populations from the Dabie Mountains (central China) was investigated with 29 microsatellite markers. The results showed that R. simsii harbored a high level of genetic diversity (HE: 0.64–0.79; HO: 0.71–0.94; I = 1.917; h = 0.826), and 84.34% of this genetic variation was maintained within populations, while variation among populations only accounted for 15.66%. The number of alleles ranged from 6 to 11, with an average of 9.069. Heterozygote excess was found, with the mean FIS and FIT values of ?0.1739 and 0.0092, respectively. The average value of gene flow (Nm) was 1.3525. Within‐population, genetic diversity (I) ranged from 1.131 to 1.681. Cluster analyses divided the eight populations into two clades: the ‘Ltjiuzihe’ population formed its own cluster, while the other seven populations clustered together. There was a weak negative correlation between genetic and geographic distance. The results are highly relevant for the conservation and sustainable utilization of wild R. simsii germplasm resources in central China.  相似文献

Oriental beech (Fagus orientalis Lipsky) is a widespread monoecious and wind-pollinated tree species. It is one of the major components of the Hyrcanian forests of Iran and it is of both ecological and economical importance. Twelve beech stands were surveyed at 9 chloroplast (cp) and 6 nuclear (n) polymorphic microsatellite loci (simple sequence repeats, SSR) to provide information on distribution of genetic diversity within and among populations and on gene conservation and silvicultural management of this species. High levels of genetic differentiation were detected for the chloroplast genome (F ST = 0.80 and R ST = 0.95), in sharp contrast to the nuclear genome (F ST = 0.06, R ST = 0.05). The analysis of molecular variance (AMOVA) showed that 48% of the total cpSSR variation was attributable to differences among regions and 30% to differences among populations within regions, suggesting multiple origins of beech populations in Hyrcanian forests. Nuclear SSRs confirmed the presence of significant differentiation among populations and among geographic regions, even if, as expected, this was less pronounced than that found with cpSSRs (based on AMOVA, differences among regions and among populations within regions each contribute 5% to total nSSR variance). A highly significant correlation between genetic (nSSRs) and geographic distances (R 2 = 0.522) was estimated, thus showing an isolation by distance effect. The application of spatial analysis of molecular variance (SAMOVA) using both marker data allowed identification of genetically homogeneous groups of populations. Possible applications of these results for the certification of provenances and/or seed lots and for designing conservation programs are presented and discussed.  相似文献

Genetic diversity of 1680 modern varieties in Chinese candidate core collections was analyzed at 78 SSR loci by fluorescence detection system. A total of 1336 alleles were detected, of which 1253 alleles could be annotated into 71 loci. For these 71 loci, the alleles ranged from 4 to 44 with an average of 17.6, and the PIC values changed from 0.19 to 0.89 with an average of 0.69. (1) In the three genomes of wheat, the average genetic richness was B>A>D, and the genetic diversity indexes were B>D>A. (2) Among the seven homoeologous groups, the average genetic richness was 2=7>3>4>6>5>1, and the genetic diversity indexes were 7>3>2>4>6>5>1. As a whole, group 7 possessed the highest genetic diversity, while groups 1 and 5 were the lowest. (3) In the 21 wheat chromosomes, 7A, 3B and 2D possessed much higher genetic diversity, while 2A, 1B, 4D, 5D and 1D were the lowest. (4) The highest average genetic diversity index existed in varieties bred in the 1950s, and then it declined continually. However, the change tendency of genetic diversity among decades was not greatly sharp. This was further illustrated by changes of the average genetic distance between varieties. In the 1950s it was the largest (0.731). Since the 1960s, it has decreased gradually (0.711, 0.706, 0.696, 0.695). The genetic base of modern varieties is becoming narrower and narrower. This should be given enough attention by breeders and policy makers.  相似文献

Japanese rice (Oryza sativa L.) cultivars that are strictly used for the brewing of sake (Japanese rice wine) represent a unique and traditional group. These cultivars are characterized by common traits such as large grain size with low protein content and a large, central white-core structure. To understand the genetic diversity and phylogenetic characteristics of sake-brewing rice, we performed amplified fragment length polymorphism and simple sequence repeat analyses, using 95 cultivars of local and modern sake-brewing rice together with 76 cultivars of local and modern cooking rice. Our analysis of both nuclear and chloroplast genome polymorphisms showed that the genetic diversity in sake-brewing rice cultivars was much smaller than the diversity found in cooking rice cultivars. Interestingly, the genetic diversity within the modern sake-brewing cultivars was about twofold higher than the diversity within the local sake-brewing cultivars, which was in contrast to the cooking cultivars. This is most likely due to introgression of the modern cooking cultivars into the modern sake-brewing cultivars through breeding practices. Cluster analysis and chloroplast haplotype analysis suggested that the local sake-brewing cultivars originated monophyletically in the western regions of Japan. Analysis of variance tests showed that several markers were significantly associated with sake-brewing traits, particularly with the large white-core structure.  相似文献

应用微卫星标记分析23个中国地方马种的遗传多样性   总被引:2,自引:0,他引:2  
为了调查中国马种的群体遗传分化与遗传结构状况, 本研究应用FAO和ISAG推荐的25对微卫星引物, 结合荧光标记PCR分析技术, 对中国23个马群体和1个英纯血马群体进行了分子遗传学研究。结果表明: 中国地方马群体的遗传多样性比较丰富, 23个中国马群体的等位基因数、多态信息含量和遗传杂合度等都高于英国纯血马。根据群体遗传距离构建的系统进化树, 能够清晰地将英纯血马从中国马中独立出来, 同时可将中国马群体分成不同的支, 基本上与其地理分布格局相符。我们用MVSP软件进行群体遗传分化分析可见, 前三个主成分的三维散点图可以明显地把英纯血马从所有群体中独立出来, 并把中国的马群体分成几个相对独立的支。进一步分析第一、二主成分的二维散点图, 可将中国的马群体分化成南方支系、藏马支系、新疆和青海支系、内蒙古支系及东北支系等5个部分。根据Structure软件分析, 推测中国马群体含有5个潜在的支系, 基本上代表了我国现在主要家马来源的基础遗传支系。这些信息可以为我国现有马种类型的划分与马种资源遗传多样性的保护提供科学依据。  相似文献

同域分布的近缘物种常常发生杂交而导致种间基因渐渗, 从而对相关物种的自然居群遗传结构产生重要影响, 近缘种间的杂交渐渗已成为进化生物学和保护生物学关注的热点。本研究采用8对cpSSR引物对我国西部高原台地向中东部丘陵平原过渡地带同域重叠分布的猕猴桃属(Actinidia)7个物种的自然居群遗传多样性、居群遗传结构和同域分布种间遗传分化进行了检测。结果表明: (1)在6个多态性位点检测到18个等位基因形成的42个单倍型, 尽管各单倍型间显示了复杂的网状进化关系, 但还是具有明显的物种特异性; (2)各物种有丰富的cpSSR遗传多样性, 但种间存在较大差异, 绵毛猕猴桃(Actinidia fulvicoma var. lanata)的遗传多样性水平最高(P = 62.50%, hT = 0.173, HT = 0.897), 美味猕猴桃(A. deliciosa)的最低(P = 37.5%, hT = 0.041, HT = 0.516); (3)尽管不同物种的居群分化程度存在较大差异, 但种内居群间存在明显分化(GST为0.319–0.780, FST为0.401–0.695), 居群间的基因流不足(Nm为0.219–0.747<1); 其中以美味猕猴桃的居群遗传分化度最高(GST = 0.780, FST = 0.695); (4)遗传分化系数GST(unordered alleles)与NST(ordered alleles)无显著差异, 揭示本研究的大多数猕猴桃属物种不存在系统地理结构, 与用Mantel检验得出的居群遗传距离和地理距离不存在显著性相关的结果一致; (5)除了中华/美味猕猴桃复合体(A. chinensis / A. deliciosa complex)的湖北五峰(HW)和广西资源(GZ)两个同域复合居群外, 同域分布的物种间遗传分化强烈(FST为0.476–0.990), 与UPGMA聚类时多数居群按各自物种聚类的结果一致。进一步分析表明, 中华/美味猕猴桃复合体近缘种间存在明显的共祖多态性和杂交渐渗现象, 近缘种植株分布的交错程度以及是否存在亚居群结构对杂交渐渗存在着重要影响。亲缘关系较远的物种间杂交渐渗事件稀少, 但存在个别同塑事件。本研究结果有助于进一步了解猕猴桃属植物自然居群cpDNA的遗传特性和渐渗杂交进化模式, 为我国猕猴桃野生种质资源保育及可持续开发利用提供基础数据和科学依据。  相似文献

Intravarietal genetic diversification associated with geographical dispersal of a vegetatively propagated species was studied using grapevine Vitis vinifera L. 'Cabernet Sauvignon' as a model. Fifty-nine clonal samples obtained from 7 countries (France, Chile, Spain, Australia, Hungary, USA, and Italy) were analyzed using 84 microsatellite markers. Eighteen polymorphic microsatellite loci (21.4%) were detected, finding 22 different genotypes in the population analyzed with a genetic similarity of over 97%. The presence of chimeric clones was evidenced at locus VMC5g7 by means of a segregation analysis of descendants by self-pollination of a triallelic Chilean clone and by somatic embryogenesis analysis, showing a mutation in L2 cell layer. Only 2 clones (obtained from France and Australia) presented the ancestral genotype, and the most divergent genotype was exhibited by another French clone, which had accumulated 5 somatic mutations. The 2 largest populations considered (from France and Chile) showed a clear divergency in the polymorphisms detected. These antecedents enabled the tracing of geographical dispersal with a phylogenetic hypothesis supporting France as the center of origin of diversification of Cabernet Sauvignon. The results obtained could help to explain diversification processes in other grapevine cultivars. The possibility that this kind of genetic variability occurs in other vegetatively propagated species is discussed, focusing on possible fingerprinting applications.  相似文献

Fourteen microsatellite loci of Phoenix dactylifera were targeted to examine the genetic diversity in Tunisian date-palms germplasm. They showed a high level of polymorphism in 49 accessions from three main oases with little geographic structure within Tunisia. The microsatellite data agrees with previous analyses of Tunisian germplasm using other molecular markers. 100% of local date-palms accessions were successfully fingerprinted and easily distinguished by the help of only three loci. The possibility of using microsatellites for large scale molecular labelling of offshoots and in vitro plantlets and their implication in the plant material certification is discussed.  相似文献

Genetic diversity among rice genotypes, including 15 indica basmati advance lines and 5 basmati improved varieties were investigated by 28 SSR markets including one indel marker. The SSRs covered all the 12 chromosomes that distributed across the rice genomes. The mean number of alleles per locus was 3.60, showing average number of polymorphism information content was 0.48. A total of 101 alleles were also identified from the microsatellite marker loci. A number of SSR markers were also identified that could be utilized to differentiate between rice genotypes. Pair wise Nei,s genetic distance between rice genotypes ranged from 0.07 to 0.95. The dendrogram based on cluster analysis by using SSR polymorphism that grouped the 20 genotypes of rice in to five clusters based on their genetic similarity. The result could be useful for the identification and selection of the diverse genotypes for the future cross breeding program and development of new rice varieties.  相似文献

The genetic diversity and population structure of hinoki (Chamaecyparis obtusa) in Japan were investigated by examining the distribution of alleles at 13 microsatellite loci in 25 natural populations from Iwaki in northern Japan to Yakushima Island in southern Japan. On average, 26.9 alleles per locus were identified across all populations and 4.0% of the genetic variation was retained among populations (G ST = 0.040). According to linkage disequilibrium analysis, estimates of effective population size and detected evidence of bottleneck events, the genetic diversity of some populations may have declined as a result of fragmentation and/or over-exploitation. The central populations located in the Chubu district appear to have relatively large effective population sizes, while marginal populations, such as the Yakushima, Kobayashi and Iwaki populations, have smaller effective population sizes and are isolated from the other populations. Microsatellite analysis revealed the genetic uniqueness of the Yakushima population. Although genetic differentiation between populations was low, we detected a gradual cline in the genetic structure and found that locus Cos2619 may be non-neutral with respect to natural selection.  相似文献

By using six polymorphic microsatellite loci, the population genetic diversity and structure of 120 individuals of Portunus trituberculatus were examined in five wild populations representing different geographical and ecological ranges along the coast of China. Middle level of genetic diversity of P. trituberculatus was revealed by observed heterozygosity, allele number and Shannon information index. Pairwise population differentiations were found between all pairs of populations except Beihai (BH) and Qingdao (QD) (P < 0.05). The greatest differentiation was detected between Yingkou (YK) and QD populations. It was inconsistent with the result in previous study using mitochondrial control region, which showed no differentiation between QD and YK population. Recent bottleneck was identified in Ningbo (NB) population under TPM and IAM models, as well as Dandong (DD) and QD populations under IAM model. Additionally, compared with YK, QD, DD and BH populations, lower historical effective population size with lower genetic diversity were detected in NB population, which called for urgent assessment of the ecological and economic potential of NB fisheries.  相似文献

Genetic diversity and population structure of 9 populations of Bufo gargarizans with total 111 samples in China were assessed using seven microsatellite loci. The analysed microsatellite markers produced 161 alleles, varied from 9 to 38 alleles each locus. The number of alleles per population per locus ranged from 4.43 to 10.29. Polymorphic information content showed that all seven loci were highly informative (mean = 0.810 ± 0.071). The average observed heterozygosity was less than the expected (0.353 ± 0.051 and 0.828 ± 0.067, respectively). All tested populations gave significant departures from Hardy–Weinberg equilibrium. Genetic differentiation among the populations was considerably high with the overall and pairwise F ST values (mean = 0.160 ± 0.039), and showed fairly high level of inbreeding (indicated by a mean F IS value of 0.504 ± 0.051) and global heterozygote deficit. In comparison to other amphibian studies; however, our results suggested that the level of genetic structuring in B. gargarizans was relatively low in the geographical scale of the study area. Interestingly, the speculated population bottleneck was found to be absent and the analyses provide only weak evidence for a recent contraction in size even though there was severe inbreeding (indicated by the F IS value) in the Chinese toad populations.  相似文献

Amochi (Arisaema schimperianum Schott) is an off-season crop plant in southern Ethiopia, grown during the dry season on residual moisture, for its edible tubers. It has gained importance as a "security crop" especially during the years of moisture stress and food shortage. Amochi is irritating in contact to the skin. Removal of this effect is an important question for breeding. As the first step, however we attempt to establish base line information of its breeding system and genetic variability using AFLPs. The extent of genetic differentiation among 11 populations (96 individuals) of amochi sampled along altitudinal gradients that varied from 1700 to 3200 m a.s.l. was investigated. The populations were classified in to three altitudinal groups: lowland (1700 to 2200 m a.s.l.), central-highland (2201 to 2600 m a.s.l.) and highland (2601 to 3200 m a.s.l.). Polymorphic loci (167) scored from four primer pair combinations, were used for principal component analysis (PCA), and analysis of molecular variance (AMOVA). Both PCA and unweighed pair group with arithmetic mean (UPGMA) clearly differentiated populations into their respective altitude groups, with large genetic distances. AMOVA analysis revealed 70.5%, 16.7% and 12.8% variability between altitude groups, between populations and within populations respectively. Average diversity indices within populations were also low. Since the largest proportion of variation is located between altitude groups, rather than within populations, we suggest future studies on the chemical composition, low irritation, and other desirable traits should consider populations from different altitude ranges.  相似文献

The edible cockle Cerastoderma edule is a marine bivalve commercially fished in several European countries that have lately suffered a significant decrease in production. Despite its commercial importance, genetic studies in this species are scarce. In this work, genetic diversity and population differentiation of C. edule has been assessed using 11 microsatellite markers in eight locations from the European Atlantic coast. All localities showed similar observed and expected heterozygosity values, but displayed differences in allelic richness, with lowest values obtained for localities situated farther north. Global Fst value revealed the existence of significant genetic structure; all but one locality from the Iberian Peninsula were genetically homogeneous, while more remote localities from France, The Netherlands, and Scotland were significantly different from all other localities. A combined effect of isolation by distance and the existence of barriers that limit gene flow may explain the differentiation observed.  相似文献

? Premise of the study: We developed microsatellite makers for Parakmeria nitida to investigate its population structure and conservation genetics. ? Methods and Results: A total of 25 microsatellite primer pairs were developed using the Fast Isolation by AFLP of Sequences COntaining repeats (FIASCO) protocol, and polymorphism was assessed in three natural populations of P. nitida. Among these markers, 11 were monomorphic and 14 showed polymorphism. ? Conclusions: These markers are potentially useful for future population genetic analyses of P. nitida and will serve as an important tool for conservation efforts.  相似文献

Assessment of the extent of genetic variability within chickpea is fundamental for chickpea breeding and conservation of genetic resources and is particularly useful as a general guide in the choice of parents for breeding hybrids. To establish genetic diversity among 60 accessions of chickpea comprising landraces, internationally developed improved lines, and cultivars, genetic distances were evaluated using 14 simple sequence repeat markers. These markers showed a high level of polymorphism; a total of 59 different alleles were detected, with a mean of 4.2 alleles per locus. The polymorphic information content (PIC) value ranged from 0.31 to 0.89. All the markers, with the exception of TAA170, TA110, GA34, and Ts35, were considered to be informative (PIC > 0.5), indicating their potential usefulness for cultivar identification. Based on the UNJ clustering method, all accessions were clustered in five groups, which indicated the probable origin and region similarity of Iranian landraces over the other cultivars. It also represents a wide diversity among available germplasm. The result has firmly established that introduction of genetic materials from exotic sources has broadened the genetic base of the national chickpea breeding program. As further implications of the findings, this study can be useful for selective breeding for specific traits and in enhancing the genetic base of breeding programs.  相似文献

The results of the study of 21 populations of Siberian spruce (Picea obovata Ledeb.) from different parts of the species natural range by microsatellite (SSR) analysis of nuclear DNA are presented. Using nine loci developed for Picea abies (L.) Karst. and Picea glauca (Moench) Voss and detecting variation in Picea obovata, the parameters of intra- and interpopulation genetic diversity, as well as the degree of population differentiation, were determined. It was demonstrated that the population of Siberian spruce in the study was characterized by a relatively high average level of intrapopulation variability (Ho = 0.408; He = 0.423) and low interpopulation differentiation (Fst = 0.048, P = 0.001) at this class of DNA markers. The genetic distance between populations ranged from 0.009 to 0.167, averaging 0.039. The isolated Magadan population, located in the extreme Northeast of Russia at a considerable distance from the main species range and characterized by the lowest genetic diversity among the studied populations, was maximally differentiated from the rest of the spruce populations. In addition, the steppe Ubukun population from Buryatia and the population from the Bogd Khan Uul Biosphere Reserve, Mongolia, were considerably different in the genetic structure from most populations of Siberian spruce, although to a lesser extent than the Magadan population. These findings are consistent with the results of previous studies of this species carried out using allozyme and microsatellite loci of chloroplast DNA and point to the prospects of using nuclear microsatellites as DNA markers to analyze the population genetic structure of Siberian spruce.  相似文献
