The effects of Pb chelater (EDTA-Pb) and ionic Pb (Pb(NO(3))(2)) on root cell death, Pb accumulation, changes of ROS, activities of antioxidant enzymes and uptake of mineral elements in response to Pb toxicity in Sedum alfredii H. were compared. Loss of plasma membrane integrity became serious by increasing Pb concentration in the medium, 200 microM Pb + 200 microM EDTA has alleviated the root cell death. The biomass was significantly affected by high concentration of Pb, and root growth was also affected by EDTA-Pb compared with ionic Pb. Lead accumulation was higher in the samples treated with ionic lead than that of the control. The concentration of reactive oxygen species (ROS) was determined by fluorescence microscopy, which indicates that the Pb stress increased the content of ROS significantly, whereas the EDTA-Pb decreased the burst of H(2)O(2). High Pb concentrations increase the activity of SOD and LOX. The Cu concentration in root increased significantly under Pb and EDTA-Pb treatment, and 200 microM Pb markedly increased the Fe content in roots. Under ionic Pb condition, the contents of Mg, Ca and K in shoots decreased, whereas they were significantly increased in case of EDTA-Pb. These results suggested that accumulating ecotype of S. alfredii roots were inefficient in uptake of higher concentration of EDTA-chelated Pb for long treatment duration, and that lead toxicity could be alleviated by EDTA.  相似文献

Phytoremediation of Pb contaminated soil with polymer-coated EDTA   总被引:1,自引:0,他引:1  
EDTA-assisted phytoextraction of lead (Pb) has been developed, but concerns have arisen due to the possibility of leaching of both Pb and EDTA to ground water caused by uncontrolled release. We developed five types of controlled-release EDTA (polymer-coated EDTA) by coating the EDTA with a polyolefin polymer. A test of the release rate showed that the duration for the release of 75% of total EDTA ranged from 3 to 210 days. A pot experiment was conducted to compare the effect of these polymer-coated EDTA and non-coated EDTA on the concentrations of Pb and EDTA in soil solution, and Pb accumulation in sorghum (Sorghum bicolor L. cv. EARLY SUMAC) in a Pb-contaminated soil. One of the polymer-coated EDTAs, C-EDTA-4, with a release period of 80 days proved to be the best in decreasing Pb and EDTA concentrations in soil solution, and increasing Pb accumulation in sorghum shoots compared to the direct application of EDTA. Our results suggest that polymer-coated EDTA has a potential for phytoextraction of Pb with a reduced environmental risk.  相似文献

The phytotoxicity and antioxidative adaptations of lead (Pb) accumulating ecotype (AE) and non-accumulating ecotype (NAE) of Sedum alfredii Hance were investigated under different Pb treatments involving 0, 0.02 mmol/L Pb, 0.1 mmol/L Pb and 0.1 mmol/L Pb/0.1 mmol/L ethylenediaminetetraacetic acid (EDTA) for 6days. With the increasing Pb level, the Pb concentration in the shoots of AE plants enhanced accordingly, and EDTA supply helped 51% of Pb translocation to shoots of AE compared with those treated with 0.1 mmol/L Pb alone. Moreover, the presence of EDTA alleviated Pb phytotoxicity through changes in plant biomass, root morphology and chlorophyll contents. Lead toxicity induced hydrogen peroxide (H2O2) accumulation and lipid peroxidation in both ecotypes of S. alfredii. The activities of superoxide dismutase (SOD), guaiacol peroxidase (G-POD), ascorbate peroxidase, and dehydroascorbate reductase elevated in both leaves and roots of AE as well as in leaves of NAE with the increasing Pb levels, but SOD and G-POD declined in roots of NAE. Enhancement in glutathione reductase activity was only detected in roots of NAE while a depression in catalase activity was recorded in the leaves of NAE. A significant enhancement in glutathione and ascorbic acid (AsA) levels occurred in both ecotypes exposed to Pb and Pb/EDTA treatment compared with the control, however, the differences between these two treatments were insignificant. The dehydroascorbate (DHA) contents in roots of both ecotypes were 1.41 to 11.22-fold higher than those in leaves, whereas the ratios of AsA to DHA (1.38 to 6.84) in leaves altering more to the reduced AsA form were much higher than those in roots. These results suggested that antioxidative enzymes and antioxidants play an important role in counteracting Pb stress in S. alfredii.  相似文献

Previous studies have shown that EDTA is necessary to solubilize soil Pb and facilitate its transport from the soil to the above ground plant tissues. These studies have also suggested that Pb is accumulated in the plant tissue with transpiration as the driving force. We conducted further studies to evaluate the relationship between EDTA soil treatment, plant transpiration, and plant accumulation of Pb and EDTA. Indian mustard (Brassica juncea) plants were grown in soils containing Pb at three different concentrations (1.5, 3.0 and 4.8 mmol/kg) for 5 weeks before being treated with EDTA concentrations ranging from 0 to 10 mmol/kg. Plant shoots and xylem sap were collected and analyzed for Pb and EDTA content using ICP and HPLC, respectively. Water loss was measured for 7 days following EDTA application. Transpiration was not affected at <5 mmol/kg EDTA but, at 10 mmol/kg EDTA transpiration decreased by 80%, whereas accumulation of Pb and EDTA increased. In the Sassafras soil, Pb and EDTA accumulation in the plant shoots continued to increase as the applied EDTA concentration increased, except at the highest level (10 mmol/kg). In soil amended with 4.8 mmol/kg Pb and 10 mmol/kg EDTA, the concentrations of EDTA and Pb in shoots decreased and visible signs of phytotoxicity were observed. The results presented herein support recent studies in hydroponic systems showing that EDTA and Pb are taken up by the plant and suggest that Pb is translocated in the plant as the Pb-EDTA complex. The results also show that the maximum Pb accumulation by plants occurs by maximizing the concentration of the Pb-EDTA complex based on the EDTA extractable soil Pb. Pisum sativum plants were treated for 3 days with an aqueous solution of 100 μM Pb(NO3)2 or with a mixture of lead nitrate and ethylenediaminetetraacetic acid (EDTA) or [S,S]-ethylenediaminedisuccinic acid (EDDS) at equimolar concentrations. Lead decline from the incubation media and its accumulation and localization at the morphological and ultrastructural levels as well as plant growth parameters (root growth, root and shoot dry weight) were estimated after 1 and 3 days of treatment. The tested chelators, especially EDTA, significantly diminished Pb uptake by plants as compared to the lead nitrate-treated material. Simultaneously, EDTA significantly enhanced Pb translocation from roots to shoots. In the presence of both chelates, plant growth parameters remained considerably higher than in the case of uncomplexed Pb. Considerable differences between the tested chelators were visible in Pb localization both at the morphological and ultrastructural level. In Pb+EDTA-treated roots, lead was mainly located in the apical parts, while in Pb+EDDS-exposed material Pb was evenly distributed along the whole root length. Transmission electron microscopy and EDS analysis revealed that in meristematic cells of the roots incubated in Pb+EDTA, large electron-dense lead deposits were located in vacuoles and small granules were rarely noticed in cell walls or cytoplasm, while after Pb+EDDS treatment metal deposits were restricted to the border between plasmalemma and cell wall. Such results imply different ways of transport of those complexed Pb forms.  相似文献

Lead (Pb) contamination of soils is a widespread problem. Mycorrhizal inoculation and synthetic chelators such as ethylenediaminetetraacetic acid (EDTA) may be useful for improving phytoremediation efficiency in Pb-contaminated soils. A greenhouse experiment was performed to study the influence of inoculation with arbuscular mycorrhizal fungus (AMF), Glomus mosseae, and addition of EDTA on phytoremediation of Pb by sunflowers (Helianthus annuus) in a calcareous soil. The experiment was a completely randomized design in a factorial arrangement with five levels of Pb, two levels of mycorrhizal treatments, and two levels of EDTA. Inoculation increased root colonization as Pb levels increased, but the addition of EDTA decreased it. Shoot and root dry matter yields increased by inoculation; however, they decreased with EDTA and Pb levels in co-application treatments. Pb concentration in shoots was significantly higher than that in roots, indicating a translocation factor greater than 1. Inoculation or addition of EDTA significantly increased Pb in roots and its translocation to shoots. The uptake index (UI) value increased in co-application of EDTA and AMF and the individual application of them; it is, therefore, concluded that both AMF and EDTA are effective in phytoremediation of Pb by sunflowers in the studied soil.  相似文献

Lead solubilization in soil and accumulation by spring wheat (Triticum aestivum L.) was studied in response to the ethylenediaminetetraacetic acid (EDTA) application method. In this study, 4 mmol EDTA kg?1 was applied using two application methods (a single dose and split doses) either alone or in combination with elemental sulfur. Results indicate that the application of EDTA in four equal splits at 1 mmol kg?1 during the growth period resulted in significantly higher shoot dry matter than its application at 4 mmol kg?1 at once 10 d before harvesting the wheat crop at the bolting stage. EDTA applied in split doses resulted in less lead (Pb) solubilization as compared with the single-dose application. The split application also significantly increased the shoot Pb concentration and Pb accumulation by wheat shoots as compared with the single-dose application. Despite its lesser effect on Pb solubilization, the EDTA application in split doses substantially increased Pb accumulation; thus, it is expected to minimize the risk of groundwater contamination.  相似文献

Greenhouse and field trial experiments were performed to evaluate the use of Chromolaena odorata with various soil amendments for phytoextraction of Pb contaminated soil Pb mine soils contain low amount of nutrients, so the additions of organic (cow manure) and inorganic (Osmocote and NH4NO3 and KCl) fertilizers with EDTA were used to enhance plant growth and Pb accumulation. Greenhouse study showed that cow manure decreased available Pb concentrations and resulted in the highest Pb concentration in roots (4660 mg kg(-1)) and shoots (389.2 mg kg(-1)). EDTA increased Pb accumulation in shoots (17-fold) and roots (11-fold) in plants grown in soil with Osmocote with Pb uptake up to 203.5 mg plant(-1). Application of all fertilizers had no significant effects on relative growth rates of C. odorata. Field trial study showed that C. odorata grown in soil with 99545 mg kg(-1) total Pb accumulated up to 3730.2 and 6698.2 mg kg(-1) in shoots and roots, respectively, with the highest phytoextraction coefficient (1.25) and translocation factor (1.18). These results indicated that C. odorata could be used for phytoextraction of Pb contaminated soil. In addition, more effective Pb accumulation could be enhanced by Osmocote fertilizer. However, the use of EDTA in the field should be concerned with their leaching problems.  相似文献

In the present study, increasing ethylenediaminetetraacetic acid (EDTA) concentration from 0 to 0.5 mmol L(-1) resulted in progressive increases in root elongation and in shoot and root dry matter (DM) of Indian mustard seedlings (Brassica juncea. L.) exposed at 0.5 mmol L(-1) of lead (Pb). The highest concentration of Pb in the shoots of Indian mustard reached 1140 mg kg(-1) dry weight (DW) in the treatment with 0.5 mmol L(-1) of Pb + 0.25 mmol L(-1) of EDTA. A significantly positive correlation was found between the concentrations of Pb and EDTA in the shoots of mustard. Roots were pretreated with an MC (methanol:trichloromethane) solution, 0.1 mol L(-1) of HCl, and 65 degrees C hot water. The plants were then exposed to 0.5 mmol L(-1) of Pb + 3 mmol L(-1) of EDTA in solution for 2 d. The pretreatments with MC, HCl, and hot water all increased the concentration of Pb in shoots by 14-, 7-, and 15-fold, respectively, compared with the shoots that had not been pretreated. Therefore, some physiological damage to roots would be useful to enhance the uptake of metal by plants and to minimize the application of doses of chelates in the practical operation of chelate-assisted phytoremediation.  相似文献

Leaching of metals due to enhanced mobility during ethylenediaminetetraacetic acid (EDTA)-assisted phytoextraction has been demonstrated as one of the potential hazards associated with this technology. This study was conducted to determine phytoextraction efficiency of Chenopodium album L. for Pb and EDTA-assisted (1.5, 3, and 9 mmol kg?1) phytoextraction and potential for leaching of Pb. The results demonstrated that BCFshoot (bioconcentration factor) was relatively higher than the BCFroot. Translocation factor in the shoot was higher than the roots. Thus, plant species would be applicable for Pb phytoextraction. EDTA enhanced translocation of Pb from roots to shoots. Lead content in the plant parts was maximum in the shoot and root of 9EDTA and 3EDTA, respectively. However, there was no significant difference between 3EDTA and 9EDTA. Lead concentration in the plant parts increased significantly from vegetative stage into flowering stage. Lead content taken up by the plant was lowest when EDTA was applied in a single dose. Therefore, application of EDTA in several increments rather than a single split reduced the leaching risk. Totally, optimum phytoextraction was observed when 3 mmol kg?1 EDTA was added in triple dosage 60 days after the plant cultivation under triple application mode. The results indicated the plant has the potential for Pb phytoextraction, but it should not be used unless the biomass containing such accumulated metal is removed for disposal. Significant improvement over current ETDA-assisted phytoextraction of Pb may be possible but should be implemented cautiously because of environmental risk.  相似文献

Enhancing phytoremediative ability of Pisum sativum by EDTA application   总被引:5,自引:0,他引:5  
The aim of our research was to demonstrate how the presence of EDTA affects resistance of pea plants to Pb and Pb-EDTA presence, and to show the effectivity of lead ions accumulation and translocation. It was determined that EDTA not only increased the amount of Pb taken up by plants but also Pb ion transport through the xylem and metal translocation from roots to stems and leaves. It can be seen in the presented research results that addition of the chelator with Pb limited metal phytotoxicity. We also demonstrated a significant effect of EDTA not only on Pb accumulation and metal transport to the aboveground parts but also on the profile and amount of thiol compounds: glutathione (GSH), homoglutathione (hGSH) or phytochelatins (PCs), synthesized by the plants. We observed a significant effect of the synthetic chelator on increasing the level of Pb accumulation in roots of plants treated with Pb including EDTA (0.5 and 1 mM). Pisum sativum plants treated only with 1 mM Pb(NO3)2 accumulated over 50 mg Pb x g(-1) dry wt during 4 days of cultivation. Whereas in roots of pea plants exposed to Pb+0.5 mM EDTA 35% more Pb was observed. When 1 mM EDTA was applied roots of pea accumulated over 67% more metal. The presence of EDTA also increased metal uptake and transport to the aboveground parts. In pea plants treated only with 1 mM lead nitrate less than 3 mg Pb x g(-1) dry wt was transported, whereas in P. sativum treated with Pb-EDTA doubled amount of Pb was observed in stems and leaves.  相似文献

Sunflower plants were grown hydroponically under controlled conditions with the root systems confined in small containers. Root confinement inhibited the growth of sunflower plants as indicated by reduction in both leaf and cotyledon area and root and shoot fresh weight. This effect was more pronounced in shoots. Root confinement favored the accumulation of potassium in the roots and shoots, and the exudation of potassium and water in excised roots. Xylem sap from root confined plants inhibited cotyledon expansion as revealed by bioassay with decapited sunflower seedlings. In addition decapited control plants incubated in ABA solution also showed cotyledon growth reduction. Xylem sap ABA analysis indicated a 7-times higher concentration in root confined than control plants. Our results suggest the synthesis of a chemical signal in the roots of plants subjected to mechanical stress which can be responsible for the inhibition of plant growth.  相似文献

Lead (Pb) is an environmental pollutant extremely toxic to plants and other living organisms including humans. To assess Pb phytotoxicity, experiments focusing on germination of wheat seeds were germinated in a solution containing Pb (NO(3))(2) (0.05; 0.1; 0.5; 1g/L) during 6 days. Lead accumulation in seedlings was positively correlated with the external concentrations, and negatively correlated with morphological parameters of plant growth. Lead increased lipid peroxidation, enhanced soluble protein concentrations and induced a significant accumulation of proline in roots. Esterase activity was enhanced in the presence of lead, whereas α-amylase activity was significantly inhibited. Antioxidant enzymes activities, such as, ascorbate peroxidase, peroxidase, superoxide dismutase, catalase and glutathione S-transferase were generally significantly increased in the presence of lead in a dose-dependent manner. The present results thus provide a model system to screen for natural compounds able to counteract the deleterious effects of lead.  相似文献

Summary The uptake of lead by roots and its transport to the shoots was examined with perennial ryegrass in solution cultures. Root uptake as measured by the decrease in concentration of lead in an aqueous solution containing 1 mg Pb/l as Pb (NO3)2 was rapid, almost complete, and unaffected by removing the shoots or killing the roots. Lead bound in the roots was not released by exchange with Ca or Ba ions. The distribution of lead within the plant was examined at intervals after a single, 3-day exposure to various levels of lead added to a nutrient solution. The total uptake, or lead burden, increased with increasing rates of addition and ranged from 281 to 9969 g/Pb per 3 plants. The proportion of the lead reaching the shoots at the first harvest (7 days after adding lead) was 3.5 to 22.7 per cent of total uptake, the lower value being for plants with the greatest burden. Transport to the shoots continued throughout the experimental periods of 21 and 28 days but did not exceed 28.9 per cent of total uptake. The concentration of lead in shoots at the first harvest ranged from 0.2 to 58.4 ppm and that in the corresponding roots from 5.5 to 5310 ppm. At later harvests, and after cutting, the concentration in the shoots decreased; an exception was in plants with the greatest lead burden. It is concluded that roots of actively growing ryegrass provide a barrier which restricts the movement of lead to the above-ground parts of plants, and so to animals or man.  相似文献

The effects of lead were investigated in bean plants (Phaseolus vulgaris L. cv. Zlota Saxa) grown hydroponically in nutrient solution and exposed to Pb(NO3)2 (0.1, 0.5, 1 mM) with or without equimolar concentrations of chelator ethylenediaminetetraacetic acid (EDTA). The roots treated only with Pb(NO3)2 accumulated up to 25 g(Pb) kg−1(d.m.), during 4-d exposure. However, in bean plants exposed to 0.5 mM Pb + 0.5 mM EDTA or 1 mM Pb + 1 mM EDTA 2.5 times less Pb was determined. In bean plants treated only with Pb, less than 6 % of total lead accumulated was transported to the aboveground parts, while in the case of plants grown with Pb + EDTA, around 50 % of total Pb was transported to the shoots.  相似文献

Root exudates of plants of Vitis vinifera L. cv. Thompson Seedless, grown in nutrient cultures with root temperatures maintained at either 20° or 30° and with shoots at a common air temperature, were assayed for cytokinin activity. After chromatography of freeze-dried sap on paper with n-butanol/acetic acid/water (4:1:1). activity was detected with a soybean callus assay. For both root temperatures, major activity appeared between RF 0.6 and RF 0.8, at about the same concentration in each case. The major difference between the 2 samples was the presence of activity at RF 0.1 to 0.2 in the 20° sample and its absence in the 30° sample.

The higher root temperature resulted in increased shoot and root elongation, increased dry matter accumulation by both shoots and roots, and also altered the morphological appearance of the roots.

The aim of this study was to determine the effect of exogenously applied Si on the growth and physiological parameters of sorghum [Sorghum bicolor (L.) Moench] cultivated in hydroponics with elevated zinc concentrations (75 μM and 150 μM Zn). Increased concentrations of Zn inhibited root growth and biomass production of roots and shoots. Application of Si individually showed a positive effect on root growth but negatively affected production of fresh and dry biomass of roots and shoots. On the other hand, silicon in combination with Zn significantly reduced the inhibitory effect of Zn on root growth but did not positively affect biomass production of roots and shoots. Accumulation of Zn in plant tissues increased with increasing Zn concentration in nutrient solution, but application of Si in combination with Zn did not significantly influence Zn accumulation in roots. Completely opposite results were found in Si accumulation in roots treated with Si in combination with Zn. Interaction of these ions resulted in considerable increase of Si accumulation in roots which almost doubled in comparison to individal Si treatment. Impact of Zn on the activity of some antioxidant enzymes was equivocal and differences were observed also between two Zn concentrations. Individual application of Si resulted in significant increase in the activity of all studied antioxidant enzymes but Si in combination with Zn mostly negatively affected their activity except the activity of catalase (CAT) which was the highest in roots grown in solution containing both Si and Zn ions. Comparing all obtained data we can assume that Si applied in combination with Zn did not significantly alleviate Zn toxicity in young sorghum except the growth of primary seminal root and further experiments are required for better understanding of their interaction.  相似文献

F1 hybrids of Sorghum bicolor (L.) Moench and their inbred parentswere analysed for NADH-nitrate reductase activity during theearly stages of seedling growth. In all the hybrids both mid-parentaland better parental heterosis were discernible in shoots whereasin roots two hybrids out of the three tested, showed heteroticlevels. The root system of wheat seedlings ( Triticum aestivum L. SUN 9E) was pruned to two seminal roots. One of the roots was supplied with different levels of NO3, the other was deprived of N. Root respiration and the increment of C and N in roots and shoots were measured to determine the C/N ratio of the phloem sap feeding the N-deprived roots. Thus it was possible to determine translocation of N from the shoots to the roots. It was calculated that the C/N ratio of phloem sap feeding roots of plants growing at optimal and suboptimal N supply was ca 54. A supra-optimal N supply reduced, whilst shading increased, the C/N ratio of phloem sap. At optimal N supply 11% of all N transported to the shoots was retranslocated to the roots. Both a supra-optimal and a limiting N supply increased translocation of N back to the roots to 18% of the N translocated to the shoot, whilst shading of the plants decreased the proportion cycled to 7%. At the optimal N supply, 40% more N was translocated to the roots from the shoot than was incorporated by them. At a lower supply of N, 80% more N was imported from the shoots than was incorporated by these roots. It is suggested that the distribution of N between roots and shoots predominantly occurs in the shoots. The specific mass transfer rate in seminal roots was determined. The highest value was found for roots grown with an optimal N supply: 1.1 mg carbohydrate s−1 cm−2 (sieve tube) which is well within the range observed for other plant organs. Roots supplied with NO3 produced more and longer laterals than N-deprived roots. It is suggested that this is due to the effect of NO3 on import of carbon and other components transported in the mass flow with carbon.  相似文献

利用聚乙二醇(PEG-6000)模拟水分亏缺条件(胁迫水势-0.2MPa,胁迫48h),研究了变水条件下紫花苜蓿(品种:阿尔冈金和陇东)和高粱(品种:抗四)根系水力学导度(Lpr)、根系活力、根叶相对含水量、水分利用效率等参数的动态变化,以期进一步明确植物水分吸收及散失过程调控的生理生态学基础。结果表明:水分亏缺限制了紫花苜蓿和高粱根系吸水,表现在Lpr的下降和根系活力的降低;继而调控了其地上部反应,引起气孔导度、光合速率、叶片相对含水量和蒸腾速率等的下降,但限制性的提高了其水分利用效率,尤其在胁迫初期。恢复到正常供水条件后,Lpr、根系活性、气孔导度等水分利用参数逐渐部分或完全恢复到了胁迫前水平,但恢复程度存在种间和品种间差异,并且根系吸水能力的恢复对于是植株地上部生长状态的恢复至关重要,尤其是水分恢复初期。紫花苜蓿根系中检测到水通道蛋白(AQPs)的存在,水分亏缺对紫花苜蓿Lpr的影响认为主要是通过影响AQPs的活性实现的。比较紫花苜蓿和高粱水分吸收与利用状况在变水条件下的动态变化,认为紫花苜蓿幼苗对干旱逆境的适应能力相对弱于高粱,品种间陇东适应能力更强。  相似文献
