A sensitive, simple spectrofluorometric technique for determination of tryptophan inamounts as small as 10 pmol is described. It is based on tryptophanase hydrolysis of tryptophan and spectrofluorometric analysis of the resulting indole. The relationship between released indole and fluorescence is linear over three orders of magnitude. The method is free from interference by other amino acids, polar indole derivatives, and a number of other compounds found in cell extracts or used in bacterial growth media. The method is rapid, reproducible, and accurate. A simple method for extraction and measurement of endogenous free tryptophan from bacterial cells is also described.  相似文献

The present work describes an alternative technique for following the rate of oxygen uptake by chemical and enzymatic systems. This method is based on spectrofluorometric monitoring of the well-known quenching effect of molecular oxygen on the emission of the photoexcited [Ru(bpy)3]2+ ion, added to the reaction mixtures. The rate of oxygen consumption determined using the present method agrees with that obtained by conventional polarographic techniques in all of the following systems: ascorbate/Cu2+, glucose/glucose oxidase (EC 1.1.3.4), and propanal/horseradish peroxidase (EC 1.11.1.7); in the last case, agreement was observed both in the presence and absence of serum albumin and of chloroplasts. Spectrofluorometric data for amphotericin autoxidation in dimethyl sulfoxide are in accord with the rate of decay of the ESR signal of a spin trap added to the reaction mixture. The advantages and limitations of the present spectrofluorometric technique relative to conventional polarographic monitoring of dissolved oxygen are discussed.  相似文献

Elevated lysosomal pH in neuronal ceroid lipofuscinoses (NCLs).   总被引:1,自引:0,他引:1  
We report here the intracellular (pHi) and lysosomal pH in fibroblasts of six forms of neuronal ceroid lipofuscinoses (NCLs). Acid extrusion rate and pH(i) values were measured by the membrane-permeant acetoxymethyl ester of the indicator dye, 2',7'-bis(carboxyethyl)-5-(and-6)-carboxy-fluorescein (BCECF) and lysosomal pH by a spectrofluorometric assay utilizing a novel acidotropic probe, Lysosensor yellow/blue. Intracellular pH was normal in all NCLs. Elevated lysosomal pH was detected in all NCL forms except CLN2 and CLN8. Elevated pH most probably disturbs the catalytic activity of lysosomes and is one important factor in explaining accumulation of ceroid and lipofuscin-like autofluorescent lipopigments characteristic of NCLs. Using the novel spectrofluorometric assay introduced in this study provides a fast and repeatable technique to measure intralysosomal pH from cell suspensions.  相似文献

A hairpin pyrrole polyamide conjugated to a Hoechst 33258 (Ht) analogue, PyPyPy-gamma-PyPyPy-gamma-Ht, was synthesized on solid-phase by adaptation of an Fmoc technique using a series of PyBOP/HOBt mediated coupling reactions. Sequence selectivity and complex stabilities were characterized by spectrofluorometric titrations and thermal melting studies. The polyamide of the conjugate was observed to bind in a hairpin motif forming 1:1 conjugate:dsDNA complexes. The conjugate is able to recognize nine contiguous A/T bps, discriminating from the sequences containing fewer than nine contiguous A/T bps.  相似文献

Green fluorescent protein (GFP) is a versatile reporter protein and has been widely used in biological research. However, its quantitation requires expensive equipment such as a spectrofluorometer. In the current study, a gel documentation imaging system using a native polyacrylamide gel for the quantitation of GFP was developed. The assay was evaluated for its precision, linearity, reproducibility, and sensitivity in the presence of Escherichia coli cells and was compared with the spectrofluorometric method. Using this newly established, gel-based imaging technique; the amount of GFP can be quantified accurately.  相似文献

We have developed a set of simple modifications of the green fluorescent protein (GFP)fragment reassembly assay in bacteria that permits: (i)fluorescent microscopy visualization of GFP reassembly only 1-2 h after induction of protein expression, thus approximating the detection of GFP reassembly to the real-time dynamics of protein complex formation in living cells; (ii) spectrofluorometric detection of reassembled GFP fluorescent signals directly in lysates from cell suspension thereby avoiding, in many cases, the need for tag-affinity isolation of protein complexes; and (iii) comparative quantification of signal intensity in numerous cell-sample lysates using a Bio-Rad IQ5 spectrofluorometric detection system (Bio-Rad Laboratories, Madrid, Spain). Collectively, the results demonstrate that the combination of microscopic and spectrofluorometric detection provides a time-saving and sensitive alternative to existing methods of fluorescence complementation analysis.  相似文献

The authors describe a novel spectrofluorometric technique based on double-labelled fluorescence imaging using immunoconjugates labelled with fluorochromes. Following isolation and characterization, cells are seeded on the surface of disks of the material(s) to be tested. After application of a primary antibody and an antibody bearing a fluorochrome, the signal emitted by the molecules in the extracellular matrix on the surface of the test disks is measured by spectrofluorimetry. Measurement is thus independent of the surface characteristics of the test material. Measured values are compared with pre-established standard curves. This technique facilitates determination of the characteristic molecules expressed by a given cell type,thus allowing accurate evaluation of the response of pertinent biological samples to implantable biomaterials. On-line estimation of biopolymer production during fermentation would be a useful adjunct to the development of strategies for process control and optimization. This study examined the applicability of spectrofluorometry, along with other on-line measurements, for the prediction of poly-ß-hydroxybutyric acid (PHB) concentrations in a high-cell density fed-batch fermentation of Ralstonia eutropha. Models previously used for modelling PHB evolution with time are not sufficiently accurate for situations where transient intermediate accumulations or PHB degradation occur. Thus, the mass balance in the model was modified to account for these situations. An estimation algorithm was developed that is based on a hybrid model consisting of a dynamic mass balance of PHB where the main reaction coefficient was regressed with respect to spectrofluorometric data. The regression between the kinetic parameter and the spectrofluorometric data was accomplished using partial least squares (PLS) regression to avoid high sensitivity to noise expected from highly correlated data, such as the spectrofluorometric readings. The model accounts for dynamics of intermediates and in this way allows the prediction of dynamic behaviour in PHB concentrations that cannot be predicted with other reported mathematical models.  相似文献

A spectrofluorometric assay was developed for quantification of bacterial biofilms grown on a microtiter plate. The method involved staining biofilms formed by gram-negative and gram-positive bacteria with wheat germ agglutinin-Alexa Fluor 488 conjugate, which selectively binds to N-acetylglucosamine residues in biofilms. The fluorescence of stained biofilms was measured with a fluorescent plate reader. This method was compared with a widely used microplate colorimetric assay involving crystal violet staining of biofilms formed by both gram-negative and gram-positive bacteria. A strong linear association existed between the two methods (r 2=0.99/0.94). Being more sensitive and specific as compared to colorimetric method, the spectrofluorometric assay provides a better alternative for quantification and characterization of bacterial biofilms.  相似文献

A highly sensitive spectrofluorometric method for quantitative estimation of certain precursors of chlorophyll biosynthesis from the mixtures of plant tetrapyrroles having overlapping fluorescence emission spectra is developed. At room temperature (293 degrees K) protoporphyrin IX is monitored from its emission maximum, 633 nm, when excited at 400 nm (E400/F633). Protochlorophyllide is estimated at 638 nm, while being excited at 440 nm (E440/F638). Mg-protoporphyrin+Mg-protoporphyrin monoester pool has emission around 589-592 nm. Therefore the integration value of the emission band that extends from 580 to 610 nm is taken to calibrate its concentration. This spectrofluorometric method designed for the determination of protoporphyrin IX, esterified and nonesterified Mg-protoporphyrin pool, and protochlorophyllide is far superior to available spectrophotometric methods and estimates as low as 1 nM concentration of plant pigments. As minute quantities of individual pigments can be quantitatively analyzed from their mixtures, this method eliminates analytical uncertainties due to recovery losses caused by chromatography. However, only dilute samples can be estimated by this spectrofluorometric method as the quantitative relation between fluorescence and concentration deviates from linearity at high, i.e., above 150 nM, concentrations of pigment to be quantified.  相似文献

Interaction of cationic dyes, pinacyanol chloride, acridine orange and phenosafranin, with Klebsiella K7 capsular polysaccharide has been investigated by spectrophotometric and spectrofluorometric measurements. The acidic polysaccharide induce a metachromatic blue shift of the absorption band of pinacyanol chloride from 600 nm to 495 nm, indicating strong metachromasy. Stoichiometry of polyanion and dye cation (1:1.5) in the polymer-dye compound formed by the interaction between pinacyanol chloride dye and K7 polymer indicate that both glucuronic acid and pyruvic acid act as the potential anionic sites for interaction. Both spectrophotometric titration of pinacyanol chloride and spectrofluorometric titration of acridine orange and phenosafranin dyes by the polymer gave quite comparable equivalent weights for the polymer. Dye-polymer interaction studies indicated induction of metachromasy in the cationic dye by the anionic biopolymer, establishing its chromotropic character.  相似文献

In vitro studies on the interactions between native herring sperm DNA (HS-DNA) and melamine as well as its related compounds (MARCs), that is, ammeline, ammelide, and cyanuric acid, have been investigated by spectrophotometric, spectrofluorometric, melting temperature, and viscosimetric techniques. It was found that any of the MARCs might interact with HS-DNA by a groove mode of binding via hydrogen bonds. The interaction constants between any of the MARCs and HS-DNA were at 10?-10? Brain slice preparations have become useful tools for studying multiple facets of normal brain function and for investigations of brain pathophysiology. Recently, a variety of neurological disorders have been linked to dysfunction of brain mitochondria. In this report we discuss optical methods for probing mitochondrial function in brain slices. Absorption spectrophotometric and spectrofluorometric techniques are described for measuring changes in the redox activity of mitochondrial cytochromes and the primary respiratory chain substrate nicotinamide adenine dinucleotide (NADH), respectively. A spectrofluorometric method is described also for measuring changes in mitochondrial membrane potential using the potential-sensitive fluorescent indicator JC-1. These methods used together have proven to be useful for studying dysfunction of mitochondria following in vitro ischemia in hippocampal slices, and might also be valuable for investigations of mitochondrial involvement in other neurological disorders.  相似文献

For quantitative estimation of coproporphyrin III and protoporphyrin IX from their mixture, a sensitive spectrofluorometric method was developed. At room temperature, coproporphyrin III fluoresces in neutral or alkaline pH at 622 nm having substantial fluorescence at 632 nm where protoporphyrin IX also fluoresces maximally. Similarly, protoporphyrin IX also has substantial fluorescence at 622 nm. Therefore, while estimating protoporphyrin IX (E400 F632) or coproporphyrin III (E400 F622) concentratton, it is essential to correct for the fluorescence due to coproporphyrin III at 632 nm and protoporphyrin IX at 622 nm. This was done by formulating equations from appropriate constants derived from pure samples of coproporphyrin III and protoporphyrin IX. As law as 1 pmole of coproporphyrin III or protoporphyrin IX could be estimated from their mixture by using the spectrofluorometric method.  相似文献

Serum activity of angiotensin converting enzyme (ACE) has been measured in 13 women (mean age 43.1 years) with primary hypothyroidism by spectrofluorometric method of Friedland and Silverstein. The mean enzyme activity was significantly lower in hypothyroid patients than in healthy persons.
