Third instar larvae of Heliothis virescens and Helicoverpa zea could be distinguished based on the hydrocarbons of their surface lipids. Hydrocarbons were the major components of the surface lipids and a distinctive capillary gas chromatographic profile could be obtained from a hexane extract of the surface lipids of a single larva. Analysis of hexane extracts of the surface lipids by capillary gas chromatography-mass spectrometry (CGC-MS) showed several obvious differences between the two species: (1) in their gas chromatographic profiles; (2) in the presence of a major alkene, hentriacontene, only in H. zea; (3) in H. virescens the CGC-MS peak with a KI of approximately 2860 was 2-methyloctacosane, but in H. zea it was a mixture of 4-methyloctacosane plus 9,13- and 8,12-dimethyloctacosanes; and (4) in the methyl branch positions of dimethyl-branched alkanes with carbon backbones of C31, C33, C35, C45, C47, C49 and C51. The methyl branch points of H. virescens dimethylalkanes were separated by seven or nine methylenes, while in H. zea the methyl branch points of the dimethylalkanes were separated by three and sometimes five methylenes.  相似文献

The structural basis of orientation stability was investigated. The stable unipolar orientation of the Melanoplus sanguinipes X-chromosome univalent is unique in that it is stable without tension created by forces towards opposite poles; tension is thought to be the principle component in stabilizing kinetochore orientations to a pole. Stable orientation of the X chromosome in Melanoplus sanguinipes was compared with unstable X orientation in Melanoplus differentialis. Ten cells (five of each species) were studied, firstly in living cultures where chromosome behavior was followed, then by serial-section electron microscopy where the structural basis for chromosome behavior was examined. Microtubules other than kinetochore microtubules were observed impinging on the X chromosomes. One end of these microtubules was buried in chromatin, while the other ran towards a pole. The X chromosomes of M. sanguinipes had more of these microtubules than did M. differentialis X chromosomes. It is suggested that M. sanguinipes X chromosomes are less condensed than M. differentialis X chromosomes and so allow more microtubules to penetrate the chromosome. The extra microtubules impinging on the M. sanguinipes X chromosome probably prevent reorientation by inhibiting the turning of the chromosome towards the opposite pole, i.e., more force is needed to turn a kinetochore towards the opposite pole than can be generated and attempts at reorientation fail. This may be analogous to the effect that tension has on the orientation stability of bivalents.  相似文献

Dietary hydrocarbons are incorporated into cuticular lipids of the grasshopper Melanoplus sanguinipes. Dietary secondary alcohols and ketones, however, are not incorporated into the cuticular lipids. In typical experiments from 8 to 28 per cent of the fed labeled n-alkanes are recovered in the cuticular lipids. Most of the radioactivity recovered from feeding the C23 n-alkane and a significant amount from the C25 was found as a secondary alcohol in the form of a wax ester. The C29 and C31 n-alkanes were recovered primarily unchanged as the n-alkane. Eighty-five per cent of injected acetate incorporated into the hydrocarbon fraction is in the branched hydrocarbons. These results show that the insect synthesizes its branched hydrocarbons, whereas a large part of the normal hydrocarbons can be dietary.  相似文献

The roles of perception, development, experience, and physiology on the swimming behavior of grasshoppers in an arena were investigated usingMelanoplus bivittatus, M. differentialis, andM. sanguinipes. The parameters of swimming included capacity (the ability to reach the edge of a 45-cm-diameter water basin within 3 min), net displacement time (time required to reach the edge of the arena), and orientation (ability to make initial contact with a target pattern).Melanoplus sanguinipes adults were most attracted to a vertical rectangle set at 90° to the water surface (the standard target). Orientation ability decreased significantly as the target became square or it declined to 60°, and there was virtually no orientation to horizontal patterns or those at 45°. All nymphal instars ofM. sanguinipes exhibited the capacity to swim and orient to a standard target. Although the net displacement time was lowest from the third instar through the adult stages, the earlier instars (first through fourth) had the greatest relative net displacement rates. As adults aged, their net displacement time and orientation ability declined significantly, particularly in males. BothM. differentialis andM. sanguinipes adults showed a decline in net displacement time over a 10-day trial period. The loss of a hind leg did not significantly alter swimming performance of eitherM. bivittatus orM. sanguinipes adults, but this alteration significantly reduced the net displacement time ofM. differentialis. DecerebratedM. differentialis adults were capable of swimming for several minutes.  相似文献

Chemistry of Dufour glands associated with the venom complex in Bracon cephi (Gahan) and Bracon lissogaster Muesebeck (Hymenoptera; Braconidae), two parasitoids of the wheat stem sawfly, Cephus cinctus Norton (Hymenoptera: Cephidae), was examined by solid phase micro-extraction (SPME) and gas chromatography/mass spectrometry. Homologous series of five chemical classes were detected in individual glands from each species. Major classes included: (1) acetate esters of saturated and unsaturated primary alcohols with parent chain lengths from C12 to C20. Hexadecanyl acetate, octadecanyl acetate, and octadecenyl acetate were major components in B. cephi. The composition of the acetate series in B. lissogaster was similar except that the octadecanyl acetate was only a minor component. (2) A homologous series of monoenes from C23:1 to C35:1 were detected in both species, with C29:1, C31:1 and C33:1 being the major components. Dienes from C31:2 to C35:2 and trienes (C33:3–C35:3) were also detected in both species. (3) A homologous series of n-alkanes from C19 to C31 was detected in both species. n-Tricosane was the major component. Minor components in both species included homologous series of both mono- and dimethyl branched alkanes. The Dufour gland hydrocarbon components in both B. cephi and B. lissogaster have some similarities to the composition of cuticular hydrocarbons of their host C. cinctus, a species with a complex pheromonal signaling system.  相似文献

The interaction between M (M?=?Ca, Yb) atom and C74 (D 3h) has been investigated by all electron relativistic density function theory. With the aid of the representative patch of C74 (D 3h), we studied the interaction between C74 (D 3h) and M (M?=?Ca, Yb) atom and obtained the interaction potential. Optimized structures show that there are three equivalent stable isomers and there is one transition state between every two stable isomers. According to the minimum energy pathway, the possible movement trajectory of M (M?=?Ca, Yb) atom in the C74 (D 3h) cage is explored. The calculated energy barrier for Yb atoms moving from the stable isomer to the transition state is 10.4 kcal mol?1 and the energy barrier for Ca atoms is 6.1 kcal mol?1. The calculated NMR spectra of M@C74 (M?=?Ca, Yb) are in good agreement with the experimental data. There are nine lines in the spectra: one 1/6 intensity signal, four half intensity signals and four full intensity signals.  相似文献

The Wittig reaction between alkylidene triphenylphosphoranes [R — CH = P(C6H5)3, where R varied from H to n — C17H35] and methyl 12-oxooctadecanoate or methyl 10-oxohexadecanoate in dimethylformamide (DMF) has been employed in the synthesis of a partial homologous series of esters of branched chain fatty acids in high yields. The effect of various ratios of reactants in both DMF and dimethylsulfoxide (DMSO) was investigated. Purification from triphenylphosphine oxide was readily accomplished by chromatography on a column of silicic acid-Celite impregnated with silver nitrate.  相似文献

The proteinaceous haemagglutinin (lectin) present in the haemolymph of Melanoplus sanguinipes (F) has been isolated and biochemically characterized. The protein was purified to homogeneity by affinity chromatography on a column of Sepharose-galactose. The purified haemagglutinin accounted for all observed haemolymphatic haemagglutination activity. Gel filtration and electrophoresis showed the haemagglutinin to be a 600–700,000 mol. wt noncovalent aggregate of 70,000 mol. wt subunits. The 70,000 subunit contained two disulfide-linked polypeptide chains of mol. wt 40,000 and 28,000, respectively. The purified haemagglutinin contained all the common amino acids and had highest amounts of acidic residues and least amounts of methionine and glucosamine. Haemagglutination activity was stable at −20°C in the presence of d-galactose but was destroyed by treatment of the haemagglutinin with trypsin, heat or EDTA. Haemagglutination inhibition studies showed that low concentrations (≤5 mM) of either d-galactosidic or d-glucosidic carbohydrates were bound by the haemagglutinin and inhibited agglutination of human asialo-erythrocytes. The purified haemagglutinin was antigenic in rabbits. Comparative experiments showed that the haemagglutinin from M. differentialis (Thomas) was identical to that from M. sanguinipes.  相似文献

Decline and disappearance of a natural population of the grasshopper Hesperotettix viridis pratensis was related to severe infection by Mermis nigrescens. In contrast the numbers of slightly infected Melanoplus bivittatus did not decrease. Uninfected M. sanguinipes, M. differentialis and M. fernur-rubrum also did not decrease. The high percentage of infection in H. viridis pratensis was related to low, wet habitat, where the grasshopper fed primarily on Solidago missouriensis; infected individuals failed to develop ovaries or mature testes. This is believed to be the first reported occurrence of a nematode parasitizing H. viridis pratensis. In juvenile M. nigrescens the unreported shape of the stoma, the stylet shape and paired oval structures in the cerebral region were photographed. Factors affecting biological control of grasshoppers by using M. nigrescens were discussed.  相似文献

Material from the testa of decorticated barley grains contained hydrocarbons, esters, triglycerides, free sterols, 5-n-alkylresorcinols, and traces of free alcohols, carbonyl compounds, and various polar, acidic materials. The hydrocarbon fraction was mainly a series of n-alkanes, extending at least from C11 to C36, in which the C29 and C31 components were prominent. Two minor series of alkanes were also present. Sometimes a trace of an unsaturated hydrocarbon was detected. The ester fraction contained sterols and alkanols esterified by fatty acids, which differed in relative amounts from the fatty acids found in the triglycerides. The triglycerides were thought to have leached from within the grain. At least five free sterols were present, including sitosterol and campesterol. The 5-n-alkylresorcinols were at least twelve members of a homologous series, of which four, C25, C27, C29, and C31, made 98% of the total. Members of the series with even numbers of carbon atoms were also present. It is suggested that they are partly responsible for excluding microorganisms from the interior of the grain. The testa membrane, with the associated pigment strand, contained an estolide of fatty acids and various hydroxyacids, a polysaccharide component, and uncharacterized material.  相似文献

The increasing CO2 concentration in Earths atmosphere is expected to cause a greater decline in the nutritional quality of C3 than C4 plants. As a compensatory response, herbivorous insects may increase their feeding disproportionately on C3 plants. These hypotheses were tested by growing the grasses Lolium multiflorum C3) and Bouteloua curtipendula C4) at ambient (370 ppm) and elevated (740 ppm) CO2 levels in open top chambers in the field, and comparing the growth and digestive efficiencies of the generalist grasshopper Melanoplus sanguinipes on each of the four plant × CO2 treatment combinations. As expected, the nutritional quality of the C3 grass declined to a greater extent than did that of the C4 grass at elevated CO2; protein levels declined in the C3 grass, while levels of carbohydrates (sugar, fructan and starch) increased. However, M. sanguinipes did not significantly increase its consumption rate to compensate for the lower nutritional quality of the C3 grass grown under elevated CO2. Instead, these grasshoppers appear to use post-ingestive mechanisms to maintain their growth rates on the C3 grass under elevated CO2. Consumption rates of the C3 and C4 grasses were also similar, demonstrating a lack of compensatory feeding on the C4 grass. We also examined the relative efficiencies of nutrient utilization from a C3 and C4 grass by M. sanguinipes to test the basis for the C4 plant avoidance hypothesis. Contrary to this hypothesis, neither protein nor sugar was digested with a lower efficiency from the C4 grass than from the C3 grass. A novel finding of this study is that fructan, a potentially large carbohydrate source in C3 grasses, is utilized by grasshoppers. Based on the higher nutrient levels in the C3 grass and the better growth performance of M. sanguinipes on this grass at both CO2 levels, we conclude that C3 grasses are likely to remain better host plants than C4 grasses in future CO2 conditions.  相似文献

Feeding by three grasshopper species, Camnula pellucida, Melanoplus packardii and Melanoplus sanguinipes, on three safflower (Carthamus tinctorius) lines for a 6-wk period from anthesis was monitored under field conditions. Ratings of feeding damage to different plant parts (leaves, floral parts, capitula, and peduncles) and measurements after termination of feeding (dry weight, seed yield, seed weight, seeds per capitulum, and capitula per row) were compared among grasshopper species and safflower lines. The Melanoplus species fed preferentially on leaves, floral parts, and capitula, while C. pellucida exhibited only peduncle feeding, which resulted in head clipping. Defoliation of 20 to 30% was associated with significant increases in total dry matter, seed yield, and number of capitula. Further defoliation resulted in decreases. The safflower lines differed in response to grasshopper feeding. S-208 was most susceptible to defoliation by grasshopper feeding, exhibiting decreased dry weight, seed yield, and capitula number. Lesaf 34C-00 was most tolerant and only M. packardii caused significant dry weight and seed yield reductions. Feeding by C. pellucida on this line resulted in an overall seed yield increase. Feeding by M. sanguinipes on Seedtec-5 resulted in yield increases of up to 16%. It appears that certain grasshopper species can increase seed yield in some safflower lines by stimulating the production of additional capitula. Therefore, moderate populations of such grasshoppers in fields of appropriate safflower cultivars do not necessarily require control.  相似文献

Apolipophorin III has been isolated and characterized from two orthopteran species, Barytettix psolus and Melanoplus differentialis. Both apoLp-IIIs have Mr ∼ 20,000 and contain 5% covalently bound sugar. Compositional analysis of the carbohydrate component of these apoLp-IIIs, as well as Locusta migratoria apoLp-III, revealed the presence of manose, fucose and N-acetyl glucosamine. In all three proteins the ratios of these sugar residues was similar (∼2:2:1). Amino acid analysis of B. psolus and M. differentialis apoLp-IIIs was performed and the results compared with those from L. migratoria and M. sexta apoLp-IIIs. The three orthopteran apoLp-IIIs possessed very similar compositions that were distinct from that of M. sexta. Similarly, N-terminal sequence analysis revealed ∼50% sequence identity between the three orthopteran apoLp-IIIs but only 9–18% identity was observed when B. psolus or M. differentialis apoLp-IIIs were compared to M. sexta apoLp-III. The implications of these findings, with respect to flight-related physiology and adipokinetic hormone stimulated lipid mobilization in these different species, is discussed.  相似文献

Four homologous series of very long-chain methyl-branched alcohols were found in the internal lipids of cabbage looper, Trichoplusia ni, pupae both as free alcohols and as esters. These alcohols were identified based on mass spectra of their TMS and acetate derivatives, and on the Kovats Indices and mass spectra of the alkanes obatained by reduction of their bromide derivatives with LiAlH4. The four homologous series (from C36 to C46) consisted of monomethyl-, two dimethyl- and a trimethyl-branched alcohol series. The major alcohols (with the corresponding alkanes in parentheses) were identified as 24-methyltetracontan-1-ol (17-methyltetracontane), 24,28-dimethyltetracontan-1-ol (13,17-dimethyltetracontane), 24,36-dimethyltetracontan-1-ol (5,17-dimethyltetracontane) and 24,28,36-trimethyltetracontan-1-ol (5,13,17-trimethyltetracontane). The minor components had an odd number of carbon atoms (C37, C39, C41 and C43) in the carbon chain and the methylalkanes formed from their reduction had methyl branches on the 18- and 14,18-positions. The methylalkanes found internally in pupae were similar to those previously reported in larval cuticular lipids (de Renobales and Blomquist, Insect Biochem. 13, 493–502, 1983). Additional novel methylalkanes identified were 2,X-dimethylalkanes, 5,15-dimethylhentriacontane, 5,23-dimethyltritriacontane, 5,17,23-trimethyltritriacontane and 5,15,23-trimethylpentatriacontane. The methyl branching positions of the major methyl-branched alcohols were different from those of the major methylalkanes.  相似文献

The SinI and EcoRII DNA methyltransferases recognize sequences (GGA/TCC and CCA/TGG, respectively), which are characterized by an A/T ambiguity. Recognition of the A·T and T·A base pair was studied by in vitro methyltransferase assays using oligonucleotide substrates containing a hypoxanthine·C base pair in the central position of the recognition sequence. Both enzymes methylated the substituted oligonucleotide with an efficiency that was comparable to methylation of the canonical substrate. These observations indicate that M.SinI and M.EcoRII discriminate between their canonical recognition site and the site containing a G·C or a C·G base pair in the center of the recognition sequence (GGG/CCC and CCG/CGG, respectively) by interaction(s) in the DNA minor groove. M.SinI mutants displaying a decreased capacity to discriminate between the GGA/TCC and GGG/CCC sequences were isolated by random mutagenesis and selection for the relaxed specificity phenotype. These mutations led to amino acid substitutions outside the variable region, previously thought to be the sole determinant of sequence specificity. These observations indicate that A/T versus G/C discrimination is mediated by interactions between the large domain of the methyltransferase and the minor groove surface of the DNA.  相似文献

The cuticular hydrocarbons (CHs) and morphology of two Ceratitis rosa Karsch (Diptera: Tephritidae) populations, putatively belonging to two cryptic taxa, were analysed. The chemical profiles were characterised by two-dimensional gas chromatography with mass spectrometric detection. CHs of Ceratitis rosa that originated from the lowlands and highlands of Kenya comprised of n-alkanes, monomethylalkanes, dimethylalkanes and unsaturated hydrocarbons in the range of the carbon backbone from C14 to C37. Hydrocarbons containing C29, C31, C33 and C35 carbon atoms predominated in these two populations. 2-Methyltriacontane was the predominant compound in both populations. Quantitative differences in the distribution of hydrocarbons of different chain lengths, mainly the C22, C32, C33 and C34 compounds of these two populations, were observed despite indistinct qualitative differences in these hydrocarbons. Morphological analyses of male legs confirmed that the flies belong to different morphotypes of Ceratitis rosa previously labelled as R1 and R2 for lowland and highland populations, respectively. A statistical analysis of the CH compositions of the putative R1 and R2 species showed distinct interspecific identities, with several CHs specific for each of the lowland and highland populations. This study supports a hypothesis that the taxon Ceratitis rosa consists of at least two biological species.  相似文献

In this work, cuticular waxes from flag leaf blades and peduncles of Triticum aestivum cv. Bethlehem were investigated in search for novel wax compounds. Seven wax compound classes were detected that had previously not been reported, and their structures were elucidated using gas chromatography-mass spectrometry of various derivatives. Six of the classes were identified as series of homologs differing by two methylene units, while the seventh was a homologous series with homologs with single methylene unit differences. In the waxes of flag leaf blades, secondary alcohols (predominantly C27 and C33), primary/secondary diols (predominantly C28) and esters of primary/secondary diols (predominantly C50, combining C28 diol with C22 acid) were found, all sharing similar secondary hydroxyl group positions at and around C-12 or ω-12. 7- and 8-hydroxy-2-alkanol esters (predominantly C35), 7- and 8-oxo-2-alkanol esters (predominantly C35), and 4-alkylbutan-4-olides (predominantly C28) were found both in flag leaf and peduncle wax mixtures. Finally, a series of even- and odd-numbered alkane homologs was identified in both leaf and peduncle waxes, with an internal methyl branch preferentially on C-11 and C-13 of homologs with even total carbon number and on C-12 of odd-numbered homologs. Biosynthetic pathways are suggested for all compounds, based on common structural features and matching chain length profiles with other wheat wax compound classes.  相似文献

Laboratory experiments were conducted to measure the feeding rates, preferences, survival rates, maturation times, and weight gain of four common species of grasshoppers (Orthoptera: Acrididae) on four lines of safflower (Carthamus tinctorius). When safflower lines were presented individually, all were readily consumed although consumption differed significantly according to grasshopper species, age and sex, and to safflower line. Consumption was greatest on a safflower line devoid of spines. However, grasshoppers also fed readily on spiny lines, and results of experiments in which all safflower lines were presented together indicated preferences that did not necessarily correspond to the results of the single-line feeding trials. Camnula pellucida, a graminivorous species, did feed on safflower, but restricted feeding almost exclusively to stem cambium, whereas Melanoplus bivittatus, M. packardii and M. sanguinipes fed mainly on the heads, flowers and leaves. Highly significant differences in grasshopper maturation time and weight gain were apparent among grasshopper species, and among safflower lines. Seedtec-5, the line which was least digestible, least consumed and least preferred nevertheless yielded rapid development and the greatest body weights of grasshoppers. All species of Melanoplus developed more quickly and gained more weight on safflower than on wheat. One species, M. packardii, was more able to utilise safflower than its congeners, and may gain competitive advantage if safflower becomes widely grown.  相似文献

Entomopoxvirus (EPV) occlusion bodies isolated from Arphia conspersa and Melanoplus sanguinipes grasshoppers were fed to 3rd and 4th instar Locusta migratoria nymphs. Locus mortality induced by A. conspersa EPV was first detected 18 days after addition of virus to the diet, and reached a level of approximately 68% of the colony population by 60 days after virus inoculation. In a similar population of L. migratoria nymphs, mortality induced by M. sanguinipes virus reached 90% 60 days after virus inoculation. Entomopoxvirus was isolated from M. sanguinipes EPV infected locust nymphs and the viral DNA was cleaved with several restriction endonucleases. The DNA fragment patterns obtained after agarose gel electrophoresis were compared with the fragment patterns from the original sample of M. sanguinipes EPV DNA cleaved with the same restriction endonucleases. No differences in the cleavage patterns were detected between the two virus DNA samples. Virus structural proteins of M. sanguinipes EPV purified from infected locust nymphs were compared by polyacrylamide gel electrophoresis with virus proteins isolated from the original sample of M. sanguinipes EPV. A total of six different virus protein bands were detected between the two poxvirus preparations.  相似文献
