Summary The characteristics of recombination of several petite (rho -) mutants of S. cerevisiae that retain the -influenced region of the mitochondrial genome, identified by the markers cap1-r, ery1-r and tsr1, are described. The petites were derived from an grande (rho +) strain and those petites which retain all three markers show recombination properties similar to those of the - parental strain. However, other rho - mutants that retain the cap1 and ery1 loci but have lost the tsr1 locus, which is located between cap1 and ery1, show markedly different properties of mitochondrial transmission and recombination, consistent with the presence of + alleles. The association of an internal deletion between the cap1 and ery1 loci with a change in phenotype provides additional evidence for the location of between these two loci.Although the petites deleted for the tsr1 locus exhibited the recombination properties of + strains, it was not possible to transmit this characteristic to rho + recombinant cells. Experiments on the kinetics of elimination by ethidium bromide of the cap1 and eryl markers from the petites and measurements of the buoyant densities of their mtDNA species did not indicate major changes (such as selective sequence repetition) in the sequences of the mtDNAs. The possible nature of the changes in the mtDNAs of these petites is discussed in light of recent studies on the physical nature of the alleles.  相似文献

Summary In order to study the effects of strainploidy on the transmission and recombination of the mitochondrial genes C, E and O conferring the resistance to chloramphenicol, erythromycin and oligomycin, respectively, haploids were crossed to diploids and the results of genetic analysis were compared with those from haploidxhaploid crosses. All haploidx diploid crosses showed an increased transmission of diploid derived alleles, relative to haploid derived ones, but the pattern of increase differed between homosexual and heterosexual crosses. In haploid x diploid homosexual crosses, the increase was of roughly equal magnitude at the C, E and O loci: there was a polar co-transmission of the diploid derived alleles. In + haploid x diploid heterosexual crosses, on the contrary, a differential increase was observed at the different loci, the magnitude being the smallest at the C locus and the largest at the O locus. As a result, there was a preferential transmission in favor of the haploid derived C alleles and of the diploid derived O alleles. A near equal transmission from both parents was observed for the E alleles. A decrease and an increase in the recombination frequency were noticed in the above haploidxdiploid homosexual and heterosexual crosses, respectively.The above phenomena were ascribed to different dosages of mitochondrial genomes from parents. Experimental data were well accorded with the theoretical expectations which were obtained on the assumptions that diploids contain twice as many mitochondrial genomes as haploids, and that random pairing and recombination would occur among mitochondrial genomes from parents. The elevation of strain-ploidy did not affect the recombination polarity which is under the control of the gene.It was theoretically predicted that a preferential transmission in favor of diploid derived alleles at all the C, E and O loci would be seen in haploid x + diploid heterosexual crosses as well as in +; haploid x +; diploid homosexual crosses, but that the magnitude of the polar transmission would vary depending upon the loci in the former crosses, while it would be the same at all the loci in the latter ones. The recombination frequency was predicted to decrease in both of these crosses.  相似文献

Summary The sequence organization of the yeast mit-DNA region carrying the large ribosomal RNA gene and the polar locus was examined. Hybridization studies using rho- deletion mutants and electron microscopy of the heteroduplexes formed between 23S rRNA and the appropriate restriction fragments, lead to the conclusion that the 23S rRNA1 gene of the + strains is split by an insertion sequence of 1,000–1,100 bp. In contrast, no detactable insertion was found in the 23S rRNA gene of the - strains. The size and the location of the insert found in the 23S rRNA gene of the + strains appear to be identical to those of the sequence which had previously been found to characterize the difference (at the locus) between the mitDNA of the wild type strains carrying the + or - alleles (Jacq et al., 1977).  相似文献

Summary A series of mutants called ebi, less inducible by ethidium bromide than the parental strain for the + mutation have been isolated after E.M.S. mutagenesis. Some of the ebi mutants also show an important accumulation of cells, in the absence of ethidium bromide. Ebi mutations are nuclearly inherited as shown by meiotic segregation. The effects of these mutants on the transmission and recombination of mitochondrial genes among the diploid progeny of crosses have been studied. Some of the ebi mutants show a non coordinated transmission of the oli1 mitochondrial marker with respect to other mitochondrial markers unexpected for homosexual crosses. This bias which is independent from will be discussed in relation to the segregation and recombination. No significant decrease of the frequency of recombinants has been detected.Abbreviations E.B. The dynamics of coupled biological oscillators can be modeled by averaging the effects of coupling over each oscillatory cycle so that the coupling depends on the phase difference between the two oscillators and not on their specific states. Average phase difference theory claims that mode locking phenomena can be predicted by the average effects of the coupling influences. As a starting point for both empirical and theoretical investigations, Rand et al. (1988) have proposed d/dt= — K sin ), with phase-locked solutions =arcsin( /K), where is the difference between the uncoupled frequencies and K is the coupling strength. Phase-locking was evaluated in three experiments using an interlimb coordination paradigm in which a person oscillates hand-held pendulums. was controlled through length differences in the left and right pendulums. The coupled frequency c was varied by a metronome, and scaled to the eigenfrequency v of the coupled system K was assumed to vary inversely with c. The results indicate that: (1) and K contribute multiplicatively to (2) =0 or = regardless of K when =0; (3) 0 or regardless of when K is large (relative to ); (4) results (1) to (3) hold identically for both in phase and antiphase coordination. The results also indicate that the relevant frequency is c/v rather than c. Discussion high-lighted the significance of confirming =arcsin(/K) for more general treatments of phase-locking, such as circle map dynamics, and for the 11 phase-entrainment which characterizes biological movement systems.  相似文献

1.  A series of CS revertants has been selected from various strains (both + and ) carrying a CR mitochondrial mutation at the RIB1 locus. The properties of mitochondrial recombination exhibited by these CS revertants in various crosses, have been examined systematically. The allele of the CS revertants has been defined in crosses with + and tester strains using two criteria: the polarity of recombination and a new criterium called relative output coefficient. We found that mutations of appear frequently associated with the mutations at the RIB1 locus selected from strains but not with those selected from + strains. A new allelic form of (n) which had not been found amongst wild type yeast strains is characterised. Similarly n mutation was found frequently associated with CR mutants at the RIB1 locus selected from CS strains but not with those selected from + CS strains. The n mutants, and the + and strains, explain the groups of polarity previously observed by Coen et al. (1970).
2.  Main features of mitochondrial crosses with n strains (+×n, ×n and n×n) are analysed. Recombination is possible between the different mitochondrial genetic markers. No high polarity of recombination is observed and the frequency of recombinants are similar to those found in homosexual crosses (+×+ and ×). A striking property, observed for the first time, exists in crosses between + +×n CS strains and some CREO mutants: the CREO are unable to integrate by recombination their CR allele into the + mit-DNA of CS strains while being capable of integrating it into + CS or CS genomes.
3.  It is proposed that the locus is the site of initiation of non reciprocal recombination events, the +/ pairing specifically initiates the non-reciprocal act while +/n or /n pairings do not.
4.  The molecular nature of the n mutation and its bearing on the structure of the locus are discussed. It is suggested that n mutations correspond to macrolesions (probably deletions) of a segment of the mit-DNA covering the and RIB1 loci. If n is a partial deletion of the sequence the + could be an additionnal deletion of the n sequence.
5.  The occurrence of spontaneous CR and ER mitochondrial mutations has been analysed by the Luria and Delbrück fluctuation test in and n isonuclear strains. Results of these tests indicate that an intracellular selection of resistant copies preexisting the action of the antibiotic occurs.

Summary The metabolic formation of either,-dodecanedioic acid or,-tridecanedioic acid from the individual n-alkane, n-alcohol, n-monoacid and,-diol with corresponding carbon chain length using K-carrageenan entrapped mutants S76 ofCandida tropicalis was studied. The immobilized cells of S76 could also directly produce-hydroxy acid and,-dioic acid from,-diol. With n-alcohol and n-monoacid as substrate, the amount of-hydroxy acid and,-dioic acid produced was also a function of the incubation time.The results demonstrated that in the immobilized cells of S76 the formation of,-dioic acid from n-alcohol can also run both via n-monoacid and via,-diol as well as in the normal cells of S76.  相似文献

An algorithm of learning in multilayer threshold nets without feedbacks is proposed. The net is. built of threshold elements with binary inputs. During a learning process each input vector x is accompanied by a teacher's decision ({1,...,M}). The pairs (x[n], [n]) appear in successive steps independently according to some unknown stationary distribution p(x,). The problem of learning of a threshold net has been decomposed to a series of problems of learning of the threshold elements. The proposed learning algorithm of the threshold elements has a perceptron-like form. It was proven that a decision rule of the threshold net stabilizes after a finite number of steps. For definite classes {p(x, )} * K of distributions p(x,), an optimal decision rule stabilizes after a finite number of steps. These classes {p(x, )} * K also contain distributions describing learning processes with perturbations.  相似文献

Biofilms were allowed to develop on wooden slides of the River Red Gum (Eucalyptus camaldulensis Dehnh., Myrtaceae) submerged in two billabongs of south-eastern Australia. The slides were placed in the photic zone and the aphotic zone, and the biofilms sampled after eight week's growth over the summer of 1989–1990 and winter of 1990. Bacterial numbers, estimated with epifluorescence microscopy, ranged from 4–78 × 106 cells cm–2. Bacteria were more abundant in the photic zone than the aphotic zone, and more abundant in summer than winter. Fewer than 0.5% of the bacteria could be cultivated on nutrient agar plates. Concentrations of phospholipids ranged from 8–79 ng cm–2, which corresponded to bacterial abundances of 2–17 × 106 cells cm–2. Fifty five phospholipid fatty acids (PLFA) were identified, of which 16:0 (13–29% of total PFLA) was the most common. Other abundant PFLA included 16:17c (6–28%), 18:26 (3–16%), 18:33 (4–12%), 18:19c (3–5%), 18:l7c (5–11%) and 18:0 (2–8%). Minor PLFA included 14:0, i and a 15:0, 15:0, 16:l5c, 16:113c, 18:36, 18:43, 20:46 and 20:53. The PLFA profiles of the biofilms were quite different from those of the sediments and plankton. There was a clear distinction between the PLFA profiles of summer and winter biofilms, but less evidence for unequivocal site or light-regime effects.  相似文献

Summary The progenies of two different rye test-crosses were analyzed for secalin proteins by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) using unreduced and reduced aqueous ethanol extracts. Segregation for two high-molecular-weight secalin bands (Glu-R1 or Sec3), one -secalin band (Gli-R1 or Sec-1), two 40K -secalin bands (Gli-R1 or Sec1) and two -type secalin bands (new locus) were studied. One recombinant between - and -secalins was found in one test-cross. The new locus, designated Gli-R3 or Sec-4, was mapped between Glu-R1 and Gli-R1, more displaced towards Gli-R1. In test-cross 1 recombination between Glu-R1 and Gli-R3 was 33.80±3.22%, and between Gli-R3 and Gli-R1, 12.04±2.21%. In the other test-cross the map distances were relatively similar but smaller, likely due to less recombination within two different species of Secale. Genes coding for 40K -secalins at Gli-R1 were likely proximal to the centromere with respect to genes coding for -secalins at the same complex locus.  相似文献

Summary Mitochondrial DNAs from six - mutants carrying the genetic locus Ribl and deleted for the rest of the genome were analyzed. Distribution of circular molecules from one mutant followed exactly the frequency rule, 1/n, for multimers with discreet classes n, 2n, 3n, etc. Another, genetically unstable mutant displayed a continuous spectrum of circular molecules of various lengths. Four other mutants contained multiple series of circular molecules. Partial denaturation maps show that the mutants analyzed show a common segment ca. 1.0 m long and differ by characteristic deletions of extremites of this segment. Short terminal deletions of the right i.e. pointing towards the Rib3 locus, terminus of this segment are correlated with modifications of the recombination properties related to the locus.  相似文献

Characteristic of [125I]-conotoxin (-CgTX) labeling using bifunctional cross linker (dithio bis[succinimidyl propionate]: DSP) was systematically investigated in crude membranes from chick whole brain. [125I]-CgTX specifically labeled 216 kDa as a main and 236 kDa as a minor bands in the crude membranes under non-reduced condition, but not labeled under reduced condition. We investigated the effect of various Ca channel antagonists on [125I]-CgTX labeling with DSP in detail, and found that there is a strong correlation between the effects of Ca channel antagonists on [125I]-CgTX labeling of the 216 kDa band and specific [125I]-CgTX binding. These results suggest that labeling of the 216 kDa band under non-reduced condition with [125I]-CgTX using DSP involves the specific binding sites of [125I]-CgTX, perhaps including one of the neuronal N-type Ca channel subunits in the crude membranes.  相似文献

Omega-conotoxin GVIA (-CTX), as a selective blocker for an N-type Ca2+ channel, has been conveniently used in many molecular biochemical and pharmacological experiments. There has been little elucidation of 125I--CTX binding sites (mainly the 135-kDa band) in the crude membranes from chick brain, although the characteristics of specific 125I--CTX binding and labeling sites in chick brain membranes have been investigated in our previous research. In this work, our goal is to further identify 125I--CTX labeling sites in chick brain membranes by using anti-B1Nt antibodies (against the N-terminal segment B1Nt of N- or P-type Ca2+ channel 1-subunits). The 125I--CTX–labeled sites in chick brain membranes could be solubilized and immunoprecipitated by using an anti B1Nt antibody. The molecular weight of the immunoprecipitated protein was determined as 135 kDa, which is inconsistent with that of the specific 125I--CTX binding protein reported previously. Moreover, the 125I--CTX–labeled protein could be purified by the method of preparative SDS-PAGE and recognized by anti-B1Nt antibodies in Western blotting analysis. These results indicated that anti-B1Nt antibodies could truly recognize 125I--CTX–labeled sites as the main band of 135 kDa from chick brain membranes, and the -CTX–labeled site (mainly the 135-kDa band) should be N-type Ca2+ channel 1-subunits.  相似文献

Summary These experiments studied the metabolic formation of a,-dodecanedioic acid using the mutant S 76 developed from the wild strain Candida tropicalis 1230 (capable of producing large amounts of a,-dodecanedioic acid).Our results show for the first time that 12-hydroxydodecanoic acid was excreted into the medium as a free acid.n-Dodecanol and n-dodecanoic acid were also detected in the n-dodecane medium. The mutant S 76 was able to produce a,-dodecanedioic acid using either n-dodecanol or dodecanoic acid as the sole carbon source. Quantitative cahnges in the concentrations of 12-hydroxy-dodecanoic acid and other intermediates were recorded during the formation of a,-dodecanedioic acid. S 76 was rapidly able to convert large amounts of 12-hydroxy-dodecanoic acid to a,-dodecanedioic acid.The formation of a,-dodecanedioic acid from n-dodecane via the sequence n-dodecanoln-dodecanoic acid 12-hydroxy-dodecanoic acid was confirmed.  相似文献

A filamentous fungus, Mortierella alpina 1S-4, is capable of producing not only arachidonic acid (AA; 20:4n-6) but also eicosapentaenoic acid (EPA; 20:5n-3) below a cultural temperature of 20°C. Here, we describe the isolation and characterization of a gene (maw3) that encodes a novel 3-desaturase from M. alpina 1S-4. Based on the conserved sequence information for M. alpina 1S-4 12-desaturase and Saccharomyces kluyveri 3-desaturase, the 3-desaturase gene from M. alpina 1S-4 was cloned. Homology analysis of protein databases revealed that the amino acid sequence showed 51% identity, at the highest, with M. alpina 1S-4 12-desaturase, whereas it exhibited 36% identity with Sac. kluyveri 3-desaturase. The cloned cDNA was confirmed to encode the 3-desaturase by its expression in the yeast Sac. cerevisiae. Analysis of the fatty acid composition of the yeast transformant demonstrated that 18-carbon and 20-carbon n-3 polyunsaturated fatty acids (PUFAs) were accumulated through conversion of exogenous 18-carbon and 20-carbon n-6 PUFAs. The substrate specificity of the M. alpina 1S-4 3-desaturase differs from those of the known fungal 3-desaturases from Sac. kluyveri and Saprolegnia diclina. Plant, cyanobacterial and Sac. kluyveri 3-desaturases desaturate 18-carbon n-6 PUFAs, Spr. diclina 3-desaturase desaturates 20-carbon n-6 PUFAs and Caenorhabditis elegans 3-desaturase prefers 18-carbon n-6 PUFAs as substrates rather than 20-carbon n-6 PUFAs. The substrate specificity of M. alpina 1S-4 3-desaturase is rather similar to that of C. elegans 3-desaturase, but the M. alpina 3-desaturase can more effectively convert AA into EPA when expressed in yeast. The M. alpina 1S-4 3-desaturase is the first known fungal desaturase that uses both 18-carbon and 20-carbon n-6 PUFAs as substrates.  相似文献

The effect of the long chain polyunsaturated fatty acid (PUFA) eicosapentaenoic acid (EPA, 20:53) on food quality for Daphnia galeata, Daphnia hyalina and a D. galeata×hyalina hybrid was examined. Somatic growth rates of all three species were determined when growing on EPA-free Scenedesmus obliquus and on S. obliquus which had been supplemented with free EPA prior to the growth experiments. Growth rates on S. obliquus were substantially higher in D. galeata(0.45day–1) than in D. hyalina(0.27day–1) and D. galeata×hyalina(0.31day–1). Supplementary EPA increased growth of D. galeata and D. hyalina, but not of the hybrid. Hence within the D. galeata/hyalina complex species differ in their ability to cope with an EPA-free diet. Analysis of fatty acids revealed that tissue concentrations of 18:33, 18:43, 20:33 and 20:43 were higher in D. galeata than in D. hyalina which indicated higher rates of assimilation and biosynthesis of PUFAs in D. galeata. This was contrasted by lower tissue concentrations of 20:53 D. galeata than in D. hyalina which suggests that 20:53 is metabolised with growth. In the D. galeata×hyalina hybrid high PUFA assimilation and biosynthesis were associated with low growth rates which explains the finding that tissue concentrations of 20:53 were highest and that growth was not constrained by the availability of 20:53.  相似文献

The structure, function, and evolutionary history of globin genes have been the subject of extensive investigation over a period of more than 40 years, yet new globin genes with highly specialized functions are still being discovered and much remains uncertain about their evolutionary history. Here we investigate the molecular evolution of the -globin gene family in a marsupial species, the tammar wallaby, Macropus eugenii. We report the complete DNA sequences of two -like globin genes and show by phylogenetic analyses that one of these genes is orthologous to embryonically expressed -globin genes of marsupials and eutherians and the other is orthologous to adult expressed -globin genes of marsupials and eutherians. We show that the tammar wallaby contains a third functional -like globin gene, -globin, which forms part of the -globin gene cluster. The position of -globin on the 3 side of the -globin cluster and its ancient phylogenetic history fit the criteria, originally proposed by Jeffreys et al. (1980), of a fossil -globin gene and suggest that an ancient chromosome or genome duplication preceded the evolution of unlinked clusters of - and -globin genes in mammals and avians. In eutherian mammals, such as humans and mice, -globin has been silenced or translocated away from the -globin locus, while in marsupials -globin is coordinately expressed with the adult -globin gene just prior to birth to produce a functional hemoglobin (2 2).  相似文献

Rheological measurements of the frequency-dependent complex elastic module G*() of entangled F-actin solutions in the frequency range 10–5 – 1 Hz were carried out in three dynamic regimes: 1.) A terminal relaxation from gel-like to liquid-like behaviour measured at frequencies < d –1 2.) a rubber-type plateau and 3.) a regime determined by chain conformational transitions at frequencies > i –1. A major point of interest was to clarify whether rheological, high precision measurements can yield quantitative information about the influence of talin and vinculin on the structure, chain dynamics, elasticity and viscoelasticity of actin filaments with time. We show that in the regime reflecting internal chain dynamics (10–2 to 1 s time domain), F-actin behaves as a random coil of the Rouse type. This contrasts with dynamic light scattering and correlation spectroscopic studies of actin filament flickering, which indicate that filaments behave as semiflexible rods. The internal chain dynamics, which are determined by thermically excited bending undulations, exhibit a persistence length of 0.3–1 m Evidence is provided that this discrepancy is due to a cross-over of semiflexible rod behaviour at excitation wavelengths () below approximately 1 gm to random-coil behaviour at 1 µ (expected at a frequency 1 Hz). The random coil behaviour is largely determined by defects in actin filaments leading to sharp bends of the chain which act as semiflexible hinges. Talin produces drastic effects on the time course of viscoelasticity during actin polymerization. It promotes the rapid formation of short filament fragments ( 1 gmm, within time scales of min) which anneal slowly into long filaments (within several hours), most probably by fusion. The viscoelasticity depends on the coexistence of short and very long filaments indicated by the elongation of the rubber plateau. The most dramatic effect is a reduction of the ratio of the terminal ('Ed) to the Rouse relaxation time of i by more than one order of magnitude (d/i = 100 compared to ratio d/i = 2000 for pure actin). From this it is concluded that talin causes a remarkable decrease in the effective segment length of the macromolecule and, thus induces an increase in chain stiffness. 1. Voltage-gated Na+ channels are responsible for initiation and conduction of action potentials. The arrival of an action potential at nerve terminal increases intracellular Na+ and Ca2+ concentrations. Calcium entry into neurons through voltage-dependent calcium channels is associated with a variety of intracellular processes. Scorpion neurotoxins have been used as tools to investigate mechanisms involved in neurotransmitter release. Tityustoxin (TsTX) is an -type toxin that delays Na+-channel inactivation. Toxin- (TiTX-) is a -type toxin that induces Na+-channel activation at resting potentials.2. In the present work, we describe the effects of both toxins on [3H]acetylcholine ([3H]ACh) release from rat cerebrocortical synaptosomes, in the presence or absence of the calcium channels blockers: -conotoxin-GVIA (-CgTx), 1 M; -agatoxin-IVA (-Aga), 30 nM; -conotoxin-MVIIC (-MVIIC), 1 M; or verapamil, 1M.3. TsTX evokes [3H]ACh release in a concentration-dependent manner with a gradual increase up to saturation at concentrations of 500 nM. However, release of ACh evoked by TiTX- was not linear regarding the toxin concentration. The [3H]-ACh release evoked by TsTX or TiTX- was partially inhibited by -CgTx or -Aga, and blocked with -MVIIC. Verapamil (1 M) had no effect. Tetrodotoxin blocked [3H]ACh release evoked by both toxins.4. These results show that different actions on Na+-channels produce different effects on [3H]ACh release with involvement of distinct presynaptic Ca2+-channels, which supports the idea that sodium channels may modulate neurotransmitter release.  相似文献

Monte Carlo simulation was used to assess the long-term effects of truncation selection within small populations using indices (I=f+m) combining mid-parent [f=(a i+a d)/2] and Mendelian-sampling (m=a-f) evaluations provided by an animal model BLUP (a=f+m). Phenotypic values of panmictic populations were generated for 30 discrete generations. Assuming a purely additive polygenic model, heritability (h 2) values were 0.10, 0.25 or 0.50. Two population sizes were considered: five males and 25 females selected out of 50 candidates of each sex (small populations, S) and 50 males and 250 females selected out of 500 candidates in each sex (large populations, L). Selection was carried out on the index defined above with = 1 (animal model BLUP), =1/2, or =0 (selection on within-family deviations). Mass selection was also considered. Selection based on the animal model BLUP (=1) maximized the cumulative genetic gain in L populations. In S populations, selection using = 1/2 and mass selection were more efficient than selection under an animal model (+ 3 to + 7% and + 1 to + 4% respectively, depending on h 2). Selection on within-family deviations always led to the lowest gains. In most cases, the variance of response to selection between replicates did not depend on the selection method. The within-replicate genetic variance and the average coefficient of inbreeding (F) were highly affected by selection with =1 or 1/2, especially in populations of size S. As expected, selection based on within-family deviations was less detrimental in that respect. The number of copies of founder neutral genes at a separate locus, and the probability vector of origin of the genes in reference to the founder animals, were also observed in addition to F values. The conclusion was that selection procedures placing less emphasis on family information might be interesting alternatives to selection based on animal model BLUP, especially for small populations with long-term selection objectives.  相似文献
