Arctostaphylos viscida Parry andA. nissenana Merriam were studied in Eldorado County, California, to check reports in the literature of hybridization between the two. Population samples were collected near Diamond Springs, where they occur sympatrically. A population sample of pureA. viscida, collected else-where, and miscellaneous herbarium specimens ofA. nissenana, were used for comparison. Data from analysis of six character differences, presented as pictorialized scatter diagrams and histograms, clearly indicate hybridization and back-crossing in the Diamond Springs populations; but we do not agree with Knight (1966) that theA. nissenana populations of this area are hybridized as a whole. Our data do not support Jepson’s (1939) opinion that the type ofA. nissenana may be of hybrid origin betweenA. viscida andA. nissenana var.arcana Jeps.  相似文献

Soils high in magnesium derived from ultramafic rocks (serpentine, peridotite, and dunite) in northwestern United States support endemic as well as wide-ranging but edaphically indifferent(bodenvag) species. The latter occur widely on diverse rock formations of the region. Severalbodenvag species are shown to respond ecotypically to ultramafic soils. Of 18 species tested, all but three are differentiated into strains either tolerant or intolerant of ultramafic soils. Tests for edaphic preferences were conducted with seedlings and mature transplants on ultramafic soils. Growth performances were determined in greenhouse pot tests, outdoor soil bins, and by transplants in the wild. Herbaceous perennials (e.g.,Achillea millefolium, Fragaria virginiana, Prunella vulgaris, Rumex acetosella) gave the clearest ecotypic differences. Woody species either showed only slight ecotypic response(Spiraea douglasii var.menziesii andGaultheria shallon) or delayed the expression of their genotypic adaptability(Pinus contorta). Where ultramafic abut non-ultramafic soils, those populations ofbodenvag species that grow in non-ultramafic habitats can have a significant proportion of individuals tolerant to ferromagnesian soils (e.g.,Achillea millefolium). This suggests gene flow between populations of contrasting edaphic sites and possibly preadaptedness for the ultramafic habitat. Strains of two introduced weeds(Prunella vulgaris andRumex acetosella) have become ecotypically tolerant to ultramafic soils, probably within the last 75 years.  相似文献

We analyzed how abiotic stress and competition interact to control the abundance and performance of the native annual grass Vulpia microstachys (Lonard and Gould; Poaceae) in a heterogeneous environment. At our study site, V. microstachys grows in nonserpentine grasslands dominated by tall invasive grasses, serpentine meadows dominated by short native forbs, and rocky serpentine slopes with a sparse native herb cover. We hypothesized that these three intermixed habitats acted as a gradient of increasing abiotic stress and decreasing aboveground competition, respectively. We further expected that the abundance and performance of V. microstachys would be highest in serpentine meadows, where neither aboveground competition nor abiotic stress were maximal. Soil and biotic variables showed roughly the expected patterns, but V. microstachys did not show the predicted peak in the middle of the gradient. Emergence, seedling survival, and abundance of V. microstachys were highest, and growth and seed production of survivors were lowest, on rocky serpentine slopes. Field experiments revealed that removal of competitors enhanced all demographic parameters, but only in the more productive habitats. An interaction between seed source and habitat, affecting emergence and survival, indicated ecotypic adaptation to the rocky serpentine slope habitat. We conclude that individual variation caused by local adaptation and phenotypic plasticity allows V. microstachys to survive in widely different habitats, none of which are optimal, resulting in considerable variation in demography.  相似文献

The gene encoding the xlnR xylanolytic activator of the heterologous fungus Aspergillus niger was incorporated into the Penicillium canescens genome. Integration of the xlnR gene resulted in the increase in a number of activities, i.e. endoxylanase, β-xylosidase, α-L-arabinofuranosidase, α-galactosidase, and feruloyl esterase, compared to the host P. canescens PCA 10 strain, while β-galactosidase, β-glucosidase, endoglucanase, and CMCase activities remained constant. Two different expression constructs were developed. The first consisted of the nucleotide sequence containing the mature P. canescens phytase gene under control of the axhA promoter region gene encoding A. niger (1,4)-β-D-arabinoxylan-arabinofuranohydrolase. The second construct combined the P. canescens phytase gene and the bgaS promoter region encoding homologous β-galactosidase. Both expression cassettes were transformed into P. canescens host strain containing xlnR. Phytase synthesis was observed only for strains with the bgaS promoter on arabinose-containing culture media. In conclusion, the bgaS and axhA promoters were regulated by different inducers and activators in the P. canescens strain containing a structural tandem of the axhA promoter and the gene of the xlnR xylanolytic activator.  相似文献

Nodule samples were collected from four alder species:Alnus nepalensis, A. sibirica, A. tinctoria andA. mandshurica growing in different environments on Gaoligong Mountains, Yunnan Province of Southwest China and on Changbai Mountains, Jilin Province of Northeast China. PCR-RFLP analysis of the IGS betweennifD andnifK genes was directly applied to unculturedFrankia strains in the nodules. A total of 21 restriction patterns were obtained. TheFrankia population in the nodules ofA. nepalensis had the highest genetic diversity among all fourFrankia populations; by contrast, the population in the nodules ofA. mandshurica had the lowest degree of divergence; the ones in the nodules ofA. sibirica andA. tinctoria were intermediate. A dendrogram, which was constructed based on the genetic distance between the restriction patterns, indicated thatFrankia strains fromA. sibirica andA. tinctoria had a close genetic relationship.Frankia strains fromA. nepalensis might be the ancestor ofFrankia strains infecting otherAlnus species. From these results and the inference of the ages ofAlnus host species, it is deduced that there was a co-evolution betweenAlnus and its microsymbiontFrankia in China.  相似文献

Background and aims

Serpentine soils impose limits on plant growth and survival and thus provide an ideal model for studying plant adaptation under environmental stress. Despite the increasing amount of data on serpentine ecotypic differentiation, no study has assessed the potential role of polyploidy. We tested for links between polyploidy and the response to serpentine stress in Knautia arvensis, a diploid-tetraploid, edaphically differentiated complex.

Methods

Variation in growth, biomass yield and tissue Mg and Ni accumulation in response to high Mg and Ni concentrations were experimentally tested using hydroponic cultivation of seedlings from eight populations of different ploidy and edaphic origin.

Results

Regardless of ploidy level, serpentine populations exhibited higher tolerance to both Mg and Ni stress than their non-serpentine counterparts, suggesting an adaptive character of these traits in K. arvensis. The effect of ploidy was rather weak and confined to a slightly better response of serpentine tetraploids to Mg stress and to higher biomass yields in tetraploids from both soil types.

Conclusions

The similar response of diploid and tetraploid serpentine populations to edaphic stress corresponded with their previously described genetic proximity. This suggests that serpentine tolerance might have been transmitted during the local autopolyploid origin of serpentine tetraploids.

Gene egl2 of secreted endo-(1–4)-β-glucanase of glycosyl hydrolase family 5 of the mycelial fungus Penicillium canescens was cloned. The gene was expressed in P. canescens under control of a strong promoter of the bgaS gene encoding β-galactosidase of P. canescens, and endoglucanase producing strains were obtained. Chromatographically purified recombinant 48 kDa protein had pH and temperature optima 3.4 and 60°C, respectively, exhibited specific activity of 33 IU, and had K m and V max in CM-cellulose hydrolysis of 10.28 g/liter and 0.26 μmol/sec per mg, respectively.  相似文献

Many of the plants found in serpentine areas are endemics and they may accumulate Ni at high concentration. High accumulation of Cr is rare, or in some views, never properly demonstrated. Generally, a very small proportion of any serpentine flora shows high accumulation of Ni, in some serpentine areas Ni accumulators are completely absent. There are approximately 570 hyperaccumulator plant species found on earth, 450 of them are Ni hyperaccumulators. A few of the Ni accumulators have potential for phytoremediation or phytomining. In the present study, 19 different Brassicaceae members growing in serpentine habitats in the district of Yahyal?, Kayseri province, Turkey viz., members of genera Aethionema, Alyssum, Arabis, Heldreichia, Hesperis, Iberis, Isatis, Microthlaspi, Odontarrhena, Pseudosempervivum and Thlaspi were investigated. Nickel concentrations in the soil and underground and aboveground parts of plants were determined by using ICP-OES. It was observed that Ni concentrations of seven taxa (Odontarrhena muralis, O. oxycarpa, Isatis cappadocica subsp. cappadocica, Microthlaspi perfoliatum, Pseudosempervivum sempervivum, Thlaspi triangulare, Thlaspi rosulare) reach the threshold criterion of 1,000 mg kg?1 for Ni hyperaccumulation. In this study Ni concentrations in aboveground tissues of I. cappadocica subsp. cappadocica are determined as 5,587 mg kg?1 (in dry weight). Accordingly, it is suggested that this taxon be added to the list of Ni hyperaccumulator plants.  相似文献

Data on habitats, food plants of larvae and adults, feeding, mating, oviposition, larval and pupal development, natural enemies, and distribution of seven weevil species (Lixus canescens F.-W., L. iridis Ol., L. myagri Ol., L. punctirostris Boh., L. subtilis Boh., L. incanescens Boh., and L. brevipes Bris.) are given. New host plants of L. canescens and L. iridis are revealed. Gall induction by L. brevipes is reported for the first time. Distribution of all the species in Ukraine and Russia (the latter based on the literature) are given in more detail. Bacteriocin production was tested in 36Klebsiella and 3Enterobacter aerogenes strains. Bacteriocins produced byK. pneumoniae were found to be active on most strains ofK. edwardsi, K. aerogenes, K. rhinoscleromatis andE. aerogenes. The bacteriocin produced byE. aerogenes 37 is also active onK. pneumoniae andK. ozaenae. The bacteriocins produced byK. rhinoscleromatis, K. edwardsi andK. aerogenes are active on only a few strains. The activity spectra of the bacteriocins of a number of strains were similar. The method of classification used for colicins could not be applied to these bacteriocins as mutants resistant to one bacteriocin were nearly always resistant to all other bacteriocins. One mutant, though resistant, still adsorbed the bacteriocin to which it was resistant and it is very likely that the same applies for all other resistant mutants. The hypothesis is made that allKlebsiella bacteriocins have the same biochemical target, or more likely, possess a common transmission mechanism.  相似文献

During investigation of the mycobiota in the Sea of Okhotsk, three new Penicillium species were isolated from subaqueous soils collected in the Sakhalin Gulf and in the northeastern part of the Sakhalin shelf, near the Piltun Bay. According to Raper and Thom’s classification, the newly described species P. piltunense, P. ochotense, and P. attenuatum belong to subsection Divaricata. Two analyses were performed to reveal the phylogenetic relationships of the putative new species with other Penicillium species. Phylogenetically, the new species are related to the members of the P. canescens group and share some morphological and physiological features with them. However, they differ in having a larger colony diameter that makes them similar to P. atrovenetum, P. coralligerum, and P. antarcticum. A detailed analysis based on ITS, combined β-tubulin and calmodulin datasets, and morphological features revealed that the new species formed a distinct group inside the P. atrovenetum subclade. Moreover, P. attenuatum differs from the other two species macro- and micromorphologically and may represent a distinct phylogenetic lineage.  相似文献

Highly arsenic resistant bacteria (27 isolates), which had a minimum inhibitory concentrations (MICs) for arsenite and arsenate of ? 40 mM and > 400 mM, respectively, were isolated from tannery wastes and agricultural soils collected in Central Thailand. On the basis of the morphological, cultural, physiological and biochemical characteristics, and on the principal ubiquinone component and 16S rRNA gene sequence analyses, they were identified as nine isolates each ofKlebsiella (Groups 1 and 8) andAcinetobacter (Groups 2, 3 and 7), four isolates each ofPseudomonas (Groups 4 and 6) andComamonas (Group 5), and one isolate ofEnterobacter (Group 9). From these 27 isolates, only one isolate, A3-3 from the genusComamonas, appeared potentially capable of oxidizing arsenite to arsenate, as determined by silver nitrate staining of arsenite agar plates after colony growth.  相似文献

Recent work in our laboratory has demonstrated that the most common contaminating fungi on different types of cheese are;Penicillium commune, P. nalgiovense, P. solitum, P. discolor, P. roqueforti, P. crustosum, P. nordicum andAspergillus versicolor. On blue cheese a new speciesP. caseifulvum has been discovered as a surface contaminant. A large number of known and unknown metabolites have been described from the above mentioned cheese associated fungi from both synthetic media and real samples. Based on chemotaxonomy our laboratory has discovered thatP. roqueforti should be divided into three species:P. roqueforti (from cheese),P. carneum (from meat) andP. paneum (from bread). SimilarlyP. verrucosum should be divided intoP. verrucosum (from cereals) andP. nordicum (from cheese and meat products). Investigations of natural populations, cytological studies, and comparisons of herbarium specimens have been used to clarify the taxonomy of the Florida species ofFlaveria. These studies indicate thatF. latifolia is a hybrid betweenF. linearis andF. floridana. Apparently the “latifolia” plants represent stabilized populations that have become adapted to the fill material dredged from the ocean and used in road building and land development in southern Florida. The wide variation noted in populations ofF. linearis andF. floridana is attributed to introgression. The paper includes a taxonomic treatment of the six species which occur in Florida. Background

Agave inaequidens and A. hookeri are anciently used species for producing the fermented beverage ‘pulque’, food and fiber in central Mexico. A. inaequidens is wild and cultivated and A. hookeri only cultivated, A. inaequidens being its putative wild relative. We analysed purposes and mechanisms of artificial selection and phenotypic divergences between wild and managed populations of A. inaequidens and between them and A. hookeri, hypothesizing phenotypic divergence between wild and domesticated populations of A. inaequidens in characters associated to domestication, and that A. hookeri would be phenotypically similar to cultivated A. inaequidens.

Methods

We studied five wild and five cultivated populations of A. inaequidens, and three cultivated populations of A. hookeri. We interviewed agave managers documenting mechanisms of artificial selection, and measured 25 morphological characters. Morphological similarity and differentiation among plants and populations were analysed through multivariate methods and ANOVAs.

Results

People recognized 2–8 variants of A. inaequidens; for cultivation they select young plants collected in wild areas recognized as producing the best quality mescal agaves. Also, they collect seeds of the largest and most vigorous plants, sowing seeds in plant beds and then transplanting the most vigorous plantlets into plantations. Multivariate methods classified separately the wild and cultivated populations of A. inaequidens and these from A. hookeri, mainly because of characters related with plant and teeth size. The cultivated plants of A. inaequidens are significantly bigger with larger teeth than wild plants. A. hookeri are also significatly bigger plants with larger leaves but lower teeth density and size than A. inaequidens. Some cultivated plants of A. inaequidens were classified as A. hookeri, and nearly 10% of A. hookeri as cultivated A. inaequidens. Wild and cultivated populations of A. inaequidens differed in 13 characters, whereas A. hookeri differed in 23 characters with wild populations and only in 6 characters with cultivated populations of A. inaequidens.

Conclusions

Divergence between wild and cultivated populations of A. inaequidens reflect artificial selection. A. hookeri is similar to the cultivated A. inaequidens, which supports the hypothesis that A. hookeri could be the extreme of a domestication gradient of a species complex.

The purpose of this study was to determine the concentration of some metals (Al, Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, Zn, Ca and Mg) in soil of serpentine and limestone sites, their bioaccumulation and impact on some biochemical parameters in T. luanica, T. kosovarica and T. albanica plants. T. kosovarica and T. albanica grows in serpentine soil, while T. luanica grow in limestone soil. The research showed that concentrations of Cd, Co, Cr, Fe, Mn and Ni were significantly higher at serpentine soil sites in comparison with limestone sites, while concentrations of Pb, Cd, Co and Cr in bulbs, leaves and seeds were under the limit of detection. The concentration of Ni in plant samples of T. kosovarica was significantly higher in comparison with its concentration in T. albanica, but it was under the limit of detection in T. luanica. Moreover, concentrations of Al and Fe in leaves of T. kosovarica and T. albanica were higher in comparison with T. luanica. The concentration of Mg was significantly higher in T. kosovarica and T. albanica than in T. luanica. The δ-aminolevulinic acid dehydratase activity, malondialdehyde and glutathione contents in leaves of T. luanica were higher in comparison with T. kosovarica and T. albanica. In addition, the amounts of total chlorophyll and δ-aminolevulinic acid (ALA) in leaves of T. albanica were higher in comparison with T. kosovarica and T. luanica. Our findings show that target organs of metal accumulation in three Tulip species appears to be leaves?>?seeds?>?bulbs, while the biochemical parameters show that limestone sites represent a less stressful habitat for growing these plant species in comparison with serpentine sites.  相似文献

In invaded environments, formerly reliable cues might no longer be associated with adaptive outcomes and organisms can become trapped by their evolved responses. The invasion of Alliaria petiolata (garlic mustard) into the native habitat of Pieris virginiensis (West Virginia White) is one such example. Female butterflies oviposit on the invasive plant because it is related to their preferred native host plant Cardamine diphylla (toothwort), but larvae are unable to complete development. We have studied the impact of the A. petiolata invasion on P. virginiensis butterflies in the Southeastern USA by comparing oviposition preference and larval survival on both plants in North Carolina (NC) populations without A. petiolata and West Virginia (WV) populations where A. petiolata is present. Larval survival to the 3rd instar was equally low in both populations when raised on A. petiolata. Mean oviposition preference on the two plants also did not differ between populations. However, we found a seasonal effect on preference between early and late season flights within WV populations. Late season females laid 99% of total eggs on A. petiolata while early season females utilized both host plants. Late season females were also less likely to lay eggs than early season females. This change in preference toward A. petiolata could be driven by the early senescence of C. diphylla and suggests a seasonal component to the impact of A. petiolata. Therefore, the already short flight season of P. virginiensis could become further constrained in invaded populations.  相似文献

Chenopodium species react on infection with tobacco mosaic virus by the formation of chlorotic or necrotic lesions and later by the abscission of infected leaves. A transition of local infection into the stem has been observed exceptionally inChenopodium quinoa, C. hybridum, andC. rubrum, but no systemic infection of the leaves followed.
