Using yeast genome databases and literature data, phylogenetic analysis of pectinase PGU genes from 112 Saccharomyces strains assigned to the biological species S. arboricola, S. bayanus (var. uvarum), S. cariocanus, S. cerevisiae, S. kudriavzevii, S. mikatae, S. paradoxus, and the hybrid taxon S. pastorianus (syn. S. carlsbergensis) was carried out. A superfamily of divergent PGU genes was found. Natural interspecies transfer of the PGU gene both from S. cerevisiae to S. bayanus and from S. paradoxus to S. cerevisiae may, however, occur. Within the Saccharomyces species, identity of the PGU nucleotide sequences was 98.8–100% for S. cerevisiae, 86.1–95.7% for S. bayanus (var. uvarum), 94–98.3% for S. kudriavzevii, and 96.8–100% for S. paradoxus/S. cariocanus. For the first time, a family of polymeric PGU1b, PGU2b, PGU3b and PGU4b genes is documented for the yeast S. bayanus var. uvarum, a variety important for winemaking.  相似文献

Chromosome numbers are reported for 19 species of North AmericanSalix, one natural hybrid, and one introduced species. The following 17 species are here examined cytologically for the first time:Salix amygdaloides Anderss.,S. arbusculoides Anderss.,S. brachycarpa Nutt.,S. Candida Willd.,S. discolor Muhl.,S. exigua Nutt.,S. kumilis Marsh.,S. interior Rowlee,S. lutea Nutt.,S. maccalliana Rowlee,S. monticola Rydb.,S. myrtillifolia Anderss.,S. pellita Anderss.,S. petiolaris J. E. Sm,S. scouleriana Barr.,S. silicicola Raup, andS. subcoerulea Piper. Sphenoptera (Tropeopeltis) barclayi sp. n. from RSA, S. (T.) kubani sp. n. from Kenya, S. (T.) makarovi sp. n. from Somalia and S. (Archideudora) karagyanae sp. n. from Tanzania are described. The name S. sansibarica Harold, 1878, nom. resurr. is resurrected, and the replacement name S. haroldi Jakovlev, 1902, syn. n. is placed to synonyms. Using yeast genome databases and literature data, we have conducted a phylogenetic analysis of pectinase PGU genes from Saccharomyces strains assigned to the biological species S. arboricola, S. bayanus (var. uvarum), S. cariocanus, S. cerevisiae, S. kudriavzevii, S. mikatae, S. paradoxus, and hybrid taxon S. pastorianus (syn. S. carlsbergensis). Single PGU genes were observed in all Saccharomyces species, except S. bayanus. The superfamily of divergent PGU genes has been documented in S. bayanus var. uvarum for the first time. Chromosomal localization of new PGU1b, PGU2b, and PGU3b genes in the yeast S. bayanus var. uvarum has been determined by molecular karyotyping and Southern hybridization.  相似文献

Till this day, there is not much known about the phylogeny of the Secale genus; therefore, in our research, we tried to shed some lights on the issue of rye (Secale genus) taxonomy. The genetic diversity and phylogenetic relationships were evaluated using 13,842 DArTseq? polymorphic markers. The model-based clustering (STRUCTURE software) separated our 84 samples into three main clusters: perennial cluster, annual cluster, and S. sylvestre cluster. The same result was obtained using Neighbour Joining tree and self-organizing maps. Secale sylvestre, S. strictum, and S. cereale are the three main species of the Secale genus. Three samples of rye are in basal positions in phylogenetic trees. These accessions share ancient morphological characters and are probably the ancestors of different lineages within Secale. Annual Secale taxa, with the exception of S. sylvestre, create one mutual taxon. We have found out that the semi-perennial taxa of S. cereale var. multicaule and S. strictum subsp. ciliatoglume are genetically closest to the annual species of S. cereale. Phylogenetic signals for semi-perennial and annual taxa are also present in S. strictum subsp. africanum. SNP-based analysis revealed that evolution of annual S. cereale has already begun in S. strictum subsp. africanum. The results showed that S. vavilovii cannot be considered as a separate species but a subspecies of S. cereale Secale cereale subsp. dighoricum is a hybrid. It is still not clear whether we can consider S. strictum subsp. strictum and S. strictum subsp. kuprijanovii as two separate species.  相似文献

Sphenoptera (s. str.) galkae sp. n. from North Pakistan and. S. (s. str.) jacobsonorum sp. n. from India (Jammu and Kashmir State) are described and compared with closely related species. New synonymy is established for the following taxa: S. hypocrita Mannerheim, 1837 (= S. torrida Jakovlev, 1898; S. ixion Kerremans, 1912, synn. n.), S. bodemeyeri Jakovlev, 1900 (= S. quadrata Kerremans, 1909, syn. n.), S. exoleta Jakovlev, 1908 (= S. politipennis Obenberger, 1927, syn. n.), S. obruta Kerremans, 1909 (= S. chalcosoma Obenberger, 1927; S. abbreviata hetera Obenberger, 1927, synn. n.), S. tragacanthae (Klug, 1829) (= S. maledicta Obenberger, 1920; S. cilicica Obenberger, 1927; S. rambouseki Obenberger, 1927; S. klickai Obenberger, 1927; S. corrosa Obenberger, 1927; S. satrapa Obenberger, 1927; S. syriae Obenberger, 1927; S. vavrai Obenberger, 1927, synn. n.), S. magna Gory et Laporte, 1839 (= S. alexandri Obenberger, 1927, syn. n.). Geometric morphometrics is a quick and reliable approach to differentiate fish stocks based on the variation of otolith shapes. In this study, morphometric analysis of otolith shapes was used to differentiate three species of Scomber. The sagittae morphology of S. scombrus otolith is totally different from that of S. japonicus and S. australasicus. Multivariate analysis consistently showed that S. japonicus was morphologically similar to S. australasicus, whereas a significant difference in otolith shapes was detected between S. scombrus and other two species of Scomber. The rostrum, antirostrum, excisural notch and dorsal-posterior margin of the otolith reflect the main variations between the three species of Scomber. Shape indices and Fourier coefficients were used to discriminate fish species using analysis of variance and Fisher discriminant analysis. The shape indices successfully differentiated 100%, 95.7% and 96.4% of otoliths in S. japonicus, S. australasicus and S. scombrus, respectively, while the Fourier coefficients only discriminated 70.0%, 61.9% and 91.3% of the sagittae in S. japonicus, S. australasicus and S. scombrus. This study indicates that the shape analysis on the sagittae morphometrics of otoliths is a better method to differentiate species of Scomber.  相似文献

The R1 gene for resistance to oomycete Phytophthora infestans (Mont.) de Bary, the causal agent of late blight disease of potato (Solanum tuberosum L.), was initially identified in S. demissum and potato varieties bred by introgressing the S. demissum germplasm. Later a sequence characterized amplified region (SCAR) marker R1-1205 of this gene was also found in S. stoloniferum and S. polytrichon. Here we describe the full-length R1 sequence cloned from S. stoloniferum. This sequence is translatable, and this evidence of structural gene integrity is reinforced by functional characterization of the S. stoloniferum R1 gene in an effectoromics experiment. When screened across a series of S. demissum and S. stoloniferum accessions, the R1 sequences differed by several single nucleotide polymorphisms and an indel; this indel served the basis for constructing SCAR markers R1-517 and R1-513 that reliably discerned two R1 orthologs. The demissum-specific marker R1-517 was found in all S. demissum accessions under study; it was also present in many demissum-derived potato varieties and hybrids. The stoloniferum-specific marker R1-513 was found in 27% of S. stoloniferum and S. polytrichon accessions; however, we failed to discern this marker in the genotypes of cultivated potato listing S. stoloniferum in their pedigrees. Most probably, such absence of R1-513 is best explained by an opportunistic breeding history of stoloniferum-derived founder lines, which were employed first and foremost in breeding for resistance to potato virus Y: eventually, these founder lines are devoid of the R1 gene.  相似文献

The genus Phaeophilacris Walk. is divided into two subgenera: Phaeophilacris s. str. and Speluncacris Sjöst. The latter name, originally proposed as a generic one and synonymized with Phaeophilacris s. l., is restored as a subgeneric name. The former genera Opilionacris Sjöst. and Cavernacris Sjöst. previously synonymized with Phaeophilacris s. l. are considered as synonyms of the subgenus Speluncacris. Four new species (Ph. (S.) malawi sp. n., Ph. (S.) boldyrevi sp. n., Ph. (S.) dmitrievi sp. n., and Ph. (S.) gigantea sp. n.) and three new subspecies (Ph. (S.) bredoides mangochi subsp. n., Ph. Pectinase (endo-polygalacturonase) is the key enzyme splitting plant pectin. The corresponding single gene PGU1 is documented for the yeast S. cerevisiae. On the basis of phylogenetic analysis of the PGU nucleotide sequence available in the GenBank, a family of divergent PGU genes is found in the species complex S. bayanus: S. bayanus var. uvarum, S. eubayanus, and hybrid taxon S. pastorianus. The genomes of the recently discovered yeast Saccharomyces eubayanus and traditional S. cerevisiae are known to be found in the yeast S. pastorianus (syn. S. carlsbergensis), which are essential for brewing. The cryotolerant yeast S. bayanus var. uvarum is of great importance for production of some wines. Based on ascospore viability and meiotic recombination of the control parental markers in hybrids, we have shown that there is no complete interspecies post-zygotic isolation between the yeasts S. eubayanus, S. bayanus var. bayanus and S. bayanus var. uvarum. The genetic data presented indicate that all of the three taxa belong to the same species.  相似文献

Biologically active substances and antioxidant activity of extracts from leaves and inflorescences of nine representatives of the genus Spiraea L. growing on the territory of the Far East of Russia were investigated. Widespread species of the genus Spiraea (S. salicifolia, S. media var. media, S. betulifolia and S. ussuriensis subsp. ussuriensis) have the highest levels of biologically active substances. The inflorescences of spiraeas there contain more flavonols (up to 3.9%), oxycinnamic acids (up to 1.2%), catechins (up to 5.7%) and saponins (up to 5.1%) compared to their leaves, and there are more tannins (up to 11.6%) in the leaves. Among the Far Eastern representatives of the genus Spiraea, S. betulifolia and S. beauverdiana (section Calospira), S. humilis and S. salicifolia (section Spiraria), S. pubescens and S. media var. media (section Chamaedryon) are promising antioxidants. Plants of the genus Spiraea probably contain water-soluble antioxidant compounds of phenolic type, because the antioxidant activity of aqueous extracts in the leaves and inflorescences of spiraeas is higher (0.16–2.79 mg/g) than that of water-alcoholic compounds (0.06–2.54 mg/g). The antioxidant activity in the leaves of spiraeas is generally higher than that in the inflorescence. A reliable positive correlation is observed between the antioxidant activity of aqueous extracts from the organs of spiraeas and a content of oxycinnamic acids.  相似文献

During a survey on corticolous Dothideomycetes in Mallorca, several collections with ascomata, asci, and ascospores matching the genus Stigmatodiscus (Stigmatodiscales, Dothideomycetes) were revealed, which did not fit any described species. Therefore, these collections were cultured and sequenced, and a multigene matrix of four loci (nuc18S-ITS-28S rDNA, rpb2, tef1 and tub2) was produced. Based on the results of the phylogenetic analyses of this matrix and of morphological investigations, three new species (Stigmatodiscus labiatus, S. oculatus, and S. pinicola) are described and illustrated, Asterodiscus is synonymised with Stigmatodiscus and the new combination S. tamaricis is proposed. A key to all currently known species of Stigmatodiscus is provided.  相似文献

This study focuses on the former Verticillium section Albo-erecta that included white or whitish species with verticillium-like, erect conidiophores. Its type species was Verticillium fungicola, which was transferred to Lecanicillium, a genus that otherwise contains species with prostrate conidiophores. Phylogenetic analysis of ITS, 18S and 28S rDNA sequences revealed that this group is heterogeneous and its members belong to at least three families. The present study is a first step to its resolution. Supported by analyses of 68 sequences of ITS, 23 sequences of 18S and 43 sequences of 28S ribosomal DNA, the species are distributed over three families. In the Cordycipitaceae, Leptobacillium gen. nov. is described with L. leptobactrum comb. nov. and its new variety, calidius. In the Hypocreaceae, Hypomyces is treated with one new combination from former Nectriopsis, H. tubariicola, and two new anamorphic species, H. ellipsosporus and H. subglobosus; in Sphaerostilbella, one new anamorphic species, S. parabroomeana, is described. In the Bionectriaceae, anamorphic isolates of Nectriopsis requiring a new combination, N. lindauiana, and a new species, N. fuliginicola, are described. Parsimony analysis of the LSU suggests that Ovicillium gen. nov. with four new species, O. attenuatum, O. oosporum, O. subglobosum and O. napiforme, is a member of the Bionectriaceae by 93 % bootstrap support. Of uncertain family classification are Chlamydocillium gen. nov. with one new soil-borne species, C. cyanophilum, and Chlorocillium gen. nov. based on Acremonium griseum Petch. A few further white verticillium-like anamorphs, particularly V. biguttatum, could not yet be reclassified.  相似文献

Two new genera (monotypic Samarabatrachus gen. nov. (S. bjerringi sp. nov.) and Syrtosuchus gen. nov. (including S. samarensis (Sennikov, 1981) and S. morkovini sp. nov) and a new species of the genus Selenocara Bjerring (S. rossica sp. nov.) are described based on revision of the original material of the form previously known as Wetlugasaurus samarensis. The genera Selenocara and Samarabatrachus gen. nov. positioned at the base of radiation of Triassic capitosaurids are combined into a new subfamily, Selenocarinae subfam. nov. The genus Syrtosuchus gen. nov., which is undoubtedly a derivative of early capitosaurids (Selenocara or a closely related genus), shows some typical benthosuchid features and is assigned to a separate benthosuchid subfamily, Syrtosuchinae subfam. nov. The diagnoses of the genera Wetlugasaurus Riabinin and Selenocara are amended. For the territory of the Obschii Syrt Highland, the genera Selenocara, Samarabatrachus gen. nov., and Syrtosuchus gen. nov. are regarded as key members of a separate Early Triassic tetrapod assemblage (Selenocara–Syrtosuchus Fauna), which is intermediate between the Tupilakosaurus and Benthosuchus faunas and dated Late Induan (Dienerian).  相似文献

A total of 39 coagulase-negative staphylococci and seven Staphylococcus aureus strains were isolated from small mammal feces, i.e., the striped field mouse (Apodemus agrarius) and the yellow-necked mouse (A. flavicollis) in two sampling areas, deciduous forest and karst plains. MALDI-TOF analysis revealed five species of coagulase-negative staphylococci: S. sciuri, S. hominis, S. warneri, S. haemolyticus, and S. xylosus. All strains were susceptible to tetracycline, linezolid, vancomycin, and teicoplanin. Three MRSA strains with the mecA gene were detected. The beta-lactamase gene blaZ was detected in ampicillin-resistant staphylococci and in the high-level resistant strains (oxacillin over 2 mg/L) mecA gene. The mecC gene was not detected by PCR. Erythromycin-resistant staphylococci harbored the ermC gene and/or the efflux gene msrA. There were no detectable dfr genes in trimethoprim-resistant staphylococci and the rifampicin-resistant strains were without mutation in the rpoB gene. In summary, wild small mammals may serve as sentinels of mecA-positive S. aureus with erythromycin resistance genes ermC and efflux msrA. Small mammals appear to be useful indicators of antibiotic resistance.  相似文献
