Abstract— Gangliosides were isolated from purified human myelin in a yield of 62 μg of lipid-bound sialic acid per 100 mg of dry myelin. Sialosylgalactosyl ceramide (G7) was found to be a major component of the ganglioside fraction, amounting to 15 per cent of the total sialic acid. It accounted for 10 per cent of lipid-bound sialic acid in adult human white matter, making it the third most abundant ganglioside on a molar basis. These results were obtained with an improved method for isolating total gangliosides in high yield, by employing DEAE-Sephadex column chromatography. Myelin from other mammalian species had considerably less G7, and there were also indications of maturational changes. Both 2-hydroxy and unsubstituted fatty acids were components of the ceramide unit, in a ratio of 3:2, respectively. The overall fatty acid pattern was very similar to that for myelin cerebroside and sulphatide. Long-chain bases included only C18 species, with sphingosine predominating (>90 per cent). These observations suggest a metabolic relationship between G7 and either cerebroside or sulphatide.  相似文献

Quantitative analyses have been made of the dietary cholesterol requirement for the growth of the larvae of Musca domestica. The larvae will not grow on diets to which no cholesterol is added, a few pupae and adults are obtained when the concentration of cholesterol is 0·05 μmol/g of diet, but the concentration has to be raised to 0·36 μmol/g of diet before the maximum numbers of pupae and adults are obtained. Further addition of cholesterol above 0·36 μmol/g diet did not have any significant effect on the weight and growth of the larvae. However, the ratios of the cholesterol to phospholipid fractions recovered from the larvae increased rapidly when the concentration of cholesterol in the diet was raised from 0·05 to 0·56 μmol/g of diet. Above this concentration only a slight increase in the ratios was observed. Larvae reared on diets containing 0·05 μmol cholesterol/g of diet contain only 25 per cent of the cholesterol content of the larvae reared on the diets containing more than 0·28 μmol of cholesterol/g of diet, the cholesterol content being expressed relative to the weight of the larvae,The absence of cholesterol synthesis has been demonstrated in the larvae by feeding [4-14C] cholesterol. The specific activity of the cholesterol recovered from the larvae is the same as that of cholesterol added to the diet. Irrespective of the cholesterol concentration of the larval diet, approximately 97 per cent of the radioactivity recovered from the larvae behaved as free cholesterol, less than 1 per cent as cholesterol esters and the rest as unidentified ‘polar sterols’. Myelin was purified from rat brain and sciatic nerve after invivo labeling with [3H]fucose and [14C]glucosamine to provide a radioactive marker for glycoproteins. The glycoproteins in the isolated myelin were digested exhaustively with pronase, and glycopeptides were isolated from the digest by gel filtration on Bio-Gel P-10. The glycopeptides from brain myelin separated into large and small molecular weight fractions, whereas the glycopeptides of sciatic nerve myelin eluted as a single symmetrical peak. The large and small glycopeptide fractions from central myelin and the single glycopeptide fraction from peripheral myelin were analyzed for carbohydrate by colorimetric and gas liquid chromatographic techniques. The glycopeptides from brain myelin contained 2.4 μg of neutral sugar and 0.59 μg of sialic acid per mg total myelin protein, whereas sciatic nerve myelin glycopeptides contained 10 μg of neutral sugar and 3.8 μg of sialic acid per mg total protein. Similarly, the gas-liquid chromatographic analyses showed that the glycopeptides from peripheral myelin contained 4- to 7-fold more of each individual per mg total myelin protein than those from central myelin. Most of the sialic acid and galactose in the glycopeptides from central myelin were in the large molecular weight fraction, and the small molecular weight glycopeptides contained primarily mannose and N-acetylglucosamine. The considerably higher content of glycoprotein-carbohydrate in peripheral myelin supports the results of gel electrophoretic studies, which indicate that the major protein in peripheral myelin in glycosylated while the glycoproteins in purified central myelin are quantitatevely minor components.  相似文献

Abstract— Explants of rat cerebellum obtained 12-24 h after birth were maintained in culture in Maximow assemblies. About 90 per cent of the cultures myelinated after 10–12 days in vitro . Cultures maintained for varying periods of time were exposed to [35S]Na2SO4; labelled sulphatide was recovered from the total homogenate. Preparation of a subcellular fraction with density properties corresponding to those of myelin indicated that labelled sulphatide appeared in this fraction. Cultures which were poorly-myelinated always exhibited a lower rate of inccrpomtion than well-myelmated cultures from littermate animals, but the distribution of labelled sulphatide into the 'myelin' fraction was similar in the two groups. The rate of incorporation of [35S]Na2SO4 into total sulphatide increased with the duration of the culture, with a low level of incorporation until the seventh day in vitro , followed by a sharp increase in rate up to the 21st day. This pattern resembles that observed for rat cerebellum in vivo .  相似文献

The freeze tolerant wood frog Rana sylvatica was studied to determine the impact of the freezing and thawing of this frog on the activity of γ-glutamyltranspeptidase in the liver. On exposure to ?2·5°C, for 1, 12 and 24 h, frogs were found to be cool, covered with ice crystals and frozen, respectively. Thawing for 24 h at 4°C recovered the frogs completely. A 45 per cent decrease in the liver weight: body weight ratio was notable after 1 h at ?2·5°C, suggestive of an early hepatic capacitance response. A glycemic response to freezing was observed: blood glucose levels exhibited a 55 per cent decrease after 1 h at ?2·5°C on cooling; a 10·5-fold increase after 12 h at ?2·5°C on the initiation of freezing; and a 22-fold increase after 24 h at ?2·5°C in the fully frozen state. Blood glucose levels remained elevated four-fold in the thawed state. Plasma insulin levels were increased twofold in the frozen state and 1·8-fold in the thawed state, while plasma ketone levels were increased 1·8-fold in the frozen state and 1·5-fold in the thawed state. Plasma total T3 levels were decreased by 22 per cent in the frozen state and normalized on thawing. In homogenates and plasma membranes isolated from the livers of Rana sylvatica, the activity of γ-glutamyltranspeptidase was found to be elevated at all stages of the freeze–thaw process. After 1, 12 and 24 h at ?2·5°C, activities were increased 2·5-, 2·3-, 2·4-fold respectively in the homogenates and 2·5-, 2·2-, 2·4-fold respectively in the plasma membranes. After thawing, activities were still increased 1·9-fold in both homogenates and plasma membranes. In homogenates prepared from the kidneys of Rana sylvatica, the activity of γ-glutamyltranspeptidase was increased 1·4-fold after 1 h at ?2·5°C after which it returned to normal. The role of thyroid hormone in producing the increase in γ-glutamyltranspeptidase in the liver of Rana sylvatica in response to freezing is discussed as is the significance of the enzyme increase in terms of hepatic cytoprotection and freeze tolerance.  相似文献

The effect of reduction in particle size on the anthelmintic efficacy of mebendazole (methyl 5 (6)- benzoyle 1–2 benzimidazole carbamate) was studied in rats undergoing a primary infection with N. brasiliensis. A single oral treatment with fine ground mebendazole (particle size spectrum—54·95 per cent of particles less than 10·62 μ dia.; 86·06 per cent less than 21·27 μ) removed more than 98 per cent of adult worms from the intestine at a dose rate of 12·5 mg/kg body wt. On the other hand the best result achieved with coarse ground mebendazole (18·47 per cent of particles less than 10·62 μ dia; 42·26 per cent less than 21·27 μ dia) was 58 per cent of adult worms removed at a dose rate of 100 mg/kg body wt. It was also shown that fine ground mebendazole adversely affected migrating third stage larvae of N. brasiliensis.  相似文献

The size and number of axons in the ventral cord connectives of the cockroach Blaberus craniifer were determined from montages constructed of electron micrographs of the left connective of each of the connective pairs examined. The fibres were grouped into three main diameter categories: fine fibres from 0·2 to 1 μm, small fibres from 1 to 6 μm, and large fibres from 6 to 24 μm. In the five different left connectives examined, the fine fibres numbered from 2006 to 8535 and composed from 56·5 to 83 per cent of the total fibres. The small fibres numbered from 1269 to 2361 and composed from 16·5 to 41 per cent of the total fibre number. The large axons ranged between 29 and 220 in number which represented from 0·5 to 2·5 per cent of the fibre population.  相似文献

The characteristics of the enzyme γ-glutamyltranspeptidase were determined in frog liver and compared to those of the rat. In Rana pipiens, tissue distribution studies indicated the order of activity to be: kidney >>> liver >> nerve > egg > lung > heart > skeletal muscle in homogenates. In the Rana pipiens relative to the Fischer 344 rat, the activity of the liver enzyme was somewhat greater (1·8-fold) and the kidney enzyme substantially less (25-fold). Frog liver γ-glutamyltranspeptidase displayed strain-dependent differences in activity with Rana pipiens and Rana sylvatica exhibiting comparable activities and Xenopus laevis exhibiting 20-fold lower activities. No influence of sex was apparent in Rana pipiens in contrast to the sex dependent differences observed in the Fischer 344 rat: ♀ : ♂ = 7:1. In homogenates and plasma membrane fractions of Rana pipiens, Xenopus laevis and the Fischer 344 rat, high, and comparable relative specific activities, were observed, 8–11, coupled with protein yields of 2·2–2·5 per cent indicating the enzyme to be plasma membrane bound and associated with the sinusoidal surface of the liver cell. Both the frog Rana pipiens and Xenopus laevis and Fischer 344 rat liver plasma membrane enzymes displayed comparable temperature-induced activation (1·51–1·74-fold) but with a peak for the frogs at 60°C and for the rat at 50°C. Both Acivicin and maleate inhibited the liver plasma membrane γ-glutamyltranspeptidase of both Rana pipiens and the Fischer 344 rat, but the frog enzyme was less sensitive (89 per cent decrease versus 97 per cent decrease) to 150 μM Acivicin and more sensitive (65 per cent decrease versus 35 per cent decrease at 150 mM maleate) to maleate. Kinetic studies indicated that the liver plasma membrane enzyme from Rana pipiens had a Km of 0·61 mM and Vmax of 55·6 nmol mg?1 min?1 and that from the Fischer 344 rat had a Km of 3·57 mM and Vmax of 71·4 nmol mg?1 min?1.  相似文献

The German cockroach is convenient for assaying gonadotrophic effects of juvenile hormone (JH) analogues. The animals can be raised synchronously from egg to adult state in 2·5 months at 30°C, a shorter time than for any of the common laboratory-reared cockroaches. Last instar larvae can be stored for up to a month at 16 to 18°C and brought to 30°C to metamorphose when needed, providing a convenient source of bioassay animals. The assay takes 2 days to complete, less time than previously published gonadotrophic assays. The growth of terminal oöcytes of the ovary responds linearly with log dose between 0·08 and 0·8 μg of Cecropia JH-I. The analogues JH-I ethyl ester and methyl-juvenate have dose response curves parallel to that for JH-I, enabling potencies, r, relative to JH-I to be calculated (r = 1·01, 0·35 respectively). There is no significant effect of the volume of hormone vehicle between 2 and 8 μl. A 30 per cent increase in the precision of the bioassay is obtained by correcting the oöcyte growth for the fed-weight of the animal. The bioassay is adequate to resolve a twofold difference in potency using as few as 36 animals per analogue.  相似文献

Anaesthesia with ether, halothane, methoxyflurane (Penthrane) and Ohio 347 (Ethrane) increased the energy stores in mouse brain as much as 1·7-fold above the control values. The greatest increases were observed in glucose and glycogen. Glucose-6-P was increased in some cases and UDP glucose was consistently lower in the anaesthetized animals. Hypothermia in conjunction with anaesthesia modified some of the observed changes. Hypothermia alone was associated with an increase in P-creatine and glucose and a decrease in UDPglucose in the brain. The cerebral metabolic rate was depressed by all the anaesthetic agents to about 50 per cent of the control value. When the body temperature was lowered to 25°, the cerebral metabolic rate fell to 73 per cent of the control rate. A temperature coefficient of 1·035 was calculated as the fractional change/degree between 25° and 34°.  相似文献

Abstract— Following intracranial injections of puromycin, the incorporation of [3H]leucine into brain protein was inhibited by 80 per cent. Conversely, incorporation of [35S]sulphate into sulphatide or [2-3H]glycerol into phosphatidyl choline was not inhibited. Under these conditions, appearance of labelled protein in myelin was inhibited by 90 per cent, while the appearance of newly labelled sulphatide and phosphatidyl choline in myelin membrane was not greatly affected. Experiments with cycloheximide gave similar results with phosphatidyl choline, but incorporation of [35S]sulphate into total sulphatide was decreased by about 30 per cent in animals given cycloheximide. Neither puromycin nor cycloheximide had any inhibitory effect on galactocerebroside sulphotransferase.  相似文献

Administration of the synthetic, double-stranded polynucleotide, (I)n·(C)n, to mice in the range from 10–100 μg per g body weight results 24 hr later in increases approaching 2-fold in total RNase II (EC 3.1.4.22) activity levels in liver and skeletal muscle. Free RNase inhibitor levels fall so that free RNase II activities approach 5-fold increases. This response of RNase II may be part of the general defensive response against infectious agents exhibited by mammalian and avian cells and could also account for accompanying effects on inhibition of cellular protein synthesis which are caused by agents such as viruses and (I)n·(C)n.  相似文献

5-Hydroxytryptamine (5HT) was measured in the nervous and heart tissues of Locusta migratoria migratorioides in concentrations of 2·34 and 2·69 μg/g wet weight respectively. Among catecholamines, dopamine was found in the nervous tissue (1·31 μg/g wet weight) as well as in the heart (2·42 μg/g wet weight). Noradrenaline was present only in the brain, but in smaller amounts (0·24 μg/g wet weight) than dopamine. Adrenaline was found neither in the ganglia nor in the heart.On the basis of their occurrence in the CNS, 5HT, dopamine, and noradrenaline, and in the heart 5HT and dopamine can be regarded as transmitter or modulator substances.  相似文献

Synthetic juvenile hormone (methyl trans,trans,cis-10-epoxy-7-ethyl-3,11-dimethyl-2,6-tridecadienoate, 1 μg/μl acetone per animal) (JH) was topically applied to 2- to 3-day-old worker honeybee larvae in the hive. Eighty per cent of the hormone-treated larvae were removed from their brood cell before pupation. Only 1 out of 42 adults showed characteristics of an intercaste. Fifty per cent of the control larvae (1 μl acetone) developed to adults, all of which were workers.After topical application of JH and feeding on royal jelly under in vitro conditions, the rate of survival is high (up to 85 per cent adults), but up to 67 per cent of queens and 44 per cent of workers exhibit eye malformations with characteristics of somatic mutation. Formation of a more solid web by the spinning larvae, shortening of the diapause by 1 to 2 days, and unusual shapes of mandibles, legs, and abdomen are a consequence of hormone treatment. The effects are less marked after application of 0·1 instead of 1 μg hormone or after its addition to the food (2 μg/g royal jelly). Treatment of the 2- to 3-day-old worker larvae and subsequent rearing on royal jelly is followed by a shift in caste differentiation from queens and workers to intercastes. In no case, are more queens developed after juvenile hormone treatment.Queen bee determinator, partially purified from royal jelly, induces a concentration-dependent shift from workers to queen differentiation. A threefold increase in the natural determinator concentration of royal jelly results in an almost exclusive (98 per cent) queen formation from 2- to 3-day-old worker larvae. In contrast to this direct effect, the influence of JH is explained as an indirect morphogenetic effect not directly coupled with honeybee caste differentiation.  相似文献

Pregnant mice were irradiated with 0.5 Gy fission neutrons on the eighteenth day of their gestation. The average litter size at birth was unchanged but mortality increased 5-6 fold in the first 3 days. The irradiated mice were the same weight as control mice at birth but showed a progressively increasing weight deficiency up to at least 36 days as compared to controls. Brain weight was 37, 45 and 25 per cent less in 2-, 3- and 52-week old irradiated animals, respectively, and the ratio of brain weight to body weight was 25, 27 and 13 per cent less. The concentrations of DNA, RNA and protein (mg/g wet tissue) were the same in irradiated and control mice in both brain and liver at all three ages. Total DNA, RNA and protein contents of whole brain after irradiation were 56-75 per cent of the control levels. No definite decrease was observed in liver. Histological study at 6 hours after irradiation showed nuclear pyknosis in the central nervous system from definite to very severe according to the part examined. It is concluded that damage to the central nervous system of the 18-day mouse foetus after neutron irradiation is mainly due to killing and/or inhibition of the differentiation of neuroblasts.  相似文献

Abstract— Synaptosomes from guinea-pig cerebral cortex contain a fetuin: sialyl glyco-protein: glycosyl transferase; evidence is presented which indicates that both a sialyl transferase; evidence is presented which indicates that both a sialyl transferase and endogenous acceptors were located in the synaptosome ‘ghost’ fractions. Following solubilization of synaptosomes with Triton X-100 and the use of fetuin minus NANA as acceptor, 25 per cent of the transferase was recovered after centrifugation and column chromatography on Sephadex G-100 and G-200 with a 64·0-fold purification. The enzyme had a pH optimum of 6·3, required no divalent metal cation for activity, and exhibited high activity with either fetuin minus sialic acid, prothrombin minus sialic acid, Tamm-Horsfall glycoprotein minus sialic acid, or orosomucoid minus sialic acid as acceptor; neither BSM nor PSM minus NANA functioned as an effective acceptor. The fetuin:sialyl transferase using fetuin minus sialic acid and CMP-sialic acid as substrates a and b, respectively, gave the following kinetic constants when using the Cleland bisubstrate model: Ka= 35μM; Kb= 3 μM; Kia, = 25 μM; Kib= 25μM; and V1= 92 pmoles. min?1.mg?1 of protein. The following divalent cations inhibited the reaction: Ba2+ > Hg2+ > Pb2+ > Cu2+.  相似文献

  • 1.1. The sialic acid content of newborn calf serum (4.8 μmol/ml) is approx. 3-fold higher than that of mature animals (1.4 μmol/ml) and decreases to 2.4 μmol/ml at 20 days of age. Colostrum-fed and colostrum-deprived calves have similar levels of sialic acid from birth to 14 days of age.
  • 2.2. The high level of sialic acid in newborn calf serum is due predominantly to N-acetylneuraminic acid, since this sialic acid accounts for 93% of the total and since <5% of the sialic acid is O-acetylated.
  • 3.3. Comparison of day 0 and day 20 serum by gel filtration and by SDS polyacrylamide gel electrophoresis demonstrates that the increase in sialic acid is associated with increased production and/or sialylation of components with MW of 45–60 kDa.
  • 4.4. A high percentage (64%) of the sialic acid in newborn calf serum is detected with the lipid-linked sialic acid assay, relative to 20 day old (25%) or mature (18%) animals.
  • 5.5. This indicates that the glycoproteins of newborn calf serum are more efficiently extracted under the conditions of this assay than glycoproteins of mature serum.

Abstract— A procedure is described for the preparation from rat brain of myelin having the same degree of purity at all ages. Such a procedure is essential for the study of myelin composition during development. Microsomal contamination was successfully eliminated by adjusting the method to maintain a constant amount of brain per unit volume in the initial density gradient step, and by including two osmotic shocks and two low-speed centrifugation steps. Myelin prepared in this way from animals ranging from 15 days to 14months of age had a total ATPase activity of 0.3-2.0 μmol of Pi.h?1.mg?1 dry wt of myelin, representing 0.1-1.2 per cent recovery of the total homogenate activity; a Na+, K+- ATPase activity of 0.1-1.6 μfnol of Pi.h?1.mg?1 dry wt, representing 0.04-1.5 per cent recovery; a nucleic acid content of 0.2-0.7 per cent of myelin dry wt, representing 0.2-2.0 per cent recovery; and a ganglioside NANA content of 0.04-0.07 per cent of myelin dry wt. representing 0.2-4.6 per cent recovery. The myelin prepared from 20-day animals had the highest content of the first three constituents; otherwise the values of the four constituents were relatively constant per unit weight of myelin. The amounts of nucleic acid and ganglioside recovered in the myelin fractions increased with increasing age and myelin yield.  相似文献

Abstract— The effect of neonatal thyroidectomy on the cyclic AMP system in the developing rat brain was examined. Administration of 131I at birth led to a 16 per cent reduction in brain weight and a 70 per cent reduction in body weight by 40 days of age. The level of cyclic AMP in the brain increased 5-fold between birth and 40 days of age and this increase was partially reduced by early thyroidectomy. A similar increase in the activity of adenyl cyclase and phosphodiesterase was observed during development, but thyroidectomy produced no detectable changes in the activity of either enzyme. The activity of the cyclic AMP-dependent protein kinase was already maximal at birth and also was unaffected by thyroidectomy.
Norepinephrine increased levels of cyclic AMP 4- to 5-fold in brain slices prepared from adult rats, but was without effect on slices prepared from newborn or 3-day-old rats. The response to norepinephrine in thyroidectomized rats did not differ from that in control rats at any of the ages examined. Our findings indicate that neonatal hypothyroidism does not deleteriously affect the development of the cyclic AMP system in the rat brain.  相似文献

In the frog Rana temporaria L., oleamide solution (10 μmole/L) applied to the isolated basal surface of the skin augmented the short-circuit current (SCC) from 59.8 ± 2.5 to 78.2 ± 1.4 μA/cm2. When applied to the serous membrane of the urinary bladder, oleamide (1 μmole/L) induced more than a 30-fold increase in osmotic water permeability. The addition of argininevasotocin against the background of oleamide further increased SCC across the skin and osmotic water permeability in the bladder. In Wistar rats, intraperitoneal injection of oleamide (0.1 μmole/L per 100 g of body weight) to non-anesthetized animals after water load reduced diuresis by 22% and increased solute-free water reabsorption and urinary sodium excretion by 31% and 55%, respectively, but did not affect urinary potassium excretion. These findings provide evidence of the similarity between the effects of oleamide and nonapeptide neurohypophyseal hormones on water and ion transport in epithelial cells of osmoregulatory organs in vertebrates.  相似文献
