Evidence for Polygenic Epistatic Interactions in Man? 总被引:2,自引:0,他引:2
Studies of multifactorial inheritance in man have ignored nonadditive gene action or attributed it entirely to dominance. Reanalyses of dermatoglyphic data on monozygotic and dizygotic twins, siblings and parents and offspring suggest that a substantial proportion of variation in total finger pattern intensity is due to epistatic interactions between additive genetic deviations, not dominance. Bootstrapping and power simulations support this interpretation of the data. We believe this is the strongest evidence so far for polygenic epistasis in man. 相似文献

Airways of asthma patients can become hyperresponsive to airway spasmogens following regular use of isoprenaline or β2-selective sympathomimetics. Hyperreactivity that results from acute exposure of animals to these drugs is pre-empted by vagal section (a procedure which does not influence spasmolytic efficacy of sympathomimetics), is not diminished by antagonism of β2-adrenoceptors and is not associated with loss of responsivity of β2-adrenoceptors in the airways. Since activation, modulation, or blockade of β2-adrenoceptors does not determine this form of hyperreactivity, the possibility that distomers may induce hyperreactivity must be considered. Ocular and vascular responses to distomers of sympathomimetics have long been recognised and, more recently, comparable observations have been made for the airways. Thus, reactivity of guinea-pig airways to spasmogens was increased following exposure to S-isoprenaline, S-salbutamol, or S-terbutaline and exposure to S-isoprenaline or S-salbutamol can intensify symptoms in asthmatics. Regular exposure to the racemate, especially during or following an allergic reaction, predisposes to expression of hyperreactivity, which is nullified, acutely, by the eutomer. Interaction between the tricyclic antidepressant desipramine and the antihypertensive agent clonidine has been investigated in five hypertensive patients in a double-blind placebo controlled study. Introduction of the tricyclic antidepressant led to loss of blood pressure control in four of the patients. The average blood pressure rise in the desipramine period compared with the placebo period was 22/15 mm Hg in the lying position and 12/11 mm Hg standing. Thus addition of a tricyclic antidepressant may lead to loss of blood pressure control in a hypertensive patient treated with clonidine. 相似文献

Sclerostin (SOST) is a glycoprotein having many important functions in the regulation of bone formation as a key negative regulator of Wnt signaling in bone. Surface plasmon resonance (SPR), which allows for a direct quantitative analysis of the label-free molecular interactions in real-time, has been widely used for the biophysical characterization of glycosaminoglycan (GAG)-protein interactions. In the present study, we report kinetics, structural analysis and the effects of physiological conditions (e.g., salt concentrations, Ca2+ and Zn2+concentrations) on the interactions between GAGs and recombinant human (rh) and recombinant mouse (rm) SOST using SPR. SPR results revealed that both SOSTs bind heparin with high affinity (rhSOST-heparin, KD~36 nM and rmSOST-heparin, KD~77 nM) and the shortest oligosaccharide of heparin that effectively competes with full size heparin for SOST binding is octadecasaccharide (18mer). This heparin binding protein also interacts with other highly sulfated GAGs including, disulfated-dermatan sulfate and chondroitin sulfate E. In addition, liquid chromatography-mass spectrometry was used to characterize the structure of sulfated GAGs that bound to SOST. 相似文献

Attention and memory cannot operate without each other. In this review, we discuss two lines of recent evidence that support this interdependence. First, memory has a limited capacity, and thus attention determines what will be encoded. Division of attention during encoding prevents the formation of conscious memories, although the role of attention in formation of unconscious memories is more complex. Such memories can be encoded even when there is another concurrent task, but the stimuli that are to be encoded must be selected from among other competing stimuli. Second, memory from past experience guides what should be attended. Brain areas that are important for memory, such as the hippocampus and medial temporal lobe structures, are recruited in attention tasks, and memory directly affects frontal-parietal networks involved in spatial orienting. Thus, exploring the interactions between attention and memory can provide new insights into these fundamental topics of cognitive neuroscience. 相似文献

Interactions between members of the genus Listeria and lectins are described. L. monocytogenes was shown to be heterogenous with respect to agglutination by lectins. L. monocytogenes serotype 4b had a pattern of lectin binding distinct from the other listeriae. Titration of the listeriae with lectins proved to be useful in further distinguishing serotype 4b. The results show that lectins may provide useful probes as diagnostic reagents for listeriae. 相似文献

Tumour necrosis factor (TNF) ligand members and their associated TNF receptor (TNFR) superfamilies have many diverse physiological
roles. TNF is thought to play a critical role in the pathophysiology of a range of diseases including refractory asthma, sepsis,
ankylosing spondylitis, lupus, type II diabetes, multiple sclerosis and psoriasis. The recent continued expansion of the novel
anti-TNF therapeutic agents (etanercept and infliximab) has seen major improvements in the treatment of some inflammatory-based
human diseases including notably rheumatoid arthritis and Crohn’s disease, with other conditions currently being trialled
using anti-TNF agents. The cellular signalling machinery used by TNFRs to achieve their many cellular responses are discussed,
as is the gonadotrophin-releasing hormone (GnRH) receptor signalling mechanisms. TNF is known to have many actions throughout
the body including effects on the hypothalamic-pituitary-adrenal/gonadal axes, with many anti-gonadotrophic effects including
a role in the development of endometriosis. These interactions between TNF, GnRH and gonadotrophs are discussed.

Summary: Diatoms and bacteria have cooccurred in common habitats for hundreds of millions of years, thus fostering specific associations and interactions with global biogeochemical consequences. Diatoms are responsible for one-fifth of the photosynthesis on Earth, while bacteria remineralize a large portion of this fixed carbon in the oceans. Through their coexistence, diatoms and bacteria cycle nutrients between oxidized and reduced states, impacting bioavailability and ultimately feeding higher trophic levels. Here we present an overview of how diatoms and bacteria interact and the implications of these interactions. We emphasize that heterotrophic bacteria in the oceans that are consistently associated with diatoms are confined to two phyla. These consistent bacterial associations result from encounter mechanisms that occur within a microscale environment surrounding a diatom cell. We review signaling mechanisms that occur in this microenvironment to pave the way for specific interactions. Finally, we discuss known interactions between diatoms and bacteria and exciting new directions and research opportunities in this field. Throughout the review, we emphasize new technological advances that will help in the discovery of new interactions. Deciphering the languages of diatoms and bacteria and how they interact will inform our understanding of the role these organisms have in shaping the ocean and how these interactions may change in future oceans. 相似文献

Prostasomes are membranous vesicles (150–200 nm diameter) present in human semen. They are secreted by the prostate gland and contain large amounts of cholesterol, sphingomyelin and Ca2+. In addition, some of their proteins are enzymes. Prostasomes enhance the motility of ejaculated sperm and are involved in a number of biological functions. In a previous work, we found that prostasome can fuse to spermatozoa at slightly acidic pH values, as demonstrated by the transfer of the lipophilic octadecylrhodamine probe. In this paper, we study the interactions of two leukocyte populations (polymorphonuclear and mononuclear) with prostasomes and find a pH-dependent adhesion (revealed by microscopic observation), but no fusion. These phenomena may be relevant for the functions of leukocytes in human reproduction. 相似文献
