Water-soluble polysaccharide fractions VO1–VO4 were isolated from the squeezed berries of the snowball tree (Viburnum opulus) by successive extraction with water at various temperatures and pH and with aqueous solutions of ammonium oxalate. These fractions were purified by ion-exchange chromatography on DEAE cellulose, and the homogeneity of the purified polysaccharides was determined by gel filtration on Sephacryl S-500. Acidic polysaccharides close to pectins in their sugar composition were found in all the extracts (fractions VO1-1, VO2-1, VO3-2, and VO4-2). Residues of galacturonic acid, galactose, arabinose, and (to a lesser extent) rhamnose are their main constituents. Neutral polysaccharides composed mainly of galactose and mannose residues were additionally found in fractions extracted with acidified water (pH 4.0) and with aqueous ammonium oxalate solutions. Partial acidic hydrolysis and digestion with pectinase of acidic polysaccharides indicated that their carbohydrate backbone consists of α-1,4-linked residues ofD-galacturonic acid. NMR spectra of acidic polysaccharides (fractions VO3-2 and VO3-3) confirmed this and demonstrated that their side oligosaccharide chains are composed of β-1,4-linked galactopyranose residues and of terminal and 2,5- and 3,5-substituted residues of α-arabinofuranose at a Gal : Ara ratio of 3 : 1. Some polysaccharides fromV. opulus were found to possess an immunostimulating activity: they enhance phagocytosis, in particular, the phagocytic index and the secretion of lysosomal enzymes with peritoneal macrophages. Calcium ions were found to be necessary for the appearance of the stimulating effect of acidic polysaccharides fromV. opulus.  相似文献

Palmaria decipiens, one of the most abundant red seaweeds of the chilean Antarctic, was collected in King George Island. The hot water extract (26% yield) showed by acid hydrolysis to contain xylose, galactose and traces of glucose. Fractionation with cetrimide gave a soluble neutral xylan and an insoluble fraction. The insoluble fraction afforded an acidic polysaccharide that contained 4.8% of uronic acids, 2.8% of sulfate and 18.9% of protein. Polyacrylamide gel electrophoresis showed that it was homogeneous. The GLC and HPLC analysis of the total acidic hydrolysis products showed that the acidic polysaccharide was composed of the neutral sugars galactose and xylose in the molar ratio 8.2:1.0 and of galacturonic and glucuronic acid in the ratio 1.5:1.0. The second-derivative FT-IR spectrum showed the characteristic amide I, II and III bands of proteins. Alkaline cleavage with 0.1 M NaOH indicated the presence of a glycoprotein with O-glycosidic linkage.Results found in this work suggest that the acidic polysaccharide extracted from Palmaria decipiens is an acidic xylogalactan-protein complex.  相似文献

The polysaccharide of the mucin secreted by the leaves of Drosera capensis is composed of l-arabinose, d-xylose, d-galactose, d-mannose, and d-glucuronic acid in the molar ratio of 3.6:1.0:4.9:8.4:8.2. For structural elucidation, methylation analysis using g.l.c. and g.l.c.-m.s. was performed on the native, the carboxyl-reduced, and the degraded polysaccharides. Partial hydrolysis, periodate oxidation, chromium trioxide oxidation, and uronic acid degradation were also performed on the native and carboxyl-reduced polysaccharides. Partial hydrolysis of the native and carboxyl-reduced polysaccharides gave various oligosaccharides that were characterized and suggest a structure containing a d-glucurono-d-mannan backbone having a repeating unit → 4)-β-d-GlcpA-(1 → 2)-α-d-Manp-(1 →. l-Arabinose and d-xylose are present as nonreducing furanosyl and pyranosyl end-groups, respectively, both attached to O-3 of d-glucuronic acid residues of the backbone. d-Galactose is present as non-reducing pyranosyl end-group linked to O-3 of d-mannose residues.  相似文献

The extracellular polysaccharide of Rhizobium meliloti 201 consists of two acidic polysaccharides, APS-I and APS-II. APS-I is composed of D-glucose, D-mannose and D-glucuronic acid in a molar ratio of 3:3:2, whereas APS-II is composed of D-glucose, D-galactose, D-mannose and pyruvic acid in a molar ratio of 4:3:2:1.APS-II was separated from the extracellular polysaccharide preparation by hydrolysing APS-I to its octasaccharide repeating unit with a specific enzyme. APS-I and APS-II were also separated by treatment with cetylpyridinium chloride and by paper electrophoresis of the depyruvylated polysaccharide.  相似文献

Energy budgets of larval stages of the Chinese green lacewing, Chrysopa sinica (Tjeder) (Neuroptera: Chrysopidae) were determined under laboratory conditions at photo‐period of 14:10 L:D, 27 ± 1°C and 75%± 2% RH. The energy used as ingestion, assimilation, respiration, productivity and feces was constructed for each developmental stage. In addition, under these experimental conditions, the potential of C. sinica as a biological control agent was evaluated according to the ingestion by this predator and the energy content of cotton aphid, Aphis gossypii (Glover) (Homoptera: Aphididae). The larval stage of C. sinica was able to consume 1281.4 1‐day‐old aphids, 1018.7 2‐day‐old aphids, 626.9 3‐day‐old aphids, 393.5 4‐day‐old aphids, 312.1 5‐day‐old aphids or 203.5 9‐day‐old aphids, respectively. No significant difference was detected between the estimated number of aphids consumed by the lacewings using energetic methods and the actual number of aphids consumed by the lacewings in this experiment. Our results showed that C. sinica is an important natural enemy of the cotton aphid, and energetic methods are very useful to quantify biological control efficacy of natural enemies.  相似文献

One new ent‐kaurane diterpenoid, 11β,16α‐dihydroxy‐ent‐kauran‐19‐oic acid ( 1 ), together with eight known analogues 2 – 9 were isolated from the aerial parts of Wedelia prostrata. One of the acidic diterpenoids, kaurenoic acid ( 3 ), was converted to seven derivatives, 10 – 16 . All compounds were evaluated for their cytotoxic activity in vitro against human leukemia (K562), liver (HepG‐2), and stomach (SGC‐7901) cancer cell lines. Only four kaurenoic acid derivatives, 13 – 16 , with 15‐keto and substitutions at C(19) position, exhibited notable cytotoxic activities on these tumor cell lines with IC50 value ranging from 0.05 to 3.71 μm . Compounds 10 – 12 , with oxime on C(15) showed moderate inhibitory effects and compounds 1 – 9 showed no cytotoxicities on them. Structure–activity relationships were also discussed based on the experimental data obtained. The known derivative, 15‐oxokaurenoic acid 4‐piperdin‐1‐ylbutyl ester ( 17 ), induced typical apoptotic cell death in colon SW480 cells upon evaluation of the apoptosis‐inducing activity by flow‐cytometric analysis.  相似文献

Summary Two types of polysaccharides were separated from the extracellular polysaccharide produced by Anabaena flos-aquae A-37 by ion exchange chromatography. The neutral polysaccharide is composed of mainly glucose with minor amounts of xylose in a molar ratio of 8:1. Glucose is believed to constitute the polysaccharide core to which xylose is attached. The acidic polysaccharide is composed of glucose and uronic acid as the major monomers with equal amounts of xylose and ribose as the minor constituents. The molar ratio of the monomers found in the acidic polymer is 6:1:1:10 as glucose: xylose: ribose: uronic acid. Chemical analyses showed that the extracellular polysaccharide consists of more neutral polymer (62%) than the acidic polymer (38%).  相似文献

Aims: Alcaligenes sp. NBRC 14130 was found as a strain hydrolysing a mixture of (±)‐trans‐ and (±)‐cis ethyl chrysanthemates to (1R,3R)‐(+)‐trans‐chrysanthemic acid. The Alcaligenes cells also have hydrolytic activity for 6‐aminohexanoate‐cyclic dimer (6‐AHCD, 1,8‐diazacyclotetradecane‐2,9‐dione). The correlation of function on the enzyme from the Alcaligenes strain with hydrolysis activities for both ethyl chrysanthemate and 6‐AHCD was demonstrated. Methods and Results: The esterase was purified to homogeneity. The purified esterase hydrolysed 20 mmol l?1 ester including the four stereoisomers to the corresponding (+)‐trans acid with a 37% molar conversion of ethyl (+)‐trans chrysanthemate. The esterase showed high hydrolytic activity for various short‐chain fatty acid esters, n‐hexane amide and 6‐AHCD. The amino acid sequence of the Alcaligenes esterase was identical to that of Arthrobacter 6‐AHCD hydrolase (EC 3.5.2.12) and similar to that of fatty acid amide hydrolase (EC 3.5.1.4) from Rattus norvegicus, having both serine and lysine residues of the catalytic site and the consensus motif Gly‐X‐Ser‐X‐Gly. Conclusion: The stereo‐selective hydrolytic activity was found in Alcaligenes sp. NBRC14130 by screening of ethyl chrysanthemate‐hydrolysing activity in micro‐organisms, and the purified esterase also acted on fatty acid esters and amides. Significance and Impact of the Study: This study has demonstrated that there are great differences in the enzymatic properties, amino acid sequence and catalytic motif of esterases in both Alcaligenes and Arthrobacter globiformis with excellent stereo‐selectivity for (+)‐trans‐ethyl chrysanthemate, but the amino acid sequence of Alcaligenes esterase is identical to that of Arthrobacter 6‐AHCD hydrolase.  相似文献

Complementation, endonuclease, strand separation, and packaging assays using mutant TerLλ’s, coupled with bioinformatic information and modeling of its endonuclease, identified five residues, D401, E408, D465, E563, and E586, as critical acidic residues of TerLλ’s endonuclease. Studies of phage and viral TerL nucleases indicate acidic residues participate in metal ion‐binding, part of a two‐ion metal catalysis mechanism, where metal ion A activates a water for DNA backbone hydrolysis. Modeling places D401, D465, and E586 in locations analogous to those of the metal‐binding residues of many phage and viral TerLs. Our work leads to a model of TerLλ’s endonuclease domain where at least three acidic residues from a ~185 residue segment (D401 to E586) are near each other in the structure, forming the endonuclease catalytic center at cosN, the nicking site. DNA interactions required to bring the rotationally symmetric cosN precisely to the catalytic center are proposed to rely on an ~60 residue region that includes a conserved α‐helix for dimerization. Metal ion A, positioned by TerLλ’s acidic D401 and E586, would be placed at cosN for water activation, ensuring high accuracy for DNA backbone hydrolysis.  相似文献

The optimum enzyme-assisted extraction conditions of cup plant (Silphium perfoliatum L.) polysaccharides (CPP) were obtained by response surface methodology (RSM), and were found to be an enzymatic hydrolysis time of 66 min, a liquid-solid ratio of 22 mL/g, pH 5.1, and an enzyme-complex dosage of 1.59 %, allowing a CPP yield of 13.69 %. DEAE-52 cellulose and Sepharose CL-6B columns were used to purify the CPP. A purified polysaccharide component, CPP-1.2, was obtained and further analyzed for its characteristics, antioxidant properties, and hypoglycemic activity. The monosaccharide composition of CPP-1.2 was mannose, galacturonic acid, glucose, galactose, and arabinose with a molar percentage ratio of 2.24:16.34:44.17:24.28:12.97. The 1,1-Diphenyl-2-picrylhydrazyl (DPPH) and 2,2-Azino-bis-(3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS) radical scavenging abilities of CPP-1.2 (1.0 mg/mL) reached 95.20 % and 93.69 % that of ascorbic acid (1.0 mg/mL), respectively. Moreover, CPP-1.2 presented significantly higher inhibitory properties on the activities of α-amylase and α-glucosidase than acarbose at each concentration tested. These results suggest that CPP-1.2 possesses the potential to be used as an antioxidant and hypoglycemic agent.  相似文献

Nineteen known triterpenoids, 1 – 19 , and one known sesquiterpenoid, 20 , were isolated from dammar resin obtained from Shorea javanica K. & V. (Dipterocarpaceae). One of the acidic triterpenoids, dammarenolic acid ( 1 ), was converted to fourteen derivatives, namely, an alcohol, 21 , an aldehyde, 22 , and twelve L ‐amino acid conjugates, 23 – 34 . Compounds 1 – 34 were examined for their inhibitory effects on the induction of Epstein–Barr virus early antigen (EBV‐EA) by 12‐O‐tetradecanoylphorbol 13‐acetate (TPA) in Raji cells, a known primary screening test for antitumor promoters. All of the compounds tested, except for compounds 4, 5, 12 – 14, 16 , and 17 , showed inhibitory effects against EBV‐EA activation with potencies either comparable with or stronger than that of β‐carotene, a known natural antitumor promoter. In addition, (20S)‐20‐hydroxy‐3,4‐secodammara‐4(28),24‐dien‐3‐al ( 22 ) exhibited inhibitory effects on skin tumor promotion in an in vivo two‐stage mouse skin carcinogenesis test based on 7,12‐dimethylbenz[a]anthracene (DMBA) as initiator, and with TPA as promoter. Furthermore, evaluation of the cytotoxic activities of compounds 1 – 34 against human cancer cell lines showed that reduction (i.e., 21 and 22 ) or conjugation with L ‐amino acids (i.e., 23 – 34 ) of compound 1 enhanced the cytotoxicity against human melanoma cell line CRL1579.  相似文献

A sulphated heteropolysaccharide was isolated from a green seaweed, Caulerpa taxifolia, by extraction with acid and purified via its copper complex. Methylation analysis of both the sulphated and desulphated polysaccharides revealed the presence of 1,4-linked xylose, 1,6-linked galactose, 1,4,6-linked mannose and non-reducing galactose end group units which are all devoid of sulphate groups. In addition 1,4-linked galactose units sulphated at C-3 are also present. Quantitative periodate oxidation showed a consumption of 1.30 and 1.60 moles of oxidant per anhydrosugar unit in the sulphated and desulphated polysaccharides respectively. The oxo-polysaccharides after reduction and hydrolysis revealed the presence of glycerol, erythritol and unoxidized galactose in the mol ratio 11.6:5.1:4.9 and 11.2:5.0:1.0 respectively, besides threitol (3.9 mol) in the desulphated polysaccharide.  相似文献

Morphological and structural characteristics of photosynthetic organs, diurnal changes in photosynthetic and transpiration rates, and the efficiency of water use were studied in three plant species from mountain-steppe ecosystems in Mongolia, Ephedra sinica Stapf, Stipa glareosa P. Smirn., and Allium polyrhizum Furcz. ex Regel. The species studied differed in the structural and functional mechanisms for the adaptation of photosynthetic apparatus to arid conditions. E. sinica has thick, vertical assimilating shoots, which are characterized by a high density (620 mg/cm3) and a small proportion of photosynthetic tissues (13%). The proportion of meso-phyll in the leaves of A. polyrhizum and S. glareosa was two and three times higher, respectively. The low content of phototrophic tissues in E. sinica shoot was compensated for by a high photosynthetic activity of single chloroplasts (25 mg CO2/(109 chloroplast h)), which was six times higher, than in two other species. Daily course of photosynthesis and transpiration inE. sinica differed from those of A. polyrhizum and S. glareosa by the absence of the midday depression. E. sinica had the highest efficiency of water use (45 mg CO2/g H2O) due to a low transpiration rate (0.25 g/g fr wt h). It is concluded that, in E. sinica, the main strategy for adaptation to arid stress is to develop in the shoot a few photosynthesizing cells of high assimilation activity. Such structural organization of photosynthetic organs in ephedra contributes to its higher efficiency of water use and stability of physiological characteristics under changing environmental conditions. These specific features of the structure of assimilating organs and their functional features contribute to a greater expansion of E. sinica with increasing climate aridization in Mongolia.  相似文献

Four neutral polysaccharides (ESBP1-1, ESBP1-2, ESBP2-1 and ESBP3-1) were successfully purified from the water extracted crude polysaccharides of Erythronium sibiricum bulbs through the combination of DEAE Sepharose CL-6B and Sephadex G-100 chromatography; their average molecular weights were 1.3?×?104, 1.7?×?104, 9.4?×?105 and 4.1?×?105 Da, respectively. Monosaccharide component analysis indicated that ESBP1-1 and ESBP1-2 were mainly composed of glucose (Glc). ESBP2-1 was composed of Glc, galactose (Gal) and arabinose, with a molar ratio of 24.3:1.1:1, whereas ESBP3-1 comprised Glc and Gal at a molar ratio of 14.8:1. In-vitro study showed that all of the four polysaccharides were able to considerably promote the proliferation and neutral red phagocytosis of RAW 264.7 macrophage cell. They could also stimulate the production of the cell lines’ secretory molecules [nitric oxide, tumour necrosis factor-α (TNF-α) and interleukin-1β (IL-1β)] in a dose-dependent manner. However, ESBP1-2 was not included in IL-1β. Overall, these results suggested that polysaccharides from E. sibiricum bulbs can be developed as immunomodulatory ingredients for complementary medicines or functional foods. However, further animal or clinical studies are required.

丽豆(Calophaca sinica)是我国华北地区特有的一种珍稀野生植物。为探明丽豆的营养价值,该文以大豆(Glycine max)为参照组,利用液相色谱-质谱联用(LC-MS)技术对其种子进行了比较代谢组学研究。结果表明:(1)丽豆和大豆中共检测到1 857种代谢产物,二者成分相同且含量相似的代谢物有1 698种(>90%),差异代谢物有159种(<10%)。(2)在差异代谢物中,成分差异的有9种,其中有5种为丽豆特有,剩余150种均为含量差异,其中48种(约30%)在丽豆中的含量高于大豆。(3) KEGG注释到8条差异代谢物显著富集(P<0.1)的通路,主要包括初生代谢物的各类氨基酸生物合成途径和次生代谢物的罗汉松脂素、花生四烯酸以及二萜类等生物合成途径。(4)丽豆比大豆含量低的化学组分主要是初生代谢产物,比大豆含量高的化学组分主要是次生代谢物,而这些次生代谢物在调节血糖、骨坏损修复、增强免疫以及消炎抗癌等生理过程中有着积极的作用。综上所述,该研究认为丽豆与大豆具有相近的营养价值,并且对改善人类亚健康状况有积极的影响;此外,该文使我们对丽豆的营养价值和代谢组成...  相似文献

Bacteriophages recognize and bind to their hosts with the help of receptor‐binding proteins (RBPs) that emanate from the phage particle in the form of fibers or tailspikes. RBPs show a great variability in their shapes, sizes, and location on the particle. Some RBPs are known to depolymerize surface polysaccharides of the host while others show no enzymatic activity. Here we report that both RBPs of podovirus G7C – tailspikes gp63.1 and gp66 – are essential for infection of its natural host bacterium E. coli 4s that populates the equine intestinal tract. We characterize the structure and function of gp63.1 and show that unlike any previously described RPB, gp63.1 deacetylates surface polysaccharides of E. coli 4s leaving the backbone of the polysaccharide intact. We demonstrate that gp63.1 and gp66 form a stable complex, in which the N‐terminal part of gp66 serves as an attachment site for gp63.1 and anchors the gp63.1‐gp66 complex to the G7C tail. The esterase domain of gp63.1 as well as domains mediating the gp63.1‐gp66 interaction is widespread among all three families of tailed bacteriophages.  相似文献

The mucilage found in the stem pith of Actinidia deliciosa contains d-glucuronic acid, d-mannose, l-fucose, l-arabinose, and d-galactose in the molar ratios 1.0:1.5:2.0:4.0. The native, carboxyl-reduced, and partially acid-hydrolysed polysaccharides were subjected to methylation analysis. Partial acid hydrolysis of the methylated, carboxyl-reduced glucuronomannan core produced a series of methylated oligosaccharides which, as their alditol derivatives, were isolated by reverse-phase h.p.l.c. and characterised by e.i.- and f.a.b.-m.s. The data suggest that the polysaccharide contains a →4)-β-d-GlcpA-(1→2)-α-d-Manp-(1→ backbone with most of the d-mannosyl and d-glucosyluronic acid residues substituted through positions 3 with oligosaccharides containing l-arabinose, α-l-fucose, and β-d-galactose.  相似文献

A chemical investigation of the endolichenic fungus Aspergillus versicolor (125a), which was found in the lichen Lobaria quercizans, resulted in the isolation of four novel diphenyl ethers, named diorcinols F–H ( 1 – 3 , resp.) and 3‐methoxyviolaceol‐II ( 4 ), eight new bisabolane sesquiterpenoids, named (?)‐(R)‐cyclo‐hydroxysydonic acid ( 5 ), (?)‐(7S,8R)‐8‐hydroxysydowic acid ( 6 ), (?)‐(7R,10S)‐10‐hydroxysydowic acid ( 7 ), (?)‐(7R,10R)‐iso‐10‐hydroxysydowic acid ( 8 ), (?)‐12‐acetoxy‐1‐deoxysydonic acid ( 9 ), (?)‐12‐acetoxysydonic acid ( 10 ), (?)‐12‐hydroxysydonic acid ( 11 ), and (?)‐(R)‐11‐dehydrosydonic acid ( 12 ), two new tris(pyrogallol ethers), named sydowiols D ( 13 ) and E ( 14 ), and fifteen known compounds, 15 – 29 . All of the structures were determined by spectroscopic analyses, and a number of them were further identified through chemical transformations and electronic circular dichroism (ECD) calculations. Preliminary bioassays of these isolates for the determination of their inhibitory activities against the fungus Candida albicans, and their cytotoxicities against the human cancer cell lines PC3, A549, A2780, MDA‐MB‐231, and HEPG2 were also evaluated.  相似文献
