The island rule is a hypothesis whereby small mammals evolve larger size on islands while large insular mammals dwarf. The rule is believed to emanate from small mammals growing larger to control more resources and enhance metabolic efficiency, while large mammals evolve smaller size to reduce resource requirements and increase reproductive output. We show that there is no evidence for the existence of the island rule when phylogenetic comparative methods are applied to a large, high-quality dataset. Rather, there are just a few clade-specific patterns: carnivores; heteromyid rodents; and artiodactyls typically evolve smaller size on islands whereas murid rodents usually grow larger. The island rule is probably an artefact of comparing distantly related groups showing clade-specific responses to insularity. Instead of a rule, size evolution on islands is likely to be governed by the biotic and abiotic characteristics of different islands, the biology of the species in question and contingency. 相似文献

Cytogenetic studies involving the family Prochilodontidae have shown that these fish can be characterized by a constant diploid
number and a conserved karyotypic macrostructure. This study focused on comparative physical chromosomal mapping using 18S
and 5S rDNA to compare the species Semaprochilodus insignis and S. taeniurus. Our results indicated the conservation of large number of conventional chromosomal markers. The molecular cytogenetic analyses
of the location of the 18S rDNA indicated the maintenance of a chromosome pair bearing these sites in both species analyzed,
and it appears to be a conserved character among the majority of the species of this family. The stability of the number of
5S ribosomal DNA sites and their chromosomal localization as has been reported for the Prochilodontidae was not, however,
confirmed for S. insignis and S. taeniurus, as these species showed multiple specific rDNA 5S sites. As such, and in spite of the fact that a number of studies indicate
that the family Prochilodontidae has a conserved karyotypic structure, the utilization of molecular tools that use chromosomal
segments as markers revealed that this presumed stability cannot be extended to the genome level for the species S. insignis and S. taeniurus. 相似文献

Temperature is an important environmental factor that influences key traits like body size, growth rate and maturity. Ectotherms reared under high temperatures usually show faster growth, but reach a smaller final size, a phenomenon known as the temperature-size rule (TSR). Oxygen may become a limiting resource at high temperatures, when demand for oxygen is high, especially in water as oxygen uptake is far more challenging under water than in air. Therefore, in aquatic ectotherms, the TSR might very well be mediated by temperature effects on oxygen availability and oxygen demand. To distinguish between the direct effects of temperature and oxygen mediated effects, growth rate and final size were measured in the aquatic ectotherm Asellus aquaticus (Linnaeus, 1758) reared under different temperature and oxygen conditions in a factorial design. Growth could be best described by a modified Von Bertalanffy growth function. Both temperature and oxygen affected age at maturity and growth. Growth responses to temperature were dependent on oxygen conditions (interactive effect of temperature and oxygen). Only under hypoxic conditions, when oxygen was most limiting, did we find a classic TSR. Moreover, when comparing treatments differing in temperature, but where the balance between oxygen demand and supply was similar, high temperature increased both growth rate and final size. Thus effects of oxygen may resolve the life-history puzzle of the TSR in aquatic ectotherms. 相似文献

The RecG protein of Escherichia coli is a double-stranded DNA translocase that unwinds a variety of branched DNAs in vitro, including Holliday junctions, replication forks, D-loops and R-loops. Coupled with the reported pleiotropy of recG mutations, this broad range of potential targets has made it hard to pin down what the protein does in vivo, though roles in recombination and replication fork repair have been suggested. However, recent studies suggest that RecG provides a more general defence against pathological DNA replication. We have postulated that this is achieved through the ability of RecG to eliminate substrates that the replication restart protein, PriA, could otherwise exploit to re-replicate the chromosome. Without RecG, PriA triggers a cascade of events that interfere with the duplication and segregation of chromosomes. Here we review the studies that led us to this idea and to conclude that RecG may be both a specialist activity and a general guardian of the genome. 相似文献

In the current study we investigated the molecular mechanisms of cytotoxicity of amyloid oligomers of horse milk lysozyme. We have shown that lysozyme forms soluble amyloid oligomers and protofibrils during incubation at pH 2.0 and 4.5 and 57 degrees C. These structures bind the amyloid-specific dyes thioflavin T and Congo Red, and their morphology and size were analyzed by atomic force microscopy. Monomeric lysozyme and its fibrils did not affect the viability of three cell types used in our experiments including primary murine neurons and fibroblasts, as well as neuroblastoma cell line IMR-32. However, soluble amyloid oligomers of lysozyme caused death of all these cell types, as estimated by flow-cytometry counting dead cells stained with ethidium bromide. The primary cell cultures appeared to be more sensitive to amyloid than neuroblastoma cell line IMR-32. Amyloid cytotoxicity depends on the size of oligomeric particles: samples containing 20-mers formed at pH 4.5 were more toxic than tetramers and octamers present in the solution at pH 2.0. Soluble amyloid oligomers can self-assemble into doughnut-like structures; however, no correlation was observed between the amount of the doughnut-like structures in the sample and its cytotoxicity. The fact that the intermediate oligomers of such an abundant protein as lysozyme display cytotoxicity confirms a hypothesis that cytotoxicity is a common feature of protein amyloid. Inhibition of intermediate oligomer formation is crucial in preventing amyloid pathogeneses. 相似文献

The inositol pyrophosphate, diphosphoinositol pentakisphosphate (IP7), is thought to negatively regulate the critical insulin signaling protein Akt/PKB. Knockdown of the IP7-generating inositol hexakisphosphate kinase 1 (IP6K1) results in a concomitant increase in signaling through Akt/PKB in most cell types so far examined. Total in vivo knockout of IP6K1 is associated with a phenotype resistant to high-fat diet, due to enhanced Akt/PKB signaling in classic insulin regulated tissues, counteracting insulin resistance. In contrast, we have shown an important positive role for IP6K1 in insulin exocytosis in the pancreatic β-cell. These cells also possess functional insulin receptors and the feedback loop following insulin secretion is a key aspect of their normal function. Thus we examined the effect of silencing IP6K1 on the activation of Akt/PKB in β-cells. Silencing reduced the glucose-stimulated increase in Akt/PKB phosphorylation on T308 and S473. These effects were reproduced with the selective pan-IP6K inhibitor TNP. The likely explanation for IP7 reduction decreasing rather than increasing Akt/PKB phosphorylation is that IP7 is responsible for generating the insulin signal, which is the main source of Akt/PKB activation. In agreement, insulin receptor activation was compromised in TNP treated cells. To test whether the mechanism of IP7 inhibition of Akt/PKB still exists in β-cells, we treated them at basal glucose with an insulin concentration equivalent to that reached during glucose stimulation. TNP potentiated the Akt/PKB phosphorylation of T308 induced by exogenous insulin. Thus, the IP7 regulation of β-cell Akt/PKB is determined by two opposing forces, direct inhibition of Akt/PKB versus indirect stimulation via secreted insulin. The latter mechanism is dominant, masking the inhibitory effect. Consequently, pharmacological strategies to knock down IP6K activity might not have the same positive output in the β-cell as in other insulin regulated tissues. 相似文献

Marine Turtles as Sentinels of Ecosystem Health: Is Fibropapillomatosis an Indicator? 总被引:1,自引:0,他引:1
Marine turtle fibropapillomatosis (FP) is a disease primarily affecting green turtles (Chelonia mydas) that is characterized by multiple cutaneous masses. In addition, the condition has been confirmed in other species of sea turtles. The disease has a worldwide, circumtropical distribution and has been observed in all major oceans. Although reported since the late 1930s in Florida, it was not until the late 1980s that it reached epizootic proportions in several sea turtle populations. Long-term studies have shown that pelagic turtles recruiting to near shore environments are free of the disease. After exposure to these benthic ecosystems, FP manifests itself with primary growths in the corner of the eyes spreading to other epithelial tissue. One or more herpesviruses, a papillomavirus, and a retrovirus have been found associated with tumors using electron microscopy and molecular techniques; however, the primary etiological agent remains to be isolated and characterized. Field observations support that the prevalence of the disease is associated with heavily polluted coastal areas, areas of high human density, agricultural runoff, and/or biotoxin-producing algae. Marine turtles can serve as excellent sentinels of ecosystem health in these benthic environments. FP can possibly be used as an indicator but correlations with physical and chemical characteristics of water and other factors need to be made. Further research in identifying the etiologic agent and its association with other environmental variables can provide sufficient parameters to measure the health of coastal marine ecosystems, which serve not only as ecotourism spots but also as primary feeding areas for sea turtles. 相似文献

Insular populations and their closest mainland counterparts commonly display body size differences that are considered to fit the island rule, a theoretical framework to explain both dwarfism and gigantism in isolated animal populations. The island rule is used to explain the pattern of change of body size at the inter-specific level. But the model implicitly makes also a prediction for the body size of isolated populations of a single species. It suggests that, for a hypothetical species covering a wide range of island sizes, there exists a specific island size where this species reaches the largest body size. Body size would be small (in relative terms) in the smallest islets of the species range. It would increase with island size, and reach a maximum at some specific island size. However, additional increases from such a specific island size would instead promote body size reduction, and small (in relative terms) body sizes would be found again on the largest islands. The biogeographical patterns predicted by the island rule have been described and analysed for vertebrates only (mainly mammals), but remain largely untested for insects or other invertebrates. I analyse here the pattern of body size variation between seven isolated insular populations of a flightless beetle, Asida planipennis (Coleoptera, Tenebrionidae). This is an endemic species of Mallorca, Menorca and a number of islands and islets in the Balearic archipelago (western Mediterranean). The study covers seven of the 15 known populations (i.e., there are only 15 islands or islets inhabited by the species). The populations studied fit the pattern advanced above and we could, therefore, extrapolate the island rule to a very different kind of organism. Community ecology entered the 1970s with the belief that niche theory would supply a general theory of community structure. The lack of wide-spread empirical support for niche theory led to a focus on models specific to classes of communities such as lakes, intertidal communities, and forests. Today, the needs of conservation biology for metrics of “ecological health” that can be applied across types of communities prompts a renewed interest in the possibility of general theory for community ecology. Disputes about the existence of general patterns in community structure trace at least to the 1920s and continue today almost unchanged in concept, although now expressed through mathematical modeling. Yet, a new framework emerged in the 1980s from findings that community composition and structure depend as much on the processes that bring species to the boundaries of a community as by processes internal to a community, such as species interactions and co-evolution. This perspective, termed “supply-side ecology”, argued that community ecology was to be viewed as an “organic earth science” more than as a biological science. The absence of a general theory of the earth would then imply a corresponding absence of any general theory for the communities on the earth, and imply that the logical structure of theoretical community ecology would consist of an atlas of models special to place and geologic time. Nonetheless, a general theory of community ecology is possible similar in form to the general theory for evolution if the processes that bring species to the boundary of a community are analogized to mutation, and the processes that act on the species that arrive at a community are analogized to selection. All communities then share some version of this common narrative, permitting general theorems to be developed pertaining to all ecological communities. Still, the desirability of a general theory of community ecology is debatable because the existence of a general theory suppresses diversity of thought even as it allows generalizations to be derived. General practitioners and psychiatrists communicate mainly by letter. To ascertain the most important items of information that should be included in these letters ("key items") questionnaires were sent to 80 general practitioners and 80 psychiatrists. A total of 120 referral letters sent to psychiatric clinics in 1973 and 1983 were studied, together with the psychiatrists'' replies, and these were rated for the inclusion of "key items." General practitioners'' letters contain less information about the family but more about psychiatric history than they did a decade ago. Overall, psychiatrists'' letters have not changed. Registrars, however, now include noticeably more "key items" than they did 10 years ago, but their letters remain twice the length of those written by consultants. It is suggested that letter writing skills are vital to good patient management and should be taught to postgraduate trainees in general practice and psychiatry. 相似文献

The majority of ectotherms mature at a larger size at lower rearing temperatures. Although this temperature-size rule is well established, a general explanation for this phenomenon has remained elusive. In this article, we address the problem by exploring the proximate and ultimate reasons for why a temperate grasshopper, Chorthippus brunneus, is an exception to the temperature-size rule. Using a complete set of life-history data to parameterize an established life-history model, we show that it is optimal for this species to mature at a larger size at higher temperatures. We also show that plasticity in adult size is determined by the relative difference between the minimum temperature thresholds for growth and development rates. The mechanism relates to aspects of the biophysical model of van der Have and de Jong. Ectotherms that obey the temperature-size rule are identified as having a higher temperature threshold for development rate than for growth rate; exceptions are identified as having a lower temperature threshold for development rate than for growth rate. The latter scenario may arise broadly in two ways. These are discussed in reference to the thermal biology of temperate grasshoppers and ectotherms in general. 相似文献

Our experiments and studies of a few other authors demonstrated that L-type calcium channels and mitochondria are involved in the induction of post-tetanic potentiation (PTP) in a number of preparations (Aplysia central nervous system, hippocampal cell cultures, crayfish neuromuscular junctions, etc.). We extend this conclusion on cortical synapses by the demonstration that inhibitors of mitochondrial Ca2+ uptake and release suppress PTP in rat neocortical cell cultures. The central‐marginal hypothesis (CMH) posits that range margins exhibit less genetic diversity and greater inter‐population genetic differentiation compared to range cores. CMH predictions are based on long‐held “abundant‐centre” assumptions of a decline in ecological conditions and abundances towards range margins. Although much empirical research has confirmed CMH, exceptions remain almost as common. We contend that mangroves provide a model system to test CMH that alleviates common confounding factors and may help clarify this lack of consensus. Here, we document changes in black mangrove (Avicennia germinans) population genetics with 12 nuclear microsatellite loci along three replicate coastlines in the United States (only two of three conform to underlying “abundant‐centre” assumptions). We then test an implicit prediction of CMH (reduced genetic diversity may constrain adaptation at range margins) by measuring functional traits of leaves associated with cold tolerance, the climatic factor that controls these mangrove distributional limits. CMH predictions were confirmed only along the coastlines that conform to “abundant‐centre” assumptions and, in contrast to theory, range margin A. germinans exhibited functional traits consistent with greater cold tolerance compared to range cores. These findings support previous accounts that CMH may not be a general rule across species and that reduced neutral genetic diversity at range margins may not be a constraint to shifts in functional trait variation along climatic gradients. 相似文献

Intrinsically disordered proteins (IDPs) have been implicated in a number of human diseases, including cancer, diabetes, neurodegenerative and cardiovascular disorders. Although for some of these conditions molecular mechanisms are now better understood, the big picture connecting distinct structural properties and functional repertoire of IDPs to pathogenesis and disease progression is still incomplete. Recent studies suggest that signaling and regulatory roles carried out by IDPs require them to be tightly regulated, and that altered IDP abundance may lead to disease. Here, we propose another link between IDPs and disease that takes into account disease-associated missense mutations located in the intrinsically disordered regions. We argue that such mutations are more prevalent and have larger functional impact than previously thought. In addition, we demonstrate that deleterious amino acid substitutions that cause disorder-to-order transitions are particularly enriched among disease mutations compared to neutral polymorphisms. Finally, we discuss potential differences in functional outcomes between disease mutations in ordered and disordered regions, and challenge the conventional structure-centric view of missense mutations. 相似文献

As far as multiculturalism is at stake, three kinds of question arise at the more or less confusing meeting point of sociology, political science and political philosophy: What are the sources and meanings of cultural difference in our societies? In what way do institutions and policy-makers in some countries deal with multiculturalism? Why should we favour or not favour multiculturalism? This article tackles these questions in turn and seeks to answer them. Cultural differences are not only reproduced, they are in the constant process of being produced which means that fragmentation and recomposition are a permanent probability. In such a situation, the problem is how to broaden democracy in order to avoid at one and the same time the tyranny of the majority and the tyranny of the minorities. 相似文献
