A sample set of registered perennial ryegrass varieties was used to compare how morphological characterisation and AFLP® (AFLP® is a registered trademark of Keygene N.V.) and STS molecular markers described variety relationships. All the varieties were confirmed as morphologically distinct, and both the STS and AFLP markers exposed sufficient genetic diversity to differentiate these registered ryegrass varieties. Distances obtained by each of the approaches were compared, with special attention given to the coincidences and divergences between the methods. When correlations between morphological, AFLP and STS distances were calculated and the corresponding scatter-plots constructed, the variety relationships appeared to be rather inconsistent across the methods, especially between morphology and the molecular markers. However, some consistencies were found for closely related material. An implication could be that these molecular-marker techniques, while not yet suited to certain operations in the traditional registration of new varieties, could be suitable methods for investigating disputable distinctness situations or possible EDV (EDV= essentially derived variety. An EDV is a variety being clearly distinct from, but conforming in the expression of the essential characteristics of, an ’initial variety’ (IV) from which it is found to have been predominantly derived) relationships, subject to establishing standardised protocols and statistical techniques. Some suggestions for such a protocol, including a statistical test for distinctness, are given.  相似文献

Distinctness, uniformity and stability (DUS) testing of varieties is usually required to apply for Plant Breeders’ Rights. This exam is currently carried out using morphological traits, where the establishment of distinctness through a minimum distance is the key issue. In this study, the possibility of using microsatellite markers for establishing the minimum distance in a vegetatively propagated crop (grapevine) has been evaluated. A collection of 991 accessions have been studied with nine microsatellite markers and pair-wise compared, and the highest intra-variety distance and the lowest inter-variety distance determined. The collection included 489 different genotypes, and synonyms and sports. Average values for number of alleles per locus (19), Polymorphic Information Content (0.764) and heterozygosities observed (0.773) and expected (0.785) indicated the high level of polymorphism existing in grapevine. The maximum intra-variety variability found was one allele between two accessions of the same variety, of a total of 3,171 pair-wise comparisons. The minimum inter-variety variability found was two alleles between two pairs of varieties, of a total of 119,316 pair-wise comparisons. In base to these results, the minimum distance required to set distinctness in grapevine with the nine microsatellite markers used could be established in two alleles. General rules for the use of the system as a support for establishing distinctness in vegetatively propagated crops are discussed.  相似文献

 A total of 145 maize inbred lines, representative of material released in France, were differentiated using RFLP markers and a set of discriminant morphological traits in order to evaluate the use of molecular markers for large-scale germplasm diversity analysis and determination of distinctness. Several criteria are proposed with respect to choice of probes, which should give reliable results for routine studies and have a known single-locus genetic determinism to avoid redundancy. A method is proposed by which to incorporate the data from different restriction enzymes obtained with the same probe. The precision of the estimation of the genetic distance is given. The relationship between molecular and morphological distances appears to be triangular, molecular divergence behaving as a limiting factor for morphological divergence. This study was carried out to assess the potential of SSR markers for variety identification by comparing SSR markers and morphological traits in tests of distinctiveness, uniformity, and stability (DUS) of pepper (Capsicum annuum L.) varieties. Twenty-seven SSR markers were polymorphic in 66 pepper varieties, revealing a total of 89 alleles. Average polymorphism information content (PIC) value was 0.529, ranging from 0.03 to 0.877. Cluster analysis of the band patterns separated the varieties into three groups corresponding to varietal types. Morphological trait-based clustering showed some degree of similarity to dendrogram topologies based on the SSR index. However, no significance correlation was found between the SSR and morphological data. SSR markers could be used to complement a DUS test of a candidate variety and to select complimentary varieties by pre-screening existing varieties in the context of protecting new varieties of pepper.  相似文献

依据菊花DUS测试指南中性状观测和分级的方法,记录了菊属、亚菊属共31个物种叶片和花序的共计8个定性多态性状,并测量了各物种叶片的14个形态指标,将其转换为7个数量多态性状,分别基于定性多态性状代码和叶片数量多态性状数值利用聚类分析研究其亲缘关系,将两次聚类结果进行比较。结果发现,基于定性多态性状和叶片数量多态性状的聚类结果均可以对菊花近缘属植物进行亲缘关系分析,并与前人利用分子标记的亲缘关系研究及植物学传统分类的研究结果一致,能够较准确地反映种间亲缘关系。而利用菊花DUS测试相关定性多态性状的信息进行亲缘关系研究,具有准确度高、采样简便、工作量小的优点。  相似文献

SRAP标记与形态学标记在西瓜DUS测试中的比较   总被引:2,自引:0,他引:2  
DUS(特异性、一致性和稳定性)测试是进行新品种申请的必要步骤。本文以28个不同的西瓜品种为研究对象,分别采用21对SRAP引物标记和54个用于DUS测试的形态学标记对其进行遗传多样性分析,其中SRAP引物在不同品种间的多态信息含量(PIC)在52.5%~89.2%之间,平均值为72.0%,计算得到的各材料间相似系数在0.92至0.99之间,而形态学标记统计得到各材料间相关系数在0.50到0.85之间。采用UPGMA法对所有材料进行聚类分析,SRAP分子标记聚类划分成四类,形态学标记将其划分为五类。对两种标记所得的结果进行相关性分析得出两者的相关系数为0.218,表明形态学标记和SRAP标记在这些材料上表现的相关性不是很高,但在品种鉴定和区别上SRAP标记表现出一定的优势,可以作为DUS测试的一种有益补充。  相似文献

Molecular markers linked to phenotypically important traits are of great interest especially when traits are difficult and/or costly to be observed. In tomato where a strong focus on resistance breeding has led to the introgression of several resistance genes, resistance traits have become important characteristics in distinctness, uniformity and stability (DUS) testing for Plant Breeders Rights (PBR) applications. Evaluation of disease traits in biological assays is not always straightforward because assays are often influenced by environmental factors, and difficulties in scoring exist. In this study, we describe the development and/or evaluation of molecular marker assays for the Verticillium genes Ve1 and Ve2, the tomato mosaic virus Tm1 (linked marker), the tomato mosaic virus Tm2 and Tm2 2 genes, the Meloidogyne incognita Mi1-2 gene, the Fusarium I (linked marker) and I2 loci, which are obligatory traits in PBR testing. The marker assays were evaluated for their robustness in a ring test and then evaluated in a set of varieties. Although in general, results between biological assays and marker assays gave highly correlated results, marker assays showed an advantage over biological tests in that the results were clearer, i.e., homozygote/heterozygote presence of the resistance gene can be detected and heterogeneity in seed lots can be identified readily. Within the UPOV framework for granting of PBR, the markers have the potential to fulfil the requirements needed for implementation in DUS testing of candidate varieties and could complement or may be an alternative to the pathogenesis tests that are carried out at present.  相似文献

To assess the potential of multiplex SSR markers for testing distinctness, uniformity and stability of rape (Brassica napus L.) varieties, we developed three multiplex SSR sets composed of five markers each. These were used to measure the extent of diversity within and between a set of ten varieties using a fluorescence-based semi-automated detection technology. Also, we evaluated the significance of any correlation between SSRs, pedigree and five of the morphological characters currently used for statutory distinctness, uniformity and stability testing of rape varieties. An assignment test was allowed to identify 99% of the plants examined, with the correct variety based on the analysis of 48 individual plants for each variety. Principal coordinate analysis confirmed that a high degree of separation between varieties could be achieved. Varieties were separated in three groups corresponding to winter, spring and forage types. These results suggested that it should be possible to select a set of markers for obtaining a suitable separation. Diversity within varieties varied considerably, according to the variety and the locus examined. No significant correlation was found between SSR and morphological data. However, genetic distances measured by SSRs were correlated to pedigree. These results suggested that SSRs could be used for pre-screening or grouping of existing and candidate varieties, allowing the number of varieties that need to be grown for comparison to be reduced. Multiplex SSR sets gave high-throughput reproducible results, thus reducing the costs of SSR assessment. Multiplex SSR sets are a promising way forward for complementing the current variety testing system in B. napus.  相似文献

对水稻DUS测试标准品种进行多样性评价,为DUS测试标准品种的选择和优化提供依据。以DUS测试指南中规定的49个水稻标准品种为材料,根据2008年在杭州田间试验对59个形态性状的调查数据,利用PopGen32软件计算了等位变异数、有效等位变异数和Shannon's多样性信息指数,对标准品种的形态性状多样性进行了评价;并根据其多样性参数采用NTSYS软件对标准品种进行了聚类分析。结果表明,59个形态性状在19份籼稻和30份粳稻品种中分别检测到201和262个等位变异,平均每个性状可以检测到3.4068(籼稻)和4.4407(粳稻)个等位变异,籼、粳稻类群中59个性状的平均Shannon's多样性指数分别为0.8318(0~1.7674)和0.9828(0~1.8547)。聚类分析结果表明,籼、粳稻群体内品种间的形态性状相似系数分别介于0.45~0.81和0.36~0.76,取相似系数0.70为阈值,可将19份籼稻和30份粳稻品种分别分为16和27类。多样性分析表明本研究的49个品种形态性状多样性丰富,具有标准品种性状描述的示例和校正作用;根据聚类分析结果,可将现有标准品种的数量减少至43个。本研究为水稻新品种DUS测试中标准品种的选择和优化提供了参考依据,同时也可供研制其他植物的DUS测试标准借鉴。  相似文献

Genotype-fitness correlations (GFC) have previously been studied using allozyme markers and have often focused on short-term processes such as recent inbreeding. Thus, models of GFC usually neglect marker mutation and only use heterozygosity as a genotypic index. Recently, GFC have also been reported (i) with DNA markers such as microsatellites, characterized by high mutation rates and specific mutational processes and (ii) using new individual genotypic indices assumed to be more precise than heterozygosity. The aim of this article is to evaluate the theoretical impact of marker mutation on GFC. We model GFC due to short-term processes generated by the current breeding system (partial selfing) and to long-term processes generated by past population history (hybridization). Various mutation rates and mutation models corresponding to different kinds of molecular markers are considered. Heterozygosity is compared to other genotypic indices designed for specific marker types. Highly mutable markers (such as microsatellites) are particularly suitable for the detection of GFC that evolve in relation to short-term processes, whereas GFC due to long-term processes are best observed with intermediate mutation rates. Irrespective of the marker type and population scenario, heterozygosity usually provides higher correlations than other genotypic indices under most biologically plausible conditions.  相似文献

本研究采用代表最大限度西瓜遗传多样性的SSR核心引物组合,分析了西瓜DUS测试指南中的24份标准品种遗传多样性与核酸指纹。以基于重测序获得的SNP标记构建的17份西瓜材料的系统发育树为参照,对24份标准品种进行了遗传多样性分析,在遗传相似系数0.80处将24份标准品种分为3大类群,分析表明:核酸指纹分类比传统形态学分类更为准确。采用二维(QR)编码构建了西瓜24份标准品种的SSR指纹图谱,并利用本技术以保护品种“京欣2号”与对照品种“京欣1号”为例,进行了DUS分子鉴定测试,共扩增32个SSR位点,“京欣1号”和“京欣2号”之间存在4个位点的差异,品种间遗传相似系数为0.89,比形态学鉴定的差异位点更多且更准。本研究建立的西瓜DUS标准品种SSR指纹图谱与分子检测技术,可以应用到西瓜品种DUS分子检测实践,同时也为西瓜品种纯度与真实性鉴定及遗传背景分析提供了技术方案。  相似文献

In the present study, a strategy was proposed for constructing plant core subsets by clusters based on the combination of continuous data for genotypic values and discrete data for molecular marker InformaUon. A mixed linear model approach was used to predict genotyplc values for eliminating the environment effect. The "mixed genetic distance" was designed to solve the difficult problem of combining continuous and discrete data to construct a core subset by cluster. Four commonly used genetic distances for continuous data (Euclidean distance, standardized Euclidean distance, city block distance, and Mahalanobls distance) were used to assess the validity of the conUnuous data part of the mixed genetic distance; three commonly used genetic distances for discrete data (cosine distance, correlaUon distance, and Jaccard distance) were used to assess the validity of the discrete data part of the mixed genetic distance, A rice germplasm group with eight quantitative traits and information for 60 molecular markers was used to evaluate the validity of the new strategy. The results suggest that the validity of both parts of the mixed geneUc distance are equal to or higher than the common geneUc distance. The core subset constructed on the basis of a combination of data for genotyplc values and molecular marker information was more representative than that constructed on the basis of data from genotypic values or molecular marker informaUon alone. Moreover, the strategy of using combined data was able to treat dominant marker informaUon and could combine any other continuous data and discrete data together to perform cluster to construct a plant core subset.  相似文献

Summary  We evaluated concordance of AFLP and RAPD markers for estimating genetic distances of 47 pepper inbred lines belonging to five varietal types. It enabled us to see the efficiency of these markers for identification, estimation of distances between varieties and variety discrimination. Genetic distance and multidimensional scaling results showed a general agreement between AFLP and RAPD markers. Based on pattern scores, dendrograms were produced by the UPGMA method. Phenetic trees based on molecular data were consistent with the classification of variety group. The precision of the estimation of the genetic distance was given. The molecular genetic distances were correlated with distances based on a set of discriminating agronomic traits measured for identification and distinctiveness tests. The relationship between molecular and morphological distances appeared to be triangular. These results and their implications in the cultivar protection purposes of pepper hybrids are discussed. We estimated the genetic distances among 10 spring wheat genotypes based on pedigree data, morphological traits and AFLP markers, used individually and combined with morphological traits, to find the best predictors of general- and specific-combining abilities among parental genotypes. Ten wheat parents were crossed in a diallel form, disregarding reciprocal hybrids, totaling 45 combinations. The F(1) hybrids, F(2) populations and parents were evaluated in the field in 2007. The experimental plots consisted of 20 plants for F(1) hybrids and 40 plants for parental and F(2) populations. All methods (pedigree data, AFLP markers and morphological traits, used individually and combined) were found to be useful for the assessment of genetic diversity. The significant coefficient correlations ranged from low (0.45) to moderate (0.67) between the distance measures and hybrid performance. There was significant agreement between the distance measures based on AFLP markers vs morphological traits + AFLP markers (r = 0.47) and between pedigree data vs morphological traits + AFLP markers (r = 0.43). The pedigree distance was positively associated with traits 100-kernel weight and grain yield per plant in F(1) (correlations of 0.67 and 0.62, respectively) and F(2) (correlations of 0.62 and 0.59, respectively) generations. These correlation values indicate that the genetic distance, based on pedigree data, could replace diallel crosses for the selection of parents with higher combining ability and with moderate reliability.  相似文献

In total, 42 accessions of Origanum vulgare L., mostly originating from Europe, were evaluated, to detect molecular, quantitative morphological, and chemotype polymorphisms and to discover possible correlations between them. Twelve traits related to morphological characteristics were measured. The components in the essential oils were identified by GC/MS analysis, and the oil contents of 18 major compounds were determined. A total of 477 molecular polymorphisms including 214 AFLP (amplified fragment length polymorphism) and 263 SAMPL (selectively amplified microsatellite polymorphic loci) were used for genotyping. Euclidean distances of morphological and chemotypic data and genetic distances (1 – Dice's similarity) of molecular markers were compared by applying Mantel tests to ascertain the congruencies between them. A relatively high correlation between chemotypic patterns and genetic markers was identified, while a lower correlation was found between the morphological and genetic matrices. Pairwise analyses of correlation among all traits showed that the stem diameter was correlated to the essential‐oil yield and the carvacrol content. Cluster analysis, population inference, and principal component analysis revealed a broad genetic and chemical variation among the accessions. The knowledge of these diversities, found in this study, will allow a plant improvement of Origanum vulgare related to pharmaceutical and spice uses.  相似文献

Simple sequence repeats (SSR) is one of the most suitable markers for variety identification as it has great discrimination power for varieties with limited genetic variation. Genetic characterization of commercial tomato varieties was investigated using 33 SSR markers and 22 morphological traits. Thirty three SSR primer pairs were screened for 63 tomato varieties. A total of 132 polymorphic amplified fragments were obtained by using 33 SSR markers. The average polymorphism information content (PIC) was 0.628 ranging from 0.210 to 0.880. One hundred thirty two SSR loci were used to calculate Jaccard's distance coefficients for UPGMA cluster analysis. A clustering group of varieties, based on the results of SSR analysis, were categorized into cherry and classic fruit type varieties. Almost all of the varieties were discriminated by SSR marker genotypes. The relationship between morphological and molecular data for 33 varieties out of 63 varieties was analyzed using Mantel matrix correspondence test. The correlation value between two methods was 0.644. However, SSR based dendrogram topology showed some similar form with morphological traits at the two main groups. Therefore, these markers may be used wide range of practical application in variety identification and pre-screening for distinctiveness test of tomato varieties.  相似文献
