Distributions of molecular size and fluorescence properties of dissolved organic matter (DOM) in the Lake Biwa and Lake Baikal watersheds were investigated using the cross-flow ultrafiltration technique and three-dimensional fluorescence measurements. From the fluorescence properties, protein-like substances were usually found in the 0.1 μm-GF/F fraction (the Durapore membrane retentate of the GF/F filtrate) of the lake DOM. The results indicated autochthonous production of protein-like organic-matters in the lake environment. Fulvic acid (FA)-like components were composed of two fractions with respect to fluorescence properties and molecular size. Two FA-like fluorescence peaks, which showed different fluorescence peak positions in the excitation-emission matrix (EEM), were partly fractionated by the molecular size of 5000 daltons (5 kDa). The FA-like fluorescence peak position of the <5-kDa fraction was observed at the shorter wavelength region compared with that of the fraction between 5 kDa and 0.1 μm (5 kDa20.1 μm fraction). A blue shift of the FA-like fluorescence peak position as well as a decrease in the molecular size of the DOM was observed in lake samples. The relative contribution of the <5 kDa fraction to the DOC concentration was high in lake waters (68%–79%) compared with river waters (44%–68%), suggesting characteristic changes in molecular size between riverine and lacustrine DOM. DOM of the 5 kDa–0.1 μm fraction was relatively higher in river waters than in lake waters. These findings coincided with in situ distributions of the fluorescence properties and molecular size of DOM found in both stream and lake environments. These results indicate that FA-like substances from forested watersheds are decomposed qualitatively and quantitatively in the river-lake environment by photochemical and biological processes.  相似文献

Bacterial utilization of dissolved organic matter (DOM) was studied in water from a humic and a clearwater oligotrophic lake. Indigenous bacteria were inoculated into either 0.2 m natural filtered lake water, or lake water enriched fivefold with colloidal DOM >100 kD but below 0.2 m. Consumption of DOM was followed from changes in concentrations of total dissolved organic carbon (DOC), dissolved combined and free carbohydrates and amino acids (DCCHO and DFCHO, and DCAA and DFAA, respectively) and by uptake of monosaccharide and amino acid radioisotopes. DCCHO and DCAA made up 8% (humic lake) to 33–44% (clear-water lake) of the natural DOC pools, while DFCHO and DFAA contributed at most 1.7% to the DOC pools. Addition of >100 kD DOM increased the DOC concentrations by 50% (clearwater lake) to 92% (humic lake), but it only resulted in a higher bacterial production (by 63%) in the humic lake. During the incubations 13 to 37% of the DOC was assimilated by the bacteria, at estimated growth efficiencies of 4–8%. Despite the measured reduction of DOC, statistically significant changes of specific organic compounds, especially of DCCHO and DCAA, generally did not occur. Probably the presence of high molecular weight DOC interfered with the applied analytical procedures. Addition of radiotracers indicated, however, that DFAA sustained 17–58% and 29–100% of the bacterial carbon and nitrogen requirements, respectively, and that glucose met 1–3% of the bacterial carbon requirements. Thus, our experiments indicate that radiotracers, rather than measurements of concentration changes, should be used in studies of bacterial utilization of DOC in freshwaters with a high content of humic or high molecular weight organic matter.  相似文献

Fluxes of dissolved organic matter (DOM) are an important vector for the movement of carbon (C) and nutrients both within and between ecosystems. However, although DOM fluxes from throughfall and through litterfall can be large, little is known about the fate of DOM leached from plant canopies, or from the litter layer into the soil horizon. In this study, our objectives were to determine the importance of plant-litter leachate as a vehicle for DOM movement, and to track DOM decomposition [including dissolve organic carbon (DOC) and dissolved organic nitrogen (DON) fractions], as well as DOM chemical and isotopic dynamics, during a long-term laboratory incubation experiment using fresh leaves and litter from several ecosystem types. The water-extractable fraction of organic C was high for all five plant species, as was the biodegradable fraction; in most cases, more than 70% of the initial DOM was decomposed in the first 10 days of the experiment. The chemical composition of the DOM changed as decomposition proceeded, with humic (hydrophobic) fractions becoming relatively more abundant than nonhumic (hydrophilic) fractions over time. However, in spite of proportional changes in humic and nonhumic fractions over time, our data suggest that both fractions are readily decomposed in the absence of physicochemical reactions with soil surfaces. Our data also showed no changes in the 13C signature of DOM during decomposition, suggesting that isotopic fractionation during DOM uptake is not a significant process. These results suggest that soil microorganisms preferentially decompose more labile organic molecules in the DOM pool, which also tend to be isotopically heavier than more recalcitrant DOM fractions. We believe that the interaction between DOM decomposition dynamics and soil sorption processes contribute to the 13C enrichment of soil organic matter commonly observed with depth in soil profiles. published online 2004  相似文献

Soil organic matter (SOM) is the largest terrestrial C pool, and retention and release of dissolved organic matter (DOM) cause formation and loss of SOM. However, we lack information on how different sources of DOM affect its chemical composition, and how DOM chemical composition affects retention. We studied seasonal controls on DOM production and chemical controls on retention in soils of a temperate coniferous forest. The O horizon was not usually the dominant source for dissolved organic C (DOC) or N (DON) as has been reported for other sites. Rather, net production of both DOC and DON was often greater in the shallow mineral soil (0–10 cm) than in the O horizon. DOM production in the shallow mineral soil may be from root exudation as well as turnover of fine roots and microflora in the rhizosphere. In the field, the two acid fractions (hydrophobic and hydrophilic acids) dominated the soil solution at all depths. A major portion of net production and removal of total DOC within the soil column was explained by increases and decreases in these fractions, although a shift in chemical composition of DOM between the O and mineral soil horizons suggested different origins of DOM in these layers. A larger loss of the free amino fraction to deep soil water at this study site than at other sites suggested lower retention of labile DON. Field DOM removal measurements suggest that field-measured parameters may provide a good estimate for total DOM retained in mineral soil.  相似文献

Effects of different molecular size fractions (< 1000 MW, < 10 000 MW, < 100 000 MW and <0.1 μm) of dissolved organic matter (DOM) on the growth of bacteria, algae and protozoa from a highly humic lake were investigated. DOM from catchment drainage water as well as from the lake consisted mostly (59–63%) of high molecular weight (HMW) compounds (> 10 000 MW). With excess inorganic nutrients, the growth rate and yield of bacteria were almost identical in all size fractions. However, in < 1000 MW fractions and with glucose added, a longer lag phase occurred. Without added nutrients both the growth rates and biomasses of bacteria decreased towards the smaller size fractions and the percentage of dissolved organic carbon (DOC) used during the experiment and the growth efficiency of bacteria were lower than with excess nutrients. The growth efficiency of bacteria was estimated to vary between 3–66% in different MW fractions, largely depending on the nutrient concentrations, but the highest growth efficiencies were observed in HMW fractions and with glucose. The growth of algae was clearly lowest in the < 1000 MW fraction. In dim light no net growth of algae could be found. In contrast, added nutrients substantially enhanced algal growth and in deionized water with glucose, algae achieved almost the same growth rate and biomass as in higher MW fractions of DOM. The results suggested that bacteria and some algae were favoured by DOM, but protozoans seemed to benefit only indirectly, through bacterial grazing. The utilization of DOM by bacteria and algae was strongly affected by the availability of phosphorus and nitrogen.  相似文献

A study of the isotopic composition of organic matter was conducted in a freshwater marsh over seasonal and diel time scales to determine the sources of dissolved organic matter (DOM) and the processes leading to its formation. Bulk C and N isotopic compositions of the bacterial fraction (0.2–0.7 m) and particulate organic matter (POM; 0.7–10 m) were compared on a seasonal basis with the change in 13C of DOM. The bulk isotopic data support the idea that DOM was, in part, derived from the breakdown of larger organic matter fractions. The bacterial fraction and POM were compositionally similar throughout the year, based on a comparison of the 13C of individual amino acids in each fraction. Annual variation in the 13C of amino acids in DOM was greater relative to the variation in larger fractions indicating that microbial reworking was an important factor determining the proteinaceous component of DOM. The 13C enrichment of serine and leucine in each organic matter fraction suggested microbial reworking was an important factor determining organic matter composition during the most productive times of year. Changes in the bulk 13C of DOM were more significant over daily, relative to seasonal, time scales where values ranged by 6 and followed changes in chlorophyll a concentrations. Although bulk 13C values for POM ranged only from –29 to –28 during the same diel period, the 13C of alanine in POM ranged from –30 to –22. Alanine is directly synthesized from pyruvate and is therefore a good metabolic indicator. The 13C of individual amino acids in DOM revealed the diel change in the importance of autotrophic versus heterotrophic activity in influencing DOM composition. Diel changes in the 13C of phenylalanine, synthesized by common pathways in phytoplankton and bacteria, were similar in both DOM and POM. The diel change in 13C of isoleucine and valine, synthesized through different pathways in phytoplankton and bacteria, were distinctly different in DOM versus POM. This disparity indicated a decoupling of the POM and DOM pools, which suggests a greater source of bacterial-derived organic matter at night. The results of this study demonstrate the use of the isotopic composition of individual amino acids in determining the importance of microbial reworking and autotrophic versus heterotrophic contributions to DOM over both diel and seasonal time scales.  相似文献

SUMMARY.
  • 1 The microbial heterotrophic utilization of dissolved organic matter (DOM) was determined experimentally in microcosms using stream water and stream-bed sediments from a third order reach of White Clay Creek (Pennsylvania, U.S.A.).
  • 2 Sources of DOM for the experiments included White Clay Creek water at baseflow and stormflow and cold water extracts of jewel weed (Impatiens capensis L.) and spicebush (Linderu henzoin (L.) Blume).
  • 3 The heterotrophic activity of the sediments was measured as uptake of the following: dissolved organic carbon (DOC), molecular weight fractions within the DOC pool, carbohydrates, amino acids and peptides, phenolics, and dissolved oxygen (DO), all in the overlying water.
  • 4 Concentrations of adenosine triphosphate (ATP), and direct microscopic counts of bacteria were used to estimate bacterial biomass in the surface sediments.
  • 5 The microcosm experiments showed that specific DOC molecular size classes and DOM functional groups were selectively removed from solution, exposure to one DOM source affected responses to a different DOM source and certain DOM sources were more readily utilized than others.
  • 6 Continued exposure to a DOM source increased microbial heterotrophic activity (a condition which persisted even after removal of the DOM source for several days).
  • 7 Rates of biotic DOC uptake ranged from 3.6 to 242.8 mg Cm-2h-1.
  • 8 Indirect estimates of biosynthesis calculated from DOC and DO data ranged from 1.6 at baseflow and 2.6–61.2 at stormflow to as high as 192.6 mg C m-2 h-1 when the community was repeatedly exposed to enriched DOM sources.
  • 9 The mean generation times of bacteria in sediments, determined from direct microscopy data, ranged from 12.5 to 46.2 h at 15°C.

Proteinaceous components from four Washington coast margin sediments were extracted with base, fractionated into one of four size classes (<3 kDa, 3–10 kDa, 10–100 kDa, >100 kDa), and analyzed for their amino acid contents. Base-extracted material accounts for ~30% of the total hydrolyzable amino acids (THAA) and each size fraction has a unique composition, regardless of where the sediment was collected (shelf or upper slope). The <3 kDa size fraction (~10% of base-extractable THAA) is relatively enriched in glycine (~30 mol%), lysine (~5 mol%), and non-protein amino acids (~5 mol%). Glycine and non-protein amino acids are common degradation products, and lysine is very surface active. We suggest that the <3 kDa size fraction, therefore, represents a diagenetic mixture of fragments produced during the degradation of larger proteins. The 3–10 and 10–100 kDa size fractions (~10% and 42% of base-extractable THAA, respectively) have similar amino acid distributions dominated by aspartic acid (~30 mol%). Enrichments in Asp is likely due to both preservation of Asp-rich proteins and the production of Asp during degradation. The >100 kDa size fraction (~38% of base-extractable THAA) is not dominated by any particular amino acid and can not be modeled by mixing the amino acid compositions of the other size fractions. We propose that the larger size fractions (10–100 kDa and >100 kDa) represent intact, or near intact, proteins. Estimates of isoelectric points and relative hydrophobicity suggest the base-extractable proteins are primarily acidic and have globular structures. Statistical comparisons to several known proteins indicates that the base-extractable component is most similar to planktonic cytoplasmic proteins.  相似文献

The Yenisei river passes every type of permafrost regime, from south to north, being characterized by increasing continuity of the permafrost and by decreasing thickness of the active layer. We used that situation to test the hypothesis that amounts and properties of dissolved organic matter (DOM) in small streams draining forested catchments respond to different permafrost regimes. Water samples were taken from eight tributaries along the Yenisei between 67°30′N and 65°49′N latitude. The samples were analysed for dissolved organic carbon (DOC) and nitrogen (DON) and DOM was characterized by its chemical composition (XAD‐8 fractionation, sugars, lignin phenols, amino acids, protein, UV and fluorescence spectroscopy), and its biodegradability. Most properties of the tributary waters varied depending on latitude. The higher the latitude, the higher were DOC, DON and the proportion of the hydrophobic fraction of DOC. The contribution of hexoses and pentoses to DOC were higher in southern tributaries; on the other hand, phenolic compounds were more abundant in northern tributaries. Mineralizable DOC ranged between 4% and 28% of total DOC. DOM in northern tributaries was significantly (P<0.05) less biodegradable than that in southern tributaries reflecting the differences in the chemical properties of DOM. Our results suggest that the differences in DOM properties are mainly attributed to differences of permafrost regime, affecting depth of active layer, soil organic matter accumulation and vegetation. Soil organic matter and vegetation determine the amount and composition of DOM produced in the catchments while the depth of the active layer likely controls the quantity and quality of DOM exported to streams. Sorptive interactions of DOM with the soil mineral phase typically increase with depth. The results imply that a northern shift of discontinuous permafrost likely will change in the long term the input of DOM into the Yenisei and thus probably into the Kara Sea.  相似文献

Monthly (or bi-weekly) water samples were collected from the Yukon River, one of the largest rivers in North America, at a station near the US Geological Survey Stevens Village hydrological station, Alaska from May to September 2002, to examine the quantity and quality of dissolved organic matter (DOM) and its seasonal variations. DOM was further size fractionated into high molecular weight (HMW or colloidal, 1 kDa–0.45 μm) and low molecular weight (LMW, <1 kDa) fractions. Dissolved organic carbon (DOC), colored dissolved organic matter (C-DOM) and total dissolved carbohydrate (TCHO) species were measured in the size fractionated DOM samples. Concentrations of DOC were as high as 2830 μmol-C l−1 during the spring breakup in May and decreased significantly to 508–558 μmol-C l−1 during open-water season (June–September). Within the DOC pool, up to 85% was in the colloidal fraction (1 kDa–0.45 μm) in early May. As DOC concentration decreased, this colloidal portion remained high (70–85% of the bulk DOC) throughout the sampling season. Concentrations of TCHO, including monosaccharides (MCHO) and polysaccharides (PCHO), varied from 722 μmol-C l−1 in May to 129 μmol-C l−1 in September, which comprised a fairly constant portion of bulk DOC (24±2%). Within the TCHO pool, the MCHO/TCHO ratio consistently increased from May to September. The C-DOM/DOM ratio and the size fractionated DOM increased from May to September, indicating that DOM draining into the Yukon River contained increased amounts of humified materials, likely related to a greater soil leaching efficiency in summer. The average composition of DOM was 76% pedogenic humic matter and 24% aquagenic CHO. Characteristics of soil-derived humic substances and low chlorophyll-a concentrations support a dominance of terrestrial DOM in Yukon River waters.  相似文献

The free-living, cosmopolitan, freshwater betaproteobacterial bacterioplankton genus Polynucleobacter was detected in different years in 11 lakes of varying types and a river using the size-exclusion assay method (SEAM). Of the 350 strains isolated, 228 (65.1%) were affiliated with the Polynucleobacter subclusters PnecC (30.0%) and PnecD (35.1%). Significant positive correlations between fluorescence in situ hybridization and SEAM data were observed in the relative abundance of PnecC and PnecD bacteria to Polynucleobacter communities (PnecC?+?PnecD). Isolates were mainly PnecC bacteria in the samples with a high specific UV absorbance at 254?nm (SUVA(254) ), and a low total hydrolysable neutral carbohydrate and amino acid (THneutralCH?+?THAA) content of the dissolved organic matter (DOM) fraction, which is known to be correlated with a high humic content. In contrast, the PnecD bacteria were abundant in samples with high chlorophyll a and/or THneutralCH?+?THAA concentrations, indicative of primary productivity. With few exceptions, differences in the relative abundance of PnecC and PnecD in each sample, determined using a high-sensitivity cultivation-based approach, were due to DOM quality. These results suggest that the major DOM component in the field, which is allochthonously or autochthonously derived, is a key factor for ecological niche separation between PnecC and PnecD subclusters.  相似文献

The degradability and chemical characteristics of water-extractable dissolved organic carbon (DOC) and nitrogen (DON) from the humus layer of silver birch (Betula pendula Roth), Norway spruce (Picea abies (L.) Karst.) and Scots pine (Pinus sylvestris L.) stands were compared in short-term incubation of soil solutions. For all extracts the degradation of DOC and DON was low (12-17% loss) and increased in the order: birch, spruce and pine. In the humus layer under pine a relatively larger pool of rapidly degrading dissolved soil organic matter (DOM) was indicated by the [3H]thymidine incorporation technique, which measures the availability of DOM to bacteria. The degradation of DOC was explained by a decrease in the hydrophilic fraction. For DON, however, both the hydrophilic and hydrophobic fractions tended to decrease during incubation. No major differences in concentrations of hydrophilic and hydrophobic fractions were detected between tree species. Molecular size distribution of DOC and DON, however, revealed slight initial differences between birch and conifers as well as a change in birch extract during incubation. The depletion of very rapidly degrading fractions (e.g., root exudates and compounds from the litter) may explain the low degradability of DOM in the humus layer under birch.  相似文献

Aims

We characterized dissolved organic matter (DOM) leached during decomposition of deciduous silver birch litter (Betula pendula Roth.), coniferous Norway spruce litter (Picea abies (L.) Karst.) and a mixture of these litters in order to find out whether the properties of DOM would explain the earlier observed signs for higher microbial activity in soil under birch than spruce.

Methods

DOM leached from decomposing litters was collected in a litter-column experiment in the laboratory. Adsorption properties (XAD-8 resin fractionation) and molecular weight as well as the degradability of dissolved organic carbon (DOC) and nitrogen (DON) were measured three times during decomposition: 1) in the early stages, 2) after the mass loss reached 20–30 % and 3) when the mass loss reached 30–40 %.

Results

The leaching of DOC hydrophilic neutrals and bases, regarded easily degradable, decreased during decomposition. The leaching of DOC in hydrophobic acids, regarded refractory, increased from spruce and especially from the mixture litter during decomposition and may be connected to the degree of litter decomposition that was highest for the mixture. Unexpectedly, the degradability of DOC differed only slightly between the litters but the degradability of DON was substantially higher for spruce than birch. Spruce DOM seemed to be more N-rich than birch DOM in the early stages of decomposition and it seemed that labile DON was mobilized earlier from spruce than birch litter.

Conclusions

We conclude that the decomposition degree of litter determines largely the properties of DOM. The observed differences in the properties of DOM sampled during the litter decomposition cannot explain differences in C and N cycling between birch and spruce.  相似文献

The photo- and bio-degradation of dissolved organic matter (DOM) in water from the Broad River were investigated in laboratory experiments using a solar simulator to control the intensity and exposure of samples to irradiation. The water samples included a natural assemblage of microorganisms, and the daily exposure of samples to irradiation was varied to distinguish the relative contributions of photochemical and biological degradation. Concentrations of dissolved organic carbon (DOC) and specific components of DOM, including chromophoric DOM (CDOM), lignin phenols and amino acids, were monitored to investigate the reactivity and predominant pathway of degradation of these DOM components. Biodegradation was primarily responsible for the overall remineralization of DOC and losses of the amino acid component of DOM, whereas photodegradation was primarily responsible for losses of the chromophoric and lignin phenol components of DOM. The rates of photodegradation of lignin phenols were strongly influenced by the presence of methoxy groups on the aryl ring. Syringyl (S) phenols have two methoxy substitutions, vanillyl (V) phenols have one methoxy substitution, and p-hydroxy (P) phenols are not substituted with methoxy groups. Photochemical decay constants were highest for S phenols, lowest for P phenols and followed a consistent pattern (S > V > P) in the experiments. The carbon-normalized yields of amino acids and lignin phenols were found to be useful molecular indicators of the highly reactive (i.e. labile) components of biodegradable and photodegradable DOM, respectively.  相似文献

Dissolved organic matter (DOM), produced through leaching from particulate organic matter (POM), is an essential component of the carbon cycle in streams. The present study investigated the instream DOM release from POM, varying in size and chemical quality. We produced large and medium sized fine particulate organic matter (L-FPOM, 250–500 μm; M-FPOM, 100–250 μm) of defined quality by feeding five types of coarse particulate organic matter (CPOM) to shredding amphipods (Gammarus spp.). Microscopic observations showed that L-FPOM and M-FPOM mainly consisted of the fecal pellets of amphipods, and incompletely eaten plant fragments, respectively. DOM release experiments were conducted by exposing CPOM and M- and L-FPOM fractions in natural stream water over a two week period. For CPOM, the release of dissolved organic carbon (DOC) by leaching was highest during the first 6 h (3.64–23.9 mg C g C?1 h?1) and decreased rapidly afterwards. For M- and L-FPOM, the DOC release remained low during the entire study period (range: 0.008–0.15 mg C g C?1 h?1). Two-way ANOVA revealed that the DOC release rate significantly differed with POM source and size fraction, both at day 1 and after a week of exposure. Multiple regression analyses revealed a significant correlation of elemental contents and lignin content to DOC release rate after a week of exposure. Overall, the results indicated that DOC release rate of FPOM, on a carbon basis, is comparable to that of CPOM after leaching, while size and source of POM significantly affect DOC release rate.  相似文献

Dissolved organic matter (DOM) in sediment porewaters from Lake Erhai, Southwest China was investigated using dissolved organic carbon (DOC) concentration, UV absorbance, fluorescence and molecular weight distribution. DOC exhibited a high concentration at the sediment–water interface with a rapid decrease to the oxic–anoxic interface at approximately 7 cm, and then increased with depth. Similar trends were also found for the UV absorption coefficients at 254 and 280 nm in the porewaters. DNA in the sediment was also measured, which confirmed the high abundance of aerobic bacteria in the upper layer of the sediment. Both humic-like (peaks A and C) and protein-like (peaks B and D) fluorescence were observed in the porewater DOM, and their fluorescence intensities exhibited a similar porewater profile as DOC concentration. A strong correlation was found between the peak fluorescence intensity ratio r(A, C) and r(D, B). Both the fluorescence index and UV absorption coefficient at 254 nm suggested a dramatic increase in aromaticity of porewater DOM across the oxic–anoxic interface. Porewater DOM exhibited a multimodal distribution of molecular weight with a relatively low polydispersity. The results of this study offer significant insight into the nature and properties of DOM in freshwater ecosystems.

Dissolved organic matter (DOM) is an important component of aquatic food webs. We compare the uptake kinetics for NH4–N and different fractions of DOM during soil and salmon leachate additions by evaluating the uptake of organic forms of carbon (DOC) and nitrogen (DON), and proteinaceous DOM, as measured by parallel factor (PARAFAC) modeling of DOM fluorescence. Seasonal DOM slug additions were conducted in three headwater streams draining a bog, forested wetland, and upland forest using DOM collected by leaching watershed soils. We also used DOM collected from bog soil and salmon carcasses to perform additions in the upland forest stream. DOC uptake velocity ranged from 0.010 to 0.063 mm s−1 and DON uptake velocity ranged from 0.015 to 0.086 mm s−1, which provides evidence for the whole-stream uptake of allochthonous DOM. These findings imply that wetlands could potentially be an important source of DOM to support stream heterotrophic production. There was no significant difference in the uptake of DOC and DON across the soil leachate additions (P > 0.05), although differential uptake of DOM fractions was observed as protein-like fluorescence was removed from the water column more efficiently than bulk DOC and DON (P < 0.05). Moreover, PARAFAC analysis of DOM fluorescence showed that protein-like fluorescence decreased downstream during all DOM additions, whereas humic-like fluorescence did not change. This differential processing in added DOM suggests slow and fast turnover pools exist for aquatic DOM. Taken together, our findings argue that DON could potentially fill a larger role in satisfying biotic N demand in oligotrophic headwater streams than previously thought. Quantitative and qualitative characterizations of dissolved organic matter (DOM) were carried out at the watershed level in central Japan by measuring dissolved organic carbon (DOC) concentration and the three-dimensional excitation–emission matrix (3-D EEM). DOC concentration was low (mean 37 ± 19 µM C) in the upstream waters, whereas, in general, it increased toward the downstream areas (mean 92 ± 47 µM C). Significant variations in DOC concentration were detected among rivers and channels. DOC concentration in the epilimnion of Lake Biwa increased during the summer period and decreased during the winter period. The lake hypolimnion has lower DOC concentration (mean 87 ± 7 µM C) compared with the epilimnion (107 ± 15 µM C). Fulvic acid (FA)-like substances in the DOM were directly characterized by 3-D EEM. The fluorescence peak for upstream DOM was found in regions with longer wavelengths (excitation/emission 386 ± 6/476 ± 5 nm) compared with downstream and lake DOM (351 ± 12/446 ± 15 nm and 341 ± 6/434 ± 6 nm, respectively). The DOC concentration is correlated with fluorescence peak intensity of FA-like substances in DOM in river waters. Such a relationship was not found in lake DOM. A blueshift of the fluorescence peak from upstream to lake DOM was observed. A decrease in fluorescence intensities was also detected during the summer period. These results may suggest that the degradation of FA-like substances in DOM occurs from natural solar irradiation. Protein-like fluorescence was significantly detected in the lake epilimnion during the summer period. A linear relationship between DOC concentration and protein-like fluorescence indicated that an autochthonous input of DOM gave rise to the increase in DOC concentration in the lake epilimnion during the summer. These results may suggest that the 3-D EEM can be used as a tool for the investigation of DOM dynamics at the watershed level with concurrent measurement of DOC concentration and the fluorescence properties of fulvic acid-like and protein-like substances.  相似文献
