We found high diversity of vegetative compatibility (vc) types in native populations of the chestnut blight fungus, Cryphonectria parasitica, in Japan and China; almost every isolate was in a unique vc type. In Japan we found 71 vc types in a sample of 79 isolates pooled from six populations. Within two populations in China, all isolates (n=28 and 11) had unique vc types; we found 15 vc types among 25 isolates in a third Chinese population where multiple isolates were collected from some trees. None of the isolates from China and only three isolates in the 71 vc types from Japan were compatible with any of 64 vc type testers from Europe, which have known vegetative incompatibility genotypes. To our knowledge this is the first report of vc type diversity for C. parasitica in Japan or of any comparisons of vc types between Asia and Europe. The most significant result of this survey is the identification fungal isolates for expanding knowledge of the genetics of vegetative incompatibility.  相似文献

Chestnut blight is controlled in Europe by using Cryphonectria hypovirus CHV1, a non-encapsulated RNA virus. The chestnut blight fungus, Cryphonectria parasitica, is weakened by the virus, and healing tissue growth occurs in the host tree. Transmission of this cytoplasmic hypovirus is restricted by the incompatibility system of the fungus, so that the hypovirus can be transmitted only between isolates of the same or closely related vegetative compatibility (vc) types. Hypovirulent isolates of C. parasitica (all of the French subtype CHV1-F1) from Castilla y León (NW Spain) were compared with virulent isolates in both laboratory (cut stems) and field inoculations (in two orchards in the province of León and one orchard in the province of Zamora). The tests were performed with the most common vc types in the region, EU1 and EU11. The cut stem assay revealed that the hypovirulent isolates of vc type EU1 did not reduce the growth of virulent cankers. By contrast, four hypovirulent strains H1, H4, H5 and H6 (all vc type EU11) reduced the growth of virulent isolates in the cut stem assay. Field tests showed that hypovirulent isolates of EU1 and EU11 were effective in reducing canker in both orchards in León with all treatments tested; however, in Zamora, where only EU11 was tested, all the treatments failed except H1, which was able to reduce growth of the canker eighteen months after the inoculation. The development of hypovirulence suggests that hypovirus subtype F1 is well adapted in the province of León. Both naturally extended and inoculated hypoviruses appear to have reduced the incidence of the canker, thus improving chestnut stands. However, the inoculations were not as effective in the orchards in Zamora. This indicates that the disease could be controlled in Castilla y León by inoculation of trees with hypovirulent strains, but that more tests should be done in provinces where the hypovirus is still not present.  相似文献

Mycological analysis of samples of sweet chestnut (Castanea sativa) collected in the northern Turkish zone of its habitat was carried out. A total of 300 strains of micromycetes belonging to 13 taxa were isolated in pure cultures. The phytopathogenic fungus Cryphonectria parasitica, the causal agent of chestnut blight, was identified among these strains. A technique for detection of C. parasitica in pure culture and host tissues using species-specific PCR markers was developed and tested. This technique can be used as an express method for detection of chestnut blight.  相似文献

In Lovran (coastal Croatia), a unique forest/orchard of evenly mixed grafted marrons and naturally growing nongrafted sweet chestnut trees exists. This old chestnut population has been devastated by chestnut blight, caused by an aggressive introduced pathogenic fungus, Cryphonectria parasitica. However, initial observations indicated recovery of naturally growing chestnut trees in that area, mediated by Cryphonectria-associated hypovirus (Cryphonectria hypovirus 1 (CHV-1)). Such recovery was not observed on grafted trees. Genotyping both, we confirmed the clonal origin of the grafted ones—marrons. No significant difference was observed between fungal strains isolated from naturally growing trees and the ones from marrons regarding fungal vegetative compatibility types or the prevalence of CHV-1. A strong correlation was observed between the types of canker: active/deep-expanding versus healing callus or superficial necrosis and the absence or presence of CHV-1 in the fungal isolates, sampled from naturally growing trees (Spearman rho 0.686, p value 7.81?×?10?5, Kendall tau 0.686, p value 5.18?×?10?7). Such correlation was not observed on marrons (Spearman rho 0.236, p value 0.235, Kendall tau 0.236, p value 0.084), because, unexpectedly, active/deep-expanding cankers were often associated with hypovirulent fungal isolates. These data indicate that the lack or unequal distribution of naturally occurring hypovirulence were not the cause of substantial marron decay in Lovran. Ecological and age-dependant differences were ruled out because all sampled trees are growing in close proximity and are of similar age. The results imply that the marron genotype is especially vulnerable and its ability to recover is limited even when the hypovirulent strain of the fungus is present in the canker.  相似文献

The number of layers of epidermis in the leaves is used as a criterion to distinguish between Castanopsis sieboldii (two layers) and C. cuspidata (one layer). An intermediate type, which has one and two layers within a single leaf, is frequently seen in the field. The origin of the intermediate type has been supposed to be a hybrid between C. sieboldii and C. cuspidata. If the intermediate type is produced by hybridization, we expect that the F1 seedlings of the intermediate type should occur in the co-occurrence area of those two species. To clarify the geographic occurrence of the intermediate type, we collected nuts of 443 mother trees from throughout the distribution area of the genus Castanopsis in Japan. A total of 7,260 seedlings germinated from these nuts were examined as to their leaf structure. The seedlings of the intermediate type occurred not only in the area where C. sieboldii and C. cuspidata coexisted, but also in the area where only C. sieboldii grows. The leaf structure of intermediate seedlings was independent of the traits of mother trees. These findings suggest that the intermediate seedlings that occurred in the area where only C. sieboldii grows are not hybrid between C. sieboldii and C. cuspidata but are a morphological variation of C. sieboldii. The difference in the structure of leaf epidermis is not always appropriate for the identification of the hybrid.  相似文献

We surveyed native populations of the chestnut blight fungus, Cryphonectria parasitica, in Japan and China, and C. nitschkei, a sympatric species on chestnut trees in Japan, to learn more about the diversity of hypoviruses and other double-stranded (ds) RNA viruses. In a sample of 472 isolates of C. parasitica and 45 isolates of C. nitschkei from six prefectures in Japan, we found 27 containing one or more dsRNAs. Twelve isolates of C. parasitica and two isolates of C. nitschkei were infected with Cryphonectria hypovirus 1 (CHV-1); four of these 12 C. parasitica isolates also contained other dsRNAs that did not hybridize to CHV-1. In China, only one of 85 C. parasitica isolates was CHV-1-infected; no dsRNAs were detected in the other isolates from China. No other known hypoviruses were found in this study. However, we found two previously undescribed dsRNAs in Japan approximately 9 kb in size that did not hybridize to each other or to any known dsRNAs from C. parasitica. We also found three additional groups of dsRNAs, one of which represents the genome of a new member of the virus family Chrysoviridae and was found only in C. nitschkei; the other two dsRNAs were found previously in isolates of C. parasitica from Japan or China. The most significant result of this survey is the discovery of novel dsRNAs that can be characterized in future research.  相似文献

Chestnut blight in south-western Germany was first reported in 1992 and is since expanding in distribution. Here we investigated the invasion history of Cryphonectria parasitica and its associated hypovirus. For this, we characterized 284 isolates collected between 1992 and 2012 for hypovirulence, vegetative compatibility (vc), mating type, and microsatellite haplotype. A total of 27 haplotypes and 15 vc types were observed, although the C. parasitica population analyzed is currently dominated to 50 % by one haplotype and to 64 % by the vc type EU-2. Structure analysis indicated two divergent genetic pools. Over 66 % of the haplotypes belonged to a pool probably originating from northern Italy. Further diversification is expected due to ongoing sexual recombination, but also to new migration and additional introductions. Cryphonectria hypovirus 1 (CHV-1) was found in four of five C. parasitica populations from Baden-Württemberg. Genetic analysis of the 35 CHV-1 isolates obtained revealed that they all belong to the German subtype, although they have clearly diverged from the first German hypovirus isolated in 1992. Our study suggests that C. parasitica has been introduced into Germany several times from two different gene pools, whereas the hypovirus most probably has a single origin.  相似文献

American [Castanea dentata (Marsh) Borkh.] × Chinese [Castanea mollissima Blume] chestnut (Fagac, ae) hybrids are a novel system in which to study influences of phytopathogenic fungi and woody plant hybridization on herbivore susceptibility, as the hybrids are well characterized with regard to resistance to the chestnut blight fungus [Cryphonectria parasita (Murr) Barr (Endothia) Diaporthales: Valsaceae] and variability is present. We chose two groups of resistance‐rated backcross chestnut that shared an F1 parent and had different American parents. Foliage from both backcross groups and the parent trees was sampled on three dates for use in feeding assays with gypsy moth larvae [Lymantria dispar (L.) [Lepidoptera: Lymantriidae], adult Japanese beetles [Popillia japonica Newman (Coleoptera: Scarabaeidae)], and fall webworm larvae [Hyphantria cunea Drury (Lepidoptera: Arctiidae)], respectively. Foliar analyses were performed concurrently and included carbohydrate, tannin, and nitrogen content, toughness, and density. Blight resistance had almost no effect on herbivore performance or foliar chemistry. When the parent trees and backcross groups were compared, however, significant differences in gypsy moth performance and Japanese beetle consumption were evident. There were no differences in fall webworm consumption. Most foliar characteristics measured differed among chestnut genotypes at some point in the season, and all varied seasonally. No clear pattern emerges with respect to the relationship among blight resistance, herbivore susceptibility, foliar properties, and plant genotype, and more research is needed to separate these effects.  相似文献

A severe foliar yellow mosaic disease was observed in horse chestnut trees (Aesculus carnea and A. hippocastanum). Reactions in woody indicator plants grafted with diseased horse chestnut suggested the presence of an ilarvirus. Virus isolates obtained by mechanical inoculation of herbaceous test plants reacted with antisera to apple mosaic virus but not with antisera to its serotype prunus necrotic ringspot virus, or to prune dwarf virus. Yellow mosaic was induced in horse chestnut seedlings grafted with tissues from herbaceous hosts infected with horse chestnut isolates or with the European plum line pattern isolate of apple mosaic virus. Virus was detected by enzyme-linked immunosorbent assay (ELISA) in embryo and endosperm of immature seed from infected trees but not in mature seed, or progeny seedlings. Strawberry latent ringspot virus was detected in one of six A. hippocastanum trees with a leaf vein yellows disease.  相似文献

Specificity of interactions between eight Lactuca species and 8 Bremia lactucae isolates was studied in seedlings and adult plants of 36 Lactuca accessions plus one L. serriola × L. sativa hybrid. Pathogenicity of the isolates and/or plant susceptibility was expressed by sporulation intensity. A highly compatible relationship was observed in all of L. serriola accession/isolate interactions tested. A differential reaction was found in numerous cases testifying to physiological specialization of the pathogen in a wild pathosystem. Nonspecific nondifferential compatibility (quantitative resistance) can be expected in L. serriola PI 281876. Age dependent resistance (seedlings versus adult plants, and vice versa) and heterogeneity of reactions were also recorded. The comparisonof B. lactucae isolates from L. sativa and L. serriola has shown a significant shift of pathogenicity in favour of L. serriola accessions. A high level of resistance was found in the L. serriola × L. sativa hybrid. The existence of basic incompatibility can be expected in L. saligna and L. virosa as well as in such taxonomically remote species as L. viminea, L. squarrosa and L. biennis. Other taxonomically remote species, i.e. L. dentata and L. alpina, exhibited a compatible reaction, although the reaction of the former was differential. A high level of sporulation was recorded in all accession/isolate interactions of L. alpina.  相似文献

In Northeast of Portugal, the macrofungal community associated to chestnut tree (Castanea sativa Mill.) is rich and diversified. Among fungal species, the ectomycorrhizal Pisolithus tinctorius and the saprotroph Hypholoma fasciculare are common in this habitat. The aim of the present work was to assess the effect of the interaction between both fungi on growth, nutritional status, and physiology of C. sativa seedlings. In pot experiments, C. sativa seedlings were inoculated with P. tinctorius and H. fasciculare individually or in combination. Inoculation with P. tinctorius stimulated the plant growth and resulted in increased foliar-N, foliar-P, and photosynthetic pigment contents. These effects were suppressed when H. fasciculare was simultaneously applied with P. tinctorius. This result could be related to the inhibition of ectomycorrhizal fungus root colonization as a result of antagonism or to the competition for nutrient sources. If chestnut seedlings have been previously inoculated with P. tinctorius, the subsequent inoculation of H. fasciculare 30 days later did not affect root colonization, and mycorrhization benefits were observed. This work confirms an antagonistic interaction between ectomycorrhizal and saprotrophic fungi with consequences on the ectomycorrhizal host physiology. Although P. tinctorius is effective in promoting growth of host trees by establishing mycorrhizae, in the presence of other fungi, it may not always be able to interact with host roots due to an inability to compete with certain fungi.  相似文献

Expanding populations are often less genetically diverse at their margins than at the centre of a species' range. Established, older populations of the chestnut blight fungus, Cryphonectria parasitica, are more variable for vegetative compatibility (vc) types than in expanding populations in southeastern Europe where C. parasitica has colonized relatively recently. To test whether vc types represent clones, we genotyped 373 isolates of C. parasitica from southern Italy, Romania, Bulgaria, Macedonia, Greece and Turkey using 11 sequence-characterized amplified region (SCAR) markers. Ten SCAR loci and six vegetative incompatibility (vic) loci were polymorphic in these samples. These populations are clonal by all criteria tested: (i) among 373 isolates, we found only eight multilocus haplotypes, and the same haplotypes were found in multiple countries, sometimes separated in time by as much as 12 years; (ii) the number of haplotypes observed was significantly less than expected under random mating; (iii) populations are in linkage disequilibrium; (iv) the two sets of independent markers, SCARs and vc types, are highly correlated; and (v) sexual structures of C. parasitica were found only in Bulgaria and Romania. One mating type (MAT-1) was found in 98% of the isolates sampled. In contrast, a population in northern Italy, in the central part of the range in Europe, had 12 multilocus haplotypes among 19 isolates. The spread of a few clones could be the result either of founder effect and restricted migration, or these clones have greater fitness than others and spread because they are better adapted to conditions in southeastern Europe.  相似文献

The availability of a system for direct transfer of anti-fungal candidate genes into American chestnut (Castanea dentata), devastated by a fungal blight in the last century, would offer an alternative or supplemental approach to conventional breeding for production of chestnut trees resistant to the blight fungus and other pathogens. By taking advantage of the strong ability of embryogenic American chestnut cultures to proliferate in suspension, a high-throughput Agrobacterium tumefaciens-mediated transformation protocol for stable integration of foreign genes into the tree was established. Proembryogenic masses (PEMs) were co-cultivated with A. tumefaciens strain AGL1 harboring the plasmid pCAMBIA 2301, followed by stringent selection with 50 or 100 mg/l Geneticin. A protocol employing size-fractionation to enrich for small PEMs to use as target material and selection in suspension culture was applied to rapidly produce transgenic events with an average efficiency of four independent transformation events per 50 mg of target tissue and minimal escapes. Mature somatic embryos, representing 18 transgenic events and derived from multiple American chestnut target genotypes, were germinated and over 100 transgenic somatic seedlings were produced and acclimatized to greenhouse conditions. Multiple vigorous transgenic somatic seedlings produced functional staminate flowers within 3 years following regeneration.  相似文献

Heterokaryosis was recently reported in the chestnut blight fungus, Cryphonectria parasitica, in which individuals contain nuclei that are isogenic except at the mating-type locus (MAT). MAT heterokaryons were found in several natural populations, including a putatively clonal population in West Salem, Wisconsin, providing an opportunity to address the question of how heterokaryons arise. We represented relationships among RFLP fingerprint haplotypes as networks in which loop formation is considered evidence of recombination. From 1990 to 1995, this population was clonal, as indicated by a simple haplotype network without loops, and the correlation of vegetative compatibility (vc) types and mating types with haplotype lineages. By 1999, we observed loops in the haplotype network involving isolates of two vc types (WS-2 and WS-3). Isolates with haplotypes in the loops were either MAT heterokaryons, carried the opposite mating type from other isolates of the same vc type, and/or had two alleles at two or more codominant SCAR (sequence-characterized amplified region) loci. Segregation of markers and recombination were evident among single-spore isolates from one heterokaryon; these single-spore isolates had novel fingerprint haplotypes, also within the loops. In contrast, vc type WS-1, which comprises 85% of the population, was represented by a simple network with no loops, indicating a clonal lineage varying only by mutation. Almost all isolates of WS-1 had the same mating type; the exceptions were five isolates that were MAT heterokaryons. These results are consistent with the hypothesis that heterokaryons formed between vegetatively incompatible individuals, and recombination occurred by a parasexual process.  相似文献

The successful development of early stages of blight‐resistant hybrid stock has increased hopes for restoration of American chestnut (Castanea dentata) to eastern North American forests. However, these forests have undergone substantial ecological change in the century since the functional extirpation of American chestnut, and it remains unknown to what extent American chestnut will be able to recolonize contemporary forests. In particular, high densities of white‐tailed deer (Odocoileus virginianus) and competition with mesophytic tree species such as maple (Acer) may impede chestnut regeneration, much as they affect oak (Quercus). We used a split‐plot analysis of variance (ANOVA) design to examine the effects of canopy gaps and herbivory on survival and growth of third generation backcrossed (BC3) hybrid chestnut seedlings over two growing seasons in central Indiana, U.S.A. Only 4 of 588 (0.7%) seedlings in closed‐canopy plots survived to the end of the study, as opposed to 264 of 589 (45%) seedlings in gap plots. Within the gap treatment, fencing was associated with reduced chestnut survival as well as reduced herbivory and increased cover of non‐chestnut vegetation. Our results indicate that herbivory may indirectly benefit chestnut regeneration by suppressing competition. However, this beneficial effect is likely context‐dependent and additional work is needed to establish the conditions under which it occurs.  相似文献

In March 2014, an outbreak of shoot cankers was observed on grafted Castanea sativa plants in a glasshouse in central Italy. Morphological characteristics led to the identification of isolates of Phomopsis recovered from cankered stems and shoots. Based on the morphological characteristics of colony appearance, shape of conidia and conidiomata as well as sequences of internal transcribed spacer regions (ITS), actin (ACT) and translation elongation factor (TEF‐1α), the fungus was identified as Phomopsis theicola/Diaporthe foeniculina. Pathogenicity test showed that P. theicola isolates were pathogenic to C. sativa when artificially inoculated, reproducing the symptoms originally observed. Koch's postulates were fulfilled by re‐isolating the pathogen. This is the first report of P. theicola/D. foeniculina causing stem and shoot cankers and dieback on C. sativa in Italy or elsewhere.  相似文献

In large parts of Europe horse chestnut trees ( Aesculus hippocastanum) suffer from severe defoliation by an alien invasive species, the specialist leaf mining moth Cameraria ohridella (Lepidoptera; Gracillariidae). Heavily infested trees have a drastically shortened period for photosynthesis. We quantified the effect of moth infestation on reproduction of horse chestnut trees in two different cities in central Europe, Bern and Munich. C. ohridella negatively affected seed and fruit weight of A. hippocastanum at both locations. In Munich, seed weight of heavily damaged trees was reduced by almost half. However, the number of seeds per fruit, the number of fruits per inflorescence, and the number of inflorescences per tree did not change due to herbivory. We conclude that C. ohridella mining affects seed quality but not seed quantity. The reduced seed weight may severely impair growth and survival of horse chestnut seedlings and thus may endanger the long-term persistence of A. hippocastanum in its endemic forests in south-east Europe.  相似文献

Graft failure that occurs in the clonal propagation of chestnuts is a practical problem which has arisen in recent years. Several hypotheses have been put forward to explain reasons for the failure but none have focused on origin and relationships of cultivars. This study was carried out to determine whether relationships of New Zealand chestnut selections and their origin reflect patterns of graft failure within the selections. Two different character data sets, random amplified polymorphic DNA (RAPD) and morpho-nut, were employed for the analyses of the relationships between the chestnut selections. Four different analyses were done to generate trees depicting the relationships of the selections. These were: morpho-nut character, RAPD character, taxonomic congruence (combination of morpho-nut and RAPD trees), and character congruence (combination of morpho-nut and RAPD data sets). When graft failure data were mapped onto the majority rule consensus tree constructed from character congruence analysis, it was found that self graft incompatibility was reflected in the origin and relationships of the chestnut selections. Information on the affinities of the chestnut selections to introduced chestnut species showed that the selections that were mostly implicated in graft failure which are from the North Island had affinities with theCastanea crenata species. But the selections (from the South Island) that were placed withCastanea sativa as well as hybrids (1002 and 1007 from the North Island) ofCastanea mollissima andC. crenata had no failed grafts. This finding indicates that graft failure in New Zealand chestnut selections does not occur by chance but is dependent on the origin and/or evolutionary history of the selections.  相似文献

Vegetative incompatibility in the chestnut blight fungus, Cryphonectria parasitica, in Europe is controlled by six unlinked vic loci, each with two alleles. Four previously identified vic loci (vic1, vic2, vic3, and vic4) were polymorphic in European vegetative compatibility (vc) types. Two new loci, vic6 and vic7, also were identified among European vc types. In one cross, vic genes segregated independently at five loci, and 194 progeny were assigned to 32 vc types; none of these loci were linked. A total of 64 vc types were identified from all crosses. All 64 genotypes possible from six vic loci, each with two alleles (26 = 64), were identified and assigned to vc types. Based on our model, vc types v-c 5 and v-c 10, which had been used in previous genetic studies, differ by only five vic genes. Future studies of vc types in C. parasitica can use knowledge of vic genotypes for analysis of population genetic structure based on vic allele frequencies and to determine the effect of each vic gene on virus transmission between vc types.  相似文献

Spatial structure of both nuclear and mitochondrial RFLPs were studied in several populations of the chestnut blight fungus, Cryphonectria parasitica, using a variety of spatial autocorrelation tests designed to detect nonrandom patterns. Fungal individuals were sampled from cankers on infected chestnut trees, and the location of each tree was mapped. Single-locus nuclear RFLPs, nuclear fingerprints, and mitochondrial DNA haplotypes were determined for each individual. Individuals with the same DNA fingerprint genotypes occurred closer together than would be expected at random in four of the five plots, while mitochondrial DNA haplotypes were aggregated in all five plots. Genetic distances between individuals, expressed as one minus the proportion of shared restriction fragment size classes for fingerprints and mitochondrial haplotypes, were significantly correlated with Euclidean distances between individuals in four of the five populations, but these correlations were very weak (r < 0.18). The same DNA fingerprint and single-copy nuclear RFLP alleles occurred on the same trees or immediately neighbouring trees more often than would be expected at random. Most of the aggregation for all three genetic markers occurred among individuals within the same cluster of chestnut stems or on neighbouring trees. Lack of spatial autocorrelation in one population was probably due to sampling on a larger scale that was too coarse to detect any patterns. Significant aggregation of genotypes in C. parasitica is most likely caused by some degree of restricted dispersal within populations. The implications of restricted dispersal are discussed in relation to the breeding system and isolation by distance in populations of. C. parasitica.  相似文献
