Taurine (Tau) and the small neutral amino acids glycine (Gly), serine (Ser), threonine (Thr), and alanine (Ala) were measured in 53 brain areas of 3- and 29-month-old male Fisher 344 rats. The ratio of highest to lowest level was 34 for Tau, 9.1 for Thr, 7.6 for Gly and Ser, and 6.5 for Ala. The heterogeneity was found in numerous areas; for example, Tau levels were more than 90 nmol/mg protein in 6 areas, and less than 20 nmol/mg protein in 10 areas. Similar heterogeneity was found with the other amino acids. The relative distribution of the small neutral amino acids showed several similarities; Tau distribution was different. With age, four amino acids decreased in 10–18 areas, and increased in only 1–3, while Thr increased in more areas than it decreased. The five amino acids of this paper, and the four of the previous paper, are among the amino acids at highest level in the brain; the sequence in their levels shows considerable regional heterogeneity.  相似文献

Particulate carbohydrates and particulate amino acids were determined in a set of samples from the Paraná River, Argentina, covering a stretch of over 1300 km. To gain an understanding of the factors which control the concentrations of these organic compounds, a multivariate statistical approach was used (R-mode varimax factor analysis). This technique separated the influence of various sources of organic matter. The floodplain of the middle reach (about 900 km long and 20–30 km wide) was the most significant source controlling the concentrations of particulate carbohydrates (PCH), providing angiosperm debris, phytoplankton and grasses. Particulate amino acids (PAA) concentrations were determined markedly both by contributions from the Paraguay basin, and the lotic and lentic environments of the floodplain. Sugars single out forested tributary basins as sources of bacteria while amino acids point toward the Paraguay River as a major source of biodegraded material; heterotrophic processes, however, appear more clearly depicted with PAA than with PCH. A downstream increase of the PCH/PAA ratio suggests a larger consumption of PAA in the lowermost reaches.  相似文献

Abstract The rate of incorporation of 14C from CO2 into glycerol, and the amount of glycerol in cells of Dunaliella tertiolecta was determined at constant salinity at two representative concentrations of NaCl. From these data, the rate of synthesis and turnover of the glycerol pool was determined. The half-time for turnover of the glycerol pool was of the order of 1 h in 170 mol m?3 NaCl and almost 6h for 700 mol m?3 NaCl. These results indicate that turnover of the glycerol pool in Dunaliella is relatively slow under steady-state conditions. Synthesis and dissimilation of glycerol do not apparently constitute a metabolic cycle in the conventional sense. Rather, glycerol metabolism resembles that of the storage polysaccharides which arc commonly produced and degraded by different pathways.  相似文献

On the basis of leaf dry wt, the protein content of six varieties of cassava varied from 29.3 to 38.6% and the estimated leaf protein production ranged from 242 to 953 kg per ha. On the basis of fr. wt of leaf, the total amino acids ranged from 8.42 to 9.4% while the essential amino acids averaged 4.21% and the sulphur-containing amino acids only 0.25%. The amino acid composition profiles for the six varieties was similar.  相似文献

Summary The ordinary aliphatic, neutral amino acids and phenylalanine have been examined for cis-inhibition of influx of alanine (J mc ala ) and lysine (J mc lys ) and trans-stimulation ofJ mc lys across the brush border membrane of rat small intestines: and their effects on the unidirectional mucosa-to-serosa flux (J ms lys ) across the short circuited intestine have been studied. The effects of alanine, -amino-n-butyric acid, leucine, and methionine on the steady-state epithelial uptake of lysine [Lys] c have also been measured. In addition the trans-effects of alanine and leucine have been examined for sodium-dependence, and alanine was tested as trans-stimulator of influx of galactose across the brush border membrane (J mc gal ).All the neutral amino acids were found to be competitive cis-inhibitors ofJ mc lys , and all, except isoleucine, were trans-stimulators ofJ mc lys . The magnitude of the trans-effect was unrelated to the efficiency of the amino acid as cis-inhibitor. As illustrated by alanine, the trans-effects are probably completely sodium-dependent. Alanine was also effective as trans-stimulator ofJ mc gal . With respect to effects on [Lys] c andJ ms lys the neutral amino acids fall into two groups: One which reduces [Lys] c and stimulatesJ ms lys , and one which increases [Lys] c and relatively inhibitsJ ms lys . These effects are not correlated with the affinities of the neutral amino acids for the two carriers involved.It is proposed that the trans-effects onJ mc lys are induced by an electrogenic, sodium-coupled efflux of the neutral amino acid across the brush border membrane, that the stimulation ofJ ms lys is brought about by a selective stimulation (of unknown nature) of efflux of lysine across the basolateral membrane (J cs lys ), assisted by competitive inhibition of lysine efflux across the brush border membrane (J cm lys ), and that the amino acids which do not stimulateJ cm lys increase [Lys] c by competitively inhibitingJ cs lys andJ cm lys .The inhibitory effect of the neutral amino acids onJ mc lys support the view that the carrier of basic amino acids serves as a second carrier of these amino acids.  相似文献

Methylmercury (MeHg) is a potent neurotoxin. The mechanism(s) that governs MeHg transport across the blood-brain barrier and other biological membranes remains unclear. This study addressed the role of the L-type large neutral amino acid transporter, LAT1, in MeHg transport. Studies were carried out in CHO-k1 cells. Over-expression of LAT1 in these cells was associated with enhanced uptake of [(14)C]-MeHg when treated with L-cysteine, but not with the D-cysteine conjugate. In the presence of excess L-methionine, a substrate for LAT1, L-cysteine-conjugated [(14)C]-MeHg uptake was significantly attenuated. Treatment of LAT-1 over-expressing CHO-k1 cells with L-cysteine-conjugated MeHg was also associated with increased leakage of lactate dehydrogenase into the media as well as reduced cell viability measured by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide reduction assay. In contrast, knock-down of LAT1 decreased the uptake of l-cysteine-conjugated MeHg and attenuated the effects of MeHg on lactate dehydrogenase leakage and CHO-k1 cell viability. These results indicate that the MeHg-L-cysteine conjugate is a substrate for the neutral amino acid transporter, LAT1, which actively transports MeHg across membranes.  相似文献

Summary The functionality of isolated brain microvessels — used as anin vitro model of the blood-brain barrier — can be influenced by interaction with cationic proteins. The various polylysines (Mr ranging from 0.9 to 180 kDa) tested affected the activity of both the Na+-dependent (A) and the Na+-independent (L) systems for neutral amino acid transport. Exposure to the 180 kDa polylysine caused a conspicuous inhibition of both transport systems, associated to an increased passive permeability. There was a constant, Mr-dependent, inhibition of the the L-system-mediated uptake of hydrophobic neutral amino acids. The activity of the A-system was enhanced, upon exposure to polymers larger than 22 kDa reaching its peak at 68 kDa and and declining at higher Mr values. The effect which was Na+-ions dependent and abolished by phloretine, could be essentially ascribed to an increased affinity of the MeAIB for its carrier (Km value decreasing from 265 to 169µM in presence of 68 kDa polylysine).  相似文献

Amino acids and neutral sugars in particulate matter were measured weekly from April in 1987 to March in 1988 in Lake Nakanuma, Japan. Changes in concentrations of total amino acids and total neutral sugars corresponded to those of chlorophylla. The composition of amino acids varied little seasonally and vertically. On the contrary, the composition of neutral sugars changed seasonally and vertically. We discuss the relationship between the changes in the biochemical components and environmental factors.  相似文献

A lysimeter study was conducted on Cajeme wheat (Triticum aestivum L.) to investigate the impact of salinity on protein and free amino acid content of the grain. Cross correlations were obtained between 16 different soil-plant-water based parameters and the concentration and total accumulation of amino acids. The results indicated that after 3 years of irrigation, the majority of protein bound and free amino acids increased in concentration in the grain. However, both free tryptophan and free proline revealed decreasing concentrations with increasing salinity. Free tryptophan showed a synergism between total accumulation, yield and concentration. Free proline concentrations decreased in association with increasing protein concentrations. Cross correlations of the 16 soil-plant-water based parameters with free and protein bound amino acids revealed significant correlations for free aspartic acid and glycine with total accumulation but not with concentrations. Only methionine plus cystine was lower than suggested FAO levels for essential amino acids and was lower in the third year than in the first year.  相似文献

The effects of four exogenous amino acids (proline, glycine, asparagine and serine) on the production of maize embryogenic callus and on its endogenous amino acid content have been investigated. For this purpose, an established embryogenic line of Type 1 callus from the inbred W64Ao2 has been used. From the results it may be concluded that a concentration of proline exceeding 6 mM is negative for the production of embryogenic callus. When proline is eliminated from the medium, other amino acids tested in certain concentrations yield a percentage of embryogenic callus production that exceeds or equals that of proline. The endogenous free proline content in embryogenic callus is significantly higher than that in non-embryogenic callus regardless of proline presence in the medium. The only exception are the glycine-containing media, in which endogenous free alanine of embryogenic callus increases at the expense of endogenous free proline. This study suggest a positive role of endogenous free proline or alanine accumulation in the embryogenic callus production which might be related to an adaptation to the metabolic changes produced by in vitro culture and embryogenesis induction. The amino acid compositions of the proteinaceous components of the gum exudates from Prosopis alba, P. chilensis, P. glandulosa, P. laevigata, P. torreyana and P. velutina, and for a sample of commercial gum mesquite, are presented. In agreement with data published previously for the polysaccharide components of their gums, only minor differences in composition are shown by these species. The amino acid compositions are characterized by very high proportions of hydroxyproline and by high proportions of proline and serine; these three amino acids account for 62.5% of those present in the gum from Prosopis velutina. The amino acid compositions of these Prosopis gums are remarkably similar to that established recently for the gum from Acacia senegal (gum arabic).  相似文献

The influence of the source of inorganic nitrogen (KNO3, (NH4)2SO4 and NH4NO3) and its concentration (5, 10, 20 and 30 mM N) on total N incorporation, as well as on N distribution into different fractions (amminiacal, amino, amide and protein) and on free amino acid levels has been determined in grape vine explants cultured in vitro.Increasing concentrations of the nitrogen source resulted in increased total N content in tissues. This effect was small for KNO3, higher for (NH4)2SO4 and maximal for NH4NO3. In addition, nitrate promoted an increase in amino-N only, whereas ammonium increased both the ammoniacal-N and the amino-N fractions. Incorporation of N into amide-N and protein-N were not affected significantly by the N sources tested.The application of increasing quantities of N enhanced the accumulation of most free amino acids, especially arginine, alanine and proline, but to different extents, depending on both the N source and its concentration. The combination of ammonium and nitrate resulted in a higher accumulation of amino acids than that observed with either one of the two forms alone.  相似文献

Developing seeds of chickpea cultivars G-130, L-550 and 850-3/27 grown under field conditions were sampled at different stages of maturity and analysed for soluble sugars, starch, soluble nitrogen, protein nitrogen and amino acids. Fr. wt of seeds of all three cultivars decreased after 28 days of flowering while the dry wt continued to increase. Rapid starch accumulation was observed between 14 and 28 days after flowering. Starch as per cent of seed dry wt started to decrease after 28 days, while starch per seed increased till maturity. The percentage of salt-soluble proteins decreased with maturation of seed. The electrophoretic pattern revealed that deposition of seed storage protein in cotyledons occurred 14 days after flowering. Most of the biochemical activity apparently occurred between 14 and 28 days after flowering.  相似文献

Changes in free amino acids were examined in the central nervous system of mice treated with caffeine for three weeks. Caffeine was administered in the drinking water, and at the end of three weeks the level of caffeine in the cerebral cortex was 113±19 g/g. When amino acid levels in cerebral hemispheres, midbrain, pons and medulla, and cerebellum were measured a significant increase in glutamine levels was found in all four regions. Glycine, alanine, serine, threonine, and GABA were significantly reduced in some regions. Caffeine appears to alter some of the metabolic or transport processes regulating amino acid pools in the brain. The decrease of GABA found in pons and medulla may contribute to the observed increase in reflex excitability after caffeine.Special issue dedicated to Dr. Elling Kvamme  相似文献

The digestive process of Oxyrrhis marina Dujardin feeding on Dunaliella primolecta Butcher commences with the capture of the phototroph, perhaps involving trichocysts which restrain the prey, followed by engulfing by a membrane that appears to originate from near the fiagella pit of the O. marina. The prey, still within the capture membrane, enters the O. marina and is digested over a period estimated to be of the order of 12-24h. During digestion the prey first becomes more electron dense and the contents coagulate. Later the prey becomes less distinct and disintegrates; by this stage remains from several prey may be enclosed within one vacuole. Small O. marina , captured by large prey-depleted O. marina , suffer the same fate. Absence of any correlation between changes in intracellular amino acids and digestion of prey probably resulted from each O. marina containing several prey at various stages of digestion. However, the value of the glutamine/glutamate (Gln/Glu) ratio remained at around 0-3, whilst the population changed from one dominated by N-replete D. primolecta to one dominated by prey-replete O. marina , N-deplete D. primolecta and prey-deplete O. marina both have Gln/Glu of < 0.1. The Gln/Glu ratio may therefore be of use in assessing the nutrient status of heterotrophic microflagellates.  相似文献

Three stands of Phragmites australis (Cav.) Trin. ex Steudel were investigated regarding the relationship between the number of efflux culms and convective ventilation efficiency affecting the hypoxic status of roots and rhizomes. The lack of old (efflux) culms after mowing the preceding winter caused a significantly higher counterpressure within the rhizome, thereby diminishing air flushing rate, i.e. oxygen supply, of rhizomes. The levels of alanine and c-aminobutyric acid in basal culm internodes increased significantly. Both amino acids indicate the hypoxic status of the root and rhizome metabolism of P. australis . Amino acid patterns of the basal culm internodes are discussed with respect to the maintenance of aerobic root metabolism and nutrient availability.  相似文献

Variations of the total free amino acid (FAA) pool and the content of specific amino acids have been measured in the muscle and hepatopancreas of adult shrimps, Penaeus japonicus, acclimatized at five water salinities: 38, 32, 26, 20 and 14%‰ The FAA content is always higher in muscle than in hepatopancreas at all tested salinites. On the other hand, the hepatopancreas exhibits the highest concentrations of essential amino acids. Two steps in the evolution of FAA content can be observed, the first one regarding decrease in salinity from 38 to 20%‰ and the second one, when salinity goes below 20%°. The first step can be characterized by a 16% decrease of total FAA content in the muscle and a 36% increase in the hepatopancreas. In muscle, the variations are mainly due to changes in non-essential FAA content, whereas in the hepatopancreas, they are linked to variations in essential FAA content. The other step is characterized by a drastic increase in moisture and decrease in FAA content in both studied organs when water salinity is 14%‰ The total FAA content is about 40% lower in shrimps at 14%° compared to 38%‰ seawater salinity. During adaptation, the FAA pool (mainly NEFAAs) of muscle seems to be directly related to osmoregulation, whereas in the hepatopancreas, its evolution seems to be linked with energy expenditure and protein synthesis. The results are evaluated in order to elucidate the role of FAA in intracellular osmoregulation and in relation to animal ecology.  相似文献

Abstract

The effect of added amino acid (glycine, alanine and serine) on the solubility of zinc citrate, zinc succinate and zinc oxalate was investigated. Equilibrated solutions were prepared and titrated to endpoint at pH 5.5 with Na2EDTA using Xylenol Orange indicator. In every case, a significant enhancement in solubility was seen as the amino acid increased from 1 to 50 molar excess. The increase was non-linear, and, with zinc succinate, showed saturation effects. In these cases, solution composition may be more complicated than initially thought, including mixed-ligand complexes and increasing percent free metal ion as the proportion of amino acid increases.  相似文献

The factorial approach to assess the amino acid (AA) requirements of pigs is based on the assumption that the AA composition of body protein is constant. However, there are indications that this assumption may not be valid because the AA composition of body protein can be affected by the AA supply. The extent to which different tissues are affected by an AA deficiency is unknown. The objective of this study was to investigate the effect of feeding pig diets with a deficient or sufficient total sulfur AA supply (TSAA; Met+Cys) from 6 to 23 weeks of age on tissue composition and meat quality. The deficient diet (TSAA–) provided 24% Met : Lys and 51% TSAA : Lys on a standardized ileal digestible basis, which are 19% and 16% below the recommended requirements, respectively. The sufficient diet (TSAA+) provided 33% Met : Lys and 60% TSAA : Lys. Diets were offered slightly below the ad libitum feed intake capacity of the pigs. Pigs offered diet TSAA– had a lower average daily gain, lower weights of the longissimus dorsi (LM) and rhomboideus muscles (RM), and of selected skin sections (P<0.05). The weight of different sections of the small intestine and the liver was not affected by the diet. The protein content of the LM and RM decreased in pigs offered diet TSAA– (P<0.05), whereas the protein content of other tissues was not affected. The TSAA supply affected the AA composition (g/16 g N) of protein in all tissues, but the Met content was changed only in the liver (P<0.05). Pigs receiving diet TSAA– had a lower Cys content in the RM and in the distal jejunum and ileum (P<0.01). The deficient TSAA supply resulted in a lower carcass weight and higher muscle glycogen stores (P<0.05), but did not affect other meat quality traits. The results of this study indicate that the muscles, jejunum and ileum respond more to a prolonged AA deficiency than the liver. In addition, the observed changes in AA composition of tissue protein question the use of a constant AA profile of retained protein to assess AA requirements.  相似文献
