Abstract: The laminar pulvinus of bean ( Phaseolus vulgaris L.) is a highly specialized organ for reorienting the lamina, and exhibits positive phototropic curvature. Structural and ultrastructural features of the pulvinus were studied to determine their possible role in its phototropic response. The vascular tissue forms a flexible, relatively inextensible central core enclosed by a starch sheath and surrounded by a multi-layered motor tissue. Phototropic curvature is a result of opposite anisotropic changes in volume of the cells in the exposed and opposite sectors of the motor tissue. Radial inflexibility of the epidermis and axial plasticity constrain these changes to the pulvinar axis. Anisotropic changes in volume of motor cells reduce the osmotic work involved. Motor cells exhibit features that are associated with high synthetic activity: thick cytoplasm with numerous ribosomes, polysomes, RER and SER, well-developed mitochondria and a large nucleus. Numerous, well-developed chloroplasts, with little starch, are increasingly abundant toward the periphery. The intercellular system is limited and partially filled with matrix. Stomata are absent and the motor tissue lacks vascularization. These features support the suggestion that the primary role of the chloroplasts in the photonastic response is photophosphorylation and photosynthetic electron transport (Koller et al., 1995[339]).  相似文献

The purpose of this study was to evaluate the temperature response of photosynthesis in two common bean genotypes differing in crop yield when grown under warm conditions. The cultivar Nobre is sensitive to high temperatures, whereas Diplomata shows better crop yield under high temperatures. Plants were grown in a greenhouse prior to transferring to a controlled environment cabinet for the temperature treatments. In a first experiment, 30 days-old plants were subjected to a short exposure (1 day) at temperatures that varied from 9 °C to 39 °C. Diplomata had lower net CO2 assimilation rate (A) at 15 °C and 21 °C, but higher from 27 °C to 39 °C. Photosynthetic parameters calculated from modeling the response of A to the intercellular CO2 concentration suggested that the different temperature responses of the two genotypes are caused by different rates of diffusion of CO2 to the assimilation site, not by differences in biochemical limitations of photosynthesis. While stomatal conductance (gs) did not differ between the genotypes, mesophyll conductance (gm) was slightly greater for Nobre at 15 °C, but much higher in Diplomata from 21 °C to 39 °C. In a second experiment, no difference was observed in biomass accumulation between the two genotypes after growth for 24 days under a 35/20 °C (day/night) regime. Hence, the differences in photosynthesis did not cause variation in plant growth at the vegetative stage. The differential genotypic response of gm to temperature suggests that gm might be an important limitation to photosynthesis in Nobre, the common bean genotype sensitive to elevated temperature. However, more studies are needed employing other methods for gm evaluation to validate these results.  相似文献

Supra-optimal levels of zinc in primary leaves of Phaseolus vulgaris increased the CO2 compensation point and inhibited net photosynthesis. Leaf morphology was modified: mesophyll intercellular area, stomatal slit length and interstomatal distance were reduced, but stomatal density increased. Internal and stomatal conductances to CO2 diffusion decreased. These changes are discussed in relation to the observed effects on leaf gas exchange and to the previously reported inhibition of different photosynthetic and photorespiratory enzymes.  相似文献

The application of 2-(3,4-dichlorophenoxy)-triethylamine to guayule (Parthenium argentatum Gray var 593) plants results in a 2-fold stimulation of rubber synthesis and a 1.5- to 3-fold increase in mevalonic acid kinase, isopentenyl pyrophosphate isomerase, and rubber transferase. The increase in these enzymic activities accounts in part for the chemical induction of rubber synthesis.  相似文献

The increase in the basal fluorescence (F0) by increases in temperature has been widely used as a screening technique to assess the temperature at which plant species suffer irreversible damage in their photosynthetic apparatus. This method has been used in this study to assess the resistance to extreme high temperature in two bean varieties, Barbucho (Chilean, non commercially bred) and Blue lake (commercially available in the UK). The results suggested that the two varieties have a similar threshold for high temperature at 42–43 °C, however, at 40 °C, Barbucho maintains its photosynthetic functions for a longer period of time compared to Blue-Lake as observed by mean of simultaneous measurement of gas exchange and room temperature fluorescence. It seems likely that inhibition at 40 °C in Blue-Lake is related to the instability of Photosystem II.  相似文献

When carbohydrates accumulate in leaves, photosynthesis is repressed. Limited nitrogen nutrition is thought to enhance this repressing effect. However, the interaction between carbohydrate and nitrogen limitation in leaf photosynthesis has not been examined intensively. In this study, we grew Phaseolus vulgaris L. plants at three different nitrogen levels, and examined the effects of sucrose feeding to the roots on the nitrogen content, carbohydrate content and photosynthetic properties of the primary leaves. Nitrogen content and photosynthetic rate were lower and the carbohydrate content was greater in plants grown with limited nitrogen than in well-fertilized plants. Sucrose feeding to the plants increased carbohydrate content and decreased photosynthetic rate and nitrogen content. The increase in carbohydrate content and the decreases in nitrogen content and photosynthetic rate occurred at the same time, and the negative relationship between the carbohydrate content and photosynthetic rate did not differ among nitrogen nutrition levels. These results show that carbohydrate accumulation in the leaves leads to a decrease in photosynthetic rate. At low nitrogen nutrition levels, carbohydrates accumulated markedly, which accelerated this effect. It appears that the nitrogen nutrition level influences leaf photosynthesis through changing the carbohydrate level rather than through modifying sensitivity of the leaf to the carbohydrate level.  相似文献

Abstract

Callus production and plant regeneration from different explants of Phaseolus vulgaris L. cv. Giza are reported. Calli cultures were induced from leaf, hypocotyl, embryo and root explants. Rapid growth of callus was achieved by leaf explants cultured on MS salts, B5 vitamins and supplemented with 2,4— dichlorophenoxyacetic acid (2, 4—D)+0.5 mg/l kinetin (kin). Addition of casein hydrolysate at 2 g/l to maintenance medium enhanced callus growth and hindered the early appearance of necrotic parts. This report also provides a detailed method for production of multiple shoots directly from the wounded edges of immature cotyledon explant via organogenesis on 1 mg/l benzyladenine (BA) or indirectly on 0.5 mg/l naphthaleneacetic acid (NAA)+2 mg/l BA. The regeneration of bean plants through the two ways described here (direct or indirect) may be of use in genetic improvement of bean.  相似文献

The major cytokinins in stems of decapitated, disbudded bean plants have been identified by enzymic degradation, Sephadex LH20 and reversed phase high performance liquid chromatography, and by combined gas chromatography-mass spectrometry as 6-(4-hydroxy-3-methylbut-trans-2-enylamino)-9--D-ribofuranosylpurine (zeatin riboside), 6-(4-hydroxy-3-methylbutylamino)-9--D-ribofuranosylpurine (dihydrozeatin riboside), and the 5-phosphates of these compounds (zeatin ribotide and dihydrozeatin ribotide). Minor cytokinins in this tissue were tentatively identified as dihydrozeatin-O--D-glucoside and zeatin ribotide-O--D-glucoside. [8-14C-]Dihydrozeatin appeared to be rapidly metabolized to dihydrozeatin ribotide when supplied to segments of stems from decapitated plants. Measurements of net photosynthesis show that in Phaseolus vulgaris L. the cultivar Michelite-62 exceeds the cultivar Red Kidney in net CO2 uptake by 23 to 31%. Data on translocation of pulse label indicate that export of a pulse of photosynthetically assimilated 14C from the source leaf of either M-62 or Red Kidney follows an exponential pattern and shows an initial rapid phase followed by a second slower phase. The steeper slope for both phases in M-62 suggests its rate of translocation of pulse label is higher than that of Red Kidney. Furthermore, only 38% of the 14C remains in the leaf of M-62 after 8 hours, while Red Kidney retains up to 60% of the label. Leaf autoradiographs obtained after pulse labeling demonstrate a much faster rate of vein loading in M-62 and are considered evidence for the higher translocation efficiency of M-62. These results provide evidence for a positive correlation between photosynthetic efficiency and translocation efficiency in M-62 and Red Kidney and give support to our hypothesis that translocation is one of the important physiological factors controlling the varietal differences in photosynthetic efficiency in Phaseolus vulgaris.  相似文献

The influence of far-red (FR; 700–800 nm) radiation on steady-state stomatal conductance and net photosynthesis in P. vulgaris has been studied. Whereas FR radiation alone was relatively ineffective, addition of FR to a background of white light (WL; predominantly 400–700 nm) resulted in increased stomatal conductance. Stomata exhibited a marked diurnal sensitivity to FR. The action maximum for enhancing stomatal conductance was near 714 nm. A combination of FR and infra-red (IR; >800 nm) enhanced net photosynthesis when added to a background of WL. When IR alone was added to WL, there was a net decrease in photosynthesis, indicating that it is the FR waveband which is responsible for the observed photosynthetic effects. The protochlorophyll(ide) forms and plastid ultrastructure were investigated in hypocotyls of dark-grown seedlings of kidney bean ( Phaseolus vulgaris L. cv. Brede zonder draad). By deconvolution of the fluorescence emission spectra into Gaussian components three protochlorophyll(ide) forms were found with maxima at 633, 642 and 657 nm, respectively. The ratio of protochlorophyll(ide) emitting at 657 nm to protochlorophyll(ide) emitting at 633 nm decreased downwards the hypocotyl. The gradient was established already after 4 days in dark-grown Phaseolus and was also seen in hypocotyls of 7-day-old dark-grown plants of 8 other species. Ultrastructural observations revealed a plastid developmental sequence along the hypocotyl. Plastids in the upper parts of the hypocotyl contained prolamellar bodies typical of etiolated leaves while those in the lower parts contained only stroma lamellae. Immunological detection of NADPH-protochlorophyllide oxidoreductase (EC 1.3.1.33) on nitrocellulose membranes after sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDSPAGE) indicated the occurrence of the enzyme in upper, middle and lower sections of hypocotyls and in the root tips.  相似文献

Accumulation of non-structural carbohydrate in leaves represses photosynthesis. However, the extent of repression should be different between sink leaves (sugar consumers) and source leaves (sugar exporters). We investigated the effects of carbohydrate accumulation on photosynthesis in the primary leaves of bean (Phaseolus vulgaris L.) during leaf expansion. To increase the carbohydrate content of the leaves, we supplied 20 mM sucrose solution to the roots for 5 d (sugar treatment). Plants supplied only with water and nutrients were used as controls. The carbohydrate contents, which are the sum of glucose, sucrose and starch, of the sugar-treated leaves were 1.5-3 times of those of the control leaves at all developmental stages. In the young sink leaves, the photosynthetic rate at saturating light and at an ambient CO2 concentration (A360) did not differ between the sugar-treated and control leaves. The A360 of sugar-treated source leaves gradually decreased relative to the control source leaves with leaf expansion. The initial slope of the A-Ci (CO2 concentration in the intercellular space) curve, and the Rubisco (ribulose-1,5-bisphosphate carboxylase/oxygenase) content per leaf area showed trends similar to that of A360. Differences in Amax between the treatments were slightly smaller than those in A360. These results indicate that the effect of carbohydrate accumulation on photosynthesis is significant in the source leaves, but not in the young sink leaves, and that the decrease in Rubisco content was the main cause of the carbohydrate repression of photosynthesis.  相似文献

The capacity for photosynthesis is often affected when plants are grown in air with elevated CO2 partial pressure. We grew Phaseolus vulgaris L. in 35 and 65 Pa CO2 and measured photosynthetic parameters. When assayed at the growth CO2 level, photosynthesis was equal in the two CO2 treatments. The maximum rate of ribulose-1,5-bisphosphate (RuBP) consumption was lower in plants grown at 65 Pa, but the CO2 partial pressure at which the maximum occurred was higher in the high-CO2-grown plants, indicating acclimation to high CO2. The acclimation of RuBP consumption to CO2 involved a reduction of the activity of RuBP carboxylase which resulted from reduced carbamylation, not a loss of protein. The rate of RuBP consumption declined with CO2 when the CO2 partial pressure was above 50Pa in plants grown under both CO2 levels. This was caused by feedback inhibition as judged by a lack of response to removing O2 from the air stream. The rate of photosynthesis at high CO2 was lower in the high-CO2-grown plants and this was correlated with reduced activity of sucrose-phosphate synthase. This is only the second report of O2-insensitive photosynthesis under growth conditions for plants grown in high CO2.  相似文献

Circumnutation of Phaseolus vulgaris L. cv. Blanc de Juillet twining shoots was examined at the cellular level. Cell growth was monitored inside the free-moving part of the shoot as a function of time and position with respect to the terminal bud. Complete elongation of epidermal cells required >7 days. A 60% increase in length of the cells in the bending zone was observed. Scanning and transmission electron microscopy studies showed the elasticity of cell walls, the extent of plasmodesmata and endoplasmic reticulum of epidermal cells. The osmotic potential in epidermal cells of the bending zone displayed significant differences only between the concave and convex sides. The pattern of cell growth in twining shoots is discussed. Circumnutation could be seen as a consequence of periodic turgor changes inducing periodic growth.  相似文献

An investigation was conducted to determine the effects of 10 pphm SO2 and 10 pphm NO2, applied both singly and in combination, on whole plant transpiration rates of Phaseolus vulgaris L. The individually applied pollutants caused short-term increases and the pollutant combination caused a decrease in transpiration rate. Aluminium toxicity is one of the major limiting factors of crop productivity on acid soils. High levels of available aluminium in soil may induce phosphorus deficiency in plants. This study investigates the influence of Aluminium (Al) on the phosphate (Pi) uptake of two Phaseolus species, Phaseolus vulgaris L. var. Red Kidney and Phaseolus lunatus L. The two bean species were treated first with solutions of Al at different concentrations (0, 25, 50 and 100 μM, pH 4.50) and second with solutions of Pi (150 μM) at pH 4.50. The higher the Al concentration the higher the Al concentration sorbed but P. vulgaris L var. Red Kidney adsorbed significantly more Al than P. lunatus L. Both species released organic acids: P. vulgaris L var. Red Kidney released fumaric acid and P. lunatus L. fumaric and oxalic acids which could have hindered further Al uptake.The two bean species showed a sigmoid Pi uptake trend but with two different mechanisms. P. vulgaris L var. Red Kidney showed a starting point of 3 h whereas P. lunatus L. adsorbed Pi immediately within the first minutes. In addition, P. vulgaris L var. Red Kidney presented significantly higher Pi uptake (higher uptake rate ‘k’ and higher maximum adsorption ‘a’ of the kinetic uptake model). The Al treatments did not significantly influence Pi uptake. Results suggest that P. lunatus L. might adopt an external Al detoxification mechanism by the release of oxalic acid. P. vulgaris L var. Red Kidney on the other hand seemed to adopt an internal detoxification mechanism even if the Al sorbed is poorly translocated into the shoots. More detailed studies will be necessary to better define Al tolerance and/or resistance of Phaseolus spp.  相似文献

Persistent decrease in the productivity of cowpea (Vigna unguiculata L.) has been partly due to attack by bruchids including Zabrotes subfasciatus and Callosobruchus maculatus. Resistance to these insects in Phaseolus vulgaris L. has been shown to be associated with arcelins, a family of seed proteins encoded by a multigenic family of lectins on the APA locus. In this work, we report the construction of an expression vector containing Arc1 gene isolated from P. vulgaris and introduced into cowpea as a strategy to confer resistance to insect attack. Following transformation and selection, feeding experiments in which C. maculatus and Z. subfasciatus were fed with transgenic (L3 and L5) and non-transgenic (control) grains showed that introduced gene protected the transgenic line. Significant differences (p < .05 and p < .01) were found in the number of eggs laid, the number of emerging insects and the loss of grain mass in L3, compared with control, for both insects. Similar observations were made in L5 with the exception of the number of laid eggs. The strategy here described may form the basis for the development of a cowpea variety tolerant to bruchids in a crop cultivated by farmers throughout Latin America and Africa.  相似文献

The glutamine synthetase (GS) isozymes in the plant fraction of nodule extracts from 62 cultivars of Phaseolus vulgaris L. and one cultivar of Phaseolus lunatus L. were analyzed by polyacrylamide gel electrophoresis. All P. vulgaris nodule extracts displayed two GS activity bands: a nodule-specific band (GSn1) and a band (GSn2) similar to the single band (GSr) present in root extracts. In nodule extracts of P. lunatus, the GSn1 band was detected, but the GSn2 band was barely detectable. In contrast to P. vulgaris, the GSn2 band and the GSr band of P. lunatus appeared to be different. The electrophoretic mobility of the GSn1 band in P. vulgaris was governed by both the plant cultivar and the development stage of the nodule. In nodule extracts of P. vulgaris and P. lunatus, the zone of GSn1 activity coincided with six to nine distinct protein bands as revealed after treatment of gels, which had previously been stained for GS activity, with Coomassie blue. All these protein bands were shown to consist of polypeptides of identical molecular weight (approximately 47,000 daltons) by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Our results indicate that P. vulgaris continuously generates isozymes of GSn1 of increasing electrophoretic mobility during the course of nodule development.  相似文献
