Field-grown large leaf Rhododendron and Kalmia latifolia were treated with one of three concentrations of paclobutrazol or uniconazol in April, June, or August in the second year from propagation. The elongation of stems was measured as was the number of flower buds initiated. Spray applications in April or June increased flowering at the lowest concentrations tested; 4 mg · L–1 paclobutrazol and 1.5 mg · L–1 uniconazol. Flowering was induced in cultivars that normally did not flower, and the number of flower buds per plant was increased in cultivars that normally flowered. All treatments that induced flowering also reduced stem elongation. Spray application in August failed to enhance flowering. At the concentrations tested, uniconazol was more effective than paclobutrazol in increasing the number of flower buds and reducing stem elongation of Rhododendron. For Kalmia, there was less response to the concentration of growth retardant, and the two chemicals enhanced flowering equally.  相似文献

Gas exchange and abscisic acid content of Digitalis lanata EHRH. have been examined at different levels of plant water stress. Net photosynthesis, transpiration and conductance of attached leaves declined rapidly at first, then more slowly following the withholding of irrigation. The intercellular partial pressure of CO2 decreased slightly. The concentration of 2-cis(S)ABA increased about eight-fold in the leaves of non-irrigated plants as compared with well-watered controls. A close linear correlation was found between the ABA content of the leaves and their conductance on a leaf area basis. In contrast, the plot of net assimilation versus ABA concentration was curvilinear, leading to an increased efficiency of water use during stress. After rewatering, photosynthesis reached control values earlier than transpiration, leaf conductance and ABA content. From these data it is concluded that transpiration through the stomata is directly controlled by the ABA content, whereas net photosynthesis is influenced additionally by other factors.Possible reasons for the responses of photosynthesis and water use efficiency to different stress and ABA levels are discussed.Abbreviations A net CO2 assimilation - ABA abscisic acid - Ci intercellular CO2 concentration - g stomatal conductance - T transpiration - WUE water use efficiency  相似文献

EAF1 regulates vegetative-phase change and flowering time in Arabidopsis.   总被引:3,自引:0,他引:3  
We have identified a new locus that regulates vegetative phase change and flowering time in Arabidopsis. An early-flowering mutant, eaf1 (early flowering 1) was isolated and characterized. eaf1 plants flowered earlier than the wild type under either short-day or long-day conditions, and showed a reduction in the juvenile and adult vegetative phases. When grown under short-day conditions, eaf1 plants were slightly pale green and had elongated petioles, phenotypes that are observed in mutants altered in either phytochrome or the gibberellin (GA) response. eaf1 seed showed increased resistance to the GA biosynthesis inhibitor paclobutrazol, suggesting that GA metabolism and/or response had been altered. Comparison of eaf1 to other early-flowering mutants revealed that eaf1 shifts to the adult phase early and flowers early, similarly to the phyB (phytochrome B) and spy (spindly) mutants. eaf1 maps to chromosome 2, but defines a locus distinct from phyB, clf (curly leaf), and elf3 (early-flowering 3). These results demonstrate that eaf1 defines a new locus involved in an autonomous pathway and may affect GA regulation of flowering.  相似文献

The plant growth retardant, paclobutrazol at 8.5 or 17.0 μM concentrations effectively inhibited the stem elongation and primary leaf expansion of bean seedlings. Although the retardant reduced the relative water content in well-watered plants, the water and pressure potentials remained high in the primary leaves. K+, Na+, Mg2+ and Ca2+ contents in the primary leaves of the paclobutrazol-treated plants were not significantly different from those in the control. The stomatal density increased on both surfaces but the length of guard cells was not reduced significantly on the adaxial epidermes of the paclobutrazol-treated primary leaves. The inhibitory effect of paclobutrazol on the abaxial stomatal conductances became more pronounced with time during the light period but the adaxial surfaces displayed similar or slightly higher conductances than those of the control. The transpiration rate on a unit area basis did not change significantly or increased in the treated leaves thus the reduced water loss of paclobutrazol-treated plants was due to the reduced leaf area. Stomatal conductances of the adaxial surfaces responded more intensively to exogenous abscisic acid and the total leaf conductance decreased faster with increasing ABA concentration in the control than in the paclobutrazol-treated leaves. Paclobutrazol, an effective inhibitor of phytosterol biosynthesis, not only amplified the stomatal differentiation but increased the differences between the adaxial and abaxial stomatal conductances of the primary leaves.  相似文献

Paclobutrazol [(2RS,3RS)-1-(4-chlorophenyl)methyl-4,4-dimethyl-2-(1h-1,2,4-trizol-1-yl)penten-3-ol] effectively decreased vegetative growth of rice (Oryza sativa L.) seedlings and increased the chlorophyll content. The number of veins in a leaf, the calculated number of stomata per leaf, and the length of guard cells were not altered by the paclobutrazol treatment, suggesting an effect on cell elongation. The allocation pattern of carbohydrates was changed by either gibberellin (GA) or paclobutrazol treatment. GA3 induced more shoot growth and less accumulation of starch than the control and paclobutrazol-treated seedlings. Photosynthetic ability was not affected by either paclobutrazol or GA3 treatment. Paclobutrazol-treated plants allocated a smaller amount of photosynthates for vegetative shoot growth and stored more as starch in the crowns than the control and GA3-treated plants. The same starch degrading activity in the crown tissue of paclobutrazol-treated seedlings as in control plants suggests that the accumulated starch is utilized in a normal activity for growth including leaf emergence, tiller formation, and root production, resulting in improved seedling quality. Apical buds of Spinacia oleracea L. cv. Matador were isolated from 7-day-old vegetative seedlings and grown in sterile culture under inductive long, or non-inductive short photoperiods. Flowering of isolated apices was inducible in long days in approximately 75% of the plants, and in short days by gibberellin treatment (about 40%) or by raising the temperature to 30–32°C (13%). In long days the percentage of flowering was further increased by gibberellin treatment (up to 90%), while it was unaffected by abscisic acid and was strongly decreased by Amo 1618 (55%). In long days the ratio of male to female plants was near 1. The percentage of female plants in long days was increased by gibberellins, and the percentage of male ones decreased by kinetin; as a consequence, the ratio of male to female plants was lowered to about 0.50 in both cases. Abscisic acid and Amo 1618 had the opposite effect, probably because they decreased the flowering in female plants, so that the sex ratio was shifted to 2.6 and 6.8, respectively. Simultaneous treatment with GA3 reversed the effect of abscisic acid on the sex ratio, but the reversal of the shift to maleness induced by Amo 1618 was only partial. In short days, gibberellins also stimulated the flowering in female plants more than in male. However, when the flowering was induced by higher temperature, most flowering plants were male and kinetin increased their percentage further. The above results have been discussed in terms of different requirements for flowering in male and female plants.  相似文献

American elm (Ulmus americana) seedlings were either non-inoculated or inoculated with Hebeloma crustuliniforme, Laccaria bicolor and a mixture of the two fungi to study the effects of ectomycorrhizal associations on seedling responses to soil compaction and salinity. The seedlings were grown in the greenhouse in pots containing non-compacted (0.4 g cm?3 bulk density) and compacted (0.6 g cm?3 bulk density) soil and subjected to 60 mM NaCl or 0 mM NaCl (control) treatments for 3 weeks. All three fungal inocula had similar effects on the responses of elm seedlings to soil compaction and salt treatment. In non-compacted soil, ectomycorrhizal fungi reduced plant dry weights, root hydraulic conductance, but did not affect leaf hydraulic conductance and net photosynthesis. When treated with 60 mM NaCl, ectomycorrhizal seedlings had several-fold lower leaf concentrations of Na+ compared with the non-inoculated plants. Soil compaction reduced Na+ leaf concentrations in non-ectomycorrhizal plants and decreased dry weights, gas exchange and root hydraulic conductance. However, in ectomycorrhizal plants, soil compaction had little effect on the leaf Na+ concentrations and on other measured growth and physiological parameters. Our results demonstrated that ECM associations could be highly beneficial to plants growing in sites with compacted soil such as urban areas.  相似文献

Ultrastructural alterations in mesophyll cells as well as variations in bulk leaf endogenous ABA and IAA concentrations were studied in water-stressed field-grown plants of Fatsia japonica. Under water deficit cellular membranes were modified and an increase in vesicles was observed. The main damage to the chloroplasts included thylakoid swelling and disruption of the chloroplast envelope. Concomitant variations in abscisic acid and indole-3-acetic acid were observed. Despite the expected increased in endogenous ABA concentration in relation to water stress, after the highest concentration of ABA, observed at predawn in severely stressed plants (29-1), there was a sharp decline from 2768 pmol g fw–1 to 145 pmol g fw–1; thus in severely stressed plants ABA levels were not related to changes in bulk leaf ABA contents. Summary The effects of ancymidol, abscisic acid (ABA), uniconazole, and paclobutrazol on asparagus somatic embryogenesis were evaluated. Calli induced from seedlings of genotype G447 were transferred to embryo induction medium (MS plus 3% sucrose, 0.1 mg L–1 NAA, 0.5 mg L–1 kinetin and 3% gelrite), with different concentrations of these compounds. After 8 weeks, the recovered bipolar or globular embryos were placed on germination medium (MS plus 6% sucrose, 0.1 mg L–1 NAA, 0.1 mg L–1 kinetin, 0.75 mg L–1 ancymidol, 40 mg L–1 adenine sulphate dihydrate, 0.17 mg L–1 sodium phosphate monobasic and 3% gelrite) for conversion to plantlets. Inclusion of ancymidol, ABA, uniconazole and paclobutrazol in the embryo induction medium did not affect the total number of somatic embryos produced relative to the control without these compounds. However, ancymidol, ABA and uniconazole significantly improved embryo development by increasing the production of bipolar embryos 250–750% and decreasing that of globular embryos 8–35% relative to the control. The bipolar embryos produced with any of the four compounds in the embryo induction medium converted to plantlets at rates 700–1100% greater than the control. None of the globular embryos converted to plantlets. Ancymidol (0.75 mg L–1) and ABA (0.05 mg L–1) were the most effective treatments; 61 and 46 bipolar embryos g–1 callus were produced, and 38% and 37% of the bipolar embryos converted to plantlets, respectively. 该试验以绿化卷材为基质材料,对沙场、渣场和混凝土屋面3种立地条件下生长的黄荆进行不同浓度(0、100、200、300、400mg·L~(-1))多效唑处理,研究根施多效唑对黄荆生长和生理特征的影响以及不同立地环境的应用差异。结果显示:(1)随多效唑浓度升高,3种立地类型黄荆株高和生物量呈降低趋势,冠幅、基径、叶面积、根幅、主根长和主根径呈减小趋势,叶片长宽比和根冠比表现出增大的趋势;多效唑处理使黄荆叶片相对含水量、叶绿素含量、可溶性糖和可溶性蛋白含量增加,使丙二醛含量下降。(2)不同立地条件下黄荆对多效唑处理的表现具有一定差异,隶属函数法综合评价显示,对沙场、渣场和屋面3种立地类型的黄荆生长调控效果最佳的多效唑浓度分别为400mg·L~(-1)、300mg·L~(-1)、100mg·L~(-1)。(3)当多效唑浓度在渣场和屋面分别为400、300mg·L~(-1)时,黄荆叶片开始受到伤害,对多效唑的耐受阈值表现为沙场渣场屋面。研究认为,多效唑可有效调控黄荆的形态和生物量分配,增强细胞渗透调节和抗氧化损伤能力,从而提高黄荆的抗逆性和环境适应性,但在应用时应考虑不同立地背景的差异,因地制宜地选择使用浓度和用量,使其更好地应用于人工植被恢复与建设。  相似文献

The effect of abscisic acid (ABA) on assimilate transport in barley was investigated in two parallel experiments. First, the effect upon [14C]sucrose transport from the flag leaf to the ear of a single ABA application made at different stages of growth of the fruits was investigated; the effect was measured 24 h after treatment. Second, the effect of a single application of ABA made at the same stages of growth as above on grain weight of the mature plant was investigated. In both types of experiments ABA was applied once to the ear of different plants as an aqueous solution (10-3–10-5 M), one to five weeks after anthesis. [14C] sucrose was applied by means of agar blocks. Parallel to these experiments, the endogenous content of ABA was investigated in the developing grains. When ears were treated with ABA two or four weeks after anthesis, an increase of up to 70% in the 14C-transport from the flag leaf to the ear was observed within a 24-h period after treatment (short duration experiments). At these growth stages the endogenous concentrations of ABA were low. In sharp contrast, ABA, especially in a concentration of 10-3 M, decreased 14C-import from the flag leaf when applied three weeks after anthesis. At this stage the endogenous ABA content had reached its maximum. Long duration experiments with a single application of ABA to the car two weeks after anthesis resulted in a marked increase of weight per thousand kernels. ABA applications made earlier or later than two weeks after anthesis either reduced the grain weight or had no effect. It is concluded that ABA is involved in the regulation of assimilate transport from the leaves to the grains, possibly by influencing the unloading of sieve tubes in the ears. Summary Mutant lines of Nicotiana plumbaginifolia resistant to the synthetic auxins 1-naphthaleneacetic acid (NAA) and indole-3-butyric acid (IBA) were isolated as germinating seedlings on selective medium. In each case, resistance was conferred by a single recessive nuclear mutation at one of 3 loci designated iba1, iba2 and iba3. Labelling studies with 14C NAA suggest that resistance was not due to changes in the uptake or metabolism of NAA. Plants homozygous for the iba1 mutation exhibit a syndrome of atypical germination and growth suggestive of a defect in the biosynthesis, metabolism or localization of abscisic acid. Wild-type seeds treated with gibberellin exhibit the same syndrome, including resistance to NAA and IBA. Short term waterlogging affected the growth of sorghum seedlings as indicated by a high mortality rate of the seedlings and a decrease in the shoot and root biomass, net assimilation rate (NAR), leaf area ratio (LAR), relative growth rate (RGR), total photosynthetic area and rate of photosynthesis. Plant resistance to the stress was exhibited by decreased stomatal index (SI), rate of transpiration and rate of respiration. When the stressed plants were exposed to normal conditions, the recovery of the seedlings was rapid and was enhanced by foliar application of kinetin at 5 mgl-1.  相似文献

A barley (Hordeum vulgare L.) mutant (cool) with leaf transpiration unaffected by the application of 1 mM abscisic acid (ABA) was isolated from the population of M2 seedlings using thermography (electronic visualization, and quantitation of the temperature profiles on the surface of the leaves). Stomata of the mutant plants were insensitive to exogenously applied ABA, darkness, and such desiccation treatments as leaf excision and drought stress. The evaporative cooling of the leaves of the cool barley was always higher than that of the wild-type barley, even without ABA application, indicating that the diffusive resistance of the mutant leaves to water loss was always lower. Guard-cell morphology and stomatal density as well as ABA level and metabolism were seemingly unaltered in the mutant plants. The plant growth retardant paclobutrazol was applied by foliar spray to seedlings of Eucalyptus globulus (Labill.) to determine its effects on reproductive activity. It was shown to induce flowering in vegetatively juvenile seedlings and increase reproductive activity in vegetatively adult material. The former case represented a 50% (three year) reduction in generation time for commercial seedlings. Paclobutrazol treatment did not appear to have a negative effect on any aspect of reproductive development, germination of new seed, or early seedling growth, while having a distinct positive effect on reproductive output. This suggests that paclobutrazol may be an effective tool enabling tree breeders to reduce generation time and increase reproductive output in E. globulus. Foliar spraying during autumn was shown to be an effective means and timing of paclobutrazol application, with results evident in the following flowering season. Considerable persistence of paclobutrazol effects was noted in the second season after application. Pot size and growth temperatures also play roles in determining the extent of new reproductive activity each season in E. globulus seedlings.  相似文献

The interactive effects of gibberellic acid, abscisic acid,and benzyladenine were studied in relation to runner formationof the everbearing strawberry. In two of three cultivars studied,Ozark Beauty and Superfection, exogenous gibberellic acid stimulatedrunner formation and inhibited flowering. In the Geneva cultivar,a shy runner producer, gibberellic acid application as a foliarspray failed to induce runners. When applied in combinationwith benzyladenine, gibberellic acid greatly stimulated runnerformation Benzyladenine alone had no effect on runnering. Exogenous abscisic acid inhibited petiole length and runnerformation and had no influence on the number of inflorescencesinitiated. Response to exogenous gibberellic acid or abscisicacid varied according to the reproductive or vegetative stagethe plant exhibited at the time of treatment. Activity of endogenous gibberellin-like substances was not substantiallydifferent between flowering and non-flowering field-grown Genevaplants when sampled in long day lengths of August. Endogenousinhibitors were highly active in flowering Geneva plants andrelatively inactive in non-flowering plants The implications of these findings on hormonal balance in thestrawberry are discussed.  相似文献

Winter wheat plants were grown in open top chambers either at 365 µmol mol–1 (AC) or at 700 µmol mol–1 (EC) air CO2 concentrations. The photosynthetic response of flag leaves at the beginning of flowering and on four vertical leaf levels at the beginning of grain filling were measured. Net photosynthetic rates (P N) were higher at both developmental phases in plants grown at EC coupled with larger leaf area and photosynthetic pigment contents. The widely accepted Farquhar net photosynthesis model was parameterised and tested using several observed data. After parameterisation the test results corresponded satisfactorily with observed values under several environmental conditions.  相似文献

Somatic embryos of Cyclamen persicum Mill. could be produced through a callus phase from juvenile explant material including anthers, ovaries and zygotic embryos. The auxin 2,4-D (1.0–1.5 mg l-1) and coconut milk (10% v/v) in MS medium were important factors for the induction of somatic embryogenesis. Somatic embryos germinated into plantlets in MS medium without growth regulators. The plants grew well in the greenhouse and flowered normally. The germination of Amaranthus paniculatus seeds was inhibited by applying paclobutrazol, a specific inhibitor of gibberellin biosynthesis. This inhibition was markedly counteracted by gibberellin A3 (GA3), suggesting that endogenous gibberellins are required for germination in this species. The inhibitory effect of paclobutrazol was also overcome by ethephon (2-chloroethylphosphonic acid) or the precursor of ethylene biosynthesis, ACC (1-aminocyclopropane-l-carboxylic acid). Thus the physiological effect of gibberellin can be mimicked by ethylene released from ethephon or synthesised from exogenous ACC.
