Ethanol (1 g/kg, orally) disturbed the hole reflex in male SHR and C57BL/6 mice and common albino rats increasing the time spent in the light part of a dark-light chamber. In mice this effect was often accompanied by an increase in number of transitions between dark and light compartments. Intraperitoneal pretreatment with endogenous metabolites of tryptophan in the kynurenine pathway of its metabolism (kynurenines)--kynurenic, picolinic and xanthurenic acids--attenuated the effect of ethanol in mice. Injection into the brain ventricles of the most active kynurenines from the group of excitatory amino acids, quinolinic acid and its precursor kynurenine, counteracted ethanol in mice and rats. The same was true in mice for intracerebroventricularly administered kynurenic, picolinic and xanthurenic acids.  相似文献

ABSTRACT. A Markov model of arthropod locomotor behaviour in choice chambers is presented. In the model, the compartments of the choice chamber and the movements of the animals from one compartment to another are treated as the states and the state transitions of a Markov chain, respectively. The model allows one to calculate the transition probabilities and the equilibrium distribution of animals in a choice chamber from direct measurements of displacement speeds and boundary turning reactions in each compartment. The compartment preferences (i.e. the proportion of time spent in each compartment) in two-compartment choice chambers were found to be strongly correlated with the predictions of the model. Klinokinesis is suggested to have negligible significance in some of the experiments reviewed. An equation is given to evaluate quantitatively the relative strength of kinetic and tactic components in the overall preference in the choice chamber; this assessment has not previously been possible.  相似文献

There is scarce information regarding the effects of anesthetic doses of the non-competitive N-methyl-d-aspartate receptor antagonist ketamine on anxiety. The current study evaluated the acute effects of intraperitoneally (i.p.) administered anesthetic ketamine (100 mg/kg) i.p. on anxiety in rats. For this purpose, the light/dark and the open field tests were utilized. The effects of anesthetic ketamine on motility were also examined using a motility cage. In the light/dark test, anesthetic ketamine, administered 24 h before testing reduced the number of transitions between the light and dark compartments and the time spent in the light compartment in the rats compared with their control cohorts. In addition, ketamine was found to exert a depressive effect on rats’ motility. In the open field test, animals treated with anesthetic ketamine 24 h before testing spent essentially no time in the central area of the apparatus, decreased horizontal ambulatory activity, and preserved to a certain extent their exploratory behaviour compared to their control counterparts. The results suggest that, in spite of its hypokinetic effect, a single anesthetic ketamine administration apparently induces an anxiety-like state, while largely preserving exploratory behaviour in the rat. These effects were time-dependent they since they were extinguished when testing was carried out 48 h after anesthetic ketamine administration.

The evidence obtained from the studies of the anxiolytic activity of calcium channel blockers in the tests for anxiety: dark-light chamber, elevated plus-maze, social interaction, incomplete sensory deprivation, and change in the excitability of the CNS is analyzed. Methodological aspects of studying the anxiolytic activity of drugs in these tests are discussed. The problems are considered related with simulation of the normal situational anxiety and pathological anxiety as well as the prospects of using the calcium channels blockers as anxiolytics. In view of these aspects, the problem of search for an "ideal" anxiolytic and possible results of revealing its activity in the tests for the normal and pathological anxiety are discussed.  相似文献

The present study examined the effects of diazepam (a widely used anxiolytic benzodiazepine) on the behavioural response of pigs to three novel experimental situations used to measure anxiety-related behaviour in rodents. Twelve weaned pigs (two pairs from each of the three litters) were tested in an elevated plus-maze at the age of 6 weeks, a light/dark test at the age of 7 weeks and an open-field test at the age of 8 weeks. Six of the pigs were pre-treated with diazepam (valium) and the other six with saline (control). In the elevated plus-maze, diazepam-treated pigs had a higher number of entries into open arms (P=0.04), spent more time on open arms (P=0.07), and had a higher number of total arm entries (P=0.05) than pigs from the control group. However, diazepam had no significant effects on behaviour in the light/dark test (i.e., latency to enter lit compartment, number of entries into lit compartment and the time spent in lit compartment) or the open-field test (i.e., number of lines crossed, number of entries into centre). In summary, the anxiolytic effects of diazepam on the pigs' behaviour were only demonstrated in the elevated plus-maze, where the time spent on open arms and the number of entries into open arms could be interpreted as measures of anxiety in pigs.  相似文献

The seeds of Griffonia simplicifolia Baill., a tropical shrub native to West Africa, are rich in 5-hydroxy-l-tryptophan (5-HTP), a direct precursor in the synthesis of serotonin (5-HT). In spite of the modern therapeutic application of Griffonia simplicifolia seed extract in mood disorders, no scientific evidence has been provided till now. For this reason the aim of our study was to investigate the effect of Griffonia simplicifolia seed extract on anxiety behavior. Griffonia simplicifolia seed extract, dosed at 1, 5, 10 and 25 mg/kg, was orally administered in rats which were submitted to the dark-light test and open field test, 60 min after the treatment. In the dark-light test, the administration of the extract at the doses of 10 and 25 mg/kg was able to significantly increase the time spent in the light compartment (P < 0.05). In the open field test, the extract dosed at 5, 10 and 25 mg/kg induced an anti-tigmotactic effect, as indicated by a significant increase of time spent in the central area of the open field (P < 0.01). In conclusion these findings indicate that Griffonia simplicifolia seed extract exerts anxiolytic-like effect in rats and suggest its potential usefulness for the treatment of anxiety in humans.  相似文献

Video tracking equipment was used to quantify the behavior of adult deathwatch beetles, Xestobium rufovillosum in a light/dark choice chamber. Effects of insect age, sex, and mating status on this behavior were analysed. The behavior recorded included the initial direction of movement, the time taken to reach the light or dark zone, the time spent in each zone, the time spent moving in each zone and the distance moved in each zone. The results show that both male and female insects 1–12 days old prefer the light zone. Insects more than 12 days old demonstrate sexually differential photoresponsive behavior. Males show a preference for the light. Females show an increasing preference for the dark, mated females more so than unmated females. The implications of these differences in behavior are discussed in relation to control programmes.  相似文献

Estrogen has been shown to affect nonreproductive behaviors in humans and rodents, including anxiety, fear, and activity levels. Rat studies have shown increases and decreases in these behaviors. Inconsistencies may be due to differences in testing conditions and the extent to which each test measures anxiety, fear, or activity. Few mouse studies have been performed. The present study was conducted to address these issues by examining the effect of estradiol benzoate (EB) in ovariectomized (OVX), C57BL/6 mice on a range of behavioral paradigms measuring anxiety [open field (OF), dark-light transition (DLT), elevated plus maze (EP)], activity [running wheel (RW)], and conditioned fear learning (FCon). In OF, vehicle (Veh) animals spent more time in the center than EB-treated animals and were more active overall. In DLT, Veh animals were more active than EB-treated animals in both the dark and light compartments and made more transitions between the two. In EP, Veh animals entered a greater number of arms. During FCon, EB animals froze more than Veh to the conditioned stimulus. In contrast, in the home cage RW, EB animals were more active than Veh. Factor analysis was used to characterize intertask correlations of females' behavior and to explore the possibility that estrogen may have an impact on a general arousal factor. In sum, estrogen treatment heightened fear responses in a range of fear and anxiety-provoking situations (OF, DLT, EP, and FCon), while increasing activity in the safer RW. We suggest that EB treatment may result in a generally more aroused animal.  相似文献

The cuboidal epithelial stem cell line Rat Mammary (Rama) 25 can differentiate in culture to droplet, alveolar-like cells that form domes, secrete small amounts of casein, and bind peanut lectin after treatment with neuraminidase. Differentiation to droplet cells is accelerated by dimethyl sulfoxide (DMSO). Morphologically intermediate states (gray and dark) which occur in the order: cuboidal----gray----dark----dark droplet----doming cells have been identified along this pathway by time-lapse cinematography. The dark and dark droplet states are associated with increased peanut lectin binding capacity whereas casein is secreted mainly by cells in domes. Cells in cultures containing low concentrations of DMSO (less than 56 mM) acquire droplets predominantly in the dark state, whereas with higher concentrations of DMSO droplet formation is seen mainly in the gray state. Kinetic analysis both from time-lapse films and conventional microscopy, shows that increasing the concentration of DMSO prolongs the time spent in the gray state, decreases the time of initial appearance of droplet cells, and increases their subsequent rate of formation, without detectable effects on the rates of the remaining morphological transitions. DMSO also reduces the average rate of DNA synthesis and increases the average cell cycle time, particularly in the second (and subsequent) cell cycles after its addition. However, neither droplet nor doming cells are terminally differentiated. Thus a linear sequence of morphological states exists between the Rama 25 stem cells and the alveolar-like or more probably alveolar bud cells in vitro, and DMSO accelerates the overall conversion predominantly by truncating one of the steps in this pathway.  相似文献

After dark-light transitions, there is a delay in photosynthetic CO2 fixation by isolated pea chloroplasts in the range of some minutes. In order to assess the physiological significance of light modulation of enzyme activity in the control of induction, we made estimates of the kinetic parameters of fructose-1,6-bisphosphatase immediately upon release from pea chloroplasts in the dark and after illumination for various time periods. The Michaelis constant for fructose-1,6-bisphosphate decreased and maximal velocities increased during induction. It seems likely that light activation of this enzyme is one of the factors contributing to the overcoming of the lag period in photosynthetic CO2 fixation.  相似文献

Several epidemiological studies have shown that consumption of large quantities of vegetables especially cruciferous vegetables (Broccoli and Brussels sprouts) can protect against chronic diseases. Sulforaphane, an isothiocynate found in cruciferous vegetables has been demonstrated to have neuroprotective effects in several experimental paradigms. This study was undertaken to examine the effect of sulforaphane on cognitive impairment in zebra fish model using a novel method of fear conditioning. Initially, the normal behaviour of zebra fishes was studied in light-dark tank for 10 min daily for 10 days. Fishes were then divided into seven groups of twelve in each. Group I served as normal, group II served as fear conditioned control, group III and group IV were sulforaphane (25 µM/L) and piracetam (200 mg/L) treated respectively. Group V served as scopolamine (400 µM/L) induced memory impairment fishes. Group VI and VII were sulforaphane (25 µM/L) and piracetam (200 mg/L) treated scopolamine induced memory impairment groups respectively. In normal behavioural analysis, fishes preferred to stay in dark compartment. The average number of entries into the dark and time spent in dark were significantly more. Fishes in group II to VII were individually subjected to fear conditioning passive avoidance task and evaluated for learned task memory. It was observed that the average number of entries into dark and time spent in dark were significantly decreased. After exposure to respective treatment fishes in group III to VII were subjected to cognitive evaluation. There was no significant difference in cognition of group III and IV fishes exposed to sulforaphane and piracetam alone respectively. Fishes exposed to scopolamine showed a significant cognitive impairment. Sulforaphane exposure prior to scopolamine significantly retained the memory of learned task. These findings suggest that sulforaphane might be a promising therapeutic agent for cognitive enhancement in Alzheimer’s disease.  相似文献

The relationship among relative size, growth rate and diel visits into a feeding compartment were studied in Arctic charr Salvelinus alpinus , a species known for its flexible activity patterns. Individual swimming activity from a refuge compartment into an uncovered feeding compartment was studied using an automated passive integrated transponder system. Approximately half of the individuals in four groups of S. alpinus spent 70–80% of their time in the feeding compartment, regardless of the time of day. The remaining individuals spent nearly as much time (70–80%) in the feeding compartment during the night, but only spent c. 20% of their time in the feeding compartment during the day. These individuals had a lower mean ± s . d . masses (13·17 ± 4·34 g) and growth rate (daily growth coefficient, G = 0·80 ± 1·19) than those individuals that spent most of their time in the feeding compartment during the day (mass = 16·65 ± 5·73 g and G = 2·04 ± 0·81). This indicated that some smaller fish were absent from the feeding compartment during daytime, possibly to avoid aggression from larger conspecifics. In the open compartment, the aggressive interactions were more frequent in daylight than during night and always low in the covered compartment.  相似文献

Unicellular, diazotrophic cyanobacteria temporally separate dinitrogen (N2) fixation and photosynthesis to prevent inactivation of the nitrogenase by oxygen. This temporal segregation is regulated by a circadian clock with oscillating activities of N2 fixation in the dark and photosynthesis in the light. On the population level, this separation is not always complete, since the two processes can overlap during transitions from dark to light. How do single cells avoid inactivation of nitrogenase during these periods? One possibility is that phenotypic heterogeneity in populations leads to segregation of the two processes. Here, we measured N2 fixation and photosynthesis of individual cells using nanometer-scale secondary ion mass spectrometry (nanoSIMS) to assess both processes in a culture of the unicellular, diazotrophic cyanobacterium Crocosphaera watsonii during a dark-light and a continuous light phase. We compared single-cell rates with bulk rates and gene expression profiles. During the regular dark and light phases, C. watsonii exhibited the temporal segregation of N2 fixation and photosynthesis commonly observed. However, N2 fixation and photosynthesis were concurrently measurable at the population level during the subjective dark phase in which cells were kept in the light rather than returned to the expected dark phase. At the single-cell level, though, cells discriminated against either one of the two processes. Cells that showed high levels of photosynthesis had low nitrogen fixing activities, and vice versa. These results suggest that, under ambiguous environmental signals, single cells discriminate against either photosynthesis or nitrogen fixation, and thereby might reduce costs associated with running incompatible processes in the same cell.  相似文献

In two experiments, hungry rats, Rattus norvegicus, were present in one side of an operant chamber while a conspecific demonstrator in the adjacent compartment moved a single lever either up or down for a food reward. During a subsequent test session, in which these rats were allowed access to the lever for the first time, all responses were rewarded regardless of their direction. In experiment 1, rats that were prevented from observing the direction of lever movement by means of a screen showed a reliable demonstrator-consistent response bias, while rats that had observed the direction of lever movement and in addition had access to any odour cues deposited on the lever did not. In experiment 2, each rat observed another rat (the ‘viewed’ demonstrator) moving a lever either up or down. They were then transferred into the test compartment of a different operant chamber in which another rat (the ‘box’ demonstrator) had moved the lever in the same direction as the viewed demonstrator or in the opposite direction. These observer rats showed a reliable preference for their box demonstrator's direction, but responded in the opposite direction to their viewed demonstrator. Taken together, the results of these experiments suggest that directional responding by rats in a vertical movement two-action test is influenced by demonstrator-deposited odour cues in addition to visual experience of a demonstrator's behaviour. Furthermore, while odour-mediated local enhancement gave rise to demonstrator-consistent responding, visual observation of a conspecific appeared to have the reverse effect. 采用split-litter法对仔鼠进行分组和处理,共5组NTS组(未经实验人员抓握和标记),PND2—9TS组和PND10—17TS组(分别在仔鼠出生后的2—9天、10—17天,每天短暂抓握和标记仔鼠),PND2—9MS组和PND10—17MS组(分别在仔鼠出生后的2—9天、10—17天,除了按TS组相同方式抓握并在不同部位标记外,每天把仔鼠与母鼠分离1h)。待雌鼠成年后,进行明/暗箱测试和一次性被动回避反应测试。结果发现与NTS组相比,PND2—9TS组和PND10—17TS组的雌鼠在明/暗箱测试中停留于明室的累计时间明显较长,在被动回避作业中的重测试潜伏期也明显较长,表明新生期的触觉刺激经历减少雌性大鼠成年后在新异环境中的焦虑,并改善情绪记忆。与相应TS组相比,MS处理组的所有行为指标都无显著性差异,说明短时间母婴分离对雌鼠成年后的焦虑和情绪记忆无明显影响。结果提示,新生期的触觉刺激和母婴分离经历对仔鼠神经系统的发育产生不同的长期效应。  相似文献

Female CD-1 mice showed reduced light avoidance at estrus compared to nonestrus in two experiments designed to control for possible effects of changes in general activity. Estrous mice were quicker to enter a light chamber from a dark chamber and slower to enter a dark chamber from a light chamber, than nonestrous mice. Also, estrous mice reared up on their hind legs more frequently than nonestrous mice in the light chamber, and there was no difference between estrous and nonestrous mice in motor activity in the dark chamber. These findings, along with previous findings of reduced shock avoidance at estrus, suggest a general decrement in avoidance motivation which may be adaptive to the process of mating and which does not appear to be secondary to a change in activity level.  相似文献

The rate of calcification in the scleractinian coral Galaxea fascicularis was followed during the daytime using 45Ca tracer. The coral began the day with a low calcification rate, which increased over time to a maximum in the afternoon. Since the experiments were carried out under a fixed light intensity, these results suggest that an intrinsic rhythm exists in the coral such that the calcification rate is regulated during the daytime. When corals were incubated for an extended period in the dark, the calcification rate was constant for the first 4 h of incubation and then declined, until after one day of dark incubation, calcification ceased, possibly as a result of the depletion of coral energy reserves. The addition of glucose and Artemia reduced the dark calcification rate for the short duration of the experiment, indicating an expenditure of oxygen in respiration. Artificial hypoxia reduced the rate of dark calcification to about 25% compared to aerated coral samples. It is suggested that G. fascicularis obtains its oxygen needs from the surrounding seawater during the nighttime, whereas during the day time the coral exports oxygen to the seawater.  相似文献

To simulate production-scale conditions of gluconic acid fermentation by Gluconobacter oxydans, different experimental setups are presented in this study. From the determination of the time constants of a production-scale reactor, it can be concluded that mixing and oxygen transfer are the rate-limiting mechanisms. This results in oxygen concentration gradients which were simulated in a one-compartment reactor in which the oxygen concentration was fluctuated by a fluctuated gassing with air and nitrogen. It could be concluded that only very long periods of absence of oxygen (ca. 180 s) results in lower specific oxygen uptake rates by Gluconobacter oxydans. From scale-down studies carried out in a two-compartment system to simulate a production-scale reactor more accurately, it could be concluded that not only the residence time in the aerated part of the system is important, but the liquid flow in between the different parts of the reactor is also an essential parameter. It could also be concluded that the microorganisms are not influenced negatively by the fluctuated oxygen concentrations with respect to their maximal oxidation capacity. The two-compartment system can also be used for optimization experiments in which the "aerated" compartment was gassed with pure oxygen. From these experiments it was concluded that also a short residence of the cells at high oxygen concentrations diminished the growth and product formation rates. These experiments show the necessity of the scale-down experiments if optimization is carried out. The two-compartment system presented in this study is a very attractive tool for reliable scale-down experiments.  相似文献

Simulated time zone transitions were performed in an isolation unit upon groups of one to four human subjects. In the first series of experiments, the adjustment of the circadian rhythm of body temperature, measured in the presence of sleep and other masking factors, was assessed by cosinor analysis and by cross-correlation methods. These methods modeled the circadian timing system either as a single component or as the sum of two components, those due to exogenous and endogenous influences. The one-component models described a more rapid adjustment of the temperature rhythm to the time zone transition than did the two-component models; we attribute this difference to the masking effects of the exogenous component. In a second series of experiments, we showed that the shift of the endogenous component, as assessed by the two-component models, was not significantly different from that measured during constant routines. The results also showed that, if the zeitgebers were phased in advance of the endogenous component, then advances of the endogenous component were produced only if this mismatch was less than about 10 hr. Mismatches greater than this, and cases where the zeitgebers were delayed with respect to the endogenous component, both produced delays of the endogenous component. We conclude that the two-component cross-correlation methods can be used to estimate shifts of the endogenous component of a circadian rhythm in the presence of masking factors. They are therefore an alternative to constant routines when these latter are impracticable to carry out.  相似文献

Dynamic equilibrium in a biological system implies that the compartment under study does not change in size during the period of observation. In many biological systems there are, however, net changes with time and this report deals with the mathematical treatment necessary to calculate unequal rates of inflow and outflow. A method is presented for the calculation of transfer rates in a two compartment system when the rates of flow between these compartments are unequal but constant. Equations were developed to calculate the amount of material transported per unit time derived from measurements of specific activity and compartment size. The problems of (1) sampling from the pool and (2) the effects of analytical errors on the estimation of rate have been evaluated. An example has been presented in which the derived equations have been applied to a study of the simultaneous passage of sodium into and out of a permanently isolated loop of bowel.  相似文献
