Osmotic stress due to high sucrose concentration was imposed on the potato aphid, Macrosiphum euphorbiae (Thomas), in an artificial diet situation. Thin layer chromatography resolved melezitose and six additional honeydew oligosaccharides. Significantly higher levels of these oligosaccharides were recovered from the honeydew of this species in response to feeding on increased dietary sucrose concentrations, pointing to an osmoregulatory response. In addition, carbohydrase activities from the potato aphid were described. A degree of specificity toward hydrolysis of the sucrose substrate over other α-glucosyl sugars was demonstrated. Also, sucrose was optimal for the formation of the oligosaccharides. A strong α-galactosidase activity was found and transgalactosylation ability was indicated. These major trends in carbohydrase activity were also found in the foxglove aphid, Acyrthosiphon solani (Kaltenbach), and the oleander aphid, Aphis nerii Fonscolombe.  相似文献

Jasmonates such as jasmonic acid (JA) are plant‐signaling compounds that trigger induced resistance (IR) to a broad range of arthropod herbivores. JA‐dependent defenses are known to reduce the growth and survivorship of many chewing insects, but their impact on piercing–sucking insects such as aphids has not been extensively investigated. In this study, induced resistance was activated in tomato (Lycopersicon esculentum Mill) (Solanaceae) using a foliar application of synthetic JA, and control plants were treated with carrier solution. The life parameters of individual potato aphids and their progeny (Macrosiphum euphorbiae Thomas) (Hemiptera: Aphididae) were evaluated on the unsprayed leaves of plants in order to access the systemic effects of the foliar treatments. IR significantly reduced the longevity and net reproduction of adult aphids, as well as the percentage of juveniles to survive to maturity. These results indicate that JA application induces systemic defenses in tomato that have a direct negative impact on aphid survivorship. This study also examined aphid honeydew excretion, in order to evaluate the potential influence of induced resistance on aphid feeding behavior. The average honeydew production per aphid was comparable on plants with or without JA treatment, indicating that JA‐dependent defenses did not deter feeding. This suggests that the observed effects of JA on aphid survivorship were due to antibiotic rather than antixenotic factors. In addition to studying the effects of JA treatment on a tomato cultivar that is susceptible to aphids, this study also examined the effects of exogenous application of JA on tomato plants that carry the aphid resistance gene, Mi‐1.2. JA application did not significantly enhance or inhibit aphid control on resistant tomato. These findings expand our understanding of the effects of JA‐dependent defenses on piercing–sucking insects, and of the potential interactions between induced resistance and R‐gene mediated aphid resistance in tomato.  相似文献

Potato aphid, Macrosiphum euphorbiae (Homoptera: Aphididae), is an abundant potato pest and vector of potato leaf-roll virus and potato virus Y in Maine and other potato growing areas. We investigated the circadian rhythmicity of its movement towards host plant odor. Effects of daily cycle (day or night) and illumination (light or dark) on the proportion of aphids colonizing potato leaflets were determined in a Petri plate arena and in a Y-tube olfactometer. In Petri dishes, both daily cycle and light had a highly significant effect on plant colonization. Increasing temperature reduced aphid colonization of the leaflets. In the olfactometer, light had a significant effect on the proportion of aphids walking towards the host plant. Interaction between time and light was also statistically significant, with the effect of illumination being smaller during the day than during the night. Our results suggest that circadian rhythm in host-finding behavior of the potato aphid is regulated by both exogenous and endogenous mechanisms.  相似文献

Fourteen microsatellite loci were isolated and characterized in the potato aphid, Macrosiphum euphorbiae Thomas, by screening a genomic library with the oligonucleotide probes (GA)10 (GT)10 and (GATA)4. Allelic diversity was estimated in samples collected from potato fields in Tunisia. Ten loci displayed polymorphism that ranged from two to four alleles per locus and the observed heterozygosity ranged from zero to one. These markers could be used to study the population genetic structure of this polyphagous aphid species.  相似文献

The potato aphid Macrosiphum euphorbiae (Thomas) is a major pest of several economic crops in Tunisia. Using 10 microsatellites, we analyzed five populations of M. euphorbiae sampled during the 2004-2005 solanaceous and cucurbit season (April through September) from five geographic origins. From 235 aphids, 61 different multilocus genotypes were identified of which three genotypes MLG1, MLG2, and MLG42 were predominant on all host plants and regions. MLG1 and MLG2 genotypes were detected in 2004 and did not reappear in spring 2005, while the genotype MLG42 was detected only in 2005. All populations showed significant deviation from Hardy-Weinberg equilibrium even in data sets including one individual per genotype. χ(2) independence tests and analysis of molecular variance showed no significant differentiation among populations collected on different host plant and from different geographic origin, but differences between populations from two successive years were significant. Factorial component analysis corroborates these results. The probable causes of this seasonal variation were discussed.  相似文献

Plant induced responses are activated by multiple biotic and abiotic stresses, and may affect the interactions between a plant and phytophagous insects. The objective of this work was to evaluate the effects of different stresses inflicted to potato plants (Solanum tuberosum) on the potato aphid (Macrosiphum euphorbiae). Abiotic wounding, biotic wounding by Leptinotarsa decemlineata and treatment with volatile methyl jasmonate (MeJA) were evaluated with regard to the orientation behaviour, the feeding behaviour and the development of the potato aphids. Dual‐choice olfactometry showed that plants treated with MeJA lost their attractiveness for the potato aphids, while both abiotic and biotic wounding did not alter the orientation of aphids. Electropenetrography revealed that the feeding behaviour of aphids was only slightly disturbed by a previous L. decemlineata wounding, while it was highly disturbed by mechanical wounding and MeJA treatment. Aphid nymph survival was reduced on mechanically wounded plants, the pre‐reproductive period was lengthened and the fecundity reduced on plants treated with MeJA. Our results bring new information about the effects of various stresses inflicted to S. tuberosum on M. euphorbiae. We showed that wounding and MeJA treatment induced an antixenosis resistance in potato plants against M. euphorbiae, which may influence aphid colonization processes.  相似文献

Transgenic plants expressing protease inhibitors (PIs) have emerged in recent years as an alternative strategy for pest control. Beneficial insects such as parasitoids may therefore be exposed to these entomotoxins either via the host or by direct exposure to the plant itself. With the objective of assessing the effects of PIs towards aphid parasitoids, bioassays using soybean Bowman-Birk inhibitor (SbBBI) or oryzacystatin I (OCI) on artificial diet were performed on Macrosiphum euphorbiae-Aphelinus abdominalis system. OCI significantly reduced nymphal survival of the potato aphid M. euphorbiae and prevented aphids from reproducing. This negative effect was much more pronounced than with other aphid species. On the contrary, SbBBI did not affect nymphal viability but significantly altered adult demographic parameters. Enzymatic inhibition assays showed that digestive proteolytic activity of larvae and adults of Aphelinus abdominalis predominantly relies on serine proteases and especially on chymotrypsin-like activity. Immunoassays suggested that OCI bound to aphid proteins and accumulated in aphid tissues, whereas SbBBI remained unbound in the gut. Bioassays using M. euphorbiae reared on artificial diets supplemented with both OCI and SbBBI showed a fitness impairment of Aphelinus abdominalis that developed on intoxicated aphids. However, only SbBBI was detected in parasitoid larvae, while no PI could be detected in adult parasitoids that emerged from PI-intoxicated aphids. The potential impact of PI-expressing plants on aphid parasitoids and their combined efficiency for aphid control are discussed.  相似文献

The effect of the tomato resistance gene, Meu1, on feeding, longevity, fecundity and developmental rate of the pink biotype of the potato aphid, Macrosiphum euphorbiae (Thomas) (Hemiptera, Aphididae), was determined using nearly isogenic tomato (Lycopersicon esculentum Mill, Solanaceae) lines. Aphid mortality was significantly higher on resistant plants, with 60% of the aphids dying by the 4th day of exposure. By the 10th day, all the aphids on the resistant plants were dead whereas 100% of the aphids on susceptible plants were alive. Meu1-mediated resistance resulted in significantly decreased fecundity with a ten-fold decrease in the net fertility rate (4.5 and 45.7 progeny per aphid on resistant and susceptible tomato, respectively). A qualitative analysis showed that honeydew was produced by aphids on resistant and susceptible plants, suggesting that aphids initiate feeding on both plant types. However, significantly lower quantities of honeydew were present when aphids were caged on resistant plants. There were also significant differences in aphid location on resistant and susceptible leaves. Experiments evaluating behavior in less than 24 h showed that aphids left resistant leaves after relatively short exposure (3–6 h). Aphids transferred from resistant to susceptible tomato at intervals between 3 h and 24 h resumed feeding as evidenced by presence of honeydew. Although the mechanism by which Meu1-mediated resistance operates is not yet known, our data suggest that resistance factors act rapidly after initiation of feeding and that lower fecundity and longevity are related to reduction in aphid feeding.  相似文献

The number of plants that sampling requires in aphid population studies often exceeds one hundred. Thus, only quick and nondestructive methods can be used to sample this pest in a reasonable time interval. We propose a visual method for estimating the density of the aphid Macrosiphum euphorbiae (Thomas) on tomato plants reared in greenhouses. After approximately 1min of visual observation plants can be assigned to abundance classes, the boundaries of which are roughly the powers of square root 10. Precise counts were collected simultaneously on sets of reference plants from the same greenhouse. Projection pursuit nonparametric regression was then used to provide unbiased estimates of aphid densities from the abundance classes and several easily gathered explanatory variables. The robustness of the method was evaluated by testing the models on the complementary data sets from plants in which the aphid densities were precisely counted. In both single and twin-row cultural conditions, for the reference and complementary data sets, the order of magnitude of the error was less than one class rank per plant. The investigation time was reduced by approximately 10-fold compared with the exact counting method. This easy-to-teach field method could be useful in large-scale population surveys and for optimizing integrated pest management strategies.  相似文献

Since the discovery of the soybean aphid, Aphis glycines Matsumura, in midwestern U.S. soybean, Glycine max L., in 2000, the aphid has become a significant economic pest. Basic information about estimating population density within fields is unknown. Therefore, we developed two sampling plans to efficiently characterize A. glycines densities. Enumerative and binomial sequential plans were developed using 89 data sets collected from 10 commercial fields sampled during 2001-2003. Re-sampling software was used to validate the enumerative plan on whole plant counts, based on Taylor's power law parameters (a = 9.157 and b = 1.543). For research applications, the enumerative plan was modified to provide an actual precision level of 0.10 (SE/mean), which resulted in an average sample number of 310 individual plants. For integrated pest management (IPM) purposes, we developed an enumerative plan with an actual precision of 0.25, which resulted in an average sample number of 38 individual plants. For IPM applications, the binomial plan will likely be more practical. Binomial plans were developed using two tally thresholds at five action thresholds. Final analysis of the operating characteristic curve for each plan indicated that the tally threshold of > or = 40 aphids per plant, and an action threshold of 0.837 (84% of the plants infested) provided the most correct treat (4%) and no-treat (95%) decisions, with very low incorrect treat (0.5%) and no-treat (0.5%) decisions. A tally threshold of > or = 40 aphids per plant and action thresholds of 84% of plants infested is equivalent to a mean density of 250 aphids per plant, a recently recommended economic threshold. Using this threshold, the minimum required sample number for the binomial plan was 11 plants.  相似文献

A non-gall-forming aphid, Macrosiphum euphorbiae, was reared within the galls of Dysaphis devecta and Aphis pomi which had been induced in apple seedlings. Similar aphids were also reared on ungalled seedling leaves of a similar age and also on mature leaves and in old galls.There is a weak positive linear relationship between lipid content and pre-reproductive adult dry weight, but aphids living on young galled or ungalled plant tissue have 30% lipid compared to 11% for aphids living on old plant tissue. Adult aphids gradually decline in weight, losing between 24% and 38% of their pre-reproductive biomass before death. The energy content of this loss subsidieses reproduction. Reproduction of aphids feeding in old galls or on mature leaves was negligible, but on younger tissues average fecundity was 31, there being little difference between aphids living on galled and ungalled tissue. There is no simple relationship between pre-reproductive adult embryo content and fecundity.The principal factor mediating honeydew production was aphid size and this factor overrides any variation caused by different feeding sites. Honeydew production averaged 0.43 mg during a life of 30 days. Aphids successfully completing their life cycle have a life-time energy consumption of 30.58 joules of which 18% is lost as heat during respiration (R), 30% is contained in honeydew (F+U), 1% is contained in exuviae (Pe). Larval growth accounts for 17% (Pg), and adult reproduction for 34% (Pr).The P/C ratio for M. euphorbiae is 53% and is broadly comparable with aphids living on herbaceous plants.

D'autres M. euphorbiae ont été élevés sur feuilles sans galles du même âge provenent de semis identiques, sur feuilles mûres et sur vieilles galles.Il y a une faible relation positive linéaire entre la teneur en lipides et le poids sec des adultes avant la reproduction, mais les pucerons élevés sur tissus végétaux jeunes avec ou sans galles contiennent 30% de lipides contre 11% pour ceux élevés sur tissus âgés. Le poids des pucerons adultes diminue progressivement, la perte avant la mort se situant entre 24 et 38% de la biomasse avant la reproduction. Le contenu énergétique de ces pertes alimente la reproduction. La production moyenne de miellat est de 0,43-mg pendant une vie de 30 jours. Les pucerons accomplissant leur cycle avec succès utilisent pendant leur vie 30,58 joules, dont 18% sont dissipés sous forme de chaleur pendant la respiration (R); le contenu du miellat (F+U) correspond à 30%, et 1% est contenu dans l'exuvie (Pe). La croissance larvaire (Pg) utilise 17%, et l'activité reproductrice des adultes (Pr) 34% de l'énergie. Le rapport P/C-53% pour M. euphorbiae-est globalement comparable à celui des pucerons vivant sur plantes herbacées.

Aphids increase production of winged individuals as a generalized response to multiple threats, including predators, competitors, and poor host plant quality. While wing formation in response to these individual threats is well documented, few investigations have evaluated whether combined threats lead to additive or non-additive outcomes. We tested the interactive effects of predation risk and plant quality on population growth and wing induction in the potato aphid, Macrosiphum euphorbiae. Plant quality was varied using phytohormonal manipulations of tomato (Solanum lycopersicum) to elevate or suppress the jasmonate and salicylate defense pathways. Predation risk was altered by exposing aphids to lethal or risk (unable to feed) individuals of the convergent lady beetle, Hippodamia convergens. Phytohormonal treatments resulted in >4-fold variation in aphid population growth and thus strongly affected plant quality; however, the percentage of winged individuals was no different across plant types. Predators similarly reduced aphid abundance, but also elicited a ~3-fold increase in wing formation, an effect that was similar in magnitude when comparing lethal with risk predators. The overall impact of plants and predators on aphids was largely additive, an outcome that was unexpected given the likelihood for interactions between these two factors and our prior results with other herbivores in this system. We discuss this discrepancy in the context of phenotypic plasticity, non-lethal predator effects, and the ecological challenges faced by wing dimorphic insects.  相似文献

The banana aphid, Pentalonia nigronervosa Coquerel (Hemiptera: Aphididae), infests banana (Musa spp.) worldwide. Pentalonia nigronervosa is the vector of Banana bunchy top virus (family Nanoviridae, genus Babuvirus) the etiological agent of Banana bunchy top disease (BBTD). BBTD is currently the most serious problem affecting banana in Hawaii. Despite the importance of this vector species, little is known about its biology or ecology. There are also no sampling plans available for P. nigronervosa. We conducted field surveys to develop a sampling plan for this pest. Ten plots were surveyed on seven commercial banana farms on the island of Oahu, HI, for the presence of P. nigronervosa on banana plantlets. We found aphids more frequently near the base of plants, followed by the newest unfurled leaf at the top of the plant. Aphids were least likely to be located on leaves in between the top and bottom of the plant. Aphid infestation on surveyed plots ranged from 8 to 95%. We developed a sequential binomial sampling plan based on our surveys. We also discovered that the within-plant distribution of P. nigronervosa is an important factor to consider when sampling for this pest. Our sampling plan will assist in the development of sustainable management practices for banana production.  相似文献

Resistance level of seven accessions of wild Solanum species (Solanaceae) to Myzus persicae and Macrosiphum euphorbiae (Homoptera: Aphididae) was evaluated by measuring survival and fecundity during sleeve cage experiments and population growth on whole plants in a controlled environment. The survival was lowest on the Solanum circaeifolium spp. capsicibaccatum, Solanum pinnatisectum and Solanum trifidum accessions for M. persicae and on the S. circaeifolium spp. capsicibaccatum, Solanum okadae and S. trifidum, accessions with M. euphorbiae. Plant species significantly influenced the fecundity of both aphid species. Aphid population growth on whole plants was negatively affected by the age of the plant, but generally followed the levels of net reproductive rate on different plant species observed during the sleeve cage experiment. The population of M. persicae varied among the seven wild potato accessions with the lowest growth rates on S. circaeifolium spp. capsicibaccatum, Solanum polyadenium, Solanum tarijense and S. trifidum. The number of M. euphorbiae also varied among accessions but the smallest population was collected from S. polyadenium. The results can be used to identify sources of resistance to aphids within those accessions already known to be resistant to the Colorado potato beetle. This study highlights the difficulties involved in developing a high-throughput screening test for aphid resistance compatible with a potato breeding programme.  相似文献

Counts of green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae), in potato, Solanum tuberosum L., fields were used to evaluate the performance of the sampling plan from a pest management company. The counts were further used to develop a binomial sampling method, and both full count and binomial plans were evaluated using operating characteristic curves. Taylor's power law provided a good fit of the data (r2 = 0.95), with the relationship between the variance (s2) and mean (m) as ln(s2) = 1.81(+/- 0.02) + 1.55(+/- 0.01) ln(m). A binomial sampling method was developed using the empirical model ln(m) = c + dln(-ln(1 - P(T))), to which the data fit well for tally numbers (T) of 0, 1, 3, 5, 7, and 10. Although T = 3 was considered the most reasonable given its operating characteristics and presumed ease of classification above or below critical densities (i.e., action thresholds) of one and 10 M. persicae per leaf, the full count method is shown to be superior. The mean number of sample sites per field visit by the pest management company was 42 +/- 19, with more than one-half (54%) of the field visits involving sampling 31-50 sample sites, which was acceptable in the context of operating characteristic curves for a critical density of 10 M. persicae per leaf. Based on operating characteristics, actual sample sizes used by the pest management company can be reduced by at least 50%, on average, for a critical density of 10 M. persicae per leaf. For a critical density of one M. persicae per leaf used to avert the spread of potato leaf roll virus, sample sizes from 50 to 100 were considered more suitable.  相似文献

大田和实验室研究均表明矿物油的施用可有效降低蚜虫对非持久性病毒的传播。我们分别于矿物油喷施马铃薯植株24 h和7 d后,调查其对马铃薯长管蚜Macrosiphum euphorbiae行为的影响;采用暗室生测法调查了蚜虫对处理的植物挥发性化合物的反应;采用刺探电位技术(EPG)评价了矿物油处理对蚜虫取食行为的影响。结果表明:矿物油处理导致植物对寄主的引诱作用失效,这种效果至少持续24 h。矿物油处理马铃薯植株24 h和7 d后,蚜虫对处理植株的取食行为发生改变。开始记录至第1次刺探所需时间降低,但只在处理7 d后显著降低,提示矿物油处理有助于蚜虫口针的穿刺。然而,流涎阶段以及韧皮部汁液吸食阶段(摄食阶段)显著缩短。本研究中观察到的蚜虫行为的改变不能充分解释施用矿物油的大田中为什么出现7 d 的保护作用,因此可能还存在其他的机制。  相似文献

A study was made of the resistance of three wild potato species, Solanum chacoense, S. stoloniferum and S. demissum, to three aphid species, namely Myzus persicae (Sulz.), Aulacorthum solani (Kalt.) and Macrosiphum euphorbiae (Thomas), which all commonly infest the cultivated potato, S. tuberosum. The resistance of each Solanum species differed with the species of aphid, with the part of the plant and with the physiological condition of the leaf. As a result, it may be difficult to utilize such resistance, since it is ineffective against some of the pest aphid species and may break down as the physiological condition of the plant changes.  相似文献

Interspecific competition between parasitoid larvae may influence the size, structure, and stability of the population, leading to a reduction in total parasitism and thus restricting the pest control. Aphidius ervi (Haliday) and Praon volucre (Haliday) are endoparasitoids that possess a wide host range and present considerable potential for the biological control of the aphid Macrosiphum euphorbiae (Thomas). The larval competition between A. ervi and P. volucre, and the possible intrinsic competitive superiority of one of the parasitoids in M. euphorbiae, have been studied. In single parasitism experiments, mated parasitoid females (n=10) were maintained individually in contact with M. euphorbiae hosts (n=30) inside petri dishes containing lettuce leaf discs and maintained in environmental chamber at 22 ± 1°C, 70 ± 10% RH, and 12-h photophase. The multiple parasitism experiments consisted of exposing single parasitized aphids (n=120) to the second parasitoid species. Two oviposition events were performed with a 4-h interval between them, namely the following: sequence A (oviposition by A. ervi, followed by P. volucre) and sequence B (oviposition by P. volucre, followed by A. ervi). Oviposition sequence A generated 24 A. ervi and 55 P. volucre adults, whereas oviposition sequence B generated 23 and 49 adults. P. volucre is an intrinsically superior competitor compared with A. ervi, and the use of the two species simultaneously may result in competitive exclusion and influence the stability of the parasitoid population.  相似文献

A survey of the parasitoids ofIllinoia liriodendri (Monell) in northern California conducted from 1988–1990 revealed the presence of 12 primary and 14 hyper-parasitoid species. The most common primary parasitoid wasAphidius polygonaphis (Fitch), which was imported from the eastern United States in the 1970's and is now established throughout the area. Striacosta albicosta (Smith) (Lepidoptera: Noctuidae), is a native pest of dry beans (Phaseolus vulgaris L.) and corn (Zea mays L.). As a result of larval feeding damage on corn ears, S. albicosta has a narrow treatment window; thus, early detection of the pest in the field is essential, and egg mass sampling has become a popular monitoring tool. Three action thresholds for field and sweet corn currently are used by crop consultants, including 4% of plants infested with egg masses on sweet corn in the silking-tasseling stage, 8% of plants infested with egg masses on field corn with approximately 95% tasseled, and 20% of plants infested with egg masses on field corn during mid-milk-stage corn. The current monitoring recommendation is to sample 20 plants at each of five locations per field (100 plants total). In an effort to develop a more cost-effective sampling plan for S. albicosta egg masses, several alternative binomial sampling plans were developed using Wald's sequential probability ratio test, and validated using Resampling for Validation of Sampling Plans (RVSP) software. The benefit-cost ratio also was calculated and used to determine the final selection of sampling plans. Based on final sampling plans selected for each action threshold, the average sample number required to reach a treat or no-treat decision ranged from 38 to 41 plants per field. This represents a significant savings in sampling cost over the current recommendation of 100 plants.  相似文献
