Tetracycline-resistant (Tet(r)) bacteria were isolated from fishes collected at three different fish farms in the southern part of Japan in August and September 2000. Of the 66 Tet(r) gram-negative strains, 29 were identified as carrying tetB only. Four carried tetY, and another four carried tetD. Three strains carried tetC, two strains carried tetB and tetY, and one strain carried tetC and tetG. Sequence analyses indicated the identity in Tet(r) genes between the fish farm bacteria and clinical bacteria: 99.3 to 99.9% for tetB, 98.2 to 100% for tetC, 99.7 to 100% for tetD, 92.0 to 96.2% for tetG, and 97.1 to 100% for tetY. Eleven of the Tet(r) strains transferred Tet(r) genes by conjugation to Escherichia coli HB-101. All transconjugants were resistant to tetracycline, oxycycline, doxycycline, and minocycline. The donors included strains of Photobacterium, Vibrio, Pseudomonas, Alteromonas, Citrobacter, and Salmonella spp., and they transferred tetB, tetY, or tetD to the recipients. Because NaCl enhanced their growth, these Tet(r) strains, except for the Pseudomonas, Citrobacter, and Salmonella strains, were recognized as marine bacteria. Our results suggest that tet genes from fish farm bacteria have the same origins as those from clinical strains.  相似文献

AIMS: To investigate the biofilm-forming capacity and the production of quorum signals in Gram-negative bacteria isolated from a food production environment, and the possible correlation between both phenotypes. METHODS AND RESULTS: Sixty-eight Gram-negative bacteria were isolated from equipment and working surfaces in a raw vegetable processing line, and tested for biofilm-forming capacity using an in vitro microplate assay. All isolates showed significantly higher biofilm-forming capacity than Escherichia coli laboratory strain DH5alpha, which was included as a negative control, and differed up to 56-fold in relative biofilm-forming capacity. Various assays based on reporter bacteria were used to detect quorum signals produced by the isolates. Twenty-six isolates produced autoinducer-2, five isolates produced N-acyl-homoserine lactones (AHLs), and none produced the Pseudomonas quinolone signal. CONCLUSIONS: No correlation was found between in vitro biofilm-forming capacity and production of quorum signalling molecules among the 68 strains isolated from the raw vegetable processing line. SIGNIFICANCE AND IMPACT OF THE STUDY: Several recent studies have shown a role of AHL-based quorum sensing in biofilm formation of specific Gram-negative bacterial strains. The current work shows that production of AHL and other quorum signals is not widespread in Gram-negative isolates from a raw vegetable processing line, and is not a general requirement for biofilm formation, at least in vitro.  相似文献

The distribution of a recently described marine bacterium, SBT 033 GenBank Accession No. AY723742), Pseudoalteromonas ruthenica, at the seawater intake point, outfall and mixing point of an atomic power plant is described, and its ability to form biofilm was investigated. The effectiveness of the antifouling biocide chlorine in the inactivation of planktonic as well as biofilm cells of P. ruthenica was studied in the laboratory. The results show that the planktonic cells were more readily inactivated than the cells enclosed in a biofilm matrix. Viable counting showed that P. ruthenica cells in biofilms were up to 10 times more resistant to chlorine than those in liquid suspension. Using confocal laser scanning microscopy it was shown that significant detachment of P. ruthenica biofilm developed on a glass substratum could be accomplished by treatment with a dose of 1 mg l-1 chlorine. Chlorine-induced detachment led to a significant reduction in biofilm thickness (up to 69%) and substratum coverage (up to 61%), after 5-min contact time. The results show that P. ruthenica has a remarkable ability to form biofilms but chlorine, a common biocide, can be used to effectively kill and detach these biofilms.  相似文献

The main purpose of this study was to isolate and briefly characterize proteolytic bacteria from a poorly known habitat – raw wool. Fleece samples were accordingly collected from Merino raw wool – a Portuguese ewe breed, at three distinct areas of their body, from animals exhibiting no symptoms or signs of abnormalities; they were then subjected to enumeration and isolation of a total of 158 bacterial strains. Said isolates were screened for protease activity, using the spot technique, on Calcium Caseinate Agar containing 1% (w/v) skim milk. The 36 isolates displaying the highest protease activity underwent a more refined assessment of enzymatic performance – by examining their cell-free supernatant extracts, using casein as substrate. Two Bacillus isolates were eventually chosen owing to their highest proteolytic activities (24.6 and 15.9 U/mL), and identified using molecular biology tools.  相似文献

The protease activities of 212 strains of rumen bacteria isolated from New Zealand cattle grazing pasture were measured. Thirty-seven per cent of strains had activity greater than or equal to the proteolytic rumen bacterium Prevotella ruminicola and 43 of these isolates were identified by morphology, carbon source utilization, Gram stain, biochemical tests and fermentation end-product analysis. Hierarchical Cluster Analysis showed that the strains formed four clusters: cluster A contained 26 strains and clustered with a reference strain of Streptococcus bovis; cluster C contained three strains and clustered with a reference strain of Butyrivibrio fibrisolvens , while clusters B (10 strains) and D (three strains) did not cluster with any of the remaining rumen bacterial type strains. Further tests identified strains of cluster B as Eubacterium budayi , while cluster D strains most closely resembled B. fibrisolvens and were described as B. fibrisolvens -like. An unclustered strain, C21a, was identified as P. ruminicola. The significance of these proteolytic bacterial populations is discussed in relation to protein breakdown in New Zealand ruminants.  相似文献

Arsenic oxidation is recognized as being mediated by both heterotrophic and chemoautotrophic microorganisms. Enrichment cultures were established to determine whether chemoautotrophic microorganisms capable of oxidizing arsenite As(III) to arsenate As(V) are present in selected contaminated but nonextreme environments. Three new organisms, designated as strains OL-1, S-1 and CL-3, were isolated and found to oxidize 10 mM arsenite to arsenate under aerobic conditions using CO2-bicarbonate (CO2/HCO3-) as a carbon source. Based on 16S rRNA gene sequence analyses, strain OL-1 was 99% most closely related to the genus Ancylobacter, strain S-1 was 99% related to Thiobacillus and strain CL-3 was 98% related to the genus Hydrogenophaga. The isolates are facultative autotrophs and growth of isolated strains on different inorganic electron donors other than arsenite showed that all three had a strong preference for several sulfur species, while CL-3 was also able to grow on ammonium and nitrite. The RuBisCO Type I (cbbL) gene was positively amplified and sequenced in strain CL-3, and the Type II (cbbM) gene was detected in strains OL-1 and S-1, supporting the autotrophic nature of the organisms.  相似文献

Hexavalent chromium [Cr(VI)] is a known carcinogen and mutagen; however, the actual mechanisms of Cr toxicity are unknown. Two approaches were used to isolate Cr(VI)-resistant bacteria from metal-contaminated river sediments. Diluted sediments were plated directly onto a peptone-yeast extract (PYE) medium containing 0 to 100 micrograms of Cr(VI) ml-1. Approximately 8.4 x 10(5) CFU g-1 were recovered on 0 microgram of Cr(VI) ml-1, whereas 4.0 x 10(2) CFU g-1 were recovered on PYE plus 100 micrograms of Cr(VI) ml-1. Alternatively, continuous culture enrichment techniques were employed using PYE and 100 micrograms Cr(VI) ml-1 input at dilution rates of 0.02 and 0.10 h-1. After six residence periods, 10(9) CFU were recovered on PYE agar containing 0 microgram of Cr(VI) ml-1 and 10(7) CFU on PYE agar plus 100 micrograms of Cr(VI) ml-1. Of 89 isolates obtained by direct plating onto PYE, 47% were resistant to 100 micrograms of Cr(VI) ml-1, and 29% were resistant to 250 micrograms of Cr(VI) ml-1. When the same isolates were plated onto PYE containing Cr(III), 88% were resistant to 100 micrograms ml-1 but only 2% were resistant to 250 micrograms ml-1. Cr, Co, Sb, and Zn were found in significantly higher concentrations at an industry-related contaminated site than at a site 11 km downstream. Total Cr in the sediments at the contaminated site averaged 586 micrograms (dry weight) g-1, and the downstream site averaged 71 micrograms (dry weight) g-1. The Cr recovered from acid-digested Ottawa River sediment samples was predominantly hexavalent. Five acid digestion procedures followed by atomic absorption spectroscopy were compared and found to be 30 to 70% efficient for recovery of Cr relative to neutron activation analysis. Biofilms are structured communities of bacteria, which are adhered to a surface and embedded in a self-produced matrix of extracellular polymeric substances. Since biofilms are very resistant to antimicrobial agents, they are at the basis of a range of problems, including quality and safety issues in food industry. Recently, major advances have been made in elucidating the different structural components of the biofilm matrix, the regulatory pathways involved in biofilm formation, and signaling molecules involved in biofilm formation and dispersal, which provide opportunities for prevention and control of these biofilms in the food industry.  相似文献

The specificities of the monoclonal antibodies and polyclonal rabbit serum used in the study were tested against a number of Vibrio strains, including 4 isolates from intestinal tracts of healthy fish and 98 isolates from sediments. V. salmonicida was detected in sediment samples from diseased farms several months after an outbreak of the disease. The bacterium was also detected in a sediment sample from a disease-free fish farm. No V. salmonicida could be detected in sediments not influenced by fish farming. The number of positive samples was generally higher with application of rabbit serum as opposed to use of monoclonal antibodies, indicating that the rabbit serum may cross-react with other bacteria.  相似文献

南海南沙海域沉积物中可培养微生物及其多样性分析   总被引:10,自引:0,他引:10  
【目的】为了从南沙海域中分离获得微生物菌种资源,【方法】本文通过沉积物采样、可培养菌分离及16S rRNA鉴定,【结果】从22个站点的沉积物样品中获得349株细菌,分属于87个种。发现产芽孢细菌分布最广,并在10个站点的分离株中占多数;它们是Bacillus,Halobacillus,Brevibacillus,Paenibacillus,Pontibacillus和Thalassobacillus。其中芽孢杆菌(Bacillus)无论在数量上还是种类上都最多,分别属于34种,其中有8个可能的新种。此外,g-Proteobacteria是分离率较高的另一亚群;其中,假单胞菌(Pseudomonas),海杆菌(Marinobacter),食烷菌(Alcanivorax)属的细菌最多。统计还发现,在深度750~2000 m之间,低GC含量的细菌最丰富,而深度2000 m以下,分离株则全部为g-Proteobacteria。【结论】南沙沉积物可培养微生物中产芽孢细菌及 g-Proteobacteria比较丰富;其中,产芽孢细菌的多样性最高,具有进一步研究开发价值。  相似文献

The increasing ecological significance of Planctomycetes and the still limited knowledge of this group prompted us to obtain cultured isolates from the sediment of a treatment water recycling tank of a marine fish farm. Presence of strains from this group was assessed in the sediments and water column of the tank. Eleven isolates were obtained from the sediment sample by exploiting Planctomycetes natural resistance to several antibiotics and their capacity to degrade organic matter. Based on morphological characteristics and resistance to antibiotics, Planctomycetes were identified. Their phylogenetic affiliation was confirmed by the sequence analysis of the 16S rRNA gene that revealed the presence of a group of 6 isolates closely related to Rhodopirellula baltica and a cluster of 5 isolates with 97.7-97.9?% of similarity to this species, which probably are a different species of Rhodopirellula. ERIC-PCR profiles showed a higher discrimination within the two groups and allowed the identification of nine different genotypes within the isolated strains. This work corroborates the association of Rhodopirellula spp. with fish farm environments.  相似文献

Sixteen environmental samples, from the United States, Germany and Norway, with histories of previous exposure to either creosote, diesel fuel or coal tar materials, were screened for bacteria which could degrade high molecular weight (HMW) polycyclic aromatic hydrocarbons (PAHs). A modified version of the spray plate technique was used for the isolations. Using fluoranthene (FLA) and pyrene (PYR) as model HMW PAHs, we isolated 28 strains on FLA and 21 strains on PYR. FLA degraders were defined as able to grow on FLA but not PYR. PYR degraders grew on both PAHs. All PYR degraders were found to be Gram-positive and all FLA degraders were Gram-negative. GC-FAME analysis showed that many of the PYR degraders were Mycobacterium spp and many of the FLA degraders were Sphingomonas spp. Comparison of the metabolic characteristics of the strains using the spray plate technique and direct growth studies revealed that more than half of the FLA degraders (59%) were able to cometabolize PYR (ie, they produced clearing zones or colored metabolites on spray plates but did not grow on the PAH) and the ability of many of these strains to cometabolize fluorene, anthracene, benzo[b]fluorene, benzo[a]anthracene and benzo[a]pyrene was significantly affected by pre-exposure to phenanthrene. Studies on the metabolic products produced from PYR cometabolism by strain EPA 505 suggested the possibility of attack at two different sites on the PYR molecule. However, the inability to derive degradable carbon from initial opening of one of the PYR rings probably accounted for the lack of growth on this PAH by the FLA-degrading strains. The PYR degraders on the other hand, were less able to cometabolize HMW PAHs, even following pre-exposure to PHE. Characterization of the FLA degradation pathway for several of the Sphingomonas isolates indicated oxidation and ring opening through to acenaphthenone as the principle metabolite. Strict anaerobic culture techniques were used to quantitatively and qualitatively evaluate the anaerobic heterotrophic bacteria present at the sediment-water interface of hyperutrophic Wintergreen Lake (Augusta, Mich.). Anaerobic plate counts remained constant from March through December, 1973, ranging from 2.4 X 10(6) to 5.7 X 10(6) organisms/g (dry weight) of sediment. The isolatable bacteria represented a small percentage of the total microbial community, which was shown by direct microscopic counts to be 2.0 X 10' organisms/g (dry weight) of sediment during June and July. Bacteria of the genus Clostridium dominated the isolates obtained, accounting for 71.8% of the 960 isolates examined. A single species, Clostridium bifermentens, comprised 47.7% of the total. Additional bacterial groups and the percentage in which they were isolated included: Streptococcus sp. (10.8%), unidentified curved rods (9.5%y, gram-positive nonsporing rods (5.6%), and motile gram-negative rods (1.9%). Temperature growth studies demonstrated the ability of all the isolates to grow at in situ sediment temperatures. Gas-liqid radiochromatography was used to determine the soluble metabolic end products from [U-14C]glucose and a U-14C-labeled amino acid mixture by representative sedimentary clostridial isolates and by natural sediment microbial communities. At in situ temperatures the natural sediment microflora produced soluble fermentative end products characteristic of those elaborated by the clostridial isolates tested. These results are considered strong presumptive evidence that clostridia are actively metabolizing in the sediments of Wintergreen Lake.  相似文献

【目的】海洋微生物在活性物质开发方面具有巨大的应用前景。为了研究胶州湾微生物的多样性和抑菌活性,选取胶州湾9个观测站点的沉积物进行了细菌多样性及抑菌活性分析。【方法】采用YPD和Z2216E培养基分离细菌,通过16S r RNA基因测序对分离菌株进行分类鉴定,继而采用牛津杯法考察分离菌株对7株指示菌的抑菌活性,最后用PCR方法筛选16株代表菌的PKSI、NRPS、CYP、Phz E和d TGD基因。【结果】从胶州湾沉积物中共分离出76株细菌,通过对16S r RNA序列分析,将其归为8个科11个属:节杆菌属、考克氏菌属、微球菌属、微杆菌属、假交替单胞菌属、Oceanisphaera、海单胞菌属、葡萄球菌属、芽孢杆菌属、硫胺素芽孢杆菌属和短芽孢杆菌属,其中分离细菌中有34株至少对1种指示菌有抑菌活性。所选取的16株菌均能检测到一种功能基因,且其中5株菌能同时检测到3种以上的功能基因。【结论】以上研究表明胶州湾海域有丰富的微生物资源,在生物活性次级代谢产物合成上有较大潜力。  相似文献

[背景] 海洋微生物在活性物质开发方面具有巨大的应用前景,而目前有关南海东海岛微生物的研究鲜少。[方法] 对从东海岛沉积物中分离纯化的海洋细菌,采用形态学观察、生理生化以及16S rRNA基因序列的系统发育分析方法进行鉴定;以大肠杆菌(Escherichia coli)、枯草芽孢杆菌(Bacillus subtilis)和金黄色葡萄球菌(Staphylococcus aureus)作为指示菌,测定其抑菌活性;对具有抑菌活性的菌株扩增聚酮合酶(Polyketide synthase I,PKSI)基因,并与已知的PKSI氨基酸序列比对;选择具有PKSI基因的代表菌株,检测菌株及其发酵抑菌物的稳定性。[结果] 分离纯化到25株海洋细菌,分属于不动杆菌属(Acinetobacter)、交替单胞菌属(Alteromonas)、芽孢杆菌属(Bacillus)、嗜冷杆菌属(Psychrobacter)、假交替单胞菌属(Pseudo-alteromonas)、海洋单胞菌属(Oceanimonas)、葡萄球菌属(Staphylococcus)、微球菌属(Micrococcus)和海杆菌属(Marinobacter)。12株菌株通过基因筛选检测到PKSI编码基因,其中6株菌株具有抑菌活性和PKSI编码基因,并分属于芽孢杆菌属和交替单胞菌属;PKSI氨基酸序列同源性分析推测菌株DHD-15和DHD-a可能产生新的I型聚酮合酶结构。菌株DHD-15和DHD-L生长温度范围为15-40℃,可耐受10% NaCl高盐以及pH 3和pH 11的酸碱条件,但不耐高温;菌株DHD-15产生的抑菌物质可耐受100℃和pH 11的高温碱性条件,在50℃、pH 9条件下制备和室温保藏条件下抑菌活性较高,其稳定性较好。[结论] 南海东海岛沉积物筛选的细菌种具有抑菌活性,具有产聚酮类活性物质的潜力。  相似文献

The requirement for proteinase inhibitors during the chromatographic isolation of tubulin from cultured cells of rose (Rosa sp. cv. Paul's scarlet) was examined by NadodecylSO4-polyacrylamide gel electrophoresis, electron microscopy and immunoblotting. Tubulin fractions isolated in the absence of proteinase inhibitors showed substoichiometric ratios of alpha-subunit to beta-subunit, and low molecular weight polypeptides, one (approximately 32 Kd) of which coassembled with polymers. Electron microscopy revealed polymorphic structures, including C- and S-shaped ribbons and free protofilaments. Immunoblotting experiments with IgGs to the individual alpha- and beta-subunits showed that some of the low molecular weight polypeptides were fragments of proteolytically degraded subunits. The use of low micromolar concentrations of the synthetic proteinase inhibitors leupeptin hemisulfate and pepstatin A protected tubulin from endogenous proteolytic activities during the isolation procedure and resulted in increased tubulin purity.  相似文献

【背景】脱硫细菌对有机硫的脱硫作用在硫的生物地球化学循环以及脱硫工业中都起着重要的作用。【目的】了解海洋沉积物中可分解有机物产生硫化氢的细菌多样性。【方法】对我国北戴河海洋沉积物中可培养的L-半胱氨酸脱硫细菌进行分离与筛选,通过对其16SrRNA基因序列测定与分析,构建系统发育树,并对其脱硫、脱氮能力进行检验。【结果】从海洋沉积物中分离得到97株细菌,从以L-半胱氨酸为硫源的培养基中筛选出62株有机脱硫专一型细菌。根据脱硫细菌的形态及其特征,从中选取12株作为典型代表做进一步分析,它们分别属于芽孢杆菌属(Bacillus)、赖氨酸芽孢杆菌属(Lysinibacillus)、动性球菌属(Planococcus)和红球菌属(Rhodococcus)。结果表明,这12株细菌均可产生半胱氨酸脱巯基酶,能够将半胱氨酸分解为丙酮酸、硫化氢和氨,即同时具备脱硫与脱氮的能力。其中有5株菌脱硫能力较强,分别属于赖氨酸芽孢杆菌属、动性球菌属和芽孢杆菌属。【结论】海洋沉积物中存在着丰富的L-半胱氨酸脱硫细菌,为进一步研究海洋中硫的生物地球化学循环提供了素材。  相似文献

Summary Bacteria with the ability to form L-phenylalanine from acetamidocinnamic acid were isolated from several soils. Among them, strain no. S-7 and strain no. N-7 were identified as Alcaligenus faecalis S-7 and Bacillus sphaericus N-7, respectively. The L-phenylalanine-forming enzyme systems in both bacteria were found to be inducible and intracellular. With intact cells of both bacteria and 40 mg/ml as wet base, 10 mg/ml acetamidocinnamic acid was utilized, and 7.7 mg/ml L-phenylalanine in a molar yield of 94% was produced after 72h incubation. The pathway of L-phenylalanine formation is considered to take the following course: acetamidocinnamic acid is deacetylated to -amino cinnamic acid; this is spontaneously changed to phenylpyruvic acid, and L-phenylalanine is formed by transamination.  相似文献

Nine lactic acid bacteria strains showing bacteriocin-like activity were isolated from various fresh fish viscera. The following species were identified based on 16S rDNA sequences: Enterococcus durans (7 isolates), Lactococcus lactis (1) and Enterococcus faecium (1). These strains were active against Listeria innocua and other LAB. Random amplified polymorphic DNA analyses showed four major patterns for the E. durans species. PCR analyses revealed a nisin gene in the genome of the Lc. lactis strain. Genes coding enterocins A, B and P were found in the genome of the E. faecium isolate. Enterocins A and B genes were also present in the genome of E. durans GM19. Hence, this is the first report describing E. durans strains producing enterocins A and B. Electrospray ionization mass spectrometry revealed that the purified bacteriocin produced by the E. durans GMT18 strain had an exact molecular mass of 6,316.89 Da. This bacteriocin was designated as durancin GMT18. Edman sequencing failed to proceed; suggesting that durancin GTM18 may contain terminal lanthionine residues. Overall, the results obtained revealed the presence of a variety of enterococci in Mediterranean fish viscera, as evidenced by their genetic profiles and abilities to produce different bacteriocins. These strains could be useful for food biopreservation or as probiotics.  相似文献
