In this paper, all known autoecious Puccinia species (rust fungi, Uredinales) on Cucurbitaceae are described and most of them illustrated. A key and information on their host range and distribution is presented. Four species, Puccinia antennata, P. arbor-miraculensis, P. hieroglyphica, and P. rhytidioderma are proposed as new. P. momordicae and P. trochomeriae are recognised as valid species different from P. cephalandrae. Uredo melothriae is a new combination for Uromyces melothriae. P. cucumeris is a new report for Namibia, P. momordicae for Zimbabwe and P. vanderystii for Kenya. The species appear to be host specific with regard to host tribes, but P. citrulli and P. cucumeris may pass tribal boundaries. Most species are morphologically very similar and are characterised by a unique set of teliospore characters. They are therefore regarded as a natural group. The distribution of this group in semiarid habitats, mainly from southern Africa to India, is discussed.  相似文献

Puccinia tatrensis Urban onGeum reptans in Czechoslovakia is recognized as a subspecies ofPuccinia sieversiae Arth.Puccinia waldsteiniae, P. gei andP. gei-parviflori appear to be the most ancient rusts known on the tribeGeeae and it is assumed that their common ancestral form was already present in the Cretaceous period.Puccinia sieversiae is more advanced and has its centre of origin and spread in Central Asia. Phylogenetically most advanced and at the same time most differentiated from each other are species ofPhragmidium, i.e.P. circumvallatum andP. miyabeanum.  相似文献

The oomycete Plasmopara obducens was first described on wild Impatiens noli-tangere in Germany in 1877. About 125 years later the first occurrence of P. obducens on cultivated I. walleriana in the United Kingdom was reported, and a worldwide epidemic followed. Although this pathogen is a major threat for ornamental busy lizzy, the identity of the pathogen remained unconfirmed and the high host specificity observed for the genus Plasmopara cast doubts regarding its determination as P. obducens. In this study, using multigene phylogenies and morphological investigation, it is revealed that P. obducens on I. noli-tangere is not the conspecific with the pathogen affecting I. walleriana and another ornamental balsam, I. balsamina. As a consequence, the new names P. destructor and P. velutina are introduced for the pathogens of I. walleriana and I. balsamina, respectively.  相似文献

In simple societies, activity of the workers can be regulated in a centralized way by a sole individual. Polistes wasps are simple in some respects because they have small, open colonies, with a single queen who is not morphologically differentiated from the workers. Worker activity in Polistes is episodic. Pause periods are followed by periods of intense activity, when most individuals are in motion. One way to investigate the regulation of worker activity is to see if activity periods are initiated by one specific individual. The queen was found regulating the resumption of activity in P. fuscatus in a centralized way, but recently, a decentralized regulation was found in P. instabilis and P. dominula. Thus, what is the pattern of regulation of worker activity in Polistes? To solve this question, broader studies of other species are desirable. Therefore, we investigated whether activity of P. versicolor workers are regulated by one individual. We tested for two mechanisms: triggering periods of colonial activity or actually physically stimulating workers to forage. The results demonstrated that no single individual was responsible to start periods of activity or physically stimulate worker’s departure. P. versicolor appears to be an example of a society with an intermediary organization of work, where the queen controls reproduction but does not organize work.  相似文献

The genusPhaseolus (Fabaceae) consists of some 50 species, all of which are distributed in the Americas. Four of these contain cultigens.P. vulgaris (common bean),P. lunatus (lima bean),P. acutifolius (tepary bean),P. coccineus subsp.coccineus (runner bean); andP. coccineus subsp.polyanthus (no English vernacular name). Biochemical markers—phaseolin seed storage protein and isozymes—have provided new evidence on the organization of the first three species. Domestication has possibly caused a strong reduction in genetic diversity inP. vulgaris andP. acutifolius. BothP. vulgaris andP. lunatus cultivars result from at least two independent domestications, in Mesoamerica and in the Andes. These two species consist of two gene pools, each of which includes wild ancestors and their respective cultivated descendants. Our findings suggest the need for additional emphasis on genetic conservation of wild ancestors and their use in breeding programs and for a comparison of inter-gene pool vs. intra-gene pool crosses in breeding programs.  相似文献

Examination of three species of batfishes (Teleostei: Epphippidae) from off Lizard and Heron Islands on the Great Barrier Reef led to the discovery of specimens of the trematode genus Paradiscogaster Yamaguti, 1934 (Digenea: Faustulidae). Morphological analysis demonstrated that the new specimens represented four morphotypes which we interpret to be new species: Paradiscogaster martini n. sp., P. vichovae n. sp. and P. brayi n. sp. from Platax orbicularis (Forsskål) and P. pinnatus (Linnaeus) off Lizard Island, and P. nitschkei n. sp. from P. teira (Forsskål) off Heron Island. Published material was re-examined and the specimens identified as P. chaetodontis okinawensis Yamaguti, 1971 from P. pinnatus from Okinawa, Japan, actually represent the new species P. brayi n. sp., demonstrating that some species of Paradiscogaster have wide geographical distributions. ITS2 rDNA data for the four morphotypes differ by 4–39 base pairs confirming the delineation of the four species proposed. A feature of this study is the recognition of Platax spp. as an important host group for Paradiscogaster, with the new species placing them as the second richest host group for these parasites after the Chaetodontidae.  相似文献

Objectives

To analyze the anti-insect mechanism of viral pesticide AcMNPV-BmK IT(P10/PH) in the host Spodoptera frugiperda 9 (Sf9) cells.

Results

Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV)- mediated expression of BmK IT, regulated by P10 protein promoter (P10) and polyhedrosis promoter (PH), promoted the replication of progeny virus in host Sf9 cells. AcMNPV-BmK IT(P10) could accelerate the budding process (or speed) of budded virus (BV) in Sf9 cells. The impact of AcMNPV-BmK IT(P10) on the nuclear polymerization of filamentous actin (F-actin) participated in regulating the accelerated budding process. Unexpectedly, both AcMNPV-BmK IT(P10) and AcMNPV-BmK IT(PH) delayed the nuclear polymerization of F-actin and promoted the clearance of F-actin in the nucleus. SfP53, an important apoptosis factor, was involved in the regulation of AcMNPV-BmK IT(P10/PH) in Sf9 cells. AcMNPV-BmK IT(P10/PH) could also delay and promote the nuclear recruitment of SfP53 after 27 h post infection (h p.i.).

Conclusion

SfP53 and F-actin are the targets of viral pesticide AcMNPV-BmK IT (P10/PH) in host Sf9 cells, which provides the experimental basis for the development of recombinant baculovirus biopesticides.

The amoeba, Mayorella viridis contains several hundred symbiotic green algae in its cytoplasm. Transmission electron microscopy revealed strong resemblance between symbiotic algae from M. viridis the symbiotic Chlorella sp. in the perialgal vacuoles of Paramecium bursaria and other ciliates. Although it is thought that the M. viridis and symbiotic algae could be model organisms for studying endosymbiosis between protists and green algae, few cell biological observations of the endosymbiosis between M. viridis and their symbiotic algae have been published. In this study, we characterized the specificity of endosymbiotic relationships between green algae and their hosts. Initially, we established stable cultures of M. viridis in KCM medium by feeding with Chlorogonium capillatum. Microscopic analyses showed that chloroplasts of symbiotic algae in M. viridis occupy approximately half of the algal cells, whereas those in P. bursaria occupy entire algal cells. The symbiotic algae in P. bursaria contain several small spherical vacuoles. The labeling of actin filaments using Acti-stain? 488 Fluorescent Phalloidin revealed no relationship between host actin filaments and symbiotic algal localization, although the host mitochondria were localized around symbiotic algae. Symbiotic algae from M. viridis could infect algae-free P. bursaria but could not support P. bursaria growth without feeding, whereas the original symbiotic algae of P. bursaria supported its growth without feeding. These data indicated the specificity of endosymbiotic algae relationships in M. viridis and P. bursaria.  相似文献

Understanding the environmental impact of bioenergy crops is needed to inform bioenergy policy development. We determined the effects of five biomass cropping systems—continuous maize (Zea mays), soybean (Glycine max)-triticale (Triticosecale ×)/soybean-maize, maize-switchgrass (Panicum virgatum), triticale/sorghum (Sorghum bicolor), and triticale-aspen (Populus alba × P. grandidentata)—on soil-saturated hydraulic conductivity (K S ) across a toposequence in central Iowa, USA. We compared data from the time of cropping system establishment in 2009 to 4 years post-establishment. Both our 2009 and 2013 data confirmed that cropping system impacts on K S vary by landscape position. We found that differences in cropping system impacts were more likely to occur at lower landscape positions, specifically, within footslope and floodplain positions. Previous research on cropping system impacts suggested that grass and woody systems were associated with a general increase in K S over time, with greater changes likely occurring at landscape positions with a higher erosive potential or lower SOC content. Our results confirmed that the triticale-aspen woody system was associated with a significant increase in K S across all landscape positions. In contrast, we did not observe an increase in K S under maize-switchgrass, which we attributed to the high density of switchgrass roots by the fourth year of study, but expect an increase in K S under switchgrass under longer measurement periods. We also found a significant increase in K S in the annual systems, likely due to the conversion to no-till soil management with cropping system establishment. We expect such differences to become more apparent over longer time scales as cropping systems continue to impact soil hydraulic properties.  相似文献

A complex of the heteropteran genera centering around Peribalus Mulsant et Rey and Holcostethus Fieber is considered. The genus Dryadocoris Kirkaldy reveals no relationship with the above genera and is believed to represent a separate clade of the family Pentatomidae. The genera Peribalus and Holcostethus are revised. The former includes three subgenera: Peribalus s. str. with two species, Asioperibalus subgen. n. (type species Cimex inclusus Dohrn) with six species, and Tianocoris subgen. n. (type species Holcostethus manifestus Kiritshenko) with two species. Holcostethus embraces two subgenera: Holcostethus s. str. and the monotypic Enigmocoris subgen. n. (type species H. fissiceps Horváth). Two new species are described: Peribalus tianshanicus sp. n. from the Tien Shan Mts. and P. przewalskii sp. n. from the northern part of China (Huan He River). P. capitatus Jakovlev and P. vernalis (Wolff) are downgraded to subspecies of P. strictus (F.). P. ovatus Jakovlev is synonymized with P. inclusus (Dohrn). Two new monotypic genera related to the revised complex of genera are established, Paraholcostethus gen. n. (type species Peribalus breviceps Horváth) and Himalayastethus gen. n. (type species H. pilosus sp. n. from Kashmir). A key to, and morphometric characters for all the taxa considered are provided. The key characters, including both male and female genitalia, are illustrated, and distributional maps are given.  相似文献

Trypophloeus klimeschi Eggers was first discovered in Xinjiang Province and had strong selection specificity for Populus alba var. pyramidalis Bunge. There was an outbreak of this beetle in the northwest shelter forest of China, resulting in significant economic losses and loss of ecological benefits. Based on a prior long-term field investigation, T. klimeschi had a different extent of injuries for different ages of P. alba var. pyramidalis and other Populus in the same area were not selected by T. klimeschi. To further explore the specificity volatile compounds, this study involved selecting host and non-host trees to analyse the volatile chemical profile of host and non-host poplars of T. klimeschi. The main volatile compounds of the host poplar P. alba var. pyramidalis for different physiological statuses and those of three other non-host poplars (P. alba L., P. tomentosa Carr., and P. dakuanensis Hsu) were analysed through solid-phase micro extraction (SPME) coupled with thermal desorption and gas chromatography-mass spectrometry (GC-MS). The major compound groups were aldehydes, esters, alcohols, ketones, phenols, terpenes and alkanes. Comparative analysis of the changes in the different physiological stages of P. alba var. pyramidalis and other non-host Populus volatile substances was conducted, and the results showed that 2-hydroxy-benzaldehyde, nonanal, decanal, 2-methyl-butanal, (Z)-3-hexen-1-ol benzoate, methyl benzoate, methyl salicylate, geraniol and salicyl alcohol might act as attractants for T. klimeschi, and 2-hexenal, hexanal, 2-cyclohexen-1-one, caryophyllene, eugenol, benzyl alcohol, and eucalyptol could be deterrents for T. klimeschi. These experiments may lead to the optimisation of a synthetic lure that may be used to detect and monitor T. klimeschi.  相似文献

A consensus rust QTL was identified within a 1.25 cM map interval of A03 chromosome in cultivated peanut. This map interval contains a TIR–NB–LRR R gene and four pathogenesis-related genes.

Abstract

Disease resistance in plants is manifested due to the specific interaction between the R gene product and its cognate avirulence gene product (AVR) in the pathogen. Puccinia arachidis Speg. causes rust disease and inflicts economic damages to peanut. Till now, no experimental evidence is known for the action of R gene in peanut for rust resistance. A fine mapping approach towards the development of closely linked markers for rust resistance gene was undertaken in this study. Phenotyping of an RIL population at five environments for field rust score and subsequent QTL analysis has identified a 1.25 cM map interval that harbored a consensus major Rust_QTL in A03 chromosome. This Rust_QTL is flanked by two SSR markers: FRS72 and SSR_GO340445. Both the markers clearly identified strong association of the mapped region with rust reaction in both resistant and susceptible genotypes from a collection of 95 cultivated peanut germplasm. This 1.25 cM map interval contained 331.7 kb in the physical map of A. duranensis and had a TIR–NB–LRR category R gene (Aradu.Z87JB) and four glucan endo-1,3 β glucosidase genes (Aradu.RKA6 M, Aradu.T44NR, Aradu.IWV86 and Aradu.VG51Q). Another resistance gene analog was also found in the vicinity of mapped Rust_QTL. The sequence between SSR markers, FRS72 and FRS49, contains an LRR-PK (Aradu.JG217) which is equivalent to RHG4 in soybean. Probably, the protein kinase domain in AhRHG4 acts as an integrated decoy for the cognate AVR from Puccinia arachidis and helps the TIR–NB–LRR R-protein to initiate a controlled program cell death in resistant peanut plants.

The stinkbug Plautia stali Scott is a notorious agricultural pest whose posterior midgut hosts specific bacteria essential for its growth and survival, highlighted as an experimental model for symbiosis studies. Some symbiotic bacteria of P. stali are cultivable, found free-living in and acquired from the environment, and, furthermore, some free-living environmental bacteria are potentially capable of establishing symbiotic association with P. stali. In this context, it is expected that such environmental bacteria may occasionally contaminate and infect the experimental insects maintained in the laboratory, which could potentially affect the functional analyses of the symbiosis. Here we report that such contamination events do occur under a laboratory rearing conditions for P. stali. When symbiont-deprived newborn nymphs from surface-sterilized eggs were reared in sterilized plastic containers with autoclaved water, most of them died as nymphs presumably as a result of aposymbiosis, but only a small fraction could attain adulthood and the adult insects were all infected with γ-proteobacteria allied to Pantoea and Enterobacter. A variety of bacteria, mainly Bacillus and also Pantoea and Enterobacter, were detected from peanuts and soybeans provided as food for P. stali. Autoclaving of peanuts and soybeans eradicated these bacteria but negatively affected the host survival, whereas ethanol sterilization of peanuts and soybeans removed Pantoea and Enterobacter, but not Bacillus, without negative effects on the host survival. On the basis of these results, we established a practical procedure for aseptic rearing of P. stali, which will enable reliable and strict analyses of host–symbiont interactions in the model symbiotic system.  相似文献

The Agave angustifolia complex, distributed from Mexico to Costa Rica, comprises four species and five varieties, including three species used for mescal production. The complex is represented in the Mexican state of Oaxaca by two wild taxa, A. angustifolia var. angustifolia and A. angustifolia var. rubescens, the cultivated form A. angustifolia “Espadín” and the partially cultivated species A. rodacantha. The aims of this study were to investigate the morphological and genetic variation of the A. angustifolia complex in the state of Oaxaca and to identify traits useful for taxonomic delimitation. Four wild and three cultivated populations of A. angustifolia from Oaxaca, one population of A. tequilana from Guanajuato and one population of A. angustifolia from Sonora were sampled for morphological, genetic and cytometric analyses. We showed that cultivated populations of A. angustifolia “Espadin,” A. rhodacantha and A. tequilana could be clearly differentiated from wild populations. Furthermore, the domesticated populations of A. angustifolia, known locally as “Espadin,” had a higher ploidy level and lower genetic variation than their related wild populations. The population of A. angustifolia from Sonora could be recognized as a different entity. Populations of A. rhodacantha need to be studied throughout their entire distribution area to further evaluate their taxonomic delimitation.  相似文献

Dastarcus helophoroides adults, a coleopteran parasitoid of Monochamus alternatus, were reared in the laboratory. The objectives of this paper are to investigate the host foraging behavior of D. helophoroides, and the evidence for learning in such behaviors. Choice and no-choice experiments were carried out. The choice experiments show that the number of D. helophoroides adults choosing the Pinus massoniana log infested with M. alternatus larvae was significantly higher than that choosing the blank (a flavorless simulative log made of brown kraftpaper), and the number of D. helophoroides adults choosing the uninfested P. massoniana log was also significantly higher than that choosing the blank. The no-choice experiment shows no significant difference between the number of D. helophoroides adults that chose the infested log and that chose the uninfested log, despite previous experience with uninfested log, infested log, or neither of them. No evidence of learning in host foraging behavior in D. helophoroides adults was found. The results indicate that the volatile chemicals emitted by uninfested P. massoniana logs and infested logs with M. alternatus larvae are equally attractive to D. helophoroides.  相似文献

Bark beetles (Curculionidae: Scolytinae) feed on the xylem and phloem of their host, which are composed of structural carbohydrates and organic compounds that are not easily degraded by the insects. Some of these compounds might be hydrolyzed by digestive enzymes produced by microbes present in the gut of these insects. In this study, we evaluated the enzymatic capacity of bacteria (Acinetobacter lwoffii, Arthrobacter sp., Pseudomonas putida, Pseudomonas azotoformans, and Rahnella sp.) and yeasts (Candida piceae, Candida oregonensis, Cyberlindnera americana, Zygoascus sp., and Rhodotorula mucilaginosa) isolated from the Dendroctonus rhizophagus gut to hydrolyze cellulose, xylan, pectin, starch, lipids, and esters. All isolates, with the exception of C. piceae, showed lipolytic activity. Furthermore, P. putida, P. azotoformans, C. americana, C. piceae, and R. mucilaginosa presented amylolytic activity. Esterase activity was shown by A. lwoffii, P. azotoformans, and Rahnella sp. Cellulolytic and xylanolytic activities were present only in Arthrobacter sp. and P. azotoformans. The pectinolytic activity was not recorded in any isolate. This is the first study to provide evidence on the capacity of microbes associated with the D. rhizophagus gut to hydrolyze specific substrates, which might cover part of the nutritional requirements for the development, fitness, and survival of these insects.  相似文献

The agronomic performances of giant miscanthus (Miscanthus x giganteus) and switchgrass (Panicum virgatum L.) grown as bioenergy grasses are still unclear in North Carolina, due to a relatively short period of introduction. The objectives of the study were to compare the biomass yield and annual N removal of perennial bioenergy grasses and the commonly grown coastal bermudagrass [Cynodon dactylon (L.) Pers.], and to determine the optimum N rates and harvest practices for switchgrass and miscanthus. A 4-year field trial of the grasses under five annual harvest frequencies (May/Oct, June/Oct, July/Oct, Aug/Oct, and October only) and five annual N rates (0, 67,134, 202, and 268 kg N ha?1) was established at a research farm in Eastern North Carolina in 2011. Across harvest treatments and N rates, greatest biomass was achieved in the second growth year for both miscanthus (19.0 Mg ha?1) and switchgrass (15.9 Mg ha?1). Grasses demonstrated no N response until the second or the third year after crop establishment. Miscanthus reached a yield plateau with a N rate of 134 kg ha?1 since achieving plant maturity in 2013, whereas switchgrass demonstrated an increasing fertilizer N response from 134 kg N ha?1 in the third growth year (2014) to 268 kg N ha?1 in the fourth growth year (2015). The two-cut harvest system is not recommended for bioenergy biomass production in this region because it does not improve biomass yield and increased N removal leads to additional costs.  相似文献

Large numbers of plant cell-wall (CW)-related genes have been identified or predicted in several plant genomes such as Arabidopsis thaliana, Oryza sativa (rice), and Zea mays (maize), as results of intensive studies of these organisms in the past 2 decades. However, no such gene list has been identified in switchgrass (Panicum virgatum), a key bioenergy crop. Here, we present a computational study for prediction of CW genes in switchgrass using a two-step procedure: (i) homology mapping of all annotated CW genes in the fore-mentioned species to switchgrass, giving rise to a total of 991 genes, and (ii) candidate prediction of CW genes based on switchgrass genes co-expressed with the 991 genes under a large number of experimental conditions. Specifically, our co-expression analyses using the 991 genes as seeds led to the identification of 104 large clusters of co-expressed genes, each referred to as a co-expression module (CEM), covering 830 of the 991 genes plus 823 additional genes that are strongly co-expressed with some of the 104 CEMs. These 1653 genes represent our prediction of CW genes in switchgrass, 112 of which are homologous to predicted CW genes in Arabidopsis. Functional inference of these genes is conducted to derive the possible functional relations among these predicted CW genes. Overall, these data may offer a highly useful information source for cell-wall biologists of switchgrass as well as plants in general.  相似文献
