Nuclear membrane fluidity is measured in rat liver by use of the fluorescence anisotropy of two probes: diphenylhexatriene and its cationic derivative trimethylammonium-diphenylhexatriene. It has been shown that, in 2-month-old rat liver cells, the bilayer surface is less fluid than the hydrophobic core. The fluidity was higher in 6-day-old rat liver nuclei, in which both the amount of cholesterol and the cholesterol/phospholipid ratio decreased. The influence of the single phospholipids, and in particular of phosphatidylcholine, has been studied by increasing the phosphatidylcholine with a choline base exchange reaction in isolated nuclear membranes. After this reaction, the fluorescence anisotropy of the bilayer surface increased, whereas at the hydrophobic core it decreased. Analysis of fatty acid composition shows an increase of phosphatidylcholine unsaturated fatty acids. The results show that the fluidity of nuclear membranes changes in relation to the lipid content and to the fatty acid composition. Effect of ethanol on rat brain polyphosphoinositides   总被引:1,自引:1,他引:0  
Abstract: Female rats were allowed to consume ethanol during gestation and lactation, and brain polyphosphoinositides of the 21-day-old pups were quantified. Ethanol intake prevented the disappearance of the metabolically labile pools of phosphatidylinositol-4-phosphate and phosphatidylinositol-4,5-bis-phosphate, which are rapidly degraded in the control group. In contrast, preweaning undernutrition left the size of these pools virtually unchanged, indicating a differential effect of the two nutritional regimens. It has been found that the rat liver nuclear matrix contains a small amount of phospholipids (2%) and neutral lipids (1.6%). The injection of hydrocortisone increased the nuclear matrix phospholipid content and reduced its neutral lipid levels. A marked increase in sphingomyelin content observed was accompanied by the reduction in phosphatidylcholine level. The decrease in neutral lipid content took place mainly at the expense of a sharp lowering of triglyceride and cholesterol ether levels. The latter may turn into free cholesterol, thus increasing its content. The data obtained testify to steroid hormone influence on lipid metabolism in intranuclear structures. The observed changes in lipid composition may be related to specific hydrocortisone-induced activation of genome.  相似文献

Synthesis and degradation of polyphosphoinositides in a rat brain synaptosome preparation were depressed by phenobarbital. Phosphatidylinositol-4-phosphate kinase (PIP-kinase), the enzyme which synthesizes phosphatidylinositol-4,5-bisphosphate (PIP2) was most strongly affected (50% inhibition at 3 mM phenobarbital); phosphatidylinositol (PI-kinase) followed (50% at 15 mM). The phosphoesterases were less sensitive: PIP-monoesterase (50% at 39 mM), PIP2-monoesterase (at 47 mM), and, least inhibited, PIP-diesterase (50% at 65 mM) and PIP2-diesterase (at 68 mM). Phenobarbital by inhibiting PIP-kinase may reduce the membrane concentration of PIP2 and thus dampen the stimulus-response which leads to the hydrolysis of PIP2 and the formation of the second messenger, inositol-1,4,5-trisphosphate (IP3), involved in mobilization of intracellular Ca2+.  相似文献

It was shown that hydrocortisone injections markedly increase the total content of liver nuclear membrane phospholipids, the greatest increase being observed in the phosphatidyl choline level. It was found also that nuclear membranes contain phospholipid metabolism enzymes. The hormone-induced increase in the phospholipid content is accompanied by a marked decrease of the activity of phospholipase A2 (5.3 times against control), phospholipase C (9.3 times) and acyl-CoA: lysophosphatidylcholine transferase (2.5 times). The results obtained are suggestive of appreciable metabolic changes in nuclear membrane phospholipids caused by hydrocortisone which, in turn, may be due to hormonal activation of the genome.  相似文献

The influence of cadmium chloride on the content of some fractions of polyphosphoinositides in the liver and kidneys of rats has been investigated in the work. We have reported that a single administration of sublethal dose of cadmium chloride leads to the long-term elevation of the content of diacylglycerol, which is responsible for the activation of protein kinase C. The increase of triphosphoinositides fraction content may be connected with activation of phosphoinositid-3-kinase and with accumulation of phosphatidylinositol-3,4-diphosphate and phosphatidilinositol-3,4,5-triphosphate, which are known as activators of some protein kinase C isoforms and also play an important role in the mitogen-activated protein kinase signaling pathway. The lipids fractions content changes were similar in the liver and kidneys, but had different time of response.  相似文献

It is shown that over 80% of nuclear neutral lipids are found in the nuclear membrane preparations, whereas only 7%--in chromatin. The injection of hydrocortisone to animals significantly decreases the neutral lipids content in both fractions. These results and the data obtained previously by the authors indicate the possible participation of nuclear membrane and chromatin lipids in the process of hormonal regulation of the genetic activity.  相似文献

The role of phosphoinositides has been thoroughly described in many signalling and membrane trafficking events but their function as modulators of membrane structure and dynamics in membrane fusion has not been investigated. We have reconstructed models that mimic the composition of nuclear envelope precursor membranes with naturally elevated amounts of phosphoinositides. These fusogenic membranes (membrane vesicle 1(MV1) and nuclear envelope remnants (NER) are critical for the assembly of the nuclear envelope. Phospholipids, cholesterol, and polyphosphoinositides, with polyunsaturated fatty acid chains that were identified in the natural nuclear membranes by lipid mass spectrometry, have been used to reconstruct complex model membranes mimicking nuclear envelope precursor membranes. Structural and dynamic events occurring in the membrane core and at the membrane surface were monitored by solid-state deuterium and phosphorus NMR. "MV1-like" (PC∶PI∶PIP∶PIP(2), 30∶20∶18∶12, mol%) membranes that exhibited high levels of PtdIns, PtdInsP and PtdInsP(2) had an unusually fluid membrane core (up to 20% increase, compared to membranes with low amounts of phosphoinositides to mimic the endoplasmic reticulum). "NER-like" (PC∶CH∶PI∶PIP∶PIP(2), 28∶42∶16∶7∶7, mol%) membranes containing high amounts of both cholesterol and phosphoinositides exhibited liquid-ordered phase properties, but with markedly lower rigidity (10-15% decrease). Phosphoinositides are the first lipids reported to counterbalance the ordering effect of cholesterol. At the membrane surface, phosphoinositides control the orientation dynamics of other lipids in the model membranes, while remaining unchanged themselves. This is an important finding as it provides unprecedented mechanistic insight into the role of phosphoinositides in membrane dynamics. Biological implications of our findings and a model describing the roles of fusogenic membrane vesicles are proposed.  相似文献

Phospholipid composition of nuclei and nuclear membranes from rat liver and hepatoma-27 were investigated. Hepatoma nuclei and nuclear membranes were found to contain cardiolipin which was absent in the same fractions of rat liver. In the nuclei and nuclear membranes of hepatoma the content of sphingomyelin was higher and that of lecithin is lower than in the corresponding subcellular fractions of rat liver. The content of acid phospholipids was much higher in hepatoma nuclear membranes than those of the normal liver. The data obtained show that the general trend of lipid dedifferentiation which earlier was demonstrated for various membranes of different tumor cells is observed also in the case of nuclear membranes.  相似文献
