Summary This study of the ultrastructure of the auditory sensilla of the New Zealand weta, Hemideina crassidens, is the first such study on a member of the orthopteran Superfamily Gryllacridoidea. Ultrastructure of the auditory sensilla is similar in all of the tibial mechanosensory organs, here called subgenual organ, intermediate organ and crista acoustica by analogy with comparable structures in Tettigoniidae.Distal to each sensory soma is a dendrite containing multiple ciliary rootlets that fuse into a single ciliary root. This splits into nine root processes that pass around the outside of the proximal basal body and then rejoin at the level of the distal basal body, distal to which the dendrite has a modified ciliary structure with a circlet of nine peripheral paired tubes and rods as it passes through the proximal extracellular space. It is then enclosed by a zone of scolopale cell cytoplasm before expanding into a dilatation within the distal extracellular space. In some sensilla this space is partially occluded by electron dense material which is part of the scolopale cell. Distal to the dilatation the cilium shrinks and ends surrounded by the scolopale cap.Accessory cells consist of glia enwrapping the sensory neuron in the region of its soma, the scolopale cell surrounding the ciliary portion of the dendrite, and the attachment cell surrounding the scolopale cell and scolopale cap and connected to them by desmosomes. The attachment cells are filled with microtubules in differing densities and orientations. Lamellae are present in the acellular matrix surrounding the attachment cells. Banded fibres, presumably of collagen, are also present in the matrix.  相似文献

Summary The statocyst ofOrconectes limosus contains static hairs arranged in four groups. All the hairs are the same in basic structure; they differ only in length and diameter and in their positions with respect to the other hairs in the group and to the statolith. In terms of functional morphology, each static hair is part of a unit consisting of an acellular lever string, three receptor cells, a scolopale cell, sheath cells, and enveloping cells. The lever string comprises two components in a characteristic longitudinal arrangement. The structure of the receptor cells resembles that of the arthropod chemo- and mechanoreceptors studied previously. The cilium and the postciliary section lie within two receptor cavities, formed by the scolopale cell and the sheath cells; the two cavities communicate with one another. The receptor cells are fixed in position by various structures. Proximally they form desmosomes with the scolopale cell, medially they are joined by filaments to the inner wall of cavity 1, and distally they are retained by a constriction between the two cavities. Two possible stimulus-mediating mechanisms are discussed: pressure changes in the receptor cavities and shearing of the base of the cilia with respect to the preciliary region. The lever string is part of the cuticle and hence is shed during molting. Summary The sensory receptor responsive to pressure applied internally to the ventral abdominal body wall of the blood-feeding insects, Rhodnius prolixus, is a single sense cell containing, at its distal end, a cilium enclosed within a scolopale, a densely staining structure characteristic of insect scolopidial sensilla. A small spherical structure lies within a dilation near the midregion of the cilium, and contains nine heavily staining bodies, the position of each corresponding to a pair of microtubules in the cilium. Proximal to the dilation, the microtubules are organized in a ring of nine pairs with one microtubule of each pair associated with dyneinlike arms. Dastal to the dilation a central pair of microtubules is present, but dyneinlike arms are absent. The scolopale cell, which gives risc to the scolopale, has cytoplasmic invaginations that form an elaborate array of extracellular compartments surrounding the body wall of the sense cell. These compartments may serve to dampen high frequency vibrations permitting the receptor to respond to pressure exerted by touch, an attribute in keeping with the receptor's proposed function of detecting abdominal distension related to the size and movement of the stomach.  相似文献

Summary The structure of a simple chordotonal organ, the presumed homologue of the noctuoid moth tympanal organ, is described in the atympanate moth, Actias luna. The organ consists of a proximal scolopidial region and a distal strand, which attaches peripherally to the membranous cuticle ventral to the hindwing alula. The strand is composed of elongate, microtubule-rich cells encased in an extracellular connective tissue sheath. The scolopidial region houses three mononematic, monodynal scolopidia, each comprised of a sensory cell, scolopale cell, and attachment cell. The dendritic apex is octagonally shaped in transverse section, its inner membrane lined by a laminated structure reminiscent of the noctuoid tympanal organ collar. A 9+0-type cilium emerges from the dendritic apex, passes through both the scolopale lumen and cap, and terminates in an extracellular space distal to the latter. Proximal extensions of the attachment cell and distal prolongations of the scolopale cell surrounding the cap are joined by an elaborate desmosome, with which is associated an extensive electron-dense fibrillar plaque. Within the scolopale cell, this plaque constitutes the scolopale rod material. The data are discussed in terms of both the organ's potential function, and its significance as the evolutionary proto-type of the noctuoid moth ear.  相似文献

In the four-eyed fish, Anableps (Atheriniformes, Anablepidae), eggs are fertilized and embryos develop to term within the ovarian follicles. Development is highly matrotrophic. During gestation, the largest term embryo of A. anableps examined had grown to a total length of 51 mm and attained a dry weight of 149 mg. The postfertilization weight increase is 298,000%. The largest term embryo of A. dowi examined had grown to a total length of 77 mm and attained a dry weight of 910 mg. The postfertilization weight increase is 843,000%. Embryonic weight increases result from nutrient transfer across the follicular placenta. This structure is formed by apposition of the maternal follicular epithelium to absorptive surface cells of the embryo's pericardial trophoderm. The latter, a ventral ramification of the pericardial somatopleure, replaces the yolk sac during early gestation. The external surface of the pericardial trophoderm develops hemispherical projections, termed vascular bulbs. Within each bulb, the vascular plexus of the trophoderm expands to form a blood sinus. Cells of the external surface of the bulbs possess microplicae. Microvilli are absent. During middle to late gestation, the juxtaembryonic follicular epithelium differentiates into two regions. One region consists of shallow, pitlike depressions within which vascular bulbs interdigitate in a “ball and socket” arrangement. Follicular pits are formed by the curvilinear distortion of the apical surfaces of follicle cells. The second region in contact with the dorsal and lateral surfaces of the embryo, is comprised of villous extensions of the hypertrophied follicular epithelium. In both regions, follicle cells appear to constitute a transporting rather than a secretory epithlium. In terms of percentage of weight increase, the follicular placenta of Anableps appears to be the most efficient adaptation for maternal-embryonic nutrient transfer in teleost fishes and closely approaches the efficiency (1.2 × 106%) of oophagy and embryonic cannibalism in lamnoid sharks.  相似文献

The fine structure of Johnston's organ in the pine sawfly, Neodiprion sertifer, was studied by electron microscopy to determine if there exists a dimorphism in this organ corresponding to the sexual dimorphism in antennal shape and surface area. The organ is made up of scolopidia that are ultrastructurally similar to those of other insects. The scolopidia, identical in both sexes, comprise three sensory cells bearing two types of sensory processes: Two are shorter and smaller in diameter than the third, which extends into the cuticle of the membrane connecting pedicel and flagellum and terminates at an epicuticular invagination. The dendrites and sensory processes are surrounded by two types of enveloping (glial) cells-a scolopale cell and an attachment cell. Other enveloping cells occur at different levels of the scolopidium. Sexual dimorphism is evident only in the numbers of scolopidial groups: Males have more groups with fewer scolopidia, but both sexes possess about the same total number of scolopidia.  相似文献

Arthritis may be the first clinical manifestation of primary hypogammaglobulinaemia. In 16 years of 281 patients who had immunodeficiency, 30 had arthritis at presentation. It was more common in Bruton''s disease (15 (22%) of 69 patients) than in other forms of immunodeficiency (15 (7%) of 212 patients). Non-septic arthritis was more prevalent than septic arthritis, particularly monoarticular arthritis in Bruton''s disease and pauciarticular disease in common variable immunodeficiency. Boys in whom a diagnosis of Bruton''s disease was delayed were likely to develop recurrent infections complicated by arthritis. The measurement of serum immunoglobulin concentrations readily differentiates immunodeficiency from conditions such as Still''s disease and dictates subsequent management.  相似文献

REM and TEM studies of the subgenual organ in Chrysoperla carnea (Neuroptera: Chrysopidae) show that it is composed of three scolopidia, each with one sensory, one scolopale and one cap cell. The distal part of the dendrite shows a cilium with a '9 + 0' structure. The cross-handing pattern of the ciliary root has a periodicity of bands of about 61 nm. The scolopale material in a certain part of the scolopale cell is organized into five rods. The cell bodies of all three cap cells form a lens-like structure. the velum, which is fixed to the leg wall and the trachea with an extracellular material. The importance of the velum is discussed. Four types of intercellular junction are found; spot desmosomes. belt desmosomes, septate junctions and gap junctions.  相似文献

The Gram stain differentiates bacteria into two fundamental varieties of cells. Bacteria that retain the initial crystal violet stain (purple) are said to be ''Gram-positive,'' whereas those that are decolorized and stain red with carbol fuchsin (or safranin) are said to be ''Gram-negative.'' This staining response is based on the chemical and structural makeup of the cell walls of both varieties of bacteria. Gram-positives have a thick, relatively impermeable wall that resists decolorization and is composed of peptidoglycan and secondary polymers. Gram-negatives have a thin peptidoglycan layer plus an overlying lipid-protein bilayer known as the outer membrane, which can be disrupted by decolorization. Some bacteria have walls of intermediate structure and, although they are officially classified as Gram-positives because of their linage, they stain in a variable manner. One prokaryote domain, the Archaea, have such variability of wall structure that the Gram stain is not a useful differentiating tool.  相似文献

利用SSR分子标记技术分析了佛坪国家级自然保护区秦岭箭竹(Fargesia qinlingensis)的克隆多样性和克隆结构,以探讨小尺度范围内秦岭箭竹自然居群遗传变异的分布特征,对该种开花特性、高山地区生态环境维护和大熊猫的保护提供重要依据。结果表明7对SSR引物共扩增出79个位点,其中多态性位点77个,多态位点百分率(PPB)为97.47%。秦岭箭竹的142个分株共形成107个克隆,最大克隆可达5 m。克隆多样性略高于其他克隆植物的平均值(D=0.62,G/N=0.17,E=0.68),基因型比率(G/N)、Simpson指数(D)、平均克隆大小(N/G)和Fager均匀性指数(E)分别为0.7535、0.9680、1.3271和0.5109。克隆空间结构分析表明秦岭箭竹的克隆构型为密集型,各克隆呈镶嵌性分布,同一克隆的分株排列紧密。克隆聚类分析表明各克隆之间聚类不明显,总体上来自同一样地的克隆被聚为一类。空间自相关分析显示在空间距离为36 m范围内,分株比基株有更显著的空间遗传结构,空间自相关系数r的取值范围分别为0.084—0.626和0.024—0.288,说明克隆繁殖在一定程度上限制了空间遗传结构的范围。样地内秦岭箭竹个体在空间距离小于44 m时存在显著的正相关空间结构,特别是在4 m处表现出最大的空间自相关系数(r=0.626),表明空间距离相距4 m内的个体最有可能属于同一克隆,4 m比5 m更能表现出清晰的克隆结构,X-轴截距为52.280,代表了秦岭箭竹不规则克隆的平均最小长度。秦岭箭竹的克隆多样性和克隆结构与初始苗补充、花粉散播方式和微环境差异有关。  相似文献

Small swellings near the base of the radial vein in each fore wing of the green lacewing, Chrysopa carnea, resemble typical insect tympanal organs, but some important differences are apparent. The swellings are bounded dorsally and laterally by thick cuticle and ventrally by thin, membranous cuticle. The ventral membrane is formed by a single, thin sheet of exocuticle with flattened hypodermis internally, but lacks the tracheal component that forms part of the tympanum in the typical insect tympanal organ. The portion of the membrane beneath each swelling is rippled while proximally it is smooth. In contrast to typical insect tympanal organs, the swellings in C. carnea are largely fluid-filled since an unexpanded trachea runs through each organ. A distal and a proximal chordotonal organ composed of typical chordotonal sensory units are associated with each swelling. The distal organ contains from five to seven units while the proximal organ is composed of from 18 to 20 units. Each sensory unit is composed of three readily identifiable cells. Distally, an attachment cell unites with the membrane and is contiguous with the scolopale cell, which surrounds the dendrite of the bipolar neuron. On the basis of the morphological evidence, one would not expect these swellings to function as sound receptors. However, the results of physiological and behavioral experiments, presented elsewhere, show that these organs are receptors for ultrasound.  相似文献

Trichobothria are present on the 3rd to 6th antennal segments in Campodea kervillei Denis and Plusiocampa cognata Condé (Apterygota : Diplura : Campodeidae). Each is composed of one closed cuticular pit with a long, thin hair, arising from the bottom. Under the pit, lies a globular structure surrounded by a fibrillar substance. The sensorial axis is composed of 4 neurons, each giving a cilium. The largest shows a tubular body and reaches the hair base. Three envelope cells wrap the sensory axis; the inner exhibits small longitudinal rods of microtubules embedded in a dense substance, similar to a scolopale.  相似文献

RNA-OUT, the 69-nucleotide antisense RNA that regulates Tn 10/IS 10 transposition folds into a simple stem-loop structure. The unusually high metabolic stability of RNA-OUT is dependent, in part, on the integrity of its stem-domain: mutations that disrupt stem-domain structure (Class II mutations) render RNA-OUT unstable, and restoration of structure restores stability. Indeed, there is a strong correlation between the thermodynamic and metabolic stabilities of RNA-OUT. We show here that stem-domain integrity determines RNA-OUT's resistance to 3’exoribonucleolytic attack: Class II mutations are almost completely suppressed in Escherichia coli cells lacking its principal 3′ exoribonucleases, ribonuclease II (RNase II) and polynucleotide phosphorylase (PNPase). RNase II and PNPase are individually able to degrade various RNA-OUT species, albeit with different efficiencies: RNA-OUT secondary structure provides greater resistance to RNase II than to PNPase. Surprisingly, RNA-OUT is threefold more stable in wild-type cells than in cells deficient for RNase II activity, suggesting that RNase II somehow lessens RNPase attack on RNA-OUT. We discuss how this might occur. We also show that wild-type RNA-OUT stability changes only twofold across the normal range of physiological growth temperatures (30–44°C) in wild-type cells, which has important implications for IS 10 biology.  相似文献

Background

A detailed understanding of an RNA's correct secondary and tertiary structure is crucial to understanding its function and mechanism in the cell. Free energy minimization with energy parameters based on the nearest-neighbor model and comparative analysis are the primary methods for predicting an RNA's secondary structure from its sequence. Version 3.1 of Mfold has been available since 1999. This version contains an expanded sequence dependence of energy parameters and the ability to incorporate coaxial stacking into free energy calculations. We test Mfold 3.1 by performing the largest and most phylogenetically diverse comparison of rRNA and tRNA structures predicted by comparative analysis and Mfold, and we use the results of our tests on 16S and 23S rRNA sequences to assess the improvement between Mfold 2.3 and Mfold 3.1.

Results

The average prediction accuracy for a 16S or 23S rRNA sequence with Mfold 3.1 is 41%, while the prediction accuracies for the majority of 16S and 23S rRNA structures tested are between 20% and 60%, with some having less than 20% prediction accuracy. The average prediction accuracy was 71% for 5S rRNA and 69% for tRNA. The majority of the 5S rRNA and tRNA sequences have prediction accuracies greater than 60%. The prediction accuracy of 16S rRNA base-pairs decreases exponentially as the number of nucleotides intervening between the 5' and 3' halves of the base-pair increases.

Conclusion

Our analysis indicates that the current set of nearest-neighbor energy parameters in conjunction with the Mfold folding algorithm are unable to consistently and reliably predict an RNA's correct secondary structure. For 16S or 23S rRNA structure prediction, Mfold 3.1 offers little improvement over Mfold 2.3. However, the nearest-neighbor energy parameters do work well for shorter RNA sequences such as tRNA or 5S rRNA, or for larger rRNAs when the contact distance between the base-pairs is less than 100 nucleotides.  相似文献

采用高通量测序技术分析了鄱阳湖典型湿地土壤细菌群落特征。测序结果表明,不同植被土壤细菌群落丰度与多样性的排序相同:苔草带苔草-虉草带芦苇带泥滩带藜蒿带。沿湖面至坡地,空间位置相近的土壤细菌群落结构具有更大的相似性,苔草-虉草带、苔草带和芦苇带的细菌群落结构相近,泥滩带和藜蒿带的细菌群落结构差异较大。变形菌门(30.0%)是湿地土壤平均相对丰度最高的门,其次为酸杆菌门(16.7%)和绿弯菌门(16.5%);多数门分类细菌相对丰度沿湖面至坡地存在一定变化趋势。硝化螺菌属是第一大属分类水平细菌群落。在土壤化学指标中,与鄱阳湖湿地细菌群落相关性较大的是总磷、铵态氮和有机质含量。以上研究结果表明,鄱阳湖湿地不同植被土壤细菌群落具有结构性差异,但沿湖面至坡地存在规律性变化。  相似文献

The fine structure of the statocyst sensilla of Neomysis integer was investigated. The statocyst contains about 35 sensilla, which are composed of two bipolar sensory cells, nine enveloping cells, and a seta. The sensory cells consist of an axon, a perikaryon, and a dendrite. The dendrite contains a proximal segment with a ciliary rootlet and at least one basal body, and a distal segment with a ciliary axoneme (9 × 2 + 0) at its base. The distal segment extends along the peripheral wall of the seta and is in close contact with the wall of the hair shaft. The enveloping cells surround the proximal and distal segments of the dendrite. The innermost enveloping cell contains a scolopale rod. It surrounds the receptor lymph cavity and secretes flocculent material into this cavity. From the tip of the cell a dendritic sheath, which encloses the distal segment of the dendrite, emerges. A peculiar feature of the second enveloping cell is the presence of a scolopale-like rod, which is more slender and less pronounced than in the first enveloping cell. The seta consists of three parts: a socket, a tubular midpart, and a gutter-like apical part, the tip of which penetrates into the statolith. The seta shows over its full length a bilaterally symmetrical axis that is coplanar with the plane in which the seta is bent toward the statolith. The structure of the seta and the position of the distal segments provide morphological evidence for directional sensitivity of the sensilla and for the magnitude of shear on the setal wall being an adequate stimulus.  相似文献

Summary The basic structure of the terminal sensilla of Locusta migratoria resembles that of Schistocerca gregaria. There are commonly six or ten neurons whose dendrites extend almost to the opening of the peg. Proximally the dendrites are clothed by a neurilemma cell which also encloses a basal cavity through which their ciliary region passes. The tormogen cell encloses the receptor-lymph cavity and actively secretes material into it. The receptor-lymph cavity and the basal cavity are quite separate.The development of new pegs at a moult is described. After apolysis the scolopale extends across the subcuticular space and protects the dendrites, which remain in a functional condition until shortly before ecdysis. As the trichogen cell grows out to form a new peg the tip is surrounded by a mass of electron-dense material, probably derived from the receptorlymph cavity. The function of this material is unknown. Regeneration of the dendrites is considered.The possible mechanism by which the tip of the peg opens and closes is considered and the general structure of the organule is discussed in relation to functioning.  相似文献

We developed a software package for improved free energy calculation, in which spherical solvent boundary potential, cell multipole method, and Nosé-Hoover equation are employed. The performance of the developed software package is demonstrated in the case of valine to alanine mutation of the 57th residue in chymotrypsin inhibitor 2. By using this package, we obtained results quantitatively comparable to experimental results. By the free energy component analysis, it is shown that leucine 51, arginine 65, arginine 67, and phenylalanine 69 residues contribute significantly to the total free energy shift, ΔΔG. Among them, contribution from the hydrophilic arginine 67 residue, which is in close contact with the mutation site, is the largest. Structure around the mutation site is largely changed by the mutation. The structure change is caused mainly by two effects, hydrophobic interaction and short-range interaction along the sequence. A conformational study of poly-L -serine has shown that it can exist in the left-handed α-helical form. A study of a pair of peptide units with the serine sidegroup attached to the α carbon atom linking the two units showed that O? H ?O hydrogen bonds between the OH group of the side chain and a carbonyl oxygen of the first peptide group in the backbone can occur in two regions of ?, namely, ? = 15°–30° for χ1 = 300° and for ? = 225°-230° for ? = 60°. The latter is close to a possible left-handed helix of poly-L -serine, stabilized by N? H ?O hydrogen bonds. From a study of contact criteria, the best conformation for this helix is found to be ? = 227°, Ψ = 238°, χ1 = 65° which has n = 3.65, h = 1.51 A. The N? H ?O hydrogen bond has a length of 2.90 A. (6°) and the O? H ?O hydrogen bond is of length 2.60 A. (0°). There are no other bad short contacts in the structure. The cylindrical coordinates of the atoms, as well as a perspective view of the structure arc given in this paper.  相似文献
