The number of microorganisms in the hindgut of dugongs (Dugong dugon) were estimated and their in vitro volatile fatty acid (VFA) production and degradation of eelgrass measured. Scanning electron microscopy showed that some rod bacteria attached to the surface of plant tissue degraded and eroded the cell walls. Number of starch-, lactate-, cellobiose-, pectin-, xylan- and cellulose-utilizing bacteria, sulfate-reducing bacteria and methane-producing bacteria were estimated at 109 ∼ 1010 colony forming units g-1. Microorganisms degraded the cellulose and noncellulolytic components of the eelgrass, and about 47.3% of dry matter was degraded after 36 h in vitro incubation. The total VFA concentration was 10.5 mmol dL-1 at 36 h incubation, which included 55.7 mol% acetate, 18.0 mol% n-butyrate and 15.1 mol% propionate. The gas composition of in vitro fermentation was 68.4% carbon dioxide, 22.2% methane and 9.4% hydrogen.  相似文献

Food consumption and digestion of male and female dugongs (Dugong dugon) was examined by analyzing long-term (1979–1998) feeding records at Toba Aquarium (Japan). Throughout all captive feeding periods, dugongs consumed eelgrass (Zostera marina) and showed steady increases in feed consumption with a consistent weight gain of 42–45?kg a year. The daily consumption of male and female dugongs increased from 10–15?kg to 23–26?kg of fresh eelgrass, accounting for approximately 14 and 7% of their body weight at one and seven years old, respectively. Both dugongs had a marginal dry matter digestibility of over 90%. Food consumption varied between seasons and individuals. There were major reductions (P<0.05) in consumption by the male in November and January and by the female in August and September. The seasonal reductions in food consumption coincided with high digestibility of eelgrass.  相似文献

Twenty‐six microsatellite loci have been isolated from a dugong (Dugong dugon). The average heterozygosity was 0.52 with two to 10 alleles per locus surveyed from 50 individuals. The markers are suitable for genetic mark–recapture (PID = 5 × 10?16) in dugongs and they could also be used to quantify physical tag loss, estimate relatedness, assign paternity, elucidate population structure and identify migrants. The loci also amplified in Florida manatees (22/26) and Asian elephants (6/26).  相似文献

The Australian dugong (Dugong dugon) and Florida manatee (Trichechus manatus latirostris) are threatened species of aquatic mammals in the order Sirenia. Sirenian conservation and management actions would benefit from a more complete understanding of genetic diversity and population structure. Generally, species-specific microsatellite markers are employed in conservation genetic studies; however, robust markers can be difficult and costly to isolate. To increase the number of available markers, dugong and manatee microsatellite primers were evaluated for cross-species amplification. Furthermore, one manatee and four dugong novel primers are reported. After polymerase chain reaction optimization, 23 (92%) manatee primers successfully amplified dugong DNA, of which 11 (48%) were polymorphic. Of the 32 dugong primers tested, 27 (84%) yielded product in the manatee, of which 17 (63%) were polymorphic. Dugong and manatee primers were compared and the most informative markers were selected to create robust and informative marker-panels for each species. These cross-species microsatellite marker-panels can be employed to assess other sirenian populations and can provide beneficial information for the protection and management of these unique mammals.  相似文献

Levels of urinary progesterone and estradiol-17 beta were measured twice a week in a female dugong, Dugong dugon, in captivity for two years from April 1996 to April 1998. The dugong showed 14 ovarian cycles during the period of study. Concentrations of progesterone ranged from 0.01 ng/mg creatinine (Cr) to 1.94 ng/mg Cr and the length of estrous cycle was 53.6+/-8.6 (mean+/-SEM) days based on intervals of urinary progesterone peak-to-peak measurements. Concentrations of urinary estradiol-17 beta ranged from 0.9pg/mgCr to 23.7pg/mgCr, and tended to peak just prior to elevations of progesterone during the first year of study. This is the first report demonstrates that the ovulatory cycle of the dugong is about 50 days. The present findings suggest that measurement of urinary progesterone is a useful method to detect ovarian cycle of the dugong in captivity.  相似文献

The in vitro digestibility of two-stage dried olive cake (TSDOC) and olive leaves (OL) unsupplemented or supplemented with increasing amounts of urea (U) or sunflower meal (SM) (0, 1.5, 2 and 2.5 g/100 g organic matter (OM) of the by-product) was determined. Chemical and amino acid composition, in vitro digestibility, in situ rumen degradability of crude protein and amino acids, and in situ–in vitro intestinal digestibility of SM CP and amino acids was determined. The in sacco rumen degradability and in vitro intestinal availability of CP and individual amino acids were also determined. Results obtained in Granadina goats and Segureña wethers were compared. SM provides arginine, glycine and aspartic and glutamic acids. The addition of increasing amounts of U or SM improved (P<0.001) the IVDMD and IVOMD of both TSDOC and OL. There was no effect (P>0.05) of the rumen inoculum origin on in vitro TSDOC digestibility. In contrast, values for OL were higher (P<0.001) for goats versus sheep. In sacco ruminal CP degradability of SM was relatively high, and similar in sheep and goats (ED=0.78 and 0.75 for sheep and goats). Individual amino acid ruminal degradability had different values, being lowest for methionine, leucine, proline, tyrosine and cysteine. Values obtained for individual amino acids differed from those of CP. Apparent intestinal digestibility of undegraded protein (AIDUP) of SM was high (0.86 and 0.98, respectively, for sheep and goats). The intestinally absorbable protein (IADP) was low (18.9 and 24.0 for sheep and goats, respectively). Results indicate that goats and sheep have the same capacity for TSDOC digestion, but goats showed a better capacity than sheep for OL utilisation. Although the amino acids supply to the intestine from SM is not important it could be a good supplement for low degradable protein feedstuffs such as TSDOC and OL.  相似文献

为明确海拔变化对干旱区山地森林土壤微生物群落的影响,揭示环境因子改变后土壤微生物群落结构特征及影响因素。对贺兰山5个海拔梯度土壤理化性质进行测定,同时采用磷酸脂肪酸(PLFA)图谱法分析土壤微生物群落组成,通过主成分分析、冗余分析(RDA)探究土壤理化性质与土壤微生物群落相对丰度之间的相关关系。结果表明:土壤养分含量在不同海拔之间差异性显著(P<0.05),土壤有机碳和全氮含量随海拔的升高而升高,全磷含量随海拔升高先升高再降低再升高;土壤微生物量随海拔升高先升高后降低,土壤微生物的相对丰度在不同海拔之间存在差异(P<0.05);主成分分析表明,与第1主成分相关性较强的微生物类群为革兰氏阳性细菌(G~+)、革兰氏阴性细菌(G~-)和真菌;与第2主成分相关性较强的微生物类群为放线菌、原生动物和非特异性细菌。非特异性细菌和真菌与各土壤因子之间均有显著相关关系,而放线菌、G~+和G~-与各土壤因子相关性较弱,原生动物与土壤全磷含量的关系密切。海拔是影响特征微生物分布的重要因素,特征微生物的含量和相对丰度随海拔的升高先升高后降低,符合山地生态学中的"中部膨胀"理论。探明了贺兰山不同海...  相似文献

In some lower rainfall regions of the world (300–750 mm), common vetch (Vicia sativa L., hereafter referred to as vetch) has been shown to have yields competitive with alternate crops and provide high-quality hay for ruminant diets, but there are few studies of vetch performance as a livestock feed, or of vetch varietal differences in livestock feeding value. This study evaluated vetch varietal differences in hay nutritive value, ruminal fermentation properties, nutrient digestibility, nitrogen retention and animal performance in fattening lambs consuming a diet comprising 20% vetch. Fifty male Hu lambs with an initial BW of 17.7 ± 0.27 kg and 2–3 months of age were assigned randomly into five groups of 10 lambs, and each allocated one of five dietary treatments for 67 days (10 days of adaptation and a 57 day experimental period). All diets contained 30% maize stover, 50% concentrate, and with a different forage source (on an as-fed basis): 20% alfalfa hay (Control), 20% vetch 333A (C333A) hay, 20% vetch Lanjian No. 1 (CLJ1) hay, 20% vetch Lanjian No. 2 (CLJ2) hay, or 20% vetch Lanjian No. 3 (CLJ3) hay. Hay CLJ3 had greater contents of ash, CP, ether extract, in vitro organic matter digestibility and metabolizable energy, and lower cell wall contents (P < 0.05) than those of C333A and CLJ1 hays, but similar to the CLJ2 hay (P > 0.05). Compared to the Control diet, the CLJ2 and CLJ3 diets resulted in greater (P < 0.05) final BW, average daily gain, total tract apparent digestibility of CP and NDF, and nitrogen balance, both when expressed as g/day and relative to nitrogen intake, while animal performance when fed diets with C333A or CLJ1 diet did not differ (P > 0.05) from the Control diet. No differences (P > 0.05) were observed between dietary treatments with respect to average daily feed intake, ruminal pH, total volatile fatty acid contents and molar proportions of acetate, butyrate, valerate, isobutyrate, and isovalerate, or total tract apparent digestibility of DM, organic matter, and ADF. The feed efficiency of tested feeds ranked CLJ3 = CLJ2 > CLJ1 > Control with C333A intermediate between CLJ1 and Control. In summary, considering hay quality, nutrient digestibility and animal weight gain, performance as a ruminant feed of hays from recently released vetch cultivars Lanjian No. 2 and Lanjian No. 3 was superior to the older cultivar C333A and the alfalfa control.  相似文献

Summary Under conditions of food deprivation, the hindgut epithelium of the experimental animals (Mesidotea entomon, Oniscus asellus, Porcellio scaber) undergoes ultrastructural changes. After the application of different diets it was demonstrated that this part of the alimentary canal contains nutrients, though it is lined by a cuticle. Experimental evidence for the formation of glycogen from glucose offered as the only diet comes from autoradiographic experiments. Amino acids, too, were detected in the hindgut cells soon after refeeding. Lipids, on the other hand, which are first absorbed by the large cells of the midgut glands, were not found in the hindgut epithelium. The existence of lipid inclusions in the hindgut epithelium some weeks after refeeding, however, supports the hypothesis that lipids reach the epithelial cells of the hindgutvia midgut glands and hemolymph.  相似文献

Okadaic acid (OA) is a diarrhetic shellfish poison (DSP) produced by a number of marine organisms including the benthic dinoflagellate Prorocentrum lima, which are often found on seagrass. As seagrass forms the basis of the diet of dugong (Dugong dugon) and green turtle (Chelonia mydas), these herbivores may potentially be exposed to OA through ingestion of P. lima found on the seagrass. In this study, the abundance of epiphytic P. lima, on seagrass, and the concentration of OA produced by these epiphytic dinoflagellates was measured in Moreton Bay, Queensland, Australia. P. lima and OA were found on all four species of seagrass collected. OA was detected in epiphytic material collected from seagrass, with a maximum of 460 ng OA/kg(wwtSG) found on Halophila spinulosa. From this information, the estimated maximum daily intake (DI) of OA by an adult dugong consuming 40 kg(wwtSG)/day was 18,400 ng/day, and an adult turtle consuming 2 kg(wwtSG)/day was 920 ng/day. Analysis by HPLC/MS/MS of 54 stranded dugongs and 19 stranded turtles did not yield OA above the detection limit of 10,000 ng/kg(animal tissue). OA was found on seagrass, however it was not detected in the tissue samples of dugongs and turtles.  相似文献

Abstract Examination of the culture fluid from model anaerobic communities in multiple-vessel chemostats showed that the most abundant organic intermediate products of substrate breakdown, acetate and propionate, existed in 3 distinct pools. Radiotracers, [14C]acetate or [14C]propionate, equilibrated between a free pool which was extracted by ether, an adsorbed or complexed pool which was only partly extracted by ether, and a third small pool which was not extracted. A microbiological assay demonstrated that the free pool was available to metabolism by bacteria, the second pool was slowly metabolised, but the third unextractable pool was refractile and not available to metabolism. These results were compared to the situation in the sediment environment.  相似文献

The dugong is a rare animal and virtually nothing is known of the time scale of its life cycle. A collection of 41 skulls and lower jaws, all from the north-eastern coastal region of Australia, was studied with the primary aim of relative age determination. It was found that they fell naturally into three groups corresponding roughly with skull size, according to number of incisors, number and state of eruption of cheek teeth and position of the cheek teeth in the jaw. These groups were arbitrarily termed adults, adolescents and juveniles. Skull measurements and state of fusion of skull sutures upheld these life stage groups, and to some extent helped to subdivide them. Further subdivision into a relative age sequence was achieved by "state of succession of cheek teeth" groups, and an index of size for the last cheek tooth. 采用宏基因组测序技术,研究了杨树人工林Ⅰ代林地和连作Ⅱ代林地根际土和非根际土、Ⅱ代林地主伐后轮作花生地和轮荒地土壤中解磷微生物类群特征及磷酸酶基因丰度变化.结果表明: 参与磷循环微生物共9属,芽孢杆菌属和假单胞菌属为优势菌群.土壤解磷微生物丰度大小顺序为轮荒地>轮作花生地>Ⅱ代林地>Ⅰ代林地;主伐更新后轮作花生地和轮荒地芽孢杆菌属和假单胞菌属丰度显著升高,节杆菌属、慢生根瘤菌属、链霉菌属丰度显著降低;但在人工林根际土壤中,与Ⅰ代林根际相比,Ⅱ代林根际土壤与此规律相反.在根际与非根际土壤中,芽孢杆菌属和假单胞菌属丰度为根际小于非根际,节杆菌属、慢生根瘤菌属、链霉菌属丰度为根际大于非根际.磷酸酶基因总丰度为Ⅰ代林地>轮荒地>Ⅱ代林地>轮作花生地,且在Ⅰ、Ⅱ代林中根际效应明显,但Ⅱ代林根际与非根际差异缩小且部分磷酸酶基因的根际丰度低于非根际丰度.解磷微生物数量与酚酸含量呈显著负相关,与pH值呈正相关.  相似文献

The effects of selenium (Se) on ruminant microbial fermentation were investigated in vitro using rumen microflora collected from a rumen-fistulated dairy cow. First, the effects ofl-selenomethionine (SeMet; at 0.2 or 2 ppm Se) in the presence or absence of wheat bran (WB, 500 mg per incubation flask) were evaluated. Second, the effects of several forms of Se (elemental Se: 50 ppm Se; sodium selenite: 2 ppm Se; SeMet: 2 ppm Se) were compared. Results showed that the amounts of short-chain fatty acids (SCFAs) tended to be increased by SeMet treatment, whereas SeMet in the presence of WB transiently suppressed fermentation. The addition of SeMet tended to increase the production of acetate while reducing the production of butyrate with and without WB supplementation. Among the different Se compounds tested, the amounts of SCFAs were greater with SeMet treatment, which yielded a higher proportion of acetate compared to other treatments. Selenite did not influence the total SCFAs concentrations; however, it increased the relative proportion of butyrate at the expense of acetate. Elemental Se did not significantly affect fermentation. Higher bacterial Se concentrations were observed for selenite than for SeMet. It was concluded that Se supplementation can influence rumen microbial fermentation and that Se compounds differ in this regard.  相似文献

In this study, the possibility of manipulating biopolymer composition in mixed culture polyhydroxyalkanoate (PHA) production from fermented molasses was assessed by studying the effects of substrate volatile fatty acid (VFA) composition and feeding regime (pulse wise versus continuous). It was found that the use of a continuous feeding strategy rather than a pulse feeding strategy can not only help mitigate the process constraints of the pulse-feeding strategy (resulting in higher specific and volumetric productivities) but also be used as means to broaden the range of polymer structures. Continuous feeding increased the hydroxyvalerate content by 8% relatively to that obtained from the same feedstock using pulse wise feeding. Therefore, the feeding strategy can be used to manipulate polymer composition. Furthermore, the range of PHA compositions, copolymers of P(HB-co-HV) with HV fraction ranging from 15 to 39%, obtained subsequently resulted in different polymer properties. Increasing HV content resulted in a decrease of the average molecular weight, the glass transition and melting temperatures and also in a reduction in the crystallinity degree from a semi-crystalline material to an amorphous matrix.  相似文献

This study was conducted to determine the effect of tamarind seed husk (TSH) as a source of tannin on various parameters of rumen fermentation in vitro. The TSH contained 14% tannin (DM basis). The biological interference of TSH tannin on rumen fermentation was assessed using polyethylene glycol (PEG) 6000 as an indicator. Three compound feed mixtures (CFM) were prepared either without TSH (CFM-I), with 2.5% TSH (CFM-II) and with 7.5% TSH (CFM-III). Parameters studied were in vitro gas production with PEG, rate of substrate degradation, and microbial protein synthesis. Addition of PEG to TSH resulted in an increase in gas production from 5.5 to 16.5 ml per 200 mg DM. Presence of TSH tannin depressed cumulative gas production by 16.8% in CFM-II, and by 29.2% in CFM-III during initial stages of fermentation (i.e. at 8 h). Rate of substrate disappearance (T1/2) was 14.4, 17.6 and 20.5 h in CFM-I, CFM-II and CFM-III, respectively. Irrespective of the carbohydrate source, presence of TSH tannin improved the efficiency of microbial protein synthesis in vitro. Thus, TSH is a natural source of tannin that can be used to beneficially manipulate rumen fermentation.  相似文献

This trial studied the effect of including mannanoligosaccharides (MOS, Bio-Mos®, Alltech Inc., USA) in the diet on the caecal volatile fatty acids (VFA) and pH of rabbits from 34 to 90 days of age. Three experimental diets were compared: control diet, zinc bacitracin (ZnBac) diet (control diet with 0.1 g ZnBac/kg feed) and MOS diet (control diet with 2.0 g MOS/kg feed). Rabbits were slaughtered at 34, 48, 69 and 90 days of age and caecal contents were collected and analyzed for dry matter, pH and VFA concentration. The empty caecum and caecal contents weights relative to live weight were also determined.Age affected (P<0.0001) VFA concentration and pH values in the caecum. The pH decreased with age whereas VFA concentration increased. Rabbits fed MOS had higher (P<0.05) VFA and tended (P=0.098) to had lower pH in the caecum than rabbits fed ZnBac and control diets. Acetic, propionic and butyric acids concentrations in the caecum increased with MOS but its molar proportions were similar among diets. Diet had no effect on empty caecum weight and caecal contents weight and dry matter concentration. There was no interaction effect between diet and age. From 34 to 90 days of age, VFA production was higher (P<0.05) in the caecum of rabbits fed MOS than in those fed ZnBac diet and control diet.The addition of MOS to the diet increased the VFA concentration in the caecum of growing rabbits from 34 to 90 days of age.  相似文献
