Data from several studies were appended to aerobic and anaerobic data sets that had been previously used to develop response surface models describing the growth kinetics of Listeria monocytogenes (Buchanan and Phillips, 1990). The expanded data sets included 709 aerobic and 358 anaerobic growth curves fitted with the Gompertz equation, and representing 189 and 150 unique combinations of four variables (temperature, pH, NaCl, NaNO2), respectively. Response surface models were developed for (1) the Gompertz B and M terms and (2) lag phase durations (LPD) and generation times (GT). In addition to modeling NaCl as a variable, a second set of response surface models was developed by substituting calculated water activity as a variable. A number of data transformations were evaluated in an attempt to better utilize no-growth data. Full quadratic models of the natural logarithm transformation of the data (no-growth data excluded) predicted values that fit the observed data well. The assignment of GT=50[emsp4 ]h and LPD=600[emsp4 ]h (the approximate maximum duration of experiments) for the variable combinations that did not support growth proved to be the most effective means of making use of the no-growth data. However, this approach did not offer any clear advantage over quadratic models where the no-growth data were excluded. Error matrices were developed for the LPD and GT models to provide 95 % confidence intervals. The agreement between observed and predicted growth kinetics was excellent considering the number and ranges of the variables encompassed in the models. The models provide reasonable predictions of the growth of L. monocytogenes in foods. The full quadratic models of LPD and GT without inclusion of the no-growth data were selected for inclusion in the USDA Pathogen Modeling Program, release 5.1.  相似文献

Polynomial equations, relating the growth rate of Yersinia enterocolitica to temperature (0–25°C) and pH (4.5–6-5) in a liquid medium were constructed for four different acidulants. The logarithm of the time for a 100-fold increase in bacterial numbers could be represented by a quadratic response surface function of pH and temperature. The interactions between pH and temperature on growth rate were found to be additive. Values for a 2 log cycle increase in growth derived from the model were in good agreement with experimental values. Predictions from the quadratic model and from a square root model were compared with experimental values in laboratory media and UHT milk. The mean square error (MSE) for the quadratic response surface model was smaller than that for the square root model in 81% of cases. In UHT milk the square root model increasingly underestimated growth rate, as the temperature decreased and would 'fail dangerous' if used for predictive purposes. This indicated that the response surface model is more reliable for predicting the growth of Y. enterocolitica under conditions of sub-optimal temperature and pH.  相似文献

The psychrotrophic bacterium Yersinia enterocolitica is characterized by temperature-dependent adaptations. To investigate Y . enterocolitica genes involved in cold adaptation, a mutant restricted in its ability to grow at 5°C was isolated from a transposon mutant library. The transposon insertion site in this psychrotrophy-defective (PD) mutant mapped 16 bp upstream of an open reading frame whose predicted amino acid sequence showed 93% similarity with the Escherichia coli exoribonuclease polynucleotide phosphorylase (PNPase), encoded by pnp . Expression of this gene was blocked in the PD mutant. However, the introduction of a second copy of pnp , including 0.33 kbp sequences upstream of its coding region, into the chromosome of the PD mutant restored pnp expression as well as the ability to grow at 5°C. Furthermore, the expression of pnp appeared to be temperature dependent: in the parental Y . enterocolitica strain, the levels of both pnp mRNA and PNPase were 1.6-fold higher at 5°C compared with 30°C. A similarly enhanced level of PNPase at 5°C was observed in the merodiploid recombinant strain, which indicates that the 0.33 kbp region upstream of pnp harboured a cold-inducible promoter. A putative cold shock promoter motif (ATTGG) was observed in this region.  相似文献

A selective medium was used to enumerate Clostridium botulinum growing in the presence of natural spoilage organisms in a model cured pork slurry. The growth responses of a mixed spore inoculum of six strains of Cl. botulinum type A were studied at 15°, 20° and 27°C with 1˙5, 2˙5, 3˙5 or 4˙5% (w/v) salt added (aw range 0961–0990). Gompertz and logistic curves, which have a sigmoid shape, were fitted to the data and lag times, growth rates, generation times and time to maximum growth rates were derived. Variation in germination rates of the spores occasionally gave a falsely extended lag time resulting in an exceptionally high estimate for growth rate. Products containing 4˙5% (w/v) NaCl would be capable of supporting growth of proteolytic strains of Cl. botulinum , even at 15°C, although the lag period would be extended. In products where absence of Cl. botulinum cannot be assured additional preservative measures are essential. The information obtained provides a framework to investigate the effects of a wider range of additives or variables on the growth responses of Cl. botulinum .  相似文献

Growth of two pathogenic and one environmental serotype of Yersinia enterocolitica under acidic conditions and at 4 and 25°C was investigated. At both temperatures the maximum growth inhibitory pH depended on the acidulant used and was in the order acetic > lactic > citric > sulphuric. At the lower temperature the maximum growth inhibitory pH was 0.3-0.5 pH units higher than at 25°C. No difference was observed between the behaviour of pathogenic and environmental serotypes in this respect. Measurement of growth at a number of sub-optimal temperatures and pH values showed that the variation of growth rate with temperature could be represented by a square root plot. The effect of different pH values could be incorporated into the model by replacing the regression coefficient b by its relationship with pH. Values of maximum growth inhibitory pH derived from the model were in good agreement with experimental values with the exception of acetic acid.  相似文献

The effect of sodium lactate and storage temperature on toxigenesis by proteolytic (Pr) and nonproteolytic (Np) Clostridium botulinum spores inoculated in processed 'sous-vide'-type beef, chicken breast and salmon was explored. Three g samples of beef and salmon homogenates with 0, 2.4 and 4.8% (w/w) lactate and of chicken with 0, 1.8 and 3.6% (w/w) lactate were placed in 24-well tissue culture plates. The samples were inoculated with 104 spores of pools of Pr (4A + 2B + 2F strains) or Np (4B + 4E strains), vacuum-packaged in barrier bags, and stored at 16 and 30°C for Pr and at 4, 8, 12 and 30°C for Np for up to 90 d. Lactate at 2.4% in beef and 1.8% in chicken delayed toxigenesis by Np for 40 d at 12°C and by Pr for 28 d at 16°C. Delaying toxigenesis for similar periods of time in salmon required 4.8% lactate and 12°C for Np, and 2.4% lactate and 16°C for Pr. Increasing levels of lactate and decreasing temperature significantly delayed toxigenesis of Cl. botulinum in the 'sous-vide' products.  相似文献

About 34% of the strains of Yersinia enterocolitica isolated from raw milk were found to produce lecithinase. A selected strain produced phospholipase C at 22°C and 37°C; production was optimum at 37°C in the stationary phase (14–16 h). A decrease in phospholipase C activity at various storage temperatures (—5°C, 4°C, 37°C) was also observed, although the enzyme was active over a wide range of temperature (5–65°C) and pH (3mD5–7mD5). The phospholipase C was partially purified by ammonium sulphate precipitation and Sephadex column chromatography, and characterized.  相似文献

The effects of temperature, sodium chloride and ascorbic acid on the aerobic growth kinetics of a clinical strain of Aeromonas hydrophila were evaluated. At 5°C, ascorbic acid (1 mmol l-1) and sodium chloride (3% w/v) inhibited the growth of the organism. At 10°C, ascorbic acid depressed only the maximum population densities (A) by approximately 2 log cycles, but not maximum specific growth rate (μm) or the lag time (Λ). On the contrary, NaCl caused A to increase, with the effect being greatest when the NaCl content was 1.5%. Temperature increase from 10 to 15°C resulted in an approximate doubling of μm and unexpectedly an apparent increase in Λ However, this apparent increase resulted from the particular manner in which the lag phase was mathematically calculated.  相似文献

Ventilation frequency patterns of Trinectes maculatus, Morone americana and Leiostomus xanthurus were used to evaluate potential thermal stress after exposure to moderate temperature increases. Fish acclimated to 5°, 15° and 25° C were exposed to a 5°C T; fish acclimated to 30° C were exposed to a 2.5° C A T. Ventilation frequencies were measured at each acclimation temperature before the fishes were exposed over a 15-min period to the increased temperatures. Ventilation rates were then measured at the elevated temperatures for the next 24 h. Significant increases in rate frequency occurred after the temperature increases in T. maculatus and M. Americana acclimated to 5°, 15° and 25° C and in L. xanthurus acclimated to 15°, 25° and 30°C. In general, rate frequencies increased as the temperature increased. Ventilation rates stabilized quickly at the higher temperatures and remained relatively constant throughout the remaining exposure period. Acclimated rate-temperatures curves (R- T curves), acute R- T curves and Q10 temperature coefficients used to assess the significance of the changes in rate frequency and to compare the species in an ecologically meaningful way, showed that several adaptive types occurred among and between species. The Q,10's of the acute R- T curves, in most cases, were found to approximate those values derived for the acclimated R-T curve. This suggests that the temperature increases had a negligible effect, that is, little or no thermal stress occurred.  相似文献

Abstract. Populations of Marenzelleria viridis in the Chester River (Kent County, Maryland) experience temperatures ranging from over 30°C in summer to near freezing in winter. Interestingly, M. viridis swims actively in winter. This observation led us to examine the relationship between locomotor capacity and temperature in individuals of M. viridis . Juvenile specimens were collected in February ("cold animals") and June ("warm animals"). Video analysis revealed that swimming is achieved by flexing the body in cyclic, helical waves. Wave frequencies were measured as an index of locomotor capacity at 5°C, 15°C, and 25°C. The mean wave frequencies of cold animals were 5.4 Hz at 5°C and 7.1 Hz at 15°C (Q10= 1.3); the mean wave frequencies of warm animals were 6.1 Hz at 15°C and 7.8 Hz at 25°C (Q10= 1.3). The effects of changes in water viscosity on wave frequency between 5–25°C were not significant. These results demonstrate that the temperature sensitivity of locomotor capacity in juvenile M. viridis is quite low. We conclude that low temperature sensitivity enables M. viridis to be active throughout the year.  相似文献

The growth kinetics of a virulence plasmid-bearing (P+) and a plasmid-cured (P−) strain of Yersinia enterocolitica serotype O:3 in pure and meat culture were investigated. Growth studies were carried out at 25 and 37 °C in supplemented phosphate-buffered saline, buffered peptone water , cefsulodin-irgasan-novobiocin broth base or supplemented broth base (CIN). The lag phase durations and growth rates under these conditions were determined by linear regression analysis. In pure culture, under most sets of equivalent conditions, P+ and P− strains had similar lag phase durations. However, under one set of conditions, i.e. CIN broth at 37 °C, the lag phase duration of the P+ strain was significantly longer than P−. In all but the most selective medium, P+ strains had slower growth rates than P− strains at 37 °C, probably due to the increased metabolic burden entailed in the maintenance of the virulence plasmid. In the most selective medium, i.e. CIN broth, P+ strains grew significantly faster than P−. This finding suggests that possession of virulence plasmid confers an enhanced ability to grow in the presence of selective agents. In meat cultures, both strains had longer lag phases than in equivalent pure cultures, with longer lag phases noted at 37 than at 25 °C. No significant differences were observed between the length of lag phases of P+ and P− strains in meat culture. Both strains of Y. enterocolitica displayed faster growth rates in meat cultures than in pure cultures, indicating that one or more components of meat enhanced the growth of this organism. The effects and interaction of incubation temperature, enrichment broth and meat on the growth kinetics of plasmid-bearing and plasmid-cured Y. enterocolitica strains are discussed.  相似文献

The effect of fluctuating temperatures on microbial growth is important in the passage of foods from production to consumption. Suspensions of Salmonella typhimurium have been subjected to sinusoidally time-varying temperatures of periods from 60 to 240 min between 4°C and 22°C, that is within and below the growth temperature range. The suspensions were prepared with two concentrations of sodium chloride and adjusted to two different values of pH. The change in the numbers of viable bacteria was measured with time and the experimental growth curves and average generation times compared with predictions based on isothermal growth data. Generally, the experimental average generation times exceeded the predictions by not more than 10% In enumerating viable bacteria in the suspensions containing 3.5% (w/v) sodium chloride it was necessary to use sodium chloride in the diluent and recovery medium in order to recover the bacteria quantitatively.  相似文献

The influence of incubation temperature, and of acetic, lactic and citric acids on the minimum pH for the initiation of growth of six strains of Yersinia enterocolitica was determined. The strains included two of serotype O : 9, two of serotype O : 3, and one each of serotypes O : 8 and O : 5, 27. In a culture medium acidified with HC1 to pH values between 4.0 and 6.0 at intervals of approximately 0.1 unit the minimum pH at which growth was detected after incubation at 20°, 10°, 7° and 4°C for 21 d was in the ranges 4.18–4.36, 4.26–4.50, 4.36–4.83 and 4.42–4.80, respectively. The minimum pH for growth was also determined in media that contained 17, 33 and 50 mmol/1 acetic acid adjusted to pH values between 5.1 and 5.9 at intervals of approximately 0.2 unit, 24, 48 and 95 mmol/1 citric acid adjusted to pH values between 41 and 4.9 at intervals of approximately 0.2 unit, and 22, 44, and 111 mmol/1 lactic acid adjusted to pH values between 4.3 and 5.7 at intervals of approximately 0.4 or 0.5 unit. The effect of these concentrations of organic acids was, in most cases, to increase the minimum pH that allowed growth. The order of effectiveness of the organic acids in raising the minimum pH for growth was acetic > lactic > citric and the minimum inhibitory concentrations were greater at higher temperatures.  相似文献

It has been reported that non-proteolytic strains of Clostridium botulinum will grow at 3.3°C, and they are therefore of concern in relation to certain chilled foods. The effects of combinations of inhibitory factors may be used to reduce the risk of growth of these bacteria in foods. The combined effect of pH values between 4.8 and 7.0, temperatures between 6° and 30°C, and sorbic acid concentrations up to 2270 mg/1 on the probability of growth from a single spore of non-proteolytic, type B strains in a culture medium has been determined. A mathematical model has been developed that enables the effect of varying combinations of these factors on the probability of growth of non-proteolytic, type B Cl. botulinum to be predicted.  相似文献

Two methods of determining the heat resistance of bacteria, a glass cup and a test tube method, were compared with a method using capillary tubes. Three strains of Yersinia enterocolitica , one of Campylobacter jejuni and two of C. coli were tested in physiological saline. The differences between the results obtained by the glass cup method and the reference method were not statistically significant for five strains and were small also for the other, a Yersinia strain. D values obtained by the glass cup method at 58, 60 and 62°C were 1.4–1.8, 0.40–0.51 and 0.15–0.19 min ( z values 4.00–4.52°C) for the Yersinia strains, and 0.42, 0.13 and 0.07 min ( z value 5.07°C) for one C. coli strain. For the other Campylobacter strains, D values of 0.71–0.78, 0.24–0.28 and 0.12–0.14 min ( z values 4.94 and 5.60°C) were recorded at 56, 58 and 60°C. D values obtained at 60°C by the test tube method were 2.7–5.0 min and were considered to be unrealistic.  相似文献

This study examined the attachment kinetics of Yersinia enterocolitica serotype O:3 to determine the optimum conditions for its isolation from meat enrichment systems using a novel surface adhesion technique. Minced beef was inoculated with Y. enterocolitica at an initial level of 10 cfu g−1 and incubated at 25 °C in an enrichment broth. Yersinia was recovered from enriched samples on polycarbonate membranes by surface adhesion and enumerated using immunofluorescence microscopy. The surface adhesion immunofluorescence technique (SAIF) had a minimum detection limit of approximately 4·0–4·5 log10 cfu ml−1 and provided good correlation between the estimation of the numbers of Yersinia in the enrichment broth derived from plate counts on Yersinia Selective agar (CIN) and those determined by SAIF ( r 2 = 0·94; rsd = ± 0·21). A derived regression equation of the SAIF count vs plate counts was used to predict Y. enterocolitica numbers in spiked meat samples stored at 0 °C for up to 20 d. The numbers as predicted by the SAIF method showed good correlation with counts derived by plating techniques ( r 2 = 0·78; rsd = ± 0·42). The application of the SAIF technique for the rapid detection of Y. enterocolitica serotype O:3 from meat is discussed.  相似文献

Expanded models for the non-thermal inactivation of Listeria monocytogenes   总被引:1,自引:0,他引:1  
Previously developed four-variable response surface models for describing the effects of temperature, pH/lactic acid, sodium chloride and sodium nitrite on the time to achieve a 4-log, non-thermal inactivation ( t 4D) of Listeria monocytogenes in aerobic, acidic environments were expanded to five-variable models that distinguish the effects of pH and acidulant concentration. A total of 18 new variable combinations were evaluated and the inactivation kinetics data appended onto a consolidation of two data sets from earlier studies. The consolidated data set, which included 315 inactivation curves representing 209 unique combinations of the five variables, was analysed by response surface analysis. The quadratic model without backward elimination regression was selected for further evaluation. Three additional quadratic models were generated using the concentrations of undissociated lactic and/or nitrous acids as variables in place of percentage lactic acid and sodium nitrite concentration. Comparison of predicted t 4D values against literature values for various food systems indicated that the models provide reasonable initial estimates of the inactivation of L. monocytogenes. The models based on the concentration of undissociated lactic and nitrous acids support the hypothesis that antimicrobial activity is associated with this form of the compounds. Evaluation of several examples suggests that these models may be useful for predicting the equivalent of the compounds'minimal inhibitory concentrations'for accelerating inactivation under various conditions.  相似文献
