The isolation of an extracellular polysaccharide from cultures of Chaetoceros affinis var. willei (Gran) Hustedt is described. The polysaccharide behaved as a homogeneous, polyanionic compound in free-boundary electrophoresis at both pH 2 and 7. It contained sulphur, presumably as sulphate half ester groups (8.7% of SO2Na), and the following monosaccharides were tentatively identified: rhamnose, fucose, arabinose, and galactose, with the two former constituting 63% of the polysaccharide preparation. The main cellular polysaccharide was a glucan and could be extracted from the cells by dilute acid. The remaining material gave, after hydrolysis, a complex mixture of monosaccharides with rhamnose as the major component. It is concluded that the extracellular polysaccharide is probably excreted from healthy cells.  相似文献

采用组织化学方法研究了六盘山鸡爪大黄茎和叶中大黄多糖贮藏和分布的特征。结果表明:大黄多糖在茎和叶内的贮藏部位各有特点,在茎中主要分布于皮层、维管束中韧皮部和木质部的薄壁细胞、髓射线和髓,随着茎的成熟大黄多糖有一定程度的增加;叶表皮、叶肉和叶脉中不同程度地分布着较少量的多糖;叶柄维管束外围的基本组织细胞中不同程度地分布着一定数量的大黄多糖,而维管束中分布较少。茎和叶中大黄多糖的贮藏和分布总体较少。  相似文献

Purified K1 polysaccharide enhanced the virulence of non-K1Escherichia coli species when given by orogastric feeding to neonatal rats. Neonatal rats developedE. coli bacteremia when K1 polysaccharide was given concomitantly with non-K1E. coli, whereasE. coli bacteremia did not develop when non-K1E. coli was given alone.E. coli K1 species did cause bacteremia and meningitis when fed to neonatal rats. The structure of the repeating unit of the capsular polysaccharide from Klebsiella type 41 has been investigated by methylation analysis of the original and the carboxyl-reduced polymer, uronic acid degradation, Smith degradation, and graded acid hydrolysis. Proton- and 13C-n.m.r. spectroscopy of the original polysaccharide and of the fragments obtained by various methods confirmed some structural features and allowed determination of the anomeric configuration of the glycosidic linkages. This polysaccharide is shown to have the following heptasaccharide repeating-unit:
This is the first polysaccharide antigen K of the Klebsiella series found to have seven sugar residues in its repeating unit, and to contain a galactose residue in its furanose form.  相似文献

A polysaccharide from the fungal pathogen Colletotrichum lindemuthianum causes browning and phytoalexin production when applied to the cut surfaces of bean (Phaseolus vulgaris) cotyledons and hypocotyls. The application of an amount of polysaccharide equivalent to less than 100 ng of glucose will elicit this response in the bean tissues. The polysaccharide has been isolated both from culture filtrates and from the mycelial walls of the fungus. Purification of the polysaccharide involved anion and cation exchange chromatography and gel filtration. The polysaccharide has an apparent molecular weight between 1,000,000 and 5,000,000 daltons, and consists predominantly of 3- and 4-linked glucosyl residues.  相似文献

An immunogenic polysaccharide prepared by phenol-water extraction of Trypanosoma cruzi was characterized by polyacrylamide gel electrophoresis and molecular sieve chromatography. The polysaccharide was shown to be a cell surface constituent by adsorption of rabbit anti-polysaccharide serum with live culture forms of the protozoa. The cell surface localization of the antigen was visualized using fluorescein- and ferritin-conjugated antibodies.  相似文献

CapR mutants of E. coli K-12 overproduce capsular polysaccharide (mucoid phenotype) and enzymes involved in capsular polysaccharide synthesis, and they are sensitive to radiation. It has been uncertain whether both properties are mediated by damage to a single cistron or by a polar effect on a second cistron in the same operon. Introduction of a polarity suppressor caused no change in the overproduction of polysaccharide, in the enzymes of polysaccharide synthesis or in radiation sensitivity of the capR mutant. Thus mucoidy and radiation sensitivity resulting from capR (lon) mutations are both the consequences of impairment of the same cistron. The experiments demonstrate the advantage of the use of polarity suppressors (over conventional nonsense suppressors) in determining whether pleiotropic effects of a mutation are the result of polarity.  相似文献

From leaves of Spinacia oleracea L. or from Pisum sativum L. and from cotyledons of germinating pea seeds a high molecular weight polysaccharide fraction was isolated. The apparent size of the fraction, as determined by gel filtration, was similar to that of dextran blue. Following acid hydrolysis the monomer content of the polysaccharide preparation was studied using high pressure liquid and thin layer chromatography. Glucose, galactose, arabinose, and ribose were the main monosaccharide compounds. The native polysaccharide preparation interacted strongly with the cytosolic isozyme of phosphorylase (EC 2.4.1.1). Interaction with the plastidic phosphorylase isozyme(s) was by far weaker. Interaction with the cytosolic isozyme was demonstrated by affinity electrophoresis, kinetic measurements, and by 14C-labeling experiments in which the glucosyl transfer from [14C]glucose 1-phosphate to the polysaccharide preparation was monitored.  相似文献

Jelly coat, a multicomponent extracellular matrix surrounding the sea urchin egg, induces the acrosome reaction in sperm. The jelly coats of the four species studied, Arbacia punctulata, Strongylocentrotus purpuratus, Strongylocentrotus drobachiensis, and Lytechinus variegatus, were found to be very similar in chemical composition. A sialoprotein (approximately 20% of the mass of the jelly coat) and a fucose sulfate polysaccharide (approximately 80%) are the major macromolecular components of the jelly coat. The acrosome reaction inducing capacity resides solely in the fucose sulfate polysaccharide. Induction of the acrosome reaction ranges from highly species specific to nonspecific. Thus, A. punctulata and S. drobachiensis sperm are induced to undergo the acrosome reaction only with their homologous jelly coat, while S. purpuratus sperm react equally well with homologous or L. variegatus jelly coat, but not with A. punctulata jelly coat. L. variegatus sperm seem to be relatively nonspecific in response. Species-specific induction of the acrosome reaction resides solely in the fucose sulfate polysaccharide, suggesting that there must be structural differences in this polysaccharide in the various species. Therefore, in some species, fertilization appears to involve sperm-egg recognition at the level of the jelly coat as well as at the level of sperm-egg receptors.  相似文献

The pneumococcal serotype 14 polysaccharide was produced in Lactococcus lactis by coexpressing pneumococcal polysaccharide type 14-specific genes (cpsFGHIJKL14) with the lactococcal regulatory and priming glucosyltransferase-encoding genes specific for B40 polysaccharide (epsABCDB40). The polysaccharide produced by Lactococcus was secreted in the medium, simplifying downstream processing and polysaccharide isolation from culture broth.  相似文献

The three intra- and extracellular polysaccharide fractions were isolated during the growth phase of Dictyostelium discoideum NC-4, and the change in content of component sugars of four fractions during the culture period was examined. Myxamoebae most extensively contain a polysaccharide fraction extracted with phenol-water (polysaccharide fraction I) in a quantity of about 15–23% per dry cell. After 15 h the uronic acid formed in the polysaccharide fraction I, and the cell, could be aggregated. The glucosamine content in the polysaccharide fraction I reached a maximum as the myxamoebae entered the exponential phase, and a large amount of galactose was produced as the cell entered the stationary phase. The phenol-water extract from the cells of the stationary phase was reacted with concanavalin-A.  相似文献

Fibroblasts from cornea, heart, and skin of day 14 embryonic chicks demonstrate the ability to make heparan sulfate-like polysaccharide when examined during the 10 hr period immediately following their removal from the embryo. Both the whole tissues from which these fibroblasts are isolated and the fibroblasts grown for 2–5 weeks in vitro also synthesize heparan sulfate. During their first few days in vitro, the three fibroblast populations display increasing rates of [35S]-sulfate and d-[1-3H]-Glucosamine incorporation into glycosaminoglycans and sharp fluctuations of those rates, yet the percentage of total [35S]-sulfate incorporated into heparan sulfate-like polysaccharide and the distribution of this polysaccharide between cells and nutrient medium do not change significantly. During their first 48 hr in vitro, skin fibroblasts, but not those from cornea or heart, show steadily decreasing discrepancies between the proportions of [35S]-sulfate and d-[1-3H]-Glucosamine incorporated into heparan sulfate, suggesting a sharp decline in the synthesis of nonsulfated glycosaminoglycans. These data support the hypothesis of Kraemer than many cell-types in vivo may normally make heparan sulfate. The data largely eliminate the hypothesis that the biosynthesis of this polysaccharide is selectively stimulated as embryonic cells adapt to growth in vitro.  相似文献

The seeds of Anthocephalus indicus contain a water-soluble polysaccharide composed of D-xylose, D-mannose, and D-glucose in the molar ratios 1:3:5. Methylation analysis afforded 2,3,4-tri-O-methyl-D-xylose, 2,3,6,-tri-O-methyl-D-mannose, 2,3,6-tri-O-methyl-D-glucose, 2,3-di-O-methyl-D-glucose, and 2,3,4,6-tetra-O-methyl-D-glucose in the molar ratios 7:21:12:15:8. Periodate oxidation and methylation data indicated 22.5% and 21.9% of end groups, respectively. The above findings, together with the results of partial hydrolysis with acid, indicate the polysaccharide to consist of a linear chain of (1→4)-linked β-D-mannosyl and β-D-glucosyl residues to which α-D-xylosyl and β-D-glucosyl groups are attached by (1→6)-linkages.  相似文献

Caulobacter crescentus cells adhere to surfaces by using an extremely strong polar adhesin called the holdfast. The polysaccharide component of the holdfast is comprised in part of oligomers of N-acetylglucosamine. The genes involved in the export of the holdfast polysaccharide and the anchoring of the holdfast to the cell were previously discovered. In this study, we identified a cluster of polysaccharide biosynthesis genes (hfsEFGH) directly adjacent to the holdfast polysaccharide export genes. Sequence analysis indicated that these genes are involved in the biosynthesis of the minimum repeat unit of the holdfast polysaccharide. HfsE is predicted to be a UDP-sugar lipid-carrier transferase, the glycosyltransferase that catalyzes the first step in polysaccharide biosynthesis. HfsF is predicted to be a flippase, HfsG is a glycosyltransferase, and HfsH is similar to a polysaccharide (chitin) deacetylase. In-frame hfsG and hfsH deletion mutants resulted in severe deficiencies both in surface adhesion and in binding to the holdfast-specific lectin wheat germ agglutinin. In contrast, hfsE and hfsF mutants exhibited nearly wild-type levels of adhesion and holdfast synthesis. We identified three paralogs to hfsE, two of which are redundant to hfsE for holdfast synthesis. We also identified a redundant paralog to the hfsC gene, encoding the putative polysaccharide polymerase, and present evidence that the hfsE and hfsC paralogs, together with the hfs genes, are absolutely required for proper holdfast synthesis.  相似文献

The capsular polysaccharide of Diplococcus pneumoniae Type XII contains residues of d-glucose and d-galactose in a molar ratio of 2:1. The methylated polysaccharide yielded upon hydrolysis 2,3,4,6-tetra- and 3,4,6-tri-O-methyl-d-glucose and 2,3,4,6-tetra-O-methyl-d-galactose as the only neutral methyl sugars. Periodate oxidation of the polysaccharide resulted in destruction of all neutral sugars and immunochemical activity against rabbit antisera. Periodate oxidation of the methyl O-methylglycosides obtained after hydrolysis of the methylated polysaccharide indicated that at least 30% of the l-fucosamine residues are substituted at C-4 in the polysaccharide. It is concluded that the polysaccharide consists of a hexosamine backbone that is substituted by d-galactosyl and kojibiosyl side-chains. The proposed terminal d-galactosyl residues apparently are sterically hindered from interacting with several d-galactose-binding proteins.  相似文献

Treatment of the O-specific polysaccharide from Shigella dysenteriae Type 3 with hydrazine in the presence of hydrazine sulphate resulted in quantitative N-deacetylation with the formation of a modified polysaccharide containing free amino groups. Oxidation of the modified polysaccharide with periodate did not destroy the 2-amino-2-deoxygalactose residues, thus indicating that they were substituted at position 3. Acid hydrolysis of the modified polysaccharide afforded 3-O-(2-amino-2-deoxy-β-D-galactopyranosyl)-D-galactose, which was identified as the N-acetyl derivative. Deamination of the modified polysaccharide with nitrous acid cleaved the 2-amino-2-deoxy-D-galactopyranosyl linkages to give a pentasaccharide as the major product, which appeared to be the modified chemical repeating unit of the O-specific polysaccharide.  相似文献

An arabinogalactan isolated from the wood of Larix sibirica L. was investigated by methylation analysis, partial hydrolysis, enzymic oxidation, and 13C-n.m.r. spectroscopy. The structural conclusions arrived at by 13C-n.m.r. spectroscopy were consistent with the data from methylation analysis. The polysaccharide is highly branched and similar in structure to those of arabino-3,6-galactans isolated from other Larix species.  相似文献

Several benthic cyanobacteria were found to produce significant amounts of extracellular flocculants. The macromolecular flocculants produced by Phormidium sp. strain J-1 and Anabaenopsis circularis PCC 6720 were characterized. The Phormidium flocculant is a sulfated heteropolysaccharide to which fatty acids and protein are bound. The polysaccharide backbone is composed of uronic acids, rhamnose, mannose, and galactose. The A. circularis flocculant is also an acidic polysaccharide containing keto acid residues and neutral sugars, but to which no fatty acids, proteins, or sulfates are linked. Both flocculants could be recovered from growth medium by precipitation with cetyltrimethylammonium bromide and were found to bind the cationic dye Alcian-blue in a linear proportion to their concentration in solution. The latter property was used to quantify flocculant concentrations in culture supernatants and natural water samples and to compute their anion densities.  相似文献

Sulfated polysaccharide isolated from tetrasporic plants of Tichocarpus crinitus was investigated. The polysaccharide was isolated by two methods: with water extraction at 80 °C (HT) and with a mild alkaline extraction (AE). The extracted polysaccharides were presented by non-gelling ones only, while galactose and 3,6-AG were the main monosaccharides, at the same time amount of 3,6-AG in AE polysaccharides was the similar to that of HT. According to methods of spectroscopy and mass spectrometry, the polysaccharide from tetrasporic T. crinitus contains main blocks of 1,3-linked β-d-galactopyranosyl-2,4-disulfates and 1,4-linked 3,6-anhydro-α-d-galactopyranosyl while 6-sulfated 4-linked galactopyranosyl resudies are randomly distributed along the polysaccharide chain. The alkaline treatment of HT polysaccharide results in obtaining polysaccharide with regular structure that composed of alternating 1,3-linked β-d-galactopyranosyl-2,4-disulfates and 1,4-linked 3,6-anhydro-α-d-galactopyranosyl residues. Native polysaccharide (HT) possessed both high anticoagulant and antiplatelet activity measured by fibrin clotting and platelet aggregation induced by collagen. This activity could be connected with peculiar chemical structure of HT polysaccharide which has high sulfation degree and contains also 3,6-anhydrogalactose in the polymer chain.  相似文献

Evidence for immunoadjuvant activity of the Helianthus tuberosus L. polysaccharide was obtained in an antibody-producing cell model. Dectin-1 and TLR-6 were identified as major receptors required for biological activity of polysaccharide in a TNF-α stimulation model. The CR3 receptor was not implicated in polysaccharide recognition in the same model. Enzyme treatment of the H. tuberosus L. polysaccharide demonstrated the presence of ß-(1 → 4) and ß-(1 → 3) glycosidic bonds.  相似文献
