An actinomycete strain, designated YIM 60475T, was isolated from the roots of Maytenus austroyunnanensis and was characterized by using a polyphasic approach. The strain was determined to belong to the genus Streptomyces, based on its phenotypic and phylogenetic characteristics. The strain produced spiral spore chains on aerial mycelium. The cell wall contained ll-diaminopimelic acid. Whole-cell hydrolysates contained galactose, glucose, and xylose. The phospholipid was type II. The DNA G+C content of the type strain was 73.3 mol%. DNA–DNA hybridization and comparison of physiological and chemical characteristics suggested that strain YIM 60475T is a new Streptomyces species, for which the name Streptomyces mayteni sp. nov. is proposed. An actinomycete strain, which was designated 172115T, was isolated from mangrove soil in Shenzhen, China. Strain 172115T fell within the genus Streptomyces in the 16S rRNA gene tree and could be grouped into this genus based on its chemotaxonomic and morphological data. The strain shared the highest 16S rRNA gene sequence similarities with Streptomyces lanatus NBRC 12787T (AB184845) (98.29%) and Streptomyces lucensis NBRC 13056T (AB184280) (98.26%). The DNA–DNA hybridization values between strain 172115T and the two most closely related type strains were low enough to justify the assignment of the strain to a novel species. On the basis of these phenotypic, phylogenetic and chemotaxonomic characteristics, 172115T represents a novel species of the genus Streptomyces, for which, the name Streptomyces shenzhenensis sp. nov. is proposed for strain 172115T (=CCTCC AA 2011001T=DSM 42034T).  相似文献

Nine strains isolated from mycetoma patients and received as Streptomyces somaliensis were the subject of a polyphasic taxonomic study. The organisms shared chemical markers consistent with their classification in the genus Streptomyces and formed two distinct monophyletic subclades in the Streptomyces 16S rRNA gene tree. The first subclade contained four organisms, including the type strain of S. somaliensis, and the second clade the remaining five strains which had almost identical 16S rRNA sequences. Members of the two subclades were sharply separated using DNA:DNA relatedness and phenotypic data which also showed that the subclade 1 strains formed an heterogeneous group. In contrast, the subclade 2 strains were assigned to a single genomic species and had identical phenotypic profiles. It is evident from these data that the subclade 2 strains should be recognised as a new species of Streptomyces. The name proposed for this new species is Streptomyces sudanensis sp. nov. A novel actinomycete, designated strain KLBMP 4601T, was isolated from the root of the medicinal plant Curcuma phaeocaulis collected from Sichuan Province, south-west China. The strain produced extensively branched substrate and aerial hyphae that carried straight to flexuous spore chains. Chemotaxonomic properties of this strain were consistent with those of members of the genus Streptomyces. The cell wall of strain KLBMP 4601T contained ll-diaminopimelic acid as the characteristic diamino acid. The major menaquinone was MK-9(H4), with minor amounts of MK-9(H6), MK-9(H8) and MK-10(H2). The major fatty acids were C16:0, iso-C16:0, C18:1ω9c and C16:1, iso G. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain KLBMP 4601T belongs to the genus Streptomyces and is most closely related to Streptomyces armeniacus JCM 3070T (97.9 %), Streptomyces pharmamarensis PM267T (97.6 %) and Streptomyces artemisiae YIM 63135T (97.5 %). The 16S rRNA gene sequence similarity between strain KLBMP 4601T and other members of this genus were lower than 97.5 %. DNA–DNA hybridization studies of strain KLBMP 4601T with the three closest species showed relatedness values of 36.3 ± 4.2 %, 27.3 ± 0.6 %, and 30.9 ± 2.5 %, respectively. On the basis of chemotaxonomic, phenotypic and genotypic characteristics, it is evident that strain KLBMP 4601T represents a novel species of the genus Streptomyces, for which the name Streptomyces phytohabitans sp. nov. is proposed. A strain YIM 33098T (= CCTCC AA001027T = DSM 41831T) was isolated from a forest soil sample collected from Nanning in Guangxi Province, China, in the course of screening for producers of new drug lead compounds. This strain was identified by using a polyphasic approach. The results showed that it should be assigned to the genus Streptomyces. An almost complete 16S rRNA gene sequence of the strain was determined and compared with those of representative Streptomyces species. Strain YIM 33098T was clustered in the same subclade with Streptomyces tendae ATCC19812T and Streptomyces eurythermus ATCC14975T. Similarities of strain YIM 33098T with the two strains were 97.35% and 97.42%, respectively. Based on the phenotypic and genotypic evidence, it is therefore proposed that strain YIM 33098T should be classified in the genus Streptomyces as a new species under the name of Streptomyces nanningensis sp. nov.  相似文献

A novel Gram-positive, halotolerant, non-sporulating, non-motile, catalase-positive, oxidase-negative and aerobic bacterium, designated strain JSM 078085T, was isolated from sea water collected from the South China Sea. Strain JSM 078085T exhibited a rod-coccus growth cycle and produced a yellow pigment. The strain was able to grow in the presence of 0–12% (w/v) NaCl and at pH 6.0–9.5 and 4–35°C; optimum growth was observed at pH 7.0 and 25–30°C in the absence of NaCl. The peptidoglycan type was A4α (l-Lys–l-Ala–l-Glu). Cell-wall sugars contained galactose and glucose. Strain JSM 078085T contained menaquinone MK-9(H2) as the major respiratory quinone and diphosphatidylglycerol, phosphatidylglycerol and phosphatidylinositol as the major polar lipids. The major cellular fatty acids were anteiso-C15:0, iso-C15:0 and anteiso-C17:0 and the DNA G + C content was 63.3 mol%. Phylogenetic analysis based on 16S rRNA gene sequence comparisons revealed that strain JSM 078085T should be assigned to the genus Arthrobacter, being most closely related to the type strain of Arthrobacter rhombi (sequence similarity 97.1%), and the two strains formed a distinct lineage in the phylogenetic tree. The level of DNA–DNA relatedness between strain JSM 078085T and the type strain of Arthrobacter rhombi was 10.6%. The combination of phylogenetic analysis, DNA–DNA relatedness, phenotypic characteristics and chemotaxonomic data supported the view that strain JSM 078085T represents a novel species of the genus Arthrobacter, for which the name Arthrobacter halodurans sp. nov. is proposed. The type strain is JSM 078085T (=DSM 21081T=KCTC 19430T). The GenBank/EMBL/DDBJ accession number for the 16S rRNA gene sequence of strain JSM 078085T is EU583729.  相似文献

A novel Gram-stain-positive, slightly halophilic, facultatively alkaliphilic, non-motile, non-sporulating, catalase-positive, oxidase-negative, aerobic bacterium, designated strain JSM 070026T, was isolated from non-saline forest soil in China. Growth occurred with 0–20% (w/v) NaCl (optimum, 2–4%) and at pH 6.0–10.5 (optimum, pH 8.0) and 5–40°C (optimum, 30°C). Good growth also occurred in the presence of 0–28% (w/v) KCl (optimum, 2–5%) or 0–25% (w/v) MgCl2·6H2O (optimum, 1–4%). The peptidoglycan type was A4α (l-Lys–Gly–l-Glu). Cell-wall sugars contained mannose and xylose. The major cellular fatty acids were anteiso-C15:0 and iso-C15:0. Strain JSM 070026T contained menaquinone 8 as the major respiratory quinone and diphosphatidylglycerol, phosphatidylglycerol and phosphatidylinositol as the major polar lipids. The DNA G + C content of strain JSM 070026T was 56.7 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain JSM 070026T was a member of the suborder Micrococcineae and most closely related to Yaniella flava YIM 70178T (sequence similarity 99.4%) and Yaniella halotolerans YIM 70085T (97.9%). The three strains formed a distinct branch in the phylogenetic tree. The combination of phylogenetic analysis, DNA–DNA relatedness values, phenotypic characteristics and chemotaxonomic data supports the proposal that strain JSM 070026T represents a novel species of the genus Yaniella, for which the name Yaniella soli sp. nov. is proposed. We previously isolated Streptomyces racemochromogenes strain 10-3, which produces a phospholipase D (PLD) with high transphosphatidylation activity. Here, we purified and cloned the PLD (PLD103) from the strain. PLD103 exerted the highest hydrolytic activity at a slightly alkaline pH, which is in contrast to the majority of known Streptomyces PLDs that have a slightly acidic optimum pH. PLD103 shares only 71–76% amino acid sequence identity with other Streptomyces PLDs that have a slightly acidic optimum pH; thus, the diversity in the primary structure might explain the discrepancy observed in the optimum pH. The purified PLD displayed high transphosphatidylation activity in the presence of glycerol, l-serine, and 2-aminoethanol hydrochloride with a conversion rate of 82–97% in a simple one-phase system, which was comparable to the rate of other Streptomyces PLDs in a complicated biphasic system.  相似文献

A novel actinomycete strain, GW25-5T, was isolated from a soil sample collected from the Fildes Peninsula, King George Island, West Antarctica. The strain was characterized by white to grey aerial mycelia, which were differentiated to straight to flexuous spore chains, with rod-shaped smooth spores. The cell wall of strain GW25-5T contained LL-diaminopimelic acid (A2pm) and traces of meso-A2pm. Whole-cell sugars were galactose and minor amounts of mannose and glucose. The predominant menaquinones were MK-9(H6) (49%), MK-9(H8) (24%) and MK-9(H4) (12%). The phospholipids contained DPG, PE, PI, PIM and PL(s). The major cellular fatty acids were iso-C16:0 and anteiso-C15:0. Genomic DNA G+C content of strain GW25-5T was 70.0 mol%. BLAST result showed that strain GW25-5 has the 16S rRNA gene sequence highest similarity of 97.5% with members of genus Streptomyces and phylogenetic analysis indicated that this strain belongs to the genus Streptomyces. DNA–DNA relatedness values of strain GW25-5T with the closest species of Streptomyces purpureus LMG 19368T and Streptomyces beijiangensis YIM 6T were significantly lower than 70% of the threshold value for the delineation of genomic species. A polyphasic taxonomic investigation based on a judicious combination of genotypic and phenotypic characteristics revealed that the organism represents a novel species of the genus Streptomyces. Thus, we propose strain GW25-5T as the type strain of this novel species, Streptomyces fildesensis (=CGMCC 4.5735T = YIM 93602T = DSM 41987T = NRRL B 24828T).  相似文献

A novel actinomycete strain, designated YIM 45720T, was isolated from a Cephalotaxus fortunei rhizophere soil sample collected from Yunnan Province, southwest China. The strain formed well-differentiated aerial and substrate mycelia. Chemotaxonomically, it contained LL-diaminopimelic acid in the cell wall. The cell-wall sugars contained ribose, mannose, and galactose with traces of glucose and xylose. Phospholipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine and phosphatidylinositol. MK-9 (H8) was the predominant menaquinone. The major fatty acids (>10%) were iso-C16:0, iso-C15:1 and anteiso-C15:0. The G + C content of the DNA was 70 mol%. Phylogenetic analysis data based on 16S rRNA gene sequence showed that strain YIM 45720T formed a distinct branch with the type strain of Streptomyces scabrisporus JCM 11712T within the genus Streptomyces. On the basis of the phenotypic and genotypic characteristics, strain YIM 45720T (=DSM 41883T = CCTCC AA 206006T) is proposed as the type strain of a novel species, Streptomyces serianimatus sp. nov.  相似文献

A novel moderately halophilic, alkaliphilic, non-motile, non-sporulating, catalase-positive, oxidase-negative, aerobic, coccus-shaped, Gram-positive bacterium, designated strain JSM 071043T, was isolated from a subterranean brine sample collected from a salt mine in Hunan Province, China. Growth occurred with 0.5–20% (w/v) NaCl (optimum 5–10%) at pH 6.5–10.5 (optimum pH 8.5) and at 10–40°C (optimum 25–30°C). Good growth also occurred in the presence of 0.5–20% (w/v) KCl (optimum 5–8%) or 0.5–25% (w/v) MgCl2·6H2O (optimum 5–10%). The peptidoglycan type was A4α (l-Lys–l-Ala–l-Glu) and major cell-wall sugars were tyvelose and mannose. The major cellular fatty acids were anteiso-C15:0, iso-C16:0 and anteiso-C17:0. Strain JSM 071043T contained MK-9 and MK-8 as the predominant menaquinones and diphosphatidylglycerol, phosphatidylglycerol and phosphatidylinositol as the major polar lipids. The DNA G + C content was 67.8 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain JSM 071043T was a member of the suborder Micrococcineae, and was most closely related to Zhihengliuella halotolerans YIM 70185T (sequence similarity 98.9%) and Zhihengliuella alba YIM 90734T (98.2%), and the three strains formed a distinct branch in the phylogenetic tree. The combination of phylogenetic analysis, DNA–DNA relatedness values, phenotypic characteristics and chemotaxonomic data supports the proposal that strain JSM 071043T represents a novel species of the genus Zhihengliuella, for which the name Z. salsuginis sp. nov. is proposed. Two novel Gram-stain positive, spore-forming, aerobic actinomycetes, designated NEAU-PCY-1T and NEAU-PCY-2, were isolated from rhizosphere soil of Urtica urens L. collected from Anshan, Liaoning Province, northeast China. The 16S rRNA gene sequence analysis showed that strains NEAU-PCY-1T and NEAU-PCY-2 exhibited 99.8% similarity with each other and are closely related to Streptomyces abietis DSM 42080T (98.2, 98.3%) and Streptomyces fildesensis DSM 41987T (98.0, 98.1%). Phylogenetic analysis based on the 16S rRNA gene sequences indicated that the two strains formed a cluster with these two closely related species. Moreover, DNA–DNA hybridization results and some phenotypic, physiological and biochemical properties differentiated the two strains from their close relatives in the genus Streptomyces. Based on a polyphasic taxonomy study, strains NEAU-PCY-1T and NEAU-PCY-2 are considered to represent a novel species of the genus Streptomyces, for which the name Streptomyces urticae sp. nov. is proposed, with NEAU-PCY-1T (=?DSM 105115T?=?CCTCC AA 2017015T) as the type strain.  相似文献

A new actinobacterium strain, designated 2AT, was isolated from a soil sample collected from Guru Nanak Dev University, Punjab (India) and characterized using a polyphasic taxonomic approach. It showed antimicrobial activity against various Gram-positive and Gram-negative bacteria including drug resistant bacteria and fungi. The strain had chemotaxononomic and morphological properties typical of the genus Streptomyces. The 16S rRNA gene sequence of the strain showed 99.9, 99.5 and 99.5 % similarity with Streptomyces flavotricini DSM 40152T, Streptomyces toxytricini DSM 40178T and Streptomyces globosus DSM 40815T, respectively. This strain formed a coherent cluster with them and shared DNA–DNA homology of 37.6 ± 0.6, 34.4 ± 0.5 and 33.1 ± 0.4 % with type strains, S. flavotricini DSM 40152T, S. globosus DSM 40815T and S. toxytricini DSM 40178T, respectively. Further, the strain was readily distinguished from the phylogenetic close relatives in a variety of morphological, physiological and biochemical properties. Based on the genotypic and phenotypic characteristics, it is proposed that strain 2AT represents a novel species in the genus Streptomyces, for which the name Streptomyces amritsarensis sp. nov. is proposed, with the type strain 2AT (=MTCC 11845T=JCM 19660T).  相似文献

A novel haloalkaliphilic, facultative anaerobic and Gram-negative Salinivibrio-like microorganism (designated strain BAGT) was recovered from a saline lake in Ras Mohammed Park (Egypt). Cells were motile, curved rods, not spore-forming and occurred singly. Strain BAGT grew optimally at 35°C (temperature growth range 25–40°C) with 10.0% (w/v) NaCl [NaCl growth range 6.0–16.0% (w/v)] and at pH 9.0 (pH growth range 6.0–10.0). Strain BAGT had phosphatidylethanolamine (PEA) and phosphatidylglycerol (PG) as the main polar lipids, C16:0 (54.0%) and C16:1 (26.0%) as the predominant cellular fatty acids and Q-8 as the major respiratory quinone. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain BAGT was a member of Salinivibrio genus, with the highest sequence similarities of 99.1, 98.4 and 98.1% to Salinivibrio siamensis JCM 14472T, Salinivibrio proteolyticus DSM 19052T and Salinivibrio costicola subsp. alcaliphilus DSM 16359T, respectively. DNA–DNA hybridization values of strain BAGT with members of Salinivibrio genus were lower than 55.0%. DNA G + C content was 51.0 mol%. On the basis of the polyphasic taxonomic results revealed in this study, strain BAGT should be classified as a novel species of Salinivibrio genus, for which the name Salinivibrio sharmensis sp. nov. is proposed, with the type strain BAGT (=ATCC BAA-1319T = DSM 18182T).  相似文献

A Gram-stain-negative, non-motile, rod-shaped bacterial strain, JW-64-1T, capable of degrading methamidophos was isolated from a methamidophos-manufacturing factory in China, and was subjected to a polyphasic taxonomic investigation. Strain JW-64-1T produced circular, smooth, transparent, yellow-colored colonies (1.0–2.0 mm) on LB agar after 2 days incubation. It grew optimally at 25–30°C and pH 7.0 without the presence of NaCl. The G+C content of the total DNA was 63.6 mol%. A phylogenetic analysis based on 16S rRNA gene sequences showed that strain JW-64-1T fell within the cluster comprising Luteibacter species. The 16S rRNA gene sequence of strain JW-64-1T was most closely related to Luteibacter rhizovicinus DSM 16549T (98.6%), followed by Luteibacter yeojuensis DSM 17673T (98.4%) and L. anthropi CCUG 25036T (98.2%). The major cellular fatty acids of strain JW-64-1T were iso-C15:0 (24.1%), iso-C17:0 (20.2%) and summed feature 9 comprising iso-C17:1 ω9c and/or C16:0 10-methyl (20.3%). The major isoprenoid quinine was Q-8 (98%), and the major polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphoaminolipid, aminolipids-1, aminolipids-2, and phospholipids. The values for DNA–DNA relatedness between strain JW-64-1T and the closest phylogenetic relatives of L. rhizovicinus and Luteibacter yeojuensis were 34.8 ± 2.6 and 25.6 ± 3.1%, respectively. On the basis of the phenotypic, chemotaxonomic, DNA–DNA relatedness and phylogenetic analysis based on the 16S rRNA gene sequences, strain JW-64-1T represents a novel species of the genus Luteibacter, for which the name Luteibacter jiangsuensis sp. nov. is proposed. A Streptomyces-like actinomycete strain, designated as YIM 78087T, was isolated from a sediment sample collected from Hehua hot spring in Tengchong, Yunnan province, south-west China. The taxonomic position of strain YIM 78087T was investigated by a polyphasic approach. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain YIM 78087T belongs to the genus Streptomyces and is closely related to Streptomyces fimbriatus DSM 40942T, Streptomyces marinus DSM 41968T and Streptomyces qinglanensis DSM 42035T (97.18, 97.05 and 97.1 % similarity, respectively). Combined with the low values of DNA–DNA hybridization between strain YIM 78087T and its closest neighbours, these analyses indicated that this new isolate represents a different genomic species in the genus Streptomyces. The predominant menaquinones of strain YIM 78087T were identified as MK-9 (H4) and MK-9 (H6). The major fatty acids were identified as anteiso-C15:0 (28.4 %), anteiso-C17:0 (23.0 %) and iso-C16:0 (15.1 %). The whole-cell hydrolysates found to contain glucose, mannose and ribose. The DNA G+C content was determined to be 73.0 mol%. Based on the comparative analysis of phenotypic and genotypic characteristics, it is proposed that strain YIM 78087T represents a novel species of the genus Streptomyces, for which the name Streptomyces calidiresistens sp. nov., is proposed. A novel actinomycete, designated strain NEAU-LA29T, was isolated from soil collected from Xianglu Mountain and subjected to a polyphasic taxonomic study. Based on a polyphasic taxonomic approach comprising chemotaxonomic, phylogenetic, morphological and physiological characterisation, the isolate has been affiliated to the genus Streptomyces. 16S rRNA gene sequence analysis showed that the isolate is closely related to Streptomyces vastus JCM4524T (98.8% identity) and Streptomyces cinereus DSM43033T (97.9%). However, multilocus sequence analysis based on five other house-keeping genes (atpD, gyrB, rpoB, recA and trpB) and low DNA–DNA relatedness values enabled the strain to be differentiated from these closely related species of the genus Streptomyces. Thus, strain NEAU-LA29T is concluded to represent a novel species of the genus Streptomyces, for which the name Streptomyces xiangluensis sp. nov. is proposed. A novel actinomycete strain YIM 31724T was isolated from a soil sample collected from Dali, Yunnan Province, People’s Republic of China. The strain is characterized by white to yellow white aerial mycelia, spiral spore chains and smooth spore surface. The cell wall of strain YIM 31724T contained LL-diaminopimelic acid (A2pm) and traces of meso-A2pm. Whole-cell hydrolysates contained mainly glucose and small amounts of galactose and xylose. The menaquinones were MK-9(H6) (31%) and MK-9(H8) (69%). Phosphatidylethanolamine was the diagnostic phospholipid. The DNA G+C content of strain YIM 31724T was 67.2 mol%. Phylogenetic analysis indicated that the strain belongs to the genus Streptomyces, with highest similarity to Streptomyces rimosus subsp. rimosus JCM 4667T (rRNA gene sequence similarity value of 98.9%) and Streptomyces erumpens DSM 40941T (rRNA gene sequence similarity value of 98.7%). Based on its phenotypic and genotypic characteristics, including low DNA–DNA hybridization results, strain is proposed as the type strain of a novel species, Streptomyces daliensis sp. nov.  相似文献

Two moderately halophilic, Gram-negative, rod-shaped bacteria, designated YIM 93003T and YIM 94343T, were isolated from a salt lake in Xinjiang province, north-west China. The two strains YIM 93003T and YIM 94343T grew at 20–40°C, pH 6–9, 0.5–24% (w/v) NaCl and at 20–40°C, pH 6–9, 0.5–23% (w/v) NaCl, respectively. No growth occurred in absence of NaCl. Phylogenetic analyses based on 16S rRNA gene sequences showed that strains YIM 93003T and YIM 94343T were phylogenetically affiliated to the genus Halomonas and exhibited sequence similarity of 97.5% and 97.4% to the type strain Halomonas anticariensis DSM 16096T, respectively. The strains possessed chemotaxonomic markers that were consistent with their classification in the genus Halomonas (Q-9 as predominant respiratory quinine; C18:1ω7c, C16:0 and C16:1 ω7c/iso-C15:02-OH as the major fatty acids). The DNA–DNA hybridization values for strains YIM 93003T and YIM 94343T, YIM 93003T and DSM 16096T, YIM 94343T and DSM 16096T were 38.1 ± 3.0, 18.3 ± 4.7, and 20.8 ± 4.6%, respectively. The G+C contents of the strains YIM 93003T and YIM 94343T were 63.4 and 64.0 mol%, respectively. Based on comparative analysis of physiological, biochemical and chemotaxonomic data, including low DNA–DNA hybridization results, two novel species, Halomonas qijiaojingensis sp. nov., and Halomonas flava sp. nov., are proposed. The type strains are YIM 93003T (=CCTCC AB 208133T =KCTC 22228T) and YIM 94343T (=CCTCC AB 2010382T =KCTC 23356T), respectively.  相似文献
