This work aims at breeding upland cotton [Gossypium hirsutum L., 2(AD)1 genome] with a reduced level of gossypol in the seeds for optimal food and feed uses, and a high gossypol level in the remaining organs for resistance to pests. Two tri-species Gossypium hybrids, (G. thurberiG. sturtianumG. hirsutum and G. hirsutumG. raimondiiG. sturtianum) including G. sturtianum (2C1) as a donor, G. thurberi (2D1) and G. raimondii (2D5) as a bridge species, were created. Recurrent selection initiated with these tri-species hybrids produced backcross (BC) progenies expressing the ”low-gossypol seed and high-gossypol plant” trait at different levels. We used AFLP markers to assess the genetic similarity among the germplasm and RFLP probes to tag the introgression of specific chomosome segments from the parental species. Five pairs of AFLP primers generated 477 fragments, among which 417 (87.4%) were polymorphic. The genetic similarity between the upland cotton and the wild species ranged from 29.5 to 43.2%, while similarity reached 80% between upland cotton and BC3 plants. Introgression of species-specific AFLPs was evident from all the parental species and confirmed the hybrid origin of the analyzed progenies. Southern-blot analysis based on 49 RFLP probes allowed us to trace the introgression of parental DNA segments in the tri-species hybrids and in three generations of backcross. Introgression was evident from 11, 8 and 7 linkage groups of G. sturtianum, G. raimondii and G. thurberi respectively. The types of introgression revealed by RFLP probes are discussed, and breeding schemes to enhance recombination are proposed. The ability to trace DNA segments of known chromosomal locations from the donor G. sturtianum through segregating generations is a starting point to map the ”low-gossypol seed and high-gossypol plant” traits.  Chickpea (Cicer arietinum L.) ranks third in the world, and first in the Mediterranean basin, for production among pulses. Despite its importance as a crop and considerable research effort, traditional breeding methods have so far been unable to produce cultivars with a large impact on chickpea production. Interspecific hybridization is known to improve yield in many crops. Therefore, an attempt was made to increase the seed yield in chickpea through the introgression of genes from wild relatives at the International Center for Agricultural Research in the Dry Areas (ICARDA), Syria, from 1987 to 1995. Four crosses, ILC 482 (C. arietinum)×ILWC 179 (C. echinospermum) and ILC 482×ILWC 124 (C. reticulatum) and their reciprocals, were made. Pedigree selection was used to advance the material. Heterosis was recorded visually in F1s, and single plant measurements for seed yield were recorded in F2 populations. Promising and uniform progenies were bulked in the F5 generation. Out of 96 F6 lines, 22 were selected on the basis of seed yield and other agronomic characters, and evaluated in a replicated trial for seed yield and 14 agronomical, morphological and quality characters. A high level of heterosis was observed in F1s. Several F2 plants produced two to three times more seed yield than the best plant from the cultigen. Nine F7 lines out-yielded the cultigen parent by up to 39%. Over 2 years, 12 lines had a higher yield than the cultigen parent. These lines were not only high yielding but also free of any known undesirable traits from the wild species, such as spreading growth habit, pod dehiscence, and non-uniform maturity. Quality traits, such as seed shape, type, colour, weight, and testa texture, protein content, cooking time and an organoleptic test of a Middle East dish, Homos Bi-Tehineh, were also similar to the cultigen parent.  In the evolutionary process of an animal lineage, interactions in secondary contacts of differentiated populations and introgressive hybridization may play an important role. In the Japanese islands, the carabid subgenus Ohomopterus (genus Carabus) exhibits a marked differentiation in body size and genital morphology. Although geographical differentiation is apparent, two or three species usually coexist at many localities. Their reproductive isolation relies on body size differences, chemical cues for mate recognition, and a species-specific genital lock-and-key system. However, these isolation mechanisms are not always effective enough to prevent interspecific hybridization. An initial assessment of the species-level phylogeny with mitochondrial gene sequences revealed that the gene genealogy is highly inconsistent with the morphology-based taxonomy. A comparison of mitochondrial and nuclear gene genealogies showed that these are strongly incongruent with each other, while the nuclear gene genealogy is more consistent with traditional taxonomy, indicating the repeated occurrence of introgression of mitochondria across species. Here, two different cases of mitochondrial introgression among Ohomopterus species are described in detail, one for parapatric species and the other for sympatric species. First, mitochondrial haplotypes and sequences were studied in Carabus insulicola and three taxa parapatric with C. insulicola, at least two of which hybridize with C. insulicola naturally. Among the four species studied, directional introgressions of mitochondria across boundary zones were detected. Second, in the Mt. Kongo area in central Honshu, which harbors five species, introgression of mitochondria among four out of the five species was detected, despite the apparent absence of on-going natural hybridization. These inferred cases of mitochondrial introgression indicate that species interactions through hybridization could have played an important role at various stages in the evolution of Ohomopterus. Forty taxa belonging to 36 species and four unclassified accessions of Helianthus were studied using RAPD technology. Single ten-mer primers were screened for those amplifying fragments common to several species. We found that when several species shared a common fragment, they belong to the same section of the genus. Moreover, we also found that some fragments are common to all species of the Helianthus. Most of the fragments were found to be of the same size in these species and to share the homology indicated by molecular hybridization. Out of 118 retained fragments, 33 were common to all Helianthus species, 56 were unique to perennial species of sects. Atrorubentes and Ciliares, 24 were unique to sect. Atrorubentes, 29 were unique to sect. Helianthus, whereas 0 were unique to sect. Ciliares. Each set of common or specific fragments was assumed to belong to a genome: (1)?the C genome carrying the fragments common to all species of the three sections, (2)?the H genome unique to sect. Helianthus, (3)?the P genome common to perennial species (sects. Atrorubentes and Ciliares), and (4)?the A genome unique to sect. Atrorubentes. The genomic structure was therefore HC for sect. Helianthus, CPA for sect. Atrorubentes, and CP? for sect. Ciliares. Molecular hybridizations with amplification products revealed homologies between Helianthus genomes and several other genera in the Helianthinae sub-tribe. The simple method used to characterize these fragments led to powerful tools for recognizing genomes which reconcile the section organization of the genus and the degree of difficulty in crossing perennial and annual forms.  相似文献

A molecular genetic map of cassava (Manihot esculenta Crantz)   总被引:12,自引:0,他引:12  
 A genetic linkage map of cassava has been constructed with 132 RFLPs, 30 RAPDs, 3 microsatellites, and 3 isoenzyme markers segregating from the heterozygous female parent of an intraspecific cross. The F1 cross was made between ‘TMS 30572’ and ‘CM 2177-2’, elite cassava cultivars from Nigeria and Colombia, respectively. The map consists of 20 linkage groups spanning 931.6 cM or an estimated 60% of the cassava genome. Average marker density is 1 per 7.9 cM. Since the mapping population is an F1 cross between heterozygous parents, with unique alleles segregating from either parent, a second map was constructed from the segregation of 107 RFLPs, 50 RAPDs, 1 microsatellite, and 1 isoenzyme marker from the male parent. Comparison of intervals in the male-and female-derived maps, bounded by markers heterozygous in both parents, revealed significantly less meiotic recombination in the gametes of the female than in the male parent. Six pairs of duplicated loci were detected by low-copy genomic and cDNA sequences used as probes. Efforts are underway to saturate the cassava map with additional markers, to join the male- and female-derived maps, and to elucidate genome organization in cassava.  The [2Fe-2S] protein from Azotobacter vinelandii that was previously known as iron-sulfur protein I, or Shethna protein I, has been shown to be encoded by a gene belonging to the major nif gene cluster. Overexpression of this gene in Escherichia coli yielded a dimeric protein of which each subunit comprises 106 residues and contains one [2Fe-2S] cluster. The sequence of this protein is very similar to that of the [2Fe-2S] ferredoxin from Clostridium pasteurianum (2FeCpFd), and the four cysteine ligands of the [2Fe-2S] cluster occur in the same positions. The A. vinelandii protein differs from the C. pasteurianum one by the absence of the N-terminal methionine, the presence of a five-residue C-terminal extension, and a lesser number of acidic and polar residues. The UV-visible absorption and EPR spectra, as well as the redox potentials of the two proteins, are nearly identical. These data show that the A. vinelandii FeS protein I, which is therefore proposed to be designated 2FeAvFdI, is the counterpart of the [2Fe-2S] ferredoxin from C. pasteurianum. The occurrence of the 2FeAvFdI-encoding gene in the nif gene cluster, together with the previous demonstration of a specific interaction between the 2FeCpFd and the nitrogenase MoFe protein, suggest that both proteins might be involved in nitrogen fixation, with possibly similar roles.  The objective of the current research was to generate a ploidy series of backcross progenies from a single triploid (2n=3x=24) Trifolium repens×T. nigrescens F1 hybrid (3x H-6909-5). The 3x H-6909-5 plant was highly sterile and produced no seeds from approximately 3000 reciprocal backcrosses to both parental species. Chromosome doubling by an in vitro colchicine method resulted in a marked increase in fertility. Pollen stainability was increased from 9.9% in 3x H-6909-5 to an average of 89.2% (range 87.7–90.9%) in the three chromosome-doubled 6x H-6909-5 plants. Subsequent backcrosses of 6x H-6909-5 and interbreeding of backcross derivatives resulted in an array of fertile hybrids at 4x, 5x and 7x levels and some aneuploids. The occurrence of 7x BC1F1 progeny from the T. repens×6x H-6909-5 (4x×6x) cross is the first unequivocal evidence of functional female 2n gametes in white clover. Meiotic pairing in F1 and BC1F1 progeny indicated the presence of allosyndetic pairing, suggesting that genetic exchange between the two species is possible. Sugarcane cultivars are polyploid, aneuploid clones derived from interspecific hybridization between Saccharum officinarum and S. spontaneum. Their genome has recently started to be unravelled as a result of the development of molecular markers. We constructed an AFLP genetic map based on a selfing population of a specific cultivar, R570.Using 37 AFLP primer pairs, we detected 1,185 polymorphic markers of which 939 were simplex (segregated 3:1); these were used to construct the map. Of those 939, 887 were distributed on 120 cosegregation groups (CGs) based on linkages in coupling, while 52 remained unlinked. The cumulative length of all the groups was 5,849 cM, which is probably around one-third of the total genome length. Comparison with reference S. officinarum clones enabled us to assign 11 and 79 CGs to S. spontaneum and S. officinarum,respectively, whereas 11 CGs were probably derived from recombination between chromosomes of the two ancestral species. The patchy size of the groups, which ranges from 1 to 232 cM, illustrates the difficulty to access large portions of chromosomes, particularly those inherited from S. officinarum. Repulsion phase linkages suggested a high preferential pairing for 13 CG pairs. Out of the 120 CGs, 34 could be assigned to one of the 10 homo(eo)logy groups already defined in a previous RFLP map owing to the use of a small common marker set. The genome coverage was significantly increased in the map reported here. Implications for quantitative trait loci (QTL) research and marker-assisted breeding perspectives are discussed. Opuntia spinosissima (Martyn) Mill. (Cactaceae) is an extremely rare taxon with a single remaining wild population of 13 plants located on Little Torch Key, Florida. The plants rarely set viable seeds and most ovaries abscise without initiation of fruits. Pollination and breeding system were examined in Little Torch Key and in plant accessions located at Fairchild Tropical Garden. Four pollination treatments were carried out to test for apomixis, self-pollination, outcrossing, and autogamy; a control treatment was also monitored. Additionally, pollen viability, pollinators, and seed set and viability were investigated. On selfed, crossed, and open (control) stigmas pollen grains germinated, but the tubes usually did not reach the base of the style. This suggests that O. spinosissima has pre-zygotic self-incompatibility. None of the pairwise crosses set seed, so the extant plants were apparently not inter-compatible. Out of 173 manipulated and control flowers, only one set fruit. Although this flower was outcrossed, no pollen tubes germinated on the stigma; this suggests agamospermy, a process common in the Cactaceae. In those flowers where pollen tubes did reach the ovary they failed to penetrate ovules, suggesting ovarian inhibition or that this taxon has lost the ability to be fertilized. Most field-collected seeds were viable, but there is no seedling recruitment under natural conditions, and vegetative reproduction is common. Based on these findings, I hypothesize that O. spinosissima is a sterile polyploid and that the 13 extant plants are asexually derived from a single lineage. The monogenic dominant genetic determinism of total resistance to powdery mildew, introduced from Muscadinia rotundifolia into Vitis vinifera, was further assessed in BC4 and BC5 full-sib families. A BC5 population of 157 individuals was used to select AFLP markers linked to the resistance gene, Run1. Thirteen AFLP markers were selected and a local map was constructed around the Run1 gene. Ten markers among the 13 were found to co-segregate with the resistance gene. The usefulness of these 13 AFLP markers for the selection of Run1-carrying genotypes was further assessed through their analysis in a set of 22 Run1-carrying resistant genotypes and 16 susceptible genotypes. Three markers out of the 13 analysed were found to be absent in all susceptible genotypes and present in all resistant individuals, and may thus represent good tools for the marker-assisted selection of grapevine varieties resistant to powdery mildew. A recombination event among the markers that were originally found to co-segregate was observed in one of the resistant individuals, showing that recombination is possible in this region and may therefore be observed in larger populations.  HLA-G is a nonclassical major histocompatibility complex (MHC) class I molecule that is expressed only in the human placenta, suggesting that it plays an important role at the fetal-maternal interface. In rhesus monkeys, which have similar placentation to humans, the HLA-G orthologue is a pseudogene. However, rhesus monkeys express a novel placental MHC class I molecule, Mamu-AG, which has HLA-G-like characteristics. Phylogenetic analysis of AG alleles in two Old World primate species, the baboon and the rhesus macaque, revealed limited diversity characteristic of a nonclassical MHC class I locus. Gene trees constructed using classical and nonclassical primate MHC class I alleles demonstrated that the AG locus was most closely related to the classical A locus. Interestingly, gene tree analyses suggested that the AG alleles were most closely related to a subset of A alleles which are the products of an ancestral interlocus recombination event between the A and B loci. Calculation of the rates of synonymous and nonsynonymous substitution at the AG locus revealed that positive selection was not acting on the codons encoding the peptide binding region. In exon 4, however, the rate of nonsynonymous substitution was significantly lower than the rate of synonymous substitution, suggesting that negative selection was acting on these codons. A method based upon targetting of intro-gressed markers in a Phomopsis-resistant line (R) of cultivated sunflower, issuing from a H. argophyllus cross was used to mark the Phomopsis resistance regions. Our study was based upon 203 families derived from a cross between an inbred line susceptible to Phomopsis (S1) and the introgressed resistant line (R). Families were checked for Phomopsis resistance level in a design with replicated plots and natural infection was re-inforced by pieces of contaminated stems. Thirty four primers were employed for RAPD analysis. Out of 102 polymorphic fragments between (S1) and H. argophyllus, seven were still present in (R) suggesting that they marked introgressions of H. argophyllus into (R). The plants were scored for the presence or absence of 19 fragments obtained from five primers, and the relationships between the presence/absence of fragments in plants and Phomopsis resistance/susceptiblity in the progenies was determined by using an analysis of variance. We found that at least two introgressed regions, as well as favourable factors from sunflower, contributed to the level of Phomopsis resistance in cultivated sunflower. Genetic variation in protoplast-derived rice (Oryza sativa L.) plants was characterized using first and second generation selfed progenies. A total of 133 regenerated plants were obtained from ten protoplasts of the japonica rice cultivar Nipponbare. Sixty two regenerated plants which set enough seeds for the subsequent field tests at the next generation and were derived from five protoplasts were selected, and their selfed seeds were used as the first selfed-seed progeny generation). Fifteen plants were selected from each of the 15 lines, and their selfed seeds were used for tests at the generation. Thirty seven lines (60%) segregated plants with detrimental mutant characters of yellow-green phenotype, dwarf stature, dense and short panicle, or low seed fertility. According to the segregation patterns in the lines having mutated plants among those originated from the same protoplasts, the stages of mutation induction were estimated. Additionally, five quantitative traits were changed in almost all and lines. Varied quantitative traits of heading date, number of spikelets per panicle, and seed fertility, were in a heterozygous state. However, culm and panicle lengths showed high uniformity, whereas reduced culm and panicle lengths were caused by mutational changes in polygenes and/or multiple genes. Studies of somitogenesis in vertebrates have identified a number of genes that are regulated by a periodic oscillator that patterns the pre-somitic mesoderm. One of these genes, hairy, is homologous to a Drosophila segmentation gene that also shows periodic spatial expression. This, and the periodic expression of a zebrafish homologue of hairy during somitogenesis, has suggested that insect segmentation and vertebrate somitogenesis may use similar molecular mechanisms and possibly share a common origin. In chicks and mice expression of the lunatic fringe gene also oscillates in the presomitic mesoderm. Fringe encodes an extracellular protein that regulates Notch signalling. This, and the finding that mutations in Notch or its ligands disrupt somite patterning, suggests that Notch signalling plays an important role in vertebrate somitogenesis. Although Notch signalling is not known to play a role in the formation of segments in Drosophila, we reasoned that it might do so in other insects such as the grasshopper, where segment boundaries form between cells, not between syncytial nuclei as they do in Drosophila. Here we report the cloning of a single fringe gene from the grasshopper Schistocerca. We show that it is not detectably expressed in the forming trunk segments of the embryo until after segment boundaries have formed. We conclude that fringe is not part of the mechanism that makes segments in Schistocerca. Thereafter it is expressed in a pattern which shows that it is a downstream target of the segmentation machinery and suggests that it may play a role in segment morphogenesis. Like its Drosophila counterpart, Schistocerca fringe is also expressed in the eye, in rings in the legs, and during oogenesis, in follicle cells.  Evolutionary change in developmental mode in sea urchins is closely tied to an increase in maternal provisioning. We examined the oogenic modifications involved in production of a large egg by comparison of oogenesis in congeneric sea urchins with markedly different sized oocytes and divergent modes of development. Heliocidaris tuberculata has small eggs (95 μm diameter) and the ancestral mode of development through feeding larvae, whereas H. erythrogramma has large eggs (430 μm diameter) and highly modified non-feeding lecithotrophic larvae. Production of a large egg in H. erythrogramma involved both conserved and divergent mechanisms. The pattern and level of vitellogenin gene expression is similar in the two species. Vitellogenin processing is also similar with the gonads of both species incorporating yolk protein from coelomic and hemal stores into nutritive cells with subsequent transfer of this protein into yolk granules in the developing vitellogenic oocyte. Immunocytology of the eggs of both Heliocidaris species indicates they incorporate similar levels of yolk protein. However, H. erythrogramma has evolved a highly divergent second phase of oogenesis characterised by massive deposition of non-vitellogenic material including additional maternal protein and lipid. Maternal provisioning in H. erythrogramma exhibits recapitulation of the ancestral vitellogenic program followed by a novel oogenic phase with hypertrophy of the lipogenic program being a major contributor to the increase in egg size. The objective of this study was to examine the distribution of rDNA loci in the genus Glycine Willd. by fluorescent in situ hybridization (FISH) using the internal transcribed spacer (ITS) region of nuclear ribosomal DNA as a probe. The hybridized rDNA probe produced two distinct yellow signals on reddish chromosomes representing two NORs in 16 diploid (2n=40) species. Aneudiploid (2n=38) and aneutetraploid (2n=78) Glycine tomentella Hayata also exhibited two rDNA sites. However, the probe hybridized with four chromosomes as evidenced by four signals in two diploid species (Glycine curvata Tind. and Glycine cyrtoloba Tind.) and tetraploid (2n=80) G. tabacina (Labill.) Benth. and G. tomentella. Synthesized amphiploids (2n=80) of Glycine canescens F. J. Herm. (2n=40) and the 40-chromosome G. tomentella also showed four signals. This study demonstrates that the distribution of the rDNA gene in the 16 Glycine species studied is highly conserved and that silence of the rDNA locus may be attributed to amphiplasty during diploidization and speciation. Seeds of English and Austrian populations of bearded wheatgrass (Elymus caninus L.) and sea barley (Hordeum marinum Huds.) growing in the vicinity of wheat (Triticum aestivum L.) fields were collected in order to search for evidence of the introgression of wheat traits into these wild relatives. Seeds were sown and plants grown for subsequent analyses using morphological and genetic (isozymes, RAPD and wheat microsatellites) markers. No F1 hybrids were found within the individuals of the two species grown, neither with morphological nor with genetic markers. Also, no evidence of introgression of wheat traits into E. caninus was observed. However, in one individual of H. marinum which had the typical morphology of this species, numerous species-specific DNA markers of wheat were amplified, thereby demonstrating previous hybridization. Consequently, the hybridization between wheat and H. marinum under natural conditions and the introgression of wheat traits into this wild relative seems to be possible. In order to estimate the potential of gene flow between wheat (Triticum æstivum L.) and jointed goatgrass (Aegilops cylindrica Host.), we carried out mixed pollinations in experimental and natural conditions. A set of species-specific RAPD (random amplified polymorphic DNA) and microsatellite markers were used to detect the presence of parental markers in the progeny of the plants used in these experiments. No hybrids were found within the offsprings of the plants used for the greenhouse experiments, while 85 Ae. cylindrica×T. æstivum hybrids were found within 2400 analyzed F1 plants resulting from the field pollinations. The hybridization rates for individuals of different populations of the wild species differed considerably: 1% for two populations known for more than 90 years versus 7% for a newly discovered population. Most of the hybrids were completely sterile, but five of them produced 13 seeds (BC1) by backcross with Ae. cylindrica. Twelve seeds germinated and generated viable and partly fertile plants. About 25% of the wheat specific RAPD markers were found in the BC1 plants, indicating that introgression of wheat DNA into Ae. cylindrica is possible. In addition, one microsatellite marker, known to be situated on the D genome (a genome shared by both species), was also found in the BC1 plants.  相似文献
